팽이버섯 내에 존재하는 GAD 효소를 발효를 통해 활 성화 시켜 MSG를 GABA로의 전환율을 높이고자 하였다. 효과적인 고농도 GABA를 생산하기 위해 나노분말 팽이 버섯에다가 수경재배한 인삼을 첨가하여 야쿠르트발효기 에서 발효한 결과 GABA 전환율은 팽이나노인삼분말 발 효군(88%) > 팽이분말 발효군(52%) > 팽이나노분말 발 효군(44%) 순으로 나타났다. 이러한 결과는 MSG를 기질 로 첨가하는 식품에서 활용할 가치가 있으리라 사료된다.
γ-Aminobutyric acid (GABA)-containing salt was prepared by crystallization of a mixture of salt water from deep sea and fermentation broth by lactic acid bacteria that contained GABA converted from glutamic acid. Salt from deep sea water has a lower sodium content but higher calcium, potassium and magnesium contents than commercial salt. Instead of monosodium glutamate (MSG), glutamic acid was used for solving the residual MSG problem. Fermentation by a lactic acid bacterium converted 90% of added glutamic acid (5%, w/v) to GABA, and continuous production of colorless fermentation broth containing more than 3% (w/v) GABA was achieved by using an activated carbon. Mixtures of salt water and fermentation broths with various GABA concentrations were co-crystallized and the GABA content was analyzed. This analysis showed that more than 90% of GABA from broth was adsorbed to salt. The appearance and surface of this prepared GABA-containing salt were examined with an image analyzer and scanning electron microscope. No difference was found with commercial sun-dried salt and no separated particles were detected, which indicates that the co-crystallization process used is suitable for the production of GABAcontaining salt.
The γ/γ´ two-phases, commonly known as a eutectic structure, are observed in the γ interdendritic region of a Nibase superalloy. However, the growth behavior of the γ/γ´ two-phases, whether it is of eutectic or peritectic nature, has not been decidedly established. Directional solidifications were, thus, performed with the planar interface at a low growth rate of 0.5 μm/s in order to promote macro segregation. Directional solidification started with the γ planar interface and the γ´ phase nucleated on the γ planar interface at the solidification fraction of 0.75. The γ/γ´ two-phases showed the γ´ rod structure as major phase and the γ minor phase between γ´ rods, and the volume fraction of the γ phase changed continuously with an increasing solidification fraction. The two-phase γ/γ´ is seen as the coupled peritectic.
We present a kinematic study of the parsec-scale radio jet in OJ 287, one of the most studied BL Lac objects, during γ-ray ares, to explore the relation between parsec-scale radio jet activity and γ-ray emission. The 22-GHz light curve of OJ 287 show three obvious are events around 2011 May, 2011 October, and 2012 March. The second radio are occurred during the γ-ray aring period, and the third radio are seemed to precede the γ-ray are by one month. One jet component moved outward with respect to the core component with an apparent superluminal speed (~ 11c) from 2010 November to 2011 November. Then it changed direction, moving apparently inward in 2011 November, when the γ-ray are occurred. The observed apparent inward motion of the jet at 22 GHz could be caused by a new jet component, unresolved at 22 GHz, in the innermost region.
Mushroom is known for anti-inflammatory and anti-oxidative potential. This study provides evidence that theinhibitory effect of mushroom on the expression of pro-inflammatory cytokines in human keratinocytes, HaCaT cells. To definethe underlying mechanisms of action, tumor necrosis factorα/IFNγ-activated human keratinocytes model was used. Mushroomsignificantly inhibited the expression of cytokines in HaCaT cells. Taken together, the results demonstrate that mushroom inhibitedinflammtion, suggesting that mushroom (DW extract: Grifola frondosa Cordyceps militaris), (Ethanol extract: Ganoderma lucidum,Lentinus edodes, Cordyceps militaris, Flammulina velutipes) might be a candidate for the treatment of skin inflammation.
The sexual maturation occurred by the changes of steroid hormones was known to sex-dependently and/or agedependently regulate the lipid metabolism in various animal species. Our current study demonstrates that lipid and its functional fatty acids can be changed depending on the status of sexual maturation. Of the functional fatty acids, γ- linolenic acid (GLA; 18:3n-6) is an important factor for maintaining human health. The purpose of our study was to investigate the level of GLA in mice with different stages of sexual maturation. To this end, the longissimus muscle (LM) of immature (3-week-old) and mature (7-week-old) female mice was analysed for the fatty acid composition by gas chromatography. Furthermore, both gene and protein level of Δ6 desaturase (FADS2) which is involved in GLA metabolism by real time PCR and Western blotting, respectively. Mature females showed greater (P<0.05) serum 17β -estradiol (E2) level and LM GLA contents than immature group. The mRNA and protein levels of FADS2, which converts precursor linoleic acid into GLA, were higher (P<0.05) in mature female mice than in immature mice. In conclusion, these results show that sexual maturation of female mice induces GLA and FADS2 contents in LM.
본 연구에서는 포장방법 (진공, 일반)과 저장온도(4℃, -20℃)를 달리한 육류 (소고기, 닭고기) 및 생선에 대해 저선량 감마선 반복조사를 적용하였을 때 나타나는 병원성미생물 E. coli O157:H7 및 S. Typhimurium의 사멸효과를 확인하였다. 소고기와 닭고기의 경우 E. coli O157: H7은 최소 2 kGy 이상의 단일조사와 0.5 kGy 이상으로 반복조사 시 생육이 확인되지 않았고, S. Typhimurium은 최소 2kGy 이상의 단일조사와 1 kGy 이상으로 반복조사 시 생육이 확인되지 않았다. 생선의 경우 최소 0.5 kGy 이상의 단일 및 반복조사 시 E. coli O157:H7 와 S. Typhimurium 의 생육이 확인되지 않았다. 육류보다 생선이 낮은 선량으로 미생물을 사멸시킬 수 있었으며, 전반적으로 포장 방법에 따른 조사선량의 차이는 없었으나 저장온도의 경우 냉장보다 냉동의 경우가 좀 더 높은 조사선량을 요구하는 것으로 확인되었다.
