본 연구에서는 수확 후 저장 기간에 따른 절화 장미의 수명 변화를 알아보았다. 절화장미 ‘Antique Curl’을 수확 직후 4℃ 암조건의 저장고에서 0일(무처리), 1일, 3일, 5일, 7일간 보존 용액 처리 후 습식 저장하였다. 저장이 끝난 절화는 24℃ 실 내 광환경으로 옮겨 절화 보존제 유무 조건에서 절화수명을 조사하였다. 그 결과, 절화수명은 저장기간 3일차까지는 20일 전후로 높았으나, 5일차 이후 유의적으로 감소했다. 절화 보 존제는 절화수명 연장에 효과적이지는 않았으나 절화의 꽃목 굽음 현상을 완화시켰다. 엽록소 형광(Fv/Fm)은 저장 기간과 관계없이 0.78~0.82 수준으로 나타났으며 저장기간에 의한 광 생리 기능저하는 확인되지 않았다. 결과적으로 절화장미 ‘Antique Curl’의 상업적 저장 기한은 최대 5일로 확인되었다.
절화장미 삽목묘 대량생산 효율을 높이기 위해 국내 육성 절화장미 ‘엔틱컬’(Rosa hybrida ‘Antique Curl’)을 대상으로 생장 활력이 높은 단경삽목묘 주요 절위를 분석하였다. 화경으로부터 순서대로 5매엽 이상을 가진 마디 6개를 삽목(6 절위, Level 1~6)하여 온실과 생장환경조절실에서 각각 30일간 관리 하였다. 그 결과, 삽목 절위가 기부 쪽으로 내려갈수록 발근율과 액아 생장이 억제되었고, 삽수 잎의 황화 및 낙엽 현상이 조기에 나타났다. 잎의 탄수화물 함량은 발근과 신초발아율에 영향을 주지 않았다. 온실환경에서 1~3 절위, 생장환경조절실 에서 1~2 절위의 생장이 가장 우수하였고, 모두 4절위까지는 정상적인 생장을 보였다. 결과적으로 상업용 장미 시설 생산에서 대량삽목번식을 할 때 화경으로부터 5매엽을 가진 네번 째 마디(level 4)까지 가능할 것으로 확인되었다.
‘티와이썬’은 농촌진흥청 국립원예특작과학원에서 육성한 토마토황화잎말림바이러스(Tomato yellow leaf curl virus, TYLCV) 저항성 품종이다. 모계로는 TYLCV에 저항성으로 알려진 자원 ‘08-9-59’를 도입, 세대진전하여 사용하였으며 부계로 사용한 ‘원예9002호’는 1997년 ‘트러스트’ x ‘모모타로 요쿠’ 교배조합에서 계통 분리하여 2002년까지 내병성 검정 및 계통 선발을 통해 육성하였다. 2010년 두 계통을 이용한 교배조합을 작성하였으며 2011년 내병성 분자마커 검정, 2012년 대만 AVRDC에서 담배가루이를 이용한 TYLCV 저항성 유묘검정을 실시하였다. 그 결과 TYLCV에 저항성이며 반신 위조병(Ve), 시듦병(I2, I3), 고구마뿌리혹선충(Rex), 세균성반점 병(Pto), 줄기마름병(Asc1), 토마토모자이크바이러스(ToMV)에 복합내병성으로 인정되어 ‘티와이썬’이라고 명명하였다. ‘티와 이썬’은 무한생장형에 과형은 원형이며 평균과중 240g 정도, 당도는 4.2°Brix 정도이다.
Bemisia tabaci is a species complex consist of about 40 cryptic species. This species is also a vector of at least 100 begomovirus including Tomato yellow leaf curl virus (TYLCV). TYLCV transmitted by B. tabaci in a circulative propagative manner but its molecular mechanism is remain unclear. We investigate the transcriptional response of B. tabaci MED to TYLCV infection using next generation sequencing (NGS) analysis. In total 16757 differentially expressed genes between viruliferous and non-viruliferous whiteflies were analyzed. Among them 289 transcripts found significant, where 116 up-regulated and 173 down-regulated. Our results reveal the whitefly-TYLCV relationship and provide important molecular information about virus transmission of vector insects.
