To date, chemical managements of plant bacterial diseases are complicated by limitations on use of antibiotics in agriculture, antibiotic resistance development, and limited efficacy of alternative control agents. In this study, thus, we performed screening of eco-friendly farming material (notice no. 2-4-064) containing tannic acid as new antibacterial-activity against 7 plant bacterial pathogens: Pectobacterium carotovorum subsp. carotovorum (Pcc), Ralstonia solanacearum (Rs), Acidovorax avenae subsp. citrulli (Aac), Xanthomonas cirti pv. citri (Xcc), Erwinia pyrifoliae (Ep), Clavibacter michiganensis subsp. michiganensis (Cmm), and Streptomycesscabies (Sc), Initial screening of antibacterial effects of eco-friendly farming material was performed using the paper disk diffusion method and co-cultivation in broth culture. Tannic acid based eco-friendly farming material showed inhibitory effect against Pcc, Rs, Aac, Xcc, Cmm, and Ss, whereas it did not show inhibition zone against Ep. However, when it tested by co-cultivation in broth culture, it showed strong inhibition effect against all pathogens that declined growth curve compared to bacterial pathogen only. Interestingly, when we observed morphological changes on those pathogens by SEM, cell morphologies of 7 pathogens were slightly changed that seem to be malfunction in their cell envelope.
The susceptibility of the predatory mite, Neoseiulus californicus to extracts of Melia azedarach, Piper nigrum, Syringa velutina and their mixtures was evaluated in laboratory bioassays. In treatments with plant extracts tested, 76-86% of adult female predators survived after 7 days after treatment. Adult female predators treated with plant extracts tested produced 68.3-81.1% as many eggs as did control females and eclosion of eggs deposited by treated predators was not affected. Moreover, treatment of plant extracts tested showed no toxic effect on N. californicus eggs and produced 100% hatchability. Survival of N. californicus nymphs was not seriously affected by exposure to the plant extracts tested. Immatures of N. californicus survived on the leaf disc treated with plant extracts tested and 78-90% of immature predators reached adulthood. These results suggest that extracts of Melia azedarach, Piper nigrum, Syringa velutina and their mixtures expected to be promising candidates for use in integrated mite management program with N. californicus.
Drought stress is a major agricultural limitation to crop productivity worldwide, especially by which leafy vegetables, plant leaves eaten as vegetable, could be more lethal. The study was carried out to know the effect of drought tolerance plant growth promoting bacteria (PGPB) on water stress of kale seedlings. A total of 146 morphologically distinct bacterial colonies were isolated from bulk soil and rhizosphere soil of leafy vegetables and screened for plant growth promoting microbioassay in greenhouse. Out of them the isolate SB19 significantly promoted the growth of kale seedlings in increasement of about 42% of plant height (14.1 ㎝), 148% of leaf area (19.0 ㎠) and 138% of shoot fresh weight (1662.5 ㎎) attained by the bacterially treated plants compared to distilled water treated control (9.9 ㎝, 7.7 ㎠, 698.8 ㎎). Shoot water content of SB19 treated kale seedlings (1393.8 ㎎) was also increased about 152% compared with control (552.5 ㎎). The SB19 isolated from bulk soil of kale plant in Iksan, Korea, was identified as species of Bacillus based on 16S rRNA gene sequencing analysis. We evaluated the effect of drought tolerance by the Bacillus sp. SB19 on kale seedlings at 7th and 14th days following the onset of the water stress and watering was only at 7th day in the middle of test. In the survey of 7th and 14th day, there were mitigation effect of drought stress in kale seedlings treated with 106 and 107 cell mL-1 of SB19 compared to distilled water treated control. Especially, there were more effective mitigation of drought damage in kale seedlings treated with 107 cell mL-1 than 106 cell mL-1. Further, although drought injury of bacterially treated kale seedlings were not improved at 14th day compared with 7th day, drought injury of 107 cell mL-1 of SB19 treated kale seedlings were not happen rapidly but developed over a longer period of time than 106 cell mL-1 of SB19 or control. The diffidence of results might be caused by the concentration of bacterial suspension. This study suggests that beneficial plant-microbe interaction could be a important role of enhancement of water availability and also provide a good method for improving quality of leafy vegetables under water stress conditions.
