        본 연구는 미역(Undaria pinnatifida) 추출물과 쇠미역(Costaria costata) 추출물의 항산화 활성 및 HepG2 세포의 알코올로 인한 산화 손상에 대한 보호 효과를 연구하였다. 미역과 쇠미역 추 출물의 총 폴리페놀과 플라보노이드 함량은 70% 에탄올 추출물에서 가장 높았다. 또한 미역과 쇠미역 70% 에탄올 추출물의 DPPH (IC50 0.33±0.21, 0.48±0.47 mg/ml), ABTS (IC50 0.34±0.30, 0.47±0.17 mg/ml) 라디칼 소거 활성이 열수 추출물 및 10% 에탄올 추출물 보다 높았다. 추출 물의 간 보호 효과를 확인하기 위하여 HepG2 세포에 알코올 산화 스트레스를 유발하여 MTT 분석을 이용하여 세포 생존력을 측정하였다. 미역 및 쇠미역 열수 추출물은 알코올 처리군 (73.95%) 대비 세포 생존율은 각각 89.91~97.63% 및 84.99~90.54%로 농도 의존적으로 증가시 켰다. 본 연구는 미역과 쇠미역 추출물이 간 보호 및 항산화 효과를 나타냄을 확인하였고 알코 올 산화 손상에 대한 간 보호 소재로 활용될 수 있을 것으로 사료된다.

        Pollution of agricultural soil by alkaline salts, such as Na2CO3, is a critical and long-lasting problem in cultivable land. The aim of the study was to examine the putative role of citric acid (CA) in alleviating Na2CO3-stress in alfalfa. In this study, Na2CO3 significantly induced leaf chlorosis, inhibited plant growth and photosynthesis related parameters, increased hydrogen peroxide (H2O2) and reduced major antioxidant enzymes (SOD ,CAD, APX) in alfalfa. However, the presence of CA these negative effects of Na2CO3-stress largely recovered. Interestingly, expression of antioxidant and ion transporter genes (Fe-SOD, CAT, APX, DHAR and NHX1) involved in Reactive oxygen species (ROS) homeostasis and oxidative stress tolerance in alfalfa. These findings suggest that CA-mediated Na2CO3 stress alleviation is an ecofriendly approach that would be useful to local farmer for alfalfa and other forage crop cultivation in alkaline soils.

        This study was carried out to investigate the effects of supercritical carbon dioxide (SC-CO2) extracts from sweet potatoes (SP) and watermelon (WM) on the oxidative stability of perilla seed oils (POs) over the existing ones. A comparison was done between the oxidative stability of perilla oil (PO) after the addition of 0.1% of SP, and WM extracts and PO without extract. The oxidative stability was measured based on the viscosity, acid value (AV), peroxide value (POV), antioxidant (DPPH) activity, p-anisidine value (p-AV), and fatty acid composition. The viscosities ranges were: PO without extract, from 53.99±0.99 to 74.38±1.61 cps, PO with SP extract, from 53.99±0.10 to 58.73±0.8 cps, and PO with WM extract, from 53.98±0.10 to 56.00±0.70 cps. While the PO containing the SC-CO2 extracts had significantly lower AV, POV, and p-AV, their antioxidant activity was approximately 10 times higher than that of the PO without extract. There were no significant differences in fatty acid composition between SC-CO2 extracts added groups and PO without extract (p<0.05). The findings of this study confirmed that the SC-CO2 extracts from sweet potatoes and watermelon enhanced the oxidative stability of perilla seed oils, and are potential natural antioxidants for use in the food industry.

