        The effect of pt-I values of 6.5, 6.0, 5.0, and 4.0 on spore germination of Bacillus cereus (B. cereus) was investigated at water activity (a%) values of 0.98, 0.96, 0.94, and 0.90 in a controlled medium at 30°C for 7 days. The best condition for germination of spores of B. cereus was observed at pH 6.5 with aw 0.98. No matter what combination of a and pH is used, a complete inhibition of spore germination was achieved at either a,.. value of 0.90 or pH value of 4.0. Spore germination was also delayed or inhibited at aw 0.98 with pH 4.0, aw 0.96 with pH 5.0, and aw 0.94 with pH 6.5, 6.0, 5.0, or 4.0. The results indicate that the combined inhibitory effects of pH and water activity on the germination of B. cereus spores in controlled medium could be applied to food preservation.

        친환경 유기농자재를 이용하여 잿빛곰팡이병균의 포자 발아, 균사생장 억제효과를 기내에서 조사하였다. 그 결과 잿빛곰팡이병의 균사생장 억제 효과는 Bacillus subtilis를 주성분으로 하는 제제가 100%의 억제효과를 보였으며, 그 외의 제제는 20% 이하의 범위에서 억제효과를 나타내었다. 포자발아 억제효과는 유기황 수화제 2종('BTB', '황스타')이 각각 97.7%, 92.3%으로 나타났다. 균사생장 억제와 포자 발아억제에 모두 효과를 보이는 제제는 없었다. 따라서 잿빛곰팡이병 방제를 위해서는 포자발아 억제를 위한 방제와 균사생장억제를 위한 제제를 각각 처리하여야 할 것으로 사료되었다. 아울러 사물 기생성을 가진 잿빛곰팡이병의 방제를 위해서는 잔재물에서 생장을 막는 것이 중요하며 이를 위해 예방을 전제로 한 방제의 경우 유기황을 주성분으로 하는 제제가 적용 가능할 것으로 생각된다.

        친환경 유기농자재를 이용하여 고추 탄저병균의 포자 발아, 균사생장 억제효과를 기내에서 조사하였다. 그 결과 균사생장 억제효과 기내검정. 탄저병의 균사생장 억제 효과는 Bacillus subtilis를 주성분으로 하는 제제가 100%의 억제효과를 보였으며, 그 외의 제제는 20~40%의 범위에서 억제효과를 나타내었다. 포자발아억제효과는 유황 수화제 2종('BTB', '황스타')이 각각 100%, 95.1%, '고려역'(95.0%), '보르스타'(99.0%), '지하부대 KM'(96.1%) 순으로 나타났다. 균사생장 억제와 포자 발아억제, 부착기 형성억제 결과를 고려하여 고추 탄저병 방제에 적용 가능한 친환경 방제제는 Bacillus subtilis, Panibacillus polymyxa 혼용제('고려역')가 가장 효과적인 것으로 판단되었으며, 예방을 전제로 한 방제의 경우 유황 수화제, 보르도액을 주성분으로 하는 제제가 적용 가능할 것으로 생각된다. 본 연구에서 선택된 제제는 고추 포장에서 탄저병 방제효과가 최종적으로 인정되면 차후 고추 탄저병 방제에 이용될 것으로 사료된다.

        The reactive oxygen species (ROS) generated during the somatic cell transfer nuclear (SCNT) procedures may cause the mitochondrial dysfunction and DNA damage, which may result in restricts the reprogramming of SCNT embryos and play a key direct role in apoptosis. The present study was conducted to investigate the effect of antioxidant treatment during the SCNT procedures on the inhibition of mitochondria and DNA damages in bovine SCNT embryos. The reconstituted oocytes were treated with antioxidants of 25 μM β-mercaptoethanol (β-ME) or 50 μM vitamin C (Vit. C) during the SCNT procedures. In vitro fertilization (IVF) was performed for controls. Mitochondrial morphology and membrane potential (ΔΨ) were evaluated by staining the embryos with MitoTracker Red or JC-1. Apoptosis was analyzed by Caspase-3 activity assay and TUNEL assay, and DNA fragmentation was measured by comet assay at the zygote stage. Mitochondrial morphology of non-treated SCNT embryos was diffused within cytoplasm without forming clumps, while the IVF embryos and antioxidant treated SCNT embryos were formed clumps. The ΔΨ of β-ME (1.3±0.1, red/green) and Vit. C-treated (1.4±0.2, red/green) SCNT embryos were significantly higher (p<0.05) than that of non-treated SCNT embryos (0.9±0.1, red/ green), which similar to that of IVF embryos (1.3±0.1, red/green). Caspase-3 activity was not difference among the groups. TUNEL assay also revealed that little apoptosis was occurred in SCNT embryos as well as IVF embryos regardless of antioxidant treatment. Comet tail lengths of β-ME and Vit. C-treated SCNT embryos (337.8±23.5 μm and 318.7 ±27.0 μm, respectively) were shorter than that of non-treated SCNT embryos (397.4± 21.4 μm) and similar to IVF embryos (323.3±10.6 μm). These results suggest that antioxidant treatment during SCNT procedures can inhibit the mitochondrial and DNA damages of bovine SCNT embryos.

