Inflammation is an important protective response mechanism that occurs against microbial invasion or injury. However, excessive inflammation may lead to cause of morbidity and mortality in diseases. The activated macrophages plays a vital role in inflammatory response by stimulation of lipopolysaccharide (LPS) and tumor necrosis factor-α (TNF-α). This activation further damages the host by inducing certain pro-inflammatory mediators such as nitric oxide (NO), interleukin-1β (IL-1β), interleukin- 6 (IL-6), TNF-α, inducible nitrous oxide (iNOS) and cyclooxygenase (COX-2). Flavonoids are bioactive compounds with potential effects as anti-cancer, anti-inflammation, anti-viral and anti-bacterial activities. Polymethoxyflavones (PMFs) are unique to citrus plants which are of specific interest owing to their biological effects that includes lipoprotein metabolism and anti-inflammatory activity. Sinensetin is one of the PMFs having five methoxy groups on the basic benzo-γ-pyrone skeleton with a carbonyl group at the C4 position. Sinensetin have been known for exerting various pharmacological activities including anti-angiogenesis, anti-diabetic and anti-inflammatory activities. However, there are no studies focused on the anti-inflammatory effects of sinensetin on skeletal muscle cells. In the present study, we investigated the antiinflammatory effect of flavonoids isolated from Sinensetin on the production of pro-inflammatory mediators mediated by nuclear factor-kappa B (NF-κB) by inhibition of signal transduction in LPS - induced L6 skeletal muscle cells.
Edible biopolymer films were developed from the exopolysaccharides (EPS) extracted from Weissella confusa 113-2. The optimum composition for film formation was determined using the response surface analysis with the explanatory variables of the EPS (0.5-5.5%) and glycerol (0.5-5.5%) concentrations and the response variable of film elastic modulus (EM). The mass ratio of distilled water to solids was set constant (14:1). Tensile strength (TS), percentage elongation at break (%E), EM, water vapor permeability (WVP) of EPS films were evaluated. The glass transition temperatures of the films were also determined by a dynamic mechanical analysis. The optimum mass ratio of EPS to glycerol was 0.754:0.375. The WVP, TS, %E, and EM of the film under the optimal composition were 3.53±0.21 g·mm/kPa·h·m2, 7.03±0.49 MPa, 84.82±12.31%, and 62.03±6.93 MPa, respectively. The glass transition temperature varied from 54 to 83 °C. The EPS film has the potential to be applied to food products as an edible film with physical and barrier properties comparable to other biopolymer edible films.
Anthriscus sylvestris (L.) Hoffm. is a perennial herb found widely distributed in various regions of Korea, Europe, and New Zealand. The root of A. sylvestris have been extensively used in the treatment for antitussive, antipyretic, cough remedy in Oriental medicine, but the physiologically active function of the leaf of A. sylvestris is as yet unknown. In this study, we investigated the anti-cancer activity and the mechanism of cell death of water extracts of leaf of Anthriscus sylvestris (WELAS), on human FaDu hypopharyngeal squamous carcinoma cells. Our data showed that WELAS treatment inhibited cell viability in a concentration- and time-dependent manner. In addition, the treatment of WELAS markedly induced apoptosis in FaDu cells, as determined by the viability assay, DAPI stain and FACS analysis. WELAS also increased the proteolytic cleavage of procaspase-3, -9 and PARP (poly(ADP-ribose) polymerase). In addition, exposure to WELAS decreased the expression of Bcl-2 (an anti-apoptotic factor), but increased the expression of Bax (a pro-apoptotic factor), suggesting that mitochondria-dependent apoptotic pathways are mediated in WELAS-induced apoptosis. Taken together, these results indicate that water extracts of leaf of A. sylvestris inhibits cell growth and induces apoptosis via the mitochondrial-dependent apoptotic pathway in FaDu human hypopharyngeal squamous carcinoma cells. Therefore, we propose that the water extracts of leaf of A. sylvestris is a novel chemotherapeutic drug, having growth inhibitory properties and induction of apoptosis in human oral cancer cells.
The purpose of this study is to verify a correlation between flexi-bar exercise and improvement of trunk strength and body composition in juvenile soccer players. The subjects were 11 teenager juvenile soccer players who conducted flexi-bar exercise half an hour daily three times a week for eight weeks. They were tested for trunk muscles strength, body composition and correlation. The result showed that trunk muscles strength improved significantly( p<.01) and weight, Rt. arm muscle mass, Rt. leg muscle mass, and Lt. leg muscle mass(p<.05), Lt. arm muscle mass, trunk muscle mass, and body muscle mass(p<.01) improved significantly as well while Lt. arm fat mass, Rt. leg fat mass, and Lt. leg fat mass decreased significantly(p<.05). In the correlation analysis, the more trunk muscle mass and trunk muscles strength increased, the more muscle mass of body regions increased, which was a positive correlation whereas fat mass of body regions decreased, which was a negative correlation. It was verified that there was a correlation between flexi-bar exercise and improvement of trunk muscles strength and body composition for juvenile soccer players.
