본 연구는 건강기능식품 프랑스해안송껍질추출물 중 catechin, caffeic acid, ferulic acid, taxifolin에 대한 동시분석법을 개발하는 연구이다. 최적분석조건을 확립하기 위해 시료 채취량, 용매 조건을 비교 검토하였으며, UPLCMS/ MS를 이용하여 각 4개 성분에 대한 정확한 분석 및 분석시간의 효율성도 향상하였다. 분석 시 사용한 컬럼은 Acquity UPLC BEH C18이며, 정량이온으로 catechin, caffeic acid, ferulic acid, taxifolin 각각 133, 135, 245 및 248을 선정하였다. 확립된 시험법에 대해 특이성, 직선성, 검출한계, 정량한계, 정확성, 정밀성 등의 밸리데이션을 수행하였다. 4개 성분 모두 50-25000 mg/L 농도에서 결정계수(R2) 0.999이상으로 높은 직선성을 확인하였다. 또한 회수율은 84.9-104%이었고, 정밀성은 1.2-4.3%의 RSD를 확인하였다. 개발된 시험법은 프랑스해안송껍질추출물 중 catechin, caffeic acid, ferulic acid, taxifolin 분석을 위한 시험법으로 활용되기에 적합한 것으로 판단된다.
breakdown of tooth-supporting tissues, producing dentition loss. Porphyromonas gingivalis (P. gingivalis), a Gramnegative anaerobic rod, is one of the major pathogens associated with periodontitis. Neutrophils are first line defense cells in the oral cavity that play a significant role in inflammatory response. Xylitol is a known anti-caries agent and has anti-inflammatory effects. In this study, we conducted experiments to evaluate anti-inflammatory effects of xylitol on P. gingivalis infected neutrophils for possible usage in prevention and treatment of periodontal infections. P. gingivalis was intraperitoneally injected and peritoneal lavage was collected for cytokine determination. For in vitro study, neutrophils were collected from mouse peritoneal cells after zymosan injection or bone marrow cells. Neutrophils were stimulated with live P. gingivalis and ELISA was used to determine the effect of xylitol on P. gingivalis induced cytokine production. IL-1β, IL-6, TNF-α concentration and neutrophil population in the peritoneal lavage was increased in P. gingivalis-infected mouse. Peritoneal cells infected with live P. gingivalis revealed significantly increased production of IL-1β, IL-6 and TNF-α at multiplicity of infection of 10. Neutrophils from bone marrow and peritoneal lavage revealed increased production of IL-1β, IL-6 and TNF-α. Xylitol significantly mitigated P. gingivalis induced cytokine production in neutrophils. Findings indicate that xylitol is an anti-inflammatory agent in neutrophils infected with live P. gingivalis, that suggests its use in periodontitis management.
Radiotherapy (RT) is a mainstay in the treatment of head and neck squamous cell carcinoma (HNSCC). For locally advanced HCSCC, concurrent chemoradiotherapy (CCRT) benefits HCSCC patients in terms of better survival and loco-regional control. In this study, we evaluated changes in oral microbiota in patients, who received CCRT for head and neck cancer. Oral rinsed samples were weekly collected before and during CCRT and at 4 weeks following treatment from HNSCC patients, who had received 70 Gy of radiation delivered to the primary sites for over 7 weeks and concurrent chemotherapy. Oral microbiota changes in three patients were analyzed by next-generation sequencing using 16S rRNA 454 pyrosequencing. On an average, 15,000 partial 16S rRNA gene sequences were obtained from each sample. All sequences fell into 11 different bacterial phyla. During early CCRT, the microbial diversity gradually decreased. In a patient, who did not receive any antibiotics during the CCRT, Firmicutes and Proteobacteria were the most abundant phylum. During the early CCRT, proteobacteria gradually decreased while Firmicutes increased. During the late CCRT, firmicutes gradually decreased while Bacteroides and Fusobacteria increased. In all the patients, yellow complex showed a gradual decrease, while orange and red complex showed a gradual increase during the CCRT. At 4 weeks after CCRT, the recovery of oral microbiota diversity was limited. During CCRT, there was a gradual increase in major periodontopathogens in association with the deterioration of the oral hygiene. Henceforth, it is proposed that understanding oral microbiota shift should provide better information for the development of effective oral care programs for patients receiving CCRT for HNSCC.
The purpose of this study was to examine the effects of communitybased rehabilitation (CBR) program on activities of daily livings (ADLs), quality of life and assistive technology satisfaction for disabled adults in South Korea. Fourteen community-dwelling subjects were participated in a home-based rehabilitation program which performed once a month for 2 months. The outcome measures included Modified Barthel Index (MBI) for assessing ADLs, EuroQol five-dimensions questionnaire (EQ- 5D) for quality of life (QoL) and Quebec User Evaluation of Satisfaction with assistive Technology 2.0 (QUEST 2.0) for assistive technology. Significant improvements in ADLs, quality of life and assistive technology satisfaction were observed after the program. This study indicated that community-based rehabilitation program is an effective method for improving ADLs, quality of life and assistive technology satisfaction for adults with disabilities.
