        In this study, 80 F7:8 recombinant inbred lines (RIL), derived from a cross between dent corn and waxy corn, were evaluated for 10 grain yield and eating-related traits over a two-year period. A total of 39 quantitative trait loci (QTLs) and 74 epistatic interactions were confirmed in 2011 and 2012. All QTLs detected in 2011 and 2012, qAC9 (amylose content), qEH4 (ear height), qSEL6 (setted ear length), and q100KW10 (fresh 100 kernel weight) had higher phenotypic variance and were observed in both years; therefore, they may be considered major QTLs. We reported that the QE interaction affects (QTLs and environmental changes) for qEH4, qSEL6, and q100KW10 in discussion. Some new QTLs identified in this study were located on different loci compared with other studies. The genetic region (bin 4.08) strongly controls plant height and ear height, and results from pleiotropy and/or tight linkage. qST3 (including stem thickness) and qEH3 were co-located within two common adjacent simple sequence repeat (SSR) markers (umc2275 and umc1273), whereas qEL6 (ear length) and qSEL6 were co-located within two common adjacent SSR markers (umc2309 and bnlg238). Thus, these SSR markers are a useful selection tool for screening grain yield and yield component traits.

        Genetic resources play a great role in crop breeding because of containing a broad array of useful genes. Currently, the harder are rice breeders trying to develop new rice cultivars with the improved traits, they are more often handicapped by the limited availability of germplasm resources. Thus, a desirable core or heuristic (HS) set of germplasm with maximum genetic diversity can be usefully exploited to breakthrough the present and future challenges of the rice breeding. As such we previously developed the rice HS sets of 166 diverse accessions out of a total 24,368 rice germplasms. Here, we report a large-scale analysis of the patterns of genome-wide genetic variations accumulated in the HS as well as Korean rice over the time. We characterized a total of about 11.8 millions of single nucleotide polymorphisms (SNPs) across the rice genome from resequencing a total of 295 rice genomes including 137 HS and 158 KB rice sets, with an average of approximately 10x depth and > 90% coverage. Using about 460,000 high-quality SNPs (HQSNPs), we specified the population structure, confirming our HS set covers all the rice sub-populations. We further traced the relative nucleotide variabilities of HQSNPs and found the level of the diversity was dynamically changing across the KB genome, which reveals the selection history of KB lines in the past and present. In addition, the results of our genome wide association study (GWAS) suggests that our HS can be also a good reservoir of valuable alleles, pinpointing those alleles underlying the important rice agronomical traits. Overall, the resequencing of our HS set re-illuminates the past, present of the germplasm utilization, which will support the Korean rice breeding in the future.

        국내 밀 재배 환경에서 수발아 저항성 밀 품종 육성의 기초 자료를 제공하기 위하여 금강밀 등 31 품종을 이용하여 2년 동안 조사한 농업 형질과 종자 특성이 수발아율 및 GI에 미치 는 영향을 평가하였다. 농업형질은 출수기, 성숙기, 간장, 수 장, 망장, 경수와 1수립수를 조사하였고, 종자 특성으로는 리 터중, 천립중, 종자의 길이, 폭과 두께를 측정하였다. 수발아 관련 특성으로는 모래묻이법을 이용한 수발아율과 종자 휴면 성과 상관 있는 GI를 조사하였다. 수장과 경수는 년차간 변이 가 없었으며, 리터중은 년차간 및 품종간 차이가 없었고, 이외 나머지 농업형질과 종자 특성 및 수발아율은 년차간, 품종간 및 품종과 년차간 상호작용에서 차이가 있는 것으로 나타났 다. 농업형질은 수발아 특성과 상관이 없었지만, 출수기와 성 숙기는 1수립수 및 종자 길이와 상관을 나타냈으며, 천립중과 종자 길이는 간장과 망장과는 정의 상관을 1수립수와는 부의 상관을 나타내었다. 수장과 경수는 종자 폭 및 두께와 상관을 보였다. 천립중은 수발아율 및 GI와 정의 상관을 나타내었으 며(r = 0.618, P < 0.001, r = 0.525, P < 0.01), 종자의 길이, 폭과 두께 역시 수발아율 및 GI와 정의 상관을 나타내었다. 백립계 품종은 적립계 품종에 비하여 리터중을 제외한 모든 특성에서 높은 값을 나타내었으며, 수발아율과 GI 값도 높게 나타났다. 적립계 품종인 수안밀, 수강밀, 알찬밀과 고소밀은 수발아 저항성이면서 휴면성이 강한 품종으로, 국내 밀 육종 프로그램에서 수발아 저항성 육성 모재로 이용할 수 있을 것 이다. 국내 밀 품종의 수발아 저항성 증진을 위한 계통 선발 에서 종피색과 종실의 무게와 크기도 고려해야 한다.

