Harlequin ladybird (Harmonia axyridis (Pallas, 1773)) is an invasive species originating from Asia, posing a potential threat to the ecosystem and the wine industry in New Zealand due to wine taint, although it can also be a useful biocontrol agent. In this study, the response profiles of antennal olfactory receptor neurons (ORNs) to 32 plant volatiles were examined in male and female H. axyridis, using the single sensillum recording technique. Various types of ORNs were identified from four types of olfactory sensilla in both male and female H. axyridis, with no sexual dimorphism. The most abundant type of sensilla contained two ORNs exhibiting highly specialized responses to methyl benzoate and β-caryophyllene, respectively. Another type of sensilla also contained two specialized ORNs, one responsive to geranyl acetate and the other to some aromatic compounds such as 2-phenylethanol, benzyl acetate, methyl benzoate, and methyl phenylacetate. In contrast, two other types of sensilla contained broadly tuned ORNs, one containing ORN(s) responsive to six-carbon alcohols such as (Z)-3-hexen-1-ol, 1-hexanol, and isomers of 2-hexen-1-ol as well as some other non-alcohol green leaf volatiles, and the other containing ORN(s) exhibiting responses to β-myrcene, geraniol, linalool, nerol, benzyl acetate, and methyl phenylacetate. This study suggests that H. axyridis possesses a set of ORNs specialized for specific plant volatiles, providing insights into the olfactory communication system of this species and potential volatiles to be used for trapping this insect.
본 연구에서는 세 가지 온도(20, 25, 30℃)에서 꼬마남생이무당벌레의 생물적 특성을 조사하였다. 복숭아혹진딧물 약충을 먹이로 사육한 꼬마남생이무당벌레 암컷의 성충기간은 20, 25, 30℃에서 각각 134.0, 101.0, 55.2일이었고, 총 산란수는 각각 508.6, 875.6, 383.4개였고, 수 컷의 성충기간은 각각 128.8, 97.8 및 46.5일이었다. 25℃에서 꼬마남생이무당벌레의 1령, 2령, 3령, 4령, 수컷 및 암컷의 복숭아혹진딧물 성충 의 일일 평균포식량은 각각 2.2, 7.3, 14.5, 29.1, 35.0 및 42.9마리이었고, 목화진딧물 성충의 일일 평균포식량은 각각 2.2, 7.5, 13.9, 29.5, 37.0 및 40.8마리이었다.
무당벌레(Harmonia axyridis)는 종내에서 초시색상패턴이 매우 다양하게 존재한다. 본 논문에서는 서로 다른 색상패턴의 무당벌레를 대상 으로 amplified fragment length polymorphism (AFLP)을 실시하여 무당벌레의 초시색상 패턴간 유전형질의 차이를 확인하고자 하였다. 총 28 개의 프라이머 조합으로 실험을 실시한 결과, 총 2,741 개의 밴드가 검출되었다. 그 중 20 개의 밴드(S1-S20)만이 특정 색상패턴에서 나타났다. 이들 가운데 9 개의 밴드를 색상에 연관된 AFLP 후보 지표로 선발하였다. 밴드 가운데 S1과 S2, S20은 Succinea 1, 2 변이형에 공통적으로 나타났으며, S3와 S5는 Conspicua 변이형에 특이적이었다. 또한 S13는 Spectabilis 변이형에, S15와 S18, S19는 Succinea 2 변이형에 특이적이었다. 특정 색상패턴에만 나타나는 9 개의 AFLP 지표들은 cloning을 통해 염기서열 분석을 실시하였고, GenBank를 이용해 다른 염기 서열과 비교를 해보았지만 아무런 상동성도 찾을 수가 없었다. 무당벌레 종 내 유전적 다양성을 평가한 결과, Spectabilis가 Conspicua보다 Succinea 변이형에 높은 유사성을 보였다. 색상에 연관된 AFLP 후보 지표를 기준으로 sequence characterized amplified region (SCAR) 지표로 변환하여 9 개의 AFLP 분자지표들 가운데에서 5 개만이 SCAR 지표로 전환될 수 있었으며, 이를 통해 AFLP 지표가 무당벌레의 색상과 연관되어 있는지 확인할 수 있었다.
