We investigated the effect of moderate- and high-intensity resistance training on hippocampal neurotrophic factors and peripheral CCL11 levels in high-fat diet (HFD)-induced obese mice. C57/black male mice received a 4 weeks diet of normal (control, CON; n = 9) or a high-fat diet (HF; n = 27) to induce obesity. Thereafter, the HF group was subdivided equally into the HF, HF + moderate-intensity exercise (HFME), and HF + high-intensity exercise (HFHE) groups (n = 9, respectively), and mice were subjected to ladder-climbing exercise for 8 weeks. The hippocampal brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) levels were significantly lower in the HF group than in the CON group (p < 0.05). In addition, in the HFME and HFHE groups were significantly higher than in the HF group (p < 0.05). The peripheral CCL11 levels were significantly higher in the HF group than in the CON group (p < 0.05). In addition, in the HFME and HFHE groups were significantly lower than in the HF group (p < 0.05). However, there was no significant difference according to the exercise intensity among the groups. Collectively, these results suggest that obesity can induce down-regulation of neurotrophic factors and inhibition of neurogenesis. In contrast, regardless of exercise intensity, resistance training may have a positive effect on improving brain function by inducing increased expression of neurotrophic factors.
본 연구는 5주령(180~200 g)의 SD계 흰쥐에 사염화탄소로 간 손상을 유발시킨 후 혼합 시료추출물(MSE)을 투여한 다음 혈액학적 및 생화학적인 parameter의 변동과 더불어 간 조직의 면역화학적 변화를 관찰하여 다음과 같은 결과를 얻었다. 실험동물의 식이량을 측정한 결과, 실험대조군(EC)군이 실험군(E1, E2)과 정상대조군(NC)과 양성대조군(PC)에 비해 통계적으로 유의하게 감소하는 경향을 나타내었고, 체중과 식이 섭취율은 실험대조군(EC)에 비해 실험군(E1, E2)과 정상대조군(NC)과 양성대조군(PC)은 통계적으로 유의하게 증가하는 간, 비장 및 흉선의 무게를 측정한 결과, 간의 무게는 실험 대조군(EC)이 정상대조군(NC)과 양성대조군(PC)에 비하여 약간 증가하였으나, 통계적으로 유의한 차이는 없었으며, 혼합 시료추출물(MSE)을 투여한 실험군(E1, E2)은 NC군 및 PC군과 유의한 차이가 없었다. 비장 및 흉선의 절대무게와 상대무게에 있어서도 모든 군에서 유의한 차이가 없었다. 실험대조군(EC)이 정상대조군(NC)에 비하여 혈청 ALT, AST, ALP와 LDH의 활성과 triglyceride(TG)의 함량이 모두 유의하게 증가하였고, 혈청 cholesterol(CHOL)의 함량은 모든 군에서 유의한 변동을 관찰할 수 없었다. 실리마린을 투여한 양성 대조군(PC)은 실험 대조군(EC)에 비하여 혈청 ALT, AST, ALP 와 LDH의 활성 및 TG의 함량 모두 유의하게 감소하였다. 그리고 혼합 시료추출물(MSE)을 투여한 실험군(E1, E2)은 실험 대조군(EC)에 비하여 혈청 ALT, AST, ALP와 LDH의 활성과 TG의 함량 모두 유의하게 감소하였다. 백혈구, 호중구, 림프구, 혈소판 수치와 혈색소의 함량 모두 실험대조군(EC)이 정상대조군(NC)에 비해 유의하게 높았고, 실험대조군(EC)에 비해 양성대조군(PC)인 실리마린 투여군이 유의하게 낮았다. 혼합 시료추출물(MSE)을 투여한 실험군 E1, E2군에서는 실험대조군(EC)에 비해 백혈구와 림프구가 유의하게 낮았고, 각 실험군(E1, E2) 간에는 약간의 차이는 있었으나, 유의성은 나타나지 않았다. 간조직의 면역조직화학적 발현 결과는 정상군대조군(NC) 및 양성대조군(PC)의 간 조직에서 SMA(smooth muscle actin) 항체에 대한 교원섬유 발현 부분이 미세한 양성으로 관찰되었다. 실험대조군(EC)에서는 간 섬유조직 발현 부분에서 얇은 띠를 형성한 강 양성의 갈색으로 관찰되어 통계적으로 유의성 있게 나타났으며, 혼합 시료추출물(MSE) 투여군인 E1과 E2군에서는 간 섬유조직 발현 부분에서 미세하게 갈색으로 염색되어 실험대조군(EC)에 비해 유의하게 낮게 나타났으며, 정상대조군(NC)과 양성대조군(PC)의 발현정도와 큰 차이가 없었다. 이상의 실험결과를 종합해 볼 때, 본 실험의 결과는 간 손상 예방과 치료, 더 나아가 건강기능성 식품 소재로서 혼합 시료추출물의 활용 가능성에 대한 기초자료로 이용할 수 있을 것으로 생각되며, 이와 관련된 추가적인 연구를 계획하고 있다.