        Genetically modified (GM) plant claims to be the solution to global poverty, and potentially solving environmental change and food requirement by increased human population. In this study, we were evaluating agronomic characteristics and chemical properties of two GM drought-tolerant rice (CaMsrB2-8 and CaMsrB2-23) compared with donor cultivars (Ilmi). Statistical analysis agronomic characteristics GM and donor rice showed no significant difference between both of them. Yield and appearance of rice grain, GM rice was a similar to the donor rice. Chemical composition analysis showed that GM drought-tolerant rice has no different with donor rice. This result indicated that GM drought tolerant rice has no big significant difference agronomic character and chemical properties; it can be solve food shortages in spite of drought condition.

        Hybrid sterility is a critical barrier of inter-subspecific crosses in rice. However, hybrid sterility can be overcome by wide compatibility variety. The HWC-line of rice had slender grain shape, tall culm length, wide compatibility with both indica and japonica cultivars. For QTL analysis of HWC-line, two F2 populations were derived from the crosses between the HWC-line and each of two Korean variety, Dasan (Korean Tongil-type cultivar) and Hwacheong (temperate japonica cultivar). In the cross between HWC-line/Dasan (HD), 93 STS markers and 13 SSR markers were mapped on 12 chromosomes. In the population from HWC-line/Hwacheong (HH) cross, 28 STS markers, 29 SSR markers and 1 FNP marker were mapped on 11 chromosomes. Eight agronomic characters were evaluated for QTL analysis in two F2 populations and parents. The F2 population from HD cross revealed 21 M-QTLs and 3 E-QTL for culm length, spikelet per panicle, spikelet fertility, grain length, grain width, grain shape and 100 grains weight. 8 QTLs of culm length, grain length, grain width and grain shape were newly detected in this study. In the F2 population from HH cross, 17 M-QTLs were detected for culm length, panicle length, spikelet fertility, grain length, grain width, grain shape and 100 grains weight. 6 QTLs of culm length, grain length, grain width and grain shape were newly found in this study. These QTLs will be able to provide basic information on putative functional genes related with agronomic characters and promote breeding new rice cultivar. HWC-line could be used as bridge for inter-subspecies crosses and in hybrid breeding.

        Our study is performed to confirm the level of genetic diversity and population structure with 80 maize inbred lines (40 waxy inbred lines and 40 flint inbred lines) and to explain the genetic basis of agronomic traits using an association mapping. The 200 SSR loci are confirmed a total of 1,610 alleles in total 80 maize inbred lines. The average number of alleles per locus was 8.05. The average GD was 0.72. The average PIC value was 0.68. The average MAF was 0.40. Population structure was revealed for K=2. Total 80 maize inbred lines were divided by groups I, II and admixed group. The 14 waxy inbred lines were assigned to group I. The 45 inbred lines include 5 waxy inbred lines and 40 flint inbred lines were contained to group II. The 21 waxy inbred lines were contained in the admixed group with lower than membership threshold 0.8. Association mapping between 200 SSR markers and 10 phenotypic traits of waxy/flint maize inbred lines were performed by Q GLM and Q+K MLM. In significant level at 0.01, 72 SSR markers were associated with 10 phenotypic traits using Q GLM. The 4 marker-trait association were detected in Q+K MLM. The results derived from this study will be used for designing efficient new maize breeding programs.

        Chili pepper (Capsicum annuum) is one of the most important vegetable crop for human being as a rich source of nutritions and spicy condiments. To make reference genome sequence of pepper, we sequenced the whole genome of Capsicum annuum, CM334 using Illumina/Solexa Genome Analyzer GA2. The genome size of CM334 is estimated as 3.5 Gb. A total of 716 Gb (205.96x coverages of the whole genome) of raw sequences were generated. After filtering out the low quality sequences, a total of 233 Gb (66.7x) of the raw sequences were used for assembly. Total assembled contig length and number were 2.93 Gb and 295,502, respectively. N50 and average length were 25.72 kb and 6,5 kb, respectively. By sequencial scaffolding with mate-pair sequences of 2 kb - 20 kb sizes, a total of 3.04 Gb of scaffold which is approximately 90% of the whole genome was assembled. The total number of scaffolds was 33,876 with N50 length of 1,605 kb. For annotation of the pepper genome, a total of 46 Gb of transcriptome sequences were generated from 12 different tissues using Illumina GA2 and Hi-seq 2000. We are under way of analyzing the characteristic traits of pepper using transcriptome data. The progress of pepper genome sequencing project including gene annotation, gene family analysis, comparative genomics studies on evolution of hot taste, genome expansion and fruit development will be presented in the meeting.

