        Collected germplasms of five representative dandelion species (Taraxacum ohwianum, T. platycarpum, T. platypecidum, T. officinale, and T. coreanum) were 104 lines from different habitates in Korea and China. Their genetic diversity was analyzed by genomic fingerprinting method using amplified fragment length polymorphism (AFLP). AFLP results of 6 primer combinations were revealed 1,176 total DNA fragments and 523 polymorphic bands with a 44.4% ratio of polymorphism. On the basis of similarity coefficient analysis by unweight pair group method with arithmetic averages (UPGMA), 104 dandelion germplasm lines were ranged from 0.64 to 0.99 and clustered distinct five group depending on the species. Furthermore, a principal coordinate analysis (PCA) by the application of multi-variate analysis indicated significantly greater differences among species than geographical origins.

        The effect on the antioxidant activity and Nitric Oxide activity production inhibitory activity of Taraxacum has not been known. Therefore, phenolics and flavonoid contents were investigated from the ethanol extracts of five different Taraxacum species. The results showed that, among the five Taraxacum, T. hallaisanensis contains the highest total phenolic and flavonoid contents. When the antioxidant activity was measured by DPPH, ABTS+ and reducing power activity, the free radical scavenging activity of T. hallaisanensis was also the highest among five Taraxacum species. However, measurement by CCK-8 assay in Raw264.7 cells indicated that the extracts of Taraxacum species have no effect on cell viability. Moreover, we also investigated the effect of Taraxacum species on NO scavenging activity in lipopolysaccharide (LPS)-stimulated Raw264.7 cells. The results clearly showed that Taraxacum species inhibited NO production, and the inhibitory effect of T. hallaisanensis was the strongest. The above results suggested that Taraxacum species affected the antioxidant and NO scavenging activity, and among the five species, antioxidant and NO scavenging activity assay of T. hallaisanensis was significantly higher than those of other four Taraxacum species. Therefore, T. hallaisanensis could be used as a potential drug with anti-oxidant and anti-inflammatory effect.

        서양민들레의 시설생산을 위해 배지조성 및 차광정도를 달리한 환경조건에 대한 생육특성과 그에 따른 생리활성 변이를 검토하고자 온실과 실험실 시험을 수행하였다. 서양민들레 시설생산을 위한 적정한 배지조합은 coco peat와 perlite를 각각 70 : 30과 50 : 50으로 혼합한 배지가 coco peat와 perlite 각각의 단독배지보다 생육 및 수량에 있어서 유의적으로 높게 나타났다(p<0.05). 차광정도에 따른 서양민들레의 초장, 근장, 엽수, 근직경, 엽면적, 엽록소 함량 및 생체중은 무차광이 50%와 70%차광보다 유의적으로 높게 나타났다(p<0.05). 총 페놀 함량과 총 플라보노이드 함량은 지하부보다는 지상부에서 높게 나타났다. 총 페놀 함량은 무차광에서 차광보다 높게 검출되었고, 총 플라보노이드 함량은 50%차광에서 가장 높았고 그 다음이 무차광과 70%차광 순으로 높았다. 한편, DPPH 라디컬 소거능과 아질산염 측정을 통한 항산화성은 추출물 농도가 증가할수록 높은 활성을 보였고 지하부보다는 지상부에서, 차광보다는 무차광에서 높은 활성을 보였다.

        Dandelion has been widely utilized for medicinal and edible purposes. This research was conducted to evaluate the effect of supplemental LED (light-emitting diode) light on germination, growth characteristics and anthocyanin content of dandelion (Taraxacum officinale) seedling using LED blue (460 nm), red (660 nm, R), blue + red (B:R=6:4) and fluorescent lamp light treatment. By LED illumination to T. officinale seed germination speed was delayed, and germination rate was the highest in the fluorescent light. The growth characteristics (plant height, number of leaves, root length and fresh weight) were greatly influenced by supplemental LED light compared with control treatment, and the growth promotion was the most effective in the red LED illumination. After 60 days of red and mixed LED light treatments, anthocyanin content of dandelion plants was significantly changed. The anthocyanin content was increased by 12~19 mg/100 g under the red LED and the mixed light conditions compared with the control and the blue LED. Results indicate that illumination with red and mix LEDs, compared with other light treatments, is beneficial for promotion of growth and anthocyanin content in dandelion.

