본 연구는 구기자 종자를 초임계유체 추출한 후 in vitro 미백활성과 항균 실험을 실시하였으 며, 추출물을 에멀션에 적용하여 안정성 평가를 수행하기 위해 pH, 점도, 에멀션 입자를 관찰하였다. 그 결과 미백활성의 경우 750 μg/mL에서 81.86 %의 미백 활성을 나타내었으며, 항균효과의 경우 Staphylococcus epidermidis, Candida albicans, Escherichia coli 균에서 항균효과가 나타났다. 또한 추 출물을 함유한 에멀션 물리적 안정성 평가에서 점도와 pH가 28일 동안 큰 변화가 없었으며 유화물 입 자 관찰 시 경시변화에 따른 합일과 분리현상이 확인되지 않았다. 이러한 결과는 화장품소재로서의 구기 자 씨앗 추출물이 유용되게 활용될 수 있는 가능성을 가진 것으로 평가된다.
This study was designed to investigate the effects of Korean Lycii fructus water extract in Al (Aluminum) administered rats. Forty-eight male Sprague-Dawley rats were divided into six groups: Control group, water extract group with 3% Lycii fructus, 1000 and 2000 ppm of Al groups, and 1000 and 2000 ppm of Al with 3% Lycii fructus water extract group. The Al content of rat tissue in the Al administered group was lower than that in rat tissue in the Al with 3% Lycii fructus water extract group. Plasma levels of renin and aldosterone activity was higher in the Al administration group, compared with the 3% Lycii fructus water extract group and Al administered group. Aspartate amino transaminase and alanine amino transaminase activities were elevated in the Al administered group and lower in the 3% Lycii fructus water extract group. Lactate dehydrogenase was lower in the 3% Lycii fructus water extract Al group than in the Al group. Choline acetyltransferase was higher in the 3% Lycii fructus water extract Al group than in the Al group.
This study examined the effects of 2% Lycii fructus powder (LFP) supplementation on lipid metabolism in rats. Male Sprague-Dawley rats were administered 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia. Then, 40 rats were divided into four diet groups: a normal diet group (NC), high cholesterol diet group (HC), normal diet plus 2% Lycii fructus powder (NC-LFP) group, and high cholesterol diet plus 2% Lycii fructus powder (HC-LFP) group. The HC group presented higher growth rates and liver weights than NC and NC-LFP however, growth rates and liver weights in the 2% LFP administered groups gradually decreased. HC also showed increased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels and decreased atherogenic index values, HDL-cholesterol, and phospholipid levels, whereas LFP group showed decreased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels as compared to HC. There were no differences in serum triglyceride, phospholipid, LDL-cholesterol, HDL-cholesterol, and free cholesterol concentrations between the normal diet groups (NC and NC-LFP). The high cholesterol diet groups (HC and HC-LFP) had significant increases in serum alanine aminotransferase (ALT), asparate aminotransferase (AST), alkaline phosphatase (ALP), and lactate dehydrogenase (LDHase) activities. And the 2% LFP administered groups had lower hepatic concentrations of total cholesterol and triglycerides than the HC groups. Overall, the results suggest that Lycii fructus powder has hypochloesterolemic effects by reducing serum and liver cholesterol contents.
This study was done to investigate the antidiabetic and antioxidant effects of Lycii fructus in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Lycii fructus was orally administrated once a day for 7 days. The contents of serum glucose, total cholesterol were significantly decreaed (p<0.05) in Lycii fructus treated group compared to the those of STZ-control group, also content of Triglyceride (TG), Altherogenic index (AI) were decreased, but not statistically significant. But high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were increased in Lycii fructus treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6PDH), glutathione-s-transferase(GST), superoxide dismutase(SOD) were significantly increased(p<0.05), glucose-6-phos-phatase(G-6pase)was significantly decreased(p<0.05) and The glutathione(GSH), glucokinase(GK) were increased. But not statistically significant In Lycii fructus treated group compared to the those of STZ-control group These results indicated that ethanol extract of Lycii fructus would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.
