        The purpose of this study was to investigate the anxiolytic-like effect of the methanol extract of Sophorae fructus (MESF) using elevated plus-maze (EPM), open field test, and horizontal wire test in mice. MESF was orally administered at doses of 50, 100, 200, or 400 mg/kg to ICR mice 1 h before behavioral evaluation. The control group was given an equal volume of 10% Tween 80, and the positive control group was given diazepam (1 mg/kg, i.p.). The administration of MESF significantly increased the percentage of time spent in open arms and the entries into the open arms of the EPM compared with the 10% Tween 80-treated control group (p < 0.05). In addition, the anxiolytic-like activities of MESF were antagonized by flumazenil (a GABAA antagonist, 10 mg/kg) but not by WAY-100635 (a 5-HT1A antagonist, 0.3 mg/kg). Futhermore, there were no changes in the locomotor activity and myorelaxant effects of the experimental group, as opposed to the 10% Tween 80-treated control group. Therefore, these findings suggest that MESF promotes the anxiolytic-like activity mediated by the GABAergic nervous system in mice.

        In order to develop as a natural source of anticancer materials of Cudrania tricuspidata, the cytotoxicity of methanol extracts by harvesting parts and times against 8 cell lines including 293 (normal kidney cells) and A-431 (epidermoid carcinoma cells) were investigated using MTT assay. All harvesting parts had hardly cytotoxicity against 293. And methanol extracts of stem bark and root bark showed very high cytotoxicities against 7 cancer cell lines. The cytotoxicity was the highest against HeLa (cervix adenocarcinoma cells) and followed by MCF-7 (breast adenocarcinoma cells), AGS (stomach adenocarcinoma cells), HT-29 (colon adenocarcinoma cells), HepG2 (hepatoblastoma cells), A549 (lung carcinoma cells) and A-431. By the way, leaf extract had a cytotoxicity against only AGS and ripe fruit extract had no cytotoxicity. Among harvesting times, the cytotoxicity of root bark were high from April to September but that of stem bark showed a little difference. These results showed that anticancer activities of Cudrania tricuspidata extracts were eventful changes by harvesting parts and times.

        The anti-oxidant properties of Lactuca indica were determined using in-vitro assay systems. The vitamin C contents of the leaf and root extracts were 24.14 and 0.38 mg/100 g, respectively. The total polyphenol contents of the leaf and root extracts were 42.8 and 7.66 mg/g, and their flavonoid contents were 23.09 and 0.77 mg/g. The leaf extract showed higher DPPH and ABTS radical scavenging ability than the root extract at all the extract concentrations. Especially, the ABTS radical scavenging ability of the leaf extract was 92.3% at a concentration of 5 mg/mL. The reducing power was increased with the increase in the concentration of extracts, and the leaf extract had a higher reducing power than the root. The Fe2+-chelating ability of the leaf and root were 97.2% and 34.3% at 14 mg/mL, respectively. The IC50 values of the leaf for DPPH, its ABTS radical scavenging ability, and its Fe2+-chelating ability were 0.19, 2.7, and 6.27 mg/mL, respectively, and the leaf extract showed lower IC50 values than root extract. These results show that the L. indica leaf extract contained high amounts of anti-oxidative compounds and had higher anti-oxidant activity levels than the root extract. It is suggested that Lactuca indica is very high in availability as a functional food and in its materials.

        서양민들레의 부위별 성분 및 생리활성 차이를 검토하고자 폴리페놀 함량, 플라보노이드 함량, 항산화성 및 세포독성을 분석하였다. Folin-Denis방법에 따른 총 페놀 함량은 꽃 추출물에서 72.0 mg kg-1으로 가장 높게 나타났으며, 그 다음이 잎, 뿌리, 꽃대 추출물 순으로 나타났다(p〈0.05). 한편, 총 플라보노이드 함량은 총 페놀 함량과 같은 경향을 보였으나 더 낮은 범위의 함량이 검출되었다. DPPH 라디컬 소거능은 추출물 농도가 증가할수록 높은 활성을 보였으며 그 중 꽃 추출물 (IC50 값 = 624.3 mg kg-1)에서 가장 높은 활성을 보였고 그 다음으로 잎, 뿌리, 꽃대 추출물 순으로 나타나(p 〈0.05), 이는 페놀 및 플라보노이드 함량이 DPPH 라디컬 소거능과 관련이 있음을 보여 주었다. MTT법에 의한 세포독성 시험에서 폐암세포주(Calu-6)와 위암세포주(SNU-601)의 세포 생존율은 꽃 추출물에서 가장 낮았고(IC50값 = 85.7과 311.4 mg kg-1), 그 다음으로 잎, 꽃대, 뿌리 추출물 순으로 뿌리 추출물이 가장 높은 생존율을 보였다. 이는 세포독성이 꽃 추출출물에서 가장 높고, 뿌리에서 가장 낮은 것을 보여 준 것이다. 결론적으로 총 페놀 함량과 총 플라보노이드 함량이 항산화성(r2=0.7280 to 0.9971)과 세포독성(r2=0.5795 to 0.9515)에 높은 상관관계가 있고, 그 함량과 활성은 식물체 부위별로 다르게 나타남을 확인하였다.

