        This study evaluated the immunogenicity of the Bacillus Calmette-Guérin (BCG) vaccine in a guinea pig model to refine preclinical assessment methods. 24 guinea pigs were divided into four groups for immunohistochemical, histopathological, and molecular analyses, including qRT-PCR and ELISA. The ELISA results revealed significant elevations in interleukin 2 (IL-2), interferon-gamma (IFN- ), and tuberculosis-specific antibodies in vaccinated guinea pigs, particularly γ notable after 6 weeks. Although lung cytokine levels remained unchanged, spleen gene expression showed significant differences in interleukin-17, interleukin-12, interleukin-1β, and C-X-C motif chemokine ligand 10 after 6 weeks. Immunohistochemistry revealed peak IL-2 expression at 8 weeks and significant IFN-γ and TNF-α expression at 6 weeks. This study confirmed the effectiveness of BCG vaccine in guinea pigs, providing crucial insights for future tuberculosis vaccine development and standardizing immune response indicators.

        Severe Fever with Thrombocytopenia Syndrome (SFTS) is a newly emerging tick-borne disease caused by the SFTS virus (SFTSV), which belongs to the phlebovirus in the Bunyaviridae family. SFTSV is enveloped with a tripartite ambisense RNA genome. The L segment encodes the viral RNA-dependent RNA polymerase, the M segment encodes the two glycoproteins, Gc and Gn, and the S segment encodes the nucleoprotein (NP) and the nonstructural protein (NSs). NP participates in ribonucleoprotein (RNP) packaging and commonly detected early after infection, suggesting that the N protein is possible to be used as a target antigen for early diagnosis of SFTSV infection. In this study, we analyzed a highly immunogenic multi-epitope using GnGc and NP genes from a consensus sequence of SFTSV strain isolated from infected patients in Korea. The selected genes are constructed to the expression vector plasmid pJHL65 and the recombinant plasmid vector was transformed into the Δasd Δlon ΔcpxR Salmonella Typhimurium attenuated strain JOL912 and the expression of these antigens was verified by immunoblotting assay. We observed the significant levels of systemic IgG and mucosal IgM responses against the JOL912-derived antigen in the immunized BALB/c mice. The level of CD3+CD4+, CD3+CD8+ T lymphocyte subpopulation and TNF-α were also highly regulated in splenic T cells re-stimulated in vitro with NP and Gn/Gc multi-epitope selected antigens. Therefore, immunized mice with NP and Gn/GC multi-epitope recombinant proteins of attenuated Salmonella delivery system elicited T cell-related immune response, inducing an effective immune response. In conclusion, the attenuated Salmonella expressing NP-GnGc multi-epitopes could be a novel vaccine candidate against the SFTS virus.

        The optimization of a prevention program utilizing a novel trivalent inactivated Salmonella bacterial vaccine to protect poultry from Salmonella infections was evaluated in this study. A total of 50 brown nick layers were divided into 5 groups, A to E, each containing 10 hens. Group B hens were immunized with the SG9R vaccine, group C chickens were immunized with the trivalent inactivated Salmonella bacterial vaccine, and group D hens were primed with SG9R and boosted with the trivalent inactivated Salmonella bacterial vaccine. Group E hens were injected with sterile PBS. All hens in groups B to E were orally challenged with a mixture of wild-type S. Enteritidis, S. Typhimurium, and S. Gallinarum (approximately 6×10⁹ CFU/ 0.2 ml/bird). Serum IgG titers, CD3+CD4+ T-cell levels, and CD3+CD8+ T-cell levels in group D were significantly higher than those in group A. Additionally, all animals in groups A to D showed no clinical symptoms and survived after the virulent challenges, whereas all chickens in group E died following the challenge. The challenge strains of S. Enteritidis, S. Typhimurium, and S. Gallinarum were not isolated from the liver, spleen, cecum, and cloaca of group D chickens. These findings indicate that priming with SG9R and boosting with a trivalent inactivated Salmonella bacterial vaccine can be an effective approach for preventing Salmonella infections by inducing robust protective humoral and cellular immune responses in chickens.

