This study aimed to analyze crude protein, amino acid and bioactive changes in Phellinus linteus HN00K9 cultured oat. The crude protein content of P. linteus cultured oat (PCO) was 12.9%, which was higher than that of uncultured oats (UCO) as control at 11.26%. The total free amino acid contents of PCO and UCO were 11,4 mg/100 g and 9,686.205 mg/100 g, respectively. Glutamic acid accounted for 21% of the total amino acids of PCO, and the histidine content was increased by more than 51% in PCO compared to UCO. The total polyphenol content of PCO was 275 mg GAE/g, which was more than twice that of UCO (135 mg). The DPPH radical scavenging activity was 15.5% in PCO, which was more than five times that of UCO (3.5%). The β-glucan content of PCO was 12.5 g/100 g, which was more than five times that of UCO (3.2 g/100 g). Therefore, it is believed that PCO can be utilized as a material for various functional foods.
Supplementing feed components is becoming increasingly difficult for various reasons, including increased shipping costs, decreased capture efficiency, and global warming. For this reason, much research has been conducted globally to find alternative protein sources. The black soldier fly (BSF; Hermetia illucens) is an important feed item for nutritional accumulation and has environmental consequences. Few studies have been conducted to determine the nutritional level of the substrate, but most of these studies have involved modifying the substrate material. The aim of this study was to determine the ideal crude protein(CP) content of a substrate for BSF rearing. The experimental treatment was separated into three CP levels (low, 13.5%; medium, 20%; high, 26.5%) in the substrate. The results showed that the BSF weight (6.46%) and protein conversion ratio (1.13%) increased as the substrate CP level increased (p<0.05). In addition, protein (14.38%) and lipid (13.12%) yields were significantly increased (p<0.05). The BSF CP level increased as the substrate CP level increased (p<0.05). CP levels in the substrate upregulated the levels of certain fatty acids, essential amino acids, and non-essential amino acids (p<0.05). In conclusion, the potential values of BSF as feed were improved as the CP level of the substrate increased; However, no difference was found between medium and high CP levels in the rearing substrate. These results suggested that the substrate CP level should be at the medium level for optimal product quality. In conclusion, BSF quality is influenced by substrate nutrient levels, necessitating the development of specialized substrates for effective rearing.
본 연구는 국내에서 흔히 유통되는 박류 단미사료의 정확한 사료 유래 대사 단백질(metabolizable protein from feed, MPfeed) 함량을 제시하기 위해 수행되었다. MPfeed를 측정하기 위해 반추위 미분해 단백질(rumen undegradable protein, RUP) 함량과 RUP의 소장소화율을 평가하였다. 시료 각각의 RUP 함량을 측정하기 위해 홀스타인 거세우 2두를 이용하여 in situ 반추위 분해율 실험을 수행하였다(0, 3, 6, 12, 24, 48, 72, 96, 120 h). 각 시료의 소장소화율을 측정하기 위해 12 h in situ 배양을 마친 시료들은 인공 4위액과 인공 소장액에 순차적으로 배양된 후 잔량 측정되었다. RUP와 소장 분해율을 측정을 통해 측정된 MPfeed는 전지대두가 45.1%로 가장 높았으며, 국산 대두박(44.3%), 팜박(42.7%), 옥수수 주정박(dried distiller’s grains with solubles, DDGS, 40.7%), 수입산 대두박(40.2%), 호마박(39.7%), 야자박(39.3%), 채종박(28.4%), 옥 글루텐 밀(26.5%), 옥 배아박(20.5%), 해바라기박(15.3%), 옥 글루텐 피드(14.5%), 아몬드박(5.25%) 순으로 나타났다. 본 연구에서 사용된 시료들의 MPfeed는 미국사양표준에서 제시하고 있는 수치와 차이를 나타내었는데, 이는 MPfeed를 측정하기 위해 사용되는 RUP 함량과 RUP의 소장소화율에서 차이를 보임에 의한 것으로 추정된다.