        Cordycepin, a specific polyadenylation inhibitor, is the main functional component in Cordyceps militaris that is one of the top three renowned traditional Chinese medicines. In this study, we performed in vitro experiments to investigate the anti-invasive and anti-metastatic activities of cordycepin using human prostate carcinoma LNCaP cells. Cordycepin were administered and their effects on LPS-induced cell migration and invasion by wound healing migration assay, measurement of TER and In vitro invasiveness assay. Within the concentrations which were not cytotoxic effects, cordycepin caused a concentration-dependent suppression of LPS-induced cell migration and invasion. The anti-invasive activity of cordycepin was also found to be associated with increased tightness of the TJ, which was confirmed by an increase in TER. The activity of MMP-2 in LNCaP cells was dose-dependently inhibited by treatment with cordycepin, and this was also correlated with a decrease in expression of its mRNA and proteins, and up-regulation of TIMPs expression. Additionally, cordycepin repressed the LPS-induced NF-kB activation and phosphorylation of PI3K/AKT. Taken together, these findings suggest that cordycepin inhibited LPS-induced migration and invasion of LNCaP cells by down-regulating the expression and activity of MMP-2, and the possible targets may be NF-kB and PI3K/AKT.

        We evaluated insecticidal and acetylcholine esterase inhibition activity of 11 Apiaceae plant essential oils and their constituents against male and female adults of Blattella germanica. Of 11 Apiaceae plant essential oils tested, dill (Anethum graveolens), carvi (Carum carvi), and cumin (Cuminum cyminum) essential oils demonstrated > 90 % fumigant toxicity against male adults of German cockroach at 5 mg/filter paper. In contact toxicity test, dill (Anethum graveolens), carvi (Carum carvi), cumin (Cuminum cyminum), and ajowan (Trachyspermum ammi) essential oils produced strong insecticidal activity against male and female adults of German cockroach. Among test compounds, carvone, 1,8-cineole, trans-dihydrocarvone, cuminaldehyde, trans-anethole, p-cymene, and γ-terpinene demonstrated strong fumigant toxicity against male and female adults of B. germanica. In a contact toxicity test, carveol, cuminaldehyde, (S)-(+)-carvone, trans-anethole, thymol, and p-cymene showed the strong contact toxicity against B. germanica male and female adults. IC50 values of α-pinene, carvacrol, and dihydrocarvone against female acetylcholine esterase were 0.28, 0.17, and 0.78 mg/mL, respectively. The toxicity of blends of constituents identified in 4 active oils indicated that carvone, cuminaldehyde, and thymol were major contributors to the fumigant or contact toxicity of the artificial blend.

        The pinewood nematode, Bursaphelenchus xylophilus is a serious forest pathogen in many countries including Japan, China and Korea. To minimize the environmental problems caused by synthetic chemicals broadly utilized in the control of B. xylophilus, we estimated the nematicidal potency of 97 phytochemicals (49 monoterpenes, 17 phenylpropenes, 16 sesquterpenes and 15 sulfides) against B. xylophilus by measuring their inhibition activity against acetylcholinesterases (ACE, EC 3.1.1.7) of B. xylophilus (BxACEs). In the primary inhibition assay using B. xylophilus crude protein, (+)-α-pinene, (-)-α-pinene and 3-carene in the monoterpenes, ο-anisaldehyde and coniferyl alcohol in the phenylpropenes and cis-nerolidol in the sesquterpenes were observed the > 50% BxACE inhibition rate and they were subsequently estimated the inhibition rate against three recombinant BxACEs. The (+)-α-pinene and coniferyl alcohol showed the high inhibition rate against BxACE-1 and BxACE-2. Interestingly, coniferyl alcohol also exhibited the high inhibition rate against BxACE-3 which would be insensitive to ACE inhibitors.