        The aim of this study was to determine total phenolics (TP) content, total flavonoids (TF) level, and antioxidant activity of methanol extracts from leaf parts of 15 Vietnamese wild plants. TP content showed the highest amount in methanol extracts from Altenanthera sessilis (409.7±1.4 ppm), and followed by Eclipta prostrata (183.6±3.8 ppm) and Cassia tora (159.3±5.7 ppm). The highest TF level also was found in A. sessilis (156.3±1.7 ppm), followed by E. prostrata, and C. tora, showing similar tendency to TP. Methanol extracts of A. sessilis, E. prostrate and C. tora at 1000 ppm exhibited the highest DPPH radical scavenging activity by 94.5, 92.6 and 92.7%, respectively. The methanol extracts from A. sessilis showed the highest ABTS and nitrite scavenging activities by 97% and 92%, respectively. The highest correlation coefficient (r2) was 0.9522 between TP and TF, and followed by 0.8919 between DPPH and ABTS activities. However, polyphenols and antioxidant activities showed low correlation coefficients, ranging from r2=0.4114 to 0.4826. It was concluded that Vietnamese wild plants contain polyphenol compounds with antioxidant activities depending on plant species.

        This study investigated the changes of antioxidant activity and α-glucosidase inhibitory effect of Jerusalem artichoke (Helianthus tuberosus) 100% methanol extracts by various heat treatment. The contents of total phenolic and flavonoid compounds in methanol extract tended to increased gradually with the rise of temperature to 180℃. The maximum yield of gallic acid (51.52 ± 2.17mg/g extract weight) and quercetin (13.39 ± 0.03mg/g extract weight) were obtained with extraction temperature of 180℃ for 120min. In addition, the improving extraction efficiency resulted in the increased biological activities, such as electronic donation ability (EDA, 90.36± 0.57%), reducing power (Abs 1.14) and α-glucosidase inhibitory effect (92.14 ± 1.14%). Overall, the results of this study indicate that the optimum conditions for the extraction process were an extraction temperature at 180℃ for 120 min, and will provide the basis for future research on the improving extraction yield of phenolic and flavonoid compounds.

        10종 양치식물의 지상부와 지하부 메탄올 추출물을 대상으로 Bacillus subtilis, Escherichia coli, Listeria monocytogenes, Propionibacterium acnes, Streptococcus mutans, Streptococcus sobrinus 등 병원성 미생물에 대한 항균활성을 broth microdilution법과 paper diffusion법을 이용하여 분석하였다. 억제환이 15 mm 이상으로 나타난 추출물을 대상으로 0.125, 0.25, 0.5 1, 2 mg·mL-1의 농도에서 3일 동안 각 미생물의 생육 억제에 미치는 영향을 분석하였다. 그 결과, B. subtilis, E. coli, L. monocytogenes에 대해서는 꿩고비의 지하부 추출물에서 억제활성이 가장 우수하였으며, 살균처리하지 않은 조추출물 상태에서 2 mg·mL-1의 농도에서 72시간 이후에도 각 39, 33, 58%의 억제활성을 보였다. P. acnes에 대해서는 꿩고비 지상부 추출물이 가장 우수한 항균활성을 보였으나, 15 mm 미만의 억제환을 보여 항균력이 약한 것으로 나타났다. S. mutans와 S. sobrinus에 대해서는 다수의 양치식물이 비교적 우수한 항균활성을 보여 양치식물의 추출물은 전반적으로 Streptococcus에 대한 감수성이 높은 것으로 생각되었다. 그러나 고비의 지하부 추출물이 2 mg·mL-1의 농도에서 72시간 후 각 28, 39%, 그 외의 추출물은 10% 내외의 낮은 항균활성을 보여 10종의 양치식물 추출물 중에는 Streptococcus에 대하여 뛰어난 항균력을 보이는 추출물은 없는 것으로 생각되었다.

