        This study was conducted in order to monitor the extraction conditions for a gel-state beverage development of the Opuntia ficus-indica stem. Moreover, the organoleptic properties of the beverage prepared by the extract were optimized using the response surface methodology (RSM). The determination coefficient (R2) value for the extraction yield of the stem was 0.95 (p<0.01). The maximum extraction yield was obtained at an extraction temperature of 93.02℃, 123 min of extraction time and 22.57 mL/g of water to sample. The beverage was prepared with the addition of xanthan gum, sugar and persimmon vinegar to the extract with a central composite design. The maximum organoleptic color of the beverage was obtained at 0.38% xanthan gum, 7.91% sugar and 0.76% persimmon vinegar. The maximum organoleptic flavor was obtained at 0.30% xanthan gum, 7.06% sugar and 1.26% persimmon vinegar. The maximum organoleptic taste was obtained at 0.22% xanthan gum, 10.36% sugar and 0.90% persimmon vinegar. The maximum overall palatability (3.92 score) of the gel-state beverage was obtained at 0.35% xanthan gum, 10.83% sugar and 1.21% persimmon vinegar.

        파종기에 따른 울금의 초장, 경직경 및 엽수는 5월 10일 파종구에서 가장 양호하였다. 생육단계에 따른 울금의 생육변화를 살펴본 결과 울금의 생육최성기인 10월에 초장은 121 cm로 가장 길게 나타났고, 경직경 또한 3.45 cm로 가장 두꺼웠다. 엽수는 9월 20일 6매로 조사되었으나 이후 엽수가 감소함이 관찰되었다. 생육단계에 따른 울금 지하경 무게 변화는 생육기간에 경과함에 따라 지하경의 무게 증가가 비례하였으며, 11월 20일 조사에서 울금의 수량이 가장 크게 나타났다. 울금을 건조하는데 가장 적합한 온도는 40℃로 확인되었다. 부형제를 달리하여 제조한 울금환의 관능검사 결과는 향, 색, 맛, 조직감 및 종합적 기호도에서 찹쌀과 꿀을 부형제로 사용한 시험구의 기호도가 가장 우수하였으며 밀가루만을 사용한 시험구의 선호도는 낮게 나타났다.

        To establish the optimal manufacturing conditions of soybean koji, soybean Koji prepared with Aspergillus oryzae 6-M-1 and Bacillus subtilis 3-B-1 isolated from traditional Korean meju. During 7 days of making Koji, the amount of amino-type nitrogen was getting more increase. The amount of amino-type nitrogen of Koji prepared with A. oryzae 6-M-1 was 686.16 mg% (w/w), that of Koji with B. subtilis 3-B-1 was 643.46 mg% (w/w) at seventh day of making Koji. The α-amylase activity of Koji prepared with A. oryzae 6-M-1 was 1472.54 unit/g, that of Koji with B. subtilis 3-B-1 was 791.00 units/g on the seventh day of the making. The acidic protease activity of Koji prepared with A. oryzae 6-M-1 was 309.00 unit/g, that of Koji with B. subtilis 3-B-1 was 135.88 unit/g at 7th day of making. The amount of amino-type nitrogen and enzyme activities of soybean Koji prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 were produced more than those of wheat flour Koji made in factory. Sensory evaluation on a commercial doenjang and doenjangs prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 was not significantly different at p<0.05.

        This study aimed to investigate the optimal enzymatic hydrolysis conditions of alginic acid using Viscozyme® L, Celluclast® 1.5L, Saczyme®, Novozym®, Fungamyl® 800L, Driselase® Basidiomycetes sp., and Alginate Lyase, for production of reducing sugar. Response surface methodology (RSM) based on central composite rotatable design was used to study effects of the independent variables such as enzyme (1-9% v/w), reaction time (10-30 h), pH (3-7) and reaction temperature (30-70oC) on the production of reducing sugar from alginic acid. The coefficients of determination (R2) of Y1, Y2, Y3, Y4, Y5, Y6, and Y7 for the dependent variable regression equation were analyzed as 0.947 0.968, 0.840, 0.926, 0.923, 0.892 and 0.825. And the p-value of Y1, Y2, Y3, Y4, Y5, Y6, and Y7 within 1% (p < 0.01) was very significant. The optimal conditions were 1.0% of the quantity of the enzyme, 10.0 hours of the response time, pH 3 and 70.0oC of the reaction temperature, where the production rate was 483.1 mg/g-alginic acid, the highest of all the enzymes used.