경기도 8개 지역에서 2010년부터 2012년 동안 식균성인 노랑무당벌레의 발생기주를 조사한 결과, 흰가루병에 감염된 12종의 식물에서 관찰 이 되었다. 특히 가장 밀도가 높았던 배과원에서 노랑무당벌레는 7월 상순부터 11월 상순까지 발견되었다. 식균성인 노랑무당벌레의 장내에서는 흰가루병 균사나 포자 외에 다른 먹이의 흔적이 발견되지 않았고, 알과 번데기를 제외한 전 발육단계에서 균을 섭식하는 특성을 볼 때 절대적 식균성 곤충으로 생각된다. 25℃에서 오이 흰가루병균을 섭식한 노랑무당벌레의 발육기간은 알, 유충, 번데기, 성충이 각각 3.9, 10.4, 4.1, 37.7일 이었고, 발육단계별 오이 흰가루병 섭식량은 45.6, 144.4, 372.2, 628.1, 473.7 mm 2로 4령, 성충, 3령, 2령, 1령 순으로 많았다. 본 연구를 통해 노랑무당벌 레의 오이 흰가루병에 대한 섭식능력을 바탕으로 향후 유용 토착천적으로써 대량사육기술, 저독성 약제 선발 등 작물 흰가루병 종합방제기술(IPM) 에 대한 연구가 필요하리라 사료된다.
철모깍지벌레의 생물적 방제를 위한 애홍점박이무당벌레의 포식능력과 기초 생태특성을 25℃± 2℃, 70% ± 5%, 16L : 8D로 설정된 사 육실에서 검정한 결과, 애홍점박이무당벌레 성충은 하루에 77.5마리의 철모깍지벌레를 포식하여, 깍지무당벌레 포식력의 2.9배, 어리줄풀잠자 리 포식력의 2.8배 이상의 뛰어난 포식능력이 관찰되었다. 1령 ~ 4령 유충 기간 동안 659.0마리의 철모깍지벌레를 포식하였고, 애홍점박이무당 벌레와 철모깍지벌레의 밀도 별 상호작용을 조사한 결과 Holling (1959)의 기능반응곡선 제 II 형과 유사한 포식반응을 확인하였다. 애홍점박이 무당벌레는 철모깍지벌레를 먹이로 공급했을 때 알에서 성충까지 21.1일 ~ 27.9일이 소요되었으며, 72.3%, 77.8%, 83.3%의 높은 부화율, 용화 율과 우화율을 보였다.
This study is the first report about the toxicity of pesticides to the mycophagous predator, I. koebelei, of powdery mildew of agricultural crops. Pesticides we tested are composed of synthetic and environmental-friendly products and being used conventionally for the control of insect or microbial pests on cucumber in Korea. our study was conducted to determine the relative toxicities of several pesticides used in Korea cucumber production to mycophagous natural enemy, I. koebelei and to provide a background for implementation of integrated powdery mildew management programs. Based on IOBC classification, three insecticides, bifenthrin + imidacloprid, acetamiprid + indoxacarb, acetamiprid + etopheprox are classified as having a Class 4 (harmful). Spiromesifen showed the low toxicity to the survival and the fecundity of I. koebelei when this chemical had been exposed to 3rd larva or newly emerged adult via feeding with cucumber powdery mildew. However, pyriproxyfen not only decreased the fecundity of female adult but also strongly prohibited from pupation. Many commercial biological or botanical pesticides can restrict the population of I. koebelei. However, Q pact (a.i. Ampelomyces quisqualis 94013), Top seed (a.i. Paenibacillus polymyxa AC-1), BT one (Bacillus thuringiensis) and Solbitchae (insecticidal microorganism) had no toxicity to I. koebelei when this chemical had been exposed to 3rd larva or newly emerged adult feeding with cucumber powdery mildew.
Cynegetis impunctata (Linnaeus, 1767), a phytophagous lady beetle, was recorded in South Korea for the first time recently. Previously, only the adult form of this species had been described. In this study, we provide a redescription of the adult and the first descriptions of the larva and pupa. Diagnostic characters for this species are given, and intraspecific color variation in Cynegetis impunctata adults is discussed.
The yellow spotless ladybug (YSL), Illeis koebelei is a kind of aboriginal ladybug in Korea. YSL feed on the fungi and extraordinary prefer to powdery mildew (PM). This study was conducted to investigate the developmental characteristic and the biological control effect on the cucumber PM by the YSL from 2010 to 2012. YSL were detected on the 12 species of host plants infected with PM. and occurrence was observed from the early of July to the early of November in Gyeonggi area. The feeding amount of YSL on the cucumber PM was in order of 4th larva, adult, 3rd larva, 2nd larva and 1st larva. The optimum developmental temperature and photoperiod for YSL was 20∼25℃ and 16L:8D respectively. Three species of fungi were selected as the short term substitution feed for YSL. Control effect on cucumber PM under 2.5% of occurrence rate was linearly increased in proportion to increased numbers of 3rd larva were applied. Combination treatment of the fungal agent with YSL enhanced suppression of cucumber PM more than either agent used alone. The safety agricultural chemicals were selected harmless to the survival of larva and adult of YSL. The survival rate of pupae and adult was over 80% after storage at 10∼12℃ for 30 days. Five species of banker plant were selected for maintenance of YSL during growing season in greenhouse.
