        Brachymystax lenok tsinlingensis (family Salmonidae), cold freshwater fish, is endemic to Asia. This species is currently distributed throughout Russia, Mongolia, China and the Korean Peninsula. B. lenok tsinlingensis in South Korea was severely affected by anthropogenic activities such as habitat destruction, agricultural run-off and water pollution, and hence this fish has recently been dramatically decreased in its population sizes and become now critically endangered. To recover the number of individuals of B. lenok tsinlingensis, stocking or translocation programs have been conducted continuously by local governments since 1970s. However, these programs made little effort to clarify populations that may have originated from stocked, translocated or introduced fish. An understanding of genetic characteristics of endangered populations is critical to develop effective conservation and restoration plans especially because genetic diversity ensues their future fate. Therefore, we assessed the “conservation status” of this species by estimating the level of genetic diversity and genetic structure among ten geographic populations including restored populations via reinforcement and supplementation. Also, we aimed to trace the genetic origins of the newly translocated population (Chiak) through a restoration practice program. Moreover, we inferred the phylogenetic relationships among Korean lenok populations as well as across the Northeast Asia. Two hundred eighteen individuals of B. lenok tsinlingensis were sampled from ten localities (Yanggu, Injae, Seorak, Bangtae and Hongcheon: North Han River basin; Pyeongchang, Chiak and Jeongseon: South Han River basin; Taebaek and Bonghwa: Nakdong River basin in South Korea). Based on mitochondrial DNA (mtDNA) control region and eight nuclear microsatellite loci, we found extremely low levels of within-population genetic diversity, which suggests small effective population sizes (Ne) within populations. For mtDNA control region, each population housed one, or at most, two haplotypes that are restricted to the respective localities, meaning that these ‘genetically unique’ lineages will be lost permanently if the local populations undergo extinction. The overall values of haplotype diversity (h) and nucleotide diversity (π) for the entire Korean population were 0.703 ± 0.024 and 0.021 ± 0.010, respectively. In the case of microsatellites, average number of alleles across the eight loci for the entire population was 9.1 and allelic richness (AR) per population ranged from 2.375 to 4.144 (mean = 3.104). The values of observed heterozygosity (HO) and expected heterozygosity (HE) were similar to each other [HO: 0.400 ~ 0.590 (mean = 0.518); HE: 0.407 ~ 0.608 (mean = 0.504)]. The inbreeding coefficient (FIS) values were generally low, ranging from 0.048 to 0.279. Consequently, the majority of the populations (except Yanggu and Pyeongchang) were not significantly deviated from Hardy-Weinberg equilibrium (HWE), suggesting random mating at these loci tested. In addition, we found that Korean lenok populations were significantly genetically isolated from each other, with private mtDNA haplotypes and microsatellite alleles, indicating limited gene flow among populations, strong effects of genetic drift due to small Ne, or a combination of both. The Mantel test of microsatellites revealed a significant correlation (r = 0.414, P = 0.04) between genetic and geographic distances for pairwise comparisons among the ten populations, while that of mtDNA showed a lack of correlation. Given the shared identical mtDNA haplotype and similar microsatellite allelic distributions between Chiak and Hongcheon populations, we suggest that the restored (introduced) Chiak population would be inferred to be genetically originated from Hongcheon population. Phylogenetic relationships among Northeast Asian populations showed that South Korean lineages have more recently diverged from China (Yellow River), than between North Korea and Russia. Although the phylogenetic relationship would be expected to be associated with geography, South-North Korea and China populations with a similar latitude was more phylogenetically closely related. These findings may suggest a possible scenario for the historical movements of B. lenok tsinlingensis in Northeast Asia during Last Glacial Maximum (LGM). It would be supported by the line of evidence that most lenok populations migrated to southward from Northern Asia such as Russia and Mongolia during LGM because the Korean Peninsula was landlocked as inland epoch and functioned as a southern shelter with Yellow River. For this reason, the Korean Peninsula is suggested to be an important geographical region for better understanding phylogenetic relationships and evolutionary histories of B. lenok tsinlingensis across the Northeast Asia. Despite large efforts made to develop several restoration programs in South Korea for B. lenok tsinlingensis, it is still unknown whether these past restoration efforts were successful or fruitless, mainly because of little attention paid to post-restoration monitoring research. Hence, there was a lack of their published official records. In the future, conservation and restoration projects of the Korean lenok populations should consider the genetic data for a better understanding of their ecological and evolutionary trajectories. And finally, we hope that our findings here can help inform on the future effective conservation and restoration plans for B. lenok tsinlingensis populatio ns in South Korea.