        본 연구에서는 단면수리실험으로 다양한 실험파 조건과 유수실 폭(B)을 변화시키면서 유공부 길이(S)와 반사율의 관계를 검토하여 유공 1실인 유공구조물의 정수면 아래 최적 유공부 길이를 제시하였다. 입사파로 불규칙파를 적용함에 따라 최저 반사계수는 상대 유수실 폭(B/Ls)이 B/Ls≈0.15에서 발생하였다. 유공부 길이와 입사파고와의 관계를 고려한 상대 유공부 길이(S/Hs)는 S/Hs≈2.5에서 최저 반사계수를 보였으며, 이는 정수면 아래 유공부 길이가 입사파고의 약 2.5배일 때 최적임을 의미하는 것이다.

        This work presents an experimental study of the influence of lifting velocity on cake formation during filtration. For design of hot gas cleanup system using ceramic filter reactor, the most important consideration is coating conditions of sorbent in filter surface (for example : lifting velocity, coating weight of sorbent, pulsing interval and removal effect for dechlorination and desulfurization). We studied the optimum operation condition as paticle size and lifting velocity using a ceramic filter reactor at 550oC. Based on the results obtained during cold and hot test, optimum lifting velocity in a ceramic filter reactor was selected 0.68 m/s. Also, the removal behaviour of the ceramic filter during filtration was studied using differential pressure. Optimum removal efficiency for dechlorination and desulfurization accomplished at differential pressure condition over 74 mmH2O.

        Recently, the recycling of end-of-life vehicles is becoming increasingly interesting for less waste discharge and recovering useful materials such as valuable metals. Hence, in Korea, the target of the recycling rate is made to 85% until 2014 (energy recovery within 5%) and the recycling rate to 95% after 2015 (energy recovery within 10%) according to the law of "regulation about resource recycling of electrical and electronic products and automobiles". However, the recycling rate is around 84% in 2010, and registered numbers of shredder residue recyclers among dismantling recyclers, crushing recyclers, shredder residue recyclers, and waste gases recyclers are very few. In order to meet the goal of 85% until 2014, Korean recycling industry of ASR should grow bigger and innovative recycling technologies have to be developed as well. In the meantime, a recycling technology of automobile shredder residue is developed in the present study, in which ASR is introduced to a copper smelting process. This process is very promising because of co-beneficial effects such as recovering copper and process heat simultaneously from ASR. In this study, lab-scale melting furnace was developed and melting tests for various ASR were carried out. From the results, the physicochemical characteristics of Korean ASR were analyzed and its melting behavior was investigated for the application to the copper smelting process. In particular, melted slag products were fabricated at different melting temperatures. Then, the basicity and pouring index in the lab-scale melting furnace were examined to find out appropriate operating conditions for the melting process. As a result, Because the amount of lump coal that included in existing process samples is a little, melting state and value of pouring index are very similar to existing process samples. In result of this experiment, in case of changing lump coal to ASR, The existing copper smelting facilities, if basicity and melting temperature are well controlled, flow of melting material is considered that is almost not affected.

        Electrochemical reductive extraction of copper from LCD manufacturing process and through hole plating process for PCB circuit board wastewater was experimented using synthetic wastewater. Copper plate which could be used as raw material through melting with extracted copper from wastewater and graphite plate were used as a cathode and an anode, respectively. The copper extraction could be optimized in pH 2 and polar space of 45mm. The extraction rate of copper per unit energy was decreased as HRT increased and initial copper concentration decreased. As the optimal HRTs which could maximize the productivity for copper on energy, 80 ~ 110 min at 50 mg/L, 64 min at 1,000 mg/L and 77 ~ 98 min at 3,000 mg/L were determined, respectively.