Oxidative stress is one of common cause of fatty changes in the liver. Antioxidant capacity was confirmed in various vegetables including black radish (Raphanus sativus L. var niger). Fermentation of vegetables using Lactobacillus plantarum has been known to generate bioactive components. This study was conducted to determine if fermented black radish (FBR) ameliorates oxidative liver injury induced by CCl4 in rats. To accomplish this, FBR (250 and 500 mg/ kg) was orally administered to rats for 7 consecutive days, single CCl4 (1.5 mL/kg) treatment or no treatment orally. Serum chemistry at 24 hours after CCl4 injury showed that FBR (500 mg/kg) significantly reduced the level of both alanine aminotransferase and aspartate aminotransferase in CCl4 exposed rats. Moreover, FBR treatment significantly increased radical-scavenging effects in livers with the reduction of lipid peroxidation in CCl4 exposed rats. Histopathologic findings including Kupffer cell activation in the liver of each group matched those of serum chemistry. Collectively, black radish, through fermentation, exerts hepatoprotective capacity in CCl4 induced liver injury in rats through anti-oxidation.
PCB 제조에서 photoresist와 Copper Clad Laminate(CCL)의 구리표면과의 부착력을 항상시 키기 위하여 사용되는 soft etching제를 제조하기 위하여 과산화수소 사용을 배제하고, 유기산과 유기과 산화물을 이용하여 산의 종류, 농도, 에칭시간 등에 따른 구리표면의 에칭속도, 표면 조도, 및 오염도 등 을 조사하였다. 또한 에칭 후의 표면의 얼룩을 제거하기 위한 안정제의 최적 배합 및 농도도 확립하였 다. 본 연구 결과 유기산의 종류 중에서는 아세트산이 초기 구리 에칭속도가 가장 빨랐으며, 농도가 0.04 M이었을 때 0.4 μm/min이였다. 유기과산화물인 APS의 농도는 높을수록 에칭속도가 가장 빨랐으나, 표면 오염이 심각하였다. 안정제 용액의 조성도 표면 오염도에 큰 영향을 주었다. 결과적 0.04 M 아세 트산, 0.1M APS에 4 g/L의 안정제(ST-1)를 첨가한 에칭액의 경우 0.37 μm/min의 에칭속도와 표면 오염이 전혀 없으며, 표면 조도도 가장 우수하였다. 즉, CCL과 photoresist와 접착력을 향상시킬 수 있 을 것으로 판단된다.