        Seagrasses, sea flowering plants, comprise approximately 60 species globally and are often called ‘ecosystem engineers’ because they create their own habitats by modifying the surrounding environments, which provide coastal zones with a number of crucial ecosystem services. Zostera marina (the common name ‘eelgrass’) is one of the seagrass beds-forming species distributed widely in northern hemisphere including the Korean coast, which plays a pivotal role in ecosystem as a primary producer and a nursery habitat or refuge for other marine organisms. However, due to global climate change and anthropogenic activities such as reclamation and dredging, there has recently been a drastic decline in population sizes of Z. marina in Korea. In order to develop effective conservation and restoration management programs of Z. marina populations, it would be helpful to consider all biological aspects of this species such as genetic characteristics as well as ecological and physiological features. This study first provides information on genetic diversity and genetic structure of Jeju Island and Namhae populations of Z. marina, which will contribute to the establishment of appropriate conservation and restoration management plans for future persistence of this species. Using six microsatellite markers, we investigated the level of genetic diversity and genetic structure among 10 geographic populations of Z. marina inhabiting Jeju Island (Hamdeok, Tokki-seom, Sungsan, Woljeong, Ojo) and Namhae (Gamak bay, Jindong bay, Nampo, Anggang bay, Geoje) on the southern coast of Korea. The level of genetic diversity within Jeju populations (mean allelic richness [AR]: 1.57 ~ 3.09) was found to be significantly lower than Namhae populations (AR: 3.09 ~ 4.29) (Mann-Whitney U-test, P < 0.05). These findings suggest that effective population sizes (Ne) of Jeju populations are generally smaller than those of Namehae populations. Within Jeju Island, Hamdeok population had the smallest population size (coverage: 138 m2) and the lowest genetic diversity (AR: 1.57), while Ojo population had the largest population size (coverage: 275,736 m2) and the greatest level of genetic diversity (AR: 3.09). Hamdeok population showed evidence of genetic bottleneck. These results again suggest that Ne of Jeju populations is generally low (except Ojo population). Among Jeju populations, all pair-wise comparisons of FST values (i.e., degree of genetic differentiation) were highly significant (FST = 0.0612 ~ 0.7168, P < 0.001) despite Jeju populations that were geographically closely located, indicating that these local populations are genetically divergent, probably due to a lack of gene flow among the populations. The observed strong population structure was substantiated by evidence that five genetic clusters are most likely, based on population assignment test (STRUCTURE). The Mantel test showed a positive relationship between genetic distance (FST) and geographic distance (km) across all the populations sampled (R2 = 0.4118, P < 0.05), suggesting that our data follow Isolation By Distance (IBD) model. Woljeong population revealed the highest level of FST values compared to other populations within Jeju Island in IBD. STRUCTURE and factorial correspondence analysis (FCA) further showed that some Woljeong individuals included genotypes of Namhae populations. Population size of Woljeong (coverage: 310m2) was approximately 50 % smaller than that of Sungsan (coverage: 841m2); however, extent of its genetic diversity (AR: 2.39) was even higher than that of Sungsan population (AR: 1.77). We speculated that Woljeong population underwent a transplantation from Namhae populations with relatively higher level of genetic diversity. FST values within Namhae populations were relatively lower (compared to within Jeju Island) despite the populations that were geographically more distant. It means that level of gene flow is higher among Namhae populations than among Jeju populations. Z. marina is known to have different life histories by water depth. In subtidal zone (deep water depth) populations predominantly undertake sexual reproduction through seeds such as annual life history, whereas those of intertidal zone (shallow water depth) undertake both sexual and asexual reproductions through horizontal rhizomes i.e., perennial life history. STRUCTURE analysis showed no clear differences between shallow and deep populations at Namhae, but some FST values were statistically significantly different despite their low values. For Geoje population sampled in 2005, intertidal and subtidal populations were not significantly different (FST = 0.0045, P = 0.033), but these populations sampled in 2015 showed a significant difference (FST = 0.0328, P < 0.001). It means that genetic structure of Geoje has been changed over the 10 year period between shallow and deep populations. Overall, the Jeju and Namehae populations analyzed in the current study have relatively low levels of genetic diversity and distinct genetic compositions, which warns the message that this ecologically important species should be conserved separately in the local populations and with high priority. We propose that future conservation and restoration plans for seagrasses should consider genetic characteristics particularly because a close relationship between genetic diversity and ecological performance in marine species has been well documented.

