We studied the effects of initial pH, different nitrogen sources, and cultivation methods (shake flask and static culture) on biomass production, exopolysaccharides (EPS), and adenosine by Paecilomyces tenuipes. Relatively low pH levels were optimal for mycelial growth and EPS production. Yeast extract was the most effective organic nitrogen source for EPS production, whereas soybean extract was the best for adenosine production. A high C/N ratio was beneficial for adenosine production; however, excessively high C/N ratios reduced adenosine production. Static fermentation significantly increased adenosine production. A Box-Behnken design was used to optimize adenosine production; the optimal conditions for adenosine production by P. tenuipes were pH 7.0, soybean concentration of 3%, and a static culture period of 20 days, with the maximum adenosine production of 141.10 mg/L (predicted value: 128.05 mg/L).

Cordyceps militaris is widely used in China, Korea, and other Asian countries as both a traditional medicinal ingredient and an edible fungus. This study aimed to optimize the growth conditions and fruiting body production of C. militaris by investigating various culture media and physical parameters such as pH, aeration, illumination, temperature, spawn materials, and oat–sawdust-based substrate formulations. After a 7-day incubation period, oats with a pH of 6.0, under sealed and illuminated conditions at 32°C, demonstrated the most effective mycelial growth. Substrates consisting of 70% oat and 30% sawdust had the shortest incubation time of 30.5 days for fruiting body formation. The basidiospores showed a typical germination pattern where the sporidium produced a single germ tube that elongated, and branched to form monokaryotic primary mycelia. In conclusion, using oats as a substrate in the cultivation of C. militaris could reduce production costs and help protect the environment.

Recently, active research in Korea and worldwide has begun to focus on gene function and cultivar development using gene editing tools. This research, in addition to studies on edible mushroom, aims to enhance the physical and biochemical characteristics of mushrooms for applications in materials and substance production. For these studies, efficient isolation of protoplasts from the target mushroom is critical. However, several commercial cell wall-lysing enzyme cocktails, including Novozyme234, Glucanex, and Lysing enzymes, have recently been discontinued. In this study, we aimed to identify alternative enzyme systems to replace the discontinued cell wall-lysing enzymes for stable isolation of protoplasts from Ganoderma lucidum. To select an optimal osmotic buffer, enzyme function in 0.6 and 1.2 M Sorbitol, Sucrose, Mannitol, and KCl was assessed. The effect of reaction time was also evaluated. Protoplast isolation efficiency of each alternative enzyme was tested using lysing enzymes from Trichoderma harzianum, Chimax-N, and Yatalase, either individually or in combination. This matrix of studies identified enzymes and optimal conditions that could replace the discontinued lysing enzymes.

Hair dyeing, hair perming, and daily hair dryer use can substantially damage hair. Consequently, the demand for products containing natural ingredients for the care of damaged hair is growing. Although polyphenols with antioxidant effects are often used for hair conditioning, few studies have focused on hair conditioning, and the potential of Sparassis latifolia mushroom extract for hair improvement has not been evaluated to date. In this study, the antioxidant activity of and polyphenol content in hot water, 70% and 100% ethanol (EtOH), n-hexane, ethyl acetate (EtOAc), and water extracts of S. latifolia were analyzed. A hair treatment containing S. latifolia extract was prepared, and its effect on damaged hair was evaluated. The highest antioxidant activity was observed in the hot water and EtOAc extracts. Moreover, polyphenol analysis using high-performance liquid chromatography and liquid chromatography-mass spectroscopy confirmed that the EtOAc fraction has relatively high contents of specific polyphenols beneficial for hair. Based on these results, a hair treatment containing S. latifolia extract was applied to damaged hair, and hair improvement was evaluated using hair thickness, tensile strength, and scanning electron microscopy. The hair treatment containing 70% EtOH extract effectively improved hair condition. We postulate that this improvement was caused by the high hydrophobic polyphenol content in the 70% EtOH extract.

This study aimed to analyze crude protein, amino acid and bioactive changes in Phellinus linteus HN00K9 cultured oat. The crude protein content of P. linteus cultured oat (PCO) was 12.9%, which was higher than that of uncultured oats (UCO) as control at 11.26%. The total free amino acid contents of PCO and UCO were 11,4 mg/100 g and 9,686.205 mg/100 g, respectively. Glutamic acid accounted for 21% of the total amino acids of PCO, and the histidine content was increased by more than 51% in PCO compared to UCO. The total polyphenol content of PCO was 275 mg GAE/g, which was more than twice that of UCO (135 mg). The DPPH radical scavenging activity was 15.5% in PCO, which was more than five times that of UCO (3.5%). The β-glucan content of PCO was 12.5 g/100 g, which was more than five times that of UCO (3.2 g/100 g). Therefore, it is believed that PCO can be utilized as a material for various functional foods.

A new oak mushroom cultivar ‘K815’ was bred from monokaryotic strains of ‘Jadam-2’ and ‘LE191732-118’. The optimal temperature for mycelial growth of ‘K815’ was 25oC, and the temperature for primordia formation and fruiting body development was 15~20oC. The fruiting body morphology is hemispherical and the fruiting form is sporadic. Total cultivation period of the new cultivar, from inoculation to its first harvest, was 133 days, which was 3 days longer than control cultivar ‘Jadam’ (129 days), with 32 days for spawn running, 89 days for browning, 4 days for primordia formation, and 8 days for fruiting body development. In the fruiting body, the thickness and diameter of pileus of the new cultivar were 32.2 mm and 58.3mm, respectively, and the length of stipe was 35.6 mm. Compared to the control cultivar ('Jadam', 'L808'), the pileus of the new cultivar was thicker and smaller and the length of stipe was shorter. The total number of fruiting bodies of ‘K815’ was 58.9, which was higher than the control varieties and total yield of 2 crop cycle of ‘K815’ was 580 g (per 3 kg substrate), which was similar to the control variety.