        본 연구에서는 가물치(Channa argus) 추출물의 신경세포 분화와 산화 스트레스에서의 효능을 분석하기 위하여 녹차와 효소를 이용한 다양한 추출 방법(상온 추출물, RE; 녹차 상온 추출물, GRE; 효소 상온 추출물, ERE; 녹차 효소 상온 추출물, GERE)을 사용하여 제조 된 추출물의 아미노산 조성과 항산화 활성을 비교 분석하였고, 신경성장인자 (NGF) 유도 신경세포 분화 및 과산화수소 처리에 의해 유도된 PC12 세포 독성에 대한 보호효과를 규명하고자 하였다. 총 아미노산 함량은 RE 및 GRE보다 효소 추출물인 ERE 및 GERE에서 훨씬 더 높았다. 효소 가수 분해물 (ERE 및 GERE)에서 ABTS 라디칼 소거 활성은 RE 및 GRE보다 높았다. 또한, RE와 ERE는 PC12 세포에서 neuronal growth factor (NGF) 매개 신경 돌기 성장뿐만 아 니라 growth associated protein (GAP)-43 및 synapsin-1의 발현을 현저하게 향상 시켰다. 과산화수소(H2O2)에 의해 손 상된 PC12 세포에 4가지 유형의 Channa argus 추출물을 첨가한 후 PC12 세포의 생존율을 측정하였다. PC12 세포 의 생존율은 RE, GRE, GERE에서 각각 77.5±1.9%, 84.0±0.8%, 81.1±0.9%이였다. 이러한 세포 생존율은 H2O2 만을 처리 한 음성 대조군(70.0±2.0%)에 비해 더 높았다. H2O2 처리에 의해 유도 된 세포 독성도 RE, GRE 및 GERE 처리에 대한 반응으로 상당히 완화되었다. 종합하면, Channa argus 추출물은 산화 스트레스와 신경 손상을 감소시키는 기능성 물질로 유용하다는 것을 시사하며, 향후 이들 소재를 활용한 다양한 기능성 제품의 개발이 필요할 것으로 판단된다.

        Larvae from the marine medaka fish Oryzias javanicus were exposed with polystyrene microplastics (MPs) for 24 h. Exposure to waterborne fluorescent MPs showed clear ingestion and egestion in feces. Under constant MPs, the concentration of dissolved oxygen significantly decreased in 24 h compared to the control. Significant intracellular reactive oxygen species and malondialdehyde contents were detected in larvae, indicating oxidative stress and lipid peroxidation. Significant elevations in mRNA expressions of heat shock protein 70 and antioxidant defense system genes (glutathione reductase, glutathione peroxidase, catalase, and superoxide dismutase) were measured with increases in enzymatic activity of oxidative stress-related proteins. Taken together, the alterations to the molecular and biochemical components suggested that waterborne MPs had an oxidative stress effect on marine medaka larvae.

        Dimethachlor is a synthetic herbicide, belonging to the chloroacetanilide group, that inhibits the undesirable growth of weeds via the suppression of very longchain fatty acid synthesis. Although dimethachlor has been shown to run off from agricultural fields into aquatic ecosystems, the toxicity of dimethachlor on aquatic invertebrates and vertebrates is unknown. In our study, we assessed the toxicity of dimethachlor on developing zebrafish embryos by analyzing viability, hatching ability, and phenotypic changes. Embryonic viability decreased from 48 h post-fertilization (hpf) at the highest concentration of dimethachlor. Decreased hatching ratio, shortened body length, and pathological changes in the eye, heart, and yolk sac were observed at sub-lethal concentrations. Additionally, dimethachlor increased the number of apoptotic cells and level of reactive oxygen species 120 hpf. Our results indicate that dimethachlor may act as an anti-developmental toxicant when accumulated in an aquatic environment.

        Gangliosides are glycolipids in which oligosaccharide is combined with sialic acids. Our previous studies have suggested an interplay between ganglioside GD1a/GT1b and meiotic maturation capacity in porcine oocyte maturation. Furthermore, ganglioside GD1a and GT1b are known for its antioxidant activity, but it is still unclear whether possible antioxidant role of GD1a and GT1b is involved in porcine embryos development competence during in vitro culture (IVC). Here, the effects of ganglioside GD1a and GT1b on the embryonic developmental competence during in vitro culture of porcine were investigated. The effects of ganglioside GD1a and GT1b on the expression of ST3GAL2 were confirmed during embryos development (2-cell, 4-cell, 8-cell and blastocyst) using immunofluorescent staining (IF). As a result, the fluorescent expression of ST3GAl2 was higher in embryos at 4-8 cells stage than blastocysts. Blastocyst development rate significantly increased in only 0.1 μM GD1a and GT1b treated groups compared with control group. To investigate the cellular apoptosis, we analyzed TUNEL assay. In case of only 0.1 μM GD1a and GT1b treated groups, the total number of cells in blastocyst compared with control group, but there was no significant difference in the rate of apoptotic cells. We identified the intracellular ROS levels using DCF-DA staining. According to the result, ROS production significantly decreased in blastocysts derived from the 0.1 μM GD1a and GT1b treated groups. These results suggest that ganglioside GD1a and GT1b improve the developmental competence of porcine embryos via reduction of intracellular ROS during preimplantation stage.

