        This study evaluated the effects of heat treatment on gastrodin and gastrodigenin content, and antioxidant activities, in Gastrodia elata Blume. Gastrodin and gastrodigenin content was analyzed post-method validation, and antioxidant activity evaluation, including assessing total polyphenol content, DPPH, and ABTS radical scavenging activities, was done. The validation of the analysis method demonstrated excellent linearity. The limits of quantification of gastrodin and gastrodigenin were 2.89 and 3.47 μg/mL, respectively. Moreover, the results of intra- and inter-day precision analysis demonstrated relative standard deviation values, within 5%. The recovery rates for gastrodin and gastrodigenin were 97.22~98.85 and 97.99~99.91%, respectively, indicating good accuracy. Under different heat treatment conditions, gastrodin and gastrodigenin content significantly increased (p<0.05), ranging from 91.15 to 310.27 and 559.66 to 830.02 mg/100 g DW, respectively. Additionally, the total polyphenol content exhibited a significant (p<0.05) increasing trend, ranging from 1,444 to 1,798 mg/100 g DW, as the temperature and time of heat treatment increased. The DPPH and ABTS radical scavenging abilities demonstrated an increasing trend at 120℃ during heat treatment. These research findings are expected to enhance our understanding of the changes in gastrodin and gastrodigenin content, and antioxidant effects in Gastrodia elata Blume during heat treatment.

        지구 온난화 이슈에서 가장 고질적인 문제 중 하나는 온실가스의 배출이다. 다양한 온실 가스 중 가장 높은 비중을 차지하는 이산화탄소(CO2)는 이를 분리하기 위해 연구자들이 지속적으로 연구를 진행해오고 있다. 이러한 관점에서 본 연구에서는 이산화탄소 기체를 분리하기 위해 poly(vinyl alcohol) (PVA) 기반 공중합체를 제조하여 기체 분리막에 활용했다. 공중합체는 자유 라디칼 중합법을 활용했으며, 곁사슬을 위한 단량체로 아크릴산(acrylic acid)를 사용하여 PVA-g-PAA (VAA) 그래프트 공중합체를 제조했다. 본 공중합체를 이산화탄소 기체분리막에 적용한 사례는 최초이며, 폴리설폰 지지체에 복합막 형태로 제조했다. 공중합체 합성 결과는 FT-IR을 통해, 합성한 공중합체 의 거동은 TEM과 DSC, TGA를 통해 분석하였다. AA 그래프팅을 통해 공중합체는 나노 구조를 형성하며, PVA의 결정화도를 급격하게 감소시켜 이산화탄소의 용해도를 증가시켰고, 이는 이산화탄소 기체 분리 성능을 향상시켰다. 이를 통해 이산화탄소 분리막 분야에 용액-확산 및 그래프팅 방법이라는 새로운 접근법을 제시하였다.

        지구상에서 가장 오래된 해양 미세조류로 알려진 스피룰리나는 인체에 필요한 영양성분을 대부분 함유하고 있는 것으로 알려져 있다. 그 구성성분으로는 phycocyanin, chlorophyll, β-carotene과 같 은 다양한 물질을 다량 함유한다고 보고되고 있으며, 노화 및 미백효과가 있다고 알려져 있다. 본 연구에서 는 스피룰리나 정제수 추출물의 UVB로 유도된 활성산소종 ROS (reactive oxygen species) 소거능과 항산 화능을 확인하였다. 스피룰리나 정제수 추출물 0.05, 0.10, 0.50 1.0 mg/mL의 DPPH 라디칼 소거능, FRAP 환원능 및 ABTS+ 라디칼 소거능을 측정하여 항산화효과를 확인하였다. 대체실험동물모델인 Zebrafish를 이용하여 스피룰리나 정제수 추출물을 0.05, 0.10, 0.50 mg/mL 농도로 처리하여 응고율, 부 화율, 심장독성을 측정하였으며, 스피룰라나 추출물을 0.05, 0.10, 0.50 mg/mL 농도로 처리한 후, DCFH-DA로 염색하여 UVB로 유도된 ROS 저해효과를 확인하였다. 항산화효과 측정 결과 양성대조군인 ascorbic acid와 비교시 DPPH, Frpa, ABTS 모두 농도 의존적으로 항산화 효과가 있음을 확인하였다. 대체 실험동물인 Zebrafish를 이용하여 응고율과 부화율, 심박수를 측정한 결과 0.5 mg/mL을 제외한 0.05, 0.10 mg/mL에서는 대조군과 비교 시 독성이 없음을 확인 하였다. UVB로 유도된 Zebrafish의 ROS 소거 능은 양성대조군에 비해 높은 ROS 감소 효과를 나타내었다. 본 연구의 결과는 스피룰라나 정제수 추출물 이 자외선 및 피부 보호 화장품 소재로 사용가치가 있음을 시사한다.

