The purpose of this study is to investigate the physicochemical properties of jelly made from fermented red ginseng concentrate (FRGC) that can be easily absorbed and digested for the health promotion of the elderly. The pH of the jellies tended to decrease with increasing concentration of FRGC. Soluble solid content has significantly higher value when added more than 2%, and the water content of the sample was significantly lower when the FRGC was added 4%. As the amount of FRGC was increased, the total color difference increased, and the hardness of samples decreased significantly. On the other hand, the total ginsenoside contents of the FRGC was 45.50 mg/g. As the concentration of FRGC increased, the content of polyphenol and flavonoids increased. The increasing pattern of polyphenols and flavonoids showed a similar trend. As the content of FRGC increased, ABTS free radical scavenging activity significantly increased (p<0.05), and in the control, the minimum value (62.6 AEAC) and the 4% sample were highest (116.2 AEAC). DPPH radical scavenging activity was like that of ABTS radical scavenging activity. However, there was no significant difference in DPPH radical scavenging activity of 3% and 4% red ginseng jelly.
진생베리는 진세노사이드 Re을 다량함유하고 있고 항염, 항암, 혈당강하 및 미백효과가 있다. 본 연구에서는 Rhizopus Oligosporus 균주를 이용하여 진생베리 발효공정을 확립하였으며 진생베리 발효 물에 대한 화장품 약리활성을 분석하였다. 발효에 의한 인삼 베리 추출물의 전자 공여 능력은 1,000 μ g/mL 농도에서 81 %를 나타냈다. ABTS+ 라디칼 소거능 1,000 μg/mL 농도에서 100.2% 효과를 나타내었다. 피부 미백과 관련된 티로시나 아제 억제 효과는 1,000 μg /mL 농도에서 57%였다. 피부 주름과 관련된 엘라스타제 억제효과는 1,000 μg/mL 농도에서 47%였다. 또한, 콜라게나제 억제 효과는 1,000 μ g/mL 농도에서 33%였다. 이상의 결과로부터 진생베리 발효물이 항염과 미백, 주름개선 효능을 가진다고 판단된다. 따라서 진생베리 발효물이 향우 항염 및 항노화 화장품 원료로서의 이용 가능성이 매우 높을 것 으로 판단된다.
인삼열매는 진세노사이드 함량이 인삼뿌리와 다르게 나타나며, 항 고혈당효과, 항암활성 등의 다양한 약리적 효능이 있다고 보고되고 있다. 본 연구에서는 인삼열매를 수확하여 Rb1 및 Rg1 등의 진세노사이드 함량이 풍부한 4회 증포 인삼열매와 Rg3를 포함한 총 진세노사이드 함량이 풍부한 7회 증포 인삼열매를 혼합하여 열수추출물과 에탄올 추출물을 얻었고, 이를 L. plantarum으로 발효시킨 추출물을 이용하여 액상 스틱을 제조한 후 10, 25, 35oC 에서 4개월 동안 저장하면서 진세노사이드 함량 변화 및 이화학적 특성을 조사하였다. 인삼열매 추출물을 함유하는 액상스틱을 35oC에서 4개월 동안 저장한 후에 pH는 4.81에서 3.81로 저하되었으나, 산도와 고형분 함량은 변화하지 않았다. DPPH 소거능은 10oC와 25oC에서 4개월 동안 저장했을 때 큰 변화를 나타내지 않았으나 35oC에서 4개월 저장했을 때 크게 감소하였다. L값(명도)와 b값(황색도)는 저장기간 동안 감소하였으나, a값(적색도)는 변화 하지 않았다. Rg1, Rb1, F2, Rg3(S), Rg3(R), Rg5 등 6종류의 총 진세노사이드 함량은 10, 25, 35oC에서 4개월 동안 저장하는 동안에 큰 변화가 없었으며, 일반세균과 대장균군도 검출되지 않았다.
