        The contents of total flavonoids, total antioxidant status and superoxide scavenger activity of edible plants were measured. Among twenty one samples of leafy vegetables and teas, mugwort (Artemesia capillaris) and green teas gave high total flavonoids contents, 48.2 mg/g sample and 44.7 mg/g sample, respectively. The highest concentration of total flavonoids among eighty nine samples analyzed was 52.1 mg/g sample from propolis. When samples were tested for total antioxidant status, a kind of groundsel showed the highest Total Antioxidant Status(TAS) value, 13.4 mM among leafy vegetables and teas and green tea gave high TAS value, 11.84 mM. On the other hand, purslane gave the lowest TAS value, 0.01 mM. The superoxide scavenge activity was measured from the extracts of all edible plants. The superoxide scavenge activity of polygonatum(81.1%) and chrysantemum (78.5%) were the highest among leafy vegetables, teas and medicinalplants. On the other hand, ginseng, poria, cassiae semen and dioscoreae rhizoma showed no superoxide scavenge activities.

        한국산 씀바귀(Lactuca dentata Makino)의 ethylacetate 추출물에서 6 종류의 flavonoid 성분이 TLC에 의해서 검출 되었으며, 이들은 극성에 따라서 A에서 F가지 명명되었다. 그들 중 주성분으로 생각되는 물질 E를 column chromatography을 이용하여 분리정제 하였다. 물질 E의 녹는 점은 249.5℃-251℃ 로 나타났으며, 그 구조를 밝히기 위하여, 물질 E와 이를 가수분해한 genin에 대한 여러가지 shift reagent에서 UV와 IR분석을 하고 또한 물질 E와 그의 acetyl 유도체에 대한 NMR 분석을 하였으며 물질 E에 결합된 당은 TLC에 의하여 확인하였다. 이와같은 분석 결과에서 물질 E는 luteolin 모핵에 glucose가 결합된 luteolin-7-0-β-D-glucoside로서 확인되었다.

        본 연구에서는 쓴메밀 74개 유전자원의 종자 표현형 및 화학 형과 관련된 8개 주요형질을 평가하였으며, 그 결과를 바탕으로 주성분분석 및 군집분석을 수행하였다. 또한 rutin 고함유 자원 등 기능성 쓴메밀 육성재료로 활용가능한 유망 유전자원을 선발하였다. 쓴메밀 유전자원의 종자크기는 일반메밀보다 작은 평균 5.2 × 3.4 ㎜였으며, 종피색은 흑갈색이 45.9%로 가장 많았다. 종자모양은 달갈형과 타원형이 주를 이루었다. 쓴메밀 종자의 유용성분 평균 함량은 rutin이 1,393 ㎎/100 g DW였다. Flavonoid 함량 범위는 253-2,669 ㎎/100 g이었으며, polyphenol 함량 범위는 209-1,823 ㎎/100 g 으로 나타냈다. 쓴메밀 유전자원의 주성분 분석 결과, 제3주성분까지 적용하였을 때 전체 분산의 68.55%를 설명할 수 있었다. 제1주성분에는 rutin, flavonoid 및 polyphenol, 제2주성분에서는 종자길이, 제3주성분에서는 종자폭의 비중이 큰 것으로 나타났다. 이를 바탕으로 5개의 군집으로 구분할 수 있었으며, 유전자원을 구분하는 가장 중요한 형질로는 rutin, flavonoid 및 polyphenol이었다. 쓴메밀 유전 자원 중 5개 자원(HLB1004, HLB1005, HLB1007, HLB1009, HLB1013)이 높은 rutin 함량을 보였으며, 이러한 유용자원들은 향후 기능성 육종소재 개발에 효과적으로 활용가능할 것으로 기대된다.

        The present study were conducted to determine physiological activities and antioxidant effects [2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, reducing power, Ferric Reducing Antioxidant Power (FRAP) and Fe2+ (ferrous ion) chelating capacity] of 70% methanol, chloroform:methanol, 2:1 volume ratio (CM) and ethyl acetate extract of turmeric (Curcuma longa L.). Bioactive compound of tannin 0.125±0.007 mg Catechin Equivalent (CE)/g dry weight. Turmeric extracts yield were 70% methanol 16.54%, CM 5.64% and ethyl acetate 4.14%, respectively. Antioxidant activity of the samples exhibited a dose-dependent increase. Results showed that extraction solvent had significant effects on total flavonoid content and antioxidant effects of ethyl acetate. But ferrous ion-chelating capacity of 70% methanol extract was higher than CM and ethyl acetate extract. From the results of this study, turmeric can be utilized as a valuable and potential nutraceutical for the functional food industry.

