        총채벌레는 우리나라에 60여 종이 알려져 있으며 다양한 시설작물에서 직접적인 섭식 피해 이외에 토마토반점위조바이러스(TSWV)를 매 개하는 간접적인 피해도 유발한다. 그동안 총채벌레 방제는 살충제에 의존해 왔는데, 이는 농업환경에 많은 부작용을 유발하고 해충의 저항성을 유발시켜 더욱 방제를 어렵게 하고 있다. 이러한 문제를 해결할 수 있는 대안으로 내성 회피를 위한 물질을 탐색하였다. 실내검정으로 약용작물 67종의 추출물을 꽃노랑총채벌레 성충에 처리하여 가장 효과가 우수한 목단피를 선발하였다. 목단피 추출물을 처리 후 1일차에 100%의 살충효 과를 보였다. 또한, 목화진딧물은 3일차 83%, 복숭아혹진딧물 3일차 97%, 점박이응애 1일차 100%의 살충효과를 보였다. 고추 포트 검정에서 꽃노랑총채벌레 방제가는 1일차 77.6%, 2일차 40%의 효과가 나타났다. 현재 추가적으로 효과를 증대시킬 수 있는 물질을 탐색하고 있으며, 총 채벌레 방제에 본 추출물을 활용한다면 효과적일 것으로 기대된다.

        Ischemic stroke causes severe neuronal damage. Chlorogenic acid is a phenolic substance present in fruits and coffee. It also exerts neuroprotective effects against various brain injuries. The 14-3-3 family protein perform a variety of functions including metabolism, signal transduction, cell differentiation, and apoptosis. The purpose of this study is to investigate whether chlorogenic acid regulates the expression of 14-3-3 protein in stroke animal models. Ischemic stroke was induced by middle cerebral artery occlusion (MCAO) surgery. Phosphate buffered saline (PBS) or chlorogenic acid (30 mg/kg) were intraperitoneally injected to adult male rats 2 h before MCAO surgery. Adhesive-removal test was performed 24 h after MCAO surgery and cerebral cortical tissues were collected for further study. MCAO damage caused severe neurological impairment and chlorogenic acid treatment ameliorated this disorder. Our proteomic approach showed a decrease in 14-3-3 expression in MCAO animals with PBS. The decrease in 14-3-3 expression alleviated in MCAO animal with chlorogenic acid. We confirmed changes in various 14-3-3 protein isoforms, including beta/alpha, zeta/delta, gamma, epsilon, eta, and tau through reverse transcription-PCR. These results explained that chlorogenic acid regulates the expression of 14-3-3 protein in MCAO-induced cerebral ischemia. 14-3-3 is considered to be an important protein for cell survival through binding to pro-apoptotic proteins. The maintenance of 14-3-3 levels is an important event in neuroprotection against ischemic injury. Therefore, we can demonstrate that the 14-3-3 protein contributes to the neuroprotective effect of chlorogenic acid in stroke animal models.

        This study analyzed the chemical characteristics and physiological activity of five kinds of fresh vegetables produced in trees in early spring and tried to use them as basic data for wild vegetable producers and processed food manufacturers using wild vegetables. The crude protein, minerals, ascorbic acid, folate, total phenol, total flavonoid, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and ACE (angiotensin converting enzyme) inhibitory activity were determined. Five spring wild vegetables contain high protein and phosphorus, indicating that they are useful food ingredients as sources of protein and phosphorus. Vitamin C content was high in R. venicifera and C. sinensis shoots, and in particular, R. venicifera and A. cortex shoots have high folic acid (folate) contents of 1,903.91 ug% and 1,525.35 ug%, respectively, which is considered a good food for folic acid intake in spring. The total phenol content was between 0.52% and 1.27%, and it was the highest in C. sinensis of 1.27%, followed by the order of R. venicifera, A.cortex, K.pictus, and A. elata, which tended to be consistent with the total flavonoid content. As for DPPH radical scavenging ability, C. sinensis (55.93%) showed the highest activity, and ACE inhibitory activity showed the highest activity in A. cortex (88.04%).

