        This study was determined optimal condition as culture medium, pH, carbohydrate source and growth regulators through axillary buds cultures of Salix gracilistyla. S. gracilistyla shoots were obtained from axillary bud on 1/2MS basal medium after 2 weeks of culture. Highest shoot growth was showed in the MS medium(GI= 2.43). Shoot growth was highest in 9% sucrose compared to other concentration. BAP affects axillary shoot multiplication in of S. gracilistyla. Among nutrients, Ca2+ was not affected shoot number of S. gracilistyla. However, NH4+, NO3 -, and K+ is greatly influenced shoot number. However, Ca2+ was affected in shoot length of S. gracilistyla. Feeding of Mg2+ was not affected in shoot length. Root number is greatly influenced by NH4 +, NO3 -, and K+, however NH4 + and K+ was not affected root number of S. gracilistyla. NH4 + was affected root length, however Mg4 + was not affected root length. Treatment of 0.1mg/L IBA promoted in vitro rooting. In vitro rooted plantlets were successfully transferred to pots for 4weeks hardening process. Later, these plants were transferred to soil with above 90% survival rate in BS+SD(2:1). These results could be easily adopted for commercial large scale cultivation.

        The aim of this study was to evaluate key properties of the prickly pear cactus (Opuntia ficus-indica var. saboten (OFI) ie, levels of key chemicals (carotenoids, flavonoids and polyphenolic compounds as well as phenolic acid) and its antioxidative potential, depending on where the plant had been cultivated in Korea. The levels of flavonoids and polyphenolic compounds in OFI were 55.45~65.32 mg (+)-catechin/g and 149.00~181.15 mg gallic acid/g, respectively. Protocatechuic acid was the most abundant phenolic acid in the ON1 and ON2 (161.90 and 196.25 μg/g DW (dry weight)). Nineteen flavonoids were identified and analyzed by LC-ESI-MS in cladodes from OFI. Narcissin was the most abundant flavonoid in all of the samples (1,241.89~1,775.10 μg/g DW). Capxanthin and zeaxanthin were the most abundant carotenoids in OFI (64.88~128.08 and 48.10~93.82 μg/g DW). The level of DPPH radical and ABTS radical scavenging activities in OFI were 10.78~25.35 and 16.85~34.16 mg AA eq/100 g, respectively. OFI by cultivar has different kind of phenolic acid, flavonoids, and carotenoids. Therefore, dietary intake of cladodes from OFI may be helpful for improving human health

        This study was conducted to investigate changes in the proximate composition, antioxidant activities, and α-glucosidase inhibitory activity of Opuntia ficus-indica (OFI) cladodes cultivated in Jeju (JJ1, JJ2, JJ3) and Jeonnam (JN1, JN2). The difference in the proximate composition (crude protein, lipid and ash content) of OFI between the two regions was not significant. Ca, Mg and Na were the major mineral components of OFI. The ascorbic acid content of OFI ranged from 57.87 to 143.72 mg/100 g. A 70% ethanol extract was used to investigate the antioxidant content and activity as well as the α-glucosidase inhibitory activity. The total polyphenol and flavonoid contents of OFI were 38.69~55.29 and 3.33~4.03 mg/g, respectively. The antioxidant activities based on the DPPH and ABTS free radical scavenging assays were 45.19~ 61.52% and 39.15~51.96%, respectively, at a concentration of 1 mg/mL. The inhibitory activity of OFI extracts against rat intestinal α-glucosidase was 29.72~45.73% at 1 mg/mL concentration, and JN1 showed the highest α-glucosidase inhibitory activity. This information could be very useful for authentication of Opuntia species with the highest potential as sources of nutritional and therapeutic elements.

        This study aimed to investigate the concentration of vitamin C in Momordica charantia (MC) by cultivar, harvest time, and maturity. The methods for determining vitamin C levels were validated by measuring their linearity, specificity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy using HPLC. Results showed high linearity in the calibration curve, with a coefficient of correlation (R2) of 0.9994. The LOD and LOQ values for vitamin C were 0.05 and 0.16 μg/mL, respectively. The relative standard deviations (RSDs) for intra- and inter-day precision of vitamin C measurements were 2.34 and 1.34%, respectively. Depending on cultivar, the concentration of vitamin C in MC varied from 20.75~107.31 mg/100 g, fresh weight, with an average level 68.85±25.57 mg/100 g, FW. When MC was analyzed by harvest time, the 20150612 MC showed the highest amount of vitamin C (113.20±1.89 mg/100 g, FW). On the other hand, the highest vitamin C content by maturity was 48.59±0.87 mg/100 g, FW (15 day old MC). This information on the comparative vitamin C levels of MC might be useful to food scientists and should be explored for functional food development.