        Background : The licorice (Glycyrrhiza uralensis) is one of the Mongolian traditional useful medicinal plants. The roots and rhizomes are rich in triterpenoids, flavonoids and glycosides. It is have been employed clinically for their antiinflammatory, antiulcer, antimicrobial and anxiolytic activities. In this study, we was undertaken to explore the phytochemical screening, antioxidant activity and total flavonoid content of G.uralensis. Methods and Results : In vitro culture related greenhouse, field growing G. uralensis and the wild type roots were dried in shade and grind in mechanical grinder. 1 g of powdered roots were extracted in 25 ㎖ of methanol with concentration of 40 ㎎/㎖. The antioxidant activity and total flavonoid contents were investigated by 1,1-diphenyl-2-picryldydrazyl (DPPH) radical-scavenging assay and aluminium trichloride method, respectively. The production of glycyrrhizic acid determined by thin layer chromatography. In the phytochemical screening showed presence of secondary metabolites flavonoids, saponins, alkaloids, terpenoids and tannin. The extract possessed potent antioxidant activity having IC50 value 2,000 ㎍/㎖ (59.97 ± 2.55) in wild type and 1,000 ㎍/㎖ (68.73 ± 3.99) in greenhouse samples. Field grown G.uralensis roots extract gave highest total flavonoid content with 30.21% compared with wild type and greenhouse grown samples (16.71% and 10.71%). Conclusion : Based on the results of investigations, G. uralensis is a potent source of novel bioactive compounds. Further investigations regarding more biological activities by high sensitive techniques such as HPLC and GC-MS need to be conducted.

        Background : Miscanthus sinensis is a C4 crop with high photosynthetic efficiency and is known as a representative cellulose-based bio-energy crop originating in East Asia such as Korea, Japan, and China. It also has a high resistance to environmental conditions such as low temperature and drying and pests. However, many studies on the antioxidant activity of the M. sinensis have not been conducted. Therefore, this study was conducted to measure total phenol and total flavonoid contents of the M. sinensis accessions collected from some countries and to select the M. sinensis accessions with rich total phenol and total flavonoid contents. The results of this study can be used as basic data for future breeding. Methods and Results : This study carried out the total phenol and total flavonoid contents analysis of M. sinensis collected from different country to preserve genetic resources. Total 205 accessions of M. sinensis were collected from the USA, China, Korea, Japan, Thailand and Zambia. Their leaves were extracted with 100% methanol and concentrated at 45℃. To determine total phenol and total flavonoid contents, we measured the total phenolic content and total flavonoid content. The highest total phenolic conten t(81.04 ± 0.02 ㎎·GAE/g) showed in the accession (PMS-178) from China. The highest total flavonoid content (32.66 ± 0.01 ㎎·QE/g) showed in the accession (KMS104) from Korea. On the other hand the lowest total phenolic content (1.11 ± 0.00 ㎎·GAE/g) and the total flavonoid content (0.81 ± 0.00 ㎎·QE/g) were obtained from the accession (M. sinensis ‘Super Stripe’) from USA. Conclusion : In this study, we analysed the total phenolic and total flavonoid contents by country of origin.