        목적 : 멀티포컬 콘택트렌즈(MFCL) 착용 시 시각 피질에서 발생하는 ERP 변화를 확인하기 위하여 단초점 콘택트렌즈 착용 시 나타나는 ERP 변화와 비교하였다. 방법 : 안질환이 없는 근시안 20명 (여성 15명, 남성 5명, 평균 연령 24.00±0.51세)을 대상으로 하였으며, 단안 시력 0.00 logMAR 이하, -0.75 D 이상의 난시안, 0.50 D 이상의 부등시안, 조절력이 비정상적이거나 시력교정술을 받은 경우는 제외하였다. 단초점 콘택트렌즈(SVCL)와 낮은 가입도 중심부-근용 멀티포컬 콘택트렌즈를착용시킨 후 NeuroScan SynAmps 2 프로그램을 이용하여 시피질에서 일어나는 시각반응을 ERP로 기록하였다. P100 분석은 O1, O2 채널을 사용하고, N170 분석은 P7, PO7, P8 및 PO8 채널을 사용하였다. MFCL 착용 후 나타난 P100과 N170의 활성도(amplitude)와 반응시간(latency)은 SVCL 착용 후 나타난 값과 비교하였다. 결과 : MFCL 착용 시 단안과 양안에서의 P100 활성도는 SVCL 착용 시와 차이가 없었으나 P100 반응시간은 우세안에서 SVCL 착용 상태보다 길었다(p=0.036). MFCL 착용 시 우세안의 N170 활성도는 비우세안보다 작았고(p=0.001), 비우세안은 단초점 콘택트렌즈 착용 시보다 작았다(p=0.008). 그러나 양안에서 N170 활성도와 반응시간은 SVCL 착용 상태와 차이를 보이지 않았다(p=0.200), (p=0.249). 결론 : MFCL 착용 후 초기시각반응과 인식과정은 우세안에서는 저하되었지만, 양안 상태에서는 SVCL 착용시와 차이가 없었다. 따라서 MFCL 착용 후 나타나는 망막에 맺힌 흐린 상은 양안 융합과정에서 극복되어 시기능에 영향을 주지 않는 것으로 사료된다.

        It has long been speculated that the visual system should use a coding strategy that takes advantage of statistical redundancies in images. But how such a coding strategy should manifest in neural responses has been less clear. Low-level image structure related to the power spectrum of natural images appears to be captured by a hard-wired efficient code in the retina of the fly and precortical structures like the LGN of cats that maximizes information content through the limited capacity channel of the optic nerve. But visual images are typically filled with higher-order structure beyond that captured by the power spectrum and visual cortex is not constrained by the same capacity limits as the optic nerve. Whether and how visual cortex can flexibly code for higher order redundancies is unknown. Here we show using psychophysical techniques that the neural response in early human visual cortex may be modulated by orientation redundancies in images such that a visual feature that is contained within a predictive pattern results in slower reaction times than a feature that deviates from a pattern, suggesting lower neural responses to predictable stimuli in the visual cortex. Our results point to a neural response in early visual cortex that is sensitive to global patterns and redundancies in visual images and is in marked contrast to standard models of cortical visual processing.

        To evaluate alterations in fecal bacterial compositions in weaning piglets after supplementation with Phellodendron Cortex extract (PCE), piglets were allowed freely to consume feed containing 10 ppm of PCE for 7 weeks. During the feeding period, fecal samples were collected from individual piglets. A total of 578 valid sequence reads were generated from 40 fecal samples of normal piglets before supplementation with PCE. Fifteen classes were identified. Approximately 90% of classifiable sequences belonged to Clostridia and Bacilli classes. The abundance of Clostridium spp. and Lactobacillus spp. was determined based on the results of the sequencing. The supplementation with 10 ppm PCE for 3 weeks increased the numbers of Clostridium spp. and Lactobacillus spp. whereas seven weeks of supplementation with PCE reduced their abundance. In vitro, PCE inhibited the growth of E. coli and Salmonella enteritidis in a dose-dependent manner.

        본 연구는 CIA로 유도된 류마티스 관절염 모델에서 지골피 추출물이 류마티스관절염 관련 인자에 미치는 영향을 평가하였다. 동물 모델에 류마티스관절염을 유발한 후 지골피 추출물을 4주간 200 ㎎/㎏/day 용량으로 경구 투여하였다. 이후 혈액, 방사선 및 조직 분석에 기초하여 효과를 평가하였다. 그 결과, 대조군에 비해 지골피 추출물은 사이토카인, 케모카인 및 면역글로불린 생성을 유의하게 감소시켰다. 또한, 지골피 추출물은 중족골을 효과적으로 보존하였으며, H&E 및 M&T 염색에서도 대조군에 비해 조직 병리학적 개선이 있음을 보여주었다. 이와 같은 결과는 지골피 추출물이 류마티스관절염 증상을 완화시켰음을 나타내고 있다. 따라서 지골피 추출물은 류마티스 관절염 관리를 위한 새로운 치료옵션이 될 수 있다.