Bemisia tabaci is a vector of Tomato yellow leaf curl virus (TYLCV) but Trialeurodes vaporariorum is not. To determine the effect of TYLCV acquisition on vector and non-vector, we compared various physiological characteristics between two species. Our results showed that TYLCV acquisition significantly affected B. tabaci but not T. vaporariorum. The B. tabaci increased susceptibility against thermal stress but weakened chill coma recovery, shortening of longevity, low fecundity and abbreviated developmental time by virus acquisition. At the molecular level, B. tabaci increased hsp70 and hsp90 levels by TYLCV ingestion. However, T. vaporariorum did not shown any changes of those characteristics. Therefore, the physiological manipulation of TYLCV was specific to vector species but not non-vector species.
The present study was conducted to control Botrytis growth of cut roses during transport by applying hot water dipping. Water temperatures ranging from 40 – 70℃ and dipping time from 10 – 90 s were evaluated in vitro and in vivo. Hot water dipping at 50℃ inhibited mycelial growth depending on dipping time. Water temperature over 60℃ completely inhibited the pathogen growth even at exposure of 10 s while water below 40℃ did not have any effect. However, temperature over 60℃ rapidly damaged the tissue of rose petals. Dipping time below 40 s at 55℃ showed no significant difference on electrolyte leakage compared to control. In vivo, dipping flower heads in hot water reduced the severity of Botrytis rot. Dipping for 20 – 90 s at a temperature of 50℃ was more effective than at higher or lower temperatures. Heat treatment increased respiration rate and decreased slightly solution uptake, transpiration, fresh weight of flowers but dipping below 50℃ for 20 s did not affect vase life.
흰가루병에 저항성이 있는 절화장미 품종을 육성하기위하여 2007년 수량이 많고 절화수명이 긴 다홍색의 스탠다드 ‘Aladin’ 품종과 화형이 우수하고 흰가루병에 저항성이 있는 적색 스탠다드 ‘Red Queen’ 품종을 각각모본과 부본으로 인공교배를 실시하였다. 2008년 획득한종자를 파종하여 219개의 실생 개체를 얻었으며 꽃잎수가 5개 미만인 것을 제외하고 2009-2011년까지 3차에 거쳐 특성평가를 실시한 후 ‘원교 D1-179’를 최종 선발하여 ‘Antique Curl’로 명명하였다. ‘Antique Curl’은 복색으로 노랑색(Y2C) 분홍색(R49A)이 혼합되어 있으며, 화형은 고심형, 꽃잎수는 56.7장으로 대조품종인 ‘MoonLight’보다 약 20장 더 많다. 화폭은 12.3cm로 대조품종보다 약간 작았다. 1m2당 연간 채화 가능한 수량은 135.5본이며 절화장은 65.8cm, 절화경경은 5.8mm의 절화 특성을 보였다. 또한 절화수명은 16.8일이며 흰가루병 저항성을 가지고 있으며 기호도 평가에서도 4.0을 획득하여 대조품종 보다 1.0 높은 기호도를 보였다. ‘Antique Curl’의품종보호권(품종보호: 제4456호)은 2013년 4월에 국립종자원을 통해 정식 등록되었다.
