        Foot-and-mouth disease (FMD), which affects cloven-hoofed animals, is economically important because of its highly contagious nature. FMD virus (FMDV), the causative agent of FMD, involves seven serotypes (O, A, Asia1, C, and SAT 1-3). Serotype Asia1 is unique to the Asian territory and is subdivided into nine genetic groups (G-I-IX) based on nucleotide variations in the VP1 sequence. Asia1 Shamir, the most representative Asia1 vaccine, is not highly protective against the Asia1/MOG/05 (G-V) lineage found in North Korea in 2007. Therefore, we investigated whether a chimeric virus strain (Asia1/MOG/Shamir), in which the VP4, VP2, and VP3 sequences of Asia1/MOG/05 were combined with the VP1 sequence of Asia1 Shamir, can simultaneously protect against both viruses. We determined the optimal viral growth conditions for the commercial utilization of this chimeric virus strain. Of the three types of cell culture media, the Cellvento medium resulted in the highest amount of antigen in the samples. The chimeric strain was proliferated in a small bioreactor to produce a test vaccine, and its immunogenicity was evaluated in pigs. The virus neutralization (VN) titer against the Asia1 Shamir virus was > 1/100 after the second immunization with the chimeric vaccine in pigs. In addition, a single dose of the test vaccine resulted in a VN titer of > 1/100 against the Asia1/MOG/05 strain. Taken together, our chimeric vaccine strain provided sufficient protection against the Asia1/MOG/05 and Asia1 Shamir viruses, suggesting its potential as a novel vaccine for both these strains.

        The domestic Pressurized Heavy Water Reactor (PWHR) nuclear power plant, Wolsong Unit 1, was permanently shut down on December 24, 2019. However, research on decommissioning has mainly focused on Pressurized Water Reactors (PWRs), with a notable absence of both domestic and international experience in the decommissioning of PHWRs. If proper business management such as radiation safety and waste is not performed, it can lead to increased business risks and costs in decommissioning. Therefore, the assessment of waste volume and cost, which provide fundamental data for the nuclear decommissioning process, is a crucial technical requirement before initiating the actual decommissioning of Wolsong Unit 1. Decommissioning radiation-contaminated structures and facilities presents significant challenges due to high radiation levels, making it difficult for workers to access these areas. Therefore, technology development should precede decommissioning process assessments and safety evaluations, facilitating the derivation of optimal decommissioning procedures and ensuring worker safety while enhancing the efficiency of decommissioning operations. In this study, we have developed a program to estimate decommissioning waste amounts for PHWRs, building upon prior research on PWR decommissioning projects while accounting for the specific design characteristics of PHWRs. To evaluate the amount of radioactive waste generated during decommissioning, we considered the characteristics of radioactive waste, disposal methods, packaging container specifications, and the criteria for the transfer of radioactive waste to disposal operators. Based on the derived algorithm, we conducted a detailed design and implemented the program. The proposed program is based on 3D modeling of the decommissioning components and the calculation of the Work Difficulty Factor (WDF), which is used to determine the time weighting factors for each task. Program users can select the cutting and packaging conditions for decommissioning components, estimate waste amount based on the chosen decommissioning method, and calculate costs using time weighting factors. It can be applied not only to PHWRs, but also to PWRs and non-nuclear fields, providing a flexible tool for optimizing decommissioning process.

