        본 연구에서 개발된 자동천공기는 콩의 재식밀도를 고려한 비닐 천공능력이 작업주행속도 0.18- 0.28m/s 조건에서 콩의 주간거리 0.20m, 0.25m, 0.30m에 대해 각각 320-500m2/h, 260-400m2/h, 210-330m2/h의 범위로 나타났다. 천공율은 칼날과 지면간 30mm 거리에서 비닐이 100% 천공되었으 며, 50mm 거리에서 원형칼날 16.7-25.2%, 사각칼날 33.0-42.3%, 육각칼날 54.5-100.0% 범위에서 천공되었다. 또한, 육각칼날 60°에서 비닐 절단면이 매끄럽고 부드러워 절단품질이 가장 우수하였다.

        Lipoxygenase(LOX) 결핍 콩(05C4 및 LS, LOX2,3-free)과 일반콩(태광, LOX1,2,3-present)으로 제조된 된장의 급이가 고지방-콜레스테롤 식이성 흰쥐의 체내 지질성분 및 항산화 효소계에 미치는 영 향을 알아보고자 하였다. SD계 흰쥐는 정상군, 고지방-콜레스테롤 급이군(HFC), 고지방-콜레스테롤 식이+5% 된장 분말 급이군[HFC+D1, 태광된장; HFC+D2, 05C4된장; HFC+D3, LS된장)으로 구분하여 5주간 사육되었다. In vitro에서 된장의 콜레스테롤 흡착활성은 4mg/mL 농도에서 LS된장이 태광된장 에 비해 1.5배 높았다. 식이효율은 정상군에 비해 대조군에서 유의적으로 높았으며, LS된장 급이군 (HFC+D3)은 대조군에 비해 유의적으로 감소되는 경향이었다. Rö hrer 및 T.M.지수와 체지방 함량은 대조군에 비해 된장 급이군에서 모두 유의적으로 감소되었으며, 특히 Lee지수 및 BMI는 LOX 결핍콩 된장 급이군에서 유의적으로 감소되었다. 혈청 총 지질 및 중성지방 함량은 대조군에 비해 된장 급이군 에서 유의적으로 감소되었으나, 총 콜레스테롤, HDL-, LDL-콜레스테롤 및 심혈관질환 위험지수는 LS 된장 급이군만 유의적인 감소를 보였다. 혈청 AST 및 ALT 활성은 HFC+D1 및 HFC+D2간에는 유의차 가 없었고, HFC+D3은 HFC+D1 및 HFC+D2에 비해 유의적으로 낮은 수준이었다. 간 조직에서 총 지 질 함량은 HFC+D3이 대조군에 비해 약 25.2%의 감소되었으며(p<0.05), 중성지방 및 총 콜레스테롤 함량은 HFC+D2 및 HFC+D3이 대조군에 비해 유의적인 감소를 보였다. 분변 중의 중성지방 함량은 된 장 급이군 모두 대조군에 비해 26.2~40.3%의 증가를 보였으며(p<0.05), 총 콜레스테롤 함량은 LS된장 급이군만 유의적으로 증가하였다. 간 조직 중 SOD 활성은 대조군에 비해 05C4 및 LS된장 급이군에서 유의적으로 증가하였으며, catalase 및 GSH-px 활성은 된장 급이군 모두 대조군에 비해 유의적으로 증가하였다. UDPGT 활성은 된장의 급이 시 정상군과 유사한 수준으로 증가되었으며, 특히 LS된장 급 이군은 정상군에 비해 유의적으로 증가하였다. 따라서 LOX 결핍 콩으로 제조된 된장은 체내 지질 개선 및 항비만 활성에 긍정적인 도움이 될 것으로 판단된다.