본 연구의 목적은 알츠하이머질환(Alzheimer’s disease: AD) 동물 모델을 대상으로 트레드밀 운 동(Treadmill exercise: TE)과 환경강화(environmental enrichment: EE) 처치가 인지기능, 근 기능, 및 밀 착연접 단백질 발현에 미치는 영향을 확인하는데 있다. AD 동물 모델을 제작하기 위해 aluminum chloride(AlCl3)를 90일간(40mg/kg/하루) 투여 하였으며 동시에 TE(10-12m/min, 40-60min/day) 혹은 EE에 노출시켰다. 그 결과 AlCl3 투여에 의한 인지기능 저하와 근 기능 감소가 TE와 EE에 의해 완화된 것 으로 나타났다. 또한, TE와 EE는 AD 질환에서 나타나는 β-amyloid(Aβ), alpha-synuclein 및 tumor necrosis factor-α(TNF-α) 단백질의 발현 증가를 감소시킨 것으로 나타났다. 게다가 TE와 EE는 AlCl3 투여에 의해 감소된 밀착연접 단백질(Occludin, Claudin-5 및 ZO-1)의 발현을 통계적으로 유의하게 증가시킨 것으로 나타났다. 마지막으로 Aβ 단백질과 밀착연접 단백질과의 상관분석을 실시한 결과 부적 상 관관계(Occludin: r=-0.853, p=0.001; Claudin-5 : r=-0.352, p=0.915; ZO-1 : r=-0.424, p=0.0390) 로 나타났다. 따라서 이를 종합해 보면 TE 혹은 EE 처치는 AD에 나타나는 병리학적 특징들을 일부 완화 시켜 인지기능과 근 기능을 일부 개선 시킬 수 있는 효과적인 운동 방법이라고 생각된다.
This study investigated the physicochemical properties of protein-fortified rice flour by mixing rice flour (RF) with untreated and fermented plant proteins. Fermented faba bean protein concentrate (FMFP) and chickpea flour (FMCF) were prepared by solid-state fermentation of faba bean protein concentrate (UTFP) and chickpea flour (UTCF) using Bacillus subtilis. FMFP and FMCF exhibited higher crude protein, reducing sugar and starch contents more than their counterparts. The increased rate of essential and branched-chain amino acids in FMFP and FMCF exceeded that of crude protein. Adding plant proteins to RF decreased swelling power (SP) and increased solubility in RF-UTFP and RF-FMFP mixtures, while SP and solubility increased in RF-UTCF and RF-FMCF mixtures. All RF-plant protein mixtures showed higher gelatinization temperature and lower gelatinization enthalpy than RF. Thermal gelation was found in all RF-plant protein mixtures, but the RF-FMCF mixture may form weak and unstable gel structures. The increase in pasting viscosity was minimal for the RF-UTFP and RF-FMFP mixtures but more pronounced for the RF-UTCF and RF-FMCF mixtures. Overall, FMFP may be a potential protein source to supplement the protein deficiency in RF with minimal changes in RF-based foods’ rheological and textural properties.
UV and O3 are materials used in the water treatment process, and many studies have been reported to remove organic matters, contaminants, and microorganisms. In this study, we were investigated effects of Chirnomidae (Chironomus flaviplumus, Chironomus riparius), which are contamination indicator species to exposure UV and O3 for the survival rate, body color change and gene expression response. The survival rate of C. flaviplumus exposed to UV decreased to about 70% after 24 hours, and C. riparius about 50%. There was no change in the survival rate of C. flaviplumus exposed to O3, and C. riparius decreased to 95% after 10 minutes of exposure, but there was no change during the subsequent exposure time. In addition, UV and O3 exposure to the two species in body color faded in a time-dependent. In the HSP70 gene expression, C. riparius showed an increase in expression after UV exposure compared to the control group, and a significant difference was shown 12 hours after exposure (P<0.05). C. flaviplumus exposed to O3 showed a relatively low expression compared to the control group, and showed a significant difference at 10 minutes and 1 hour after exposure (P<0.05). These results reported the ecotoxicological effects on Chironomidae according to UV and O3 exposure. Therefore, the results of this study can be used as basic data to understand the effects of UV and O3, which are disinfectants used in water treatment plants, on Chirnomidae entering plants. 한우에게 급여하는 사료 내 에너지원의 종류에 따라 반추위 미생물의 아미노산 조성에 차이가 있는지를 조사하고자 본 연구를 수행하였다. 국내 한우 비육우 사육에 주로 이용되는 에너지원 사료인 옥수수(T1), 생미강(T2), 소맥(T3) 그리고 소맥피(T4)로 반추위 환경과 유사한 연속식 배양기를 이용하여 72시간까지 배양을 진행하였다. 배양이 진행되는 동안 6시간 간격으로 배양액의 발효 성상을 확인하였으며 pH, NH3-N이 일정하게 유지되는 것을 확인하였으며, 배양 종료 시점인 72시간대에 미생물체 단백질 합성량(MPS), 미생물의 아미노산 조성 및 미생물 균총 변화를 분석하였다. 