        Background: The grading of Hanwoo (Korean native cattle) is based on four economic traits, and efforts have been continuously made to improve the genetic traits associated with these traits. There is a technology to predict the expected grade based on the 4 economic genetic SNP characteristics of Korean cattle calves using single nucleotide polymorphism (SNP) technology. Selection of highly proliferative, self-renewing, and differentiating satellite cells from cattle is a key technology in the cultured meat industry. Methods: We selected the Hanwoo with high and low-scored of genomic estimated breeding value (GEBV) by using the Hanwoo 50K SNP bead chip. We then isolated the bovine satellite cells from the chuck mass. We then conducted comparative analyses of cell proliferation, immunocytochemistry, qRT-PCR at short- and long-term culture. We also analyzed the differentiation capability at short term culture. Results: Our result showed that the proliferation was significantly high at High scored GEBV (Hs-GEBV) compared to Low scored GEBV (Ls-GEBV) at short- and long-term culture. The expression levels of Pax3 were significantly higher in Hs-GEBV bovine satellite cells at long-term culture. However, there were no significant differences in the expression levels of Pax7 between Hs- and Ls-GEBV bovine satellite cells at short- and long- term culture. The expression levels of MyoG and MyHC were significantly high at Ls-GEBV bovine satellite cells. Conclusions: Our results indicated that selection of bovine satellite cells by Hanwoo 50K SNP bead chip could be effective selection methods for massive producing of satellite cells.

        With the aging of nuclear power plants (NPPs) in 37 countries around the world, 207 out of 437 NPPs have been permanently shutdown as of August 2022 according to the IAEA. In Korea, the decommissioning of NPPs is emerging as a challenge due to the permanent shutdown of Kori Unit 1 and Wolsong Unit 1. However, there are no cases of decommissioning activities for Heavy Water Reactor (HWR) such as Wolsong Unit 1 although most of the decommissioning technologies for Light Water Reactor (LWR) such as Kori Unit 1 have been developed and there are cases of overseas decommissioning activities. This study shows the development of a decommissioning waste amount/cost/process linkage program for decommissioning Pressurized Heavy Water Reactor (PHWR), i.e. CANDU NPPs. The proposed program is an integrated management program that can derive optimal processes from an economic and safety perspective when decommissioning PHWR based on 3D modeling of the structures and digital mock-up system that links the characteristic data of PHWR, equipment and construction methods. This program can be used to simulate the nuclear decommissioning activities in a virtual space in three dimensions, and to evaluate the decommissioning operation characteristics, waste amount, cost, and exposure dose to worker. In order to verify the results, our methods for calculating optimal decommissioning quantity, which are closely related to radiological impact on workers and cost reduction during decommissioning, were compared with the methods of the foreign specialized institution (NAGRA). The optimal decommissioning quantity can be calculated by classifying the radioactivity level through MCNP modeling of waste, investigating domestic disposal containers, and selecting cutting sizes, so that costs can be reduced according to the final disposal waste reduction. As the target waste to be decommissioning for comparative study with NAGRA, the calandria in PHWR was modeled using MCNP. For packaging waste container, NAGRA selected three (P2A, P3, MOSAIK), and we selected two (P2A, P3) and compared them. It is intended to develop an integrated management program to derive the optimal process for decommissioning PHWR by linking the optimal decommissioning quantity calculation methodology with the detailed studies on exposure dose to worker, decommissioning order, difficulty of work, and cost evaluation. As a result, it is considered that it can be used not only for PHWR but also for other types of NPPs decommissioning in the future to derive optimal results such as worker safety and cost reduction.

        Once a radioactive material is released from the nuclear power plant (NPP) by accident, it is necessary to understand the behavior of radioactive plume to protect residents adequately. For this, it is essential to measure the radiation dose rate around NPPs at important locations. Our previous study developed a movable radiation detector that can be installed quickly in an accident to measure gamma dose rate in areas where environmental radiation monitoring system is not installed. The data measured by the detector are transmitted to the server in real-time through LoRA wireless communications. There are two methods to use LoRA communications; one is self-network, and the other is the network provided by the mobile carrier. A signal receiver, called a gateway, should be equipped near the installation location of radiation detectors to use a self-network without using the mobile carrier’s system. In other words, the movable radiation detectors we made can function if there should be any gateway near them. The distance capable of communication between gateway and detector is about 8 km in an open area without significant obstacles. Korea has many significant obstacles, such as mountains around most NPPs. Thus, the gateways could be installed in the proper position before the accident to operate the movable radiation detectors without problems. If the gateway is located at a high position like a mountain top, it could cover a wide area. In this study, the elevation database in the area around the NPPs was collected and analyzed to determine where gateways should be installed. The analysis range is limited in the urgent protective action planning zone. The optimization was also performed to minimize the number of gateways.