        본 연구는 적작약 꽃 추출물의 활성산소 억제와 항염증 및 MMP-1 발현 억제능 효과에 관해 알아보고 기능성 화장품 소재로써의 가능성을 확인하고자 하였다. 본 실험 방법으로는 세포 내 ROS 측정을 통한 활성산소 억제효과와 세포 독성 평가 및 항염증 측정, HDF 세포에서의 MMP-1의 발현 억제능 효과를 측정하고자 하였다. 실험 결과, RAW 264.7 세포와 HDF 세포 내에서 ROS로 억제효과를 확인하였고, 세포 독성평가는 적작약 꽃 추출물 5, 10 μg/mL 처리 농도에서 90% 이상의 세포 생존율, 그 외 처리 농도에서는 80% 이상의 세포 생존율을 확인하였다. 또한 RAW 264.7 세포에서의 NO 생성 억제와 HDF 세포에서 MMP-1의 발현 억제능이 유의하게 억제되는 것을 확인하였다. 이상의 결과를 종합하면, 적작약 꽃 추출물의 세포 내 ROS 생성억제와 NO 생성 억제로 항산화와 항염증 효과, 피부세포에 대한 낮은 독성, MMP-1의 발현 억제를 통한 노화 효과가 확인됨에 따라 기능성 화장품 소재로써의 활용 가능성을 확인할 수 있었다.

        Calocybe indica (Purkayastha, 1974), Milky mushroom, is a relatively new to the world of mushroom industry, which is belong to Lyophyllaceae of Basidiomycota, and commonly used as edible mushroom in India and Southern Asia country. This study was conducted to investigate the free radical scavenging, skin whitening and anti-collagenase activities of methanol extract from fruiting body of C.indica cultivated in Bangladesh. In addition, the polyphenol compounds of the extract were analyzed by HPLC. The mushroom extract showed good activity in lipid peroxidation which ranged from 29.35% to 55.39% at the concentration 0.125~2.0 mg/mL. The scavenging activity of the extract on 1,1-diphenyl-2-picrylhydrazy radicals were from 20.22~83.93% at the same tested concentration, which were comparable with the positive control BHT. The hydroxyl radical scavenging activity of the extract was 76.24~93.92%. The skin whitening effect of the mushroom extract was performed with UV light protecting, tyrosinase and DOPA inhibitory activities. The methanol extract absorbed UV-B wavelength with maximum level of 0.356 at 280 nm. The tyrosinase and DOPA inhibitory activities of the extract were ranged from 30.36 ~ 66.25% and 7.13 ~ 30.25% at the concentration 0.125~2.0 mg/mL, respectively. Furthermore, anti-collagenase activity were determined by measuring collagenase and elastase inhibitory activity. The collagenase and elastase inhibitory activity of the extract were 42.77% and 51.08% at the concentration 1.0 mg/mL, respectively. The experiment results suggested that fruiting body of Calocybe indica could be used as natural skin care and antioxidant agents.