This study was performed to investigate the effect of Lycii fructus beer on serum lipid profiles and antioxidant activity in rat Sprague-Dawley (SD) rats weighting about 190g were divided into the following 5 groups ; distillate water (Control), 5% ethanol in distillate water (Ethanol), commercial beer (CB), Lycii fructus beer (LFB) and 5% alcohol red wine diluted with distillate water (RW). Body weight, total food intake, FER and percent organ (liver, kidney) weight per body weight were not significantly changed by Lycii fructus beer drinking. After 6 weeks, serum total cholesterol, triglyceride and HDL cholesterol level were not significantly different. But, Lycii fructus beer intake tended to decrease serum triglyceride level and atherogenic index. Also, GOT and GPT levels were expressed lower than Ethanol group. There was not significantly different in hepatic glutatiione (GSH) content and glutathione-S-transferase (GST) activities among 5 groups. Lipid peroxidation in the hepatic was decreased by Lycii fructus beer intake. The results demonstrated that Lycii fructus beer was potential and effective antioxidant that can protect the decrease associated with alcohol.
This study was conducted to investigate the effect of physical properties and antioxidant activities of the beer made by Lycii fructus for development of functional beer. Physical properties such as Brix˚, pH and hunter values were determined and compared with commercial beer. L(lightness) value was not significant difference among beer, although a(redness) and b(yellowness) values were higher in Lycii fructus beer than those of the commercial beer. Total phenolic acid contents were 0.790 mg/mL in Lycii fructus beer and 0.603 mg/mL in commercial beer. Electron donatin ability was 93% in Lycii fructus beer and 87% in commercial beer. Lycii fructus beer showed higher SOD-liked activities than in the commercial beer. The relative antioxidant effects of the Lycii fructus beer showed 19% inhibitory effect on the peroxidation of egg yolk lecithin.
The lipid stability of walnut added with water extracts of Ganoderma lucidum, Camellia sinensis and Lycii fructus was studied. The results are as follows : 1. when the extracts were added to walnut, the lipid stability was improved. The antioxidative activities of water extract of Camellia Sinensis and Lycii Fructus were considerably higher than that of Ganoderma lucidum. The antioxidative activity was decreased in the rank order Lycii Fructus$gt;Camellia Sinensis$gt;Ganoderma lucidum. 2. This clearly suggests that water extract of Lycii Fructus at 4℃ Bx level can be used as an alternative natural antioxidant for the lipid stability of walnut.
The storage stability from powdered anchovy and its preparation cooperated with three additives (water extract from green tea and boxthorn and BHT) was studied. The results are as follows: 1. When the additives were added to powdered anchovy, the storage stability was improved. The antioxidative activities of water extract from green tea and boxthorn were considerably higher than BHT. The antioxidative activities were decreased in the rank order boxthorn $gt; green tea $gt; BHT. 2. The more the concentration of natural additives was increased, the more the storage stability was improved. The rank order was 60° $gt; 40° $gt; 20° Bx. 3. The AV and POV of powdered anchovy were increased in the rank order to 6 $lt; 12 $lt; 18 $lt; 24 $lt; 30 months by storage term. 4. This clearly suggested that water extract at 20° Bx level from boxthorn was possible utilization as a new natural antioxidant for storage stability of powdered anchovy.
Background: To enhance the taste and physiological characteristics of Lycii fructus (Gugija) extracts, we investigated the changes in the physiological characteristics of Gugija extracts caused by adding white ginseng (WG) and red ginseng (RG)
Methods and Results: Gugija extracts, including 10G10, 10GW-G8 : 2, -G6 : 4, -G4 : 6, -G2 : 8, and -G0 (mixtures made by replacing 0, 20, 40, 60, 80, and 100% of Gugija with WG), as well as 10G10, 10GR-G8 : 2, -G6 : 4, -G4 : 6, -G2 : 8, and -G0 (mixture made by replacing 0, 20, 40, 60, 80, and 100% of Gugija with RG) were extracted with water at 10 times the respective mixture's volume. The antioxidant activities of Gugija extracts were investigated by assessing their 1,1-diphenyl-2-picrydrazyl (DPPH) and 2,2’-azinobis(3ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, ferric reducing antioxidant potential (FRAP) activity, nitrite scavenging activity, and angiotensin converting enzyme (ACE) inhibitory activity. As the amount of WG added increased, the DPPH, and, ABTS radical scavenging activity, and FRAP activity of the Gugija extract decreased. The half maximal inhibitory concentration (IC50) value of 10G10, 10GW-G6 : 4, 10GR-G6 : 4, and 10GR-G0 for DPPH radical scavenging activity were 25.50 ± 1.04, 52.06 ± 1.46, 16.87 ± 1.24, and 9.50 ± 0.16 ㎕/㎖, respectively. On the other hand, the physiological activity of Gugija extract increased with the addition of increasing amounts of RG. However, ACE inhibitory activity was the highest (50.25 ± 2.58%) in the Gugija 10-fold extract without any added RG.
