        Background: Sperm quality and the number of sperm introduced into the uterus during artificial insemination (AI) are pivotal factors influencing pregnancy outcomes. However, there have been no reports on the relationship between sperm concentration at AI and sperm quality in Hanwoo cattle. In this study, we examined sperm quality and pregnancy rates after AI using sperm inseminated at different concentrations. Methods: We evaluated the motility, viability, and acrosomal membrane integrity of sperm at different concentrations (10, 15, 18, and 20 million sperm/straw) in 0.5-mL straws. Subsequently, we compared the pregnancy rates after AI with different sperm concentrations. Results: After freeze-thawing, sperm at the assessed concentrations showed similar viability and acrosomal membrane integrity. After AI, cattle in the 10 million group had significantly lower pregnancy rates compared to those in the 18 and 20 million groups. Conversely, there were no statistically significant variances observed between cattle in the 10 and 15 million groups. Conclusions: Sperm at concentrations of 10, 15, 18 and 20 million per straw exhibited comparable motility, viability, and acrosomal membrane integrity. However, a concentration of at least 18 million sperm per straw is required to achieve a consistent rate of pregnancy rate in Hanwoo cattle after AI.

        In the present study, we examined if deep uterine artificial insemination (DUAI) can improve the pregnancy rate of artificial insemination (AI) using epididymal spermatozoa (ES) in Hanwoo cattle. The estrus cycles of 88 Hanwoo cows were synchronized, and 17 cows were artificially inseminated using the DUAI method with ES, 20 cows were artificially inseminated via the uterine body (BUAI) method with ES, and as a control, 51 cows were inseminated by using the BUAI method with ejaculated spermatozoa from 1 proven bull after frozen thawing. The pregnancy rate of the DUAI method (58.8%) was higher than that of the BUAI method (25.0%, p = 0.0498). The motility of ES was examined immediately after thawing and after 3 and 6 h of incubation. The rapid progressive sperm motility of the control group was significantly higher than that of the ES group immediately after thawing and after 3 and 6 h of incubation (p < 0.05). The straight line velocity and average path velocity of the ES group after 6 h of incubation were significantly lower than those in the control group (p < 0.05). The linearity and amplitude of lateral head of ES were lower than those at 6 h (p < 0.05). The flagellar beat cross frequency and hyperactivation of ES were lower than the control spermatozoa immediately after thawing and at 3 h (p < 0.05). These motility parameters suggested that ES had a low motility and fertilization ability compared to the control spermatozoa. After frozen-thawing and 3 h of incubation, the percentage of live spermatozoa with intact acrosomes in the ES was significantly lower than that in ejaculated spermatozoa (p < 0.05). Our findings suggested that the DUAI method can overcome the low pregnancy rate of ES, despite the low motility, viability, and fertilization ability of ES.

        Sexed sperm can contribute to increase the profitability of the cow industry through the production of offspring of the craved sex, such as males for meat or females for dairy production. Therefore, the utilization of sexed sperms plays a very important role in the production of offspring of superior cattle. In this study, we examined the pregnancy rates and calves sexing proportion of male and female calves produced using AI, both performed using sexed and conventional sperm. In the result, the conception rates after ET were 73.3% (33/45) sexed semen and 52% (55/104) conventional semen. Thus, the sex ratio for sexed-semen inseminations was 70% (21/30) females for singleton births within a 272 to 292 day gestation interval. The sex ratio for conventional semen was 61% (34/56) females for births. As a result, it is suggested that the use of sex classification sperm will play a very important role in the offspring production of Korean bovine.

        본 연구는 Canada에서 수입한 동결정액으로 인공수정된 모돈의 번식성적을 조사하여 동결정액을 이용한 인공수정(동결정액 인공수정)의 효율을 개선할 수 있는 통찰력을 얻기 위해 수행되었다. 이를 위해 A(GGP) 농장에서 2016년 5월과 9월 사이 총 626회의 동결정액 인공수정과 B(GGP) 농장에서 2015년부터 2017년 기간 동안 수행한 총 2,429 동결정액 인공수정의 결과 기록을 분석하였다. A종 돈장에서 동결정액을 썼을 때 총산자수 및 실산자수는 9월 보다 5월에 높았다(p<0.05). B종돈장의 분만율, 총산자수 및 실산자수는 조사연도 간에 차이가 없었다. A종돈장 결과와 B종돈장 결과를 통합하여 분석했을 때, 분만율은 A종돈장이 높았으나(p<0.01) 총산자수와 실산자수는 두 종돈장간에 차이가 없었고, 동결정액 인공수정을 했을 때가 액상정액 인공수정을 했을 때보다 낮았다(동결정액:액상정액 인공수정에 대한 총산자수와 실산자수는 각각 10.9±0.3:13.4±0.1두 및 10.0±0.3:12.0±0.1두 이었음; p<0.01). 결론적으로, 이상의 결과는 동결정액 인공수정이 액상정액 인공수정보다 번식 효율이 낮긴 하지만 연중 적절한 시기에 동결정액 인공수정을 실시하면 번식 효율이 증가될 수도 있음을 시사한다.