        There is much observational evidence that active star formation is taking place in the Hii regions Sh 2-255 – 257. We present a photometric study of this star forming region (SFR) using imaging data obtained in passbands from the optical to the mid-infrared, in order to study the star formation process. A total of 218 members were identified using various selection criteria based on their observational properties. The SFR is reddened by at least E(B −V ) = 0.8 mag, and the reddening law toward the region is normal (RV = 3.1). From the zero-age main sequence fitting method it is confirmed that the SFR is 2.1 ± 0.3 kpc from the Sun. The median age of the identified members is estimated to be about 1.3 Myr from a comparison of the Hertzsprung-Russell diagram (HRD) with stellar evolutionary models. The initial mass function (IMF) is derived from the HRD and the near-infrared (J, J −H) color-magnitude diagram. The slope of the IMF is about 􀀀 = −1.6 ± 0.1, which is slightly steeper than that of the Salpeter/Kroupa IMF. It implies that low-mass star formation is dominant in the SFR. The sum of the masses of all the identified members provides the lower limit of the cluster mass (169M⊙). We also analyzed the spectral energy distribution (SED) of pre-main sequence stars using the SED fitting tool of Robitaille et al., and confirm that there is a significant discrepancy between stellar mass and age obtained from two different methods based on the SED fitting tool and the HRD.

        Insect growth regulators (IGRs) are compounds that induce developmental anomalies such as premature molting or supernumerary larval stages. IGRs can be divided into 3 groups based on their mode of action, one of which is known as the juvenile hormone agonists (JHAs). Previously, we have developed a novel screening method which can identify juvenile hormone antagonists (JHANs) using a yeast-two hybrid system incorporating Aedes aegypti juvenile hormone receptor complex proteins (MET and FISC). It has been reported that plants may use IGRs in their defenses against insect herbivores. In this study, 171 plant essential oil samples were screened using the yeast-two hybrid system in order to identify JHAs and/or JHANs. Eight out of 171 samples, 4 JHA and 4 JHAN candidates, were selected and their insecticidal activities were investigated against A. aegypti larvae. They showed high levels of insecticidal activities, with LC50 values ranging from 11.2~21.4 ug/ml. These results indicate that plants do use JHAs and/or JHANs as a part of their defenses against insect herbivores. Also, JHAs and JHANs identified in this study could be exploited as effective biological control agents.

        Insect growth regulators (IGRs) are insecticides that disrupt the normal development of target insects by inducing symptoms such as premature molting or supernumerary larval stages. IGRs are categorized into three types based on their mode of action. One of the them is classified as juvenile hormone agonists (JHAs). Recently, we developed a yeast-two hybrid system that can identify potential juvenile hormone antagonist (JHAN) candidates using Aedes aegypti juvenile hormone receptor complex proteins, known as MET and FISC. In order to identify novel JHAN compounds, approximately 8,700 basic chemical compounds were screened using the yeast-two hybrid system. Among them, 35 compounds were first selected based on consistency of JHAN activity, and their insecticidal activities were investigated against A. aegypti larvae. These compounds showed insecticidal activities about 1.7~3 times higher than that of pyriproxifen, which is a well known JHA insecticide. These results suggested that these JHANs can be used as effective JHAN insecticides.

        Insect Growth Regulators (IGRs) are insecticides that disrupt the normal development of target insects by inducing symptoms such as premature molting or supernumerary larval stages. Juvenile hormone systems become the targets of two types of IGRs: the Juvenile Hormone Agonists (JHAs) and Juvenile Hormone Antagonists (JHANs). Pyriproxyfen is one of the chemical compounds widely used as JHA to control many kinds of insects while Kanakugiol is a plant-extracted compound which acts as JHAN. The small brown planthopper, Laodelphax striatellus, is one of the most serious pest insects of rice plants because it can transmit the rice stripe virus which often causes significant reduction of yield in the field. In order to analyze the differential gene expressions of L. striatellus upon JHA and JHAN treatment by using next generation sequencing technique, we sprayed Pyriproxyfen and Kanakugiol on 4th instar nymphs of L. striatellus respectively, and extracted total RNA for RNA-seq. The quality-filtered Illumina sequence reads of the control, JHA, and JHAN treated samples were mapped to the reference gene sequences by using the Bowtie2 software. Then the results of mapping by Bowtie2 were analyzed by eXpress software to quantity the differential gene expression.

