        Background : Platycodon grandiflorum root (PGR) was one of the primary herbs used in a phlegm-relieving herb from the past. Platycoside compounds on PGR may exhibit neuroprotective, antimicrobial, anti-inflammatory, anti-cancer, anti-allergy, improved insulin resistance, and cholesterol-lowering properties. In order to developing a concentrate product that improved the functionality and preference of PGR, it was fermented using lactic acid bacteria (Lactobacillus plantarum N76-10 and 56-12). Methods and Results : The concentrate products were created by PGR-concentrate (PGRC, 60 ºBrix) mixed with fermented PGR-extract (FPGRE, 2 ºBrix) at the level of 0, 50, 100, 150 and 200%. Sweetness and preference were supplemented by other added materials including honey, oligosaccharide, concentrate of jujube (60 ºBrix) and pear (60 ºBrix), and cactus Chounnyouncho extract (2 ºBrix). The products were put into investigation for their preference of taste, antimicrobial activity in accordance with amount of FPGRE. When it comes to preference of taste, the most favor is adding 100% of FPGRE on PGRC. The product added 150% FPGRE exhibited a strong microbial anti-proliferation in all four kinds (Corynrbacterium diphtheriae, Klebsiella pnneumoniae, Staphylococcus aureus, and Streptococcus pyogenes) of bacteria inducing bronchus diseases and was higher antimicrobial activity than concentrate without FPGRE. In terms of the sensory evaluation (taste, texture and visco-elasticity), concentrate mixed with FPGRE (10), jujube concentrate (2), pear concentrate (10), cactus Chounnyouncho extract (10), oligosaccharide (2), honey (1) and xanthan gum (0.02) showed the highest scores. Conclusion : Thus, A PGR concentrate was made by adding FPGRE (100%) and it was increased organoleptic quality, antimicrobial activity. These studies may provide new product development for effective utilization on Platycodon grandiflorum root.

        Background : This study aimed to identify the mechanisms of the antinociceptive effects of PG in the fibromyalgia (FM)-like animal model. Methods and Results : To assess the possible effect of PG on FM symptoms, we constructed a FM animal model induced by intermittent cold stress with slight modification. All mice underwent nociceptive assays using electronic von Frey anesthesiometer and Hargreaves equipment. To assess the relation between PG and the expression levels of brain-derived neurotrophic factor (BDNF), cAMP response element-binding protein (CREB), and phosphorylated CREB (p-CREB), western blotting and immunohistochemistry analyses were performed. In behavioral analysis, nociception tests showed that the pain threshold was significantly decreased in the FM group compared to control group. Western blot and immunohistochemical analyses of the medial prefrontal cortex and hippocampus showed downregulation of BDNF and p-CREB proteins in the FM group compared to control group. PG recovered these changes at behavioral tests and protein level. These results provide evidence that the effects of PG extract in the FM model may be related to its modulating effect on the BDNF signaling pathway in the hippocampus and medial prefrontal cortex. Conclusion : Our animal model may be involved in the mechanism by which PG extract is effective as a therapeutic agent for FM.

        Background : This experiment was conducted to select suitable industrial crop after Panax ginseng cultivation under the greenhouse in the middle area. Methods and Results : This experiment was conducted in greenhouse of Chungbuk Agricultural Research Service and extension services from 2015 and 2016 years. Ginseng grown four years in greenhouse. After cropping of Panax ginseng was transplanted Cynanchum wilfordii, Platycodon grandiflorm, Dioscorea opposita, and Codonopsis lanceolata. Non-cultivated field that were grown sesame. Cynanchum wilfordii, Platycodon grandiflorm, and Codonopsis lanceolata was collected in the Chungbuk area and Dioscorea opposita was collected in Andong-si. Each industrial crop after panax ginseng cultivation under the greenhouse was sowing from late April to early May 2015 years. The growth and yield of crop examined in the experiment were good on the whole except Codonopsis lanceolata without disease due to continuous cropping of Panax ginseng. The yield of Cynanchum wilfordii, Platycodon grandiflorm, Dioscorea opposita, and Codonopsis lanceolata was 803kg, 1,393kg, 1031kg, 412kg per 10a respectively, and the every decline yield index was by 1%, 8%, 12%, and 31% respectively. The proper crop in after cropping of Panax ginseng was cynanchum wilfordii, which was yielded 1,393kg per 10a by fresh tuber and it was 1% decline compared to the non-cultivated field. Conclusion : In this study, Cynanchum wilfordii, Platycodon grandiflorm, Dioscorea opposita was recommended as a crop after Panax ginseng cultivation under the greenhouse in the middle area.

