        본 연구에서는 한국 및 중국산 감초(Glycyrrhiza uralensis), 그리고 우즈베키스탄산 감초(Glycyrrhizaglabra)를 대상으로 추출용매, 추출온도, 추출시간 등 추출조건별 추출물을 제조하고 이들 추출물들의 추출 수율과 항산화 활성을 비교하여 최적의 추출조건을 선정하였다. 항산화 활성 중 자유 라디칼(1,1-phenyl-2-picrylhydrazyl,DPPH) 소거활성은 85% 에탄올로 60 ℃에서 6 h 동안 추출한 조건에서 한국 감초로부터 얻은추출물이 가장 높은 활성(46.05%)을 나타내었다. 루미놀 발광법을 이용한 추출물들의 총 항산화능 측정 실험과피부 광노화에 중요한 1O2으로 유도된 세포막 손상에 대한 추출물들의 세포 보호 효과를 측정한 실험 모두에서도 위의 조건에서 가장 높은 항산화 활성을 나타내었다. 특히, 한국 감초는 τ50이 116.4 min으로 비교 물질인(+)-α-tocopherol (28.5 min)보다 약 4 배나 높은 세포 보호 효과를 나타내었으며, 추출 수율은 18.75%로우즈베키스탄 및 중국 감초보다 각각 1.2 배 및 2.5 배의 추출 수율을 나타내었다. 따라서, 본 연구 결과는 항산화 소재로 화장품에 응용하기 위하여 감초로부터 추출물을 얻기 위한 최적의 조건은 85% 에탄올로 60 ℃에서6시간 동안 추출하는 것임을 보여주었다.

        본 연구는 색소의 종류가 다른 흑미와 적미의 생리기능 성을 구명하여 천연 기능성 식품 소재로의 이용을 위한 기초자료를 제공하고자 일반 벼인 일품을 포함하여 흑미인 흑광, 흑설 및 적미인 적진주, 홍진주을 실험재료로 항산화 활성 및 암세포 성장 억제에 대한 실험을 수행하였다. 추출 수율 및 총 페놀함량은 흑미가 적미 보다 높았다. 총 페놀함 량에서 흑미와 적미는 일반벼 보다 4배 이상 높은 수준이었 다. DPPH radical 소거능은 적미와 흑미 간에는 차이가 없었 으나 품종 간에는 홍진주, 흑광, 흑설, 적진주 순서로 높았 다. 반면, ABTS 항산화력은 적미보다 흑미가 높았다. 이는 총 페놀화합물 함량 결과와 경향이 같았다. 암세포 성장 억제 능 측정 결과, 적미는 폐암세포 성장 억제 효과가 흑미 보다 높았다. 반면, 유방암 세포 성장 억제 능은 흑미와 적미 간에는 차이가 없었으며, 품종 간에는 적진주, 흑광, 홍진주, 흑설 순으로 높았다. 흑미와 적미의 생리기능성 비교 결과, 항산화 활성은 흑미가 높은 반면, 적미는 폐암세 포 성장 억제 효과에서 높았다.

        This study was designed to analyze the chemical composition of essential oil in ‘Shiranuhi’ immature fruitand to test their biological activities. ‘Shiranuhi’ immature essential oils (SIEO) were obtained by steam distillation fromfruits collected from Jeju Island and were analyzed using gas chromatograph (GC)-flame ionization detectors (FID) andGC-MS. Fourteen components were identified in the essential oil. Limonene (75.21%) and terpineol (8.68%) were the majorcomponents in SIEO. Since acne vulgaris is the combined result of a bacterial infection and the inflammatory response tothat infection, we examined whether SIEO possessed antibacterial against skin pathogens. As a result, SIEO showed excel-lent antibacterial activities against drug-susceptible and -resistant Propionibacterium acnes and Staphylococcus epidermidis,which are acne-causing bacteria. In this study, SIEO was examined on DPPH radical scavenging activities, which showedmoderate antioxidant activity (SC50, 15.36µL/mL). In order to determined whether SIEO can be safely applied to humanskin, the cytotoxicity effects of SIEO were determined by colorimetric MTT assays in normal human fibroblasts and kerati-nocyte HaCaT cells. They exhibited low cytotoxicity at 0.5µL/mL in both celllines. Based on these results, we suggest thepossibility that essential oil of ‘Shiranuhi’ maybe considered as an antibacterial and antioxidant agent.