The purpose of this study was to assess drought tolerance of groundcover plants for extensive green roof. Form July to November 2013, plant height, chlorophyll content, soil moisture, evapotranspiration and survival rate was measured. As results, after unirrigation started, Aster koraiensis reached 0% of soil moisture earliest followed by Sedum kamtschaticum, Hemerocallis fulva, Hylotelephium spectabile, Iris sanguinea, Hosta longipes and control. Among the plants, A. koraiensis withered earliest on 19th day of unirrigation experiment, followed by H. longipes and H. fulva (27th day); and I. sanguinea (29th day). S. kamtschaticum and H. spectabile survived even after 120th day of the experiment. As for chlorophyll content, I. sanguinea showed constant values in early stage than rapidly decreased right before its withering. Chlorophyll content of A. koraiensis and H. fulva showed rapid decrease from the beginning, while that of H. spectabile and S. kamtschaticum showed continuous reduction for first 30 days and the reduction was slowed down since then. In conclusion, H. longipes, I. sanguinea and H. fulva were found most applicable for extensive green roof considering scenic aspect.
우리나라의 식품관련 폐기물정책은 식품자원 흐름의 하류단계인 가정과 식당에서 발생하는 음식물쓰레기를 중심으로 추진되고 있으나 동 폐기물은 성상이 균질하지 않고, 계절적으로 성상이 변동하며, 이물질이 섞여 있어 재활용의 가치가 적다. 한편 식품제조업체에서 발생하는 식품폐기물은 성상이 균질하고 발생원 1개소 당 발생량이 많아서 가정과 식당에서 발생하는 음식물쓰레기에 비하여 재활용의 경제적 효과가 크고 가축의 사료로 재활용할 수 있어 부가가치가 높다. 우리나라의 폐기물관리법에서는 식품제조업에서 발생하는 식품폐기물은 동식물성잔재물로 분류하여 관리하고 있으며, 연간 1,000톤 이상 배출하는 다량배출사업장은 사업장폐기물 감량화제도에 의해 감량과 재활용계획을 수립하고 그 결과를 보고하도록 하고 있으나, 이보다 배출량이 적은 업체에 대해서는 중앙정부나 지방정부 차원에서 감량이나 재활용 정책을 추진하고 있지 않다. 본 연구에서는 식품제조업체 중 전분제품과 당류 제조업, 동물성 및 식물성 유지 제조업을 대상으로 동식물성잔재물이 발생 및 처리실태를 파악하였으며, 배출에서 재활용 및 사용, 처리의 전 과정의 물질흐름을 추정하고, 동 폐기물의 재활용에 대한 경제적 효과를 분석하였다. 우리나라의 동식물성잔재물은 유상으로 판매하는 것과 무상이나 처리비를 지급하면서 위탁 처리하는 경우가 있었다. 또한, 참깨박과 들깨박과 같이 부패성이 없고 유상으로 재활용업체에 판매되어 사료로 생산되고 있었으며, 수요가 안정적으로 확보되어 있어서 방치될 우려가 없기 때문에 순환자원으로 인정하여 폐기물에서 제외하여도 관리상에 문제가 없는 것도 있었다. 또한, 일본의 식품폐기물의 재활용관련 법제도를 분석한 결과 식품재활용사업계획인정제도는 국내의 식품제조업체에서 발생하는 동식물성잔재물의 재활용을 활성화하기 도입이 필요한 것으로 판단되었다.
