The effectiveness of a crystalline natural barrier in providing sealing capabilities is based on the behavior of numerous fractures and their intersections within the rock mass. It is important to evaluate the evolving characteristics of fractured rock, as the hydro-mechanical coupled processes occurring through these fractures play a dominant role. KAERI is actively developing a true tri-axial compression test system and concurrently conducting hydro-mechanical experiments using replicated fractured rock samples. This research is focused on a comprehensive examination of coupled processes within fractures, with a particular emphasis on the development of true tri-axial testing equipment. The designed test system has the capability to account for three-dimensional stress conditions, including vertical and both maximum and minimum horizontal principal stresses, realizing the disposal conditions at specific underground depths. Notably, the KAERI-designed test system employs the mixed true tri-axial concept, also known as the Mogi-type, which allows for fluid flow into fractures under tri-axial compression conditions. This system utilizes a hydraulic chamber to maintain constant stress in one direction through the application of oil pressure, while the other two directional stresses are applied using rigid platens with varying magnitudes. Once these mechanical stress conditions are established, control over fluid flow is achieved through the rigid platens in contact with the specimen section. This pioneering approach effectively replicates in-situ mechanical conditions while concurrently observing the internal fluid flow patterns within fractures, thereby enhancing our capacity to study these coupled phenomena. As future research, numerical modeling efforts will be proceeding with experimental data-driven approaches to simulate the coupled behavior within the fractures. In these numerical studies, two distinct fracture geometry domains will be generated, one employing simplified rough-walled fractures and the other utilizing mismatched rough-walled fractures. These investigations mark the preliminary steps in the process of selecting and validating an appropriate numerical model for understanding the hydro-mechanical evolution within fractures.
This study examines the changes in chlorogenic acid (CGA), an antioxidant, and one of its decomposition substances, caffeic acid, at various roasting stages and extraction conditions. Based on the CGA content for each roasting stage, at 3℃ after the beginning of the 1st crack, the CGA decreased for washed beans and natural beans by more than 50% compared to that of green coffees. The CGA continued to decrease sharply by more than 75% at the end of the 1st crack for washed beans and at 5℃ after the end of the 1st crack for natural beans. At the peak of the 2nd crack, it had decreased by more than 90% for both beans. The Caffeic acid content gradually increased for both washed and natural beans, then rapidly increased from the beginning of the 2nd crack to the peak of the 2nd crack. However, its contents were very small in quantity. Additionally, the content of CGAs for differing extraction conditions were in the order of 3-CGA, Crypto-CGA, and Neo-CGA. Crypto-CGA content was about half that of 3-CGA and Neo-CGA content was approximately 100 ppm less than that of Crypto-CGA. This study was conducted in order to help make coffee that has the most antioxidant effect.
In the coagulation/sedimentation (C/S) process of the water treatment process, the inflow of massive algal bloom causes many problems including fouling of filter media. This study was conducted to find out the way to remove the algae’s harmful effects by addition of pre-treatment prior to C/S process. Many Jar-tests were conducted such as ① ACF (Algae Coagulation·Flotation) process using natural algae coagulant (Water Health®), ② ACF + C/S process and ③ C/S process with a variety of conditions using cultured algae. The average values of turbidity were ① 0.42 NTU for ACF process, ② 0.13 NTU for ACF + C/S process and ③ 0.25 NTU for C/S process. It was shown that the treatment efficiency of ACF process could get low turbidity results, and ACF + C/S process could achieve more efficient results than those of C/S process. Any negative effects of ACF process to the efficiency of C/S process were not observed in ACF + C/S process. In order to reduce the unfavorable effects of algae, it was found out that the introduction of ACF process in the forms of ① ACF or ② ACF + C/S could be one of the effective and alternative solutions.
This study was performed to investigate improving immune activities of natural water-soluble sulforaphane extracted from Brassica oleracea var. italica by nano encapsulation process. The nanoparticles of the sulforaphane extracted with ultrasonification process at 60℃ promoted human B and T cell growth, about 7~35% compared to the control. The secretion of IL-6 and TNF-α from T cells were also enhanced as 2.6×10-4pg/cell and 2.1×10-4 pg/cell, respectively, by the adding nano samples. NK cell activation was improved about 8%, compare to the control in adding cultured medium of T cell added nano samples. It was also found that sulforaphane extracted from B. oleracea var. italica had highly inhibitory activity on hyaluronidase as IC50 about 200 μg/ml. It can be concluded that natural water-soluble sulforaphane samples by nano-encapsulation, each size is 200 nm, extracted from B. oleracea var. italica has high immune activities through higher efficiency of bio-activation than conventional extracts.