        Foot-and-mouth disease (FMD), which affects cloven-hoofed animals, is economically important because of its highly contagious nature. FMD virus (FMDV), the causative agent of FMD, involves seven serotypes (O, A, Asia1, C, and SAT 1-3). Serotype Asia1 is unique to the Asian territory and is subdivided into nine genetic groups (G-I-IX) based on nucleotide variations in the VP1 sequence. Asia1 Shamir, the most representative Asia1 vaccine, is not highly protective against the Asia1/MOG/05 (G-V) lineage found in North Korea in 2007. Therefore, we investigated whether a chimeric virus strain (Asia1/MOG/Shamir), in which the VP4, VP2, and VP3 sequences of Asia1/MOG/05 were combined with the VP1 sequence of Asia1 Shamir, can simultaneously protect against both viruses. We determined the optimal viral growth conditions for the commercial utilization of this chimeric virus strain. Of the three types of cell culture media, the Cellvento medium resulted in the highest amount of antigen in the samples. The chimeric strain was proliferated in a small bioreactor to produce a test vaccine, and its immunogenicity was evaluated in pigs. The virus neutralization (VN) titer against the Asia1 Shamir virus was > 1/100 after the second immunization with the chimeric vaccine in pigs. In addition, a single dose of the test vaccine resulted in a VN titer of > 1/100 against the Asia1/MOG/05 strain. Taken together, our chimeric vaccine strain provided sufficient protection against the Asia1/MOG/05 and Asia1 Shamir viruses, suggesting its potential as a novel vaccine for both these strains.

        The purpose of this study is to review the discourse on vaccination from a critical perspective by considering coronavirus disease 2019 (COVID-19) news on web portals to be practical arguments advocating for certain actions. For this purpose, this study analyzed the argument patterns of the discourse on vaccination with the keyword COVID-19 vaccine side effects and examined discourse characteristics highlighted by media reports to evaluate their meaning. Contrasting patterns were observed between pro- and anti-vaccine arguments, which consisted of the necessity of vaccination and medical evidence and which focused on personal choice and vaccine side effects, respectively. The characteristics of the political discourse were observed in the contest between these arguments. Rhetorical phrases, which are often used in the political discourse, represented the misleading arguments that lacked evidential accuracy and argumentative validity by maximizing fear, instead of alleviating fear about a health crisis with scientific information and discussion. As fake news trending on social media was introduced to online news portals, which are regarded as public discourse platforms, and undermined trust in the public discourse, it served as an opportunity to politicize the discourse on vaccination.

        This study evaluated the effect of a combination of acetaminophen (AAP), vitamin C (VC) and thioctic acid (TA) on reducing serum cortisol concentrations in mice with foot-and-mouth disease (FMD) vaccination. For 5 days from 3 days before FMD vaccination to 2 days after vaccination, mice were orally administered with AAP 600 mg/kg feed (n=25, AAPT), combination of AAP 600 mg and VC 200 mg per kg feed (n=25, AVCT) and combination of AAP 600 mg, VC 200 mg and TA 20 mg per kg feed (n=25, AVTT), respectively. From day 1 to day 7 after FMD vaccination, the cortisol concentration of all groups treated with the drug was significantly decreased compared to that of the positive control group with FMD vaccination (p<0.05). In conclusion, the combined treatment of AAP, VC and TA was the most effective in relieving stress from FMD vaccination compared to the single treatment.