배양액의 pH는 모든 처리구에서 배양 기간동안 5.5~7.0을 유지하였다. Total VFA 농도는 T1이 23.13 mM으로 가장 낮았고, T4가 29.93 mM으로 가장 높았다(p<0.05). A (acetate) :P (propionate) 비율은 T1이 1.48로 가장 낮았으며, T4가 2.81로 가장 높게 나타났다 (p<0.05). Butyrate는 T1이 2.37 mM으로 가장 낮았으며, T2, T3 그리고 T4는 4.37~4.58 mM으로 차이가 없었다(p<0.05). MPS는 T2 (332.5 mg/L)와 T3 (320.2 mg/L)가 높았고 T1 (137.5 mg/L)와 T4 (154.2 mg/L)로 낮게 나타났다(p<0.05). DGGE band 분석결과 모든 처리구는 57.5 % 이하의 유사도가 나타났다. 미생물의 총 아미노산 함량은 T1 (31.59 %)과 T3 (31.33 %)가 가장 높았으며, T2 (20.09 %)가 가장 낮았다 (p<0.05). 이는 급여된 사료 내 총 아미노산 함량과 반대되는 결과로 나타났다. 따라서, 본 연구에서 한우 급여 에너지원 사료에 따른 반추위 내 미생물 발효 특성과 미생물체 단백질 합성량이 미생물 군집에 영향을 미치며 이에 따라 미생물의 총 아미노산 함량에 영향을 미친다는 것을 확인하였다.
This study was conducted to evaluate in vitro antioxidant activity of goat meat hot water extracts and the changes in apoptosis-related protein expression levels in the cancer cells treated with these extracts. Goat meat hot water extracts were prepared using different cuts of goat meat, including foreleg, hindleg, loin, and rib. Among these extracts, the foreleg and hindleg extracts displayed higher (P<0.05) ABTS radical scavenging activity than the other two extracts. Protein expression levels of BAX, p53, and p21 were not different in the cells treated with the extracts from different cuts, regardless of the cell type. Only p53 expression in HT-29 cells was elevated (P<0.05) after loin extract treatment. These results suggest that antioxidant activity and apoptosis-related effects of goat meat hot water extract varied with cut of meat under in vitro conditions. Because all data was obtained from the in vitro experiment, the ability to generalize conclusions is limited. Additional in vivo studies are necessary.
Protein is an essential nutrient for humans to sustain life, but it is predicted that it will be challenging to secure protein through the traditional livestock industry in the future. Microalgae has high future value as an alternative protein food source due to resource utilization and sustainability advantages. In order to increase productivity, the culture conditions of microalgae, Chlorella vulgaris, Dunaliella salina, and Scenedesmus obliquus were examined in this study. The optimal culture conditions of C. vulgaris were mixotrophic culture, 25oC culture temperature, 7.0 initial pH, 10% initial inoculation, stirring culture, 3000 Lux light intensity, and 24L:0D light/dark cycle period with red LED. For D. salina, the optimal culture conditions were mixotrophic culture, 20oC culture temperature, 8.0 initial pH, 10% initial inoculation, stirring culture, 6000 Lux light intensity, and 12L:12D light/dark cycle period with white LED. For S. obliquus, the optimal culture conditions were mixotrophic culture, 30oC culture temperature, 8.0 initial pH, 10% initial inoculation, stirring culture, 4500 Lux light intensity, and 14L:10D light/dark cycle period with fluorescent light. These findings can be used as important information for increasing the production of microalgae as an alternative protein material resource in the future.
In this study, the protein content and functional changes in soybeans cultured with Phellinus linteus HN00K9 were analyzed. P. linteus HN00K9 was cultured on soybeans. The crude protein content in soybeans cultured with HN00K9 (PMS) was 41.99%, which was higher than that in soybeans not cultured with the mushroom (UCS). The total free amino acid content in PMS increased to 39,963 mg/100 g, which was higher than that in UCS (36,817 mg/100 g). In particular, in PMS, glutamic acid accounted for 18.5% of the total amino acids at 7,413 mg/100 g. The total polyphenol content in PMS was 2.66 mg GAE/g, which was more than 45% higher than the amount in UCS (1.45 mg GAE/g). Additionally, PMS showed a DPPH radical scavenging activity of 33.3%, which was 3 times higher than that exhibited by UCS (11.5%), reflecting its high antioxidant content. Therefore, the PMS in this study has potential for use as a functional food material.