        메탄올을 용매로 12종 양치식물의 지상부와 근경의 건조시료를 초음파추출하여 페놀성 물질 함량 및 DPPH와 ABTS radical 소거능을 측정하였다. 총 폴리페놀의 함량은 건조시료 1 g에 2.793~140.014 mg·g-1, 생시료 1 g에 0.779~49.579 mg·g-1으로 다양하게 나타났으며, 넉줄고 사리와 더부살이고사리의 지상부에서 총 폴리페놀 함량이 높았다. 총 플라보노이드 함량은 건조시료 1 g에 2.559~34.909 mg·g-1, 생시료 1g에 0.714~11.487 mg·g-1으로 나타났다. 건조시료 중에는 관중 지상부, 생시료 중에는 더 부살이고사리의 지상부에서 총 플라보노이드 함량이 가장 높았다. 총 폴리페놀은 지상부보다 근경에 많이 함유된 경향을 보였으나, 총 플라보노이드는 지상부에 많았다. DPPH와 ABTS radical 소거능은 지상부보다 근경 추출물에서 높게 나타났다. 총 폴리페놀은 지상부보다 근경에 많이 함유된 경향을 보였으나, 총 플라보노이드는 지상부에서 함량이 많았다. DPPH와 ABTS radical 소거능은 지상부보다 근경 추출물에서 높게 나타났다. DPPH radical 소거능과 ABTS radical 소거능 모두 나도히초미 근경 추출물에서 가장 우수하였다. 그러나 시료의 수분함량과 추출수율을 고려하여 RC50을 나타내는데 필요한 생시료의 양을 계산한 결과, 나도히초미 근경보다는 더부살이고사리 지상부가 DPPH와 ABTS radical 소거용 식물소재로 적합하였다. 양치식물의 추출물은 특히 ABTS radical 소거능이 매우 우수하였으며, 가지고비고사리 추출물을 제외한 대부분의 시료가 ascorbic acid 또는 BHT와 유사한 소거활성을 보였다.

        To evaluate the availability of Cudrania tricuspidata Bureau as a natural source of antimicrobials, the antimicrobial activity of methanol extracts of harvested parts was investigated using the paper disc diffusion method. The extracts from leaves and root bark had broad antimicrobial activity against various bacteria, including Bacillus subtilis, Bacillus cereus, Staphylococcus aureus, Streptococcus mutans, Listeria monocytogenes, Enterobacter aerogenes, Enterococcus faecalis, Salmonella choleraesuis subsp. choleraesuis, Vibrio vulnificus, and Pseudomonas aeruginosa and inhibited Bacillus cereus, Staphylococcus aureus, and Listeria monocytogenes, agents of food poisoning especially well. The extract from ripe fruit had a very high antimicrobial activity against Staphylococcus aureus with a 20.2 mm of clear zone at 50 mg/mL sample concentration. These results indicated that Cudrania tricuspidata could be used as new source for developing natural antimicrobial agents.

        To obtain the information on antioxidant activity by harvesting parts and time in Cudrania tricuspidata Bureau, total phenolic contents, flavonoid contents, superoxide dismutase (SOD) like activity and 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity of methanol extracts from Cudrania tricuspidata were investigated. Total phenolic contents was the highest in leaf [30.2~38.8 mg/g dry weight (DW)] and followed by root bark, stem bark, fruit, root except bark and stem except bark. Among harvesting time, total phenolic contents of leaf was the highest as 38.1~38.8 mg/g DW in May and June, and then decreased to October. Root bark harvested in April and May contained 26.6~27.8 mg/g DW total phenolic compounds and total phenolic content fell to a very low values at 1.8 mg/g DW in February. Flavonoid contents was very high in leaf and root bark. Especially, root bark harvested in April had the highest of flavonoid contents as 23.2 mg/g DW. As a result of simple linear regression analysis of total phenolic contents on flavonoid contents according to harvesting parts, it showed a high correlation (p 〈 0.001) with 0.57~0.97 R2 (coefficient of determination). As RC50 value, that was, the concentration of sample required for 50% reduction of DPPH absorbance, was 13.7~20.5 μg/mL in leaf so DPPH radical scavenging activity was very high. But SOD like activity of 1,000 μg extract/mL MeOH was a low value of 0.3~9.2% in all samples.

        In order to verify the efficacy of Lespedeza cuneata G. Don which have been used as a folk traditional herb medicine from ancient times, the test of antioxidant activity and the analysis of constituents were carried out. Methanol extracts from L. cuneata G. Don showed a radical scavenging activity corresponding to that of (+)-catechin as positive control at 250 ~mug /mL (ca 90% inhibition), and inhibited more than 30% of DPPH radical at low concentration of 10~mug /mL. In the present study, as a fundamental study for the specification of antioxidant components from L. cuneata G. Don, the analysis for constituents for L. cuneata with HPLC was preformed although there was no results to identify functional flavonoids such as quercetin.