The Wnt/β-catenin signaling pathway is mandatory in adipogenesis. In this study, we investigated the applicability of functional materials for the treatment of obesity by examining Wnt/β-catenin pathway reporter activity related to adipocyte differentiation inhibiting with korean native plant extracts. The luciferase activity of HEK 293-TOP cells increased the reporter activity approximately 152% and 130% by treatment with Sanguisorba officinalis and Thuja orientalis, respectively. Ricinus communis were represented about 90% higher activity, two samples(Rosa rugosa and Sophorae Flos) showed 80% higher activity than the control. Three samples of plant extracts (Zanthoxylum piperitum, Pueraria thunbergiana, Solanum nigrum) were about 70% higher activity compared with the non-treated control. Cytotoxicity of plant extracts was not detected in the rat neural stem cells. These results suggest that the selected eight plant extracts are safe compounds. Our findings indicate that Wnt/β-catenin pathway reporter activity could be used for high throughput screening system. In addition, the plant extracts selected as candidates for adipocyte differentiation inhibiting may be potential therapeutic agents for obesity, it will be exploring the possibility of developing an anti-obesity materials through further experiments with selected plant extracts.
This study is aimed at identifying the distribution of vascular plants growing at Byeollipsan(Mt.), Bongchunsan (Mt.) and Gucksoosan(Mt.) in Ganghwa isand. Field surveys were conducted for each season from March 2015 to October 2015. The flora of study area were found to be of 455 taxa, 93 families, 280 genus, 392 species, 4 subspecies, 53 varieties and 6 forma. Rare plants were found to be of 3 taxa. Among them, rare plant species consisted of endangered species (EN degree):1 (Thalictrum simlex var. brevipes Hara), vulnerable species (VU degree): 2 (Wikstroemia trichotoma (Thunb.) Makino, Scorzonera albicaulis Bunge), respectively. In all the surveyed areas, a total of 6 taxa (Clematis brachyura Maxim. etc.) were found to be endemic to Korea. The floristic special plants found in the surveyed areas were 1 taxa of grade Ⅴ, 4 taxa of grade Ⅳ and 8 taxa of grade Ⅲ, respectively. The naturalized plants were identified as 29 taxa and included Fallopia dumetorum (L.) Holub, Ambrosia artemisiifolia L., Aster pilosus Willd. etc..
The final goal of this research is to develop a miniaturized botanical biofilter using a wick-typed automatic humidifier for stabilizing soil moisture content (SMC) and purifying indoor air pollutants by the biofilter. This new biofilter equipped with wick-typed automatic humidifier was manufactured as more compacted design removing an absorption tower-typed humidifier compared with the previous big-sized biofilter made in 2015. This study was performed to compare changes of SMCs among floors depending on the number of wicks installed on the humidifier within the novel biofilter, and to compare changes of SMCs and plant growth parameters before and after planting Spathiphyllum wallisii ‘Mauna Loa’ on the biofilter. SMCs among floors depending on the number of wicks were similar, and all regression lines of SMCs showed almost horizontal lines because of long-term stability on SMCs. Comparing plant growth parameters of S. wallisii ‘Mauna Loa’ before planting and at 30 days after planting on the biofilter, all growth parameters were not statistically significant. Thus, SMCs of the biofilter were more stabilized using this humidifying appar
The genus Tilia is characterized by linear form bracts of which the lower part is attached to the peduncle of a cyme. This character is distinguished from the others genus of Malvaceae. The purpose of this study is verifying the phylogenetic relationship of genus Tilia. Phylogenetic analyses were conducted to evaluate relationships of 10 taxa of Tilia in Korea and Japan including one outgroup (Gossypium hirsutum). The molecular phylogenetic analyses were conducted with sequences based on ITS, trnL-F and rpl32-trnL region. The combined data result of ITS, trnL-F and rpl32-trnL was formed by 6 clades. T. kiusiana situated as the most basal clade. T. amurensis, T. taquetii and T. rufa are composed a clade. T. koreana, T. insularis and T. japonica was formed independent clade. T. insularis has the closest relationship with T. japonica. T. miqueliana, T. mandshurica, and T. megaphylla are composed a clade and showed a sister relationship than other species.
