Mushrooms(Pleurotus ostreatus, Agaricus bisporus and Flammulina velutipes) are popular food sources in Korea, and have been reported as therapeutic foods, useful for preventing various diseases. In this study we researched HPLC conditions for the determination of nucleic acids in extracts of the three type of mushrooms. The method for nucleic acids analysis of mushrooms was developed using HPLC with UV detection. To determine the nucleic acids, mushroom extracts were extracted in hot water at 90℃ by reflux extraction for 1 hr. Then, the extracts were hydrolyzed by enzymes RP-1G and 50000G. The HPLC conditions were simple, rapid, and sensitive, and were applicable for the analysis of 4 nucleosides (cytidine, uridine, guanosine and inosine) and 3 mono-nucleotides(5`-CMP, 5`-UMP, and 5`-IMP) in the mushrooms. The nucleic acids in the mushrooms were cytidine, guanosine, inosine, uridine, 5`-CMP, 5`-IMP, and 5`-UMP. The analysis results for total nucleic acids in the mushroom extracts(Pleurotus ostreatus, Agaricus bisporus, and Flammulina velutipes) indicated levels of 25.28, 27.75, and 19.87㎎/g, respectively. In conclusion, this method can be used successfully for qualitative and quantitative analysis of nucleic acids in Pleurotus ostreatus, Agaricus bisporus, and Flammulina velutipes.

1-Deoxynojirimycin(1-DNJ) was hardly detected by general UV detector. Therefore, in this study, we analyzed 1-DNJ, a effective functional material in which was contained the leaves of Cudrania tricuspidata and its tea by gas-chromatography. Synthesized a TMS derivative by attaching trimethylsilyl group to 1-DNJ, analyzed this by GC, and could detect a good peak. The leaves of Cudrania tricuspidata contains 1154.83±56.67 ug/g.d.w of 1-DNJ and tea of Cudrania tricuspidata leaves does 8.01±0.61 ug/g.d.w. This means the contents of 1-DNJ was nearly identical to each other. The larger size of the leaves had the more contents of 1-DNJ and the middle region of collection was the highest than any other collective regions of leaves.

본 연구는 항만과 지역사회의 관계를 파악하기 위한 방법의 하나로서 항만과 해항도시 지역주민의 삶의 질(QOL)간의 관계를 분석하였다. 즉, 항만이 당해 지역 지역주민의 QOL에 어떠한 영향을 미치는지를 분석하였다. 그리고 이러한 연구내용을 진행하기 위하여 선행연구의 고찰을 통해 총 18개의 QOL 측정지표들을 선정하였다. 분석의 결과, 부산항과 인천항은 당해 지역의 QOL 측정지표인 인구 1인당 일반세출결산액, 인구 1인당 제조업체 수, 주택 보급률, 인구 1인당 금융기관 수, 인구 1인당 문화재 수, 인구 1인당 학교 수, 인구 1인당 병상 수, 인구 1인당 복지비 규모 등에 공통적으로 의미 있는 영향을 미치는 것으로 나타났다.

LC-DAD-ESI/MS를 이용하여 국내 자색벼 품종에 대해 개별 안토시아닌 조성 및 함량을 평가한 결과는 다음과 같다. 1. 자색벼 품종에서 분리된 모든 개별 안토시아닌의 화학구조는 MS fragment 패턴을 확인하여 cyanidin을 base로 한 unknown 화합물 1종을 포함, cyanidin 3,5-diglucoside, cyanidin 3-glucoside, peonidin 3-glucoside의 총 4가지 개별성분이 분리 및 동정되었다. 2. Cyanidin 3-glucoside 및 peonidin 3-glucoside이 주요성분으로 cyanidin 3-glucoside의 경우 90% 이상의 가장 높은 함량 비중을 나타냈으며, 개별성분별 평균 함량은 cyanidin 3-glucoside > peonidin 3-glucoside > cyanidin 3,5-diglucoside > unknown(cyanidin based)의 순으로 나타났다. 3. 흑진주벼의 총 안토시아닌 함량은 408 mg/100 g으로 흑남벼보다 약 6배 정도 높은 함량을 나타내었다.