The sweetpotato whitefly Bemisia tabaci (Hemiptera: Aleyrodidae) is a disastrous pest in horticultural plants worldwide. B. tabaci is a species complex including at least 24 biotypes in the world. In Korea, B-biotype has been invaded in 1998, Q-biotype in 2005 and widely spread into the country. B. tabaci is also a vector of more than 100 plant viruses, especially begomoviruses. Since 2008, Tomato yellow leaf curl virus (TYLCV) has been invaded into Korea and severely damaged tomato cultivar in all over of the country. B. tabaci is the only vector insect of TYLCV. Here we demonstrated whether TYLCV influence on the vector physiology during virus transmission. Pesticide susceptibility of whiteflies on TYLCV acquisition was determined in B. tabaci using the two-layered parafilm feeding chamber which containing 20% sugar solution including different doses of imidacloprid. Our result showed that TYLCV-viruliferous whiteflies were more susceptible to imidacloprid ingestion than non-viruliferous whiteflies. This study suggests that plant virus can manipulate the physiological conditions of vector insects.
Cross knee curl-up is an ideal variation of abdominal curl up exercise to strengthen abdominal
musculature without excessive lumbar flexion which can increase the loads on the disc and ligaments. In addition, slowly forced expiration can facilitate the activation of the abdominal musculature. The purpose of this study was to determine the effects of slowly forced expiration on activity of abdominal muscles, such as rectus abdominis (RA), external oblique (EO), and transverse abdominis/internal oblique (TrA/IO), while cross knee curl-up. Eleven young and healthy subjects (6 males and 5 females) participated. All subjects performed the cross knee curl-up slowly forced expiration and natural breathing. Paired t-test was performed in normalized electromyogram (EMG) muscle activity of the bilateral RA, EO, and TrA/IO to compare the differences between the cross curl-up with slowly forced expiration and natural breathing. Statistical significance was set at .05. There were no significant differences in normalized EMG muscle activity of the bilateral RA, EO, and TrA/IO between the cross curl-up with slowly forced expiration and natural breathing. The finding of this study designates that slowly forced expiration does not induce increasing activity of abdominal muscle in cross knee curl-up; hence, learning step of breathing control might not be necessary to strengthen abdominal muscle in cross knee curl-up.
Abdominal curl-up exercise may excessively increased superficial neck flexor such as sternocleidomastoid (SCM) muscle. Also, the muscle activity of the abdominal muscles haven’t investigated during abdominal curl-up with craniocervical flexion (CCF). Therefore, the purpose of our study was to determine the effect of CCF on the muscle activity of the abdominal and SCM muscles during abdominal curl-up. Twelve healthy subjects (six men and six women) with no history of abdominal or lower back pain within 6 weeks were recruited. Surface electromyographic signals were collected on SCM, rectus abdominis (RA), internal oblique (IO), and external oblique (EO) muscles bilaterally during performing the traditional abdominal curl-up and the abdominal curl-up with CCF. Paired t-tests were used to compare the differences in the muscle activity of the bilateral SCM, RA, EO, and IO muscles between the traditional abdominal curl-up and the abdominal curl-up with CCF (p<.05). There was significantly lower electromyogram (EMG) activity of the both SCMs during the abdominal curl-up with CCF (Right SCM, 39.50±15.29%MVIC; Left SCM, 38.24±17.31%MVIC) than with the traditional abdominal curl-up (Right SCM, 54.85±20.05%MVIC; Left SCM, 53.18±26.72%MVIC) (p<.05). The activity of abdominal muscles were not significantly different between the traditional abdominal curl-up and the abdominal curl-up with CCF. The abdominal curl-up with CCF requires significantly less muscle activity of SCM. Consequently, the abdominal curl-up with CCF is recommended to prevent excessive activation of superficial cervical flexors during abdominal curl-up exercise.