According to survey for development of container to producers who carry out container cultivation, the most important thing is ‘Realization of cultivation techniques (Prune roots, Water saving, Heat conservation)’ as the basis for the design of container and this survey showed that the best one is ‘small and medium-sized container of less than 50L’ as the size of container suitable for cultivation. To investigate the growth characteristics of plants according to the type of container, we classified the container into three types and then planted public plants Pinus densiflora, Cornus kousa, Chionanthus retusa in 12types of containers repeatedly three times. After Comparison of the growth of the Root-collar diameter, Rib-type is showed the highest growth of the Root-collar diameter in case of Pot-type. So, active effects of prune roots impact on positive the growth of plants. Grow inside the ground-type is showed the highest in case of Growing Bag-type and Root skirt-type is showed the highest in case of Bag in pot-type. After, In subsequent studies, based on this study, produce development prototype of design container and test them. and then to investigate the possibility of applying to the farmhouse.
Investigation of the plant growth affected by plug cell volume/pot size and plant age in herbaceous native plants belonging to 11 family and 24 species showed that growth of aerial part was different according to both plant age and plug cell volume/pot size. Among them, especially Iris dichotoma, Iris ensata var. spontanea, Iris pseudoacorus Lysimachia vulgaris var. davurica, Campanula glomerata var. dahurica and Solidago virgaurea subsp. asiatica showed more prominent difference depending on plug cell volume compared to other species. Regarding growth of subterranean part, as decrease plug cell volume/pot size root quality index became high while root weight became low, indicating that growth restriction was happened by exceed increase of root density due to limited root zone area. As increase cell volume/pot size, root development became active in bigger than 150 ㎖ of cell volume. Plant quality got better as increase cell volume/pot size, and 60 days old plants (DOP) revealed higher quality than 90 DOP revealed, indicating that marketing is possible from 60 DOP. Considering both plant age and cell volume (pot size), plant quality was better in smaller cell volume when cultivate plants in short time, while better in bigger cell volume when cultivate plants long time. Accordingly, when extend cultivation periods in small volume of plug cell, plant quality became bad. Therefore, this study showed that in order to produce high quality of plants, plants should be grown in proper plug cell volume depending on cultivation periods. Among plants tested Hosta longipes, Aquilegia buergeriana var. oxysepala, Aquilegia japonica, Veronica linariifolia, Hemerocallis fulva, Iris ensata var. spontanea, Iris pseudoacorus, Allium senescens, Hypericum ascyron, Caltha palustris var. palustris, Gentiana scabra, Campanula glomerata var. dahurica, Dendranthema boreale, Dendranthema zawadskii var. latilobum, Aster spathulifolius, Aster maackii, Aster koraiensis and Solidago virgaurea subsp. asiatica were determined to be able to produce standard size nursery plants using plug tray, and proper plug cell volume was bigger than 150 ㎖ (32 cell tray).
This study was focused on the relationship between horticultural preference and parenting attitude of parents having elementary school students. Because a human’s preference of plant was inhered an image of one’s life. This study carried out to define the variables related to horticultural preference and parenting attitude, for promotion in horticultural activity program of parent. For this study, the data was collected from 193 parents having elementary school child from May to July, 2015 at Seoul, and Jeonju city. The major variables related to parenting attitude were grade of children, dialogue hour with their children, understanding of their children, intimate, number of breakfast, occupation, family income, growing activity. And the relationship between parenting attitude and horticultural preference were marked statistical difference at plant classification into need a supporting stick, foodstuff, life time of plant, individual or community growth, cook and industrial arts or ornamental plants. Especially it was concerned in achievement, rationality, affection of parenting attitude.