        생식력을 보존하고자 하는 가임기 여성 암환자는 항암치료 전, 자신의 난소를 동결보존할 수 있으 며, 암 치료 후 동결된 난소를 해동, 이식하는 방법으로 임신에 성공할 수 있다. 하지만 이 과정 중 난소과 난포는 동결 손상을 입게 되며, 난소 조직의 생존률은 크게 저하될 수 있다. 그 후 난소 이식 에 따른 허혈성 손상 등의 이유로 암 생존 후 동결 해동된 난소조직이 성공적으로 난소 기능을 유지 하는 것은 매우 어려운 일이다. 최근 난소의 동결과 보존 그리고 이식은 전세계적으로 활발한 연구 가 진행되고 있는 주제이지만 아직까지 획기적인 방법이 개발되지는 못하였다. 따라서 가임력 보존 의 가장 궁극적인 방법인 난소동결보존을 최적화하여 그 효율성을 높이고, 이식후 생존기간을 연장 시킬 수 있는 기술의 탐색이 지속적으로 이루어져야 할 것이다.

        This study was conducted to establish the extraction conditions for the optimization of the biological activities of black garlic using the response surface methodology (RSM). The extraction conditions were based on the central composite design, with 15 kinds of variations in the extraction temperature (50-90℃), extraction time (3-15 hrs), and ethanol concentration (0-100%). The total phenol, flavonoids, thiosulfinate contents, and anti-oxidant activity of black garlic extract were significantly higher at the J condition (90℃ for 9 hrs with a 50% ethanol concentration). In this condition, the biological activities such as DPPH radical scavenging (66.10%), ABTS radical scavenging activity (75.02%), and reducing power by of FRAP (375.4 μM/mL) were excellent. Overall, the extraction conditions for the optimized biological activities of black garlic via RSM were expected to be at 89.68℃ for 9.79 hrs with a 55.72% ethanol concentration. The actual values were 96.4-114.8% of the predicted values.

        This study was conducted to develop a sauce prepared with sweet pumpkin and Korea Doenjang. The optimum conditions for manufacturing sweet pumpkin-doenjang sauce were investigated using the response surface methodology, based on the central composition design. The amount of stock added, the thickening agent, and doenjang were used as the independent variables, and the sensory characteristics (taste, flavor, color, and overall acceptability) were used as the dependent variables to evaluate the optimum conditions for the preparation of the sauce. The optimum conditions for the maximized-responses variables in the preparation of the sauce were 448.5 g of sweet pumpkin stock, 331.5 g of the thickening agent, and 20.0 g of doenjang. The quality characteristics of sweet pumpkin-doenjang sauce that was manufactured at optimum conditions were as follow: 89.55% moisture content, 0.70% crude protein, 0.10% crude lipids, and 0.71% crude ash. The pH of the sauce was 5.96; total acidity, 0.08%; and soluble solids, 6.80 °Brix. The total polyphenol content of the sauce was 5.70 mg/L. The electron-donating ability and reducing power of the sauce were, 14.24% and 1.64 OD, respectively.

        Enzymatic action and microbial growth degrade the quality of fresh-cut lettuce. Browning, a bad smell, and softening during storage are the major forms of quality deterioration. Health-oriented consumers tend to avoid foods treated with chemicals to maintain their freshness. This study was conducted to evaluate the change in the quality of fresh-cut lettuce with combined low-temperature blanching (LB) and ultrasonication (US). The optimum condition was selected using the response surface methodology (RSM), through a regression analysis with the following independent variables; the ultrasonication time (X1), blanching temperature (X2), blanching time (X3), and dependent variable; ΔE value (y). It was found that the condition with the lowest ΔE value occurred with combined 90s US and 45℃ 90s LB (US+LB). The combined treatment group (US+LB) was stored at 10℃ for 9 days with the control group and each single-treatment group, with low-temperature blanching and ultrasonication. Overall, the US+LB group had a significantly high L* value, which indicates significantly low a*, b*, ΔE, browning index, PPO, and POD activity values, and a low total bacteria count (p < 0.05). The US+LB group also had the highest sensory score (except for aroma and texture; p > 0.05).

        Several tests and experimental work have been done for identifying the best growth conditions and accumulated amount of lipid moiety in B. braunii, a microalga(UTEX 572) in terms of media composition. The specific growth rate was found to be the highest at 0.15 g/L-day when the phosphorus concentration was doubled with the other ingredients at the normal level. Experiments for varied media compositions revealed that the accumulation of lipid was the highest at 48% (dry cell weight based) in the nitrogen deficient medium and its corresponding specific growth rate was comparative to that in the normal BG 11 medium. In the bubble column experiments, carbon dioxide containing air produced four times more cell mass than air only. Light and glucose addition also enhanced cell mass with maximum, 1.8 g/L and accordingly 42% of lipid composition, which turned out to be a better strategy for higher lipid-producing microalgal culture.