        시판되는 남해산 흑마늘 4제품(A, B, C 및 D)을 이용하여 항산화 활성, 아질산염 소거 및 nitrosodimethylamine(NDMA) 생성억제 활성을 비교하였다. 마늘 중 total pyruvate와 thiosulfate 함량은 생마늘보다 흑마늘에서 높았으며, B제품에서 가장 높은 함량이었다. 흑마늘의 물 및 에탄올 추출물에서 총 페놀과 플라보노이드 함량은 물 추출물에서 높았으며, 흑마늘은 생마늘 추출물보다 높은 함량으로 A제품에서 다소 높게 정량되었다. 항산화 활성으로 DPPH, ABTs, hydroxyl 및 nitric oxide 라디칼 소거활성을 비교한 결과, 흑마늘이 생마늘보다 높았으며, 물 추출물이 에탄올 추출물보다 높은 소거활성을 보였고, A와 B제품간의 항산화 활성은 비슷한 경향이었다. 아질산염 소거활성과 NDMA 생성억제 활성은 항산화 활성과 동일한 경향이었으며, A제품의 활성이 유의적으로 높았다. 따라서 흑마늘의 NDMA 생성억제 활성이 항산화 활성과 유사한 패턴을 보이고 있는 바, 흑마늘은 생체 내 암 발생의 억제에도 영향을 줄 것으로 예상되었다.

        The objective of this study was to determine the effect of macrophages on growth of human colon cancer cells. The results showed that co-culture of colon cancer cells with macrophages inhibited the growth of colon cancer cells (HCT116 and SW620) depending on the number of macrophages, RAW 264.7 cells, and activated THP-1 cells accompanied by down regulation of pSTAT3 in cancer cells. We also found that expression and release of cancer cell growth inhibitory cytokines, IL-1 receptor antagonist (IL-1ra) and IL-10, was increased in macrophages. Blocking of the STAT3 pathway with specific inhibitor and siRNA of STAT3 abolished the growth of colon cancer cells and expression of IL-1ra and IL-10. In addition, neutralization of IL-1ra and IL-10 with antibodies resulted in reversal of macrophage-induced inhibition of cancer cell growth. These data showed that IL-1ra and IL-10 released from macrophages inhibit growth of colon cancer cells through inhibition of the STAT3 pathway.

        약용식물 중 산형과 식물 18종 32점에 대한 항산화 및tyrosinase 저해 활성을 검정한 결과는 다음과 같다. 산형과 식물 15종의 32점은 항산화 활성, tyrosinase 저해 활성을 나타내기 위한 실험재료로 사용되어졌다. Bupleurum falcatum L.(Shoot), Torilis japonica D. C. (Whole)에서 폴리페놀과 플라보노이드 함량이 높게 나타났다. Foeniculum vulgare Gaeriner (Flower) 는 항산화 활성과 tyrosinase 저해 활성이 높게 나타났다. 항산화 활성은 총 폴리페놀 함량과 고도의 유의한 부의상관관계를 보였고, 총 플라보노이드 함량 간에는 유의한 부의 상관관계를 보였으며 tyrosinase 저해 활성은 총 폴리페놀과 총 플라보노이드 함량은 상관관계를 보이지 않았다. Tyrosinase 저해활성은 항산화 활성 함량과 상관관계를 보이지 않았다. 총 폴리페놀은 총 플라보노이드 함량 간에는 고도의 유의한 정의 상관관계를 나타냈다. 이 중 Foeniculum vulgare Gaeriner (Flower)는 항산화 활성과 tyrosinase 저해 활성이 가장 높게 나타났다. 이들 식물들에 대한 추출용매 및 추출조건에 따른 추출효율을 검토하고, 추가적인 생리활성 능력을 검정하는 등 보다 정밀한 연구가 필요할 것으로 사료된다.

