        Background: In this study, microwave extraction was used, which is an effective method to extract useful bioactive substances as it requires low quantities of solvent and short time periods. The aim of this study was to determine the optimal extraction conditions for Agrimonia pilosa Ledeb. Methods and Results: The independent variables were ethanol concentration, microwave power, and extraction time, each of which had five levels. The dependent variables were total polyphenol and total flavonoid content, and DPPH radical scavenging activity. To determine the optimal extraction conditions for bioactive compounds, a response surface methodology was employed. Contour maps were generated from polynomial equations. The optimal conditions were then assumed by superimposing these contour maps. Based on the resulting graph, the optimal microwave extraction conditions for Agrimonia pilosa Ledeb were determined as 42 - 48% ethanol concentration, 240 - 280W microwave power, and 13 - 20 min of extraction time. Conclusions: Ethanol concentration had a significant effect on microwave extraction, in terms of total polyphenol and total flavonoid content, as well as DPPH radical scavenging activity. Microwave power and extraction time influenced the total polyphenol content, but not the total flavonoid content or the DPPH radical scavenging activity.

        Electrochemical reductive extraction of tin from semiconductor plating process wastewater was experimented using synthetic wastewater. Copper and graphite plate were used as a cathode and an anode, respectively. The tin extraction could be optimized in pH 0.5 and polar space of 60 mm. The extraction rate of tin per minute was increased as current and initial tin concentration increased, and more than 87% and 97% of tin could be extracted within 80 minutes at 500 mg/L and 1,000 mg/L of initial tin concentration, respectively. The electrochemical reaction orders and kinetic coefficients were 1.24 ~ 1.26 and 0.004 ~ 0.006 (L/mg)(n − 1)min−1. The residual concentration of tin could be expressed as Ct= (Co −0.246+ 0.0012t)−4.065.

        To establish the optimal manufacturing conditions of soybean koji, soybean Koji prepared with Aspergillus oryzae 6-M-1 and Bacillus subtilis 3-B-1 isolated from traditional Korean meju. During 7 days of making Koji, the amount of amino-type nitrogen was getting more increase. The amount of amino-type nitrogen of Koji prepared with A. oryzae 6-M-1 was 686.16 mg% (w/w), that of Koji with B. subtilis 3-B-1 was 643.46 mg% (w/w) at seventh day of making Koji. The α-amylase activity of Koji prepared with A. oryzae 6-M-1 was 1472.54 unit/g, that of Koji with B. subtilis 3-B-1 was 791.00 units/g on the seventh day of the making. The acidic protease activity of Koji prepared with A. oryzae 6-M-1 was 309.00 unit/g, that of Koji with B. subtilis 3-B-1 was 135.88 unit/g at 7th day of making. The amount of amino-type nitrogen and enzyme activities of soybean Koji prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 were produced more than those of wheat flour Koji made in factory. Sensory evaluation on a commercial doenjang and doenjangs prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 was not significantly different at p<0.05.

        This study aimed to investigate the optimal enzymatic hydrolysis conditions of alginic acid using Viscozyme® L, Celluclast® 1.5L, Saczyme®, Novozym®, Fungamyl® 800L, Driselase® Basidiomycetes sp., and Alginate Lyase, for production of reducing sugar. Response surface methodology (RSM) based on central composite rotatable design was used to study effects of the independent variables such as enzyme (1-9% v/w), reaction time (10-30 h), pH (3-7) and reaction temperature (30-70oC) on the production of reducing sugar from alginic acid. The coefficients of determination (R2) of Y1, Y2, Y3, Y4, Y5, Y6, and Y7 for the dependent variable regression equation were analyzed as 0.947 0.968, 0.840, 0.926, 0.923, 0.892 and 0.825. And the p-value of Y1, Y2, Y3, Y4, Y5, Y6, and Y7 within 1% (p < 0.01) was very significant. The optimal conditions were 1.0% of the quantity of the enzyme, 10.0 hours of the response time, pH 3 and 70.0oC of the reaction temperature, where the production rate was 483.1 mg/g-alginic acid, the highest of all the enzymes used.

        Electrochemical reductive extraction of copper from LCD manufacturing process and through hole plating process for PCB circuit board wastewater was experimented using synthetic wastewater. Copper plate which could be used as raw material through melting with extracted copper from wastewater and graphite plate were used as a cathode and an anode, respectively. The copper extraction could be optimized in pH 2 and polar space of 45mm. The extraction rate of copper per unit energy was decreased as HRT increased and initial copper concentration decreased. As the optimal HRTs which could maximize the productivity for copper on energy, 80 ~ 110 min at 50 mg/L, 64 min at 1,000 mg/L and 77 ~ 98 min at 3,000 mg/L were determined, respectively.

        This study was performed to develop new anti-obesity agents for functional foods from medicinal plants and herbs. Methanol extracts from 560 kinds of medicinal plants and herbs, and their lipase inhibitory activities were investigated. Methanol extract from Desmodium oxyphyllum Dc showed the highest lipase inhibitory activity at 74.2%. This lipase inhibitory activity was maximally extracted when powder of Desmodium oxyphyllum Dc was treated with 80% methanol (1 : 20) for 48 h.

        The objective of this research is to evaluate optimal conditions of permeability and selectivity on the polysulfone membrane for efficiency of separation of CH4 by checking four factors which are temperature, pressure, gas compositions and gas flow rates. When higher pressure was applied at the input, lower efficiency of recovery of CH4 and higher efficiency of separation of CH4 were shown. It has the tendency to show lower efficiency of recovery of CH4 and higher efficiency of separation of CH4 at the output as higher temperature at input. The lower flow rates make higher efficiency of recovery of CH4 and lower efficiency of separation of CH4. Finally, over 90% efficiency for CH4 separation and recovery conditions are temperature (-5℃), pressure (8 bar), gas composition rate (6:4) (CH4:CO2) and gas flow rate (5 ℓ/min). These conditions make higher separation and recovery efficiency such as 90.1% and 92.1%, respectively.