 ,  , The effects of gamma irradiation on the survival, development, fecundity, and sex ratio of the multicolored Asian ladybird beetle (Harmonia axyridis) were investigated. A dose of more than 50 Gy dose of irradiation on eggs completely inhibited hatch, and 1st and 3rd instar larva irradiated with over 40 Gy almost died. Adverse effects of gamma irradiation over 70 Gy were also observed in the survival and development of pupae and adults. tail indicating DNA damage was showed in survival individuals exposed to gamma irradiation.Although irradiated individuals usually survived, they had longer periods of larval and pupal development than untreated individuals. Slight female-biased sex ratios were observed in some treatments. However, sex ratio differences soon equilibrated. Generally, inherited harmful effects of gamma irradiation were observed in F1 and F2 generations. Comet tail indicating DNA damage was showed in survival individuals exposed to gamma irradiation.
The seven-spotted lady beetle, Coccinella septempunctata (Coleoptera: Coccinellidae), known also as the seven-spot ladybird, is natural enemy for aphids and has a broad ecological range, living almost anywhere there are aphids for it to eat. In order to understand the genetic diversity and geographic variation of the species we sequenced a portion of mitochondrial COI gene (658 bp) and complete nuclear internal transcribed spacer 2 (ITS2) collected from nine Korean localities. A total of 21 haplotypes (CSCOI01 ~ CSCOI21), with the maximum sequence divergence of 4.56% (30 bp) were obtained from COI gene sequence (from 78 individuals), whereas 65 sequence types (CSITS201 ~ CSITS265), with the maximum sequence divergence of 2.06% (11 positions) were obtained from ITS2 (from 79 individuals), indicating substantially larger sequence divergence in mitochondrial gene sequence. Both COI gene and ITS2 shows the distribution pattern that only a few haplotypes or sequence types are widely distributed, whereas majority of them are highly restricted in one geographic location, even represented as a single individual. Unlikely the ITS2 sequence types the mitochondrial COI haplotypes evidenced the presence of two main phylogenetic groups, reciprocally monophyletic to each other. Geographically, these two groups are spread in all localities surveyed. Considering both COI gene and ITS2 sequence together, current our data may suggest the presence of ancestral polymorphism, rather than on-going speciation, but more scrutinized analysis will be performed soon. Due partially by the presence of both COI groups in all surveyed localities, the genetic diversity estimates of all localities are similar from the perspective of COI gene, but ITS data showed extremely lower genetic diversity of one islet locality, Anmyeon-do (locality 2; 0.002530 vs. 0.008054 ~ 0.012060). Analysis of gene flew estimates between localities indicates that most populations are highly interconnected to each other. However, one islet locality, Anmyeon-do (locality) has shown statistically significant distance from the remaining localities on the basis of only ITS2 data (FST = 0.19 ~ 0.34), requiring scrutinized phylogeographic inference on this population with expanded sampling. As more scrutinized analysis is performed, further fruitful inference on the geographic contour of the species might be available.
The seven-spotted lady beetle, Coccinella septempunctata (Coleoptera: Coccinellidae) has a broad ecological range, living almost anywhere there are aphids for feeding. In this study, we isolated and characterized a total of 10 microsatellite loci from the species. The loci were validated and characterized using 25 samples collected from five Korean localities. The number of alleles and heterozygosity observed at each locus ranged from 4 to 16 and from 0.37 to 0.89, respectively. None of the loci deviated significantly from Hardy-Weinberg equilibrium and there was no indication of significant linkage disequilibrium among pairs of loci. These microsatellite markers should be very valuable markers for population genetic studies of Coccinella septempunctata.