        본 연구에서는 물사과 가지 추출물 및 용매 분획물의 항산화 활성과 세포보호효과를 검색하고 유효성분 을 분리하여 화학구조를 규명하였다. 물사과 가지 추출물 및 용매 분획물의 DPPH 및 ABTS+ 라디칼 소거 활성 을 측정한 결과, 에틸아세테이트 및 부탄올 분획물에서 우수한 라디칼 소거 활성이 있음을 확인하였다. 또한 H2O2로 유도된 세포 손상에 대한 세포보호효과를 확인한 결과, 20 μg/mL의 농도에서 추출물 및 에틸아세테이 트 분획물에서 보호효과가 나타남을 확인하였다. 항산화 활성이 좋은 에틸아세테이트 분획물에서 유효성분을 찾고자 medium pressure liquid chromatography (MPLC)를 실시하여 2개의 화합물을 분리하였으며 1H 및 13C NMR 데이터 분석 및 문헌 비교를 통하여 화학구조를 동정하였다; pinocembrin (1), desmethoxymatteucinol (2). 분리된 화합물에 대한 DPPH 및 ABTS+ 라디칼 소거 활성을 측정한 결과, 화합물 1, 2 모두 농도의존적으로 우수한 라디칼 소거 활성이 나타났다. 100 μM의 농도에서 화합물 1, 2는 H2O2로 유도된 세포 손상에 대하여 세포보호효과를 나타냈다. 이러한 연구 결과를 바탕으로 물사과 가지 추출물은 천연 항산화 소재로써 활용 가능할 것이라 사료된다.

        Background : Sorghum (Sorghum bicolor) is the fifth most important crop in the world. Although it is known as a crop with many medicinal action, Studies on sorghum breeding are inferior to other crop. Therefore, it is necessary to cultivate excellent varieties of sorghum which is stable and has excellent physiological activity. So, this study was conducted to investigate the total phenolic content and the total flavonoid content of the sorghum accessions to select the sorghum accessions with rich antioxidants. The results of this study can be used as basic data for future breeding. Methods and Results : The seeds for study were supplied from the Center for Agricultural Genetic Resources of National Institute of Agricultural Sciences. The seeds were extracted with 100% methanol and concentrated at 45℃. To determine antioxidant, we measured the total phenolic content and total flavonoid content. The highest total phenolic content showed in the accession from IT No. 155552 (55600.34 ± 17.78 ㎎·GAE/g) and the total flavonoid content showed the highest in IT No. 143716 (116563.20 ± 149.39 ㎎·QE/g). On the other hand the lowest total phenolic content showed in the accession from IT No. 135777 (20.14 ± 4.29 ㎎·GAE/g) and the total flavonoid content showed the lowest in IT No. 152910 (701.25 ± 5.53 ㎎·QE/g). Conclusion : Among 300 sorghum accessions, we analysed the total phenolic contents and total flavonoid contents.

        The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1'-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

        Background : Kenaf (Hibiscus cannabinus L.) is a member of the malvaceae family and has been prescribed in traditional folk medicine in Africa and India. It showed broad biologicas activities such as hepatoprotective activity, antioxidative activity and haematinic activity. Kenaf leaves contain a lot of vegetable calcium, protein, iron and vitamins, so have high potential as functional food. Thus, the present study purposed to analyze useful substances contained in kenaf leaves and their activities and look for their possibilities as the materials of food and medicine. Methods and Results : We sowed the seeds of three varieties of kenaf, namely, Dowling, Everglade-41 and Tainung-2 at planting space of 20 × 20㎝ on the field and collected plants on the 98th , 127th and 141st days from sowing. The total polyphenol content was measured using a slightly modified Folin-Denis method, which uses the phenomenon that a phenolic substance turns blue by reaction with phosphomolybdate. Conclusion : We analyzed SOD enzyme activity related to the scavenging of superoxide anion radical (․O2-) through NBT reduction. As for the SOD activity of leaf extract by variety, the activity was 92.6%, 91.0% and 92.6%, respectively, in Tainung-2 on day 98, 127 and 141 from sowing, slightly higher than those in the other two varieties but not significantly different. The activity was slightly higher in the middle period of growth than in the harvest period, but not much different according to harvest time. As for the DPPH radical scavenging activity of leaf extract by variety, the activity was 80.87% and 80.71%, respectively, in Tainung-2 on day 98 (30th of October)and day127 (28th of September), slightly higher than the other two varieties.