The presence of distinct bacterial species is found to be dependent on age, diet, and disease. We compared the detection rate of several oral bacterial strains in a cohort of 36 subjects including healthy volunteers, periodontal patients, and oral cancer patients. Gargling samples were obtained from these subjects from which DNA was then extracted. Specific primers for 29 bacterial species were used for PCR detection. In the oral cancer patients, Capnocytophaga ochracea, Gemella morbillorum, and Streptococcus salivarius were detected more frequently compared with the healthy volunteers and periodontitis patients. Fusobacterium nucleatum/ polymorphym and Prevotella nigrescens were significantly less prevalent in oral cancer patients than the other groups. In periodontitis patients, Porphyromonas gingivalis and Treponema denticola were more frequently found compared with the healthy volunteers. In the healthy volunteer group, Peptostreptococcus anaerobius was more frequently found than the other groups. The detection rate of several oral bacterial species was thus found to differ between healthy volunteers, periodontitis patients and oral cancer patients.
Xylitol is a five-carbon sugar alcohol that reduces the incidence of caries by inhibiting the growth of oral streptococci, including Streptococcus mutans. Since xylitol is transported via the fructose phosphotransferase system, we hypothesized that it could also affect the growth of other oral bacteria strains. We tested the effects of xylitol against non-periodontopathogenic oral bacteria frequently found in healthy subjects as well as periodontopathogens including Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia. With 5% xylitol, Streptococcus vestibularis and Gemella morbillorum showed marked growth inhibition. With 10% xylitol, all of the tested periodontopathogens and Actinomyces naeslundii showed marked growth inhibition, whereas the growth inhibition of Neisseria mucosa, Neisseria sicca and Veillonella parvula was mild only. Xylitol is a widely used sweetener and the concentration used in our experiment is easily achieved in the oral cavity. If xylitol reduces the growth of periodontopathogens more preferentially, it could also reduce the prevalence of these pathogens and have clinical utility in the prevention or treatment of periodontal disease.
청국장 제조를 위한 분리균은 포자형성율이 높은 Bacillus subtilis로 대사력이 활발한 대수증식기의 것을 콩에 1, 000CFU/g 이상의 균수를 접종하여 40℃와 상대습도 90%에서 배양하면 발효가 빠르게 진행되었다. 발효가 종료되면 포자형성율이 95% 이상 되어야 저온의 후숙과정에서 재번식이 쉽게 일어나지 않았으며 발효취는 발효 중에서만 형성되었다. 중립콩과 대립콩의 발효율이 가장 좋았다. 저온의 후숙과정은 5℃에서 1∼2일 후 경과하면 Bacillus subtilis는 더 이상의 증식이 일어나지 않고 발효취가 소멸되므로 저온의 유통과정을 거쳐야 불쾌취가 없는 구수한 청국장의 맛을 유지할 수 있었다.
청국장 제조 시 사용되는 균주인 Bacillus sp. HJ18-3과 B. subtilis KACC 15935균주를 청국장 제조 시 starter로 접종하여 발효시켜 품질특성과 isoflavone 비배당체 함량을 측정하였다. a-amylase와 cellulase활성은 control과 B. subtilis HJ18-3 균주를 접종한 청국장이 B. subtilis KACC 15935 균주를 접종한 청국장보다 각각 2.56, 2.59배 그리고 1.13, 1.23배 높은 결과를 보였다. 그러나 protease활성은 B. subtilis KACC 15935과B. subtilis HJ18-3의 활성이 높은 효소활성을 보였다. 아미노태질소 함량은 Control, B. subtilis KACC 15935, B. subtilis HJ18-3의 함량이 각각 88.20±13.86, 129.62±1.15, 118.40±48.65 mg%로 나타났으며 이는 protease 효소활성의 결과와 유사한 경향을 보였다. 환원당 함량은 control, KACC 15935, HJ18-3의 함량이 각각 1.95±0.72, 2.62±1.17, 2.59±0.76%로 전반적으로 control에 비해 처리구가 더 높은 경향을 나타냈다. 호기적 총균수와 미비한 차이로 B. subtilis HJ18-3의 균수가 적은 경향을 나타낸 것은B. cereus와Candida albicans에 대해 항균력이 우수하다고 보고된 영향으로 해석된다. control과B. subtilis KACC15935, B. subtilis HJ18-3의 isoflavone 비배당체 형태인 daidzein과 genistein, glycitein을 합한 함량은 각각 158.20±6.55, 131.60±4.00, 200.79±7.54 ug/g으로 나타났다. 이는 초기 raw soybean의 aglycone함량이 3.96 ug/g인 것에 비하여 39∼50배 전 환율을 보인다. 또한 β-glucosidase활성 이 있는 B. subtilis HJ18-3균을 접종하여 제조한 청국장이 control과 Kacc15935균을 접종하여 제조한 청국장보다 1.27, 1.53배 더 높은 aglycone 전 환율을 보였다. 따라서 본 연구에서 사용한 메밀 속성 장으로부터 분리된B. subtilis HJ18-3 균주는 amylase, protease, cellulase와 같은 세포의 효소분비능이 우수하며, 아미노태질소와 환원당 함량이 높고, isoflavone 비배당체 함량을 증가시키는 기능을 함유한 균주로 확인되었으며, 앞으로도 우리나라 전통 청국장의 세계화를 위해 유용 starter로 찾는 노력이 더 필요할 것이다.