토마토 TYLCV는 병이 발생되면 약제나 다른 방법으로 방제가 어려우므로 내병성 품종의 개발이 반드시 필요하다. 특히, TYLCV 저항성 유전자 Ty-1,2,3(부분우성)을 한 개체 내로 집적을 시켜서 저항성이 증대된 품종의 개발이 필요하다. 토마토 TYLCV 저항성 11-TY1-AV 등 7계통을 대상으로 보독인 담배가루이를 이용하여 표현형을 조사하였다. 이병성 대비(11-TY5-AV; 슈퍼썬로드)와 F3 세대 3계통 및 F2 세대 4계통을 TYLCTHV 균주를 이용하여 접종하였다. 접종 13일 후에 이병성 대비가 100% 이병증상을 나타내었으며, 이때 11-TY4-AV는 48개체 중에서 23개체가 이병되었으며, 11-TY6-AV는 42개체 중에서 14개체, 11-TY7-AV는 48개체 중에서 14개체, 11-TY8-AV는 48개체 중에서 15개체가 이병 되었다. F2 세대 4 집단 모두 p>0.05 수준에서 TYLCV 접종 후 표현형이 정상적인 유전분리비(3:1)를 보이는 것을 확인 하였다. 또, TYLCV에 저항성인 동일한 자원을 부계로 사용하더라도 모계로 어떤 것을 사용 하느냐에 따라서 저항성 정도가 다르게 나타나는 것을 확인할 수 있었다.
Sweetpotato whitefly, Bemisia tabaci, is a vector of more than 100 plant-diseased viruses, as well as a serious pest of various horticultural plants. This species harbors a primary endosymbiont Portiera along with several secondary endosymbionts such as Cardinium and Hamiltonella. We investigated whether or not TYLCV acquisition alters the densities of endosymbionts in the body of B. tabaci using quantitative real-time PCR. Our results showed that the densities of both Cardinium and Hamiltonella, but not Portiera, increased upon acquisition of TYLCV. In addition, expression of GroEL, a molecular chaperone produced by Hamiltonella, was significantly upregulated in TYLCV-infected whiteflies. Our results suggest that endosymbionts may play an important role in TYLCV transmission mechanism within the body of B. tabaci.
The sweet potato whitefly, Bemisia tabaci (Gennadius, 1889) (Hemiptera: Aleyrodidae), is one of the most important agricultural pests by direct feeding and begomovirus transmission in the world. B. tabaci species complex consisting of about 30 species worldwidely has been mainly distinguished by molecular methods because of difficulties detecting morphological differences. In Korea, distribution of two exotic groups, the Mediterranean (Q biotype) and the Middle East-Asia Minor 1 (B biotype), were confirmed by comprehensive molecular methods in 2012. Of them, the Q biotype as a vector of Tomato yellow leaf curl virus (TYLCV) has caused significant crop yield losses.
Tobacco leaf curl virus (TbLCV), one of four begomoviruses in Korea, was first reported in 2011. It damaged, although not frequent, to greenhouse tomatoes at Iksan, Gimje, Sunchang, and Jeju. The vector transmitting TbLCV, however, is not known yet. In this study, we sampled an unknown B. tabaci populations on leaves of the Japanese honeysuckle (Lonicera japonica) at Iksan in outdoor conditions. PCR analysis revealed that some populations were infected with TbLCV. We identified an unknown indigenous genetic group named as JpL and clarified its phylogenetic relationships based on nucleotide sequences of mitochondrial cytochrome oxidase I gene.
The sweetpotato whitefly, Bemisia tabaci, acts as a vector of more than 100 plant viruses. B. tabaci is known to harbor a primary endosymbiont (Portiera) and 6 secondary endosymbionts (Arsenophonus, Cardinium, Fritschea, Hamiltonella, Rickettsia and Wolbachia). These endosymbionts play important roles in the acquisition and transmission of plant viruses. Using PCR analysis, we identified endosymbiotic bacteria in various B. tabaci populations collected from different places of Korea. Distribution of endosymbionts was different according to the biotype of B. tabaci. Subsequently, their relative densities of endosymbionts were compared between TYLCV-viruliferous and non-viruliferous populations of the Q biotype using quantitative realtime PCR. We found that the densities of Portiera, Cardinium and Hamiltonella are higher in viruliferous than non-viruliferous whiteflies. Our results suggest the role of endosymbiont for the TYLCV transmission of whiteflies.