        우엉 종자를 이용하여 항산화 기능성 새싹채소를 개발하기 위하여 실험을 수행하였다. 우엉 종자는 25℃ 명조건에서 발아가 가장 왕성하였으며, 4일만에 82%가 발아하였다. 발아 종자를 암조건에서 재배한 결과, 20℃에서 길이 생장이 가장 왕성하였다. 녹화처리는 새싹의 길이생장을 억제하였으나, 자엽의 길이 및 부피생장을 촉진하였다. 수확 후 예냉하여 포장한 우엉 새싹채소는 4℃보다 10℃에서 저장할 때 저장 기간이 증가되었다. 포장용기의 통기구는 저장 기간에 영향을 주지 않았다. 0, 1, 2, 3일 녹화처리한 새싹채소의 항산화 활성을 분석한 결과, 우엉 새싹채소의 radical 소거능 및 ferrous ion chelating 효과는 시판중인 브로컬리, 적채, 완두의 새싹채소 및 콩나물보다 우수하였다. 총 폴리페놀 및 플라보노이드의 함량 또한 우수하였으며, 특히 3일 녹화시킨 우엉 새싹채소에서 가장 높은 함량을 보였다. 지질과산화 억제활성은 녹화처리가 길어질수록 우수하였다. 우엉 새싹채소는 우엉의 성체 잎줄기 보다 항산화 효과가 우수하였다. 따라서 우엉 새싹채소는 기능성 채소로써 이용가치가 높은 것으로 생각된다.

        하늘타리 (Trichosanthes kirilowii)로부터 조직배양 기법을 이용하여 약리작용을 나타내는 생리활성물질인 trichosantin을 생산하기 위한 연구로 Agrobacterium rhizogenes ATCC15834 의 감염을 통하여 기내 배양된 유묘의 잎절편으로부터 형질전환 모상근을 유도하였다. 유도된 모상근 clones으로부터 성장속도와 분지화 정도가 빠르며 단백질 합성능이 가장 우수한 TR-03 clones을 선발하였으며, 이를 사용하여 생체량과 배지내 가용성 단백질의 생산을 최대로 높일 수 있는 최적 배양조건을 조사하였다. TR-03 clone을 4% sucrose가 첨가된 MS 배지에 초기 접종농도 2 g (생중량)을 접종하여 100 rpm으로 배양시 생중량과 배지내 가용성 총단백질의 함량이 플라스크당 약 2.4 g (건중량)과 28.3ug/l으로 최대치를 나타내었다. 한편, 배지내 가용성 단백질에 대한 RIP활성 검정에서 배양 4주 후 21.3 unit로 최대치를 보였다.

        Monascus purpureus MMK2를 이용하여 황색색소 생산을 위한 배양 조건의 최적화에 관한 결과는 다음과 같다. 실험 균주가 생산하는 황색색소의 최적 배양 조건은 탄소원으로 wheat flour 3.0% 첨가, 질소원으로 0.15%, 인산염으로 의 농도가 0.25% 및 의 농도가 0.15%일 때 가장 높은 황색색소 생성을 나타내었다. 배양 온도는 , 초기 PH가 6.5 및 배양 시간 7일 일 때 황색색소의 생성능이 28.91 unit

        Scopolamine and hyoscyamine are important anticholinergic compounds obtained from Hyoscyamus niger. Adventitious roots induced from rhizome of H. niger and roots were cultured in SH medium supplemented with 3% (w/v) sucrose and 0.5 mg/L IBA. Roots were grown rapidly after 10 days of cultures. Scopolamine production was increased 7 times and hyoscyamine production was increased 12 times after 10 day of cultures. SH medium was best in root growth. But, highest scopolamine productivity was observed in WPM medium, followed White medium and best hyoscyamine productivity was resulted in MS medium. Sucrose was increased scopolamine and hyoscyamine production were increased the medium supplemented by sucrose comparing to than those by other carbon sources.

        Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. The highest growth yield was achieved in 1/2 MS medium, while the lowest growth yield was obtained in 4 MS medium. The patterns of polysaccharide formation were a little similar in all cases, but on MB5 medium, the po]ysaccharide contents of plant were higher than others. Among the modified nitrate levels, effective growth level were obtained in 1/4 N and 1/2 N. High contents of polysaccharide were obtained in 4 N. Different concentration of potassium and calcium did not improve the growth and polysaccharide production. The micropropagated shoots were successfully acclimatized artificial soils.

        A biosurfactant-producing microorganism was isolated from activated sludge by enrichment culture when grown on a minimal salt medium containing n-hexadecane as a sole carbon source. This microorganism was identified as Pseudomonas sp. and it was named Pseudomonas sp. EL-G527. It's optimal culture condition is 2% n-hexadecane, 0.2% NH4NO3, 0.3% KH2PO4, 0.3% K2HPO4, 0.02% MgSO4ㆍ7H2O, 0.0025% CaCl2ㆍ6H2O, 0.0015% FeSO4ㆍ7H2O in 1ℓ distilled water and initial pH 7.0. Cultivation was initiated with a 2% inoculum obtained from starter cultures grown in 30 ㎖ of the same medium in 250 ㎖ flask. They were cultivated at 30 ℃ in reciprocal shaking incubator and the highest biosurfactant production was observed after 4 days.