        Most previous studies on dinoflagellates in Korean coastal areas were conducted without morphological descriptions and illustrations of the observed dinoflagellates. This indicates that the species and diversity of dinoflagellates may have been respectively misidentified and underestimated in the past, probably due to cell shrinkage, distortion and loss caused by sample fixation. This study provides information on the morphological observations of four dinoflagellate orders (Prorocentrales, Dinophysiales, Gonyaulacales and Gymnodiniales) from Jangmok Harbour in Jinhae Bay, Korea. The unfixed samples were collected weekly from December 2013 to February 2015. A total of 13 genera and 30 species were identified using light and scanning electron microscopy, although some samples were not clarified at the species level. Harmful dinoflagellates, Prorocentrum donghaiense, Tripos furca, Alexandrium affine, A. fundyense, Akashiwo sanguinea and Cochlodinium polykrikoides, were identified based on the morphological observations. The results also reflect the occurrence and identification of dinoflagellates that had not been previously recorded in Jangmok Harbour.

        The aim of this study was to investigate the method validation for the determination of loganin content in Cornus officinalis (CO). This medicinal plant reportedly mainly included loganin. The specificity, linearity, precision, accuracy, limit of detection (LOD), and quantification (LOQ) were measured by HPLC/DAD. Our results showed that high linearity in the coefficient of calibration correlation (R2) for loganin standard was 1. LOD and LOQ for loganin were 0.007 and 0.026 mg/mL, respectively. The recovery rate of loganin was revealed to be in the high range of 95.14-105.45%. The relative standard deviation of intra- and inter-day precision of loganin in CO was 1.8% and 2.3%, respectively. The loganin content of CO from Gurye, Uiseng, Ichoen, and China were 7.81, 3.41, 7.42, and 6.52 mg/g, respectively. In conclusion, these methods were validated for the detection of loganin in CO.

        This study was performed to investigate the content of carotenoid (α-carotene, β-carotene, β-cryptoxanthin, capxanthin, lutein, lycopene, zeaxanthin) and physicochemical characteristics (size, color, dietary fiber, total tannin content) of unripen and ripen persimmons (Diospyros kaki) by seven cultivars. The length of the unripen and ripen persimmons ranged from 40.87-64.75 and 48.80-80.29 cm, the width were from 44.03 to 66.73 and 57.65 to 91.53 cm, respectively. Ripen persimmons were showed in the highest Hunter’s L, a and b values. Total tannin content were resulted of unripen- and ripen persimmons ranged from 7.58 to 19.80 and 2.36 to 5.85 mg/g. Total carotenoid content were from 1.09 to 6.45 and 7.46 to 29.46 μg/g, respectively. From the above results, unripen- and ripen persimmon fruit could be useful for the health functional food.

        This study performed to investigate the changes in S-allyl-L-cysteine (SAC) content of garlic with different cooking methods. Methods for determining SAC was validated by determining specificity, linearity, limit of detection (LOD), limit of quantification (LOQ), precision and accuracy using HPLC-FLD system. Results showed high linearity in the calibration curve with a coefficient of correaltion (R 2 ) of 0.9999. The LOD and LOQ values for SAC were 0.15 and 0.49 μg/mL, respectively. The relative standard deviations (RSDs) for intra- and inter-day precision of SAC were less than 5%. The recovery rate of SAC was in the range from 97.35% to 97.47%. The SAC content of raw garlic was 2.77 mg/g, and there are no significantly difference among blanching and microwave treatment in SAC content. However, SAC content of boiling, autoclaving at 110℃ and 121℃ increased in the range of 3.50~9.19 mg/g, 6.52~16.21 mg/g and 14.15~50.24 mg/g as increasing cooking temperature and time, respectively.

        Biological control has been tried for integrated pest management. It is often comparable, safe, and environment-friendly, making itself an alternative for chemical agents. Filamentous microorganisms, i.e., fungi and streptomystes, produce many kinds of useful metabolites, and some of them have been developed as a biocontrol agent. However, they still have a long way because of the concern of manufacturing cost. Therefore, process development was intensively studied to meet cost-effectiveness. Operating conditions of bioreactor, e.g., agitation and aeration, had an effect on biological and physiological responses such as mycelial morphology, oxygen and nutrient transfer. Understanding relationship between operating parameters and microbial responses in terms of growth, substrate and oxygen consumption, and production yield was critical for process development. This study dedicated to build strategies for mass production of biological control agent using aerobic filamentous microorganisms.