Pleurotus eryngii (also known as king trumpet mushroom, french horn mushroom, king oyster mushroom) is an edible mushroom native to Mediterranean regions of Europe, the Middle East, and North Africa, but also grown in parts of Asia. It has the ability to produce various biologically active compounds and possesses a well-developed ligninolytic enzyme system that participates in the degradation of lignin and different aromatic compounds. In this study, we investigated the protective effects of the ethyl acetate extract of Pleurotus eryngii (PEE) on the development of atopic dermatitis (AD) in human keratinocyte HaCaT cells. Keratinocytes, one of major cell types in the skin, can be induced by TNF-α and IFN-γ to express thymus and activation-regulated chemokine (TARC/CCL17), which is considered to be a pivotal mediator in the inflammatory responses during the development of inflammatory skin diseases, such as AD. In addition, normal T-cell–expressed and secreted chemokine (RANTES) is a (C-C) chemokine released by T lymphocytes, other inflammatory cells, and platelets and plays an important role in allergic inflammatory processes. Pretreatment of HaCaT cells with PEE suppressed TNF-α/IFN-γ-induced protein and mRNA expression of CCL17 and RANTES. PEE significantly inhibited TNF-α/IFN-γ-induced NF-κB activation. These results suggest that PEE may exert anti-inflammatory responses by suppressing TNF-α and IFN-γ-induced activation of NF-κB in the keratinocytes and might be a useful tool in therapy of skin inflammatory diseases.

Protective effect of Corydalis ternata against the carbon tetrachloride-induced toxicity was investigated. Carbon tetrachloride(CCl₄) induces hepatotoxicity due to the reactive free radical(CCl₃*) generated by cytochrome P-450 enzyme. We examined effects of hexane, chloroform, butanol and water fractions prepared from the Corydalas ternata methanol extract. Rats were treated with those for 3 days, and liver microsomes and cytosols were prepared at 24 hour after last treatment. Hepatoprotective activity. of the water fraction was higher than that of other fractions. To examine mechanism of the hepatoprotective effect of Corydalis ternata, we measured contents of malondialdehyde(MDA), cytochrome P450(CYP), glutathione, calcium as well as the activities of NADPH-CYP reductase, glutathione S-transferase(GST), superoxide dismutase(SOD), glutathione peroxidase(GPX) and catalase. The fraction inhibited production of MDA, content of CYP and calcium in liver of water fractions - treated rats as compared with those of CCl₄ treated rats. The GST activity was increased. We speculate that the O₂ radical scavenging activities of the water fraction might play a key role in the mechanism opposing the progression of CCl₄ induced hepatotoxicity, but the activities of SOD, GPX, CAT were decreased. These results suggest that the mechanism might be mainly due to the decrease of CYP contents, act as calcium channel Mocker and increase of GST activity rather than O₂ radical scavenging activities.
Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl₄ induced hepatotoxicity. The hepatoprotective effect of the subfractions of butanol fraction has been evaluated by analyzing oxygen free radical scavenging enzyme activities and histopathological examinations. In BS-5 subfraction treated group, the activity of superoxide dismutase has been significantly increased as compared with that of CCl₄ treated rats. Antioxidant activity has been evaluated by the examination of the scavenging activity against 1,1-diphenyl-2-picrylhydrazyl radical. BS-5 subfraction has shown strong antioxidant activities. The histopathological examination showed that the treatment of BS-5 subfraction has relieved the ballooning degeneration of hepatocytes which had been generated by CCl₄. It appears that the protective effect of BS-5 subfraction would be mediated of the attenuation of lipid peroxidation by acting as a free radical scavenger, which were based on the increase of superoxide dismutase activity.
Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl₄-induced hepatotoxicity. The hepatoprotective effect of subfractions has been evaluated by analyzing blood and hepatocyte biochemical analyses and biotransformation enzyme analyses. Treatment of BS-5 subfraction has significantly decreased the activities of alanine aminotransferase and aspartate aminotransferase. In addition, the levels of cholesterol and triglyceride in liver have been decreased as compared with that of CCl₄ treated rats. The hepatoprotective effect of BS-5 subfraction on the CCl₄-induced hepatotocicity would be mediated of the attenuation of the level of cytochrome P450 and the enhancement of the activity of glutathion S-transferase.