본 연구에서는 김치로부터 분리된 L. brevis SM61을 생 화학적 특성과 16s rRNA 염기서열 분석을 통해 분리 및 동정하였으며, 흰점박이꽃무지 유충 열수추출물과 산양삼 분말을 L. brevis SM61을 이용하여 48시간동안 발효를 수행하여 발효물을 제조하였다. 그리고, 흰점박이꽃무지 유충 열수추출물과 L. brevis SM61을 사용한 발효물에 대해 농도에 따른 세포 독성은 없는 것으로 판단되었다. 그리고, 흰점박이꽃무지 유충 열수추출물과 L. brevis SM61을 사용한 발효물에 대해 발효 시간에 따른 폴리페놀 함량을 분석한 결과 발효 초기에는 약 1.49 μg/mL 비해 48시간 이후 약 5.09 μg/mL 수준으로 증가하였다. 또한, 항산화능 은 흰점박이꽃무지 유충 열수추출물이 약 60%였으나, L. brevis SM61을 이용한 발효물이 약 98%로 높아졌다. 항균 활성의 경우 L. brevis SM61을 이용한 발효물에서만 항균 효과가 확인되었다. 따라서, 본 연구를 통해 분리된 신규 L. brevis SM61을 이용한 흰점박이꽃무지 유충의 열 수추출물에 대한 발효물은 식용 곤충의 기능성을 강화시 킬 수 있을 것으로 판단되며, 해당 유산균을 활용하여 다양한 식용 곤충에도 적용이 가능할 것으로 사료된다.
This study was investigating the anti-obesity effects of diets with a mixture of cheese, red ginseng and fermented mature fruit of Rubus coreanus Miquel. We fed the experimental diets (SC5, SC10) supplemented with 5% or 10% cheese containing of fermented mixture, then orally administered the extract of fermented mixture (RB100, RB300) at a concentration of 100 or 300 mg/kg body weight to SD rats with high fat diet (HFD). The results were as follows: Sample cheese groups decreased visceral fat mass and adipocyte size of stomach compared to that of HFD group. Additionally, lipid droplets of liver in sample cheese groups were smaller than that in HFD group. The serum triglyceride (TG), total cholesterol (tChol), glucose, leptin, and insulin levels in sample cheese group were lower than that in HFD group. But, the serum adiponectin and HDL-cholesterol (cHDL) levels were higher than that in HFD group. These results suggest that fermentation of red ginseng and Rubus coreanus mixed with cheese might be helpful in preventing obesity in high fat diet-fed rats.
생체 내 실험에서 발효 인삼꽃 추출물(FM), 발효하지 않은 인삼꽃 추출물(FD)과 대조군으로 생리식염수를 2주간 마우스 체중 ㎏ 당 100, 200 ㎎/㎏ B.W.의 농도로 마우스에 경구 투여한 후 LPS에 의해 활성화된 복강 대식세포가 분비하는 염증성 사이토카인 IL-6, TNF-α의 생성량을 측정하였다. 그 결과, IL-6는 발효 LPS로 자극한 경우, 두 가지 농도에서 모두 처리한 군에 비해 높은 증식능을 나타내었고, LPS로 자극한 결과, 특히 발효 인삼꽃 추출물 200 ㎎/㎏ B.W. 농도에서 유의적으로 낮은 IL-6 분비량을 보였다. TNF-α의 경우, 100 ㎎/㎏ B.W.와 200 ㎎/㎏ B.W. 두 농도 모두에서 LPS로 자극하지 않은 경우, 낮은 증식 효과를 보여주었고, 자극한 경우, 인삼꽃 시료를 첨가한 군이 대조군보다 낮은 TNF-α 분비량을 보였으며, FM에서 FD보다 더 TNF-α를 억제하는 효과가 큰 것을 볼 수 있었다. 이상의 결과에 따르면 FD의 사이토카인 IL-6, TNF-α 생성 효과는 200 ㎎/㎏ B.W. 농도 투여 시 효과적으로 면역 세포와 면역 기관의 주요 기능을 증진시킬 가능성이 있을 것으로 사료된다. 이러한 연구결과를 토대로 앞으로 인삼꽃 발효를 이용한 기능성 사료 개발과 더불어 산업적 측면에서 보다 긍정적인 효과를 얻을 수 있을 것으로 판단된다.