Conclusions: From the above results, we suggest that adding RG to Gugija extracts increase their antioxidant, FRAP, and nitrite scavenging activities.
Background : The fruit of Lycii fructus (Gugija) has beeen used as a tonic medicine and a long-term healthy food without side effect in Asia. An increase in the demand for natural healthy food, Gugija has been thought as a source of healthy foods, and then the extracts of dried Gugija have been frequently used as food ingredients. But, its extracts with Gugija alone has a slightly bad taste. Physiological characteristics of Gugija extracts were investigated by adding white ginseng (WG) and red ginseng (RG) to enhance taste.
Methods and Results : Gugija extract were prepared by adding 0, 20, 40, 60, 80 and 100% of ginseng (WG and RG) to the weight of Gugija, respectively. Antioxidant activities of Gugija extracts were investigated DPPH and ABTS radical scavenging activity, SOD-like activity, FRAP, activity. As the amount of WG added increased, DPPH, ABTS radical scavenging activity, and FRAP activity of Gugija extract decreased. On the other hand, the physiological activity of Gugija extract increased with increasing amount of RG. The SOD-like activity of Gugija extract added up to 60% of WG and to 40% of RG was continuously lowered, in the addition amount thereafter, the activity increased as the addition amount increased. In components, As the amount of WG and RG added increased, the ginsenosides content of Gugija extract increased, the content of ginsenosides was higher in extract added RG than WG, but betaine and total flavonoid content decreased.
Conclusion : From the above results, we may suggest that Gugija extracts by adding RG increased the antioxidant activities, SOD-like activity, and the preference degree.
Background : ROS produced by oxidative stress damaged endothelial cells, and cause a variety of vascular complications. In diabetic hyperglycemia state, ROS increase. The polyol pathway occur in diabetic complications, the excess glucose is absorbed into the polyol pathway when aldose reductase increased, NADPH changes it to sorbitol. Glutathione (GSH) removes ROS. GSH level is reduced by glutathione reductase, using NADPH as an electron donor. Activation of the polyol pathway decrease NADPH, and GSH also reduced. As a result, ROS is increased. In diabetic hyperglycemia state, Glycolysis increases. Effects of increased glycolysis, protein kinase C (PKC) is increased. NAD(P)H oxidase, stimulated by PKC-dependent pathway, increases ROS in the cell. In this study, we measured the ROS scavenging activity of 5 natural products (Lycii fructus, Astragalus membranaceus, Cassia Tora, Polygonatum odoratum, Rubus Coreanus), to confirm the efficacy as diabetic antioxidants. Methods and Results : We extracted 5 natural product by distilled water and ethanol. DPPH radical scavenging activity was significantly higher in Lycii fructus, Rubus coreanus. ABTS radical scavenging activity was better Rubus coreanus, Lycii fructus, Cassia Tora. In addition to, Rubus coreanus, Cassia Tora, Lycii fructus was comparatively higher reducing power activity than other natural products. And total phenolic and flavonoid contents were much higher in Rubus coreanus compared with other extracts. Conclusion : These results suggest that Lycii fructus, Rubus coreanus can be applied as diabetic antioxidant that prevent vascular complications caused by ROS.
This study was conducted to investigate the effects of Lycii fructus and Astragalus membranaceus mixed extracts on immunomodulators and prevention in a streptozotocin-induced diabetes rat model. A total of 28 male rats were divided into four dietary groups and fed a commercial diet (A), commercial diet plus induced diabetes by a streptozotocin (STZ) injection (B), induced diabetes by STZ plus medicinal crop extracts(I&H®) diet (C), and medicinal crop extracts (I&H®) diet (D). Immunoblotting analyses revealed cytokine expression, and ELISA analyses revealed immunoglobulin E and nitric oxide production. As a results, the tumor necrosis factor-α (TNF-α) and inducible nitric oxide synthase (iNOS) as a inflammatory cytokine were decreased. Interleukin-6 (IL-6) and signal transducer and activation of transcription 3 (STAT3) cytokine related in diabetes expression through JAK/STAT3 pathway were also decreased. Furthermore, immunoglobulin E and nitric oxide production were decreased in the serum and lens, respectively. These results suggest that Lycii fructus and Astragalus membranaceus mixed extracts provide positive effects on immunomodulators and prevention in diabetes and eye disease complications.