        Background:The study aimed to obtain data on the effects of cultivation and soil reduction of green manure crop on the quantity and quality of organically cultivated Cynanchum wilfordii Hemsley.Methods and Results:The experiment comprised four treatments: control, hairy vetch, barley, and hairy vetch + barley (3 : 2). The plant height in the hairy vetch treatment (86.3㎝) was significantly different from that in the other treatments, whereas the stem diameter leaf area, and special product analysis division (SPAD) value did not differ across the treatments. The largest soil reduction of green manure crop was recorded in the barley treatment (440 ㎏/10 a), whereas the smallest was recorded in the single treatment with hairy vetch (80 ㎏/10 a). The hairy vetch + barley (60 : 40) treatment showed 63% more soil microorganisms than control. Radical scavenging activity estimation revealed that the total polyphenol content was highest (1,740 ㎎/㎏), and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) was 92.6% in the barley treatment. The 2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) activation was highest in the control (51.1%), and the root yield was the highest in the barley treatment (310 ㎏/10 a).Conclusions:The root yield, total polyphenol content, and antioxidant activity of Cynanchum wilfordii (Maxim.) Hemsley increased in presence of the green manure crop barley.

        The aim of this paper is to investigate the syntactic structure of the so-called transparent relative clause. What in the standard free relative clause has been treated as a unit including a head NP and a relative pronoun together. In the case of transparent free relatives, however, a what-clause reveals some peculiar characteristics different from standard relative clauses. To explain those characteristics, many linguists have suggested several different analyses. The analyses can be categorized into three groups: Backward Deletion, Shared Structure analysis, (Rightward) Movement and Deletion analysis. Because of their drawbacks, I have suggested Leftward Movement and Deletion analysis, and adopted Winkler's(2005) D(rivational)-Model of Grammar to explain the interactions between the PF deletion and LF interpretation.

        Background : This study was investigated the effect of organic fertilizer application and stem training methods on the growth and yield of Cynanchum willfordii. Methods and Results : Traditional species, C. willfordii conducted the test to the field in Chungbuk ARES, Korea. Two methods, non-supporting (custom) and I-shape supporting was used for stem training method. Planting spacing was adjusted to 30 (interrow spacing) x 20 cm (intrarow spacing) and other key management is the followed the cultivation manual for standard medicinal crops. Organic fertilizer application is conducted in mid-March before planting of C. willfordii. Registered organic agricultural materials such as fungal cultures (CC), mixed organic materials (MO), fermented and mixed expeller cake (EC), and fermented fowl dropping (FD) was used. Application rate is based on the nitrogen application rate after soil testing. Plant height was both good in all at I-shape supporting (266.7 ㎝) and all in non-supporting (160.8 ㎝); however, stem diameter was more thicker in custom (4.6 ㎜) of I-shape supporting and EC (4.8 mm) of non-supporting. Number of branched stem were good at EC from I-shape supporting and non-supporting, as well as leaf growth. SPAD value was evaluated in MO (52.7) of I-shape supporting and EC (52.3) of non-supporting. Number of branched root per organic fertilizer were showed 7.0 at MO from I-shape supporting, but 7.3 at FD from non-supporting. While length of main root were 27.0 cm in MO of I-shape supporting and 31.3 ㎝ in FD of non-supporting. Root weight also more heavier in EC (66.3 g) of I-shape supporting and FD (53.0 g) of non-supporting. When applied organic fertilizer, total density of soil microorganisms were changed into 38.4 ppm in custom; however, it more plentiful of 90.7 ppm in MO, it showed good effect on the replication of soil microbiota. Conclusion : From the investigated results, MO of I-shape supporting was good at overall growth, including good tendency of roots growth.