The study was surveyed to 269 respondents for investigating psychological response to indoor foliage plants grouped by four type of design materials. 21 indoor foliage plants grouped as filler, mass, form, and line plant, are shown to each respondent for surveying the preference and emotion by SD (semantic differential) scale. It was statically significant that the filler plant group was the least preferred one between the indoor foliage plant and the preference of line plant group was the highest. In filler plant Scindapsus aureus is significantly favored, in mass plant Aglaonema crispum and Syngonium podophyllum, in form plant Ficus banghalensis in line plant Dracaena fragrans ‘Lemon Lime’, respectively. The green is the most preferred color of leaf and gray is the least one. When investigated on the major emotions felt with four types of indoor foliage plant with SD scale, filler plant felt as ‘Bright’, ‘Light’, ‘Pale’, mass plant ‘Warm’, ‘Sharp’, form plant ‘Simple’, ‘Luxurious’ and line plant ‘Sophisticated’, ‘Luxurious’, ‘Pleasant’, ‘Soft’ and ‘Fancy’, respectively. This result will be valuable when the indoor garden is designed with an indoor foliage plant.
Background : Korean Indigenous Hylotelephium erythrostictum is widely distributed in South Korea and is used in Korean traditional medicine. In this study, the phylogenetic analysis of Korean native Hylotelephium erythrostictum and related plants on Internal Transcribed Spacer (ITS) sequences were investigated to distinguish its origin. Methods and Results : The phylogenetic analysis of 6 species of Hylotelephium were investigated by ITS. The dendrogram was constructed by UPGMA(Unweighted Pair Group Method with Arithmetic Mean) clustering algorithm based on genetic similarity of ITS. In the ITS sequence analysis, the size of total was varied from 676 to 779 bp. The size of ITS 1 was rated at 287bp, while ITS 2 was rated at 123bp. The G+C content of ITS region was ranged from 60 to 66%. In the ITS tree, six species of Hylotelephium were monophyletic, and H. viviparum was the first branching within the clade. Conclusion : H. spectabile formed a clade with H. erythrostictum, while H. verticillatum formed with H. viviparum.
Background : Compositae is one of the largest plant families which has high probability of diversity and mutation. Though the various researches about the Compositae are ongoing, it is incomplete and need to be conducted the molecular genetic researches to back up the previous studies. This research was performed to identify the genetic divergence of Compositae plants based on the DNA barcoding for cpDNA-matK and rbcL regions. Methods and Results : For this studies, the genetic sequence analysis (SNP/InDel) and phylogenetic analysis were conducted by using Neighbor-Joining algorithm as targeting the nineteen specimens from 7 species which received a IT number along with NCBI Genbank database (http://ncbi.nlm.nih.gov) sequences. The result of matK sequence analysis, 68 SNP and 2 InDel regions (at nt 527-538bp and 695-706bp positions) were confirmed. Also 10 SNPs were found in rbcL region. The genetic divergence showed 0.000-0.059% in matK regions, and the mean was 0.024%. The highest distance were observed between Ligualria fischeri and the group composed with Aster tataricus and Solidago virgauria (2 and 3). The sequence divergence for rbcL regions showed 0.000-0.018%, and the mean was 0.005%. The highest sequence distance in rbcL region were observed between L. fischeri group and S. virgauria (HE574593). In result of phylogenetic analysis in matK region, the most species formed independent clade. A. tataricus in Aster genus and two samples of S. virguaria in Solidago genus were formed one same clade. S. virguaria(1) and A. spathulifolius(2) has been separated into independently for the plants belonging to same genus, respectively. A. spathulifolius showed differences with NCBI data. The rbcL formed one same clade except L. fischeri and Synurus deltoides. Conclusion : This study indicates that matK is more valuable than rbcL for the distinction among the species of Compositae. This results are expected to be used for the establishment of the classification system of Compositae as well as for the studies in the development of an authentication marker.