본 연구에서는 어린이 기호식품 중 하나인 과자류(과자, 캔디류, 츄잉껌, 빙과류)에 대하여 1,005건(과자 250건, 캔디류 255건, 츄잉껌 250 건, 빙과류 250건)에 대해 위생 지표 미생물인 일반세균 수, 대장균군/대장균과 더불어 식중독 원인균인 바실러스 세레우스, 클로스트리디움 퍼프 린젠스 및 황색포도상구균에 대한 미생물 검사를 실시하였다. 과자류에 대한 일반세균 수의 경우 전체 시료의 26.8%에서 검출되었고, 검출된 세균 수는 1.7 log CFU/g을 나타내었다. 유형별로 살펴보았을 때에는 과자 250건 중 45.2%의 시료에서 검출되었으며, 빙과류 28.8%, 캔디류 20.0% 및 츄잉껌 13.2% 순으로 검출되는 것으로 확인되었다. 다른 위생지표 세균인 대장균군/대장균의 경우는 모든 시료에서 검출되지 않았다. 식중독균을 분석한 결과에서는 캔디류를 제외한 나머지 유형에서 바실러스 세레우스만 검출이 되었고, 평균 검출량은 1.4~1.6 log CFU/g로 현행 식품공전상의 미생물규격인 3 log CFU/g 이하 수준으로 모두 적합한 제품이었다. 이는 식품공전 상에서 정 하고 있는 미생물 규격에 비해서는 낮은 수치로 식품의 제조·유통에는 문제가 없는 것으로 보이지만, 식중독 민감도가 높은 어린이들에게 안전한 식품을 제공하기 위해 합리적이고 과학적인 미생물 기준규격을 고려해야 한다.
Despite many researches related with in-vitro culture of porcine spematogonial stem cells (SSCs), adherent culture system widely used has shown a limitation in the maintenance of porcine SSC self-renewal. Therefore, in order to overcome this obstacle, suspension culture, which is known to have numerous advantage over adherent culture, was applied to the culture of porcine SSCs. Porcine SSCs retrieved from neonatal testes were suspension-cultured for 5 days or 20 days, and characteristics of suspension-cultured porcine SSCs including proliferation, alkaline phosphatase (AP) activity, and self-renewal-specific gene expression were investigated and compared with those of adherent-cul-tured porcine SSCs. As the results, the suspension-cultured porcine SSCs showed entirely non-proliferative and significantly higher rate of AP-positive cells and expression of self-renewal-specific genes than the adherent-cultured porcine SSCs. In addition, long-term culture of porcine SSCs in suspension condition induced significant decrease in the yield of AP staining-positive cells on post-day 10 of culture. These results showed that suspension culture was inappropriate to culture porcine SSCs, because the culture of porcine SSCs in suspension condition didn’t stimulate proliferation and maintain AP activity of porcine SSCs, regardless of culture periods.
The purpose of this study was to investigate spirometric lung pattern, respiratory function and degree of fatigue by lung function tests and fatigue tests of 39 elderly people in a care facility aged 65 and over. The respiratory function tests were used to the Spirovit SP-1 and fatigue tests were used modified Piper fatigue scale(mPFS). Regarding the respiratory function, the FVC was 1.41±0.36ℓ, the FVC % predicted was 69.10±14.98%, the FEV1 was 1.02±0.31ℓ, the FEV1 % predicted was 63.27±16.05%, the FEV1/FVC was 72.77±13.40%, and the fatigue score was 5.83±1.09. As for the spirometric lung pattern, 19 patients had a restrictive pattern(48.7%), followed by 11 with a mixed pattern(28.2%), 5 with an obstructive pattern( 12.8%), and 4 with a normal pattern(10.3%). Regarding the respiratory function and fatigue by spirometric lung pattern, the FVC and the FVC % predicted of patients with a normal pattern or an obstructive pattern were greater than other groups at a statistically significant level. As for the FEV1, that of patients with a normal pattern was significantly higher than others, and for the FEV1 % predicted, that of patients with a normal pattern or a restrictive pattern was significantly higher(p<.001). Fatigue score by patients with a normal pattern was significantly less than patients of other patterns(p<.001). Therefore, pulmonary physical therapy is considered necessary to improve respiratory function and fatigue degradation of elderly in a facility.