본 연구는 발효된 대두, 미강, 홍삼 3종을 배합을 하여 시너지 효과를 확인함으로써 개별 시료보다 더욱 더 좋은 혼합물을 만들기 위해서 시행되었다. 3가지 혼합비율 262, 622, 442를 비교하였을 때 442 비율이 항산화활성과 항노화활성이 뛰어난 것으로 나타났다. 또한 442 혼합비율은 개별 재료의 효과와 비교하였을 때 개별 홍삼보다 높은 시너지 효과를 보여 주었다. 그러므로 홍삼을 원료로 하여 화장품을 제조할 때는 홍삼을 단독으로 사용하는 것보다 미강과 대두와 혼합된 442 혼합비율 재료가 항산화활성과 주름개선에 더 높은 활성을 가지는 기능성 화장품의 중요 원료로 선호될 수 있다. 앞으로 이 물질에서 주요한 유효 성분을 분리, 정제하여 항산화 검증과 항노화 검증의 필요성이 있다고 사료된다.
The study was conducted to investigate the condition for mixed fermentation of Rhodilola sachalinensis with red ginseng using Lactobacillus acidophillus 128 and the changes of physicochemical properties and antioxidant activities before and after the lactic acid fermentation was examined. In the single fermentation of Rhodiola sachalinensis extract, the pH and titratable acidity rarely changed, and the number of lactic acid bacteria decreased greatly. On the other hand, in the lactic acid fermentation of Rhodiola sachalinensis-red ginseng mixed extract of 50% red ginseng content, the pH decreased, whereas the titratable acidity and the number of lactic acid bacteria increased. The solid content of optimal mixed extract for lactic acid fermentation was 0.5%. Sugar content decreased during fermentation, but total phenolic compounds tended to increase during fermentation. The salidroside and p-tyrosol content of the initial Rhodiola sachalinensis-red ginseng mixed extract was 419.5 ㎎% and 60.1 ㎎%, respectively; after fermentation, the salidroside content after lactic acid fermentation decreased greatly to 81.8 ㎎%, and the amount of p-tyrosol increased greatly to 324.9 ㎎%. The DPPH scavenging activity of Rhodiola sachalinensis-red ginseng mixed fermentate was 78.1% at 0.1% concentration, showing a tendency to increase as compared to 50.3% of Rhodiola sachalinensis-red ginseng mixed extract before the fermentation (p<0.05); it was a higher antioxidant activity as compared to the single fermentation of Rhodiola sachalinensis or red ginseng.
A tissue cultured wild ginseng (TCWG) suspension was inoculated with lactic acid bacteria and fermented to improve the functionality of TCWG. The utilization of TCWG was increased directly using the freeze-dried powder. The optimal ratio of TCWG powder and water for fermentation was 1:19 (5%), which was selected by measuring the fluidity and viable cell count according to concentration. The effects on ADH activation and immune cell activation by each ferments with 10 kinds of Lactobacillus sp. strains were examined. The ferments with the Lactobacillus casei KFRI 692 strain showed 5.4 times higher ADH activity and 1.3 times higher ALDH activity than the non-fermented TCWG powder (control). The level of NO production and cytotoxicity was also measured by Raw 264.7 cells. The ferment with the Lac. casei KFRI 692 strain showed the highest level of NO production and lower cytotoxicity than the others. Therefore, the Lac. casei KFRI 692 strain was selected as a strain for fermentation of a TCWG suspension to maximize its functionality. To identify the optimal fermentation time of the selected Lac. casei KFRI 692 strain on the 5% TCWG suspension, the viable cell count of lactic acid bacterial and the changes in pH were observed for 72 hours. 24-hrs was found to be the optimal fermentation time. In this way, fermented TCWG with lactic acid bacteria showed higher ADH activation efficacy and immune cell activation than non-fermented TCWG.
In this study red ginseng was extracted with ethanol and then fermented by yeasts including Lactobacillus casei and Bifidobacterium longum. Fermented red ginseng extracts(FRGE) were found to be more effective antioxidants in vitro with regards to 1, 1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging activity than red ginseng extracts(RGE). In FRGE, the contents of ginsenosides Rb1, -Rb2 and -Rc were much lower than in RGE, however, the contents of ginsenosides 20(S)-Rg3, 20(R)-Rg3 and compound K were higher than RGE. FRGE was added to Yanggaeng(0, 5, 10, 15, 20%), and physicochemical and sensory evaluations of the Yanggaeng were conducted. The L and b values of Yanggaeng with added FRGE were decreased by increasing the ratio of FRGE, while the a value was increased. Sensory evaluations for, taste, color, flavor, texture and overall acceptability of Yanggaeng with addition of FRGE (10%) were applicable for improving the Yanggaeng product.