        This study evaluated whether fevernill, which is primarily composed of acetaminophen, vitamin C, and anhydrous citric acid, is effective in relieving stress caused by foot-and-mouth disease virus (FMDV) vaccination in calves. Three-month-old calves from a farm in Korea were randomly assigned to one of three groups (n = 10 per group: control [untreated], group A [FMDV vaccination], and group B [FMDV vaccination + addition of fevernill 0.2% to feed for five days after vaccination]). Body weights, antibody formation, clinical symptoms, serum parameters were measured in the 14 days following vaccination. Total weight gain and average daily gain during the experiment period were group A (12.00±1.15 kg, 0.86±0.08 kg/day) compared to group B (13.57±0.98 kg, 0.97±0.07 kg/day) was significantly improved (p<0.05). There was no significant difference in the formation of antibodies against the FMDV vaccine between group A and group B, though the antibody value of group B tended to be higher than that of group A. The pre-inflammatory cytokine TNF-α value was 74.47±19.26 pg/ml in group A and 59.05±11.88 pg/ml in group B on the 5th day of the experiment, which was significantly lower in group B than in group A (p<0.05). Also, cortisol concentrations were significantly lower in group B than in group A (p<0.05). In conclusion, the feeding of fevernil is judged to be helpful in mitigating the side effects caused by FMD vaccination, and thus it is thought to be able to prevent the decrease in productivity caused by vaccination.

        Long-term non-surgical contraceptive methods for cats, especially community cats, are of global interest for cost-effective and humane reasons. This study aimed to investigate the effectiveness of a gonadotropin-releasing hormone (GnRH)-based vaccine for immunocontraception and to confirm its safety in intact female cats. Recombinant Salmonella typhimurium flagellin fljB (STF2)-GnRH protein was expressed in Escherichia coli. We divided female cats into vehicle control group (n = 4) and two experimental groups (100 μg injection group [n = 7] and 1000 μg injection group [n=7]), and immunized them twice intramuscularly (0.2 mL/cat at zero week and 4 weeks later into the other leg). Breeding trials started on day 120. All control cats (n = 4/4), 71% of the 100 μg injection group (n = 5/7), and 57% of the 1000 μg injection group (n = 4/7) became pregnant within 203 days after the introduction of male cats. The 1000 μg injection group had significantly a longer median time to conception following treatment (166 days) than the control (17 days, p < 0.05). Average litter size was significantly lower in the 1000 μg GnRH-vaccinated cats (2.8 ± 0.7) than in the control cats (4.5 ± 0.5, p < 0.05). Injection site reactions were not observed in any cat. The E. coli-expressed STF2-GnRH vaccine did not provide contraception in a sufficient proportion of the cats. However, it might be effective to suppress fertility through infertility vaccines before inducing permanent infertility through the trap-neuter-return.

        This study compared the immune responses, stress relief and weight gains of needle or needle-free intramuscular and needle-free intradermal vaccination in pigs. When the same amount of a foot-and-mouth disease (FMD) vaccine was administered to pigs, antibody titers at 4 weeks after the 1st and 2nd FMD vaccination were not significantly different between the needle (IM-S) and needle-free (NM-P250) intramuscularly vaccinated groups, but the weight gain of NM-P250 was significantly increased compared to that of IM-S at 8 weeks after the 2nd FMD vaccination (p<0.05). In addition, serum cortisol concentrations of NM-P250 were considerably decreased compared to those of IM-S on the 5th and 7th day after the 1st and 2nd FMD vaccination (p<0.05). However, the antibody titers of IM-S vaccinated with 2 mL of FMD vaccine were significantly increased compared to those of the needle-free intradermal vaccinated group with 0.5 mL of FMD vaccine at 4 weeks after the 1st and 2nd FMD vaccination (p<0.05). In conclusion, the needle-free intramuscular injection for the FMD vaccination can be chosen for weight gain and stress relief in pigs.