        Background : This study was performed to investigate the morphological and growth characteristics of tetraploid Polygonum multiflorum L. with colchicine treatment, and to clarify its effective induction and time for the increase of rhizome hypertrophy and effective components. Methods and Results : The induction of tetraploid P. multiflorum were done with colchicine treatment of 0.1, 0.5, and 1.0% conc. and the time was treated with 12, 24, and 48 hrs, respectively. DMSO 1% is treated for the effective penetration of colchicine with constant-temperature oven at 24C. Leaf explant was added the HR-A solution stained with HR-B solution, and then determined whether DNA contents of the doubles or not. Seed stand rate showed lower tendency as higher treatment concentration and as delayed the treatment time. When treated in DMSO 1% with 48 hr, seed stand was so small rate with 2%, and treated in 0.5% with 12 hr, seed stand was recorded higher rate with 33%. When treated in 0.5% colchicine with 24 hr, chimera and tetraploid induction rates were highest and obtained 29 individual chimera and 5 individual tetraploid. On overground growing, mean stem diameter of tetraploid (11.3 ㎜) is two times thicker than that of diploid (6.4 ㎜), with the vigorous growth. leaf length and width of tetraploid was 9.1 and 6.2 ㎝, respectively, and that of diploid was 6.8, and 5.1 ㎝, respectively. Stem diameter of a scape was also more thicker tendency in tetraploid than in diploid, chlorophyll contents over 29% existed in tetraploid. Investigated result from stoma no. and size of leaf with 400 magnification, stoma no. of diploid (26 individual) was four times higher than that of tetraploid (7 individuals), the size of tetraploid (38 ㎛) is twice longer in size of diploid (18 ㎛) and stoma area are broaden in four times. Seed length of tetraploid (3 ㎜) is longer than that of diploid (22 ㎜); but, seed width of tetraploid (22 ㎜) is widen than that of diploid (14 ㎜) in seed size comparison. That is, seed length is bigger in 1.4 times and seed width is bigger in 1.6 times, and seed surface area is broaden in two times. Conclusion : From above results, overground growing of tetraploid P. multiflorum is fairly better than that of native P. multiflorum. In the future, this tetraploid variety will promote to register when recognize the excellence than native species after tested the disease tolerance, underground growing characteristics, and key index component.

Purified platycoside compounds from the roots of PGR may exhibit neuroprotective, antimicrobial, anti-inflammatory, anti-cancer, anti-allergy, improved insulin resistance, and cholesterol-lowering properties. To evaluate preference and functionality of PGR extracts, PGR was fermented by several lactic acid bacteria. Lactic acid bacteria used were Leuc. mesenteroides N12-4 and N58-5, L. plantarum N76-10 and 56-12, L. brevis N70-9 and E3-8. Methods and Results : This study was performed in order to investigate the changes of platycosides, as well as the antimicrobial activities on bronchus diseases inducing bacteria(C. diphtheriae, K. pnneumoniae, S. aureus, S. pyogenes) of Platycodon grandiflorum root(PGR) fermented by using lactic acid bacteria(Leuc. mesenteroides N12-4, Leuc. mesenteroides N58-5, L. plantarum N76-10, L. plantarum N56-12, L. brevis N70-9, L. brevis E3-8). Growth of L. plantarum on PGR was the most active during lactic acid fermentation by some different strains. Total platycoside, platycoside E, platycodin A, polygalacin D2, polygalacin D and diapioplatycoside E contents of PGR fermented for 96 hours at 37℃ by Leuc. mesenteroides and L. plantarum were increased, while platycodin D and platycodin D3 were decreased. The antimicribial activity on PGR fermented by L. plantarum N56-12 exhibited a strong microbial proliferation in all four kinds of bronchus diseases inducing bacteria and was higher than non-fermented PGR extract. Conclusion : Thus, this results showed antimicrobial activities on bronchus diseases inducing bacteria and platycosides content of PGR by L. plantarum N56-12 were higher than non-fermented PGR extract.

