Salivary gland dysfunction is a common complication of diabetes. Decreased saliva production and changes in saliva composition may cause oral diseases. Reactive oxygen species (ROS) generation in the salivary glands results in the loss of acinar cells and decreased saliva secretion. Glucagon-like peptide 1 (GLP-1) is the incretin hormone that regulates blood glucose level and can suppress ROS production and inflammation through its antioxidant effects. Dipeptidyl peptidase-4 (DPP-4) is an enzyme that breaks down GLP-1. In this study, we evaluated the pathological role of DPP-4 and GLP-1 on salivary gland dysfunction in type 2 diabetic db/db mice. We observed reduced salivary secretion and histopathological alteration of salivary glands in the db/db mice. The increased DPP-4 and decreased GLP-1 levels in the salivary glands were also detected in the db/db mice. Furthermore, the db/db mice had increased apoptosis and oxidative injury in salivary glands. There was an accumulation of advanced glycation end products and mucus in the salivary glands of the db/db mice. In conclusion, these results showed the possible involvement of DPP-4 and GLP-1, leading to increased ROS-induced apoptosis in diabetes-related salivary gland dysfunction. DPP-4 and GLP-1 may be a pharmacological target for patients with diabetes-related salivary gland dysfunction.
This study was designed to evaluate antihyperglycemic and antihyperlipidemic effects of ethanol extracts of Taraxacum mongolicum (T.m.) on streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley rats were randomly assigned to five groups: normal (NC), STZ-control (DC), and three experimental groups. Diabetes was induced in Sprague-Dawley rats with a single intravenous injection [45 mg/kg body weight (b.w.)] of STZ. An ethanol extract of T.m. was orally given to diabetic rats for 14 days. Three experimental groups were additionally treated with T.m. extract at doses of 1 g/kg b.w./day for T.m.-1, 2 g/kg b.w./day for T.m.-2, and 3 g/kg b.w./day for T.m.-3. Oral administration of T.m.-2 significantly increased their body weights. T.m.-1 and T.m.-2 significantly decreased aspartate aminotransferase (AST) levels than DC. T.m.-1 and T.m.-2 group significantly decreased blood glucose levels. Total cholesterol, triglycerides, and free fatty acids were significantly decreased whereas high-density lipoprotein cholesterol was significantly increased in groups treated with T.m. extract than those in the DC group. These results support the fact that administration of T.m. extract can reduce hyperglycemia and hyperlipidemia risk in diabetic rats.
Diabetic encephalopathy is a major complication with cognitive impairment and neurodegeneration in patients with type 1 or type 2 diabetes mellitus (DM). DM-induced glucolipotoxicity is a risk factor for Alzheimer’s disease–like phenotype, including amyloidogenesis, tau hyperphosphorylation, and neuronal apoptosis. Although the detailed mechanism underlying the pathogenesis of diabetic encephalopathy remains unclear, mitochondrial oxidative stress is emerging as a key factor for diabetic complications and neurodegeneration. A deeper understanding of the regulatory mechanism of mitochondrial oxidative stress under hyperglycemic conditions will provide insights into the development of therapeutic strategies for diabetic encephalopathy. Here, we review the role of mitochondrial oxidative stress in diabetic encephalopathy and the regulatory mechanisms by which high glucose induces the generation of mitochondrial reactive species oxygen species in neuronal cells. This review also summarizes the mitochondrial-dependent and -independent pathways (O-linked-N-acetylglucosaminylation, calcium, and glycogen synthase kinase 3β signaling) that regulate mitochondrial oxidative stress in a DM model.
Purpose: The purpose of this study was to analyze factors influencing diabetic retinal examination in community-dwelling older adults with diabetes, in order to provide basic data to improve screening rates of diabetic retinal examination. Methods: This study used raw data from the National Community Health Survey, 2020, which is a cross-sectional survey. The subjects of this study are 16,031 old adults aged over 65 years who had been diagnosed with diabetes. The demographic characteristics, diabetes and diseaserelated characteristics, social distancing and fear of COVID-19 according to the diabetic retinal examination were analyzed with chi-square test. Logistic regression analyses were used to examine the factors influencing diabetic retinal examination. Results: We identified that diabetic retinal examination was associated with age, residence, level of education, income, type of diabetes care, period of diabetes, and fear of COVID-19. Conclusion: Health care workers in the community should strongly recommend yearly diabetic retinal examination for diabetic patients who older adults, have lower levels of education and income. A multiple approach is required to raise the low screening rate of diabetic retinal examination, including lower the cost burden.