The objective of this study was to prepare bioactive ginseng yogurts containing compound K, which is transformed from ginsenosides, and to investigate the compound’s cytotoxicity against tumor cells. Milk containng ginseng was fermented by Bifidobacteria KK-l and KK-2, and their activities for transforming ginsenosides to compound K were measured. Among the tested concentrations of ginseng in the milk, compound K was effectively produced in the 3% and 6% ginseng yogurts fermented for 48 hrs. These fermented ginseng yogurts were extracted with BuOH, and their cytotoxicities against tumor cells were examined. The BuOH extract of the yogurt made from the 3% ginseng milk showed cytotooxic activity against P388 and HeLa tumor cells. However, the nonfermented ginseng milk did not exhibit cytotoxicity against these cells. Therefore, we deem that the ginseng yogurt, which contained compound K, could be developed as a potential fermented drink product.
This study investigated the physicochemical properties of Pekin duck breast meat obtained from ducks fed diets consisting different types of sipjeondaebo-tang by-products and red ginseng marc with fermented red koji during storage. A total of 180 Pekin ducks (0-day old) were divided into four groups, each consisting of three replicates (15 ducks per pen). This study investigated diets with four types of treatments: control (basal diet),1% blend powder, pelleted 1% blend, and coated pellets of 1% blend; the blend was a mixture of sipjeondaebo-tang by-products powder and red ginseng marc with fermented red koji. There was no significant difference (p>0.05) in duck breast meat pH on storage days 3 and 7, TBARS on storage days 0 through 7, and DPPH radical scavenging on storage days 0 and 7. However, the pH values on storage day 0 and DPPH radical scavenging on storage day 3 were significantly different (p<0.05) in the meats from control and treated diet fed ducks. Especially, on storage day 7, the breast meat from ducks treated with different types of sipjeondaebo-tang by-products and red ginseng marc with fermented red koji showed lower TBARS values and increased DPPH radical scavenging activity compared to the control. In conclusion, addition of different types of sipjeondaebo-tang by-product and red ginseng marc with fermented red koji to 1% blend might be helpful in increasing antioxidant effects and reducing product wastage.
Background : Recently, wild ginseng cultured ginseng cultured in a bioreactor is mass produced using biotechnological tissue culture technology. PgTRx1 gene which is involved in the production of useful substances in fermented wild ginseng cultured root was selected and introduced into a model plant (Nicotiana benthamiana) to investigate transformation useful gene expression and possible production of useful substances.
Methods and Results : The PgTRx1 gene was amplified and isolated from fermented wild ginseng cultured root. Isolated PgTRx1 gene was ligated to the plant expression vector pMBP1. Overexpression genes were recombined and cloned into E. coli. Agrobacterium tumefaciens LBA4404 was transformed, cultured A. tumefaciens LBA4404 was agro-infiltrated into a model plant for transient assay. Agro-infiltration model plants were sampled on days 0, 1, 2, and 3, and cDNA synthesis was performed after total RNA extraction. The expression level of PgTRx1 gene increased with time, and NbNR, NbHSR, NbAPx, NbSIP, NbPAL, NbPR1a and NbNOA1 genes showed a decrease in the expression level. The samples were taken to determine antioxidant activity, acetylcholine hydrolase inhibitory activity and glutamate content at 0 h, 12 h, 14 h, and 36 h. The highest antioxidant activity was observed at 24 h of sample, acetylcholine hydrolase inhibitory activity at 12 h, and glutamate at 36 h.
Conclusion : The possibility of introducing the model plant of the PgTRx1 gene derived from fermented wild ginseng cultured root was confirmed. The results showed that various activities were increased with time of agro-infiltration.
This study was conducted to investigate the effects of dietary supplementation of various types of fermented red ginseng marc with red koji to laying hens on eggshell and egg yolk quality characteristics. A total of 240 Hy-line Brown laying hens (40 wk of age) were randomly allotted to 24 pens (6 replicates per treatment and 10 laying hens per replicate). Experimental diets consisted of 4 treatments containing basal diet (control), 1% fermented red ginseng marc powder with red koji, 1% fermented red ginseng marc pelleted with red koji and 1% fermented red ginseng marc coated with red koji. During the 8-wk feeding trial, there were no significant differences in eggshell strength, eggshell thickness and eggshell color among the treatments, except for eggshell strength at 4 and 8 wk and eggshell thickness at 0 wk. In addition, no differences in egg yolk color and egg yolk index were found for all treatments throughout the 8-wk feeding period, except for egg yolk color at 0 wk. Thus, using various types of fermented red ginseng marc with red koji to laying hens did not improve eggshell and egg yolk quality characteristics.