        We conducted diagnostic investigations to analyze the causes of abortions (46 cases, 65.7%), deaths (22 cases, 31.4%) and muscular lesions (2 cases, 2.9%) occurred after foot and mouth disease (FMD) vaccination in livestock farms in Korea. Bacterial culture, enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) were performed to detect the causative agents of abortion in bovine and caprine. The diagnostic results showed that 36 (51.4%) cases, referring as “Identified”, were occurred by influence of underlying disease including bovine viral diarrhea (12 cases, 17.1%), neosporosis (7 cases, 10.0%), septicemic colibacillosis (5 cases, 7.1%), Q fever (4 cases, 5.7%) and other abnormal conditions (8 cases, 11.4%) not by vaccination. Other 2 (3.0%) cases were suspected to be vaccine-associated adverse reaction on the basis of pathological findings (shock lung, oil-component-induced granuloma) and clinical symptoms (dyspnea with pulmonary edema). The other 32 (45.7%) cases were determined “Unknown” because any pathogens and pathological changes were not identified. However, many of the “Unknown” cases were presumptive to be the vaccine-related adverse reaction based on epidemiological investigation, especially, the cases which showed the clinical signs within 2 days after the vaccination. It is important to conduct pathological, microbilogical and epidemiological investigation to diagnose whether the cases are from vaccine-associated adverse reaction or not.

        The abnormal meat could caused by abscess, fibrosis and granuloma in the region of the neck by the adverse effect of foot-and-mouth disease (FMD) vaccination practice for Korean swine. After FMD vaccination was implemented in 2010, the number of incidences of abnormal meat has been increasing. In this study, it was evaluated whether needless intradermal injection (NII) of FMD vaccine reduces abnormal meat at the injection site in comparison to conventional intramuscular injection (IM). In addition, the onset of humoral immunity was analyzed in order to see if NII of this vaccine is able to mount comparable antibody response with IM. This study was carried out in four Korean pig farms. The O-type FMD vaccine was implemented via NII at three farms and the control group was vaccinated via IM. Antibody titers were compared from the serum samples taken from 10 random pigs at age of 8, 12, 16 and 22~24 weeks of age. The O+A type FMD vaccine was also compared with the same protocol. When test animals were slaughtered, incidence, type and size of abnormal meat were recorded. The results of this study showed that the antibody titers between intradermal and intramuscular group were similar but a significant difference in the incidence of abnormal meat formation was observed between two groups (p < 0.05). In summary, needless intradermal injection of FMD vaccination was proven to be effective for the onset of humoral immunity and reducing abnormal meat formation.

        Fowl adenovirus serotype-4 (FAdV-4) infection, also known as hydropericardium-hepatitis syndrome (HHS), is one of the most prevalent diseases in the Korean poultry industry. Therefore, an effective vaccination against FAdV-4 should be developed for prevention of HHS infection. However, a standard animal model with significant pathologic characteristics has not been established for evaluation of HHS vaccine efficacy. In this study, we comparatively evaluated FAdV-4 pathogenicity from specific- pathogen-free (SPF) chickens using FAdV-4 strain (ADL091024) isolated in laying chickens in Korea according to chicken age, passage number and virus titers. In brief, FAdV-4 passaged four times in yolk sac of SPF eggs with the highest viral titers could induce the most severe clinical signs in 1-day-old chickens, similar to natural HHS outbreaks. Furthermore, the efficacy of a newly developed attenuated live FAdV-4 vaccine candidate was successfully determined using the established 1-day-old SPF chickens. There was no significant pathogenicity based on the criteria such as mortality, body weight, gross lesions, histological lesions and virus detection rates in the vaccination and non-challenge control groups. However, the challenge group without vaccination showed significant pathogenicity including hepatic necrosis, histological lesion scores in the liver and heart, virus replication and decreased body weight gain. In conclusion, the chicken challenge model established in this report was proposed for consistent and reliable evaluation of FAdV- 4 vaccine efficacy.