γ-Aminobutyric acid (GABA) is present in many vegetables and fruits, but not in dairy products. GABA is known to be beneficial for preventing neurological disorders and hypertension. The objective of this study was to measure the physicochemical changes, number of lactic acid bacteria, and GABA content in fermented goat milk containing rice (1%). The levels of pH and titratable acidity (TA) in each test samples were found to be pH 4.4∼4.5 and 0.74∼0.8%, respectively. The number of viable lactic acid bacteria between test samples ranged from 8.63 to 8.95 Log CFU/ml. Therefore, pH, TA, and number of viable cells in each test sample had no difference. Furthermore, the Lactobacillus delbrueckii subsp. bulgaricus SP5 (Lb. bulgaricus SP5) showed the highest GABA production in goat milk containing rice. Its maximum GABA yield was 3.4 mg/100 mL. The results suggest that GABA contents of fermented goat milk containing rice can be enriched using Lb. bulgaricus SP5.
Our previous research on sulfated polysaccharide purified from Ecklonia cava, a brown alga found in Jeju island, Korea, showed that sulfated polysaccharides modulate the apoptotic threshold of intestinal cells, thereby preventing intestinal damage caused by ionizing radiation. In this study, we investigated the ability of sulfated polysaccharide to augment restoration of small intestinal stem cells from γ-ray-induced damage. In our results, sulfated polysaccharide treatment increased the numbers of Ki-67-positive cells as well as inducible nitric oxide synthase (iNOS)-expressing cells in the small intestine compared with those of irradiated only mice. Meanwhile, exposure to irradiation increased the number of paneth cells, which are frequently associated with intestinal inflammation, whereas sulfated polysaccharide treatment reduced the number of paneth cells in the small intestinal crypt. Conclusively, our data suggest that reduction of iNOS-expressing cells and paneth cells in sulfated polysaccharide-treated mice contributes to the inhibition of radiation-induced intestinal inflammation.
The Lactobacillus brevis 340G strain isolated from traditional Korean fermented food (kimchi) produced 15.50mMof γ-aminobutyric acid (GABA) after 48 h of cultivation in MRS media containing 1% L-monosodium glutamate(MSG). The culture conditions of Lb. brevis 340G were optimized for GABA production. Lb. brevis 340G was cul-tivated at 30oC in optimized MRS media containing 3% sucrose and 2% yeast extract with 3% MSG, resulting inmaximum GABA production (68.77mM) after 54 h of cultivation. Skim milk fermented with Lb. brevis 340G pro-duced 4.64mM of GABA in the presence of 1% MSG. These results suggest that Lb. brevis 340G could be usedas a starter for functional fermented foods and skim milk fermented with Lb. brevis 340G could be further devel-oped to become functional dairy food fortified with GABA.
We analyzed the contents of γ-oryzanol, which is contained in brown rice of the nation rice varieties Chucheong, Heukjinju and Sindongjin, by HPLC. Furthermore we also performed experiments on its biological activity, to prove the effectiveness of rice bran. The contents of γ-oryzanol contained in hulled rice showed 1, 587 ppm for Heukjinju, followed by Chucheong(1, 038 ppm), and by Sindongjin(472 ppm). In anti-oxidative activity, we performed an experiment, by measuring the radical scavenging activity of DPPH. Heukjinju showed the best effect, and Chucheong showed the worst effect. In cholesterol lowering activity, Heukjinju showed the best activity and Sindongjin showed the worst effect. In anti-bacterial activity as well, Heukjinju showed the best activity, and Sindongjin showed the worst effect. Through these experiments, we compared the contents of γ-oryzanol, which is contained in hulled rice(Chucheong, Heukjinju, Sindongjin). Also, we found the anti-oxidation effect, cholesterol lowering effect, and anti-bacterial activity of the γ-oryzanol extracts. Based on our research, we expect that γ-oryzanol can work as a new drug, or nutritional supplement.
Corn has been used for a long time as a traditional remedy, as well as a food source. We previously reported that in vitro supplementation of corn water extracts enhanced the proliferation of splenocytes, compared to the control group. In this study, we examined the immunomodulative effect of a water extract of corn. Seven to eight weeks old mice(Balb/c) were fed an ad libitum chow diet, and were orally administrated a water extract of corn every other day, for four weeks, at two different concentrations(50 and 500 ㎎/㎏ B.W). Cytokine production(IL-2, IL-10 and IFN-γ) by macrophages stimulated with LPS or not stimulated with LPS was detected by ELISA assay using the cytokine kit. In an ex vivo study, the cytokines IL-2, IL-10 and IFN-γ were detected at 500 ㎎/㎏ b.w. supplementation group with LPS stimulation in all cases. Also, the ratio of IFN-γ to IL-10 was in the range of 0~3 with mitogen stimulation, such as con A and LPS. In conclusion, this study suggests that in mice, corn extracts may enhance immune function by regulating the cytokine production(IL-2, IL-10 and IFN-γ) of the activated macrophages.
