        급격한 수온의 변화는 어류의 생리학적인 측면에서 스트레스를 유발한다. 본 연구에서는 넙치 (Paralichthys olivaceus)로부터 각 수온별(9, 12, 15, 18 및 21℃) 조건에 따라 24 및 48시간 동 안 노출시킨 후에, 혈액생리학적 분석, 스트레스 단백질로 알려진 Hsp70 mRNA 발현 및 산소 소비량을 조사하였다. 혈액학적 분석에서 hematocrit (Ht) 및 hemoglobin (Hb), 혈장 코티졸 및 글루코스의 변화, aspartate aminotransferase (AST) 및 alanine aminotransferase (ALT), NH3, 삼 투질농도(osmolality) 및 총단백질(total protein, TP)은 9℃ 및 12℃에서 다른 수온별 실험구에 비 해 대부분의 항목에서 유의적인 차이를 보였다. Hsp70 mRNA 발현은 9℃ 및 12℃에서 다른 실험구에 비해 높은 발현량을 확인하였고, 산소소비량은 9℃ 및 12℃에서 21℃에 비해 낮았다. 이러한 결과는 넙치 종자의 장거리 수송을 위한 수온자료로 활용할 수 있다

        We determined the effects of different water temperatures (15, 20, and 25℃) and photoperiod cycles (24L:0D, 12L:12D, and 0L:24D) on the oxygen consumption of the offspring of a cultured Japanese strain (JJ), a selected Korean strain (KK), and intraspecific hybrid strains (JK and KJ) of red seabream, Pagrus major, under starvation conditions. The different fish strains, water temperatures, and photoperiod cycles had effects on the mean oxygen consumption of fish. Oxygen consumption increased with increasing water temperatures for all photoperiod treatments (p <0.001). Fish held in continuous darkness (0L:24D) used consistently less oxygen than fish exposed to continuous light (p <0.05). The oxygen consumption of fish exposed to the light phase in a 12L:12D photoperiod was higher than that of fish in the dark phase of the 12L:12D cycle, and differences were significant in three of the strains: JJ (15℃), KK (15 and 20℃), and KJ (25℃). The oxygen consumption of the inbred (JJ and KK) and intraspecific hybrid (JK and KJ) strains varied with differing water temperatures and photoperiod cycles. The JK strain displayed significantly higher oxygen consumption than the other strains under all experimental conditions except 15℃ with a 0L:24D photoperiod. The JK and KJ strains usually showed the highest and lowest oxygen consumption values, respectively, whereas the inbred strains exhibited intermediate values. Oxygen consumption in the JJ and JK strains was usually higher than that of the KK and KJ strains. We propose that differences in the thermal sensitivity and photosensitization properties of the strains contribute to differences in their ability to adapt to changes in water temperature and photoperiod, thus resulting in differences in the amplitude of their metabolic rates.

        Recently, several approaches have been used to measure the oxygen consumption rates of individual embryos, but relationship between oxygen consumption and pregnancy rates of Hanwoo following embryo transfer has not yet been reported. In this study, we investigated the correlation between oxygen consumption rate and pregnancy rates of Hanwoo embryo using a SECM. In addition to, the expression of apoptosis-related genes was determined using real-time PCR by extracting RNA according to the oxygen consumption of in vivo embryo. First, we found that the oxygen consumption significantly increased in blastocyst-stage embryos (blastocyst) compared to early blastocyst stage embryos, indicating that oxygen consumption reflects the embryo quality (Grade I). The oxygen consumption or GI blastocysts were significantly higher than those of GII blastocysts (10.2x1014/mol s-1 versus 6.4x1014/mol s-1, p<0.05). Pregnant rate in recipient cow was 0, 60 and 80% in the transplantation of embryo with the oxygen consumption of below 10.0, 10.0~12.0 and over 12.0x1014/mol s-1, respectively. Apoptosis regulatory genes, Hsp-70.1 were significantly increased in over-10.0 group than below 10.0 group but in Caspase-3, Bax and P53 gene, there was no significant difference. In conclusion, These results suggest that measurement of oxygen consumption maybe help increase the pregnant rate of Hanwoo embryos.