        Background : The effect of ridge-up bed cultivation and stem training method on the plant growth and yield of Cynanachum willfordii was investigated. Methods and Results : Domestic variety of Cynanachum willfordii was tested at Chungbuk ARES under the following conditions. Nonsupporting (custom) and I-form supporting was used for stem training method, and ridge height was set as 30 (custom) and 60 cm (high ridge). Planting spacing was adjusted to 30 (interrow spacing) × 20 cm (intrarow spacing), and other major management was followed the method of standard cultivation for medicinal crop. Investigated result from leaf characteristics, leaf length was longer in high ridge cultivation (HRC) as 11.1 cm than custom cultivation (CC) as 10.6 cm. Leaf width is proved to be 12.8 (HRC) > 11.2 cm (CC). Leaf number is proved to be 294 (HRC) > 254/plant (CC), with higher number of 44/plant at HRC and weight/10 leaves were more heavy at HRC (4.9 g) than that of CC (2.6 g). It was more fruit setting at HRC over 15/plant. According to the stem training method and ridge height from nonsupporting cultivation, main root was 4.0 (CC), higher than that of HRC over 0.5/plant. However, root length was more longer in HRC (28.6) than that of CC (25.0 cm). Main root diameter was also showed more thicker pattern in HRC. From staking cultivation, root number of HRC was 7/plant, it was recorded more 3/plant than that of CC, and it was also same pattern in main root length and root diameter. By the standard of commercial root, yield of living roots in nonsupporting cultivation were 59.0g/plant (HRC), it was more heavy over 10.4g/plant than that of CC. In staking cultivation, HRC were recorded as 74.2g/plant, more heavy 6.9g/plant than that of CC. Yield from I-form support stem training and ridge-up bed cultivation of HRC was higher approxmiately 52.7% than that of CC. Conclusion : Overall growth by high ridge cultivation of I-form support was good and yield of HRC was also increased over 52.7% than that of CC.

        Livestock wastewater has high potential as one of energy resources because this wastewater is including high organic matter. Therefore the studies attempting to bio-gasification and bio-electricity generation using livestock wastewater is being tried. The pre-treatment system used in this study was the purpose to control the ammonia nitrogen in conjunction with the system and the microbial fuel cell. Because ammonia nitrogen is to inhibit the electricity generation efficiency of microbial fuel cell. These studies were to ascertain the effect of oxidants on the nitrogen removal in the pre-treatment system using catalyst and microbubbles to treat the ammonia nitrogen. The three kinds of oxidant such as air, oxygen (O2), and hydrogen peroxide (H2O2) were used to know the ammonia and nitrate nitrogen removal. This system was operated with four kinds of conditions. First method is O2 gas with 100 mL/min with 1ml of 30% H2O2 was supplied to the wastewater. A second method, the O2, with 400 and 1,000 mL/min was supplied through the flow meter before livestock wastewater was flow in the reactor. The last method, air was supplied 800 mL/min. The nitrate removal had no significant difference all conditions except the air supply. On the other hand, the ammonia and nitrate nitrogen removal when oxygen was supplied with 1000 mL O2/min was higher than that of the other conditions. The removal rate when air was supplied 800 mL/min was similar to the value in the supplied with 400 ml O2/min. This result showed that oxidant was important factor to improve the ammonia nitrogen removal rate.