        본 연구에서는 포도즙의 양(0 g, 50 g, 100 g, 150 g, 200 g)을 다르게 첨가하여 포도 양갱을 제조하고, pH, 당도, 색도, 기계적 물성, 총 페놀 함량, DPPH, 관능평가를 실시하였다. 포도양갱의 pH는 포도즙의 첨가량이 증가할수록 pH 값이 낮아지면서 유의적인 차이를 보였으며, 당도는 대조군이 1.47 Brix로 가장 낮게 나타났고, 포도즙 첨가량이 증가할수록 당도가 점점 높아져 유의적인 차이를 보였다. 색도는 명도값(L값)과 황색도(b값)이 포도즙 첨가량이 증가할수록 감소하였으나, 적색도(a값)는 그와 반대로 증가하였다. 물성 측정 결과, 포도즙 첨가가 증가할수록 경도는 증가하는 경향을 보였지만, 탄력성, 응집성, 점착성, 씹힘성은 모두 감소하는 경향을 보였다. 총 페놀 함량은 포도즙 첨가량이 증가할수록 높은 값을 보였으며, DPPH radical 소거능도 총 페놀 측정 결과와 마찬가지로 대조군에서 가장 낮은 활성을 나타내었으며, 포도즙 첨가가 증가할수록 높은 항산화 활성을 보였다. 관능평가 결과, 색, 향, 맛 등의 전체적인 기호도에서 포도즙 150 g 첨가군이 가장 높은 점수를 받았다. 이상의 결과를 종합하여 볼 때 항산화 실험에서 포도즙 200 g 첨가군의 결과가 가장 좋았으나, 관능검사 결과, 포도즙 150 g 첨가군을 가장 선호하는 결과를 함께 고려해 볼 때는 포도즙 150 g이 첨가된 양갱시료의 실용화가 바람직 할 것으로 사료된다.

        Russula compacta, a wild mushroom, belongs to Russulaceae, Russulales of Basidiomycota. This study was conducted to evaluate the free radical scavenging, anti-inflammatory, anticholinesterase and anti-α-glucosidase effects from fruiting bodies of R. compacta extracted with methanol and hot water. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging effects, the methanol and hot water extracts showed good scavenging effects comparable with positive control, BHT. The chelating effect of methanol and hot water extracts of the mushroom were significantly higher than the positive control, BHT. The reducing power of the methanol and hot water extracts of the mushroom were lower than the positive control at the concentrations tested. In the HPLC anaysis of phenolic acids profile of the mushroom extract, 7 phenolic acids such as gallic acid, vanillin, rutin hydrate, resveratol, quercetin formononetin, and biochanin-A were detected. Nitric oxide (NO) production in lipopolysaccahride (LPS) activated RAW 264.7 cells was inhibited by 1.5-fold with the treatment of methanol extract when compared with the control. In the anti-cholinesterase activity assay, the methanol extract inhibited the acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) effects by 73.9% and 81.05% at the 1.0 mg/mL concentration, whereas galanthamine, the standard drug, inhibited the AChE and BChE activities by 97.80% and 81.12%, respectively at the same concentration. The methanol and hot water extracts of the mushroom inhibited the α-glucosidase activity by 55.44% and 62.00%, respectively at the 2.0 mg/mL concentration, while acarbose, the positive control inhibited the α-glucosidase activity by 81.81% at the 2.0 mg/mL concentration. From the experimental results, the fruiting bodies of R. compacta contained natural antioxidant, anti-inflammatory, anti-cholinesterase, and anti-diabetic substances, which might be used for health foods.

        Coprinellus miaceus, belongs to Coprinaceae of Agaricales, Basidiomycota, has been used for an edible purposes in asian countries. This experiment was initiated to evaluate the free radical scavenging, free radical scavenging, anti-acetylcholinesterase, anti-inflammatory and α-glucosidase inhibitory activities of C. micaceus fruiting bodies extracted with methanol and hot water. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the scavenging activities of methanol and hot water extracts were lower than that of positive control, BHT. The chelating effects of methanol and hot water extracts were significantly higher than positive control, BHT at the concentrations of 0.125-2.0 mg/mL. In the reducing power assay, methanol and hot water extracts exhibited the lower activities than the positive control, BHT at the 0.125-0.2 mg/mL concentration. In the HPLC analysis of phenolic acids profile of the mushroom fruiting bodies, 4 phenolic compounds including procatechuic acid, chlorogenic acid, (-)-epicatechin, and naringin were detected. The tyrosinase inhibitory activities of methanol and hot water extracts were 91.33% and 91.99% at 2.0 mg/mL concentration, while the inhibitory activity of kojic acid, the positive control, was 99.61%. Nitric oxide (NO) production in lipopolysaccahride (LPS) activated RAW 264.7 cells were inhibited by the methanol extract in a concentration dependent manner. In the acetylcholinesterase (AChE) inhibitory activity assay, methanol and hot water extracts of the mushroom inhibited the AChE by 94.64% and 74.19%, respectively at the 1.0 mg/mL concentration, whereas galanthamine, the standard drug inhibited the AChE activity by 97.80% at the same concentration. The methanol and hot water extracts of the mushroom inhibited the α-glucosidase activity by 62.26% and 67.59%, respectively at the 2.0 mg/mL concentration, while acarbose, the positive control inhibited the α-glucosidase activity by 81.81% at the same concentration. Therefore, it is concluded that fruiting bodies of C. micaceus contained natural antioxidant, anti-inflammatory, anti-acetylcholinesterase and α-glucosidase inhibitory substances which might be used for promoting human health.