        Background : Buckwheat (Fagopyrum spp.) is an annual crop belonging to the polygonaceae family and cultivated in most of Asian and European countries. Nowadays, many people take interest in the utilization of buckwheat seed because of its high nutritional and pharmaceutical values. Especially, tartary buckwheat is drawing attention for its high rutin content, which is beneficial to health. Methods and Results : Tartary buckwheat sprout (TBS) was powdered and two grams of powder was mixed with 4 ㎖ H2O in a glass petri disc (100 x 20 ㎜) and exposed to far infrared irradiation (FIR) at different temperature (80, 100, 120, 140, 160℃) for an hour each. Further, the FIR treated powdered sprout samples were suspended in 200 ㎖ of 80% ethanol (v/v) and kept overnight in a shaker at room temperature. The extracts were filtered through Advantec 5B Tokyo Roshi Kaisha Ltd., Japan and dried using a vacuum rotatory evaporator (EYLA N-1000, Tokyo, Japan) in a 40℃ water bath. Dried samples were weighed and kept at 4℃ for further analysis. Conclusion : Total polyphenol was evaluated by Folin-Ciocalteau assay and total flavonoid by aluminum nitrate colorimetric assay, while antioxidant properties were evaluated based on DPPH free radical scavenging activity, metal chelating property and total antioxidant capacity. This study showed that FIR treatment to TBS caused a decrease in total antioxidant capacity and metal chelation property. However, there was a slight increase in total polyphenol and total flavonoid content from 80 to 120℃. Similarly, DPPH free radical scavenging activity also increased in the same way as TP and TF in TBS. The HPLC result revealed that quercetin production was directly proportional to temperature, and the production (average 14.87 ㎎/g dw) of quercetin was highest at 120℃ (an hour’s treatment), which was 13.54 times higher than the control in TBS.

        This study attempted to investigate the antioxidant properties of four different species of sand dune plants (Calystegia soldanella, Messerschmidia sibirica, Vitex rotundifolia and Rosa rugosa). In order to validate the antioxidant activity of these plants, we first determined the total amount of flavonoid versus phenolic contents (TFC/TPC) and extracted crude flavonoids for measuring antioxidant activities that were determined by DPPH, ABTS and FRAP assays with radical scavenging effects. We found that highest amounts of TPC were detected in R. rugose with values of 110.20 ㎎/g (leaves) and 65.71 ㎎/g (stems), while the highest amounts of TFC in V. rotundifolia with values of 38.07 ㎎/g (leaves) and 6.55 ㎎/g (stems). We further examined how closely related the amounts of TFC/TPC in antioxidant activities and found that R. rugose has the highest activities of radical scavenging with values of 63.4 ㎍/㎖ and 51.2 ㎍/㎖ determined by DPPH and ABTS assays compared with the value of 21.2 ㎍/㎖ by FRAP assay. It is of note that there is a statistically significant correlation between the resulting antioxidant activities and the total ratio of TFC and TPC, suggesting that the different amounts of TFC/TPC may directly contribute to the various antioxidant activities.

It showed broad biologicas activities such as hepatoprotective activity, antioxidative activity and haematinic activity, Recently, immunomodulatory effect of kenaf extract has been elucidated. However, depigmenting activity from kenaf extract has not been evaluated. In the present study, the tyrosinase inhibitory effect of kenaf leaf extract was investigated before and after subjecting the extract to together with infrared(FIR) irradiation. Methods and Results : FIR irradiation involves electromagnetic waves with wavelengths ranging from 4 to 15 μM. It has been hypothesized that FIR treatment during extraction of polyphenols from plant cells stimulates exudation of chemical components in cells without destroying the cells by radiant heat and breaks covalent bonds of polymerized polyphenols resulting in the release of active, natural antioxidants with a low molecular weight. The purpose of this study was to evaluate not only the changes in antityrosinase activity but also the chemical transformation of kenaf extract exposed to FIR(Scheme 1). Conclusion: The tyrosinase inhibitory activity of kenaf xtract was evaluated after far-infrared (FIR) irradiation. The ethanol extract of kenaf was prepared and its main component was analyzed as a kaempferitrin (kaempferol-3,7-O-a-dirhamnoside). Inhibitory activity of kenaf extract was not detected in tyrosinase assay. However, tyrosinase inhibitory activity was observed in kenaf extract treated with FIR irradiation. After 60 min of FIR irradiation onto kenaf extract at 60oC, a ethanolic extract was prepared and it showed significant tyrosinase inhibitory activity (IC50=3500ppm). According to HPLC analysis, kaempferol, afzelin and minor product were detected(. The inhibitory activity may be due to the existence of kaemperol, afzelin and minor product. This study showed that FIR irradiation method can be used as a convenient tool for deglycosylation of flavonoid glycoside.