Background : Allergy is a common disease caused by type I hypersensitivity reaction of the immune system. Unfortunately, there is no proper treatment for allergy. Therefore, the discovery of therapeutic drugs for allergy is essential. In this study, the crude extracts of 56 plant parts were screened for anti-allergy effects in RBL-2H3 cells. Methods and Results : IgE-sensitized RBL-2H3 cells were individually treated with 56 extracts of medicinal herbs at the final concentration of 20 ㎍/㎖ and stimulated with the antigen DNP-BSA. β-Hexosaminidase release, interleukin-4 (IL-4) secretion, and cell viability in the sample treated cells were measured. Among the tested samples, extracts from the root of Polygonatum stenophyllum Maxim., aerial part of Acer triflorum Kom., and leaf of Pyrus pyrifolia var. culta (Makino) Nakai showed inhibitory effects on β-hexosaminidase release. The aerial part of Peucedanum japonicum Thunberg and seed of Panax ginseng C.A.Mey. showed suppressive activities on IL-4 secretion. All of the extracts were not cytotoxic at the tested concentration. Conclusion : From the result, six extracts including Polygonatum stenophyllum Maxim. (root) and Acer triflorum Kom. (aerial part) inhibited both β-hexosaminidase release and IL-4 secretion in IgE-sensitized RBL-2H3 cells. The use of these extracts for developing anti-allergy materials is suggested.
Background : As a part of ongoing research to elucidate and characterize anti-inflammatory nutraceuticals, six kinds of plant extracts (aerial part of Nepeta cataria, leaves of Lonicera maackii, leaves of Platycarya strobilacea, flower of Fagopyrum dibotrys, flowers and fruits of Solanum nigrum, stem of Physostegia virginiana) were tested for their ability to suppress inflammation. The anti-inflammatory has been studied in lipopolysaccharide (LPS)-stimulated RAW264.7 cells which cells synthesized nitric oxide (NO) from L-arginine by nitric oxide synthase (NOS). In this study, NO synthesis inhibitory activity of six kinds of plant extracts on LPS-stimulated RAW 264.7 mouse macrophages was evaluated. Methods and Results : Six kinds of plant extracts were parceled out from RDA (Rural Development Administration). RAW 264.7 cells (1.5×105 cells/well) were seeded onto 96-well plates with DMEM media containing 10% FBS and 1% antibiotics. The cells were pretreated with the extracts and LPS-stimulated cells for 24 h. Cellular NO production was stimulated by adding 1 μg/mL of LPS. After incubation, Griess reagent was used to determine NO production. Absorbance was measured at 520 nm by microplate reader. NO synthesis inhibitory activity potential of these extracts was evaluated by assessing NO production by LPS-stimulated RAW 264.7 cells in the presence. As a result, inhibition rate of NO production was about 40% of L. maackii, 33% of F. dibotrys, 23% of P. strobilacea and 17% of P. virginiana. Meanwhile, there was no significant results in aerial part of N. cataria and flowers and fruits of S. nigrum. Conclusion : From the above results, we be able to confirm that leaves of L. maackii and flower of F. dibotrys appeared dose-dependent NO synthesis inhibitory activity and leaves of P. strobilacea appeared NO synthesis inhibitory activity in low-concentration. As screening NO synthesis inhibition of six extracts, they may be a good candidate for delaying the progression of human inflammatory diseases and warrants further studies.
Background : Platycodon grandiflorum has been used as food material and a traditional medicine in Korea. In order to develop an efficient in vitro micropropagation technique for a rare plant species and conservation for inbred line of plant breeding. Methods and Results : Plant regeneration via organogenesis and somatic embryogenesis was investigated in Platycodon grandiflorum. Leaf, stem, root tissues of 7-day-old seedlings were cultured on 1/2MS medium containing various concentration (0, 0.5, 1 and 2 ㎎/L) of IBA, BA and NAA. The results showed that 1/2MS medium supplemented with BA+NAA 2.0 ㎎/L yielded the highest callus formation ratio of 73.5%. When various tissues (leaf, stem, root) were tested on 1/2MS medium containing BA 2.0 ㎎/L+ NAA 2.0 ㎎/L for somatic embryogenesis, the optimum tissue for embryogenic shoot induction was stem tissue. Callus were cultured on MS medium containing various concentration (0, 0.5 and 1 ㎎/L) of BA and NAA. The best rooting rate was achieved by three different medium (B5, 1/2MS and MS) and 1/2MS medium cultured the highest rooting ratio (82%). Conclusion : This study suggested that above micropropagation techniques can be used for rapid multiplication as well as in vitro or in vivo conservation of the Platycodon species.