We developed a new oyster mushroom cultivar named 'Dawon-tari', which has a longer stipe and higher yield than those of ‘sootari’.‘Dawon-tari’ was crossed by mating monokaryons isolated from ‘Sootari’ and ‘Daejang2ho’. The optimal temperature for mycelial growth was determined to be 20~25°C on potato dextrose agar medium, while the optimal temperatures for primordia formation and fruiting body growth were 15~17°C on a sawdust substrate. During bottle cultivation, the mycelial growth phase required approximately 26 days. Additionally, primordia formation required 5 days, and fruiting body growth took 4 days. The fruiting bodies exhibited a shallow funnel shape; were grayishbrown; and the stipes were characterized by a long, thin structure. The yield of fruiting bodies was 185 g per 1,100 mL bottle, which was 5% higher than that of ‘Sootari’.

The oyster mushrooms have known to be a major product in Gyeonggi-do, with production accounting for 69% of the entire country. The ‘Daeseon’ cultivar, which has white and straight stem, was developed. This cultivar was developed by mating monokaryons isolated from the ‘Heuktari’ and ‘Hwaseong-2ho’ varieties. The optimum temperature for the mycelial growth was 26~32oC on PDA medium and that for the primordia formation and the growth of fruit body of ‘Daeseon’ was 16~20oC on sawdust media. It took 35 days to complete spawn running, 3 days for finish primordia formation, and 4 days for finish fruit body growth in the bottle culture. It has shallow funnel-shaped pileus and a white straight stipe. The yield per bottle was 201 g/1,100 ml and was 16% higher than that of control cultivar ‘Suhan-1ho’. Based on above results, we expect this cultivar to be suitable for small packaging.

This study was conducted to evaluate the market potential of 'Hwanggeumsantari', a new oyster mushroom variety developed in Gyeonggi Province in 2019, and to derive effective market entry strategies. To objectively assess the marketability of 'Hwanggeumsantari’ from various perspectives, a panel of 50 consumers was formed, and surveys and interviews were conducted. Additionally, two Focus Group Interviews were held with distributors in the agri-food sector. The results showed that potential consumers were positive about the color and texture of ‘Hwanggeumsantari', but expressed relatively less satisfaction with its shelf life and aroma. Distributors, through tow Focus Group Interviews, evaluated that the product had sufficient market competitiveness, but called for efforts to improve cultivation techniques and enhance price competitiveness. Based on the results of market evaluations by consumers and distributors, a SWOT analysis was conducted. The main strengths identified were its outstanding color and texture, while the main weakness was its freshness issue. Meanwhile, the growing demand for healthy foods and various online sales channels could provide market entry opportunities for ‘Hwanggeumsantari', but high sensitivity to price in the case of premium pricing could pose a threat. Consequently, a market entry strategy for ‘Hwanggeumsantari’ was proposed using a 4P mix approach based on the SWOT analysis results. The product strategy emphasized premium positioning, small packaging, and the development of packaging technology to maintain freshness. The price strategy proposed premium pricing and the operation of incentive programs. The distribution strategy suggested channel diversification, direct stores, or direct sales, while the promotion strategy emphasized storytelling and collaboration with influencers.

Oyster mushroom is one of the most widely cultivated and consumed mushrooms in Korea, and mechanization and automation of cultivation systems have enabled mass production. Many cultivars have been developed to replace the old ones such as ‘Suhan‘ and ‘Chunchuneutari 2 ho,‘ which have been cultivated for over 20 years. Among these, ‘Soltari‘ was developed in 2015. Although it has excellent quality, its cultivation is challenging and the productivity is somewhat lower. To address these issues, the Mushroom Division at the National Institute of Horticultural and Herbal Science selected the genetic resource KMCC05165 and attempted hybridization between monokaryons from KMCC05165 and ‘Soltari(KMCC04940)’. Through repeated cultivation tests and evaluation of fruiting body characteristics, the superior strain ‘Po-2019-smj22’ was selected and finally named ‘Otari‘. The optimal mycelial growth temperature of ‘Otari’ was between 25 and 30°C and optimal fruiting body growth temperature was between 13 and 18°C. Mycelial growth on PDA medium was best at 25°C, and at the same temperature, mycelial growth was similar across four media: PDA, MEA, MCM, and YM. In 1,100 mL bottle cultivation, the yield was approximately 174 g, which is about 5% higher than the control cultivar ‘Soltari‘, and the number of valid individuals was also higher at about 25. The diameter and height of the pileus were 29.8 mm and 17.6 mm, respectively, slightly smaller than ‘Soltari‘, and the stipe was thin and long with a thickness of 12.2 mm. Additionally, the pileus’ lightness index (L index) was 30.7, indicating a darker brown color compared to 'Soltari.' With excellent mycelial growth, ease of cultivation, and high yield, the new cultivar ‘Otari‘ is expected to be widely adopted by domestic oyster mushroom farms.
