        Gray leaf spot caused by Stemphylium spp., is a major disease of tomatoes, and it threatens its cultivation worldwide, especially in warm and humid areas. This study was conducted on 223 tomato germplasm conserved at the National Agrobiodiversity Center to select the resources resistant to the gray leaf spot pathogen strain previously isolated in Korea, using a bioassay and genotypic analysis of the resistance gene (Sm). Two weeks after inoculation with Stemphylium lycopersici, the disease index (rated on a scale of 0-4) of gray leaf spot was assessed in detached tomato leaves. The results showed that 22 resources were resistant, with a disease index of 0-1. Additionally, 65 genetic resources were found to be moderately resistant, with a disease index between 1.0 and 2.0. Subsequently, Hybridization Probe Melting (HPM) analysis of the 22 resistant genetic resources confirmed the genotype of the gray leaf spot resistance gene (Sm). Among them, 20 genetic resources showed a homozygous resistant genotype. The resources selected in this research may contribute to the breeding of new tomato varieties resistant to gray leaf spot and may serve as a basis for further genotypic analysis studies.

        본 연구는 세계채소센터의 고추 유전자원들에 대한 침수조건에서 생육특성을 조사해 침수 저항성 자원을 선발하기 위해 수행하였다. 세계채소센터 유전자원센터에서 보유중인 4종의 고추 8자원을 선발하여 유리온실에서 종자를 파종하여 2개월 간 육묘한 후 본 실험을 위해 비닐하우스로 이동 후 침수시험을 수행하였다. 시험에 이용된 고추는 고온인 여름철에 시험이 수행되어 침수에 대한 반응이 매우 빠르게 나타났다. 1. 침수 처리구에서 생육이 전체적으로 감소 되었으나 저항성 자원들은 대조구와 비교하여 초장에 있어 유의성 있는 차이를 보이지 않았다. 2. 침수 처리 후 고추자원의 엽온 조사에서는 No. 29 (9656-13) 자원을 제외한 모든 침수구에서 엽온이 대조구에 비해 높게 유지되었다. 3. 감수성 자원들은 위조지수가 1.5 이상을 나타냈으나 내침 수성 자원들은 0.5로 낮게 유지되었다. 4. SPAD 값을 조사한 결과에 있어서는 침수로 인해 전체적으로 대조구에 비해 10% 낮은 수치를 나타냈다. 5. Proline 함량에 있어서는 침수구에서 전체적으로 Proline 함량이 높게 나타났으나 내침수성이 강했던 No. 8과 22는 대조구와 유의차가 없이 낮았다. 6. 침수자원들에 대한 침수와 고온에 따른 화분활력 조사에서는 침수에서 일부 자원의 화분활력 감소를 확인하였으며, 고온처리에서 처리구 대부분의 화분 활력이 유의하게 감소하는 결과를 나타냈다. 7. 본 시험을 통해 이번 시험의 고추자원 중 Capsicum bacatum 종인 No. 8 (VI028788), No. 12 (VI044312) 자원 이 침수저항성이 강한 자원으로 판단되어 1차 선발하였다.

        1. 배추(Brassica rapa L. ssp. pekinensis) 국내외 수집 유 전자원 170점에 대한 TuMV(Turnip mosaic virus, TuMV) 저항성 유전자원을 선발하기 위하여, 유전자원의 잎에 국내 분리 계통인 TuMV-HY을 즙액 접종하였다. 2. TuMV를 접종한 배추 유전자원 개체들은 20oC±2oC에서 재배하였으며 4주 동안 바이러스 병징 발현을 육안으로 조사하여 저항성을 평가하였다. 3. 배추 유전자원은 약한 모자이크, 얼룩에서 심한 모자이크 등의 전신 감염 병징을 보였으며 TuMV 감염이 역전사중합효 소반응으로 확인되었다. 4. 바이러스 병징을 보인 배추 유전자원들은 TuMV에 대한 감수성 유전자원들로 판정되었다. 이들 중 공시한 국내 수집 자원들 모두가 감수성으로 판정되었다. 5. 13개 배추 유전자원들은 감수성 개체들에서 관찰된 모자이크 등 전신 감염 병징이 발현되지 않았으며 35일 동안 병징이 관찰되지 않았다. 6. 역전사중합 효소 연쇄반응으로 조사한 결과 13개 배추유 전자원들의 상위 잎들에서 TuMV가 검출되지 않았으므로 13 개 배추유전자원들이 TuMV에 대한 저항성 계통들로 판정되었다.