        This study was conducted to analyze the polyphenol contents and antioxidant activities of small-black-bean- Chungkukjang-added black food (SBCB) and soybean Chungkukjang (SC) extracts for the development of functional materials. The yields of the hot-water-and-ethanol SC extracts were higher than those of the hot-water-only SC extracts, which were higher than those of the ethanol-only SC extracts. The total phenol contents of the hot-water-and-ethanol SBCB extracts were higher than those of the other extracts. The EDA values of the hot-water-and-ethanol SBCB extracts were higher in the 0.625~5.0 mL/mL extract concentration than those of the other extracts, and those of the SC extracts were high in the 10 mg/mL extract concentration. The SOD-like activities of the hot-water- and ethanol-only SC extracts were higher than those of the other extracts. The nitrite scavenging ability of the ethanol-only SC extracts was higher at pH 1.2 than that of the hot-water-only SC extracts. The xanthine oxidase inhibitory activities of the hot-water- and ethanol-only SBCB extracts were higher in the 0.625 mL/mL extract concentration than those of the other extracts, and increased along with the extract concentration. The inhibitory activities of tyrosinase also increased along with the extract concentration, and the reducing power increased along with the extract concentration and was high in the hot-water-only SBCB and SC extracts.

        This study was carried out to optimize the filtration, clarification, anti-browning processing conditions of clear pear juice and to investigate changes in antioxidant activity of pear juice produced through different heating treatment. For the filtration with cheese cloth, filter paper, or centrifugation (10 min at 3,000 rpm), the pear juice was most efficiently filtered with centrifugation because it showed the highest lightness (L value) and lowest yellowness (a value). Among various clarifying agents, 1% of gelatin or bentonite clarified effectively pear juice but tannin or egg albumin did not. Among anti-browning agents (0.1%) like L-ascorbic acid, NaCl or citric acid, L-ascorbic acid prevented the browning of pear juice with the lowest browning index value (2.62), compared to that of NaCl (2.74), or citric acid (2.87). Fructose, sucrose, glucose and sorbitol were present in the pear juice, the fructose and glucose contents increased but that of sucrose decreased in the heated pear juice. The total polyphenol content of the heated pear juice significantly increase, and did the total flavonoid contents in the clear and heated pear juice. The DPPH radical scavenging activity and nitrate scavenging activity were higher in the clear and heated pear juice than in the fruit crush.

        The antioxidant and photoprotective effects of various extracts from the roots of Rumex crispus L. were evaluated. The concentrations (IC50) of various extracts required to exert a 50% reducing effect on a DPPH radical were found to be 0.005~0.093 mg/mL. The ethyl acetate extract showed a more remarkable effect than the positive control ascorbic acid. The concentrations (QC50) of the butanol and ethyl acetate extracts required to exert a 50% reducing effect on the singlet oxygen 1O2 were found to be 0.464 and 0.365 mg/mL, respectively. Both extracts were also found to protect the in vitro biological system from the detrimental effect of a singlet oxygen 1O2 on type II photosensitization in E. coli and genomic DNA. Among all the tested extracts, the ethyl acetate and butanol extracts contained higher amounts of total phenolic contents. The results suggest that our study may contribute to the development of new bioactive products with potential applications to the reduction of photo-produced oxidative stress involving reactive oxygen species in living organisms

        This study was conducted to investigate the antioxidant and antimicrobial activities of the hot-water and methanol extracts of raw and black Doraji to increase its utilization. In order to prepare the black Doraji, it was steamed for 15 days at a temperature of 60℃ and then it was dried at a temperature of 30℃ for 3 hr. The methanol extract from the black Doraji (BM) contained the highest levels of total polyphenols among the extracts, and the total polyphenol content of the extract from a black Doraji was higher than that of the extract from a raw Doraji. The total flavonoid contents of the hot-water extract from a black Doraji (BW) was the highest (7.94 mgQE/g) among the samples. The DPPH and ABTS radical scavenging activities increased according to the increase in the concentrations of the Doraji extracts. The BM has the highest radical scavenging activity among the extracts. Each extract showed a slight difference in the antibacterial activity according to the tested strains. The Black Doraji showed a higher antimicrobial activity compared to the raw Doraji. The hot-water extracts demonstrated higher activities than the methanol extracts, and the BW revealed the strongest activity. In this study, the black Doraji showed more effect of the antioxidant and anti-microbial activities than the raw Doraji. These results will provide fundamental data for improving the sitological value and the black Doraji can be used as a valuable resource for the development of nutraceutical foods.