Protein can be provided by cultivating various microbes, which contain more than 30% protein content by cell dry weight. This study compared intracellular protein concentrations of various wild-type yeasts from different sources to select the best yeast strain with high protein concentration. Among them, Saccharomyces cerevisiae KCCM 34709, used for molasses fermentation, exhibited 4.1-fold higher protein concentration than a laboratory yeast strain, S. cerevisiae D452-2. In this study, an approach consisting of random mutagenesis coupled with the Bradford protein assay-based screening method was applied to enhance the S. cerevisiae KCCM 34709 protein content. Among 1,000 mutants, the #180 mutant strain produced 5,041±519 mg/L total amino acid in 48 h, which was 31% higher than the parental S. cerevisiae KCCM 34709 strain. These results demonstrate that the #180 mutant strain can be an attractive cell factory for animal-free protein production.
Microbial proteases are more economical than plant- and animal-derived proteases due to their ease of production and high activity. This study aimed to optimize the production of proteases from fermentative food-derived microorganisms. Five strains with proteolytic activity among 50 Bacillus sp. were first screened. Two strains with high protease activity were identified: Bacillus amyloliquefaciens SRCM 102139 and Bacillus subtilis SRCM 104999. SRCM 102139 strain and SRCM 104999 strain had the highest protease activity in 0.8% glucose and 0.3% yeast extract, and in 0.8% starch and 0.1% soy peptone, respectively. The production of protease for two strains was optimized by the Central Composite Design (CCD) under response surface methodology. The optimal conditions for protease production in SRCM 102139 were 0.5% and 0.347%, pH 6.0, for carbon (glucose) and nitrogen (yeast extract springer 0202) sources, respectively, with a predicted value of 0.929 U/mL. Additionally, the optimal conditions for protease production in SRCM 104999 were 0.5% and 0.5%, pH 6.7, for carbon (starch) and nitrogen (soy peptone HSP-349) sources, respectively, with a predicted value of 0.431 U/mL. The actual protease activities of SRCM 102139 and SRCM 104999 under the established conditions were 0.926 U/mL and 0.428 U/mL, respectively, closely matching the predicted values.
The purpose of this study was to analyze the physicochemical properties by mixing ratio of soybean and peanut protein to secure basic data for developing alternative protein foods. As a result of analyzing the protein molecular weight pattern, it was confirmed that the specific molecular weight was affected by the soybean and peanut protein mixture. The content of glutamic acid, aspartic acid, arginine, glycine, serine, alanine, and tyrosine among nonessential amino acids was higher as the mixing ratio of peanut protein to soy protein was higher. However, the higher the peanut protein mixing ratio, the lower the water absorption capacity. Based on the results of this study, further studies, such as selecting soybean and peanut cultivars for determining the optimal mixing ratio of soybean and peanut protein and processing methods to improve physical properties, are necessary
본 연구는 Tetraselmis suecica와 T. tetrathele의 영양물질 증진효과를 가져올 수 있는 배양시스템 구축을 위하여, 발광다이오드 (LED)의 파장별(청색; 450 nm, 황색; 590 nm, 적색; 630 nm) 탄수화물, 단백질, 지질 함량을 측정하였다. 두 종 모두 단백질 비율(42~69%)이 가장 높았으며, 생장속도가 낮았던 황색파장에서 탄수화물, 단백질, 지질의 높은 함량을 보였고, 생장속도가 가장 높았던 적색파장에서는 낮은 함량을 보였다. 이러한 결과는 세포 분열 속도의 감소로 인해 단백질 합성과 함께 세포의 화학적 조성과 효소 활동에 변화를 주어 지질과 탄수화물 함량이 증가한 것으로 생각된다. 따라서, T. suecica와 T. tetrathele의 유용한 생화학적 물질의 증대를 위해 대수생장기 초 기와 중기는 적색 LED 그리고 대수생장기 후기에는 황색 LED를 주사하는 2단계 LED 배양을 제안하였다.