        본 연구는 기능성 곡물로 이용되고 있는 아마란스(Amaranthus ssp.)를 어린잎채소 용도로 개발하기 위하여 국내에 보유되어 있는 유전자원에 대한 어린잎 생육특성, 엽 적색도 발현 정도 및 기능성 물질의 함량 변이를 평가함으로써 어린잎채소로서 이용 가능한 유망 아마란스 자원 선발을 위하여 수행되었다. 국내에 수집․보존되어 있는 아마란스 유전자원 262 종에 대하여 색도색차계로 측정한 엽 적색도 발현 정도를 기준으로 전체 자원의 상위 15%에 해당하는 39종을 1차 선발하였다. 1차 선발된 유전자원에 대하여 고온기 및 저온기의 재배를 통하여 어린잎 생육과 유전자원 별 적색소 발현을 검증 평가하여 안정적인 적색 강도를 보이는 고색도 자원을 최종 선발하였다. 아마란스의 엽 적색도는 모든 1차 선발 계통들에 있어서 고온기에 비해 저온기 재배 시 적색도 발현이 우수하였으며 2회의 재배를 통한 평균 적색도 발현 정도를 기준으로 IT199970 등 11종의 자원을 선발하였다. 이들 선발된 유전자원들에 대한 어린잎 생체중 및 총페놀 함량, 총플라보노이드 함량 정보를 제시하였다. 이들 선발 종들은 5개의 아마란스 종에 속하였으며 엽 적색도 발현이 우수하여, 향후 적색 풍미를 더하는 어린잎 샐러드 채소의 소재로서 이용 가능할 것으로 판단된다.

        Genomic DNA prepared from fruitbdies of 25 Grifola frondosa strains were amplified with the ITS primers and the PCR reaction products were enzymed. The ITS bands have same electrophoresis patterns but part of them have different restriction enzyme cutting site. These strains were divided into three broad categories. SRAP amplification employing 47 SRAP primer pairs were carried on and 138 polymorphic bands were detected. The phylogram tree showed that: ten strains were differentiated from the others effectively and fifteen strains have no obvious differences between each other. The results implied that the ITS-RFLP and SRAP markers were effective methods for strains identifica tion and germplasm evaluation but other markers were also needed to be used in conjunction.

        39 accessions of sickle alfalfa (Medicago falcata L.) collected from different regions of China were evaluated for their tolerance to drought. Differences were observed among accessions in their reactions to drought and 8 good tolerant accessions at populations level and 18 excellent tolerant accessions at individual level were identified based on drought response index (DRI).

        한국재래종과 도입종을 포함한 62계통의 고추를 경북대학교 부속농장에 재배하여 종자를 증식하는 동시에 개화소요일수를 포함한 16가지 형질의 특성을 조사 기록하였다. 개화소요일수에 있어서는 KC85(칠곡 동명)와 KC116(PI308791), KC154(Cherry Sweet) 및 KC185(조선고추)가 83일로 가장 조생이었고 KC139(Serrano Chili)와 KC103(PI297488)이 126일로 가장 만생이었다. 과실을 따는데 걸리는 힘에 있어서는 KC85(칠곡 동명)가 0.45kg으로 가장 쉽게 따지고 KC162(PI224444)가 3.34kg으로 가장 많은 힘이 걸렸다. 흰가루병에는 KC10(Pimiento), KC23(PI224423), KC51(PI257044), KC56(PI257053), KC126(PI358812), KC119(PI322719), KC143(Emerald Giant), KC148(VR-2), KC163(PI224445), KC166(Papari Mild)이 발병이 적었다. 탄저병 발생은 KC39(Serrano Chili)에서 가장 적었다. 고추 품종의 수집 및 도입활동을 계속하여 한국재래종 14점과 도입종 25점을 추가 확보하였다.