        국내 대학과 국립식량과학원 간의 협력 연구를 통해 확보된 32개 벼 형질전환 계통들에 대하여 주요 작물학적 특성및 수량성을 평가하였다. 이들 계통들은 수량성, 내병성, 스트레스저항성, 미질변이 등에 관여하는 17종의 유전자가 니폰바레, 낙동벼, 동진벼 등에 형질전환 되어 육성된 계통들이다. 주요 결과는 아래와 같다.1. 형질전환 계통들은 출수기, 간장, 수장, 수수 등의 주요작물학적 특성뿐만 아니라 수량성에서도 모품종과 상당한 차이를 보였다.2. 조사된 벼 형질 전환계통들에서 조사된 농업형질에 대한다변량분석 결과, 모품종에 비해 각 형질전환 계통군 내에서 관찰된 작물학적 상이성은 일정한 방향성이 없는무작위적이었으며, 그 변화폭도 3개의 모품종들의 작물학적 차이보다 훨씬 크게 관찰되었다.3. 따라서 우량 벼 형질전환 계통 육성의 효율성을 제고하기 위해서는 모품종에 형질전환 되어 발현될 유전자의우수성뿐만 아니라, 계통육성과정에서 다양한 유전학적요인에 의해 야기되는 ‘표현형 왜곡현상’을 적절히 제어할 수 있는 육종전략도 함께 고려되어야 할 것으로 사료되었다.

        본 연구는 국내‧외에서 수집한 동부 유전자원 245점에 대한 작물학적 특성을 조사하여 품종육성 기초자료로 활용하고자 수행하였으며 그 결과는 다음과 같다.1. 개화일수는 41∼50일(51.5%), 51∼60일(43.7%), 등숙일수는 21∼30일(53.9%), 31∼40일(23.7%), 생육일수는 71∼80일(26.9%), 81∼90일(23.4%) 순으로 많이분포하였다.2. 신육형은 무한형 72.7%, 중간형 25.7%, 및 유한형1.6%, 초형은 포복형 78.8%와 직립형 21.2%, 잎 모양은 심장형 98.4%와 피침(결각)형 1.6%, 꽃색은 보라색85.2%, 백색 13.6% 및 연주색 1.2% 씩 분포하였다.3. 성숙 꼬투리색은 갈색 54.7%와 황갈색 37.6%, 착협위치는 아래로 향함 90.6%, 중간 5.7% 및 곧추섬 3.7%씩 분포하였다.4. 종피색은 갈색 25.3%, 흑색 23.3%, 백색 20.8%, 종자모양은 계란형 66.9%, 장방형 24.9%, 신장형 8.2% 씩분포하였다.5. 협장은 10.1∼20.0 cm 89.0%, 20.1∼30.0 cm 8.6%,협당립수는 12.1∼15립 62.0%, 9.1∼12립 25.7%, 15.1∼18립 9.1% 씩 분포하였다.6. 백립중은 15.1∼20.0 g 37.6%, 10.1∼15.0 g 28.6%,주당수량은 100.1∼200.0 g 52.7%, 100.0 g 미만 22.9%,200.1∼300.0 g 15.9% 씩 분포하였다7. 선발한 7자원 종실의 전분 함량은 44.1∼57.0%, 단백질 함량은 23.3∼27.5% 범위로 유의차가 있었다. Sucrose함량은 자원에 따라 1.46∼2.03% 범위에 포함되어 유의차가 인정되었다.

        These studies were conducted to provide basic information on Korean ginseng cultivars and breeding lines (Panax ginseng C. A. Mey.) and to identify the variations that can be utilized in ginseng breeding programs. The agronomic characteristics was used to clarify the genetic relationships among Korean ginseng cultivars and breeding lines and to classify them into distinct genetic groups. Angle of petiole and number of fibrous root showed a wide variation from 15.0~67.8˚ and 0~5, respectively. The average plant length was 54.2cm with a range of 37.9~64.8cm and the average stem diameter was 5.6mm with a range of 4.0~7.5mm. The average stem length was 31.9cm with a range of 21.8~37.9cm and the average root weight was 38.1 g with a range of 23.0~52.0 g. The 24 Korean ginseng cultivars and breeding lines were classified into 4 groups based on agronomic characteristics using the complete linkage cluster analysis. The I, II, III and IV groups included the 60.8%, 7.4%, 13.1% and 8.7% of the cultivars and breeding lines, respectively. The breeding lines in group I could be characterized as the group with the highest growth characters and yield components, such as plant length, stem diameter and root weight. The root weight, the yield component, had highly significant positive correlations with stem diameter, plant length and stem length.