        Ionizing radiations have been effective mutagen to overcome the limitation of the useful genetic resources in natural environment. This study was conducted to investigate an effect of proton beam on germination, growth patterns in the irradiated dandelion (Taraxacum officinale), and genetic variation in 12 morphological mutants induced in proton-beam irradiated dandelion. Percentage germination rate was drastically decreased over 250Gy. The lethal dose 50 (LD50) of germination was estimated between 250 Gy to 500 Gy. Significant decreases in growth patterns (plant height, number of leaf and fresh weight) were observed by increase of dose (Gy) of proton beam irradiation. According to the correlation analysis between dosage and growth factors, the orders of compactness of correlation were germination, plant height, fresh weight and number of leaf, respectively. Twelve morphological variants such as, dwarf, color, plastid, growth and leaf shape were screened at 50 to 250 Gy of the beam irradiation. As a result of ISSR analysis of the 12 variants, out of 33 bands detected overall, 8 bands were identified to be polymorphic with a rate of 24.2% at the control group. While 33 bands detected overall, 21 bands were identified to be polymorphic with a rate of 63.6% at the proton beam irradiation. The result indicates that the dandelion with proton beam treatment might be promoted variation at DNA level

        Dandelion has been widely used as a folkloric medicine for treatment of diverse diseases. This study was conducted to evaluate the effect of light spectrum using red (660 nm), blue (460 nm), red and blue mixed (Red : Blue = 6 : 4) LED (light emitting diodes) and fluorescent lamp on growth and functional components of dandelion (Taraxacum officinale). When LED was illuminated to T. officinale cv. Goldenboll, seed germination was delayed, and germination rate was the highest in the control (fluorescent light). The growth (plant height, root length and fresh weight) except leaf number was increased under the LED treatments compared with the control, and the growth promotion was the most effective in the red LED illumination. Total polyphenol contents in dandelion irradiated with the red and blue mixed or the red LED were 121.77 mg% or 115.36 mg%, respectively, which were greater than those in dandelion treated with blue LED and fluorescent lamp. Asparagine, proline, serine, threonine, glutamic acid and arginine were the predominant amino acids in dandelion and total amino acid was the highest under the Red LED illumination. The results indicate that application of the red and the mixed LED illumination promote growth and increase functional components during cultivation of dandelion

        This study was conducted to evaluate the effect of light spectrum using red (660 nm), blue (460 nm), red and blue mixed (Red : Blue = 6 : 4) LED (light-emitting diode), and fluorescent lamp on antioxidant activities and functional components of dandelion (Taraxacum officinale). Total polyphenol contents in dandelion irradiated with the red and blue mixed or the red LED were 121.77 mg/100 g or 115.36 mg/100 g, respectively, which were greater than those in dandelion treated with blue LED and fluorescent lamp. Asparagine showed the highest content among amino acids in leaves and roots regardless of treatments. Total amino acid was the highest when illuminated with the red LED. DPPH (1,1-diphenyl-2-picrylhydrazyl) radical-scavenging activity and SOD (super oxide dismutase) activity of ethanol extracts were increased under all of the LED treatments compared with fluorescent lamp, and the antioxidant activities were increased by the red and the mixed LED illumination. The results indicate that application of the red and the mixed LED illumination promote antioxidant activity and increase functional components during cultivation of dandelion.

        Genus Taraxacum has been widely used as a folkloric medicine for treatment of diverse diseases. The genetic diversity and relationship among 32 accessions belonging to five Taraxacum species (T. mongolicum T. coreanum, T. coreanum var. flavescens, T. officinale and T. laevigatum) which collected from field, mountain, island and seaside of Korea were evaluated using ISSR markers. A total of 142 ISSR loci detected in the overall species were all polymorphic loci (100%) and interspecies polymorphisms obtained from Korean native and naturalized species were 98.2% and 94.5%, respectively. The genetic similarity matrix (GSM) among 32 accessions ranged from 0.025 to 0.860 with an average of 0.303. According to the clustering analysis, the Korean native species and naturalized species were divided two major clusters. In addition, the different species were divided into independent groups except for the T. coreanum and T. coreanum var. flavescens, and all the 32 accessions could be classified into 7 categories. The study findings indicate that Taraxacum accessions have a high genetic diversity and the dandelion accessions as breeding materials can be effectively utilized for the improvement of Taraxacum breeding.

