Fluorescent bacteria were isolated from sporocarps that browned into various mushrooms during survey at places of the production in Korea. We examined the pathogenicity, biodiversity, and genetic characteristics of the 19 strains identified as Pseudomonas tolaasii by sequence analysis of 16S rRNA and White Line Assay. The results emphasize the importance of rpoB gene system, fatty acid profiles, specific and sensitive PCR assays, and lipopeptide detection for the identification of P. tolaasii. As a result of these various analyses, 17 strains (CHM03~CHM19) were identified as P. tolaasii. The phylogenetic analysis based on the 16S rRNA gene showed that all strains were clustered closest to P. tolaasii lineage, two strains (CHM01, CHM02) were not identified as P. tolaasii and have completely different genetic characteristics as a result of fatty acids profile, specific and sensitive PCR, lipopetide detection, rpoB sequence and REP-PCR analysis. Pathogenicity tests showed 17 strains produce severe brown discolouration symptoms to button mushrooms and watersoaking of sporophore tissue within three days after inoculation. But two strains did not produce discolouration symptoms. Therefore, these two strains will be further investigated for correct species identification by different biological and molecular characteristics.

This study investigated the microscopic characteristics and genetic relationships of Ganoderma applanatum fruiting bodies. Basidiospores were brown, ellipsoid, and had one or two large vacuoles and a double wall. The surface of basidiospores was smooth or wrinkled and most had numerous small and shallow holes. The length and width of basidiospores of Ganoderma applanatum isolates GBGA-01, GBGA-02, ASI 50167, ASI 52821, ASI 52822, ASI 52823, and ASI 53399 were on average 7.6×4.8 mm, 7.9×4.6 mm, 7.7×4.9 mm, 8.2×5.3 mm, 7.7×5.0 mm, 8.0×4.9 mm, and 7.9×4.9 mm, respectively. In contrast, the basidiospores of Ganoderma lucidum isolate ASI 7125 were 7.7×5.2 mm. Using the universal ITS1/ITS4 primer set, the ITS region of the isolates were amplified and sequenced. The ITS sequences were very closely related to G. applantum isolate GBGA-01, GBGA-02, ASI 50167, ASI 52821, ASI 52822, ASI 52823 and ASI 53399, but were not the same species. Whereas, G. lucidum isolate ASI 7125 belongs to different group.

This study analyzed the effects of different nitrogen sources in substrate composition on the growth of Pleurotus ostreatus, as well as the subsequent changes in flavor and antioxidant activity. The T2, composed of poplar sawdust, beet pulp, cotton seed dregs, and cotton seed coat in a ratio of 40:20:20:20, exhibited the highest yield at 156.6 g. The total polyphenol content and ABTS and DPPH radical scavenging activities were 8.25 mg GAE/g, 70%, and 49%, respectively, showing higher radical scavenging activity compared to the Control and T1. Additionally, varying nitrogen content resulted in distinct aroma patterns and is presumed to influence taste profiles such as sourness, umami, and saltiness.

This study was conducted to selection and investigate appropriate conditions for mass production of antagonistic microbes to control cobweb disease caused by Cladobotryum mycophilum. A grampositive bacterium was isolated from spent substrate of Agaricus bisporus and showed significant antagonistic activity against Cladobotryum mycophilum. The bacterium was identified as Bacillus altitudinis. based on the cultural, biochemical and physiological characteristics, and 16S rRNA sequence. The isolate is saprophytic, but not parasitic nor pathogenic to cultivated mushroom whereas it showed strong inhibitory effects against C. mycophilum cells in vitro. The control efficacy of B. altitudinis HC7 against cobweb disease of C. mycophilum was up to 78.2% on Agaricus bisporus. The suppressive bacterium may be useful for the development of biocontrol system. To define the appropriate conditions for the mass production of the Bacillus altitudinis HC7, we have investigated appropriate culture conditions and effects of various nutrient source on the bacterial growth. The appropriate initial pH and temperature were determined as pH 6.0 and 30oC, respectively. The appropriate concentration of medium elements for the growth of pathogen inhibitor bacterium(Bacillus altitudinis HC7) was determined as follows: 3.0% soluble startch, 10% soytone, 1.0% (NH4)2HPO4, 1.0 mmol KCl, and 0.5% L-asparagine.

In this study, we investigated the microbial community of oyster mushrooms at different growth stages at the species level. Gram-positive bacteria were predominant in the presterilized medium. On the other hand, Gram-negative bacteria were predominant in the culture-completed medium, post-harvest medium, and fruiting bodies. In addition, Pseudomonas tolaasii, which is known to cause disease in mushrooms, was confirmed in the cultured medium, post-harvest medium, and fruiting bodies, and it was determined that the mycelium culture stage was contaminated, and the reason why no disease occurred was Sphingobacterium psychroaquaticum. It was confirmed that this was because the growth of Pseudomonas tolaasii was suppressed by producing a component called tolacin. As a result of confirming the diversity of microorganisms, it was confirmed that the presterilization medium contains a variety of microorganisms compared to other growth stages, and the diversity decreases in the order of culture completion medium, fruiting body, and post-harvest medium. showed a trend. As a result of microbial similarity analysis, it was confirmed that the cultured medium and the post-harvest medium showed similar microbial communities, and in the case of fruiting bodies, there were some similarities but overall differences.