        There is a growing global interest in food choices that could affect health. Consumer food choices are dictated by numerous factors such as attitudes, beliefs, desires and preferences stemming from psychological and physiological attributes. The present study aimed to understand consumers’ internal perceptions about cream soup, cream sauce, and tomato sauce using ZMET (Zaltman’s Metaphor Elicitation Technique). The 9 step ZMET interview process was conducted for a sample of 36 consumers (12 consumers for each sauce). A content analysis of the survey results was carried out. From this, 56 concepts of cream soup were derived, and 15 constructs that met with the agreement of more than one-third of the total sample consumers were extracted. These 15 constructs included ‘Soft’, ‘Recollection’, ‘Familiar’, ‘Warm’, ‘Comfortable’, etc. Similarly, for cream sauce, 67 concepts and 20 constructs ‘Silky’, ‘Warm’, ‘Restaurant’, ‘Family’, ‘Memory’, etc. were deduced. A total of 66 concepts and 20 constructs for tomato sauce ‘Sourness’, ‘Sauce Bottle’, ‘Pleasant’, ‘Ingredient’, ‘Cooking’, etc. were derived. The analysis of consumers’ consensus maps through this study provides a deep and useful understanding of consumers and their latent needs. Also, the results of this study indicate that exploring consumers’ internal perceptions is critical to understanding their healthy food choices. This can be used as basic data for formulating marketing strategies.

        The emulsion and micro-encapsuled formulation from three essential oils, cassia, thyme white and lemon grass were tested for their insecticidal activities against Lycoriella ingenua (Dufour) adults at 0.3% concentration. As a result, the micro-encapsules than emulsion formulation was confirmed to show greater activity. Based on this result, insecticidal activities at several levels of concentration were examined, resulting in more than 80% insecticidal activity after 30 minutes at 0.6% level of cassia formulation. And also, insecticidal activity of three oil-based micro-encapsuled formulations lasted for up to 12 hours at 0.9% concentration. The main ingredients derived from three essential oils were evaluated on the insecticidal activity against Lycoriella ingenua. As a result, in case of thymol from thyme white, it showed more than 80% mortality after over 30 minutes at 0.25%, and in cinnamic aldehyde and citral, exhibited 88% and 82% mortality after 30 minutes at the 0.5% level, respectively. These three compounds were examined for whether the insecticidal activity was maintained depending on being over time. In case of thymol, it maintained good insecticidal activity against Lycoriella ingenua at 0.5% level until 120 hours, while cinnamic aldehyde and citral showed lower activity after 12 hours.

        In the previous study, the plant essential oils such as clove bud , thyme white and garlic oils gave potent toxicity with the range of 1.5 to 4.5×10-3 ul cm-3 of LC50 values against Bemisia tabaci. Based on the bio-assay results, three plant essential oils(clove bud, thyme white and garlic) were applied to 30% emulsion type. The 100, 200 and 400 fold dilution of these emulsions was showed mild toxicity under port scale treatment against Q biotype. At each fold dilution, clove bud showed 44, 26 and 14%, and thyme white produced 44, 39 and 27%, respectively. Of garlic emulsion, its toxicity was evaluated with 27, 26 and 11%, respectively. But, in case of clove bud and thyme white mixture, their toxicity was more good under the mixture than under the single treatment. Mixture with clove bud 200 fold dilution adding to thyme white 200 fold dilution showed potent activity with 86% mortality. And also 200 adding to 400, and 400 to 400 exhibited 61%, 76%, respectively. These essential oil mixtures showed similar strong toxicity against B biotype under port scale test. In particular, the mixture with clove bud 200 fold dilution adding to thyme white 200 fold dilution showed the highest synergistic effect. Toxicity to plants, except the 100 fold dilution of these emulsions was not observed.