Natural Ginger extract was extracted from Ginger component using ethanol as a solvent, and we tested various pharmaceutical characteristics in this extract. Characteristics experiment to use natural Ginger extract tested antimicrobial experiment using microbe, and analyzed organic and inorganic component with analysis instrument. From the result of characteristics experiment, some conclusions are obtained as follow. In this experiment, natural Ginger extract was obtained about 22.50 g-Ginger extract(extraction ratio : 1.80%) as liquid state of some viscous yellow-brown color. The result of antimicrobial experiment with staphylococcus aureus and aspergillus niger in microbe decreased more and more according to passage of reaction time. This phenomenon could know that Ginger extract has influenced to antimicrobial effect. In the result of instrumental analysis, aromatic organic components of benzimidazole(1.283), propyl isothiocyanate or isothiocyanic acid(1.477) etcs from Ginger extract were detected with GC/MS and inorganic component of Ca(27.6 ppm), K(24.08 ppm), Mg(6.03 ppm), Na(1.02 ppm), Zn(1.02 ppm), Sb(0.711 ppm) and Li(0.079 ppm) etcs from Ginger extract were detected with ICP/OES.

근적외선 분광 분석법을 이용하여 참깨의 지방함량의 신속측정 가능성에 대해서 연구하였다. 검량식의 기본 모형은 다중 회귀 모형으로 하였으며, 회귀 모형의 독립 변수의 선택은 step-wise법을 이용하였다. 1. 참깨의 121점 중 지방산 함량 분포는 palmitic acid(IT028899-7.2%, IT169359-10.0%), stearic acid(IT028897-4.5%, IT028910-7.0%), oleic acid(IT170020-32.2%, IT156371-46.5%), linoleic acid(IT156348-36.5, IT169926-49.6%), 그리고 linolenic acid(IT169316-0.2%, IT156367-0.6%)로 나타났다. 2. 지방함량을 예측하기 위해서 검량선 작성시 종래의 화학적 방법에 의한 분석치와 NIRS 분석치 와의 상관계수는 oleic acid(r2-0.834) and linoleic acid(r2-0.925)로 판단되어 linoleic acid의 이용은 가능할 것으로 판단되었다.

A simple, selective and sensitive procedure for the confirmation of 14 sulfonamide antibacterials in milk was developed. The milk samples were homogenized, extracted and deproteinized by acetonitrile and defatted by n-hexane. Analysis was performed by liquid chromatography with tandem mass spectrometry (LC-MS/MS) in positive mode for all 14 analytes. Mass spectral acquisition was performed in the multi reaction monitoring mode (MRM), selecting two structurally significant transitions per compound. The calibrations were performed in sample matrixes and the interference effect of sample matrixes on the ionization was effectively eliminated. Good linear relationship (R2=0.992～ 0.999) were observed at 6 concentrations of 2.5～100 ng/g. Satisfied recoveries (86.3～110.2%) of all sulfonamides were demonstrated in spiked milk at three levels from 5 to 20 ng/g. The limits of quantitation (LOQs) for sulfonamides ranged from 0.25～2.1 ng/g.

This study was performed to comprehend the plasma proteins expressed specifically during early pregnancy in pregnant or non-pregnant Hanwoo using proteomic analysis technique. Plasma samples (0, 2, 3, 4, 7, and 11 weeks after AI) were obtained from pregnant (P, n=3) or non-pregnant (NP, n=4) Hanwoo, respectively. To evaluate proteins differentially expressed, 2-dimensional electrophoresis (2DE) was conducted. Normalized protein spots were selected for the significant expression variation deviated over two fold in its expression level between two groups. Molecular functions of the proteins were DNA binding, protein binding, hemoglobin binding, ferrochelatase and transporter activity and arylestera, respectively. According to western blotting, haptoglobin was specifically expressed only in NP group during early pregnancy; however, paraoxonase 1 was highly expressed in pregnant group. Based on these results, pregnancy was maintained successfully by the activation of specific plasma proteins associated with immune system and antioxidant regulation during early pregnancy in Hanwoo