Declined salivary gland function is commonly observed in patients with diabetes. Advanced glycation end products (AGEs) are believed to contribute to the pathogenesis of diabetes-induces hypofunction of the salivary glands. Polydatin (resveratrol-3-O-β-mono-D-glucoside) is a polyphenol that can be easily accessed from peanut, grape, and red wines. Although polydatin is known to have anti-glycation, anti-oxidation, and anti-inflammation effects, its effect in the salivary gland is not known. In the present study, we evaluated the AGEs burden in the salivary gland in streptozotocin (STZ)-induced diabetic rats and the protective effect of polydatin on diabetes-related salivary hypofunction. Polydatin (50 and 100 mg/kg/day) was orally administered in the STZ-induced diabetic rats for 4 weeks. The results showed that the salivary flow rate of the STZ-induced diabetic rats was reduced compared with that in the normoglycemic control rats. The circulating AGEs in serum and secreted AGEs in saliva increased in the STZ-induced diabetic rats. The reactive oxygen specises (ROS) were highly generated in the salivary gland in these diabetic rats. The salivary gland from the diabetic rats showed increased acinar cell apoptosis compared to normoglycemic control mice. However, polydatin suppressed all of these diabetes-related salivary changes. Overall, polydatin could provide a beneficial option for diabetes-related salivary hypofunction.
This study was performed to investigate the antioxidant and antidiabetic activities of soybean fermented with Monascus. Also, the changes in the content of isoflavones and Monacolin K were analyzed. It was observed that the glycoside forms of daidzin and genistin were converted to aglycones of daidzein and genistein within 6 days of fermentation. The product can be used as a health functional material that can increase bioavailability. Monacolin K production was found to increase significantly with the progression of fermentation with an increase to 0.04 mg/g and 0.44 mg/g on 6 and 12 days of fermentation, respectively. The DPPH radical scavenging activity of soybean fermented with Monascus was significantly increased compared to that of soybean. The protein expression of inflammation-related genes (TNF-α, IL-6, and COX-2) in the MIN cell was significantly increased in the presence of alloxan compared to the normal group, but a decrease was observed in the presence of soybean fermented with Monascus. In conclusion, soybean fermented with Monascus showed the highest antidiabetic and antioxidant effects. These results suggest that soybean fermented with Monascus has the potential to be used as a beneficial ingredient with antidiabetic and antioxidant effects.
고중성 지방혈증은 잘 알려진 급성 췌장염의 원인 중 하나이다. 당뇨병성 케톤산증, 고중성 지방혈증 및 급성 췌장염이 연관이 있다는 것은 알려져 있으나, 이 세 가지 질환의 연관성이 어떠한 기전에 의해 발생하는지 정확히 알려지지 않았다. 보통 중성지방이 1,000 mg/dL 이상으로 상승하였을 때 급성 췌장염의 위험도가 증가한다고 알려져 있다. 그러나 본 저자들은 당뇨병성 케톤산증 환자에서 중성지방이 200 mg/dL 미만임에도 급성 췌장염이 발생한 증례를 경험하여 이를 보고하고자 한다.
The purpose of this study was to examine the effects of Atractylodes lancea (A.l.) in a dose-dependent manner on lipid levels and plasma glucose in diabetic rats induced with streptozotocin (STZ). The Sprague-Dawley rats were randomly classified into five groups: normal, STZ-control and three experimental groups [A.l.-1, diabetic treated with ethanolic extract of A.l. (1 g/kg b.w.), A.l.-2, diabetic administered with ethanolic extract of A.l. (2 g/kg b.w.), and A.l.-3, diabetic administered with ethanolic extract of A.l. (3 g/kg b.w.)]. The normal and STZ-control group were fed an AIN-93 diet and the three experimental groups were administered with A.l. extract at doses of 1, 2, and 3 g/kg b.w./day, respectively, for 14 days. The plasma glucose levels in all the experimental groups were significantly lower than the STZ-control group after 14 days of treatment. The total cholesterol of the A.l.-3 and triglyceride levels, atherogenic index (AI) of all three experimental groups were significantly lower than the STZ-control group. The ALT and AST activities at A.l.-2, A.l.-3 were significantly lower than the STZ-control group. This result that demonstrate the administration of Atractylodes lancea can reduce hyperglycemia and hyperlipidemia risk in diabetic rats.