        Oxygen consumption is a useful parameter for evaluating mammalian embryo quality, since individual bovine embryos was noninvasively quantified by scanning electrochemical microscopy (SECM). Recently, several approaches have been used to measure the oxygen consumption rates of individual embryos, but relationship between oxygen consumption and pregnancy rates of Hanwoo following embryo transfer has not yet been reported. In this study, we measured to investigate the correlation between oxygen consumption rate and pregnancy rates of Hanwoo embryo using a SECM. In addition to, the expression of pluripotent gene and anti-oxidant enzyme was determined using real-time PCR by extracting RNA according to the oxygen consumption of in vivo embryo. First, we found that the oxygen consumption significantly increased in blastocyst-stage embryos (blastocyst) compared to early blastocyst stage embryos, indicating that oxygen consumption reflects the embryo quality (Grade I). Oxygen consumption of blastocyst was measured using a SECM and total cell number of in vitro blastocyst was enumerated by counting cells stained by propidium iodide. The oxygen consumption or GI blastocysts were significantly higher than those of GII blastocysts (10.2 × 1015/mols—1 versus 6.4 × 1015/mols—1, p<0.05). Total cell numbers of in vitro blastocysts were 74.8, 90.7 and 110.2 in the oxygen consumption of below 10.0, 10.0∼12.0 and over 12.0∼1015/mols—1, respectively. Pregnant rate in recipient cow was 0, 60 and 80% in the transplantation of embryo with the oxygen consumption of below 10.0, 10.0∼12.0 and over 12.0 × 1015/mols—1, respectively. GPX1 and SOD1 were significantly increased in over —10.0 group than below 10.0 groups but in catalase gene, there was no significant difference. On the other hand, In OCT-4 and Sox2, pluripotent gene, there was a significant difference (p<0.05) between the below-10.0 (0.98 ± 0.1) and over 10.0 (1.79 ± 0.2). In conclusion, these results suggest that measurement of oxygen consumption maybe help increase the pregnant rate of Hanwoo embryos.

        Yellow clay dispersion has been applied to minimize fisheries impact by the red tide Cochlodinium polykrikoides blooms in Korean coasts since 1995. The present preliminary study documents the effect of yellow clay on Korean rockfish, Sebastes schlegelii, in terms of oxygen consumption rate (OCR). The OCR in the low clay suspension (0.05 and 0.23 %, w/w) showed normal level compared to the control. In contrast, the OCR for each one of three replicates in the high clay suspension (1.16 and 5.58 %, w/w) was not returned to the previous level that clay was not treated, indicating that high clay suspension (≥1.16%, w/w) might give negative effect on Korean rockfish. Overall, this result suggests that field application of clay to control Harmful Algal Blooms (HABs) may not give impact on Korean rockfish once the clay is dispersed in a low concentration (≤0.23%). In order to understand the changes of OCR in the repeated exposure to clay, it is required to do further studies on the changes of OCR when the fish is exposed to clay repeatedly after recovery in the normal seawater.

        To evaluate the biological response of sea squirt Halocynthia roretzi with different body size to heavy metals and its suitability for ecotoxicity assays, the effects of Cr, Cu and Zn on its clearance and oxygen consumption rates were investigated. Clearance and oxygen consumption rates of H. roretzi with various body sizes were calculated at different metal concentrations. Both clearance and oxygen consumption rate were negatively correlated with body sizes. Clearance rate of H. roretzi decreased gradually with increasing concentration of heavy metal, the decreasing rate was in an order of Cr¤Cu¤Zn. The oxygen consumption rate first increased at low metal concentration (below 100 μg L-1) and then decreased rapidly with increasing metal concentrations. The decreasing rate was in an order of Cu¤Cr=Zn. There was a trend that the clearance rate and oxygen consumption rate decreased drastically under a concentration of 400 μg L-1, and then decreased smoothly when the metal ion concentration increased continually. So the oxygen consumption and clearance rate at a concentration of 400 μg L-1 Cu could be thought as a suitable biological tool for exotoxicology analysis.

        Oxygen consumption has been regarded as a useful indicator for assessment of mammalian embryo quality. This study was performed to investigate whether oxygen consumption reflects morphological grade of in vivo derived bovine blastocyst-stage embryos (blastocyst). The oxygen consumption of in vitro produced blastocyst was compared to its total cell number. In addition, pregnant rate was measured after transplantation of in vivo blastocysts with different oxygen consumption. The quality of blastocyst collected on day 7 after artificial insemination was categorized as grade I and II (G I and G II) based on microscopic observation of the morphology. Oxygen consumption of blastocyst was measured using a scanning electrochemical microscopy (SECM) and total cell number of in vitro blastocyst was enumerated by counting cells stained by propidium iodide. Pregnancy of recipient cow was confirmed with rectal palpation after 60 days of embryo transfer. The oxygen consumptions of G I blastocysts were significantly higher than those of G II blastocysts ( versus , p<0.05). Total cell numbers of in vitro blastocysts were 74.8, 90.7, and 110.2 in the oxygen consumption of below 10.0, 10.0~12.0, and over respectively. Total cell number was significantly increased in embryos with high oxygen consumption (p<0.05). Pregnant rate in recipient cow was 0, 50, and 85.7% in the transplantation of embryo with the oxygen consumption of below 10.0, 10.0~12.0, and over , respectively. These results suggest that measurement of oxygen consumption may help increase the pregnant rate of bovine embryos.