        Since the 2010 foot and mouth (FMD) epidemic, the Korean government has applied a FMD vaccination to pigs. A FMD vaccine is injected to pigs by intramuscular (IM) route. One of the drawbacks in FMD vaccine IM injection is that it would result in an abnormal meat on the injection spot. An abnormal meat due to FMD vaccine IM injection would cause economic loss to pig farmers. An intradermal (ID) injection would be an alternative method for FMD vaccination. The goal of current study was to compare the antibody formation rate between FMD vaccine IM injection and ID injection. The antibody formation rate was measured by the FMD serotype O vaccination percent inhibition (PI) value. In total 350 pigs (175 for FMD vaccine IM injection and 175 for FMD vaccine ID injection) were included in the study. In results, the PI values of FMD vaccine IM injection group were significantly higher than it of FMD vaccine ID injection group. However, the proportions of pigs with PI value was higher than 50, which is a legislative requirement for marketing pigs, for both FMD vaccine IM and ID injection groups at week 20 or 23 were not significantly different. The current results indicated that a FMD vaccine ID injection could be an alternative method of IM injection.

        Fucoidan(후코이단)은 주로 갈조류에서 추출되는 fucose를 함유한 함황 다당류의 일종으로, 항 균, 항바이러스 및 항종양 효과와 함께 다양한 경로로 면역력을 향상시키는 생리 기능성물질 로 알려져 있다. 최근 연구에 따르면, 인체 백신 분야에서는 fucoidan의 백신 adjuvant(항원보조 제)로서의 가능성이 제시되었다. 수산업 분야에서는, 보조사료로서의 fucoidan의 기능에 관한 연구는 보고되고 있으나, 수산용 백신 개발을 위한 adjuvant 연구는 전무한 실정이다. 동물세포 에서 fucoidan의 adjuvant에 대한 긍정적인 검토와 함께 안전성을 증명한 연구는 많이 있지만, fucoidan을 어류 백신용 adjuvant로 사용하기 위해서는 어류에서도 이를 확인할 필요가 있다. 또한 fucoidan의 분자량에 따라 세포 내 흡수율이 각기 다르다는 점과 병원체의 인위감염에 따 른 항체 생성을 포함한 어류의 특이면역 반응 시스템에 대한 연구가 많이 부족하다는 제약이 있다. 따라서 이러한 분야에 대한 적극적인 연구가 뒷받침 된다면 안전하고 효과적인 adjuvant 로 사용할 수 있을 것이다. 본 연구에서는 fucoidan이 사람과 동물을 포함하여 어류의 면역자극 즉 체액성 및 세포성 면역에 미치는 영향에 대한 연구를 검토하고, 수산업 분야에서 fucoidan 의 사용과 어류 백신용 adjuvant로서의 가능성을 고찰하였다.

        The virus-like particle (VLP) is similar to a pathogenic virus and has a high immunogenicity. However, in the selection of various structural proteins to form VLP, all expression systems commonly consume most of the time and suffer from various difficulties. Therefore, there is a need for a system that can rapidly determine the structural proteins required for effective VLP production. This study aims to construct a transient expression platform using insect cells to solve this problem. Plasmid-based expression vectors and baculovirus-inducible expression vectors were constructed. The vectors were evaluated for overexpression using EGFP. We also confirmed the formation of virus like particles through overexpression of virus structural proteins.

        This study evaluated the effect of reduced glutathione (GSH) for the reduction of stress and inflammatory response in calves inoculated with foot-and-mouth disease (FMD) vaccine. Twenty-five calves were divided into five groups of 5 calves. The negative control (NC) did not receive any vaccination or drug treatment. The positive control (PC), GSH-25, GSH-50 and GSH-100 were intramuscularly injected with GSH at concentrations of 0, 25, 50 and 100 mg / 10 kg body weight (BW), respectively, for 3 days after FMD vaccination. On day 3, 5 and 7 post-treatment, the serum cortisol and tumor necrosis factor- α (TNF-α) levels in GSH-50 and GSH-100 were significantly decreased compared with those in PC (p < 0.05). However, there was no significant difference in the serum cortisol and TNF-α levels between GSH-100 and NC 3 and 5 days post-treatment, and between GSH-50, GSH-100, and NC 7 days post-treatment. The results from this study suggest that treatment of 50 mg / 10 kg BW GSH for 3 days is useful for the reduction of stress and inflammatory response caused by FMD vaccination in calves.