        This study examined the effects of learner-initiated noticing through comparison and teacher-initiated noticing through grammar instruction on EFL middle school learners' English writing process and uptake. For the study, 18 second-year middle school students with intermediate-high English proficiency were divided into three groups. After the first writing composition, one group (Group C) underwent a comparison stage, in which they compared their original writings and reformulated versions. Another group (Group I) received reactive grammar instruction on the four most frequent types of grammar errors, while the other group (Group IC) underwent both stages. All three groups revised their original writings in the next stage. To examine the learners' cognitive processes, think-aloud protocols were analyzed. The results showed that Group IC performed better in uptake than the other two groups, while no major differences were found between Groups I and C. This suggests that a mixture of learner-initiated noticing and teacher-initiated noticing influenced the learners' uptake.

        본 연구는 청국장 제조 시 Bacillus subtilis HJO-2, 18-9와HJ25-8의 3가지 균주를 starter로 접종하여 발효시킨 청국장의품질특성과 Isoflaveone의 함량을 측정하였다. 환원당을 유리하는데 관여하는 α-amylase 효소활성의 경우 Bacillus subtiliHJO-2와 HJ18-9 접종구에서 대조구보다 높은 활성을 보였다.청국장의 단백질을 분해하여 특유의 구수한 맛 성분을 유리는 protease 활성의 경우 Bacillus subtilis HJO-2, 대조구,HJ25-8, HJ18-9의 순으로 높은 활성을 나타내었다. 아미노태질소와 암모니아태질소의 함량은 Bacillus subtilis HJO-2 접종구에서 높았으며, 청국장 isoflavone 비배당체 함량은 Bacillussubtilis HJ18-9과 HJ25-8접종구에서 187.00±2.85, 210.77 ±11.75 μg/g으로 높았다. 이러한 결과로 본 연구를 통해 선별한균주를 스타터로 접종하여 구수한 맛에 관련된 protease, 아미노태질소, 환원당의 높은 값을 기대하기 위해서는 Bacillussubtilis HJO-2를 접종하여 청국장을 제조해야 하며, 체내이용률이 높은 isoflavone aglycone의 값을 기대하기 위해서는Bacillus subtilis HJ18-9과 Bacillus subtilis HJ25-8등으로 제조하는 것이 바람직하다. 청국장 제조에 알맞은 균주를 개발,평가하여 가공품으로 개발의 기초연구가 되고자 하였다.

        본 연구에서는 숙성 온도와 기간에 따른 마늘의 품질특성 변화를 살펴보기 위하여 통마늘을 60℃에서 60일간, 70℃에 서 40일간 숙성 처리하여 pH, 총산도, 갈변도, 5-HMF 함량, fructose 함량, SAC 함량, 총 폴리페놀 함량 및 DPPH radical 소거활성 측정하였다. 숙성 온도가 높고 숙성 기간이 길어질 수록 마늘의 pH는 감소하고, 총 산도, 갈변도 및 5-HMF 함량 은 증가하는 경향을 보였다. 특히, 5-HMF 함량은 60℃ 숙성 처리 시 0.08~3.30 mg/100 g으로 증가량이 적은 반면, 70℃ 숙 성 처리 시 0.05~106.07 mg/100 g으로 숙성 25일차부터 급격 히 증가하는 것으로 나타났다. Fructose 함량은 60℃의 숙성 온도에서는 0.77~14.57%으로 지속적으로 증가한 반면, 70℃ 숙성 온도에서는 30일차까지 1.07~19.75%까지 증가 후 15.11% 로 감소하는 것으로 나타났다. SAC 함량은 생마늘의 경우 48.11 mg/100 g이었으며, 60℃에서 20일 숙성 시 100.50 mg/ 100 g으로, 70℃에서 5일 숙성 시 85.55 mg/100 g까지 증가한 후 숙성 기간이 경과할수록 SAC 함량은 감소하였다. 생마늘 의 총 폴리페놀 함량은 98.38 mg/100 g이었고, 60℃ 숙성 처 리 시 591.82 mg/100 g까지, 70℃ 숙성 처리 시에는 665.22 mg/100 g까지 생마늘에 비해 각각 6.01배 및 6.67배 증가하였 다. DPPH radical 소거활성의 경우, 숙성 온도가 높고 숙성 기 간이 길어질수록 증가하는 경향을 보였다. 따라서 마늘 섭취 가 용이하고 항산화 활성이 우수한 숙성 마늘 제조를 위해서 는 70℃에서 30일, 숙성 마늘의 유효성분인 SAC의 다량 생산 을 위해서는 60℃에서 15일간 숙성 처리하는 하는 것이 적합 할 것으로 판단되며, 이상의 결과는 항산화 효과가 강화 및 SAC 함량 증진 등 선택적인 숙성 마늘의 제조에 활용이 가능 할 것으로 기대된다.