        Human embryonic stem (ES) cells are derived from the inner cell mass of the preimplantation embryo. Human ES cells have the capacity to differentiate into various types of cells in the body. Human ES cells are indefinite source of cells for cell therapy in various degenerative disorders including neuronal disorders. Directed differentiation of human ES cells is a prerequisite for their clinical application. The objective of this study is to develop the culture condition for the derivation of neural precursor cells from human ES cells. Neural precursor cells were derived from human ES cells in a stepwise culture condition. Neural precursor cells in the form of neural rosette structures developed into neurospheres when cultured in suspension. Suspension culture of neurospheres has been maintained over 4 months. Expressions of nestin, soxl, sox2, pax3 and pax6 transcripts were upregulated during differentiation into neural precursor cells by RT-PCR analysis. In contrast, expression of oct4 was dramatically downregulated in neural precursor cells. Immunocytochemical analyses of neural precursor cells demonstrated expression of nestin and SOX1. When induced to differentiate on an adhesive substrate, neuro-spheres were able to differentiate into three lineages of neural systems, including neurons, astrocytes and oligo-dendrocytes. Transcripts of sox1 and pax6 were downregulated during differentiation of neural precursor cells into neurons. In contrast, expression of map2ab was elevated in the differentiated cells, relative to those in neural precursor cells. Neurons derived from neural precursor cells expressed NCAM, Tuj1, MAP2ab, NeuN and NF200 in immunocytochemical analyses. Presence of astrocytes was confirmed by expression of GFAP immuno-cytochemically. Oligodendrocytes were also observed by positive immuno-reactivities against oligodendrocyte marker O1. Results of this study demonstrate that a stepwise culture condition is developed for the derivation of neural precursor cells from human ES cells.

        Acute myocardial infarction (AMI) as an acute form and a catastrophic manifestation of coronary artery disease (CAD) is the major cause of morbidity and mortality worldwide. The prevalence and incidence of myocardial infarction can be influenced by demographic, biological and psychosocial factors, and it is rare in childhood and adolescent years. Contrary to the elderly, where cardiac attack is mainly caused by CAD, exercise-induced cardiac attack is relatively more prevalent in a young adult. We report here on a case of ST-segment elevation myocardial infarction (STEMI) in a young adult during vigorous physical activity and we review the relevant literatures.

        Background : The effect of ridge-up bed cultivation and stem training method on the plant growth and yield of Cynanachum willfordii was investigated. Methods and Results : Domestic variety of Cynanachum willfordii was tested at Chungbuk ARES under the following conditions. Nonsupporting (custom) and I-form supporting was used for stem training method, and ridge height was set as 30 (custom) and 60 cm (high ridge). Planting spacing was adjusted to 30 (interrow spacing) × 20 cm (intrarow spacing), and other major management was followed the method of standard cultivation for medicinal crop. Investigated result from leaf characteristics, leaf length was longer in high ridge cultivation (HRC) as 11.1 cm than custom cultivation (CC) as 10.6 cm. Leaf width is proved to be 12.8 (HRC) > 11.2 cm (CC). Leaf number is proved to be 294 (HRC) > 254/plant (CC), with higher number of 44/plant at HRC and weight/10 leaves were more heavy at HRC (4.9 g) than that of CC (2.6 g). It was more fruit setting at HRC over 15/plant. According to the stem training method and ridge height from nonsupporting cultivation, main root was 4.0 (CC), higher than that of HRC over 0.5/plant. However, root length was more longer in HRC (28.6) than that of CC (25.0 cm). Main root diameter was also showed more thicker pattern in HRC. From staking cultivation, root number of HRC was 7/plant, it was recorded more 3/plant than that of CC, and it was also same pattern in main root length and root diameter. By the standard of commercial root, yield of living roots in nonsupporting cultivation were 59.0g/plant (HRC), it was more heavy over 10.4g/plant than that of CC. In staking cultivation, HRC were recorded as 74.2g/plant, more heavy 6.9g/plant than that of CC. Yield from I-form support stem training and ridge-up bed cultivation of HRC was higher approxmiately 52.7% than that of CC. Conclusion : Overall growth by high ridge cultivation of I-form support was good and yield of HRC was also increased over 52.7% than that of CC.