        본 연구에서는 느릅나무버섯의 자실체에서 메탄올과 열수를 이용해 추출한 물질의 항산화, 항염증 및 MelanomaB16/F10 세포에서의 멜라닌 생성 저해 효과를 탐색하였다. DPPH 라디칼 소거능과 환원력을 이용해 항산화 효과를 측정한 결과 양성대조군으로 사용한 BHT에 비해 항산화 효과는 낮았지만 다른 종류의 버섯에 비해 효과가 우수한 것으로 나타났으며, 철 이온 제거 항산화 실험에서느릅나무버섯의 열수 추출물은 양성대조군인 BHT에 비해 철 이온을 효율적으로 제거하여 항산화 효과가 높게나타났다. 느릅나무버섯 자실체의 염증 저해 실험에서는배양 중인 RAW 264.7 대식세포에 자실체의 메탄올과 열수추출물을 각각 처리 한 후 염증 유도 물질인 LPS를 투여하여 추출물의 NO 생성 저해효과를 조사한 결과, 처리한 추출물의 농도 의존적으로 염증 유발의 주요 인자인NO의 양이 현저하게 감소하는 우수한 소염효과를 보였으며, carrageenan에 의해 흰쥐 뒷발에 유도된 부종을 저해하는 실험에서는 투여한 추출물의 농도가 증가함에 따라흰쥐의 뒷발에 유도된 부종의 용적도 비례적으로 감소하여 항염증 효과가 높은 것으로 나타났다. Melanoma 세포에 버섯추출물을 처리하고 생합성된 멜라닌의 양을 측정한 결과 농도 의존적으로 melanoma 세포내 멜라닌의 양이 감소하여 멜라닌 저해 효과가 높은 것으로 나타났다.따라서 느릅나무버섯 자실체의 항산화, 항염증 및 멜라닌생성 저해 선분은 앞으로 항산화제, 소염제 및 미백제로의 이용이 가능할 것으로 사료된다.

        This study investigated the antioxidant activities of Amaranth (Amaranthus spp. L.) seed extracts. Methanol and hotwater extracts of the seeds were evaluated for their total polyphenol, flavonoid, and tannin content, antioxidant activ-ities, and protective effects against oxidative stress on L-132 cells. The hot water extracts showed higher total phe-nol (4.23mg GAE/g) and tannin (0.511mg TAE/g) content, while the methanol extracts showed higher flavonoidcontent (1.53mg CE/g). The methanol extracts performed better than the water extracts for DPPH radical-scavengingactivity, while the water extracts performed better for ABTS radical-scavenging activity and SOD-like activity. WithL-132 cells, the water extracts showed strong protective effects against oxidative stress in a dose dependent mannerwith an effect three-fold higher than that of the methanol extracts at a concentration of 500µg/mL. A higher inhi-bition activity of nitric oxide in RAW 264.7 cells was observed with the water extracts than with the methanolextracts at a concentration of 250µg/mL. In summary, the water extracts showed the highest total polyphenol andtannin content and a more powerful protective effect against oxidative stress and inhibition effect of NO. Our resultssuggest that amaranth seeds may be useful as natural antioxidant compounds.

        Free-radical copolymerization of glycidyl methacrylate(GMA) and N-phthalimidoethyl acrylate(NPEA) were carried out at 60℃ in dimethylformamide(DMF) solution in the presence of benzoylperoxide(BPO) at low conversion. The polymers were characterized by IR and 1H-NMR. The compositions of the copolymer was analyzed by ultra violet(UV/Vis) spectrophotometry. The reactivity ratios of the monomer was determined by the application of Fineman-Ross(FR) and Kelen-Tudos(KT) methods. The monomer reactivity ratios of the system and Alfrey-Price's resonance effect(Q) and polar effect(e) value for NIEA were determined as follow. The reactivity ratios of the monomer obtained from FR and KT are found to be r1=0.87, r2=0.98 and r1=0.88, r2=0.99 respectively. The Q and e values of poly(GMA-co-NPEM) calculated from r1 and r2 was Q= 1.31, e=0.75 respectively.