        Phenolics level, total flavonoid content, and antioxidant were determined from the methanol extracts of the eight medicinal plants using roots. Total phenolics were found as the highest levels in the methanol extracts of Arctium lappa, and followed by Youngia sonchifolia and Cirsium japonicum. Total amount of the each phenol compounds were detected in C. japonicum extracts (319.2 mg kg-1) as the greatest component, and followed by A. lappa (96.3 mg kg-1) and Y. sonchifolia (22.9 mg kg-1). Total flavonoid content showed the highest amount in methanol extracts from A. lappa (68.1 mg 100 g-1) and followed by Y. sonchifolia (11.2 mg 100 g-1) Lycoris radiate extracts (87.2%) had the highest nitrite scavenging activity and followed by A. lappa (81.5%) and Y. sonchifolia (77.5%). Methanol extracts of A. lappa at 25 mg 100 g-1 exhibited the highest DPPH radical scavenging activity by 90.8%, even though less activity than synthetic antioxidants Vitamin C or butylated hydroxytoluene (BHT). Level of polyphenols was highly correlated with anti oxidative activity (r2 = 0.85). The results suggest that several medicinal plants selected had the potent biological activities, and that their activities were differential depending on plant species.

        Phytotoxicity, antioxidant activity, and cytotoxicity of the aqueous or methanol extracts from the young sprouts of the six Compositae medicinal plants were determined. Aqueous leachates at 40g dry tissue L-1 (g L-1) Cirsium japonicum and Aster yomena showed the highest inhibitory effect on alfalfa (Medicago sativa L.). Total phenolic content showed the highest amount in methanol extracts from Ixeris dentata, and followed by A. yomena, and Cephalonoplos segetum. Methanol extracts of C. segetum and I. dentata at 25μg mL-1 exhibited the highest DPPH radical scavenging activity by 87.2, and 52.8%, respectively. By means of HPLC analysis, MeOH extracts of C. segetum had the highest amount of antioxidant chlorogenic acid. Based on MTT assay, the methanol extracts from Y. sonchifolia (IC50 = 65.7μg mL-1) showed the highest cytotoxicity against Calu-6. These results suggest that plant extracts had a dose-dependent biological potentials including phytotoxicity, antioxidant activity, and anticancer activity, and that their activities exhibited differently depending on plant species.

        Antioxidant activity, total phenolics level and cytotoxicity of the methanol extracts from the young sprouts of 5 Korean woody salad plants were determined. Methanol extracts of Kalopanax pictus had the highest DPPH radical scavenging activity, with an IC50 value of 23.5mg 100g-1, and followed by Valeriana fauriei(43.1mg 100g-1), and Morus alba(〉100mg 100g-1). Total phenolic content showed the highest amount in methanol extracts from Kalopanax pictus(23.7mg 100g-1), and followed by Valeriana fauriei(22.7mg 100g-1), Aralia elata(16.8mg 100g-1) and Morus alba(14.2mg 100g-1). In a MTT assay, methanol extracts of Aralia elata with IC50 values of 151.0 and 140.7μg mL-1 showed the most potent cytotoxicity on Calu-6 and MCF-7, respectively. On the other hand, methanol extracts of Kalopanax pictus(IC50=96.5μg mL-1) showed the highest activity against HCT-116, and followed by those of Aralia elata(123.3μg mL-1), and Actinidia arguta(162.0μg mL-1). Total phenolic content of the tested plant extracts was correlated with the DPPH radical scavenging activity, suggesting the phenolic compounds may contribute to the antioxidant properties of Korean salad plants.

        본 연구는 배암차즈기 메탄을 추출물의 생리활성 및 기능성을 검토하기 위해서 항산화효과 및 아질산염 소거능을 측정하였다. 그 결과, 배암차즈기 메탄을 추출물의 전자공여능(RC50)은 51.10 μg/ml로 나타났고 SOD 유사활성은 추출물(0.92mg/ml)을 첨가하였을때 62.11%로 가장 높게 관찰되었으며, linoleic acid에 대한 항산화력은 추출물 (0.2mg/ml)을 첨가하여 지질과산화물을 측정한 결과 배양시간 1, 2, 4일 경과후 각각 72.86%, 77.18%, 78.32% 감소율을 보임으로써 효과적인 항산화 효과를 나타내었다. 총 페놀함량은 gallic acid equivalents (GAE)로 측정되었고, 202.35± 1.26 GAE μ extract으로 조사되었다. 아질산염 소거능은 복합 생약추출물(0.2mg/ml)을 첨가하였을 때 pH 1.2조건에서 44.29%로 가장 높게 나타났으며 pH가 증가할수록 감소하였다. 이상의 결과로 배암차즈기 메탄을 추출물은 우수한 항산화 효과와 아질산염 소거능을 나타냄으로써 기능성 식품 소재 및 식품 첨가물로써 사용가능 하리라 사료된다.
