        상토에 기비로 혼합된 질소 시비수준 차이가 ‘녹광’ 고추의 플러그 묘 생장에 미치는 영향을 구명하기 위해 본 연구를 수행하였다. 코이어더스트, 피트모스 그리고 펄라이트를 용적기준 35, 35 및 30%로 혼합한 상토를 조제할 때 질소를 0, 100, 250, 500, 750, 1,000 및 1,500mg·L-1로 농도를 조절하여 첨가하였고, 질소를 제외한 필수원소는 모든 처리에서 동일한 농도로 조절하였다. 비료를 포함한 상토를 50-cell 트레이에 충진한 후 종자 를 파종하였다. 파종 후 매주 pH와 EC 측정, 파종 0, 3 및 7 주 후 상토의 다량원소 농도 분석, 그리고 파종 7 주 후에 지상부 생장 조사와 식물체 무기원소 함량을 분석하였다. 파종 전 상토의 pH는 질소수준별 차이가 크지 않았지만 육묘기간이 길어질수록 처리간 차이가 커지는 경향이었다. 상토의 EC는 파종 전 질소 시비수준 별 뚜렷한 차이를 보였지만, 파종 4주 이후부터 처리간 차이가 적어졌고, 7주 후에는 모든 처리에서 유사한 수 준으로 측정되었다. 상토 추출용액의 NH4-N 및 NO3-N 농도는 EC와 유사한 경향을 보이며 낮아졌고, 다량원소 농도 역시 파종 3주 이후에 감소폭이 더 커졌다. 파종 후 7주 후 조사한 고추 유묘의 지상부 생장은 500 및 750mg·L-1 처리구에서 우수하였으며, 1,000mg·L-1 이상의 처리구에서는 질소 무시비구와 비슷한 수준으로 생장이 저조하였다. 파종 7주 후 분석한 식물체내 N 함량은 질소 시비수준이 높아질수록 직선적으로 증가하였으며, 지상부 생장이 우수하였던 500 및 750mg·L-1 시비구가 각 각 5.13 및 5.31%로 분석되었다. 이상의 결과를 고려하 였을 때 고추의 유묘 생장을 위해서는 기비로서의 질소 시비수준을 500 또는 750mg·L-1으로 조절하는 것이 바람직하며, 건물중에 기초한 N 함량이 5.1~5.3% 수준으 로 시비농도를 조절하는 것이 바람직하다고 판단하였다.

        Mesenchymal stem cells (MSCs), which are present in all tissues, can differentiate into cells with various specific functions. Recently, cell-based therapies using MSCs have been increasing in the veterinary research and related fields. In this study, we investigated the cellular morphology, proliferating capacities, expression of cell surface markers such as CD13, CD34, CD44, CD45, CD90, and CD105, mesodermal differentiation potentials, and expression of senescence-related markers of p53, p21, and telomerase reverse transcriptase in equine adipose tissue-derived MSCs (eAD-MSCs) after cryopreservation. The eAD-MSCs were analyzed immediately and after being frozen in liquid nitrogen for 1 year (< 1 year, G1) and more than 3 years (> 3 years, G2), respectively. After cryopreservation for 1 - 3 years, G2 eAD-MSCs showed similar cellular morphology, proliferating capacities, and expression of cell surface markers when compared with G1 eAD-MSCs. Moreover, cryopreservation did not affect the adipogenic, chondrogenic, or osteogenic differentiation potentials of G1 and G2 eAD-MSCs. Collectively, cryopreservation for (or over) 3 years maintained the stem cell phenotype and differentiation potentials of eAD-MSCs. These results will be an advantage that can be effectively used for future development of cell-based therapies.

        Transient receptor potential melastatin 8 (TRPM8) plays a crucial role in innocuous cool sensation, acute cold pain and cold-induced hyperalgesia during pathologic conditions. To help understand TRPM8-mediated cold perception in the dental pulp and periodontal tissues, we examined the distribution of TRPM8-immunopositive (+) axons in molar and incisor pulp and periodontal tissues using transgenic mice expressing a genetically encoded axonal tracer in TRPM8+ neurons. In the radicular pulp of the molar teeth, a small number of TRPM8+ axons were observed. TRPM8+ axons branched frequently and extensively in the core of coronal pulp, forming a network in the peripheral pulp. Some TRPM8+ axons ascended between odontoblasts and were observed in the dentinal tubule. TRPM8+ axons were linear-shaped in the radicular pulp, whereas many TRPM8+ axons showed portions shaped like beads connected with thin axonal stands at the peripheral pulp. TRPM8 was densely expressed in the bead portions. In the incisor pulp, TRPM8+ axons were occasionally observed in the core of the coronal pulp and rarely observed at the peripheral pulp. TRPM8+ axons were occasionally observed and showed a linear shape rather than a bead-like appearance in the periodontal ligament and lamina propria of the gingival tissue. These findings, showing differential distribution of TRPM8+ axons between radicular and coronal portions of the molar pulp, between incisor and molar pulp, and between dental pulp and periodontal tissues, may reflect differential cold sensitivity in these regions.