        The purpose of this study is to minimize the PCH(Propylene CHlorohydrins) as a by-product in HPMC(HydroxyPropyl MethylCellulose) manufacturing process. HPMC is made of cellulose which is natural high polymer. And HPMC is applicable to several industrial areas. Especially it can be used in food industry as an effective additive. PCH is the by-product which is generated in chemical reaction in HPMC manufacturing process. So it is essential to eliminate PCH for the improvement of product quality. Therefore we have studied to minimize the amount of PCH. It is expected that the application of HPMC could be enlarged as the result of this study.

        Yoghurt는 쉽게 접할 수 있는 기호성 식품으로 생체 조절 기능을 갖는 기능성과 다양한 맛을 부여할 수 있는 퓨전성을 동시에 강화시킬 수 있기 때문에 여러 가지 기능성 소재들을 첨가하여 새로운 yoghurt를 개발하기 위한 연구가 활발하게 수행되고 있다. 본 연구는 probiotics 유산균이 생성하는 천연의 세포 외 다당류(EPS; exopolysaccharide)를 이용하여 품질이 개선된 Helicobacter pylori을 억제하는 기능성 yoghurt를 제조하고자 우리나라의 식품공전에 식물성 재료로 허가되어 있어 사용 시 안전성에 문제가 없으며 전래된 여러 효능에 착안하여 감초를 이용한 기능성 yoghurt를 개발을 검토하였다. 감초 용매 추출물(1 mg/mL)의 H. pylori KCCM 40449에 대한 항균 활성을 측정한 결과, 40% methanol 추출물은 13.2±0.6 mm, 40% ethanol 추출물은 14.0±0.2 mm, 80% ethanol 추출물은 14.3±0.3 mm, 40% isopropanol 추출물은 15.1±0.3 mm의 생육저지환을 나타내었으며(p < 0.05), 최소 저해 농도(Minimum Inhibitory concentration: MIC)는 각 용매 추출물의 경우 25-100 μg/mL로 나타났다. 40% 에탄올 감초 추출물을 우유 배지에 0.05, 0.1, 0.2%(v/v)의 농도로 첨가하고 Lactobacillus amylovorus DU-21을 접종하여 37oC에서 72시간 동안 발효시키면서, yoghurt의 이화학적 특성 및 관능적 특성을 관찰하였다. pH는 균 접종 직후, 6.44-6.47에서 72시간 발효 후, 3.41-3.51를 나타내었으며, 적정산도는 0.109-0.111%에서 1.021-1.091%로 변화하였다. 점도와 syneresis를 측정한 결과, yoghurt의 점도는 발효 12시간까지 모든 실험구가 급격히 증가하였으며, 그 이후 24시간까지 증가 폭이 다소 둔화된 후, 72시간까지 완만한 증가 양상을 나타내었다. Syneresis는 발효 시간이 증가할수록 감소하였다. 72시간 발효 하였을 때, 감초추출물 0.2% 첨가구에서 12.0±0.4%로 가장 적은 값을 나타내었다. 산생성이 진행됨에 따라 점도가 증가하였으며 syneresis가 감소하는 것을 알 수 있었다. Yoghurt의 Helicobacter pylori KCCM 40449에 대한 항균활성은 첨가군에서 항균활성이 뚜렷이 나타났다(p < 0.05). 이러한 결과는 yoghurt 상등액의 낮은 pH에 의한 영향이라기 보다는 발효 중, 생성된 항균성 물질과 첨가된 감초 추출물의 영향이라고 판단된다. 발효유의 관능검사를 실시한 결과 모든 배양시간에서 첨가구들이 대조구에 비하여 낮은 점수를 받았다(p < 0.05). 이는 감초 특유의 향과 맛에 대해 익숙하지 않는 점과 과도한 산 생성으로 인해 오히려 기호도를 낮춘 것으로 사료된다. 관능검사 결과, 40% 에탄올 감초 추출물을 0.05% 함유한 yoghurt의 향, 맛, 조직감, 색택 및 전반적인 기호도가 다른 처리군에 비하여 월등히 높았다. 이에 감초 추출물을 함유한 yoghurt의 제조 시, 적절한 감초 추출물의 배합비율은 총 중량대비 0.05%인 것으로 판단된다. 식물자원에는 다양한 생리활성물질들이 서로 상호작용하면서 혼합되어 있는 형태로 다양한 질환에 대한 조합치료(combination therapy)에 이용될 수 있으며, 생체 내에서 다중 타겟에 영향을 미치므로 식물작원에 함유되어 있는 다양한 성분이 함유되어 있는 조추출물(crude extract), 용매 분획물(solvent fraction)을 의약품으로 이용하거나, 이로부터 분리된 단일성분 화합물 또는 이에 대한 유도체를 합성하여 새로운 의약품에 대한 개발이 시도되고 있다. 감초의 지표 성분이며, H. pylori에 대한 항균활성이 있는 glycyrrhizin은 비당질의 고감미에 속하는 천연 감미료로서 설탕의 약 200배의 감미를 가지고 있으며, 항산화성을 가지고 있어서 식품 성분의 산화를 방지하여 품질을 안정화시키는 작용을 하는 것으로 알려져 있기 때문에 H. pylori 억제능 이외의 부수적인 효능을 볼 수 있을 것이다. 기호성이 유지되면서 항균활성이 강화된 기능성 yoghurt의 개발을 위해서는 감초 추출물 0.2% 이상 첨가할 수 있도록 적절한 향을 조합하거나, 항균효과와 기호성을 개선할 수 있는 생약제를 이용하여 감초 추출물의 항균 활성을 강화하는 연구가 필요할 것으로 사료된다.