        Genome sequencing researches for considerable numbers of crops and wild plants are being developed. Cytogenetic researches according to chromosome number and size are essential to confirm and comprehend ploidy level and genome size before genome sequencing project is actually conducted. Cytogenetic researches on six food crop plants were carried out by DAPI staining and fluorescence in situ hybridization (FISH) method. Fagopyrum esculentum Moench showed 2n=2x=16, each chromosome length of 1.42㎛ to 1.77㎛, total chromosome length of 13.31㎛, and karyotypic formula of 2n=8m; Phaseolus angularis W.F. Wight, 2n=2x=22, 2.01㎛ to 3.84㎛, total 28.03㎛, 2n=9m+2sm, Perilla frutescens var. japonica Hara, 2n=2x=40, 1.73㎛ to 2.76㎛, total 44.36㎛, 2n=5m+13sm+2st. Chromosome sizes of the other three species such as, Panicum miliaceum L., 2n=2x=36, total chromosome length of 30.83㎛, Sesamum indicum L., 2n=2x=26, 27.39㎛, lpomoea batatas L., 2n=2x=30, total 33.51㎛ were too small for each chromosome type to be identified and analyzed. The result of FISH analysis using 5S and 45S rDNA probe showed species-specific chromosome locations in the genome. These preliminary analyses were carried out to decide which food crop to prioritize for genome sequencing.         Mungbean (Vigna radiata) is a fast-growing, warm-season legume crop that is primarily cultivated in developing countries of Asia. We constructed a draft genome sequence of mungbean to facilitate genome research into the subgenus Ceratotropis and to enable a better understanding of the evolution of leguminous species. The draft genome sequence covers 80% of the estimated genome, of which 50.1% consists of repetitive sequences. In total, 22,427 high confidence protein-coding genes were predicted. Based on the de novo assembly of additional wild mungbean species, the divergence of what was eventually domesticated and the sampled wild mungbean species appears to have predated domestication. Moreover, the de novo assembly of a tetraploid Vigna species (Vigna reflexo-pilosa var. glabra) provided genomic evidence of a recent allopolyploid event. To further study speciation, we compared de novo RNA-seq assemblies of 22 accessions of 18 Vigna species and protein sets of Glycine max and Cajanus cajan. The species tree was constructed by a Bayesian Markov chain Monte Carlo method using highly confident orthologs shared by all 24 accessions. The present assembly of V. radiata var. radiata will facilitate genome research and accelerate molecular breeding of the subgenus Ceratotropis.

        Cardiovascular system is the primary organ to develop and reach a functional state, which underscores the essential role of the vasculature in the developing embryo. The vasculature is a highly specialized organ that functions in a number of key physiological works including the carrying of oxygen and nutrients to tissues. It is closely involved in the formation of heart, and hence it is essential for survival during the hatching period. The expression of genes involved during vascular development in the olive flounder (Paralichthys olivaceus) in the days after hatching is not fully understood. Therefore, we examined the expression patterns of genes activated during the development of flounder. Microscopic observations showed that formation of blood vessels is related to the expression of the vimentin gene. Also, the temporal expression patterns of this vimentin-like gene in the developmental stages and in the normal tissues of olive flounder. The purpose of this study was to examine the expression patterns of vimentin in normal tissues of the olive flounder and during the development of the vascular system in newly hatched olive flounders and HIF-1 plays a vital role in the formation of blood vessels during development. Vimentin expression was strong at the beginning of the development of blood vessels, and was present throughout all developmental stages. Our findings have important implications with respect to the roles of vimentin and HIF-1 in the development and evolution of the first blood vessels in olive flounder. Further studies are required to elucidate the vimentin-mediated hypoxic response signal transduction and to decipher the functional role of vimentin in developmental stages.