        This paper aims to suggest Middle and Medio-passive categories in Korean with cross-linguistic data. K. Kim(2009) argues that there are three kinds of non-active Voice categories in Korean: I-inchoatives, Zero inchoatives and Passives. I, however, argue that I-inchoative and Zero inchoative show morphological, distributional and semantic differences, and thus they should be respectively categorized into Middle and Anticausative. The so-called –i morpheme is regarded traditionally as passive morpheme, but I show that it is not just a passive but Non-active Voice morpheme in Korean. I have also argued that Medio-passive constructions should be assumed in Korean, because cross-linguistically they show distributional and semantic differences from middles and passives. An Instrumental phrase and an Agent phrase or an Agent phrase and‘by itself’can appear together in the medio-passives, and semantically they can have middle interpretation too.

        Genome sequencing researches for considerable numbers of crops and wild plants are being developed. Cytogenetic researches according to chromosome number and size are essential to confirm and comprehend ploidy level and genome size before genome sequencing project is actually conducted. Cytogenetic researches on six food crop plants were carried out by DAPI staining and fluorescence in situ hybridization (FISH) method. Fagopyrum esculentum Moench showed 2n=2x=16, each chromosome length of 1.42㎛ to 1.77㎛, total chromosome length of 13.31㎛, and karyotypic formula of 2n=8m; Phaseolus angularis W.F. Wight, 2n=2x=22, 2.01㎛ to 3.84㎛, total 28.03㎛, 2n=9m+2sm, Perilla frutescens var. japonica Hara, 2n=2x=40, 1.73㎛ to 2.76㎛, total 44.36㎛, 2n=5m+13sm+2st. Chromosome sizes of the other three species such as, Panicum miliaceum L., 2n=2x=36, total chromosome length of 30.83㎛, Sesamum indicum L., 2n=2x=26, 27.39㎛, lpomoea batatas L., 2n=2x=30, total 33.51㎛ were too small for each chromosome type to be identified and analyzed. The result of FISH analysis using 5S and 45S rDNA probe showed species-specific chromosome locations in the genome. These preliminary analyses were carried out to decide which food crop to prioritize for genome sequencing.         These studies were conducted to discriminate Cynanchum wilfordii, C. auriculatum, Metaplexis japonica, andPolygonum multiflorum by morphological characters. Cynanchum wilfordii, C. auriculatum, and Metaplexis japonica(Ascle-piadaceae) were easily distinguished from Polygonum multiflorum by twining stem to the left, producing opposite leaves,having laticiferous stems and leaves at cutting cross-sectionally, and lacking lignification of the stem. Four species had differ-ent morphological characters of flower. Comparing four species to fruit type, three species of Asclepiadaceae had follicle,while Polygonum multiflorum had achene with three wings. Follicle shape was lanceolate, narrowly lanceolate, and widelylanceolate in C. wilfordii, C. auriculatum, and Metaplexis japonica, respectively. Whereas seed color of four species was darkbrown similarly, seed shape and 1,000 seed weight were useful characters to discrimate between three species ofAsclepiadaceae and Polygonum multiflorum. Shape and color of root were thickened and yellowish brown in C. wilfordii andC. auriculatum. They were thin and long and white in Metaplexis japonica, and fusiform and reddish brown in Polygonummultiflorum. They would be useful characters to distinguish Cynanchum spp., Metaplexis japonica, and Polygonummultiflorum, however, impossible characters to discriminate between C. wilfordii and C. auriculatum.

        충북농업기술원 마늘연구소에서는 2000년부터 수집한 유전자원 중 한지형인 ‘단양종’ 보다 조숙다수성이며 불완전추대의 특성을 가진 계통을 5년 동안 지속 선발하여 ‘단산’ 품종을 육성하였다. ‘단산’ 마늘의 추대 특성은 불완전 추대형으로 ‘단양종’의 완전 추대형과 뚜렷한 구별성을 갖는다. 따라서 화경이 출현하지 않아 수확 전에 쫑을 뽑을 필요가 없으며 숙기도 6일 정도 빨라 수확기 때 장마를 피할 수 있는 장점을 갖고 있다. 2차 생장비율은 3.3%로 ‘단양종’이 17.3% 보다 매우 낮아 이차생장율이 적은 상품성 마늘을 생산할 수 있다. ‘단산’의 통의 크기는 ‘단양종’ 보다 크고, 쪽 껍질의 색은 갈색을 띠며 안토시아닌 줄무늬가 존재한다. 수량은 1,131 kg/10 a로 ‘단양종’ 보다 18% 정도 높다.