        The HWC-line of rice showed wide compatibility with both indica and japonica cultivars, tall culm length, long and slender grain shape. For QTL analysis, two F2 populations were derived from the crosses between the HWC-line and each of two Korean variety, Dasanbyeo (Korean Tongil-type cultivar) and Hwacheongbyeo (temperate japonica cultivar), respectively. A total of 190 F2 plants were evaluated in each of two F2 populations. Eight agronomic characters were measured for QTL analysis in F2 populations and parents. Two molecular linkage maps were constructed. In the F2 population from cross between HWC-line / Dasanbyeo (HD) cross, 93 STS markers and 13 SSR markers were mapped on 12 chromosomes, covering a total length of 1942.6 cM, with an average distance of 18.33cM between adjacent markers. In the F2 population from HWC-line / Hwacheongbyeo (HH) cross, 28 STS markers, 29 SSR markers and 1 FNP marker were mapped on 11 chromosomes, spanning a total length of 925.53cM, with an average distance of 15.96cM between adjacent markers. In the F2 population from HD cross, 16 M-QTLs and 1 E-QTL were detected for culm length, spikelets per panicle, spikelet fertility, grain length, grain width, grain shape and 100 grains weight. 7 QTLs of spikelet fertility, grain length, grain width and grain shape were newly identified in this study. In the F2 population from HH cross, 15 M-QTLs were detected for culm length, panicle length, spikelet fertility, grain length, grain width, grain shape and 100 grains weight. 6 QTLs of culm length, grain length, grain width and grain shape were newly identified in this study. The QTLs identified in this study would provide basic information on putative functional genes related agronomic characters and facilitate breed new rice cultivar.

        In order to clarify the chromosomal location of quantitative trait loci (QTL) associated with the yield and agronomic traits in waxy corn and sweet corn (Zea maysL.), we were conducted identifying of QTLs associated with yield and agronomic traits by employing genetic linkage map of F2:3 population. A total of 14 QTLs each for days to silking (DTS), plant height (PH), ear height (EH), ear height ratio (ER), ear length (L-Ear) and kernel setting length (L-Sear) were detected in the 158 F2 families. The number of QTL per each trait was ranged from 1 to 6, and also phenotypic variance was ranged from 3.55 to 16.86%. For DTS, one QTLs was found to be controlled by genomic regions at locations chromosomes 1 contributing 9.21% of phenotypic variance. While three QTLs for PH, were found to be controlled by 3 genomic regions at locations chromosomes 1 and 2 contributing 6.68, 6.85 and 8.17% of phenotypic variance, respectively. For EH, six QTLs were found to be controlled by 6 genomic regions at locations chromosomes 1, 7, 8 and 10 range from 3.55 to 11.44% of phenotypic variance. The one QTLs for ER was found at locations chromosomes 1 contributing 7.25% of phenotypic variance. For L-Ear, two QTLs were found to be controlled by 2 genomic regions at location chromosome 7 and 10 contributing 7.40 and 11.63% of phenotypic variance, respetively. The one QTLs for L-Sear was found at locations chromosomes 3 contributing 16.86% of phenotypic variance. Among them, three QTLs, such as qEH8 (11.44%), qLEar10 (11.63%), and qLSear3 (16.86%) may be considered as a major QTLs, while the remaining 11 QTLs might be regarded as minor QTLs. This study may provide valuable information for the further identification and characterization of genes responsible for agronomic traits in waxy corn and sweet corn.