        The objective of this study was to evaluate the effect of treating dairy cattle with exogenous human chorionic gonadotrophin (hCG), five (5) days post artificial insemination (AI) on serum progesterone (P4) concentration and pregnancy rate. In this experiment, five days after AI, cows were assigned randomly to two groups namely: a) treated group (67) which were administrered with 1500 IU hCG (Chorulon) and b) control group (61), which received no treatment. On day 5, 10, 15 and 20 after the artificial insemination, blood samples from a total of 8 cows (4 from each group) were collected and were analyzed for serum P4 concentration. Cows were detected for estrus according to standing heat by visual observation. Cows that were detected still in estrus after days 18-24 were re-inseminated and recorded as not pregnant (open). Pregnancy diagnosis was conducted by ultrasonographic examination and transrectal palpation of the uterus on approximately 60 days in cows that observed to be not in estrus. The conception rate in hCG treated and control groups were 52.5 and 36.1%, respectively. The results proved that there were no significant differences in conception rate between two groups (p=0.0568). However, pregnancy rates were reduced by hCG treatment. Average serum P4 concentrations did not differ between Hcg-treated and control groups on day 5 (0.377 versus 0.375 ng/ml). On day 20 serum P4 concentrations were greater in the treated group compared with the control group (3.085 versus 2.010 ng/ml). The treatment with hCG seemed to increase P4 level compared with the control. In conclusion, the results of this study showed that 1500 IU of hCG administered on 5 day post AI increased conception rate in dairy cows. This was supported by the results on serum P4 concentration which was greater in hCG treated group.

        Control mating is important aspect in bee breeding programs. The technique of artificial insemination is the possible one that can surely control mating of the selected drones with the virgin queen. This is the first time applied artificial insemination technique to control mating of A. cerana in Korea. Altogether 18 queens were artificially inseminated, and 2,000 drones of Korean A. cerana were used to evaluate amount of semen collection. Semen of A. cerana is much difficult to separate from mucus in comparing with A. mellifera. The average amount of semen can be collected from one A. cerana drone was 0.09 μl, whereas the A. mellifera was more than 6 times (0.58 μl semen per A mellifera drone). Obtaining 1 μl of semen have to collect from 11.94 drones that successful semen ejection and have to kill 17 A. cerana drones. Queens artificially inseminated with 4 μl of semen (once insemination) or 8 μl of semen (twice insemination, each with 4 μl of semen) started laying egg later than naturally mated queens 5.3 and 2.5 days, respectively. The onsets of oviposition of artificially inseminated queens were 12.5 to 15.3 days. Queens received twice inseminations started laying eggs 2.8 days earlier than those received only once insemination. Artificially inseminated queens produced exclusively brood and were similar as the naturally mated ones. The brood production of the queens received once insemination with 4 μl of semen was insignificantly different than those received twice inseminations or naturally mated ones, suggesting that one artificial insemination with 4 μl of semen is favorable.

        This study was conducted to investigate the optimal artificial insemination (AI) time with diagnostic kit at ovulation time. We already applied the patent about the protein in the cow heat mucose in external reproductive tract. And we would examine the accuracy for detection of cow heat by the kit produced with the protein. Evaluation of optimal heat detection was tried two time at 12 hrs and 24 hrs after the heat. And then, AI service also performed two times with no relation to the results of heat diagnosis by heat detection kit and pregnancy rates were checked with rectal palpation on 60th day after AI. Heat diagnostic results by kit in natural heat after 12 hrs in Hanwoo cows were showed 31.3~75.0% on positive in first heat detection and 33.3~100.0% on positve in second heat detection. In the 1st positive results were significant different (p<0.05), but 2nd positive were not. The results of heat detection showed different result on regional influence and individual cow effects. The pregnancy rates of first trial of heat detection were showed 34.4~78.7% on positive and 21.3~68.8% on negative after the diagnosis by heat detection kit. And the pregnancy rates of next trial of heat detection were showed 33.3~85.7% on positive and 14.3~66.6% on negative after the heat diagnosis. Both positive results of first trial and next trial also were showed significant different (p<0.05), but negative results were not. In positive result, first trial of total pregnancy rates was higher than the next trial of pregnancy, but there showed opposite results on negative results. In conclusion, the optimal heat detection kit is suitable to ordinary Hanwoo cows and it suggested that we have to improve the kit’s accuracy by detecting the materials like proteins related optimal AI time.