        This study aimed to identify specific psychological and brain activation responses relating to the processing of negative emotions in patients with alcohol dependency. The authors hypothesized that patients with alcohol dependency would demonstrate the abnormal functioning of brain regions involved in negative emotions. Eleven male patients diagnosed with alcohol dependence in an inpatient alcohol treatment facility and 13 social drinkers with similar demographics were scanned using functional magnetic resonance imaging (fMRI) as they viewed film clips that evoked negative emotions. During exposure to negative emotional stimuli, the control group evinced significantly greater activity in the right anterior cingulate cortex (ACC) in comparison to patients with alcohol dependency. Correlation analyses demonstrated a negative association in the relationship between beta values from the right ACC and amygdala in participants classified in the control group. No statistically significant relationship was observed for blood oxygenation level-dependent (BOLD) changes between the two regions in the patient group during the elicitation of negative emotions. On the other hand, patients exhibited a greater activation of the amygdala as negative emotions were induced. These results suggest that alcoholism presents pathophysiology of brain activation that is distinct from the responses of healthy individuals functioning as controls.

        To elucidate the effect of cellular phone electromagnetic wave (EMW) exposure on the developing cerebellar cortex of neonatal Sprague-Dawley rats, animals were exposed to cellular phone electromagnetic waves for 1 hr per day for 3 weeks. At the end of the experimental period, animals were sacrificed by cardiac perfusion, after which histological samples were prepared and observed microscopically. In the EMW exposure group, external granule cells were remained partially in the external granular layer without migrating into the internal granular layer. In addition, dark stained shrunken Purkinje cells with pyknotic nuclei increased and the outline of cells became irregular and showed degenerative signs, such as mitochondrial swelling and disrupted cristae. Moreover, the cisternae of rough endoplasmic reticula and Golgi complex were severely swollen. Bergmann glial cells adjacent to the dark stained Purkinje cells were swollen and cytoplasmic organelles were scant. Dark stained shrunken granule cells were also observed and the outline of cells was irregular. The results of the present study suggest that cellular phone EMW exposure to neonatal Sprague-Dawley rats leads to a partial delay of early migration of cerebellar cortical cells and degenerative changes in Purkinje cells, Bergmann glial cells and granule cells.

        Transcranial direct current stimulation (tDCS) is a neuromodulatory technique that delivers low-intensity direct current to cortical areas, thereby facilitating or inhibiting spontaneous neuronal activity. This study was designed to investigate changes in various sensory functions after tDCS. We conducted a single-center, single-blinded, randomized trial to determine the effect of a single session of tDCS with the current perception threshold (CPT) in 50 healthy volunteers. Nerve conduction studies were performed in relation to the median sensory and motor nerves on the dominant hand to discriminate peripheral nerve lesions. The subjects received anodal tDCS with 1 mA for 15 minutes under two different conditions, with 25 subjects in each groups: the conditions were as follows tDCS on the primary motor cortex (M1) and sham tDCS on M1. We recorded the parameters of the CPT a with Neurometer(R) at frequencies of 2000, 250, and 5 Hz in the dominant index finger to assess the tactile sense, fast pain and slow pain, respectively. In the test to measure CPT values of the M1 in the tDCS group, the values of the distal part of the distal interphalangeal joint of the second finger statistically increased in all of 2000 Hz (p=.000), 250 Hz (p=.002), and 5 Hz (p=.008). However, the values of the sham tDCS group decreased in all of 2000 Hz (p=.285), 250 Hz (p=.552), and 5 Hz (p=.062), and were not statistically significant. These results show that M1 anodal tDCS can modulate sensory perception and pain thresholds in healthy adult volunteers. The study suggests that tDCS may be a useful strategy for treating central neurogenic pain in rehabilitation medicine.