        Oxygen consumption has been regarded as a useful indicator for assessment of mammalian embryo quality. However, there was no standard criterion to measure the oxygen consumption of embryos. Here, we measured oxygen consumption of bovine embryos at various developmental stages was measured using a scanning electrochemical microscopy (SECM). We found that the oxygen consumption significantly increased in blastocyst-stage embryos compared to other stage embryos (from 2-cell-stage to morula-stage), indicating that oxygen consumption reflects the cell number ( versus , p<0.05). In the morula-stage embryos, the oxygen consumption of in vivo derived embryos was significantly higher than that of in vitro produced embryos ( versus , p<0.05). However, there was no significant difference in consumption of oxygen by in vivo and in vitro-derived bovine blastocyst-stage embryos (p>0.05). In the frozen-thawed blastocyst-stage embryos, live embryos showed significantly higher oxygen consumption than dead embryos ( versus , p<0.05). These results indicate that the measuring oxygen consumption by SECM can be used to evaluate bovine embryo quality.

        The purpose of this study was to establish modified physiological cost index (PCI) for predicting energy consumption by heart rate (HR) at isokinetic ergometer exercise testing. The subjects were twenty-eight healthy men in their twenties. All of them performed upper and lower extremity isokinetic ergometer exercise tests which had six loads (400, 500, 600, 700, 800, and 900 kg-m/min) and five loads (400, 500, 600, 700, and 800 kg-m/min) respectively. The exercise sessions were finished when HR was in plateau. HR and oxygen consumption were determined during the final minute. Resting heart rate and oxygen consumption were used for calculating heart rate, oxygen consumption changes and modified PCI. Regression analysis established the relationship between each variable to work load, HR and oxygen consumption. The results were as follows: 1) In the lower extremity ergometer exercise test, oxygen consumption increased continuously as work load increased, but in the upper extremity ergometer test, oxygen consumption only increased until work load was 700 kg-m/min. 2) HR increased as work load increased in both exercise tests, but in the upper extremity ergometer test, HR decreased from the 700 kg-m/min. 3) The modified PCI increased as work load mcreased until the 700 kg-m/min point in the lower extremity ergometer test and until the 500 kg-m/min point in the upper extremity ergometer test when it started to decrease in both tests. 4) In the lower extremity ergometer exercise test, regression analysis established the relation as = -.0215HR - .2141 where is given in l/min and HR in beat/min ( = .2677, p = .000). ln the upper extremity ergometer exercise test. regression analysis established the relation as = -.0115HR + .2746 ( = .1308, p = .000). The results of this study were similar to previous studies but were different under high work load conditions. So modified PCI should be used with only low intensity work load testing. Subjects for upper extremity ergometer exercise testing should complete a prescribed training course prior to testing, and only low intensity work load should be used for safety considerations.

        We investigated the effects of temperature changes on the oxygen consumption rhythm in Japanese eels, Anguilla japonica, using an automatic intermittent flow respirometer (AIFR). The endogenous rhythm of the oxygen consumption rate (OCR) in the eels (n = 18; 44-74 cm, 145-690 g), freshly collected by bag net from estuaries, was nearly synchronous with the tidal pattern of the estuarine collection site. The magnitude of mean OCR (mOCR) of eels showed variable range of 82.2 - 116.5 ml O2 kg-1 ww h-1 under constant conditions. In case of increasing temperature from 25 to 38℃, the OCR of eels exhibited a gradually increasing trend with a rhythmic pattern until 36℃. Above 36℃, the rhythms of the OCR dampened and the OCR decreased rapidly at around 36 - 37℃. The OCR of the eels exhibited the maximum value at 38℃, and then it sharply decreased. The results suggested that the critical thermal maximum (CTM) regarding the endogenous rhythms of the eels was at around 36 - 37℃ when water temperature increased at 0.5℃/14 h following the acclimation at 25℃. In case of decreasing temperature (0.5℃/14 h) from 25 to 0℃, the OCR of the eels displayed a abrupt decrease up to 23℃, and between at 23 and 20℃, there was an agitation which showed a slight increase in the OCR with a duration of 1-2 days. Below 9℃, the OCR rhythm of the eels showed a constant state regardless of temperature decreasing. These results suggest that the Japanese eel has an upper incipient lethal temperature at 36℃, with a lower thermal limit at 9℃. The biochemical aspects of the eels influenced by water temperature need to be further studied.

        Eco-physiological research and the control of Cochlodinium polykrikoides was carried out to elucidate eco-physiological characteristics of red tide organism through culture experiment depending on the condition of photon irradiance. Oxygen consumption of C. polykrikoides was high with a value of 1.12mg/L/hr in the dark compared with that of 0.13mg/L/hr at 100μEm-2s-1. DO values in a circular chamber with the lapse of time in seawater containing C. polykrikoides were declined in the dark period. DO values of seawater containing C. polykrikoides in the dark were declined from 7.01mg/L to 2.65mg/L in 30cm depth and from 7.01mg/L to 6.63mg/L in 5cm depth depending on the depth of circular culture vessel. Olive flounder, Paralichthys olivaceus and file fish, Stephanolepis cirrhifer exposed to Cochlodinium showed the separation of the lamella epithelium from gill filament, which disrupted the respiratory process at the gill level.