        Twenty four fractions by solvent extraction and/or acid precipitation from fruit body and culture broth of Inonotus obliquus were prepared, and their inhibitory effect against acetylcholinesterase (AChE) was investigated. Among these fractions, acid (1 M HCl) precipitates from cell-free culture broth and fruit body exhibited the highest inhibitory effect on AChE in vitro. Acid precipitates inhibited AChE activity in a concentration-dependant manner and IC50 values of both acid precipitates were 0.53 mg/mL. The inhibition pattern was general non-competitive inhibition. The energetic parameters were also determined by dual substrate/temperature design. Both acid precipitates increased the values of Ea, ΔH, ΔG and ΔH* decreasing the value of ΔS for AChE. The results implied that the acid precipitates from I. obliquus increased the thermodynamic barrier, leading to the breakdown of ES complex and the formation of products as inhibitory mechanism.

        To explore the role of histone deactylase (HDAC) activation in an in vivo model of hypertrophy, we studied the effects of Trichostatin A (TSA). TSA subjected to thoracic aortic banding (TAB)-induced pressure stress in mice. In histological observations, TAB in treated mice showed a significant hypertrophic response, whereas the sham operation remained nearly normal structure with partially blunted hypertrophy. TSA treatment had no effect (measured as HW/BW) on sham-operated animals. TAB animals treated with vehicle manifested a robust ~50% hypertrophic response (p<0.05 vs sham). TAB mice treated with 2 mg/kg/day TSA manifested a blunted growth responses, which was significantly diminished (p<0.05) compared with vehicle-treated TAB mice. TAB mice treated with a lower dose of TSA (0.5 mg/kg/day) manifested a similar blunting of hypertrophic growth (~25% increase in heart mass). Furthermore, to determine activity duration of TSA in vitro, 1 nM TSA was added to H9c2 cells. Histone acetylation was initiated at 4 hr after treatment, and it was peak up to 18 hr, then followed by significantly reduced to 30 hr. We also analyzed the expression of p53 following TSA treatment, wherein p53 expression was elevated at 4 hr, and it was maintained to 24 hr after treatment. ERK was activated at 8 hr, and maintained till 30 hr after treatment suggesting an intracellular signaling interaction between TSA and p53 expression. Taken together, it is suggested that HDAC activation is required for pressure-overload growth of the heart. Eventually, these data suggest that histone acetylation may be a novel target for therapeutic intervention in pressure-overloaded cardiac hypertrophy.

        뽕나무병원성세균에 대한 Beaveria bassiana 6균주의 항균활성을 paper disc법으로 조사하였고 최소성장저해농도(MIC)를 액체배지희석법으로 조사하였다. 항균활성은 B. bassiana J200에서만 나타났으며, Erwinia rhapontici KACC 10407에서는 13mm, Pseudomonas syringae KACC 10390와 Xanthomonas campestris KACC 12134에서는 각각 17 mm의 항균활성이 나타났다. MIC 측정 결과 E. rhapontici KACC 10407, P. syringae KACC 10390 및 X. campestris KACC 12134 모두 4.0%에서 MIC를 보였다. 이상의 결과로 B. bassiana의 뽕나무병원성세균에 대한 생물학적 방재제로서의 이용 가능성을 확인하였다.