Periodontitis is a bacteria-induced inflammatory disease associated with alveolar bone loss. Osteoclast is a macrophage-lineage cell that exhibits phagocytic activity; however, osteoclast phagocytic activity has not been demonstrated under pathological conditions. Diabetes is a pathological condition that exacerbates alveolar bone loss via periodontitis; therefore, we examined phagocytic osteoclasts in diabetic rats that had periodontitis. The rats were divided into the control (C), periodontitis (P), and diabetes with periodontitis (DP) groups. Diabetes and periodontitis were induced by streptozotocin injection and ligature of the mandibular first molars, respectively. On days 3 and 20 after the ligature, the rats were sacrificed, and osteoclasts containing inclusions were quantified by tartrate-resistant acid phosphatase staining. On day 3, there were more osteoclasts containing inclusions in the DP group than in the C group. Among inclusions, osteocyte-like cells and dense bodies were more frequently observed in the DP group than in the C group. Cytoplasm-like structures were elevated more in the DP group than in the C and P groups. However, no differences were observed on day 20. Interestingly, some osteoclasts were in contact with the osteocytes within the exposed lacunae and contained several inclusions within a large vacuole. Thus, the elevation of phagocytic osteoclasts in rats with diabetes and periodontitis provides insight into the role of osteoclast phagocytic activity under pathological conditions.
본 연구의 목적은 내당능장애를 유도한 쥐들을 대상으로 다른 운동 강도의 지구성 및 저항 성 운동을 중재하여 당뇨관련 혈액인자들에 미치는 영향을 비교 분석하는데 있다. 37주령 C57BL/6 쥐 54마리를 (1) 일반식이대조군(CO, n=9), (2) 내당능장애군(IGT, n=9), (3) 내당능장애 + VO2max 50% 지구성운동군(IGT50A, n=9), (4) 내당능장애 + VO2max 75% 지구성운동군(IGT75A, n=9), (5) 내당능 장애 + 1RM의 50% 저항성운동군(IGT50R, n=9), 그리고 (6) 내당능장애 + 1RM의 75% 저항성운동군(IGT75R, n=9)으로 분류하였다. 지구성 운동 프로그램은 동물 실험용 트레드밀을 사용하여 주5일, 1일 40분씩 8주간 트레드밀운동을 실시하였다. 저항성 운동은 주5일 8주간 사다리 저항 운동을 반복사이 2분간의 휴식을 주어 총 8번의 Climbing을 실시하였다. 운동 프로그램 후 인슐린은 통계적으로 모든 그룹간의 차이가 없었다. 공복 혈당은 대조군보다 IGT 유도군들에서 통계적으로 유의하게 높게 나타났으며, 운동군들간의 차이는 없었으나 IGT군이 IGT75R군에 비해 유의하게 높은 수치를 보였다. HOMA-IR은 대조군과 IGT군에서만 유의한 차이를 보였다. 당화혈색소 검사에서는 IGT군이 다른 모든 집단들에 비해 높은 수치를 보였으며, IGT75A군과 IGT50R군에서 유의한 차이를 보였다. 총 콜레스 테롤은 대조군이 내당능장애 유도군들과 비교하여 유의하게 낮은 수치를 보여주었다. 반면 중성지방은 그룹간의 차이가 없었다. HDL-C는 지구성 운동군들이 대조군과 IGT군에 비해 유의하게 높은 수치를 보였으나 저항성 운동군들과의 차이는 없었다. LDL-C는 대조군이 IGT군과 운동군들에 비해 유의하게 낮은 수치를 보였으며, IGT75A군은 IGT군과 IGT50R군에 비해 낮은 수치를 보였다. 결론적으로 75%의 저항성 운동은 혈당에 보다 긍정적인 영향을 주었으며, 75%의 지구성 운동은 당화혈색소와 LDL-C 의 감소에 긍정적인 영향을 미치는 것으로 나타났다. 또한 지구성 운동은 저항성 운동에 비해 HDL-C 의 증가에 보다 효과적임을 보여주었다.