Although the greater part of Coccinellidae are all pure entomophagous insects (Weber and Lundgren, 2009), many species of phytophagous, mycophagous and polyphagous ladybirds have been reported in worldwide. This study was conducted to investigate the morphological characteristics of mouthparts of mycophagous ladybird, I. koebelei, comparison with aphidophagous species, Propylea japonica (Coccinellidae: Coccidellini), and developmental charicteristics of I. koebelei at different temperature under incubation conditions. Four species of powdery mildews that I. koebelei fed were observed, and their host plants were pear tree, persimmon tree, red pepper and cucumber. The most conspicuous elements of I. koebelei mouthparts is the mandible. The distal incisor region of the mandible of both species is formed by two teeth, giving them a bifid apex. and the terminal teeth spilt near top and the ventral tooth is smaller than the dorsal one. but the ventral tooth of I. koebelei divided into a row of additional teeth was different with P. japonica. Furthermore, while the inner mandibular cutting edge of P. japonica is smooth, that of I. koebelei is covered in minute teeth, forming a comb. These structures are presumed to help I. koebelei rake fungal spores growing on leaf surface. The developmental period and reproduction ability of were significantly different according to temperature conditions when I. koebelei was fed the powdery mildew from cucumber leaves. The optimum temperature for I. koebelei rearing was from 23 to 25℃ that is the similar condition of cucumber powdery mildew occurrence.
The seasonal fluctuation of the large 28-spotted lady beetle, Henosepilachna vigintioctomaculata (Motschulsky), was observed in potato fields of Gangneung region, Gangwondo Province. The effects of their leaf consumption over a 24 hour period and developmental periods by different temperatures were investigated, and then the developmental threshold (DT) and effective cumulative temperature (ET) of each stage were calculated. Various developmental stages of H. vigintioctomaculata were captured in the fields from May to September, with three peaks in late May, late June, and late August. Leaf consumption by larval stages increased with the development of instar. The 4th instar larvae consumed potato leaves much more than other stages. Egg hatchability and emergence rate of H. vigintioctomaculata were lower at 10℃ and 30℃, but higher at around 20∼23℃. The developmental periods of each stage became shorter as temperature increased, 90 days at 10℃, 40 days at 20℃ and 25 days at 25℃. Base on developmental velocity to temperature, calculated DT and ET were 7.3℃ and 83.
There was different between two differential geographical and environmental condition areas on elytra color expression patterns of the multicolored Asian lady beetles (Harmonia axyridis). Especially, it was investigated that expression rates of melanic patterns (conspicua, spectabilis and axyridis) relatively increased in overwintering populations collected in highly mean temperature and longer cumulative daylength area. In addition, in the same collection site, the seasonal difference had influenced on color patterns of H. axyridis. Although these effects didn"t were not observed in the laboratory, environmental conditions such as temperature or cumulative daylength might be factors that gave an effect on color pattern formation.
Menochilus sexmaculata는 무당벌레아과에 속하며 몸길이가 3.3-6.2mm의 중대형 무 당벌레로서 오스트레일리아구, 에티오피아구, 동양구와 일본(류큐, 혼슈, 시코쿠, 큐 슈, 쓰시마)에 이르는 지역에 분포하는 종이었다. 대부분의 분포지역에서 이 종은 무 당벌레과 내의 우점종으로 진딧물 천적으로 유용하게 이용되어 왔다. 형태적으로 이 종의 개체들은 체색과 무늬 변이가 매우 심하며, 그로 인해 Micraspis discolor나 Chilocorus nigrita와 같은 종으로 오동정된 사례가 매우 많다. 국내에서도 1985과 1986년 무당벌레 목록에 이 종이 수록되었던 적이 있으나, 표본 확인 결과로 Harmonia yedoensis 변이체로 확인된 바 있다. Menochilus sexmaculata의 국내 존재는 2001년 부산 고신대의 소장표본에서 2개체가 확인되면서이다. 2007년 고신대의 소장 표본을 2차로 조사하여 총 31개체의 표본을 확인하였다. 체색 변이는 온대형으로, 구조형질로서 수컷생식기는 일본 및 대만산 대 조표본과 일치하는 것으로 나타났다. 채집량은 2000년과 2003년을 제외하고는 매년 채집이 되었으며, 부산 북구와 부산 동쪽해안을 제외한 대부분의 부산광역시(1군 10 개구)를 아우르는 것으로 나타났다. 특히 임의채집을 계속해온 고신대 무당벌레과의 종류별 표본 수로 비교해 볼 때, 이 종은 5위에 해당하는 것으로 비교적 흔한 종에 속하는 것으로 판단되었다. Menochilus sexmaculata의 발견과 분포 양상으로 볼 때, 최근에 부산일대에 정착하 여 확산되는 종으로 의심된다. 따라서 향후 이 개체군의 생태를 밝히고, 확산 양상을 모니터링할 필요가 있을 것으로 판단된다. 아울러 타이완, 일본 본토와 오키나와 등 의 인접 지역의 집단과 분자수준의 비교분석을 통하여 이입 경로의 검토 역시 수행 되어야 할 것이다.