        Inhibition of α-glucosidasesinvolved in the digestion and absorption of carbohydrates can decrease the postprandial increase of blood glucose level after a mixed carbohydrate diet. Therefore, we investigated the changes in α-glucosidase inhibitory activity of wheat during germination process. We also investigated total phenolics and oxygen radical absorbance capacity (ORAC) during germination process of wheat. Wheat was germinated for 0, 12, 24 and 48 hrs and results showed that both phenolic contents and α-glucosidase inhibitory activities increased with germination time. More specifically, total phenolic content increased from (293mg/100g-F.W.) at 0 hrs germination to a high (4,082mg/100g-F.W.) achieved after 48 hrs germination. Additionally, alpha-glucosidase inhibitory activity was dramatically increased from a low 10% inhibition at 0 hrs germination to a high 60% inhibitory activity observed at 48 hrs. These results suggest that wheat, depending on the germination time, has the potential to contribute as a dietary supplement for controlling hyperglycemia and oxidative stress-linked diabetes complications. It may due to the natural response of plant seed to overcome the biotic/abiotic stress from environment during germination period.

        The organic acids, fuel oil contents and antioxidant activities of Yakju with the addition of rice varieties were investigated. The rice cultivars that were used in this study were Ilmi rice, Hopyung rice, Hopum rice, Hanarum rice, Anda rice and Indica rice. The study results are as follows: the pH values of Ilmi Yakju and Hanarum Yakju were 4.14 and 4.07, respectively, and those of the other Yakjus, 3.92~3.98. The total titratable acid content of the Yakju for which Indica rice was imported from Thailand was highest among the samples. The reducing sugars content of the Yakju for which Indica rice was used was higher than that of the other samples. The ethanol content of the Hanarum Yakju was higher than that of the other Yakjus, and the lowest ethanol content was found for the Indica Yakju. The major organic acid component of the Yakju was lactic acid. The total organic acid content was highest in the Indica rice Yakju. The amounts of the following main fusel oils were highest, in the following order: 2-butyl alcohol, 2-pentanol, N-propyl alcohol, acetaldehyde, ethyl acetate, methyl alcohol and 2-methyl-1-propyl alcohol. The DPPH free radical scavenging activity of the Indica rice Yakju was much higher than that of the Ilmi rice Yakju and the Hanarum rice Yakju. Also, the SOD activity of the Indica rice Yakju was strongest among all the samples. The total polyphenol content of the Indica rice Yakju was the highest among all the samples.

        The nutritional composition, bioactive components and antioxidant activities of the first and second sprouts of Kalopanax pictus, Cedrela sinensis and Acanthopanax cortex were investigated to increase the utilization of these sprouts. The moisture and crude lipid contents of the first sprout were higher than those of the second sprout, and the crude fiber and carbohydrate contents were higher in the second sprout. The organic acid content of the first sprout was higher than that of the second sprout. The second sprout had higher free amino acid contents in K. pictus and C. sinensis, and the first sprout had a higher content in A. cortex. Especially, the second sprout of K. pictus and C. sinensis had a higher level of glutamic acid, which is a major taste component of foods, than the first sprout. The second sprouts of all the samples had higher mineral levels than the first sprouts. In contrast, the first sprouts had higher total polyphenol and flavonoid contents and showed a higher antioxidant activity level, except for the DPPH radical scavenging activity of the A. cortex. The results of this study show that the nutritive value of the second sprout was higher than that of the first sprout, but the bioactive components value of the second sprout was lower than that of the first sprout. Therefore, additional research is needed on the cultivation condition required to maintain the nutritive values and the antioxidant activities of these sprouts.