        메밀 속성 장 프리믹스의 최적 조건을 확립하고자 품질특 성을 조사하였다. 소금량(8, 10, 12%)에 따른 품질특성 결과, 대장균 군수는 소금량 10, 12%(21일), 8%(28일) 이후로 검출 되지 않았으며, B. subtilis HJ18-4를 starter로 첨가한 군은 발 효 7일부터 검출되지 않았다. 아미노태 질소 함량은 21일까 지 급격히 증가하였으며, 그 후로 완만히 증가하였다. 그 결 과, 소금 함량 10%, starter 첨가 시 보다 안전한 발효가 이루어 졌다. 가수량(1:1, 1:1.5. 1:1.8)의 따른 품질특성 결과, 총균수 와 아미노태 질소 함량은 가수량이 적을수록 높았으며, 1:1 혼 합(35일)일 때 아미노태 질소 함량은 169.5 ㎎%로 가장 높았 고, 발효 28일 이후 대장균 군이 검출되지 않았다. 용기별(항 아리, 유리, 플라스틱)의 품질특성 결과, 총균수와 아미노태 질 소 함량은 유리 용기 대비 플라스틱과 항아리 용기가 높았으 며, 대장균군도 보다 빨리 감소하였다. Batch size(1.25 ㎏, 2.5 ㎏)를 달리하여 제조된 메밀 속성 장의 품질특성 결과, 총균수와 아미노태 질소 함량은 batch size에 따른 큰 차이가 없었으 며, 대장균 군 측정 결과, 14일째 2.5 ㎏(3.00×102 CFU/g), 1.25 g (1.25×103 CFU/g)으로 나타났다. 메밀 속성 장 프리믹스의 최적 조건은 가수량 1:1, 소금의 양 10%, B. subtilis HJ-18-4를 starter로 첨가, 항아리 및 플라스틱 용기는 3주, 유리 용기는 4주 발효시키는 것이 바람직하였다.

        국내 전통식품 인증을 획득한 된장을 대상으로 간 섬유화 억제를 위한 기능 소재로서의 활용가능성을 알아보고자하였다. 국내 농산물로만 제조되어진 전통식품인증 된장총 24개 제품을 전국에서 수집하여 70% 메탄올로 추출한다음, MEF 세포주에 처리하여 간섬유화의 지표인 α-SMA의 발현 억제활성을 측정하였다. 공장식 생산과정을 거치는 일반 시판된장과 대조군 대비, 경북지역에서 수집된No. 13 전통인증된장 추출물이 약 74%의 α-SMA 발현억제활성을 나타내어 뛰어난 간 섬유화 억제활성을 나타내었다. 이는 의약품으로 사용되는 silymarin 84%과 비교시10% 정도밖에 차이나지 않았으며, 2년 이상의 발효시간을거친 제품임을 감안할 때 향후 발효시기에 따른 기능성의향상에 대한 기대도 가능할 것으로 예상된다.

        Proteinaceous insecticidal proteins, Cry proteins, from Bacillus thuringiensis (Bt) are insecticidal proteins that are highly active against several species of Lepidoptera. Thus, cry genes encoding these Cry proteins have been widely applied for construction of transgenic crops resistant to pest insects. In this study, through the 3D structure prediction and accompanying mutagenesis study for the Mod-Cry1Ac, 7 and 16 amino acid residues from domain I and II, respectively, responsible for its insecticidal activity against larvae of Spodoptera exigua and Ostrinia furnacalis were identified. We used site-directed mutagenesis to improve the insecticidal activity of Mod-Cry1Ac, resulted 31 mutant cry genes. These mutant cry genes encodes potent insecticidal proteins in the form of crystalline protoxins of 95 kDa. SDS-PAGE analysis of the recombinant polyhedra revealed that expressed Cry proteins was occluded into polyhedra and activated stably to 65 kDa by trypsin. When the insecticidal activities of these mutant Cry proteins against to larvae of P. xylostella, S. exigua and O. furnacalis were assayed, they showed higher or similar insecticidal activity compared to those of Cry1Ac and Cry1C. Especially, Mutant-N16 is considered to have the potential for the efficacious biological insecticide since it showed the highest insecticidal activity.

To construct novel cry genes with enhanced insecticidal activity, we randomly mutated these 23 amino acid sequences by in vitro muti site-directed mutagenesis, resulting in totally 24 mutant cry genes. For further characterization, these mutant cry genes were expressed as a fusion protein with polyhedrin using baculovirus expression system. SDS-PAGE analysis of the recombinant polyhedra revealed that expressed Cry proteins was occluded into polyhedra and activated stably to 65 kDa by trypsin. When the insecticidal activities of these mutant Cry proteins against to larvae of P. xylostella and S. exigua were assayed, they showed higher or similar insecticidal activity compared to those of Cry1Ac and Cry1C. Especially, among them Mutant-N16 showed the highest insecticidal activity against to both of P. xylostella and S. exigua. Therefore, Mutant-N16 is considered to have the potential for the efficacious biological insecticide.