        황백(Cortex Phellodendri: CP)은 황벽나무(Phellodendron amurense)의 건조된 수피로부터 얻어진다. 이 수피는 한국의 전통 한약제로서 설사, 황달, 무릎과 발의 통증, 요도관 및 피부 감염증에 폭넓게 사용되어 왔다. 이들 기능성성분의 분리 및 정제는 박층 크로마토크래피, 컬럼 액체 크로마토크래피 및 HPLC와 같은 여러 분석법들이 동양의 약초연구에 이용되어 왔다. 본 연구는 CP로부터 berberine을 분리하기 위해 향류분배 크로마토크래피법(CPC)으로 효과적으로 수행하였다. 두 용매의 CPC 최적조성은 n-butanol: acetic acid: water(4:1:5 v/v/v)이었다. 이동상의 유속은 1,000 rpm 회전력에서 상승법으로 분당 3 mL 속도로 전개시켰다. CPC에서 분리된 분획분은 prep-HPLC로 정제하였다. 1H-NMR 스펙트럼은 4.10과 4.20 ppm에서 3H(-OCH3), 6.10 ppm에서 2H의 (-OCH2O-) proton signal의 공명이 관찰되었다. 2개의 방향족 proton은 이중결합 패턴을 보였다. H-11과 H-12 doublet은 각각 7.98과 8.11에서 나타났다. 13C-NMR 스펙트럼에서는 C2와 C3의 methylenedioxy group(-OCH2O-), C9과 C10에 methoxy group(-OCH3)이 4개의 치환된 형태로 보였다. 분리․정제된 berberine의 화학구조는 1H, 13C-NMR, ESI-MS 데이터 분석으로 확인하였다.

        The purpose of this study was to investigate the antioxidant effects of Mori cortex radicis powder and to determine the optimal composite recipe by testing different amount of Mori cortex radicis powder and sugar in cookies prepared with Mori cortex radicis powder. In regard to its antioxident effects, Mori cortex radicis powder had a total phenolic content and DPPH free radical scavenging activity of 149.56 mg GAE/g and 137.77 μg/mL, respectively. The response surface methodology was used to obtain ten experimental points (including two replicates for Mori cortex radicis powder and sugar) and Mori cortex radicis cookie formulation was optimized using rheology. The results of the sensory evaluation produced significant values for color (p<0.05), texture (p<0.05), sweetness (p<0.01) and overall quality (p<0.05), and the results of instrumental analysis showed significant values in sweetness (p<0.001), redness (p<0.01) and spread ratio (p<0.5). As a result, the optimum formulations obtained by numerical and graphical methods were found to be 16.84 g of Mori cortex radicis powder and 64.42 g of sugar.

        The purpose of this study was to investigate the antioxidative effects of Mori Cortex Radicis powder and to determine the optimal mixing ratio of Mori Cortex Radicis powder and water in the preparation of bread. The optimal sensory composite recipe was determined by producing bread with different levels of Mori Cortex Radicis powder and water. The analysis was performed using response surface methodology and a sensory evaluation was performed with the data. Ten experimental recipes, including two with reference points in the composition, were selected. In terms of the antioxidative effects of Mori Cortex Radicis powder, the IC50 for total phenolic content and DPPH free radical scavenging activity were 149.56 GAE/g dry powder and 137.77 /mL respectively. Measurement results of the mechanical properties showed differences in volume (p〈0.05), baking loss (p〈0.05), yellowness (p〈0.01), lightness (p〈0.01), redness (p〈0.01), hardness (p〈0.01) and springiness (p〈0.05). The sensory measurements showed significant values for color (p〈0.05), appearance (p〈0.05), flavor (p〈0.01), taste (p〈0.01), and overall quality (p〈0.01). Overall, based on numerical and graphical methods, the optimal formulation was determined to be 21.16 g of Mori Cortex Radicis powder and 372.47 g of water.

        This study was performed to investigate the antidiabetic study effect in ethanol extract of Ailanthicortex(A.C) radici in Streptozotocin(STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45㎎/㎏ dissolved in citrate buffer. The ethanol extract of A.C radici was orally administrated once a day for 7 days at a dose of 500㎎/㎏. The content of serum glucose, triglyceride(T.G), total cholesterol were significantly decreased in treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased in treated group compared to the those of STZ-control group. but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was significantly increased. These results indicated that ethanol extract of A.C would have antidiabetic effect in STZ-induced diabetic rats.

        This study was conducted to examine the antioxidative effect and nitrite scavenging ability of extract from Acanthopanax cortex shoot. The total phenolic compound and flavonoids contents of extract from Acanthopanax corex shoot were 116.33±6.09 ㎎ GAE/g and 65.07±4.10 ㎎ RE/g, respectively. Antioxidative activities were measured by various in vitro models such as DPPH radical scavenging activity, FRAP, reducing power, ABTS radical scavenging activity, ORAC assay. This results showed that the extract of Acanthopanax cortex shoot was effective in scavenging radicals and protecting oxidation when assessed various in vitro systems. Similarly, the nitrite scavenging ability of extract was increased in a dose dependent manner. Moreover, ORAC value at a concentration of 0.1 ㎎/㎖ was 103.4±5.6μM TE/g. Considering high consumer demand beneficial health effects, Acanthopanax cortex shoot can be utilized to develop functional food health-promoting and natural antioxidant agents.