        This study examined the bioactive compound content and the antioxidant activities of bitter melon (Momordica charantia Linn.) leaves. The content of vitamin C, beta-carotene, and total carotenoids was 69.77, 45.68, and 65.08 mg/100 g, respectively. To investigate the antioxidant capacity, bitter melon leaves were extracted using various concentrations of ethanol (60, 80, or 100%). Highest content of total polyphenols (18.07 mg gallic acid equivalent/g) and flavonoids (4.53 mg cathechin equivalent/g) was found in the 100% ethanolic extract of the leaves (E100). Also, the E100 extract showed the highest levels of 2,2'azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and α-α-diphenyl-β-picrylhydrazyl (DPPH) free radical scavenging activities. Reducing power was also the highest (39.21 mg Trolox equivalent/g) in E100 extract. The E100 extract effectively inhibited lipid peroxidation by 91.45% compared to the control group. Also, the E100 extract showed a cytoprotective effect against oxidative stress in HepG2 cells and decreased the generation of intracellular reactive oxygen species. These results suggest that bitter melon leaves could be regarded as a potential source of natural antioxidants.

        Baicalin is a flavonoid compound with many advantages, including anti-inflammatory agents and antioxidants. Lipopolysaccharide (LPS) is an endotoxin that induces neuronal damage through inflammatory response and oxidative stress reaction. This study was investigated the protective effects of baicalin on the oxidative stress and histopathological changes caused by LPS in hippocampus. Adult mice were divided into four groups; vehicle-treated, baicalin-treated, LPS-treated, and LPS and baicalin co-treated animals. Baicalin (10 mg/kg/day) and/or LPS (250 μg/kg/day) were intraperitoneally administered for seven consecutive days, and body weight was measured. Reactive oxygen species (ROS) level and lipid peroxidation level in the hippocampus were examined. Histopathological study was performed using hematoxylin and eosin staining manuals. LPS treatment decreased body weight and increased ROS and oxidative stress in the hippocampus. However, co-treatment with baicalin alleviated these changes caused by LPS. Severe histopathological changes were observed in the hippocampus of LPS-treated animals. Baicalin co-treatment attenuated the changes and preserved neuronal cells from LPS damage. These results showed that baicalin suppresses LPS-induced neuronal damage by alleviating oxidative stress in the hippocampus. Thus, this study demonstrated that baicalin exerts protective effects against LPS-induced oxidative stress in hippocampus.

        We propose a novel stripping solution containing acids (HCl and HNO3), an oxidant [(NH4)2S2O8], and complexing agents (NaCl and citric acid) to remove surface passivation layers from 14K gold alloys fabricated using an investment casting process. The optimized solution employing only HCl acid is determined by varying molar fractions of HCl and HNO3 on 14K yellow gold samples. Stripping properties are also identified for red and white gold alloy samples under the optimized stripping conditions. The removal of passivation layers, weight loss, and microstructure evolution are characterized using Raman spectroscopy, a precision scale, and optical microscopy. The proposed stripping solution effectively removes passivation layers more rapidly than conventional cyanide stripping. Weight loss increases linearly for up to 5 min for all 14K gold alloys. Red gold exhibits the greatest weight loss, followed by yellow gold and white gold. The results of microstructural analysis reveal that the conformal stripping occurs according to time. These results imply that the proposed oxidative chloride stripping might replace conventional cyanide stripping.

