The objective of this study is investigation of contamination levels and assessment of health risk effects of heavy metals in herbal pills. 31 Items and 93 samples were obtained for this investigation from major herbal medicine producing areas, herbal markets and on-line supermarkets from Jan to Jun in 2010. Inductively coupled plasma mass spectrometer method was conducted for the quantitative analysis of Pb, Cd and As. In addition, the mercury analyzer system was conducted for that of Hg without sample digestion. The average contents of heavy metals in samples were as follows : 0.87 mg/kg for Pb, 0.08 mg/kg for Cd, 2.87 mg/kg for As and 0.16 mg/kg for Hg, respectively. In addition, the average contents of heavy metals in different parts of plants, including cortex, fructus, herba, radix, seed, algae and others were 0.63 mg/kg, 3.94 mg/kg, 1.42 mg/kg, 1.05 mg/kg, 0.16 mg/kg, 22.31 mg/kg and 10.17 mg/kg, respectively. After the estimations of dietary exposure, the acceptable daily intake (ADI), the average daily dose (ADD), the provisional tolerable weekly intake (PTWI) and the relative hazard of heavy metals were evaluated. As the results, the relative hazards compared to PTWI in samples were below the recommended standard of JECFA as Pb 3.1%, Cd 0.9%, Hg 0.5%. Cancer risks through slope factor (SF) by Ministry of Environment Republic Korea and Environmental Protection Agency was 4.24 × 10−7 for Pb and 3.38 × 10−4 for As (assuming that the total arsenic content was equal to the inorganic arsenic). Based on our results, possible Pb-induced cancer risks in herbal pills according to parts used including cortex, fructus, herba, radix, seed, algae and others were 1.95 × 10−7, 1.45 × 10−6, 2.14 × 10−7, 6.27 × 10−7, 1.99 × 10−8, 3.61 × 10−7 and 9.64 × 10−8, respectively. Possible As-induced cancer risks in herbal pills by parts used including cortex, fructus, herba, radix, seed, algae and others were 1.54 × 10−5, 7.24 × 10−5, 1.23 × 10−4, 2.02 × 10−5, 3.25 × 10−6, 2.18 × 10−3 and 5.67 × 10−6 respectively. Taken together, these results indicate that the majority of samples except for some samples with relative high contents of heavy metals were safe.

In order to investigate phenotype and genotype of Salmonella enterica subsp. enterica serovar Enteritidis, Forty-eight S. Enteritidis isolates from diarrhea patients were analysed using antimicrobial resistance typing, Phage typing, and Pulsed field gel electrophoresis in Seoul from 2004 to 2005. All of S. Enteritidis were resistant to streptomycin(SM, 37.5%), ampicillin(AM, 43.8%), t icarcillin(TIC, 43.8%), chloramphenicol(CM, 29.2%), t etracycline(TE, 10 .4%) and nalidixic acid(NA, 18.8%) among 16 antimicrobial drugs. Of 48 S. Enteritidis, 8 isolates(16.7%) were resistant to 1 drug, 3 isolates( 6.3%) to 2 drugs, 1 isolate (2.1%) to 3 drugs and 17 isolates(35.7%) to 4 drugs. The basic pattern of 4 drugs resistance was SM, TIC, TE, and CM but 1 drug resistant isolates represent all nalidixic acid resistance. Among 30 antibiotic r esistant S . Enteritidis, 2 1 isolates(70 %) were phage type 2 1, 8 i solates(26.7%) were phage t ype 2 3 and 1 isolate( 3.3%) was RDNC, respectively. Of the phage types observed, all of phage type 23(8 isolates) were nalidixic acid resistant and phage type 21 were AM-TIC-SM-CM multi-resistance(13 isolates; 43.3%), AM-TIC-SM-TE(4 isolates; 13.3%), AM-TIC-SM(1 isolate; 3.3%), AM-TIC-CM(1 isolate; 3.3%), and AM-TIC(2 isolates; 6.7%) resistance and 1 isolate of RDNC was NA-TE resistance. PFGE divided the isolates into two major clusters, A(n=14) and B(n=14). There were four different resistance profiles with resistance to AM, TIC, SM, TE, NA within PFGE A. Also resistance to AM, TIC, SM, CM was common within PFGE B. The PFGE A strains typed as PT21(n=5), PT23(n=8), and RDNC(n=1), While all the PFGE B strains typed as PT21(n=14). In consequence, there was the highly significant concurrence between resistance typing, phage typing and PFGE.

A total of 136 coliform bacteria isolated from spring water (112 strains) and ground water (24 strains), submitted to Seoul Health and Environmental Research Institute from June to July in 1997, were characterized biochemically and microbiologically. Colonical characteristics of each isolate were also noted, including color and texture on EMB agar. Among the 136 isolates, 50.7% were greenish metallic sheen color, 44.2% were pink and 5.1% were violet. The sixty four percent were smooth, 34.6% were mucoid and 0.7%. were rough. Twenty three bacterial species were identified by IMViC and API 20E test. Among the 136 coliform bacteria known to species, 39 isolates (28.6%) were Escherichia spp., 32 isolates (23.5%) were Klebsiella ssp., 30 isolates (22.1%) were Enterobacter spp., 19 isolates (14.0%) were Serratia spp., 6 isolates (4.4%) were Citrobacter spp., 4 isolates (3.0%) were Kluyuera spp. and 7 isolates (5.1%) were other bacterial species. Strains, which were gas-positive in lactose broth but gas-negative in Kligler Iron Agar were Ent. intermedium, Ser. liquefaciens, Ser. marcescenes and Salmonella arizoae. Strains, which were H2S production were also Kleb. pneumoniae, Kleb. oxytoca, Kleb. ornithinolytica, Ent. sakazakii, Ent. cloacae, Ser. liquefaciens, Ser. ficaria, Cit. freundii and Sal. arizoae. In the present study, most of coliform isolated from spring and ground water were E. coli, Klebsiella spp. and Enterobacter spp. Since coliform with pink colony in EMB agar was isolated as frequent as coliform with greenish metallic sheen colony, coliform with pink colony should be considered as important colony. Our results suggested that new coliform strains may be emerging on the basis of biochemical and microbiological testes.

The logarithmic mean of staphylococci was 1.60±0.59 CFU/g, and in accordance with fish and shellfish, shellfish 0.01±0.00 CFU/g, echinodermata 3.51±0.60 CFU/g, fish 1.68±0.64 CFU/g, crustacean 0.34±0.33 and mollusc 2.90±0.11 CFU/g. The logarithmic mean of coliforma was 2.2±0.32 CFU/g, and in accordance with fish and shellfish, echinodermata 3.58±0.89 CFU/g was highest, shellfish 3.25±0.30 CFU/g, crustacean 3.23±0.49 CFU/g, fish 2.18±0.63 CFU/g, peeled shellfish 1.80±0.51 CFU/ g and mollusc 1.55±0.95 CFU/g. As the results of research of the change of the contaminated indicator microflora in working with storage period at 10℃, 20℃ and 30℃, total viable counts was increased without storage temperature and enterococci were decreased slowly at 10℃, but increased at 20 and 30℃ Vibrios were decreased slowly at 10℃, decreased at 20℃ and 30℃ in 2 days after increased rapidly. Staphylococci were increased promptly without storage temperature in 2 days, then the total viable counts were maintained. Coliforms were increased at 10℃ by 7 days, then decreased or maintained after 14 days, changed at 20℃ in accordance with fish species in 2 days, then returned to the initial total viable count, and decreased rapidly at 30℃ on 2 days. By the way, there were no difference among the species.