        This study was conducted in order to evaluate the alleviating effects of Phellodendrin cortex water extract (PCWE) on skin aging in hairless mice via observation of morphogical and histological changes. Skin aging was induced by UVB irradiation and application of squalene monohydroperoxide (Sq-OOH) to the back skin of hairless mice for six weeks. And, at the same time, saline (C), jojoba oil (VC), PCWE (E), and 0.01% retinoic acid diluted with polyethylene glycol (PC) were applied topically twice per day, six days per week, for a period of six weeks. Improved wrinkle formation in a pattern of shallow furrows and thin and narrow crests was observed in the retinoic acid and PCWE application groups, compared to the C group. On the morphologic analysis for skin wrinkles, the E group showed lower levels in skin roughness, maximum roughness, average roughness, smoothness depth, and arithmetic average roughness by 13.1, 17.2, 18.4, 15.4, and 16.1%, respectively, compared with the C group, indicating that PCWE inhibited potential formation of wrinkles in the skin. In the C group, structures of lipid lamellae and collagen fibers were broken or deformed with an irregular arrangement. Application of retinoic acid and PCWE protected against the deformity of lipid lamellae and collagen fibers. Elastic fibers in dermis of the C group also showed severe transformation; however, applications of retinoic acid and PCWE resulted in a significant decrease in the number of denatured elastic fibers. Therefore, PCWE could have an alleviating effect on skin aging induced by UVB irradiation and application of Sq-OOH.

        This study was conducted for evaluation of the alleviating effects of Phellodendrin cortex water extract (PCWE) on skin inflammation in hairless mice. Skin inflammation was induced by UVB irradiation and application of squalene monohydroperoxide (Sq-OOH) to the back skin of hairless mice for six weeks. At the same time, saline (C), jojoba oil (VC), PCWE (E), and 0.01% retinoic acid diluted with polyethylene glycol (PC) were applied topically twice per day, six days per week for a period of six weeks. The skin erythema index of the E group was lower than that of the C group. Epidermis and dermis of the C group were remarkably thickened, compared to the PC or E group. In the C group, infiltration of many inflammatory cells, including neutrophils and lymphocytes, was observed in dermis, and a large number of mast cells were observed in dermis and hypodermis; the degree of degranulation was remarkable. However, these phenomena were alleviated in the PC and E2 groups. The E group showed a lower activity in skin xanthine oxidase but a higher activity in skin superoxide dismutase, compared to the C group (P<0.05). The VC, PC, and E groups also showed a high activity of skin catalase by 25.3%, 58%, and 42%, respectively, compared to the C group. Taken together, these results indicate that PCWE could have an alleviating efficacy on skin inflammation induced by UVB irradiation and application of Sq-OOH in hairless mice.

        The majority of strokes are caused by ischemia and result in brain tissue damage, leading to problems of the central nervous system including hemiparesis, dysfunction of language and consciousness, and dysfunction of perception. The purpose of this study was to investigate the effects of Poly(ADP-ribose) polymerase(PARP) on necrosis in neuronal cells that have undergone needle electrode electrical stimulation(NEES) prior to induction of ischemia. Ischemia was induced in male SD rats(body weight 300g) by occlusion of the common carotid artery for 5 min, after which the blood was reperfused. After induction of brain ischemia, NEES was applied to Zusanli(ST 36), at 12, 24 and 48 hours. Protein expression was investigated using immuno-reactive cells, which react to PARP antibodies in cerebral nerve cells, and Western blotting. The results were as follows: In the cerebral cortex, the number of PARP reactive cells after 24 hours significantly decreased(p<.05) in the NEES group compared to the GI group. PARP expression after 24 hours significantly decreased(p<.05) in the NEES group compared to the GI group. As a result, NEES showed the greatest effect on necrosis- related PARP immuno-reactive cells 24 hours after ischemia, indicating necrosis inhibition, blocking of neural cell death, and protection of neural cells. Based on the results of this study, NEES can be an effective method of treating dysfunction and improving function of neuronal cells in brain damage caused by ischemia.