        This study was carried out to evaluate the influencing factors that affect the reproductive performance of cows at the Monirampur upazila in Jessore district of Bangladesh. A total of 224 cows were brought to the upazila livestock hospital for artificial insemination (AI). The cows were inseminated between 12 to 18 hours from the onset of estrus and data was obtained from the owner. Out of 224 cows, 133 became pregnant and 91 were non pregnant. In this study, the overall pregnancy rate was 59.29%. Among the age variability, the highest pregnancy rate (70.27%) was at the age of 4 years old. In case of breed variation, the highest pregnancy rate was observed in local breed (69.07%) compared with other crossbred cows. Hence the breed variations significantly influence the conception rate of cows. According to the parity, we found that the pregnancy rate was increasing with their parity but decreasing after 4th parity. The highest conception rate was observed in 3rd parity (67.74%) which was significantly higher than that of heifers (Parity-0). Here we also found that the types of bull semen used for AI had no significant effect for pregnancy rate. The skills of AI technician for AI to cows were significantly affecting the pregnancy rate. However, this study is not enough for rating and comment about the reproduction performance of cows. Therefore, further extensive study is needed for rating and recommendation for the cattle up gradation at that particular area.

        본 연구는 인공수정 횟수가 모돈의 번식성적에 미치는 영향을 알아보기 위하여 수행되었다. 총 48두 의 F1 모돈(Yorkshire×Landrace)을 공시하였으며, 완전임의화배치법(CRD)에 의하여 4개의 처리구에 배치되었다. 처리구는 인공수정 횟수(1회, 2회, 3회, 4회)이며, 발정확인 후 12시간 뒤부터 인공수정을 진행하였다. 모돈의 발정확인은 9시와 21시에 웅돈을 접촉시켜서 확인하였으며, 재귀발정일은 5-6일 인 모돈들을 대상으로 구배치 하였다. 모돈의 승가허용 확인 후 12시간 간격에 따라 9시와 21시에 처 리구에 따른 인공수정을 실시하였다. 인공수정 횟수가 모돈의 임신율, 수태율에는 유의적인 영향을 미 치지 않으며, 분만율에서는 3회 인공수정한 처리구가 다른 처리구들에 비해 낮은 수치를 보였다. 총산 자수, 생시자돈수, 복당체중과 같은 분만성적에서도 인공수정 횟수에 따른 통계적 차이는 나타나지 않 았다. 인공수정 횟수의 1회 감소는 7,000원의 정액비용을 절감할 수 있다. 따라서 발정확인 후 인공수 정을 1회하여도 다른 처리구들과 비교해 동일한 번식성적을 가지며, 1회 인공수정을 통한 정액비용의 감소는 농가의 생산비를 절감할 수 있을 것으로 사료된다.

        본 논문은 자연환경의 변화가 젖소의 인공수정 수태율에 미치는 영향을 분석하였다. 먼저 종속변수로는 32개 농가의 인공수정자료를 이용하여 60개월(2008년 1월~2012년 12월)의 월별수태율을 구해 패널 자료로 구성하여 이용하였다. 젖소의 인공수정 수태율에 미치는 주요한 설명변수로 자연환경 변수로는 기온, 강수량, 그리고 풍속을 이용하였고, 월별 효과를 제어하기 위해 월별더미변수도 사용하였다. 분석 결과를 보면, 자연환경 설명변수 중 기온 변수가 가장 유의성이 큰 것으로 나타났다. 평균 기온이 1℃ 상승하면 수태율은 약 –0.6% 하락하는 것으로 추정되었다. 더미변수를 활용한 추정에서는 1월, 4월, 6월, 7월, 8월의 더미변수가 유의성이 큰 것으로 나타났다. 추정치를 보면 평균 수태율은 1월의 경우 약 44%, 4월의 경우 약 33%, 6월은 약 32%, 7월은 약 31%, 8월은 약 27%인 것으로 나타났다. 수태율은 기온이 높아지는 여름이 될수록 낮아지며 겨울의 경우 수태율이 높은 것으로 분석되었다.