Background : The Production of high quality ginseng seedling become increasingly important in the cultivation of ginseng because the quality of ginseng seedling is closely related to yield of ginseng. However, it has become difficult to produce high quality ginseng seedling due to environmental instability such as an increase in the occurrence of abnormal climate and pests, and the difficulty in obtain new place to cultivate ginseng seedling. This study was carried out to analyze the growth characteristics of ginseng seedling according to application of fermented compost during ginseng seedling cultivation Methods and Results : In order to evaluate the effect of fermented compost on the growth of ginseng seedling, fermented compost containing antagonistic microorganism was prepared and applied 300kg per 10a to planned place for ginseng cultivation. After seeding of the ginseng seeds, the growth of ginseng was analysed. The pH, EC, organic matter, P2O5 and NO3-N of the compost treated group were 6.1, 2.75 ds/m, 18.97 g/㎏, 1,202 ㎎/㎏ and 287.4 ㎎/㎏, respectively, while those of non-treated group were 6.6, 0.25 ds/m, 14.67 g/㎏, 1,055 ㎎/㎏ and 8.77 ㎎/㎏, respectively. As a result of analyzing the growth characteristics of ginseng seedling, the growth of aboveground part was not significantly different between the treated and non-treated group. The root length and root diameter were 13.0 ± 1.50 ㎝ and 5.5 ± 0.27 ㎜, respectively, which were not significantly different from non-treated group. However, root weight was 1.20 ± 0.14 g, which is 1.4 times higher than that of control. Conclusion : It was confirmed that the quality of ginseng seedling was improved by compost application. These results suggested that fermented compost could be used as a material for nutrients management in ginseng seedling cultivation.
Background : Recently, ginseng (Panax ginseng C.A. Meyer.) berry has been used as a health-promoting supplements. Also, Mulberries (Morus alba L.) fruit have been used in traditional herbal medicine to treat and prevent diabetes. In this study, we measured the cytotoxicity after fermentation of the extracts in Panax Ginseng Berry and Mulberry Fruit. Methods and Results : The extracts were prepared by decoction for 3 hours in distilled water (100 g/L). The dried extract was then dissolved in phosphate-buffered saline (PBS) in preparation for use. Cell viability was examined by an MTT assay. RAW 264.7 cells were seeded at 1 × 104/mL densities in 96-well plates. Each grouping had a non-treated group as the control. The extracts were added to each well and incubated for 24 hours at 37°C and 5% CO2. The MTT solutions (5 mg/mL) were added to each well, and the cells were cultured for another 2 hours. The supernatant was then discarded, and 100 μL of dimethyl sulfoxide was added to each well. The optical density was read at 540 nm. Conclusion : Probiotics and prebiotics modulate the composition of human and domestic animal gut microbiota. The beneficial effects may result from suppression of harmful microorganisms or stimulation of organisms which contribute in a positive way to the nutrition and health of human and domestic animal. Recently, fermentation using microorganisms for the production of more effective compounds has been extensively studied. In particular, the novel pharmacological effects of a new compound generated by fermentation have been reported. Some previous studies have demonstrated that Fermented herbal medicine extract showed better bioactivity than normal herbal Plants extract when used at the same concentration.
방사선에 의해서 유도된 간 손상에 대한 백삼과 발효인삼추출물의 보호효과를 비교 연구하였다. ICR계 생쥐에게 코발트-60 감마선의 5Gy조사 7일 전부터 백삼과 발효인삼추출물(150㎎/㎏/day)을 각각 투여하였 다. 대조군은 생리적 식염수를 투여하고 방사선조사군은 생리적 식염수를 투여하면서 5Gy를 조사하였다. 그리고 각각의 실험군에서 간조직의 H2O2, catalase, MDA를 측정하였다. 그 결과 방사선조사군과 보다 백 삼과 발효인삼추출물 투여군에서 catalase함량이 유의성 있게 증가하여 간의 보호효과가 있었으며 H2O2와 MDA함량도 유의성 있게 감소하였다. 백삼과 발효인삼이 간 조직에 대한 방사선조사로 부터 매우 우수한 방호제라고 할 수가 있다.