        This study aims to interpret the characteristics of the townscape of the market area in the Hongju Eupseong walled town based on the formation and transformation of modern urban tissue. Based on an analysis of the town plan using a cadastral map, land register, building management register, and aerial images, the change process of the urban tissue and townscape in the market area is divided into three phases: the expansion phase of the market’s function from the 1910s to 1940s, which featured an open market-based townscape; the declining phase of the market’s function from the 1940s to 1960s, which featured a route-shop-based townscape; and the expiration phase of the market’s function after the 1960s. More specifically, with the opening of roads, the functions of the open market gradually diminished from the 1940s on, and after the war, especially during the rapid urbanization of the 1960s, the market area changed into a roadside shop and inner residential area, and the foundation of the current townscape was fixed. Above all, it is noteworthy that since the 1960s, the urban tissue and townscape have formed on the foundation of the former open market based on Japanese capital.

        This study is for the consideration of the existence tendency of Kudoa septempunctata in olive flounder. In general, muscle has shown a strong PCR positive reaction in spores containing tissues rather than non-containing tissues. However, blood PCR results showed opposed tendency. In various organs of the tested fish containing spores in muscle tissue, heart had shown positive reaction along with muscle at PCR analysis. Muscle fiber necrosis was observed at the histological observation, and this degeneration was common in both samples. The one sample was the PCR positive muscle containing spore and the other was the PCR positive muscle non-containing spore. Both of muscle tissues indicated a positive reaction at ISH (in-situ hybridization) against K. septempunctata.

        Connective tissue growth factor (CTGF, CCN2) is one of the multi-functional secreted proteins which belong to CCN family of cysteine-rich growth factors. CTGF is known to have pivotal roles in embryonic endochondral ossification but its role in relevance to periodontitis is never been determined. To identify new molecular mediators associated with periodontitis-induced bone resorption, we have analyzed publicly available GEO database and found the markedly augmented CTGF mRNA expression in periodontitis gingival tissues. The existence of CTGF significantly enhanced mature osteoclasts survival which accompanied by reduction in TUNEL-positive nuclei and PARP cleavage. These results may provide another line of evidence the CTGF mediated prolonged osteoclast survival and subsequent increased bone resorption in the periodontitis patients.

        목 적 : 수술 전·후 연부 조직 종양과 전이암 의심 환자에 대해 역동적 조영증강 검사와 확산 강조영상을 적용하여 도출된 매개변수의 수치와 시간-농도 곡선 그래프를 분석해 악성 병변의 진단 유용성을 확인하고자 한다. 대상 및 방법 : 본 연구는 후향적 연구로서 임상윤리 위원회의 승인을 얻어 진행하였으며 연구 기간은 2015년 02월 01일부 터 2016년 07월 31일까지 핵의학 검사나 전산화 단층 촬영을 선행하고 정확한 병변의 진단을 위해 MRI 검사를 의뢰받은 환자 51명을 대상으로 하였다. 연구에 사용된 프로토콜은 본원에서 시행되고 있는 역동적 조영증강 자기공명 검사에 최적 화된 검사기법과 확산 강조영상을 사용하였다. 정량적 평가는 획득된 데이터를 Tissue 4D를 사용하여 매개변수를 도출하였으며, 확산 강조영상에서 확산 제한되는 부분과 현성 확산 계수에서 신호 감소를 보이는 병변에 관심 영역을 설정하여 계측 하였다. 통계적 분석은 독립 표본 t-test와 상관 분석을 사용하였다. 결 과 : 계측된 매개변수의 평균값은 연부 조직 종양의 경우 Ktrans, Kep, Ve, ADCminimal, ADCmaximal, ADCmean, TCCpattern/type은 각각 0.161 ± 0.012, 0.661 ± 0.041, 0.268 ± 0.003, 0.828 ± 0.393, 1.218 ± 0.502, 1.304 ± 0.435, 2.675 ± 0.674, 5.500 ± 1.874이며, 전이암 의심 환자의 경우는 0.126 ± 0.019, 0.711 ± 0.012, 0.219 ± 0.024, 0.879 ± 0.392, 1.285 ± 0.412, 1.074 ± 0.393, 2.787 ± 0.606, 5.590 ± 1.161로 나타났다(p>0.05). 과 혈관성으로 인하여 확산 제한, 초관 류, ADC 신호감소가 동시에 나타난 경우는 연부 조직 종양은 20명 중 8명이고 평균적인 값들은 Ktrans, Kep, Ve, ADC 순으 로 각각 0.176 ± 0.121, 0.655 ± 0.297, 0.274 ± 0.146, 0.701 ± 0.203 이었다. 전이암의 경우는 31명중 9명의 평균값은 Ktrans, Kep, Ve, ADC 순으로 각각 0.133 ± 0.083, 0.698 ± 0.202, 0.187 ± 0.111, 0.889 ± 0.283 이었다. 혈관종, 탄력 섬유종 연골육종, 내연골종의 경우는 확산 제한과 초관류를 동시에 나타내지 않았다. 결 론 : 악성의 경우 매개변수의 기준값은 Ktrans, Kep, Ve, ADCminimal, ADCmaximal, ADCmean은 0.14 ± 0.09, 0.73 ± 0.47, 0.21 ± 0.11, 0.84 ± 0.40, 1.02 ± 0.40, 1.21 ± 0.43, TCC의 pattern은 Ⅲ(3)이고 type은 ⅴ(5), ⅵ(6), ⅶ(7) 이었다. 악성 종 양의 경우과 혈관성으로 인해 확산 제한과 초관류가 동시에 나타났으며, 역동적 조영증강에서 도출된 Ktrans, Kep, Ve의 값과 TCC 그래프의 pattern 분석을 상호보완적으로 적용했을 때 고식적 검사에 비해 정확한 질환 판별이 가능하였다. 그리고 수 술 전·후 병변에 잔존하는 종양과 항암 치료 후 개선되는 양상을 평가하는데 있어 매우 유용할 것이라 사료된다.