Background : The Aralia cordata and Dendropanax morbifera, belonging to the family Araliacea, is a perennial herbaceous species. Although its beneficial effects for several chronic diseases are well-known, the role of its effects on chronic intestinal inflammation, colon carcinogenis are limited. Inflammatory bowel diseases (IBD) are chronic inflammatory disorders of gastrointestinal tract. It has been reported that IBD is associated with colon cancer which is one of the most common types of cancer diagnosed and a major cause of cancer-related mortality. Methods and Results : The present study hypothesized that Araliacea family would exert a protective effect on inflammation and dextran sulfate sodium (DSS)-induced colitis mouse model. Effect of Aralia cordata extracts (ACE) and Dendropanax morbifera leaf extracts (DMLE) on pro-inflammatory markers, mitogen-activated protein kinase signaling (MAPKs), activation of nuclear factor κB (NF- κB) in THP-1 human monocyte and HT-29 colon epithelial cell model were investigated. In addition, colon inflammation inhibitory effects of ACE and DMLE on the characteristics of in DSS-induced colitis model. Results showed that ACE, DMLE attenuated the severity of DSS-induced colitis, as assessed by disease activity index scores, shrinkage of colon length, and histopathologic changes and infirmatory cytokine concentration. Furthermore, ACE treatment significantly suppressed the colon carcinogenesis in AOM/DSS induced colon cancer model. The apoptosis induction abilities of the DMLE were studied by analyzing the expressions of Bcl2 family protein, caspase, Annexin V/PI staining and DNA fragmentation. According to our results, DMLE significantly induced cell death in SW-480 cells. Apoptosis was determined by cell morphology and electrophoresis of DNA fragmentations. Furthermore, treatment with DMLE also induced the increase in caspase activity, pro-apoptotic proteins (Bax and Bad) and the decrease in anti-apoptotic proteins (Bcl-2 and Bcl-xL). In addition, DMLE induce cell cycle arrested at the G1/S phase in SW-480 cells and exhibited significant inhibition on the expressions of G1/S phase specific CDK, cyclin and up regulation of CDK inhibitors. Conclusion : Taken together, these results provide evidence that ACE and DMLE prevention colon cancer by regulating inflammation, colitis, and colon carcinogenesis which indicates its benefit for colon health.
Background : Glycosylation of natural compounds results in great diversity of secondary metabolites. Glycosylation steps are implicated not only in plants growth and development but also in plant defense responses to various environmental stresses. This process is mediated by members of a multigene superfamily glycosyltransferase (GT), which catalyze the transfer of single or multiple activated sugars to a wide range of substrates, thus influences their chemical property and bioactivity. Although its activity has been recognized for a long time and genes coding UGTs in several higher plants have been identified, specific function of GTs in detail still remains elusive. Methods and Results : Spatial and temporal expression patterns of a ginseng UDP-dependent glycosyltransferase, was analyzed by qRT-PCR. It was expressed highly in rhizome, upper root and youngest leaf compared that of other organs. Spacial expression was observed by GUS histochemical assay after generating promoter::GUS fusion. Noticeably, it expressed axillary branch as well as other organs tested by qRT-PCR. Overexpression of PgUGT in Arabidopsis resulted in fused organ in axillary branch. Stress responsiveness against various abiotic stresses and subcellular localization in Arabidopsis are also addressed. Conclusion : PgUGT phylogenetically closed to PgUGT71A27 involved in ginsenoside compound K (C-K) production. Considering that the C-K is not reported in raw ginseng material, further characterization of this gene may shed light on the biological function of C-K in ginseng growth and development. Organ fusion phenotype could be caused by defective growth of cells in boundary region, commonly regulated by phytohormones such as auxins or brassinosteroids, which in needs to be analyzed further.