        In this study, we analyzed the utensils, covers and mats that were used for making meju, the shape of meju, and the heating method used for making meju from the 225 ways of preparing jang mentioned in the 32 volumes of the ancient cook books from 530 AD to 1950. The heating method of traditional meju bean and starch included 57 kinds of steaming, 59 of boiling, 21 of roasting + boiling, and 2 of cooking. The shape of meju included 41 kinds of egg, 27 of ball, 22 of lump, a kind of doughnut, 8 kinds of hilt, 6 of flat, 4 of chip, and a kind of square. Among the 72 gochoojang meju, the heating method of bean included 9 kinds of boiling, and 6 kinds of steaming; whereas the heating method of starch included 19 kinds of steaming of dough, 11 of rice cooking, and 5 of boiling of dough. The utensils for molding of bean meju were 49 kinds of straw sack, 14 of round straw container, 11 of heating bed, 7 of large straw bowl or Japanesesnailseed, 5 of jar, 4 of ditch, 3 of straw bowls, 2 of pottery steamer of dough, 2 of gourd, and a kind of long round bamboo bowl and sack of straw. The cover and the mat used for molding of meju included 36 kinds of straw, 17 kinds of paper mulberry leaf, 15 of wide straw seat, 14 of mugwort, 11 of pine tree leaf, 10 of soybean leaf, 6 of cocklebur leaf, 6 of sumac leaf, 6 of barley straw, 6 of mulberry leaf, 5 of fallen leaf, 5 of cogon grass, 4 of reed seat, 3 of scrap of cloth, 2 of Indian bean tree leaf, a kind of reed. There were only 5 kinds of hanging.

        The soybean aphid, Aphis glycines Matsumura, was introduced from East Asia (EA) into North America (NA) and is now widely established in NA. To compare soybean aphid populations between the native and invasive regions, we examined 689 individuals obtained from 28 different collections in NA and EA. A total of 8 microsatellite loci were used for population genetics statistics. Gene diversity and mean number of alleles in NA populations averaged 0.40 and 2.70, respectively, whereas in EA they averaged 0.55 and 4.32, respectively. Structure analysis of all populations revealed two distinct structures in the invaded and in the native regions. Among EA populations, certain Korean populations were genetically closest to NA populations, especially those from Ohio and Delaware. An approximate Bayesian computation test also supports an introduction into NA from Korea.

        In our previous study, COPA (coatomer subunit alpha) gene from the two-spotted spider mite, Tetranychus urticae, exhibited RNA interference (RNAi)-based lethality when its double-stranded RNA (dsRNA) was systemically delivered via multi-unit chambers or its hairpin RNA was in planta-expressed by agroinfiltration. The cumulative mortality of T. urtcae was 55.0 ±14.2% in soybean plants agroinfiltrated with COPA gene. To investigate the temporal expression profiles of hairpin RNA following agroinfiltration, the amount of hairpin RNA expressed in plants was quantified over time by quantitative real-time PCR. Relative transient expression levels of T. urticae COPA hairpin RNA was highest at 46 h post-agroinfiltration and the extent of COPA gene knockdown was lowest at 12 h post-infestation on soybean plants. To investigate small interference RNA (siRNA) profiling, northern blot assay is currently under progress.

        Soybean peptide (SP) exhibited low intestinal absorption at oral administration due to its fragile structure under gastric digestion. Therefore, we have attempted to encapsulate peptide by cross-linkage interaction between positive charged chitosan (CS) or chitosan oligosaccharide (CSO) and negative charged peptide. The CS (or CSO) with SP nanoparticles were prepared by using ultrasonification technique. The objective of this study was to find the optimal processing method by changing concentration, pH, and homogenizing conditions. We measured physicochemical properties such as particle size, zeta-potential, encapsulation efficiency (EE%), release rate (RR) and antioxidant ability of samples. The results showed that the optimal processing method was using 0.5% (w/v) CSO (diluted by pH 3 Acetic acid buffer) mixed with 0.5% (w/v) SP (diluted by pH 6 buffer) by 9:1 ratio. Afterwards, using high-speed mixer at 12,000 rpm for 3 min, and then passed 2 times through an ultrasonicator (50% power, 3 min). In this way for processing, the particle sizes of CSO/SP nanoparticles were approximately 300 nm, zeta-potential were approximately 45 mV. In addition, the EE% and RR of CS/SP nanoparticles was higher than the CSO/SP nanoparticles. The increase in antioxidant ability of SP was attributed to the affected by CS/CSO microcapsules. In conclusion, this research can befoundation for the manufacturing process of CS/SP nanoparticles, and it was expected that the future application of this nanoparticle in food matrix.