        서양민들레의 부위별 성분 및 생리활성 차이를 검토하고자 폴리페놀 함량, 플라보노이드 함량, 항산화성 및 세포독성을 분석하였다. Folin-Denis방법에 따른 총 페놀 함량은 꽃 추출물에서 72.0 mg kg-1으로 가장 높게 나타났으며, 그 다음이 잎, 뿌리, 꽃대 추출물 순으로 나타났다(p〈0.05). 한편, 총 플라보노이드 함량은 총 페놀 함량과 같은 경향을 보였으나 더 낮은 범위의 함량이 검출되었다. DPPH 라디컬 소거능은 추출물 농도가 증가할수록 높은 활성을 보였으며 그 중 꽃 추출물 (IC50 값 = 624.3 mg kg-1)에서 가장 높은 활성을 보였고 그 다음으로 잎, 뿌리, 꽃대 추출물 순으로 나타나(p 〈0.05), 이는 페놀 및 플라보노이드 함량이 DPPH 라디컬 소거능과 관련이 있음을 보여 주었다. MTT법에 의한 세포독성 시험에서 폐암세포주(Calu-6)와 위암세포주(SNU-601)의 세포 생존율은 꽃 추출물에서 가장 낮았고(IC50값 = 85.7과 311.4 mg kg-1), 그 다음으로 잎, 꽃대, 뿌리 추출물 순으로 뿌리 추출물이 가장 높은 생존율을 보였다. 이는 세포독성이 꽃 추출출물에서 가장 높고, 뿌리에서 가장 낮은 것을 보여 준 것이다. 결론적으로 총 페놀 함량과 총 플라보노이드 함량이 항산화성(r2=0.7280 to 0.9971)과 세포독성(r2=0.5795 to 0.9515)에 높은 상관관계가 있고, 그 함량과 활성은 식물체 부위별로 다르게 나타남을 확인하였다.

        민들레의 종류별 성분 및 생리활성 차이를 검토하고자 폴리페놀 함량, 플라보노이드 함량, 항산화성 및 항암성을 분석하였다. 민들레 종류별 1000mgkg1의 메탄올 추출물의 총 페놀 함량은 지상부 추출물이 50.2~76.8mgkg1범위로 지하부 추출물 24.9~40.0mgkg1범위보다 높게 나타났으며, 종별로는 민들레가 가장 높았고, 그 다음으로 흰민들레와 서양민들레 순으로 나타났다. 민들레의 지상부 및 지하부 추출물에서 각각 76.8과 40.0mgkg1를 보여 유의적으로 가장 높은 함량을 보였다(p < 0.05). 한편, 총 플라보노이드 함량은 총 페놀 함량과 유사한 경향을 보였으나 총 페놀 함량보다 낮은 함량 범위(6.5~36.4mgkg1)를 보였다. DPPH 라디컬 소거능은 추출물 농도가 증가할수록 높은 활성을 보였으며 민들레의 지상부와 뿌리 추출물 1,000mgkg1에서 DPPH 라디컬 소거능은 각각 89.6와 83.4%의 소거능을 보여 다른 두 종보다 높은 활성을 보였다. MTT법에 의한 세포독성 시험에서 민들레 종류별 지상부와 지하부 추출물의 폐암세포주(Calu-6)에 대한 세포 생존율은 민들레 추출물에서 가장 낮았고(IC50값 = 83.4과 66.4mgkg1), 그 다음으로 흰민들레와 서양민들레 순으로 높은 생존율을 보여 민들레가 다른 두 종에 비해 높은 세포독성이 있음을 보여 주었다. 한편, 위암세포주(SNU-601)에 대한 세포 생존율은 폐암세포주에 비해 상대적으로 높은 경향으로 이는 추출물이 더 낮은 항암활성을 갖고 있음을 나타냈다. 각 성분과 생리활성 항목간의 상관관계에 있어서 총 페놀 함량과 항산화 활성 또는 세포독성 항목간의 상관관계(r2 =0.0097~0.6213)는 총 플라보노이드 함량과 항산화 활성 또는 세포독성 항목간 상관관계(r2 =0.0027~0.4627) 보다 높게 나타났다. 결과적으로 총 페놀 함량과 총 플라보노이드 함량이 항산화성 및 세포독성 과 높은 연관성을 보이며, 그 함량과 활성은 민들레 종류에 따라 다르게 나타났다.

        This study was carried out to investigate the physiochemical and microbiological changes of dandelion during fermentation. Thirty and fifty percentage raw sugar groups (SFE30 and SFE50) were introduced into dandelions and fermented for 120 days at 15-20℃. This study was conducted to investigate the utilization of sucrose on lactic acid bacteria from dandelions and their effect on the pH, titratable acidity, microorganism and formation of organic acids in dandelions during fermentation. The number of lactic acid bacteria increased remarkably up to 15 days of fermentation and then decreased rapidly thereafter. The maximum number of lactic acid bacteria, 7.9 log CFU/mL was reached at pH 4.17 and the pH of dandelion showed a slight decrease during fermentation and decreased steadily up to 90 days to reach an optimum pH of 4.0. The titratable acidity of dandelions fermented increased during fermentation. The concentration of organic acid, amino acid and free sugar in SFE30 was higher than both SFE50 and DWE. The results of dandelions fermented were remarkably retarded in the 50% raw sugar group compared to the 30% raw sugar group.