        Pig producers have been shown keen interest of the number of spermatozoa in a semen dose since pig artificial insemination introduce. However, determining the minimal number of spermatozoa need per AI without detrimental effect on overall reproductive performances is not an easy question to answer. To increase the efficiency of semen utilization in pig AI, optimum number of spermatozoa per dose needed to determine. The objective of this study was to determine the reproductive performance and factors that affect on-farm application of low-dose semen insemination in sows. Data were collected from Darby Genetics AI studs from 4th of June to 7th of July, 2012 (n=401). The numbers of parturition were 84, 234 and 83 in sows inseminated with doses of 1.5×109, 2.0×109 and 2.5×109 spermatozoa in 100ml extender, respectively. There were no significant differences on reproductive performances such as gestation pe-riod, total born, total born alive, stillbirth and mummy in sows inseminated with different semen doses. The average number of born alive was 10.5, 11.0 and 10.4 from sows inseminated with 1.5×109, 2.0×109 and 2.5×109 sperms, re- spectively. Also, number of spermatozoa per dose did not affect litter size (p>0.10). There were no significant differ-ences of maternal genetic line difference on gestation period, total number born, number born alive, born dead and mummy. The estimated correlation coefficients of the different semen doses with total number born, number born alive, born dead and mummy were r=—0.00, —0.01, 0.02 and 0.02, respectively. Taken together, the result of this study suggested that when semen was appropriately inseminated after induced ovulation, insemination with low-dose (1.5∼2.0×109) semen dose not adversely affect sow’s fertility.

        The efficiency of artificial insemination (AI) for horses remains unsatisfactory. It is mainly because each process of AI causes a detrimental effect on semen quality. To sustain quality of semen properly, several factors including libido of stallions and sperm damage during sperm processing and preservation should be considered. Stallions with decent libido produce a high ratio of sperm to seminal plasma in their ejaculates, which is the ideal semen composition for maintaining sperm quality. Thus, to maximize the fertility rate upon AI, stallions should be appropriately managed to enhance their libido. Seminal plasma should have a positive effect on horse fertility in the case of natural breeding, whereas the effects of seminal plasma on both sperm viability and quality in the context of AI remain controversial. Centrifugation of semen is performed during semen processing to remove seminal plasma and to isolate fine quality sperm from semen. However, the centrifugation process can also result in sperm loss and damage. To solve this problem, several different centrifugation techniques such as Cushion Fluid along with dual and single Androcoll-ETM were developed to minimize loss of sperm and to damage at the bottom of the pellet. Most recently, a new technique without centrifugation was developed with the purpose of separating sperm from semen. AI techniques have been advanced to deliver sperm to optimal region of female reproductive tract at perfect timing. Recombinant equine luteinizing hormone (reLH) and low dose insemination techniques have been developed to maximize both fertility rate and the efficiency of AI. Horse breeders should consider that the entire AI procedure should be optimized for each stallion due to variation in individual horses for a uniformed AI protocol.

        This study was conducted to investigate optimal time of artificial insemination (AI) to Hanwoo female after natural estrus. AI was occurred 12 and 24 hours after natural estrus in both heifer and multiparous recipient then pregnancy and parturition rates were estimated. Results indicated that AI performed at 24 hours after natural estrus showed significant (p<0.05) higher pregnancy rate in both heifer and multiparous recipient groups with significantly (p<0.05) higher abortion rate. However, there are no significant differences of parturition rate, twin birth and sex ratio in both heifer and multiparous recipient groups. Therefore, our results may suggest that performance of AI at 24 hours after natural estrus promise higher pregnancy rate than AI at 12 hours after natural estrus in both heifer and multiparous recipient.

        This study was carried out to investigate the influence of artificial insemination (AI) failure in 1,619 Korean native cows at Gangwon East area, Korea. The average AI failure rate was 37.02% in the cows and the highest rate was 40.85% in Yangyang-city. Based on the parity in the cows, the AI failure rate was 49.14% and 29.91% in the first and fifth parity cows, respectively. Whereas cows until fifth parity were decreased in AI failure, cows with sixth or more parity showed an upturning AI failure trends with the increasing of parity number. AI failure rate incidence according to the rump fat thickness measured by ultrasound was 28.9% and 33.4% at 5 mm to 10 mm and over than 15 mm, respectively. There was a positive correlation (0.2186) between AI failure rate of mother and that of their offspring cows. That is, offspring of dams with high AI failure rate showed also higher AI failure than those of dams having lower AI failure rate. In conclusion, these results indicate that the AI failure rate was closely related to the rump fat thickness, parity number, and conception rate of mother cows. In addition, these results might strengthen the basis to improve the reproductive performance in Korean native cows.