This study was carried out to identify how a self-stretching exercise program affects pain for each body area, pain relief and job satisfaction for care workers. 20 of 40 care workers with musculoskeletal symptom were randomly selected and participated a self-stretching exercise program consisting of 15 motions. The intervention was done five times or more per weeks for 8 weeks and 1 session lasted within 15 minutes. 'Musculoskeletal symptom survey table' of the Korea Occupational Safety and Health Agency(KOSHA) and JDI(Job Descriptive Index) was used for pain on the musculoskeletal symptom and job satisfaction. Survey were done twice before and after the program. The result of this study showed that self-stretching exercise program group(SSPG) relieved from pain significantly in the shoulders(p<.01) and lumbar(p<.05), comparing to the non selfstretching exercise program group(NSPG). Although no significant difference on variations in the JDI appeared in SSPG, the significant reduction appeared from the colleague relationship and organization in NSPG(p<.05). SSPG showed the significant increase on variations in JDI from the job and organization comparing to NSPG. Especially, the improvement on satisfaction for the organization was shown(p<.05). Accordingly, the self-stretching exercise program for care workers can be said to positively affect the overall pain relief and increase on the JDI.
In this case report, we investigated the effects of robot-assisted gait therapy in a chronic stroke patient using motor assessment and gait analysis. A patient who suffered from the right hemiparesis following the left corona radiata and basal ganglia infarction received 30 minutes of robot-assisted gait therapy, 3 times a week for 4 weeks. Outcome was measured using Motoricity index(MI), Fugl-Meyer assessment(FMA), modified motor assessment scale(MMAS), isometric torque, body tissue composition, 10-meter gait speed and gait analysis. After robot-assisted gait therapy, the patient showed improvement in motor functions measured by MI, FMA, MMAS, isometric torque, skeletal muscle mass, 10-meter gait speed. In gait analysis, cadence, single support time, double support time, step length, walking speed improvement in after robot-assisted gait therapy. The results of this study showed that robot-assisted gait therapy is considered to facilitate locomotor recovery of the chronic hemiparetic stroke patient.
The purpose of this study was to compare the effects of treadmill walking in underwater and overground which affects gait and physical function of people who have had a stroke. Twenty people after a stroke who have become hemiplegic over 6 months were participated. Participants were divided into two groups: underwater treadmill group(UTG) and overground treadmill group(OTG). The intervention was done 4 times per weeks for 6 weeks and 1 session lasted for 30 minutes. Gait and physical function elements were measured at baseline, at the middle(3 weeks) and at the end of the intervention(6 weeks). For the elements of gait, walking velocity, affected stance phase, affected weight bearing were assessed. For the elements of physical function, Short Form 8(SF-8) health survey was used. The result of this study showed that both groups improved similarly in walking velocity. However participants in UTG improved more than those in OTG in affected stance phase(p<.05), affected weight bearing( p<.05) and emotional aspect(p<.001). Based on the results of this study, it can be suggested that treadmill walking both in underwater and on the ground can be effective in improving hemiplegic gait and physical function of people who have had a stroke. The result also suggest that the underwater treadmill exercise can be more effective than overground treadmill in restoration of gait in people after stroke.
Multivessel spontaneous coronary artery dissection is extremely rare. There are no guidelines regarding optimal treatment of spontaneous coronary artery dissection. The treatment depends on clinical symptoms, extent and location of the dissection, and the hemodynamic status of the patient. Although percutaneous coronary intervention might be required for all of the dissecting arteries, this case demonstrated the importance of an appropriate selection of treatment modalities for spontaneous coronary artery dissection.
Genetically modified (GM) papaya (Carica papaya L.) line 55-1 (55-1), which is resistant to papaya ringspot virus infection, has been marketed internationally. Many countries such as the European Union, Japan, and Korea have a mandatory safety assessment, approval and labeling regulations for GM foods. Thus, there is a need for specific methods for detecting 55-1. In this study, we established a real-time PCR detection method applicable to 55-1 for a variety of papaya products. The limit of detection was possible for fresh papaya fruit up to dilutions of 0.005% and 0.01% (weight per weight [w/w]) for homozygous SunUp and heterozygous Rainbow cultivars, respectively, in non-GM papaya. The 55-1 event-specific detection method observed parallelism (r2>0.99) between the concentration of line 55-1 cultivars and Ct values obtained in amplification plots at concentrations of 0.005-10% for SunUp DNA and 0.01-10% for Rainbow DNA. The method was applicable to the qualitative detection in various types of processed products (cocktail fruit, dried fruit, juice, etc.) containing papaya as a main ingredient. Monitoring papaya products for the presence of GM papaya were demonstrated using a P35S and T-nos real-time PCR detection method but no amplification signals were detected.