흰쥐의 뇌로부터 읽는구조 전체를 포함하는 1.8kbp 크기인 IP_3K를 암호화 하는 IP_3KcDNA 유전자속의 NotⅠ부위 GCGGCCGC를 GCAGCCGC로 site-directed mutagenesis 하여 얻은 변이 IP_3KcDNA를 pSP72·Not2 운반체의 EcoR I 부위에 클로닝하여 증폭시키고, 유전자 재조합 과정을 거쳐서 포유동물의 발현 운반체인 pZIP·NeoSV(X)의 NotⅠ부위에 IP_3KcDNA를 서브클론하였다. 이것을 CCL39 hamster lung fibroblasts 세포에 transfection 하여 발현시키고, anti-IP_3K antibody를 사용하여 Western blot 법으로 효소량과 활성도를 각각 측정한 결과, 효소량은 5배, 효소의 활성도는 무려 16배로서 기대하였던 것보다 훨씬 발현효율이 높았다. 또한 흰쥐의 각 조직속에 IP_3K의 분포와 생화학적인 특성이 논의되었다.
The effect of water extract in fruits of Omija (Schizandra chinensis Ballion) on liver function in expermental liver injuries induced by CCl4. The levels of GPT, GOT activities in serum were decreased apparently on the dosage of water extract in fruits of Omija correspond to raw Omija 1g compared to control group. The levels of Microsomal protein, glycogen, and pyruvate in liver and of Urea nitrogen, GPT, GOT, LDH in serum showed a trend toward restoration of normalization. No effective of levels of hepatic G6P DH activity and serum free fatty acid by water extract in fruits of Omija.
본 연구는 4종의 별의별간(SS) 음료의 간보호 소재로서 의 이용 가능성을 조사하고자 항산화능 평가, t-BHP 와 CCl4로 산화적 손상 및 급성 간독성 유도한in vitro, in vivo 모델을 활용하여 간보호능을 평가하였다. 실험결과, 별의별간 01~04는 50 μM vitamin C 와 유사한 항산화 효과를 나타내었다. HepG2 세포에 t-BHP로 산화 스트레스를 유도한 뒤 나타나는 세포독성에 대해 별의별간 01 및 04에서 농도 의존적인 세포 보호효과를 보였으며, ROS 생성 억제에서 별의별간 01, 03, 04에서 농도의존적인 억제를 나타내었다. 미나리가 혼합된 별의별간 04에 대한 급성 간손상in vivo 모델을 활용하여 간보호능 검증 결과, 별의별간 04는 CCl4로 증가된 혈중 ALT, AST의 유의적 감소, 간 조직중 증가된 MDA 함량 감소 및 감소된 GSH의 유의적 증가를 나타내었다. 또한, 혈청 및 간 조직에서 증가 된 중성지방과 콜레스테롤을 유의적으로 감소시켰다. 이러 한 결과를 종합하며, 별의별간 04는in vitro 및in vivo 모델 에서 산화적 손상에 대해 간보호 효과를 나타내었다
Considering the high potential of the widely-used halogenated hydrocarbons on the global warming and ozone depletion, the development of effective thermal destruction methods of these compounds are quite urgent and indispensible. As part of the research efforts of this area, the destruction of CCl4 and flame characteristics have been investigated numerically by the co-firing CCl4 with CH4 in an industrial LNG-fired combustor as a function of molar ratio of the CCl4 to CH4 using a commercial code of STAR-CCM+. Considering a broad range of Damkohler number associated with the process of intensive CHCs (Chlorinated hydrocarbons) combustion with auxiliary fuel together with the inhibition reaction especially near flammability limits, a proper combustion modeling of CCl4 thermal destruction is quite desirable. In this study, however, after careful review of the literature about the flame characteristics of halogenated hydrocarbon together with the previous study about the modeling of the CCl4 flame based on the data of burning velocity, the eddy breakup turbulent combustion model was employed since it is quite reasonably assumed that chain branching reaction looks dominant in most flame region over the halogenated inhibition effect in strong turbulent reacting flows. One of the most useful results based on this study is that; without any incorporation of flame inhibition effect, the length of co-fired flame increases steadily as the ratio of CCl4 to CH4 (R) increases from 0.0, 0.1, 0.2 to 0.5, and 1.0 together with the increase of the maximum flame and exit gas temperature. The reason of the increase of the flame length with the increase of flame temperature can be explained by the presence of the additional CCl4 fuel with low heating value. Further a detailed discussion has been made on the thermal destruction of CCl4 together with the Cl2 concentration by Deacon reaction.