본 연구는 서로 다른 운동유형이 당뇨처치 그룹의 베타 아밀로이드, BDNF 및 인지기능에 미치는 영향을 알아보기 위하여 24마리의 C57BL/6 쥐를 당뇨처치를 하지 않은 그룹 6마리(통제그룹: 6)와 당뇨처치 그룹 18마리로 무작위 할당하여, 이를 다시 당뇨처치 그룹의 경우 통제그룹 6마리, 유산소 운동그룹 6마리, 저항운동 그룹 6마리로 나누었고 운동그룹은 8주간 주 5회 저강도로 각각 트레드밀 운동과 사다리 운동을 실시하였다. 그 결과 베타아밀로이드는 8주 후 당뇨처치 그룹의 통제그룹 (DM.G.)이 나머지 세 그룹에 비하여 높은 수준을 나타내었으나 통계적으로 유의한 차이는 나타나지 않았다. BDNF의 경우는 당뇨통제그룹이 나머지 세 그룹에 비하여 낮은 수준을 나타냈으며 통계적으로 유의한 차이를 나타냈다(p<.05). 인지기능을 알아보기 위한 Y-미로 검사에서도 당뇨통제 그룹이 나머지 세그룹에 비하여 낮은 수준을 보였으며 통계적으로도 유의한 차이를 나타냈다(p<.05).
녹두(Phaseolus aureus L.)가 당뇨 유발 흰쥐에 있어서 혈당 및 지질성분, 단백질, 전해질 농 도 등에 미치는 영향을 확인하기 위하여 기본 식이를 섭취시킨 BD군, 기본 식이군에 5% 녹두 급여군 (BM군), 당뇨 유발 실험군(BS군)과 BS군에 5% 녹두를 섭취시킨 실험군(SM군)으로 나누어 실험을 수행 하였다. 혈청 지질성분(총 콜레스테롤, LDL-콜레스테롤, 중성지질, 인지질, 유리 콜레스테롤, 콜레스테롤 에스테르), 혈당 및 유리지방산(non esterified fatty acid, NEFA)의 농도와 동맥경화지수 (atherosclerotic index, AI), 심혈관 위험지수(cardiac risk factor, CRF)는 당뇨 유발군에서 여타 실험군과 비교하였을 때 유의적인 차이를 보이며 증가를 나타내었고(p<0.05), 5% 녹두 급여군에서 감소되는 것으로 나타났다. 당뇨유발군인 BS군에 비해 5% 녹두를 첨가시킨 SM군에서 고밀도 지단백 콜레스테롤 (high density lipoprotein-cholesterol, HDL-콜레스테롤) 및 총 콜레스테롤에 대한 HDL-콜레스테롤의 농도 비는 유의적인 차이를 보이며 증가를 나타내었다(p<0.05). 혈청 단백질 농도에 있어서, 당뇨 유발 군(BS군)에 5% 녹두를 급여(SM군)한 흰쥐의 혈청 알부민(albumin) 농도와 알부민/글로불린 비 (albumin/globulin ratio, A/G 비)는 증가 하는 것으로 관찰되었다. 나트륨(Na) 및 염소(Cl)는 SM군에 서 BS군 보다 농도가 감소된 것으로 확인하였다. 이상의 결과, 5% 녹두는 흰쥐의 혈당 및 지질대사 기 능 개선에 도움이 되는 것으로 판단된다.
제2형 당뇨 마우스로 8주간 50%, 75% 강도의 유산소성 및 저항성 운동을 수행하여 항 당뇨 및 지질 개선에 미치는 영향을 비교 분석하였다. 39주령 C57BL/6 마우스 48마리를 일반식이정상군 (Normal, n=8)을 제외하고, Streptozotocin (STZ)으로 당뇨를 유발하여 제2형 당뇨군(DM, n=8), 제2형 당뇨+VO2max 50% 유산소운동군(DM50A, n=8), 제2형 당뇨+VO2max 75% 유산소운동군(DM75A, n=8), 제2형 당뇨+1RM 50% 저항운동군(DM50R, n=8), 그리고 제2형 당뇨+1RM 75% 저항운동군 (DM75R, n=8)으로 구분하였다. 유산소성 운동은 주 5일, 1일 40분씩 8주간 트레드밀 운동을 실시하였으며, DM50A군은 1~4주는 8m/min, 5~8주는 8~10m/min으로, DM75A군은 1~4주는 12m/min, 5~8주는 12~14m/min으로 점증하여 실시하였다. 저항성 운동은 주5일 8주간 사다리 저항운동을 실시하였으며, DM50R군은 1RM의 50%, DM75R군은 1RM의 75%로 운동 반복 사이 2분간의 휴식을 주어 총 8번의 Climbing을 실시하였다. 8주간의 운동 후 공복 혈당은 DM군에 비해 DM운동군에서 유의적으로 낮았으 며, 인슐린은 운동에 따른 유의한 차이가 없었으나, HOMA-IR은 DM군에 비해 DM운동군이 유의하게 낮은 것으로 나타났다. HbA1c는 DM군에 비해 DM50R 및 DM75R군이 유의하게 낮은 것으로 나타났다. 총콜레스테롤과 LDL-C는 DM운동군 간의 차이가 없었으나, HDL-C는 DM75A군이 가장 높은 수준으로 나타났고, 중성지방은 DM75R군이 가장 낮게 나타났다. 심혈관 위험 지수는 Normal군 및 DM75A군이 가장 낮게 나타났다. 따라서 T2DM 마우스에서 중강도의 저항성 운동은 혈당 및 인슐린 저항성 조절에 더 나은 개선을 보여주었으며, 중강도의 유산소성 운동은 HDL-C 수준 증가를 통한 심혈관 위험 지수를 감소 하는데 효과적이었다.