Propylea japonica is small ladybug(approximately 4-5mm) and is met with everywhere in Korea. When there was no aphid, the survival rate of P. japonica was 0% among another instar bugs and 26.7~32.8% among same instar bugs after 48 hours. When there were sufficient aphids, the cannibalism of P. japonica was low, so the survival rate of P. japonica was 78.6~95.8% among another instar bugs and 80.6~100% among same instar bugs after 48 hours. In the comparison of the number of eggs at different oviposition site, P. japonica preferred the corrugated cardboard. In experiment using T-tube, the rate of decoy was 81% at cotton aphid compared with pepper leaf and 62% at cotton aphid compared with artificial diet. In the artificial diet using shrimp and chicken liver, the developmental period from egg to adult was 17days and the survival rate from egg to adult was 66.7% but the vitality was not good. These data suggest that P. japonica can be used effective natural enemy for control of aphids and is needed to research about artificial diet, mass rearing system and control effect in field.
The surface of most coccinellids (particularly the elytra) has characteristic colour patterns, which show great variability within many species. Individuals of a species of ladybird often differ from one another in colour and pattern. In case of Harmonia axyridis, environmental factors, such as temperature or food, can probably influence colour and pattern. However, virtually no work has been carried out in the influence of such factor. Therefore, in this study, according to rearing temperature, photoperiod and diet, variation of elytra color patterns were investigated. First instar larva on each color pattern were reared in an incubator at one of two temperatures; 25 and 30℃, under the following photoperiod; L16: D8, L12: D12 and L8: D16 and were provided three species of aphids. And their color pattern of adults were estimated.
The multicolored Asian ladybird beetle, Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae), is common to a wide range of natural and agricultural habitats. Applications of gamma irradiation minimized the losses of stored food and the death or failure of emergence in larval and pupal stages. On the other hand, degrade toxin waste as one of alternative to chemical pesticide for both quarantine and sprout control purposes of storage crop pest. So, we have investigated whether gamma irradiation exposed to eggs, lava, pupa and adults of H. axyridis. It may be affected the emergence, fertility, fecundity, development period and sex ratio of H. axyridis. Some changes of physiological characteristics may be applied to more efficient agents as biological control of several aphids. Insects were exposed to gamma irradiation from 0 to 500 Gy of 60Co depended on their developmental stages. The results showed that the first instar, eggs, third instar, pupae, and adults were more sensitive in order of irradiation dose. And fecundity and fertility of female adults were significantly decreased with increasing gamma irradiation dose at all tested individuals.
The multicolored Asian lady beetles (Harmonia axyridis) has characteristic color patterns, which show great variability within species. Up to now, it has been well known that main factors affected on individual color pattern variations in the population of H. axyridis are external, physical, and environmental characteristics. Indeed, there is as yet no evidence to indicate whether the variation is genetic or environmental factors. Also the factors which produce this variation are unknown in this species, although it is suspected that much of the variation is under genetic control. However, the genetic relationships among many of color types were investigated by observing the progeny of each particular pairs. It is worth mentioning a few particular breeding cases to illustrate certain facets of variability, and to indicate examples suitable for genetic analysis of the color pattern variation.
The multicolored Asian ladybird beetle (Harmonia axyridis) has been commonly used with biological control agents for control of several kinds of aphids in agroecosystems. Also, environment friendly agricultural materials have been commonly applied in crop fields because the government held down pesticide application and environment friendly agricultures are gradually increased with consumer"s desires. The multicolored Asian ladybird beetles may be directly or indirectly under the influence of environment friendly agricultural materials in crop fields. The insecticidal environment friendly agricultural materials (IEFAMs) might be safe against each developmental stage of multicolored Asian ladybird beetle. Fungicidal environment friendly agricultural materials (FEFAMs) had a miner effect to each developmental stage of multicolored Asian ladybird beetle with the exception of FEFAM A. Environment friendly agricultural materials contained useful soil microorganisms (EFAMSMs) C and H might be down the hatching rate of eggs, and EFAMSM A and F had a killing effect to 1st instar of lady beetles. Environment friendly agricultural materials contained plant extracts (EFAMPEs) A and D might be suffered effect a deathblow of egg hatching with lady beetles. Otherwise, there was a miner effect to lady beetles with the rest of tested environment friendly agricultural materials.