        Identification of Fusarium oxysporum f. sp. raphani and investigation on fusarium wilt development by isolates and inoculation methods were conducted to establish a screening method for fusarium wilt-resistance in radish (Raphanus sativus L.) germplasm. Pathogenicity of F. oxysporum f. sp. raphani isolate, Heonggye-1 and Heonggye-2, to radish plants was confirmed by seedling test. Radish seedlings were inoculated by root-dipping and soil-drenching with or without root-wounding. For Heonggye-1 isolate, mean disease indexes were 4.13 and 3.91 by root-dipping and soil-drenching with root-wounding, respectively, but those were 1.87 and 1.88 without root-wounding. For Heonggye-2 isolate, mean disease indexes were 3.83 to 4.37 regardless of inoculation methods. Two-hundred sixty accessions of radish germplasm collected from 9 countries of Asia and Europe were evaluated for fusarium wilt-resistance by soil-drenching with root-wounding with Heonggye-2 isolate. Fifty-four resistant accessions with higher than 70% of the percentage of resistant seedlings in accession (PR) and lower than 20% of the percentage of susceptible seedlings in accession (PS) was found. Eighteen susceptible accessions with lower than 20% of PR and higher than 50% of PS were selected. These accessions could be used as breeding and research materials after re-evaluation of disease-resistance and characterization of agronomical traits.

        This study was conducted to characterize the amylose and protein contents of 4,948 rice landrace germplasm using the NIRS model developed in the previous study. The average amylose content of the germplasm was 20.39% and ranged between 3.97 and 37.13%. The amylose contents in the standard rice were 4.99, 18.63 and 20.55% in Sinseonchal, Chucheong and Goami, respectively. The average protein content was 8.17% and ranged from 5.20 to 17.45%. Protein contents in Sinseonchal, Chucheong and Goami were 6.824, 6.869 and 7.839%, respectively. A total of 62% germplasm were distributed between 20.06% and 27.02% in amylose content. Germplasm of 81.60% represented protein content of 6.78-9.75%. The distinguishable ranges of amylose contents according to origin were 16.58-20.06% in Korea, 20.06-23.25% in Japan, 23.25-27.02% in North Korea, and 27.02-37.13% in China. In the protein content, approximately 30% of Chinese resources ranged from 9.75 to 17.45%, whereas less than 10% were detected in other origin accessions. Fifty resources were selected with low and high amylose ranging from 3.97-6.66% and 30.41-37.13%, respectively. Similarly, fifty resources were selected with low and high protein ranging from 5.20-6.09% and 13.21-17.45%, respectively. Landraces with higher protein could be adapted to practical utilization of food sources.

        The aim of this study is to select the superior resources of high yield, high content of functional material optimal to mechanical harvesting by the evaluation of crop growth and yield characteristics in sorghum germplasm. One hundred accessions of sorghum germplasm were used in this experiment. Days from seeding to heading date showed the range from 68 to 94 days with the highest frequency proportion was the group from 80 to 85 days, which occupied 34% (34 plant resources) of 100 germplasm. Ear types of 100 sorghum germplasm could be classified as 7 types of broom-tillering, half broom-tillering, extreme open-loose type, open-loose type, intermediate type, compact type, extreme-compact type of which intermediate type was the highest ratio of 28% (28 plant resources) of 100 germplasm. Yield showed the range from 106 to 365 ㎏/10a with the highest frequency proportion of it was the group from 150 to 200 ㎏/10a, which occupied 44% (44 plant resources) of 100 germplasm. Among 100 sorghum germplasm, 18 ideal resources of high yield and short plant height appropriate for mechanical harvesting were selected. In order to evaluate high content of functional substance, selected 18 resources were analyzed for total polyphenol content, DPPH radical scavenging activity and total anthocyanin content. Finally, we selected 5 resources of short plant height, high yield, high content of total polyphenol and high DPPH radical scavenging activity among 18 genetic resources.