시험장에서 육성한 품종인 명안, 불로, 청대, 장명, 청운과 육성중인 청양6호, 청양7호 및 청양재래의 가용성 무질소물, 조단백질, 조지질, 회분, 총 폴리페놀, 엑스, 베타인의 함량을 8, 9, 10 및 11월의 수확시기별로 조사한 결과, 가용성 무질소물 및 엑스는 육성중인 청양6호가 가장 높은 함량을 보였고, 조단백질, 총폴리페놀 및 베타인의 함량에서는 육성중인 청양 7호가 가장 높은 함량을 보였으며, 조지질은 청대에서, 조회분은 청운에서 가장 높았다. 건과비율은 육성중인 청양6호에서 가장 높았고, 건과비율이 높을수록 가용성 무질소물의 함량은 증가 (r = 0.70)하는 반면 조단백질 (r = -0.83) 및 조회분 (r = -0.71)등은 감소하였다. 일반성분 중에는 서로 일정한 상관을 보였는데, 가용성 무질소물의 함량이 증가하면 엑스의 함량 (r = 0.81)은 높아지는 반면, 단백질 (r = -0.88), 지질 (r = -0.82) 및 회분 함량 (r = -0.81)은 감소하고, 총폴리페놀 함량이 증가하면 베타인 함량이 증가 (r = 0.90)하는 경향이었다. 품종특성과 성분을 분석한 결과 육성중인 청양 6호는 가용성 무질소물 및 엑스분이 높게 관찰되었을 뿐만 아니라, 건과비율이 높아 타 품종에 비해 쉽게 건조될 수 있어 건조비용을 줄일 수 있는 것으로 예상되었고, 청양 7호는 조단백질, 총폴리페놀 및 베타인 함량이 높아 기능성 품종으로서의 가치를 보여주었다.
구기자(枸杞子)의 품질향상(品質向上)과 품질평가(品質評價)의 척도(尺度)등을 제시(提示)하는 효률적(效率的)인 건조조건(乾燥條件)을 구명(究明)하기 위하여 건조온도(乾燥溫度)와 시간(時間)을 달리하여 구기자(枸杞子)를 건조(乾燥)한 다음, 건조조건별(乾燥條件別)로 구기자(枸杞子)의 수분함양(水分含量) 변화(變化), 색도(色度) 및 점도를 조사(調査)하였으며 각처리별(各處理別) 성분(成分)을 분석(分析)하여 다음과 같은 결과(結果)를 얻었다. 1. 수분함량(水分含量)의 변화(變化)에서 고형물(固形物)은 초기온도(初期溫度) 50℃에서 4시간(時間) 건조후(乾燥後) 60℃에서 28 시간(時間) 건조(乾燥)한 경우가 함양(含量)이 가장 많았다. 2. 색도(色度)의 변화(變化)에서는 1번 처리구(處理區)에서 적색도(赤色度)와 황색도(黃色度)가 각각(各各) 5.0, 51.3으로 외관상(外觀上) 좋았으며 명도(明度)에서도 66으로 고유(固有)의 상품가치(商品價値)를 지니고 있었다. 3. 점도는 1번 처리구가 3.5로 나타나 다른 처리구에 비하여 높은 점도를 보였으나 수율은 4번 처리구가 높았고 나머지는 차이(差異)가 없었다. 4. 건조온도(乾燥溫度)와 시각(時閣)에 따라 성분조성(成分組成)에 차이(差異)가 있었는데 무기성분(無機成分)인 Zn, CaO는 4번 처리구가 K2O, P2O5에서는 3번 처리구가 많았으나 조단백질, 총당, 환원당둥有 기성분(機成分)은 1번 처리구가 가장 높아 주요성분(主要成分)이 많음을 알수 있었다. .5. 열풍(熱風) 건조방법(乾燥方法)은 온도조절(溫度調節)이나 건조시간(乾燥時間)이 상품(商品)의 질(質)에 큰 영향(影響)을 끼치므로 구기자(拘杞子)는 2단계(段階)로 조건(條件)을 달리해 건조(乾燥)하는것이 바람직한데 시험결과(試驗結果) 초기온도(初期溫度) 50℃에서 2시간(時間), 후기온도(後期溫度) 60℃에서 24시간(時間) 건조(乾燥)하는것이 구기자(拘杞子) 품질(品質) 및 경제적(經濟的) 측면(側面)에서 최적(最滴)의 조건(條件)이었다.