        The increase in the meat quality and milk production of cows, which breed Korean Native Cow (KNC) and Holstein cow, is not improving reproductive efficiency. In this study, we examined the effect of interferon (IFN) supplementation on motility of frozen-thawed semen and pregnancy rate after artificial insemination of KNC and Holstein cow. In experiment 1, we investigated the effect of IFN-tau concentration (10,000 IU and 20,000 IU) on the percentage of total motility (TM) and progressive motility (PM) of frozen-thawed spermatozoa. In experiment 2, KNC were infused 20,000 IU IFN-tau at insemination or after insemination. In experiment 3, KNC or Holstein cow were inseminated with frozen-thawed semen and infused 20,000 IU IFN-gamma or -tau after insemination. In experiment 1, the average of TM (23.9% to 30.9%) and PM (18.5% to 21.9%) were similar between control and treatments. In experiment 2, the pregnancy rates of IFN infusing times were not different from 45.8% to 53.6%. In experiment 3, the pregnancy rates of Holstein cow infused different kinds of IFN were similar (control, IFN-gamma, IFN-tau; 42.9%, 40.5%, 48.0%). In the case of KNC, however, the pregnancy rate of control was 55.6%, which was significantly lower than that of IFN-gamma (68.9%; p<0.05). Thus, IFN is effective on the improvement of reproductive efficiency, but further study is needed.

        The purpose of this study was to examine the effect of growing stages of the Korean Native Striped Bull (KNSB) on the freezability and fertility of frozen-thawed semen. First, we investigated the total motility (TM) and progressive motility (PM) according to the diluent used for semen freezing. Second, we examined the effect of the age of KNSB on semen volume, TM and PM of fresh and frozen-thawed semen. Third, we examined the effect of frozen semen from the different age of KNSB on the fertilization rate, and the artificial insemination pregnancy rate. The diluents used in this experiment were Triladyl and Tris-egg yolk extender (EYE). Semen was collected from 5 KNSB in the growing stage (15 months) and 5 adult KNSB (36 months). When Triladyl or Tris-EYE extender was used for semen freezing, there was no difference of the mean TM and the mean PM. However, the mean TM was significantly higher in Bull No. 1885 than Bull No. 4283 ( <0.05). The mean volume of semen collected from the 15-month-old bulls (2.3 ml) was significantly lower ( <0.05) than that from the 36-month-old bulls (5.0 ml). The mean semen concentration was similar for the 15-month-old ( spermatozoa/ml) and 36-month-old ( spermatozoa/ml) bulls. For the 15-month-old and 36-month-old bulls, the mean TM of fresh semen were 93.7% and 88.3%, respectively, and the mean PM were 97.0% and 88.3%, respectively; the 15-month-old bulls showed a particularly high PM ( <0.05). For the 15-month-old and 36-month-old bulls, the mean TM (56.0% and 58.0%, respectively) and the mean PM (64.0% and 70.7%, respectively) of frozen-thawed semen did not differ. The development rates of embryos after fertilization and the pregnancy rate after artificial insemination using frozen-thawed semen did not differ according to the bull's age. In summary, semen volume differed according to the bull's age, but semen concentration and survival rate, the fertilization rate, and the pregnancy rate did not differ according to the stripe bull's age. Accordingly, semen from bulls in the growing stage can be collected and frozen for the preservation and multiplication of rare livestock.

        This study was carried out to investigate artificial insemination (AI) failure status and frozen semen characteristics in Korean proven bulls‘ number (KPN) semen used for AI of Hanwoo cows in Gangwon East region (Gangneung, Donghae, Taebaek, Samcheok, Sokcho, Yangyang, Goseong). Among semen used for AI, AI failure rate showed lowest at KPN506 (27.6%), whereas highest at KPN593 (77.2%). Correlations of AI failure in between Korean proven bulls semen and cows was 0.2941, which means that AI failure rate of Korean proven bulls semen may have respectable effect on reproduction of Hanwoo cow. In addition, present study was conducted to investigate spermatozoal viability rate, ruptured acrosome rate and active mitochondria in frozen Korean proven bulls semen with flow cytometry. The semen of KPN593 showed significantly (p<0.05) higher viability rate in KPN593 (30.49%) than that in KPN637 (37.34%). Furthermore, percentage of ruptured acrosome was lower in KPN637 as 21.37% than in KPN637 (21.37%), but it was not statistically significant. In conclusion, these results indicate that choice of Korean proven bulls semen may correlate positively with conception rate in Hanwoo cow. Therefore, KPN with high AI failure rate might be avoid to increase conception rate and characteristics of frozen semen might be evaluated before its use for AI.