        The current study was carried out to investigate the effect of addition of Rhus veniciflua Stokes oil (RVSO) and black garlic extract (BGE) on the lipid oxidation in Hanwoo (Korean cattle) beef model systems. The RVSO at 0.2% inhibited the TBARS (2-thiobarbituric acid reactive substances) formation when tested in liposome model system. The antioxidant effect of RVSO was further found to be similar to 0.2% butylated hydroxy toluene (BHT) and 0.01% vitamin E. On the other hand, BGE at 0.1% also showed the inhibition of TBARS formation in 4% NaCl-added Hanwoo beef patty and found to have slightly lower (p<0.05) effect than 0.1% vitamin E but higher (p<0.05) effect than 0.1% BHT. Results of this study indicated that both RVSO and BGE possess strong antioxidant effects and help to increase the oxidative stability in Hanwoo beef products.

        The purpose of this study was to investigate the effects of virtual reality (VR) therapy with compensation inhibition and feedback (CIF) on upper extremity function in chronic stroke patients. Seven chronic stroke patients participated in this study, which was a randomized controlled trial with a crossover design. Self upper extremity exercise, conservative VR therapy, and VR therapy with CIF were performed for one hour per session, 5 times per week, over a 3 week period. The main outcome measures involved range of motion (ROM) including shoulder, elbow, and wrist joints, a Manual Function Test (MFT), and a Motor Activity Log (MAL). Data were calculated as posttest and pretest changes in every session and were analyzed using Friedman and Wilcoxon signed-rank tests at p<.05. The results were as follows: 1) Statistically significant increase in ROM measurements of shoulder and elbow joints were seen with VR therapy with CIF compared to VR therapy and self upper extremity exercise (p<.05), whereas no significant increasing was noted for the wrist joint (p>.05). 2) Statistically significant increase in the MFT was seen with VR therapy with CIF compared with VR therapy and self upper extremity exercise (p<.05). 3) VR therapy with CIF also resulted in statistically significant increase in both activity of use (AOU) (p<.05) and quality of movement (QOM) (p<.05) on the MAL test when compared with VR therapy and self upper extremity exercise, respectively. In conclusion, VR therapy with CIF was more effective than conservative VR therapy and self upper extremity exercise in improving the upper extremity function in hemiplegic patients with chronic stroke.

        Aphids feed on host plants by penetrating the stems or leaves with stylets. The feeding behavior of aphids consists of probing, penetration, salivation, and sap ingestion. To assess the effects of sound on feeding behavior, we monitored the stylet activity of the green peach aphid, Myzus persicae (Sulzer), using electrical penetration graph (EPG). The use of EPG was critical for determining the stage, frequency, and duration of feeding in aphids. We played back three acoustic stimuli of sine waves with frequencies of 100, 1000, and 5000 Hz to adult aphids. In the sound treatment group, the frequencies of probing, penetration, and salivation increased, whereas the duration of sap ingestion decreased. The results of EPG revealed that the acoustic stimuli may restrict aphid feeding by inhibiting sap ingestion.

        The green peach aphid, Myzus persicae (Sulzer), is one of the most important insect pests in the world. We have explored the possibility of using sounds as a control agent in this species. Sine waves with four different frequencies (100, 500, 1000, 5000 Hz) were used as acoustic stimuli in the playback experiments. As a behavioral bioassay, we recorded honey dew production (HDP), wagging, and walking. HDP occurs regularly at each stage of nymphal and adult periods, and the rate of HDP may indicate a degree of food consumption. Aphids conduct wagging to determine a host plant or to find a feeding site. Walking may be regarded as dispersal. All acoustic stimuli significantly reduced the rates of HDP and increased the rates of wagging. There were no significant effect of acoustic stimuli on walking. Based on the bioassay, the acoustic stimulus with 5000 Hz seemed to be the most effective on inhibiting feeding behavior of this aphid species at an individual level.