        Understanding the host defense mechanisms in response to brown leaf spot disease caused by Cochliobolus miyabeanus is very important for production of resistant plant. In this study, two-dimensional gel electrophoresis (2-DGE) in conjunction with mass spectrometry was utilized to unravel changes of stress inducible proteins in rice leaves infected with C. miyabeanus. For this purpose, we firstly observed disease developmental process of C. miyabeanus in rice using trypan blue, anilin blue, acid fuchsin staining, and DAB staining for ROS detection and expressional abundance of ROS related proteins in rice leaves inoculated was confirmed by Western blotting. Proteins were extracted by PEG fractionation and their expression patterns were analyzed by 2-DGE and subjected to image analysis using the ImageMaster 6.0 2D Platinum software, resulting in the identification of 86 differentially expressed protein spots with significantly changes (p<0.05) compared with control. MALDI-TOFTOF-MS analysis revealed that 69 proteins including 42 and 27 significantly up- and down-regulated proteins, respectively, were identified. Based on gene ontology analysis, identified proteins were classified according to their functional groups: metabolism (20%), oxygen-detoxifying (13%), protein stress/defense (24%). Thus, these results for the first time suggest that differentially induced proteins may play a key role for understanding host defense mechanisms during rice -C. miyabeanus interaction.

        The rice blast disease caused by Magnaporthe oryzae (M. oryzae) is one of the most serious diseases of cultivated rice (Oryza sativa L.) in most rice-growing regions of the world. In order to investigate early responsible genes in rice in response to M. oryzae, we analyzed transcriptomics analysis using 300 K tilling microarray chip. The quality of RNA samples was initially validated by 4 defense related genes and phytoalexins measurement using RT-PCR and HPLC, respectively, which are well known defense markers. We determined that accumulation of 608 genes showed statistically significant changes in the level of transcription (>2 fold change, P<0.05). Among them, 261 genes were more up-regulated in incompatible interaction than that of compatible one. We further analyzed GO enrichment analysis of the 41 and 231 which were 2 fold up-regulated genes at 12h and 48h in incompatible interaction, respectively, using Rice Oligo nucleotide Array Database (http://ricearray.org). Furthermore, MapMan analysis (http://mapman.gabipd.org/) revealed that 21 and 85 genes including 18 receptor-like genes which were more induced in incompatible interaction compared to compatible interaction were found to be involved in biotic stress. Thus, this study suggests that early inducible genes including receptor-like protein kinases in incompatible interaction may play a key role in disease resistance against M. oryzae attacks.

        Here, we first demonstrate that identification of rice brown spot disease fungus (Cochliobolus miyabeanus, C. miyabeanus) proteins is possible in infected tissues using in planta apoplastic proteome with non-destructive tissues. In planta apoplastic proteins from rice leaves inoculated with C. miyabeanus, CM2 (compatible race), were isolated by vacuum infiltration with CaCl2/Na-acetateextractionbuffer, separated by SDS-PAGE, and identified by MudPIT. Of the 529 proteins that were identified by MudPIT, a large proportion (490) was from the rice. Numerous carbohydrate metabolic process (48), oxidation and reduction (44), response to oxidative stress (20%) were identified and confirmed their expression at RNA levels using microarray. Bioinformatic analysis showed that 176 and 39 of these proteins have a signal peptide in rice and rice brown spot fungus, respectively, using Signal P. The large proportion of proteins interestingly identified from the in planta apoplast were involved inprotease, hydrophobin, and host cell wall hydrolysis (Xylanase, beta-glucosidase) derived from pathogen. Thus, we suggest that in planta rice apoplastic secretome will be an important clue to understand the rice-rice brown spot fungus interactions.