        In the present study, acetone extract and sub-fractions of Albizia amara stem bark were evaluated for their free radical potential. The results showed that the crude extract and all the fractions exhibited antioxidant and free radical scavenging activities under different in vitro assays. Among the different fractions, the ethyl acetate fraction exhibited higher DPPH and ABTS radical scavenging activities than the standard quercetin. A. amara stem bark might be valuable source of natural antioxidants that could be used for medicinal and food applications.

        The polysaccharides from fruit body of Auricularia auricula and Tremella fuciformis were extracted using hot water, and partially purified through ethanol precipitation and dialysis. Free radical scavenging activities of the crude and purified polysaccharides were examined and compared each other. Free radical scavenging activities of the partially purified polysaccharides were higher than those of crude polysaccharides. DPPH free radical, ABTS radical and SOD-like activities of partially purified polysaccharide at 1 mg/mL of concentration from A. auricula were 61.7, 9.6 and 38.9%, respectively, while those of T. fuciformis were 9.6, 5.7 and 15.3%, respectively. Results of site and nonsite specific hydroxyl radical scavenging activities indicated that the partially purified polysaccharide fractions from A. auricula and T. fuciformis exhibited the hydroxyl radical scavenging effect by hydrogen donating ability and iron ion chelating ability. Also, reducing powers of A. auricula and T. fuciformis were 77.1 and 14.7% of BHT (0.1%) as standard, respectively. It was suggested that antioxidant activities of A. auricula were about 1.4~6.4 times higher than those of T. fuciformis due to different levels of polyphenol content.

        Flavonoids are a large family of polyphenolic compounds synthesized by plants that have a common chemical structure. Flavonoids may be further divided into subclasses, Anthocyanidins, Flavan-3- ols, Flavanones, Flavonols, Flavones, Isoflavones. Flavonoids have antioxidant and antiradical activities. The antiradical efficacy of flavonoids have established structure-activity relationships (SAR) of the antioxidant activity. Reactivity of flavonoids is highly dependent on the configuration of OH groups on the flavonoid B and C rings, there being little contribution from A ring to antioxidant effectiveness. This short article presents the current knowledge on structural aspects on their free radical reactions of flavonoids.

        Methyl methacrylate(MMA) was grafted onto microcrystalline cellulose(MCC) with ceric ammonium nitrate(CAN) as a redox initiator at the various conditions. The cellulose triacetate(CTA) composite films added MCC and MMA-grafted MCC powders were prepared on a glass plate. The graft yield(GY) and graft efficiency(GE) of the grafted MCC were calculated with the simple equations by the weight balance method. The double bond of C=O on the grafted MCC surfaces was confirmed by the fourier transform infrared spectroscopy with attenuated total reflection(FT-IT ATR) spectrophotometer. After grafting, the degree of crystallinity of cellulose powders was decresed by judging from x-ray diffraction(XRD) data. Scanning electron microscope(SEM) photos showed the only solvent and CAN solution could change the roughness of MCC powders and the effect of powder dispersions in composite matrix. The tensile strength of MCC/CTA composite films was decreased with increase of MCC powder contents. When 5% grafted MCC was added, the tensile strength of grafted MCC/CTA composite films was increased from 82.3 MPa to 97.2 MPa. The thermal property of powders was also analyzed by the thermogravimetric analysis(TGA).

Various extracts were prepared from this cultured mushroom according to three experimental settings and their antioxidant properties were examined. Antioxidant activity of extracts from Tremella fuciformis was evaluated by scavenging activity of 2,2´-azinobis-(3-ethylbenzothiazoline- 6-sulfonic acid) radical cation (ABTS+), 1,1- diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase(SOD)-like activity. Among the extracts, the chloroform extract from in experimental setting I showed the most potent radical scavenging activity in each assay, showing 56% of DPPH radical scavenging activity at 1,000 ppm and 35.7% of ABTS+ scavenging activity at 1,000 ppm. Total phenolic content were estimated in various extract by using caffeic acid, chlorogenic acid, catechin, and ferulic acid as standards. Correlations of phenolic content in each extracts with antioxidant activity will be discussed in this study.