        In this study, optimization of anaerobic co-digestion for food and livestock wastes was studied by an experimental design method. A central composite design (CCD) was applied in designing experiments. Selected two independent variables for this study were initial substrate concentration and mixing rate of livestock wastes. The ranges of experiment for initial substrate concentration and mixing rate of livestock wastes were 2~10 g-VS/L and 0~100%, respectively. Selected responses were methane yield, maximum methane production rate and volatile solids (VS) removal rate. The experimental design was analyzed using a response surface methodology (RSM). Models obtained by the RSM were analyzed by analysis of variance (ANOVA). ANOVA demonstrated that the models were highly significant. Optimal conditions obtained for the models were initial substrate concentration of 2.1 g-VS/L and mixing rate of livestock wastes of 48.8%, respectively. The measured values under the optimal conditions were well in agreement with the predicted values from the models. Thus, it showed that the CCD and RSM were appropriate for determination of an optimal mixing condition in the anaerobic co-digestion process for food and livestock wastes.

        This study evaluated the germination rate of Rheum palmatum L. in Korea as affected by storage temperature, germination temperature, and a soaking treatment. The germination rate of R. palmatum L. stored at various conditions for 8 weeks was 〉 60%. The highest germination rate occurred at 25℃. The germination point of R. palmatum L. with soaking was started 1 day after application, and non soaked seeds germinated in 2 days. When R. palmatum L. was soaked with water, the germination rate was lower, but mean germination velocity was the highest. These results may help our understanding of variations in germination characteristics for seeds treated under different germination conditions.

        This study was conducted to develop a functional feed additive for pig with spent Lycium chinense Mill fruit. We investigated the optimum conditions for the extraction of polyphenol from spent Lycium chinense Mill using methanol. Methanol concentration as a solvent for extraction, extraction time and the volume of solvent per a gram of solid (ground spent Lyceum chinense Mill) were selected as parameters. Three levels of parameters were configured according to Box Behnken experiment design, a fractional factorial design, and total 15 trials were employed. Total polyphenol concentration from each trial was used as response from experiment system and effects of parameters on total polyphenol extraction efficiency were determined using response surface model. As a result, all terms in analysis of variance, regression (p = 0.001), linear (p = 0.002), square (p = 0.017) and interaction (p = 0.047) was significant and adjusted determination coefficient (R2) was 94.7%. Total polyphenol extraction efficiency was elevated along increased methanol content and decreased solvent to solid ratio. However extraction time did not affect the efficiency. This study provides a primary information for the optimum extraction conditions to maximize total polyphenol recovery from spent Lycium chinens Mill fruit and this result could be applied to re-use of argo-industrial by-products and to develop of functional feed additives in organic farming.

        본 실험은 반응표면분석법을 이용하여 진피 에탄올 추출물의 이화학적 특성을 모니터링 하였다. 진피의 에탄올 추출조건의 최적화를 위하여 예비실험 결과에 따라 요인변수들 중 시료대 용매비를 20 mL/g로 고정하여 반응표면을 나타내었다. 추출조건에 따른 수율 및 총 폴리페놀함량의 최대값은 예측된 정상점에서 능선분석을 실시하여 본 결과, 36.31%와 13.86 mg/mL이었다. 추출조건별 총 플라보노이드함량은 3.19~6.67 mg/mL의 범위였으며,

        본 연구는 증절간 고구마 제조를 위하여 냉풍건조 방법을 이용하여 건조조건별 증절간 건조 고구마의 건조특성, 물성 및 관능적 특성을 연구하였다. 건조시 수분함량은 48시간에서 4.98%로 가장 낮은 수치를 보였고, 색차에서 건조온도가 낮을수록, 건조시간이 짧을수록 L값이 높아졌으며 a, b, 값은 건조온도가 높을수록 건조시간이 길어짐에 따라 증가하는 경향을 보였다. 가용성 고형분은 건조 조건별로 19~70 의 범위를 나타내었고, 환원당은 98.7