        한국자원 128개와 중국자원 214개 및 미국자원 37개를 포함한 총 379개의 국내외 콩 유전자원에 대한 지방 함량과 지방산 조성을 평가하고 유용자원을 선발하여 고품질 콩 품종 개발의 기초자료로 활용코자 수행되었다. 한국자원과 중국자원은 농촌진흥청 농업유전자원센터로부터, 미국자원은 경북대학교 식물유전육종연구실로부터 분양 받았다. 콩 유전자원의 지방함량은 평균 19.1%이었고, 12.7∼26.0%의 범위이었고, 포화지방산인 palmitic acid의 함량은 평균 10.6%이었고 범위는 4.1∼18.1%, stearic acid의 함량은 평균 2.9%이었고 범위는 1.9∼12.7%이었다. 불포화지방산인 oleic acid의 함량은 평균 28.2%이었고 범위는 15.3∼56%, linoleic acid의 함량은 평균 51.2%이었고 범위는 24.4∼68%, linolenic acid의 함량은 평균 7.1%이었고 범위는 3.4∼11.1%이었다. linoleic (ω-6)/linolenic(ω-3) acid 함량의 비율은 평균 7.5이었고 범위는 3.8∼17이었다. 포화지방산이 평균 13.5%이었고 범위는 6.7∼21.7%, 그리고 불포화지방산의 함량은 평균 86.5%이었고, 범위는 78.3∼93.4%이었다. 고지방품종 개발을 위한 지방함량이 25%이상인 고지방함량 자원으로는 중국자원인 Suinong 14, Dongnong 434 및 Dongnong 46의 3개이었다. Oleic acid 함량이 50%이상인 고 oleic acid함량 자원으로는 미국자원인 KLG12073과 KLG12074, 중국자원인 Jilin 14 및 한국자원인 Kwangan과 Bongeui의 5개이었고, linoleic acid의 함량이 60%이상인 고 linoleic acid 함량 자원으로는 미국자원 KLG12083과 KLG12081을 포함한 8개이었다. Linolenic acid의 함량이 4%이하인 저 linolenic acid 함량 자원으로는 미국자원 KLG12093와 KLG12096을 포함한 7개이었다. Linoleic acid(ω-6)/linolenic acid(ω-3)의 비율이 4.5이하로 낮은 자원은 미국자원 KLG12074 (3.8)와 KLG12073 (3.8), 한국자원 Kwangan (3.8)와 Bongeui (4.5) 및 중국자원 Dongnong 47 (4.3)의 5개이었다.

        Low temperature germinability (LTG) is an important trait for breeding of varieties for use in direct-seeding rice production systems. Although rice (Oryza sativa L.) is generally sensitive to low temperatures, genetic variation for LTG exists and several quantitative trait loci (QTLs) have been reported. The objective of this study was to develop and employ high-efficiency molecular markers for evaluation of LTG in rice germplasm. A panel of japonica rice accessions (n=180) from temperate regions in Asia was evaluated for LTG and genotyped with markers from regions previously reported to harbor other LTG QTLs. ANOVA revealed that four markers on chromosome 2, 4, and 11 from previously reported QTLs showed highly significant value (p < 1.0e-04) and their R2 ranged 0.083 (qLTG11-1) to 0.190 (qLTG4b-1). An association analysis was conducted using SNP data generated by sequencing of the panel. Eight SNP markers were found to be significantly associated with LTG using general and mixed linear models. Three SNP-based CAPS and dCAPS markers from these results were developed and showed higher accuracy in predicting sensitive LTG germplasm. These new LTG markers will be useful for molecular evaluation of germplasm, particularly to identify sensitive or weak LTG accessions.

        Sesame (Sesamum indicum L.) is one of the most important oilseed crops with high oil contents and rich nutrient value. The development of a core collection could facilitate easier access to sesame genetic resources for their use in crop improvement programs and simplify the genebank management. The present study was initiated to the development and evaluation of a core collection of sesame based on 5 qualitative and 10 quantitative trait descriptors on 2,751 sesame accessions. The accessions were different countries of origin. About 10.1 percent of accessions were selected by using the power core program to constitute a core collection consisting of 278 accessions. Mean comparisons using t-test, Nei’s diversity index of 10 morphological descriptors and correlation coefficients among traits indicated that the existing genetic variation for these traits in the entire collection has been preserved in the core collection. The results from this study will provide effective information for future germplasm conservation and improvement programs in sesame.

        Soybean [Glycine max. (L.) Merr] is one of the most important legumes in the world. However, soybean varieties are sensitive to flooding stress and their seed yields are substantially reduced in response to the flooding stress. 192 soybean germplasm collection was screened to identify flooding tolerant germplasm at an early vegetative growth stage (V1). Soybean plants at V1 stage were waterlogged for 4 to 10 days. To evaluate flooding tolerance, survival rate were investigated as a time dependent manner. Jangbaegkong, Danbaegkong, Sowonkong, Socheong2 and Suwon269 showed flooding tolerance, while Shillog, T201, T181, NTS1116 and HP-963 showed flooding sensitivity. We also investigated effects of flooding stress on soybean morphology. The adventitious root development was greatly increased in flooding tolerant plants compared to it in flooding sensitive plants. In addition, root length and root number were analyzed. The significant reduction of root length and root number was observed in flooding sensitive plants. Thus, these results indicate that the morphological changes in roots are important for acclimation to flooding stress. Taken together, the relationship between the morphological changes in the roots and flooding tolerance may be useful in selecting a flooding tolerant soybean germplasm.