        The study was conducted to investigate functional materials as skin whitening and anti-inflammatory agent from Taraxacum officinale and Taraxacum coreanum. The total polyphenol and flavonoid content in the ethanol extract of Taraxacum officinale were found to be 64.07mg/g and 32.46mg/g, respectively. In tyrosinase inhibitory activity, the hot water extract of Taraxacum coreanum was higher than the other extracts. However, in nitric oxide (NO) scavenging ability, the ethanol extract of Taraxacum coreanum was higher than the other extracts. the ethanol extract of Taraxacum coreanum showed strong NO production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability measurement by MTT assay and the lactate dehydrogenase (LDH) assay against L929 cell, the extracts were exhibited fine cell viabilities and normal LDH release levels as nontoxic result in sample concentration of 250~1000μg/ml. As a result, the ethanol extract and the hot water extract of Taraxacum coreanum could be applicable to functional materials for anti-inflammatory and skin whitening related fields, respectively.

        천연 갈변저해제 소재를 발굴하기 위하여 민들레와 감국을 열수 및 80% 에탄올로 추출하고, 그 추출물에 대하여 PPO 저해활성과 DPPH radical 소거능, 총페놀 함량 등을 통하여 항산화 효과를 조사하였다. 또한 추출물을 사과슬라이스에 침지하여 외관의 변화를 관찰하였다. 감국 추출물이 민들레 추출물보다 높은 PPO 저해활성을 보였으며 DPPH radical 소거능 그리고 총페놀 함량 등에 대해서 높은 함량을 나타내었다. 각 추출물에 대하여

        In this study, we investigated antioxidative activity, antibacterial activity against pathogenic strains including methicillin resistant Staphylococcus aureus (MRSA), and tyrosinase inhibitory activity in 75% ethanol extract of Taraxacum coreanum and its fractions. The total polyphenol and flavonoid contents of the extract were 238.59mg/g and 33.18mg/g and the total polyphenol and flavonoid contents of the ethyl acetate fraction were 427.81mg/g and 148.90mg/g as the highest content of fractions. In DPPH radical scavenging ability, SC50 values of the ethyl acetate and butanol fraction were 38.40μg/ml and 82.28 μg/ml, respectively. In antibacterial activity by the disc diffusion assay against S. aureus, S. epidermidis and MRSA, the ethyl acetate fraction showed stronger antibacterial activity than other fractions and the extract. Especially, the ethyl acetate fraction was exhibited effective antibacterial activity against MRSA. In the cytotoxicity measurement by MTT assay, the extract and fractions were exhibited Raw 264.7 cell viabilities of 96.32~143.21% as nontoxic result in concentration of 5~100 μg/ml. As a result, the ethyl acetate fraction of the 75% ethanol extract from T. coreanum could be applicable to functional materials for related fields.

        In this study, we established a novel somatic embryogenesis and plant regeneration system through cell suspension culture of white dandelion (Taraxacum coreanum NAKAI.). Embryogenic calli could be initiated from leaf and root explants of sterile seedlings on solid Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) after 3-week cultures. To proliferate embryogenic calli rapidly, cell suspension culture was performed with transferred to liquid MS medium with various combinations of plant growth regulators (PGRs) including 2,4-D, α-naphthalene acetic acid (NAA), indole-3-acetic acid (IAA), N6-benzylamino purine (BAP), thidiazuron (TDZ), and kinetin. During suspension cultures, embryogenic calli not only greatly proliferated, but shoot organogenesis also simultaneously occurred from the surface of somatic embryos. Among them, TDZ at lower concentration, 0.1 mg/L produced the highest efficiency of somatic embryo formation and shoot organogenesis. Rooting of embryogenic calli with adventitious shoots was done on solid MS medium containing 0.1 mg/L NAA and 0.3% activated carbon. Nearly 80% of embryogenic calli with shoot organogenesis could be rooted normal. Well-rooted plantlets were transferred into pots under a greenhouse condition, and plants derived from this system appeared phenotypically normal.

        The genetic diversity and the genetic relationship among 30 genetic resources of T. officinale and T. coreanum collected from 20 regions in Korea were evaluated by using ISSR markers. Out of 127 loci detected overall, 122 were identified to be polymorphic with a rate of 96.0% at the 30 individuals. The intraspecific polymorphism between T. officinale and T. coreanum was 92.6% and 88.2%, respectively. The genetic similarity matrix (GSM) revealed a wide range of variablility among the 30 accessions, spanning from 0.179 to 922. According to the clustering analysis, different species T. officinale and T. coreanum, were divided into independent groups and all of the accessions could be classified into 7 categories. Especially, all of the mountain collected accessions belonged to independent groups. The study findings indicate that T. officinale and T. coreanum accessions have a high genetic diversity and accordingly carry a germ-plasm qualifying as good genetic resources for breeding.