The sweetpotato whitefly, Bemisia tabaci, is a vector of more than 100 plantdiseased viruses as well as a serious pest to various horticultural crops. Virus acquisition affects the vector’s development and reproduction, but its mechanism is largely unknown. Here we compared the temperature responses between non-viruliferous and TYLCV-viruliferous Q biotype of B. tabaci. When both non-viruliferous and viruliferous whiteflies were exposed for 1 and 3 h at 4, 25, and 35°C, the mortality rate of viruliferous whiteflies is higher than nonviruliferous after exposure at 4°C and 35°C, but no differences at 25°C between them. Analysis of the expression levels of heat shock protein (hsp) genes using the quantitative realtime PCR showed that viruliferous whiteflies has higher expression in hsp70, and hsp90 at both 4°C and 35°C, but no differences at 25°C. The results suggest that vector insects may not be durable to unfavorable temperature conditions when they acquisite plant viruses.
The purposes of this study were to compare abductor hallucis (AbdH) muscle activity during toe curl exercise according to position of interphalangeal joint (IPJ). Fifteen healthy subjects with neutral foot were recruit for this study. All subjects performed toe curl exercise with towel while maintaining the IPJ in flexion (condition 1) and extension (condition 2). Toe curl exercise with towel was perform three trials for five second periods in each condition. Surface electromyography (EMG) activities were recorded from three muscles (AbdH, tibialis anterior, peroneous longus) in each condition. EMG activity was normalized to the value of maximal voluntary isometric contraction (%MVIC). The EMG activities acoording to position of IPJ were compared using a paired t-test. This study showed that the EMG activity of AbdH during toe curl exercise with IPJ extension significantly increased compared to those during toe curl exercise with IPJ flexion (p<.05). However, the EMG activity of tibialis anterior and peroneus longus were not significantly different between the conditions (p>.05). These results suggest that toe curl exercise with towel must be performed with extension of IPJ in order to strengthen intrinsic muscle in subjects with overuse injuries related to excessive pronation.
Cherry tomato ‘TY Candy’ F₁ Hybrid was bred in Tomato Life Science and Research Institute from 2008 to 2014, which has indeterminate plant type, oblong fruit shape, average fruit weight 17 ~ 22 g and sugar content brix 8 ~ 9. Female line was selected and fixed with vigorous plant, firm fruit texture, Fusarium and Cladosporium resistance. Male line was selected and fixed with medium internode, deep leaf color, high brix, Yellow leaf curl virus and Nematode resistance. F₁ combination was made and pollinated in 2011 year, hybrid selection and sample seed production were progressed in 2012 year, farmers’ field trial was conducted at major tomato cultivation area and resulted good performance in 2013 ~ 2014. The distinctive characteristics of ‘TY Candy’ are higher yield, fruit quality and TYLCV, Nematode, Cladosporium etc. multi-disease resistance compared to existed varieties.
Tomato yellow leaf curl disease is a devastating disease of tomato (Solanum lycopersicum), which is caused by begomoviruses generally referred to as tomato yellow leaf curl virus (TYLCV). The breeding for TYLCV resistance has been based on the introgression of the Ty-3 resistance locus. Knowledge about the exact location of the Ty-3 on tomato chromosome 6 is needed to understand the genomic organization of the Ty-3 locus. In this study, we conducted a genetic analysis using a segregating population derived from a cross between resistant accession S. lycopersicum “A45” and susceptible accession S. lycopersicum “A39”. The F1 plants showed resistance to TYLCV and F1 was self-pollinated to produce F2 progeny. To screen the TYLCV resistance in 145 F2 plants, a leaf agroinfiltration method was used. F2 plants showed a classical Mendelian seregation (106 resistance : 39 susceptibility) for resistance to TYLCV respectively. SCAR and CAPS markers linked to the Ty-3 were tested for genotyping F2 plants and .genotyping and agroinfiltration results were cosegregated in the newly developed F2 population.