        Recently, Bacillus thuringiensis (Bt) cry genes encoding insecticidal Cry proteins have been widely applied for the construction of transgenic crops resistant to insect pests. This study aimed to construct novel mutant cry1Ac genes for genetically modified crops with enhanced insecticidal activities. Using multi-site directed mutagenesis, 34 mutant cry1Ac genes were synthesized and converted at 24 amino acid residues, located on domain I (8 residues) and domain II (16 residues). These mutant genes were expressed as a fusion protein with polyhedrin using the baculovirus expression system. The expressed proteins were occluded into polyhedra and activated stably to 65 kDa by trypsin. Among these, Mut-N04, N06, and N16 showed high levels of insecticidal activites against larvae of Plutella xylostella, Spodoptera exigua, and Ostrinia furnacalis. Mut-N16, which showed the highest insecticidal activity, is expected to be a desirable cry gene for introduction into transgenic crops. This study could provide useful means to construct mutant cry genes with improved insecticidal activities and expanded host spectrum for transgenic crops.

        미이라병은 Diaporthe/Phomopsis complex에 의해 유발되는병으로 콩 재배기간 중 따뜻하고 습한 환경에서 종자가 성숙되면 감염률이 높아지며 감염된 콩 종자는 외관상 품질뿐만아니라 종자 활력이 저하된다. 미이라병에 대한 연구를 수행하기 위하여 대만에 위치한 아시아채소개발연구센터(AVRDC)의 콩 시험포장에서 미이라병 병징을 보이는 콩 줄기를 채집하고 이로부터 3개의 곰팡이 균주(isolate)를 분리하였다. 배지위에서의 곰팡이 균사의 생육특성, 현미경하에서 관찰된 알파,베타 분생자(conidia)의 모양 그리고 PCR-RFLP 분석으로, 3개의 균주는 Diaporthe phaseolorum var. sojae 으로 확인되었다. 한편, 미이라병 저항성 육종을 위해서는 유전자원과 계통의 검정이 선행되어야 하는데, 인공접종을 위해서 분생자의최적 배양조건을 탐색하였다. 그 결과 배지는 PDA, 온도는24oC에서 잘 배양되었으며, 일장은 암조건에서는 균사체만 유도되고 분생자는 유도되지 않았으며, 24시간과 15시간의 일장에서는 균사체 유도 및 분생자의 유도 정도에 차이가 없었다.또한 잎-줄기와 꼬투리, 두 개의 접종 부위에 따른 미이라병감염률을 조사하였는데, 두 접종 부위에 따른 미이라병 감염정도는 통계적인 유의차는 나타나지 않았으나 잎-줄기에 접종한 개체 보다 꼬투리에 접종한 개체의 종자 감염률이 높은 경향을 보였다.

        Multiple starters consisting of two Bacillus amyloliquefaciens strains (MJ1-4 and EMD17), Pichiafarinosa SY80, and Rhizopus oryzae were used for Doenjang making. Bacillus strains were selected based on their abilities to inhibit toxinogenic fungi and Bacillus cereus, fibrinolytic activity, and their ability to confer good flavor to Cheonggukjang. P. farinosa SY80 and R. oryzae, previously isolated from soy sauce, were selected because they were not inhibited by two bacilli. Doenjang was prepared by inoculation of multiple starters (A1 Doenjang). Control Doenjang was prepared by inoculation of B. subtilis KACC 16750 (Natto strain) and Aspergillus oryzae KCCM 60166 (A2 Doenjang). Another control (A3 Doenjang) was prepared by inoculation of microorganisms present in rice straw. Doenjang samples were fermented for 70 days at 20℃. pH of 3 samples decreased from the initial value of 6.4 to 5.8~6.0 and titratable acidity (TA) increased from 0.6 to 1.1~1.3. The amount of amino-type nitrogen increased during fermentation. There were slight differences in moisture, crude-protein, and crude-fat contents after 70 days. Contamination of fungi was observed only in A3 Doenjang and B. cereus was not detected from all 3 samples. A1 Doenjang showed the highest fibrinolytic activity and A2 Doenjang the second. These results indicate that Doenjang made with carefully selected starters was functionally improved and microbially more safe.