        Cryoprotectant is a substance used to protect biological tissue from freezing injury. However, there was few research paper on application of cryoprotectant in food stuff although its benefits was approved from the biological cell tissues. The objective of this study was to investigate the effect of the sugar addition as a cryoprotectant on the properties of frozen soybean sprouts. Before freezing process, the samples were blanched at 100°C for 1 min to observe the influence of blanching treatment. The blanched or non-blanched soybean sprouts was immersed in sugar solution as cryoprotectant, and continuously, the samples were frozen at -18°C for 24 h. Their physicochemical properties such as drip loss, hardness, color and cellular tissue were analyzed after thawing in running water. In our study, the drip loss of blanched sample without sugar was 43%, and comparatively, blanched one with sugar was 20% which was the lowest value among all samples. There was no significant difference of hardness between sample with sugar and without sugar. From our results, it was supposed that sugar can protect the soybean sprouts during freezing process regardless blanching process.

        Biogenic amines (BAs) are toxic nitrogenous compounds and formed by the microbial decarboxylation of amino acids during fermentation. Consumption of fermented foods high in BAs can pose the adverse health effects. The objective of current study was to develop a reduction method of BAs in fermented soybean paste by applying catechins during fermented koji manufacturing step. Analysis of BAs in fermented soybean paste was conducted using High Performance Liquid Chromatography (HPLC). The levels of Putrescine decreased for 63.7%, 60.1%, 76.1%, 73.4% for 0.3% addition of Epicatechin, Epicatechin gallate, Epigallocatechin, Epigallocatechin gallate, respectively. The levels of Cadaverine decreased for 24.3%, 34.3%, 42.6%, 37.2% for 0.3% addition of Epicatechin, Epicatechin gallate, Epigallocatechin, Epigallocatechin gallate, respectively. Especially, The best reduction effect of BAs appeared for 0.3% addition of Epigallocatechin. Current study demonstrated the potential use of catechins for reduction of biogenic amines in fermented soybean paste model. Finding from this research can practically guide fermented food industry by providing practical application of additives for reduction of BAs.

        The introduction of stress up-regulated genes into heterologous plants is found to confer stability trait to plants in otherwise hostile environments. The soybean cold inducible factor-1 gene(SCOF-1) encoding a zinc finger transcription factor was introduced into Populus alba L.(white poplar) by co-culture with Agrobacterium tumefaciens GV3101 that carried the binary vector, pGA643/SCOF1. The putative P. alba transformed plants were selected on Murashige and Skoog medium supplemented with 2.22μM 6-benzylaminopurine, 1.18mM carbenicillin, and 28.47μM hygromycin. The integration of SCOF-1 into the P. alba genome was also established by polymerase chain reaction(PCR), and the expression of the gene was confirmed by reverse transcription PCR. After exposure for 4h at 4℃ followed by 48h recovery at 27℃, the transgenic plants completely recovered to normal growth status. The chlorophyll content in transgenic plants was higher than that in wild type plants after chilling treatment. However, the moisture content and total phenolics was also not significantly different in transgenic and wild type plants after cold treatment.

        울금의 주요 성분인 커큐민과 대두 추출물인 이소플라본의 피부 미용 측면에서 생리활성을 연구하여 화장품 소재로서 가능성 여부를 규명하고자 하였다. 본 연구는 세포실험을 통해 커큐민과 대두 추출물을 피부 세포에 대한 독성 및 항염증을 확인하고, HPLC을 이용하여 대두 추출물의 성분분석과 피부에 적용하였을 때 수분과 유분, 홍반변화를 측정하였다. 본 연구 결과 커큐민이 RAW 264.7 세포에 대한 독성이 적은 것으로 확인되었으며, 항염증에 대한 효과를 확인하였다. 8주 동안 커큐민과 대두 추 출물이 함유된 화장품과 식이를 병행하여 사용하였을 때 수분함량 변화, 유분함량 변화, 홍반 지수의 감 소가 통계적으로 유의미한 변화를 나타내었으며, 여드름 피부에 유의한 효과를 가질 수 있음을 확인하였 다. 따라서 본 연구는 커큐민과 대두 추출물이 화장품 소재로 사용 시 여드름 피부 개선에 효과적인 화 장품 소재로서 활용 가능성이 있을 것으로 사료된다.