        The objective of this study was to rapidly evaluate fatty acids in a collection of millet (Panicum miliaceum subsp. miliaceum) of different origins so that this information could be disseminated to breeders to advance germplasm use and breeding. To develop the calibration equations for rapid and nondestructive evaluation of fatty acid content, near-infrared reflectance spectroscopy (NIRs) spectra (1104-2494 nm) of samples ground into flour (n =100) were obtained using a dispersive spectrometer. A modified partial least-squares model was developed to predict each component. For foxtail millet germplasm, our models returned coefficients of determination (R2 ) of 0.89, 0.89, 0.89, and 0.92 for palmitic acid, oleic acid, linoleic acid, and total fatty acids, respectively. The prediction of the external validation set (n=10) showed significant correlation between references values and NIRs values (r2 =0.64, 0.90, 0.79, and 0.89 for palmitic acid, oleic acid, linoleic acid, and total fatty acids, respectively). Standard deviation/standard errors of cross-validation (SD/SECV) values were close to 3 (2.62, 2.40, 1.85, and 2.23 for palmitic acid, oleic acid, linoleic acid, and total fatty acids, respectively). These results indicate that these NIRs equations are functional for the mass screening and rapid quantification of the oleic and total fatty acids characterizing millet germplasm. Among the samples, IT153514 showed an especially high content of fatty acids (48.14mg~;g-1 ), whereas IT123909 had a very low content (34.44mg~;g-1 ).

        We collected and examined germplasm resources of lettuces, edible chrysanthemums and mallows for organic farmers. We found that 3 genotypes (OL-51, OL-52, and OL- 63) are suitable for organic cultivation. The main use of treatment for grey mold among eco-friendly pest control methods is helpful to increase efficiency of seedgathe끼ng after collecting and evaluating a total of 64 kinds of lettuces- collected 43 South Korea local varieties, 8 kinds cultivated by the Highland Agriculture Research Center, 4 kinds of cultivated lettuces, and 9 kinds for sales- to choose approp꺼ate kinds of lettuces for organic cultivation in South Korea. We chose those with more than 300 leaves (OC-18, OC-44, and OC-51) after evaluation of yield characteristics of a total of 62 edible chrysanthemums. We covered each genotype of seed of edible chrysanthemums when gathe끼ng the seeds by ourselves so as not to decrease the purity of each genotype. We chosed OM-38, OM-39, and OM-40 for their yield performance and germination rates among the total 41 genotypes of mallows. We selected 9 genotypes of lettuces, editable chrysanthemums and mallows with possibility of organic cultivation and found that they can be used permanently through self seed-gathering on organic farms.

        Maize (Zea mays.L) is the third important crop after wheat and rice in Afghanistan. The productivity is low 2.14 ton/ha due to lack of diversity and well-adapted varieties. 65% of maize production is used as feed and 17.5% as food. Afghanistan is facing seriously malnutrition and poverty, therefore more maize is required to recover food security. Popcorn is also important snack food in Afghanistan, but it is luck of breeding donors. The main objective of this work is to evaluate and identify the introduced hybrids of Korean germplams. International Corn Foundation (ICF), Kyungpook National University (KNU) and Institute of Asian Culture and Development (IACD) collaboratively planted three different types of maize (Silage1, KNU waxy1, Popcorn) in two locations (Isharak and Sherabad) in Mazar-i-sharif. All hybrids have shown good performances and can easily be adapted in Afghanistan climate. Among them KNU waxy1 was the most attractive which has been planted first time. Farmers showed highly interest on new taste and they were so enthusiasm to replant. Maize breeding laboratory at KNU is developing new popcorn cultivar for commercial use in Korea that has shown outstanding result in Afghanistan climate. This experiment described that introducing of new maize hybrids especially KNU waxy1 and popcorn can have great potential to fill up food demand in Afghanistan.