This study includes the clinical features of diabetic patients with fascial space infections of head and neck and those of non-diabetic patients. We retrospectively studied 62 patients who were treated for fascial space infections of the head and neck, and compared the patients’ background, the aetiology of the infection, clinical variables, laboratory values, and treatment process. Statistical analyses of the differences between groups were made with Student's t test, the chi square test, and analysis of variance, as appropriate. Diabetic patients had more spaces involved, had longer hospital stays, and developed more complications. The main microorganism of diabetic group was Klebsiella pneumonia and otherwise, Streptococcus spp. was for non-diabetic group. We conclude that diabetic patients are more likely to develop complications and it is necessary to consider about dominant microorganism when using medication for diabetes patients
Streptozotocin (STZ, 45 mg/kg body weight)으로 유도된 Sprague Dawley (SD)계 수컷 흰쥐에 5% 울금을 섭취시켰을 때, 혈청 크레아티닌(creatinine) 및 blood urea nitrogen (BUN)의 농도와 alkaline phosphatase (ALP), lactate dehydrogenase (LDH), aminotransferase (AST, ALT), 카탈라아제(catalase), 아밀라아제(amylase) 및 리파아제(lipase) 활성에 미치는 영향을 알아보기 위해 본 실험을 수행한 결과, 기본식이를 급여시킨 대조군(control)인 정상군은 BD군, 기본식이 실험군에 5% 울금 급여군(BT군), 당뇨 유발 실험군(BS군)인 질환 모델 대조군(control-diabetic)과 질환 실험군(diabetic, BS군) 에 5% 울금을 급여시킨 실험군(ST군)으로 나누었다. 흰쥐의 혈청 크레아티닌 및 BUN 농도와 ALP, AST 및 ALT 활성은 유사한 경향으로 5% 울금의 급여가 감소시킨 것으로 확인되었다. SD계 수컷 흰쥐의 혈청 LDH 활성은 BS군에서 유의적인 차이를 나타내며 높은 활성을 보였으나(p<0.05), 5% 울금의 급여가 감소 효과를 나타내었다(p<0.05). 카탈라아제 활성은 BS군에서 유의적으로 낮은 활성을 나타내었고, ST군에서 활성을 증가시켰다(p<0.05). 혈청 아밀라아제 및 리파아제의 활성은 5% 울금의 급여가 감소시킨 것으로 확인되었다. 본 연구 결과, SD계 수컷 흰쥐를 통한 in vivo 실험에서 5% 울금의 섭취가 당뇨, 내인성 항산화 효소 및 간 기능 개선에 효과가 있는 것으로 나타났다.
Curcumin is a hydrophobic polyphenol extracted from turmeric that exhibits a variety of biological functions has albeit with limited efficacy as a functional food material owing to its low absorption when administered orally. The newly developed curcumin powder formulation exhibits improved absorption rate in vivo. This study evaluates the anti-oxidant effects of Theracurmin® (TC), which is highly bio-available in curcumin powder. The antioxidant activity of TC was investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, ferrous reducing antioxidant power (FRAP) assays, NO radical, superoxide radical, H2O2 scavenging activity, and total antioxidant capacity (TAC). Additionally, we evaluated the antioxidant activity of TC in high-fat diet (HFD)-fed streptozotocin (STZ)-induced Type 2 diabetic rats. As a result of oral administration of TC for 13 weeks in type 2 diabetic rats, the group administration of 2,000 mg/kg significantly increased FRAP, superoxide dismutase (SOD), and reduced the level of glutathione (GSH) in liver tissue 1.9, 1.2, and 1.2-times, respectively. Furthermore, serum TAC levels increased by 1.3-fold after the rats were administered with a dose of 500 mg/kg. These results were consistent with the in vitro assay results. In conclusion, TC exhibited its potential as a functional food material through its antioxidant properties.