        Analysis of nutritional compositions of soybean sprouts cultivated with bamboo ash was carried out. Bamboo ash was utilized as sprouting water of soybeans and adjusted to 0.2, 0.6, 1.0, 1.4, 2.0, 6.0 and 10.0 g/L. Stem length and contents of isoflavone (daidzin, glycitin, genestin, daidzein, glycitein, and genestein) and vitamin C in soybean sprouts cultivated with 0.2 g/L were higher than those in soybean sprouts cultivated with only water. Potassium, magnesium, and calcium of all cultivation methods were detected in higher contents than others. In particular, potassium showed a high absorption rate in the soybean sprouts. The major amino acid was asparagine (616.05~849.15 mg/100 g, soybean eq.), and contents of lysine, leucine, and ornithine in soybean sprouts cultivated at 0.2 g/L were higher than those of methods by only water and addition of 6-benzylaminopurine. According to the results, soybean sprouts cultivated with 0.2 g/L of bamboo ash were effective for increasing nutritional compositions.

        The vitamin E enhanced transgenic soybean was developed by introducing a perilla γ- tocopherol methyltransferase gene (γ-TMT) under the control of pea vicilin promoter and a selection marker, phosphinothricin acetyltransferase (PAT) gene. With regard to the potential problems of safety, the non-target organism evaluation is required as an essential element for the environmental risk assessment of genetically modified (GM) crops. We studied the effects of the vitamin E enhanced transgenic soybean feeding on survival of Daphnia magna which is commonly used as a model organism in ecotoxicological studies. The Daphnia magna was fed on vitamin E enhanced transgenic soybean and non-genetically modified (non-GM) soybean (Willams 82) at 0, 1,000, 1,800, 3,240, 5,830, 10,500 and 20,000 mg/L concentrations, respectively. The GM soybean used for the test was confirmed to have the γ-TMT/PAT gene expression by the enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) analysis. The results showed that there was no significant differences between cumulative immobilities of Daphnia magna fed on GM soybean and non-GM soybean. The 48hr-EC50 values showed no significant differences between GM soybean (2,416 mg/L) and non-GM soybean (2,408 mg/L). The results of this study suggested that there was no significant differences in toxicity for Daphnia magna between GM soybean and non-GM counterpart.

        본 논문은 히스토그램과 그레이레벨 프로파일의 VS(valley size)를 이용한 문턱치 기반의 대두 선별알고리즘 을 제안한다. 대두의 변형은 크게 표면 변색 및 외형 변형으로 나눌 수 있다. 본 연구에서는 대두 개체를 건전립, 부 분변색립, 전면변색립, 표면변형립으로 구분하고 히스토그램 기반 문턱치와 VS기반 선별알고리즘을 이용하여 표면의 변색 및 변형에 따른 불량 대두와 건전립을 선별하였다. 제안한 방법은 대두와 같이 개체수가 많은 농작물의 실시간 선별을 목적으로 하여, 정수덧셈 연산기반의 선별 알고리즘으로 구현 되었다. 제안한 방법을 실제 선별에 적용한 결 과 선별의 정확도는 94.5%이며 기존의 문턱치 기반 대두 선별 알고리즘에 비교할 때 상대적으로 적은 연산량에서 동 등한 수준의 정확도를 나타내는 것을 확인하였다.

        The effects of drying temperature on the drying characteristics of soybeans at the different position in the dryer with 28.1% (±0.8) of initial moisture content were studied. Drying temperatures varied at 35, 45 and 55ºC, with a constant inlet air velocity (3 m/s). The local air velocities at a position 1, 5, and 9 were 0.150 m/s (±0.012), 0.247 m/ s (±0.018) and 0.795 m/s (±0.036), respectively. The drying rate increased as the local air velocity increased from 0.150 m/s to 0.795 m/s. The cracked grain ratio increased as the drying rate increased. To prevent quality degradation of soybeans during hot-air drying, the local air velocity should be considered. Thin-layer drying models were applied to describe the drying process of soybeans. The Midilli-Kucuk model showed the best fit (R2>0.99). Based on the model parameters, the drying time to achieve the target moisture content (15%) was successfully estimated. The drying time was strongly dependent on the position in the dryer and the drying temperature.