        This study investigated the effects of changes to the pulsation factor of pulsed direct currents on wound healing. Patients with a pressure ulcer at a care hospital for the elderly were randomly divided into three groups: Group 1 involved the application of 100㎲ in pulse duration, 10 ㎳ in pulse period, 100 pps in a pulsation factor, 15 ㎃ in pulse amplitude, and polarity red+ by using pulsed direct currents; Group 2 involved a change of pulse period to 8 ms; and Group 3 received general wound management. Although there were no statistically significant differences in the changing stages of pressure ulcers among the groups, all the groups dropped in numerical stages. In the two groups to which pulsed direct currents were applied, there was a statistically significant reduction in the stages of pressure ulcers from the initial assessment to the 12-week assessment (p<.05). Even though there were no statistically significant differences in changes to the area of pressure ulcers among the groups, a statistically significant decrease was found in pulsed direct current group 2 whose pulse period was shortened (p<.05). There was no difference in the healing rate of pressure ulcers among the groups, but it made a numerical increase in pulsed direct current group 1 and group 2 and a numerical decrease in group 3. There were no significant differences in the characteristics of those who had a full recovery among the groups. Those findings indicate that pulsed direct currents have positive effects on the wound healing of patients with a pressure ulcer and that a treatment with pulsed direct currents whose pulsation factor is raised by reducing the pulse duration is especially effective.

        We define hydrogel as a polymer network containing a large amount of water or biological fluid in a 3-D structure. Because of the physical or chemical chains present in a hydrogel, it is stable in aqueous environment. Therefore, it has been used in diverse medical fields. In addition, by controlling the gelation degree of polymer solution (the state prior to hydrogel) hydrogels can be easily applied to damaged tissue area. This unique structure and properties of hydrogel shares a similarity with ECM (Extracellular matrix) in that it has a potential to be applied in tissue engineering field. Especially, the injectable property and ECM like structure can be applied to bone regeneration. Out of several polymers can be form hydrogels, silk fibroin (SF) has an excellent biocompatibility, biodegradability and it can be used to create bone regeneration scaffold in the form of hydrogel. In this study, we fabricated a SF hydrogel containing hydroxyapatite nanoparticle (HAp). To improve the dispersibility of HAp in the SF aqueous solution, we chemically modified the surface of HAp particles using hyaluronic acid (HA) – dopamine (DA) conjugate. Since SF aqueous solution has a long term gelation time, we utilized ultra-sonication method to induce a rapid gelation. Stability of HAp in SF aqueous solution was measured by ELS and TGA. Finally, FT-IR and WAXD were used to evaluate the changes of secondary structure of silk hydrogel according to concentrations of hydroxyapatite nanoparticle concentration.

        This study was conducted to investigate the effect of biological membrane transfer modifier, lysophospholipd (LPLs) on the parameters from in vitro rumen simulated fermentation. Commercially available LPLs product (LipidolTM) was supplemented into experimental diets which consisted of orchard grass and concentrate diet (60:40) in different levels (0.1%, 0.3% and 0.5%). Then in vitro rumen simulated fermentation was performed. Although, a declining trend of pH was found in treatments, all pH values were detected in a range relevant to normal rumen fermentation. Gas production, ammonia nitrogen and total VFA production were greatly influenced by the supplementation of LPLs. All parameters were increased along with increased levels of LPLs in diet. As a result, 0.1% of LipidolTM is recommended based on the determined in vitro rumen fermentative parameters in this study.