        Evodiae Fructus is the dried unripe fruit of Evodia rutaecarpa, and has traditionally been used for treating stomachache and diarrhea. Evodiamine and rutaecarpine, the major biologically active compounds of Evodiae Fructus, are reported to have anti-oxidative and anti-inflammatory effects, as well as inhibit proliferation and metastasis of various cancer cells. The current study investigates the anti-oxidative and anti-cancer effects of the Evodiae Fructus extract, considering varying concentrations of methanol extraction (40, 80, and 95%). High contents of total phenolic compounds were determined in the order of extracts 80, 95, and 40%. Evaluating contents of the 95, 80, and 40% extracts revealed 36.77, 7.29, and 1.86 μg/mg evodiamine, respectively, and 53.02, 17.16, and 3.79 μg/mg rutaecarpine, respectively, with the highest content of both compounds obtained in the 95% extract. DPPH radical scavenging activity was observed to be inversely proportional to the contents of total phenolic compounds, with decreasing SC50 values obtained in the order 80, 95, and 40% extract. The 95 and 80% extracts exerted toxicity to AGS gastric cancer cells, but the 40% extract was non-toxic. Evodiamine is a known anti-cancer agent, and could be responsible for the observed toxicity. Cleavage of PARP, and Caspase-3, -7, -8 and -9 was observed in the 95% extract-treated AGS cells, indicating that cell toxicity exerted by the 95% extract could be attributed to apoptosis.

        The purpose of this study was to investigate the protective effect on oxidative stress induced PC12 cells, and volatile flavor composition of essential oils derived from medicinal plant seeds- Gossypium hirsutum L. (G. hirsutum), Coix lachryma-jobi (C. lachryma-jobi) and Oenothera biennis (O. biennis). The essential oils were obtained by the solvent (hexane) extraction method from the seeds. The essential oils of the seeds were analyzed by the solid-phase micro-extraction gas chromatography mass spectrometry (SPME-GC/MS). The major compounds of G. hirsutum, C. lachryma-jobi and O. biennis were cyclonexanol (16.65%), β-asarone (14.29%) and ylangene (50.01%). The DPPH radical scavenging activity (IC50) was the highest value of 8.52 mg/mL in the O. biennis. Additionally, IC50 values of G. hirsutum and C. lachryma-jobi were 26.76 mg/mL and 36.81 mg/mL. For the oxidative stress on PC12 cells, we treated with hydrogen peroxide (H2O2). The pretreatment of oxidative stress induced PC12 cells with all the essential oils preserved or increased their cell viability and G. hirsutum and O. biennis attenuated the ROS generation (by 68.75% and 56.25% vs. H2O2 control). The results of this study suggest that the essential oils derived from medicinal plant seeds could be used as valuable back data as a natural essential oil material to prevent neurodegenerative diseases by protecting neuro-cells.

        본 연구에서는 감국 꽃 추출물의 항산화 및 항염활성을 검색하고 유효성분을 분리하여 화학구조를 규명하였다. 감국 꽃 에탄올 추출물의 DPPH 및 ABTS+ 라디칼 소거 활성을 측정한 결과 우수한 라디칼 소거 활성이 있음을 확인하였다. 또한 LPS로 유도된 RAW264.7 대식세포에서 세포독성 없이 우수한 nitric oxide (NO) 저해 활성을 나타내었다. 감국 꽃 에탄올 추출물의 유효성분을 찾고자 medium pressure liquid chromatography (MPLC)를 실시하여 2개의 화합물을 분리하였으며 1H 및 13C NMR 데이터 분석 및 문헌 비교를 통하여 화학구조를 동정하였다; Cynaroside (1), Apigetrin (2). 분리된 화합물을 HPLC를 통하여 정량분석을 수행하였으며 감국 꽃 추출물의 주요 성분으로 확인되었다. 화합물 1, 2의 항염활성을 확인한 결과 모두 농도의존적으로 우수한 NO 저해활성을 나타내었다. 이러한 연구 결과를 바탕으로 감국 꽃 추출물은 천연 항산화 및 항염 소재로써 활용 가능할 것으로 사료된다.