        The earth has been facing a rapid warming during past several decades. To figure out the impact of high temperature on agronomic performance of rice especially on yield, we cultivated 89 rice varieties of various origin in Suwon Korea, Shanghai China and IRRI Philippines(Wet season and Dry season). Days to heading, culm length, panicle length, panicle number, spikelet number, spikelet fertility, grain weight and grain yield were comparatively investigated. Overall grain yield displayed significantly lower values in Shanghai and IRRI(wet and dry) compared with in Suwon. Meanwhile minimum values were much lower in Shanghai and IRRI than in Suwon. However, some varieties such as Keunseom, Taichung178 showed similar performance for grain yield in both Suwon and IRRI(wet season), and some varieties such as Hangangchal, Dasan showed similar performance for grain yield in Suwon, Shanghai and IRRI(wet season), Nampung showed very high yield in Suwon comparing to other two locations. For most varieties, grain yield was the highest in Suwon and followed by in Shanghai and at IRRI(wet season). However, in dry season at IRRI, yield trend was quite different from the expectation. Further studies are in progress to find out the genotype by environment interactions in order to obtain basic information for breeding high temperature tolerant rice.

        A Transgenic Kimch cabbage has been developed harboring T-DNAs expressing delta-endotoxin insecticidal protein, herbicide (basta) resistant protein, and antisense transcript of AsMADS2 gene. Three transgenic lines, #24, #45, and #51, originated from the same T0 plant were analyzed in terms of molecular characterization, phenotype, and agronomic traits. Flanking sequence analysis confirmed that T-DNA, with 7132 bp intact structure, was inserted onto the pseudochromosome A10 of B. rapa and all the genes in T-DNA were functionally active. Three of GM cabbage showed 69.2~75.3% of plant height and 81.8~89.7% of diameter to those of the isogenic variety ‘Nowon’, respectively. Curving upward leaf lamina attitude was observed on GM cabbage, while straight or slight concave on non-GM cabbage. In addition, an average range of 86~91.5% of head height and 87.4~94.8% of head diameter were observed on GM cabbage to those of the isogenic variety ‘Nowon’, respectively Moreover, curled inwards or slight overlap of head-forming leaf overlap at terminal region was observed on GM cabbage, but curled outwards or erect on non-GM cabbage. AsMADS2, a transcription factor reported to be involved in early flowering, was stably expressed to RNA in the GM cabbage, but it was not shown the significant influences to flowering time.

        The objective of this study were to identify QTLs for agronomic traits using a set of introgression lines carrying wild rice (Oryza rufipogon) segment in cultivated rice (ssp. japonica cv. Hwaseongbyeo). Ninety-six ILs were evaluated for seven agronomic traits, amylose and protein contents. The proportion of the recurrent genome in ILs ranged from 87.8 to 100%, with an average of 96.7%. The mean number of homozygous and heterozygous donor segments were 2 (ranging 0-7) and 1.7 (ranging 0-6), respectively, and the majority of these segments had size less than 10 cM. A total of 22 quantitative trait loci were identified for 9 traits and each QTL explained 7.2% to 56.6% of the phenotypic variance. Some QTLs were clustered in a few chromosomal regions. A first cluster was located near RM527 on chromosome 6 with QTLs for culm length, panicle length, days to heading, 1000-grain weight and protein content. Three ILs with high spikelets per panicle compared to the recurrent parent were selected to detect and fine map the wild segments responsible for this variation. The results will be discussed.

        Discovery, identification, and informatics of low molecular weight peptide are extensively rising in the field of proteomics research. In this study, we analyzed protein profiles to discover peptide based biomarker for twelve different soybean seeds with three different agronomic types using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS). For optimization of SELDI-TOF MS in soybean seed proteome analysis, four different extraction buffers were tested with urea solubilization buffer, thiourea/urea solubilization buffer, phenol extraction buffer, and modified trichloroacetic acid (TCA)/acetone precipitation/urea solubilization extraction buffer. Two different type of ProteinChip arrays, cation exchange (CM10) and anion exchange (Q10), applied to profile peptides. Among the four different extraction buffers, phenol extraction was selected to protein extraction methodology. Numbers of detected peak cluster in twelve soybean seeds were 125 at CM10 and 90 at Q10 array in the mass range from 2 to 40 kDa. Among them, 82 peak clusters at CM10 and 33 peak clusters at Q10 array showed significantly different peak clusters at p<0.00004 (CM10) and p<0.00005 (Q10) among twelve different soybean cultivars. Moreover, 29 peak clusters at CM10 and 17 peak clusters at Q10 array were detected in all cultivars as an ‘universally existed peptide’. In comparison with three different agronomic types, total of 55 peak clusters (CM10) and 23 peak clusters (Q10) were significantly different peak clusters at p<0.00004 and p<0.0001, respectively. In these probability levels, soybean seeds were well discriminated into different cultivar and different type with each other. Also we could find several specific peptide biomarkers for agronomic type.