        The green pale plant bug, Apolygus spinolae was one of the main insect pests that damaged leaves and fruit in grapes and its damage status was firstly reported in 2000 in grape orchards. This research was conducted to evaluate the distribution and difference in damage rate depending in management type of grapevine orchards (domestic sale farm vs export farm) in the export complex area of Korea (Hwangsung in Gyeonggii, Sangju and Yeongcheon in Gyeongbuk, Namwon in Junbuk and Yeongdong in Chungbuk) from 2010 to 2012. Damage by A. spinolae occurred in all 62 survey farms and damage rate differed depending on locality and individual farms in the same area. Damage rate was lower in export farms than in domestic sale farms, and damage rate of leaves was highly correlated with damage rate of new shoots. 15 species of hemipteran insect were attracted to sticky traps and A. spinolae was the dominant species. The attracted number of A. spinolae in the sticky traps differed depending on locality, and more occurred in domestic sale farms than expert farms. A. spinolae was continually attracted to sticky traps in the harvest period in grapevine orchards.

        Artificial insemination (AI) has been performed widely in swine industry using fresh liquid sperm instead of frozen type of sperm. However fresh sperm are not able to preserve more than three days with optimal motility and other sperm parameters for the successful fertilization, since in vitro stored sperm has an oxidative stress that resulted increase of abnormality and acrosome reation. To overcome these major problems, novel preservative formulation is needed to neutralize the oxidative stress and to provide suitable physiological environment for sperm in in vitro. In this study, naturally derived substances such as Poncirus trifoliate (Trifoliate orange), Garcinia mangostana (Mangosteen), pig placenta and testis extracts were tested as sperm preservative agents. Placenta extracts (PE), trifoliate orange extracts (TOE), testes extracts (TE) and mangosteen extracts (ME) were applied to analyze specific parameters for sperm motion characteristics individually and combinatorial. Each individual extract treatment can accelerate the sperm motility but noticeably TOE, TE and ME treatments exhibited the considerable and significant preservation of sperm motility. PE, TE and ME showed a significant (p<0.05) increase in ALH after one week. Further we evaluated the five different combinations of these extracts on sperm motility and its motion characteristics. Surprisingly even after one week ME, TOE and TE combination significantly preserved the sperm motility about 75%. It is noteworthy that unlike individual extract treatment, combination of ME, TOE and TE simultaneously protect the sperm motility and its motion characteristics. Taken together these data conclude that addition of ME, TOE and TE can be effective for preservation of pig sperm.

        본 연구는 베트남 하노이 지역에서 8가지 한국 토마토 품종 (핑크탑, 큐피랑, 슈퍼도태랑, 선레드, 선글러브, TP-7 플러스, 러블리 250, 광복)과 1가지 베트남 품종(FM 120)을 대상으로 봄-여름 및 가을-겨울 작기에 따른 수확량 및 과실 특성을 비교 분석하였으며, 그 결과는 다음과 같다. 1. 작기에 따라 초장, 경장, 주당 과실수 및 수확량(kg)은 유의성이 있었으며 가을-겨울 작기에서 높게 나타났다. 전체적으로 한국품종이 베트남 품종에 비해 높은 생육특성을 보인 반면, 과실수와 수확량에서는 베트남 품종이 우수하였다. 2. 토마토 과실의 주요 특성인 과장, 과폭 및 과중은 작기에 따라 유의한 차이를 보였으며 가을-겨울 작기에서 높은 수치를 나타내었지만 TSS 함량은 봄-여름 작기에서 높았다. 건물중과 비타민C 함량은 작기에 따라 유의한 차이가 없었다. 품종별 과실 특성은 한국 품종이 우수하였지만 비타민C 함량은 베트남 품종이 높았다. 3. 봄-여름 작기에서 한국품종의 경우 수확량 및 과실특성에서 가을-겨울 작기 재배에 비해 큰 감소를 보인 반면 베트남 품종은 큰 차이를 보이지 않았다. 따라서 본 실험에 이용된 8가지 한국 품종의 경우 봄-여름 작기보다는 가을-겨울 작기 재배가 적합할 것으로 판단된다.

        True hermaphrodites are animals of equivocal sex in which both male and female gonads develop simultaneously. The frequency of true hermaphroditism is higher in pigs than in other domestic animals. Two Korean pigs were diagnosed with true hermaphroditism showing ovotestes, epididymes, penes, and uteri. Histomorphologically, the testicular tissues consisted of Sertoli cells that were devoid of spermatogenic germ cells and showed proliferation of interstitial cells. However, the uteri were of normal architecture and had well-developed uterine endometrial glands. The samples were 38, XX female karyotype without the sex-determining region Y (SRY) gene.