        We studied the total polyphenol content, DPPH radical scavenging activity, hydroxyl radical scavenging activity and α-glucosidase inhibition of water extracts from 17 medicinal plants. Total polyphenol contents ranged from 10.0 (Coix lachryma-jobi L, CL) ~ 279.7 (Perilla sikokiana, PS)mg/g. The water extract from medicinal plants were evaluated for its free radical scavenging activities and compared with a commercial antioxidant, ascorbic acid. DPPH radical scavenging activity of Pyrus pyrifolia (PP), Chamaecyparis obtusa L. (COL), Chamaecyparis obtusa F. (COF), and PS were higher than positive control. Higher hydroxyl radical scavenging activity were shown in Acanthopanax senticosus (AS) and Cordyceps militaris (CM) than the other plants. The highest anti-α-glucosidase activity was observed in Cornus officinalis (CO) and Paeonia suffruticosa Andrews (PSA) water extracts. PSA showed not only the higher DPPH radical scavenging activity but also the anti-α-glucosidase activity. The results of our study that PP, COL, COF, PS, AS, CM, CO and PSA could be potential candidates for natural antioxidants.

        Dried leaves of Raphanus sativus L. and Angelica keiskei L. extracts have strong antioxidant potential and in consequence profound effects on ulcerative colitis. Present study was conducted to explore the effect of diet mixtures containing dried leaves of Raphanus sativus and Angelica keiskei powder on ulcerative colitis in mice and antioxidant potential of radish green and Angelica extracts as well. Both dried leaves of Raphanus sativus and Angelica keiskei extracts exhibited higher antioxidant activity due to the presence of polyphenols, favonoids and antioxidant enzymes like superoxide dismutase (SOD). Analyzed data indicate that Angelica keiskei extract had higher polyphenol and flavonoid contents compared with radish green, while maximum SOD activity was noted in dried leaves of Raphanus sativus extract Likewise, higher antioxidant (348.72 ± 31.65 μg/mL) and ABTS+ radical scavenging (17%) activities were noted in dried leaves of Raphanus sativus extract compared with Angelica keiskei.

        This study was conducted to investigate the antimicrobial activities and antioxidant activities of the Korean mistletoe extract and its solvent fractions (e.g. n-hexane, ethyl ether, ethyl acetate, butanol). Ethyl ether fraction against Bacillus cereus showed stronger activities than benzoic acid (2.5 mg/mL). The MIC of korean mistletoe extract and slovent fractions were in the range of 6.25-25 mg/mL. The MIC (6.25 mg/mL) of ethyl acetate fraction onto Staphylocossus aureus was the lowest among them. Ethyl ether fraction which showed the strongest antioxidant activities by DPPH (1,1-diphenyl-2-picryl-hydrazyl) and FRAP (ferric ion reducing antioxidant power) methods had the highest total phenolic contents. It is suggested that Korean mistletoe could be utilized as natural preservative material through the study of the active compounds from ethyl ether fraction.

        This study was conducted to analyze the antioxidant activities of fermented black jujube and to compare these with those of dried jujube, for the development of functional materials. The antioxidative activities of dried jujube and fermented black jujube extracts were analyzed by electron-donating ability (EDA) using 1,1-diphenyl-2-picryl hydrazyl (DPPH), superoxide-dismutase-(SOD)-like activity by pyrogallol, nitrite-scavenging ability, and xanthin oxidase. The yield of the fermented black jujube extracts was higher than that of the dried jujube extracts, and that of the ethanol extracts was higher than that of the hot-water extracts. The total phenol contents of the hot-water extracts from fermented black jujube were higher. The EDA values of the hot-water and ethanol extracts from fermented black jujube and dried jujube increased with an increase in extract concentration, and were about 85% in a 1000 ㎍/mL extract concentration. The SOD-like activity increased with an increase in extract concentration. The SOD-like activity of the hot-water extract from fermented black jujube was higher than that of the other extracts. The nitrite-scavenging ability at pH 1.2 of the hot-water extracts from dried jujube was higher than that of the other extracts. The xanthine oxidase inhibitory activities of the hot-water and ethanol extracts from fermented black jujube were higher than those of the other extracts, and increased along with the concentrations of the extracts.