        Aneurysmal bone cyst (ABC) in maxilla is a rare and benign lesion but shows extensive bony destruction, occasionally accompanied with secondary osseous lesions, i.e., central giant cell granuloma, ossifying fibroma, fibrous dysplasia, etc. As the pathogenesis of ABC has not been clearly defined, ABC is diagnostically challenged due to its variable histological features. A 17-year-old boy showed a huge radiolucent lesion at right anterior maxilla, which was accidentally found in routine dental-radiological examination for orthodontic treatment. He had no medical history of systemic disease, and did not remember any traumatic experience on his right anterior maxilla. The radiolucent lesion involved periapical area from right central incisor to right first premolar, and was clinically diagnosed as odontogenic keratocyst. During surgical operation a cyst-like sac was enucleated with severe hemorrhage. In the histological observation the thick fibrous sac showed no lining epithelium, and its luminal side disclosed multiple aneurysmal spaces which were shrunken and almost obliterated. The fibrous sac itself was hyperplastic with abundant vascular channels, and produced fibromatous thickening associated with ossifying trabecular bones. This fibro-osseous tissue was hamartomatous, which was not directly connected and organized with marrow bone of maxilla. Finally, the present case was diagnosed as secondary type ABC differentially from traumatic bone cyst (TBC), odontogenic cyst, and central reparative granuloma. And it was presumed that the hamartomatous proliferation of fibro-osseous tissue in the cystic sac of ABC could produce the swelling pressure effect in the bone marrow similar to the overgrowth of central giant cell granuloma, ossifying fibroma, fibrous dysplasia, etc., in the secondary type ABC.

        Browning is one of the key factors that influenced the callus subculture of tree peony (Paeonia suffruticosa Andr.). Effects of medium composition and exogenous hormones: macro elements of Murashige and Skoog (MS salts) and iron salt (Fe2+), pH, agar and 6-benzylaminopurine (6-BA), 1-naphthaleneacetic acid (NAA) and kinetin (KT) on the callus browning of P. suffruticosa ‘Shan Hu Tai’ in vitro were studied in this paper. Results showed that the browning of P. suffruticosa callus were more sensitive to KT than 6-BA in different concentrations of 6-BA and KT separately with different concentrations of NAA, and reduced to the lowest (13.3%) under 0.5 mg·L-1 NAA plus 0.3 mg·L-1 KT. 1/4 × MS plus 1/4 × Fe2+ was the best basic medium in which the browning rate was only 18.2%. The browning rate of the callus was the lowest of 4.0% under pH 6.5 and the callus grew better in 7.0 g·L-1 agar than others. This study indicated that the best medium preventing P. suffruticosa callus in vitro from browning was: 1/4 × MS medium supplemented with 6.95 mg·L-1Fe2+, 0.3 mg·L-1 KT, 0.5 mg·L-1 NAA, 6.0 g·L-1 agar and 30 g·L-1 sucrose in pH 6.5.

        산업화를 위한 지구자 및 12가지 식물성 원료 추출물의 생리학적 효과를 평가하기 위하여, 만성 에탄 올을 투여한 마우스 혈청 내에서의 생리학적 지표 및 간과 뇌 조직 내에서의 산화적 스트레스에 대한 보호효과를 확인하였다. 에탄올을 투여한 마우스의 혈당은 정상 대조군 그룹(NG)과 에탄올 투여 그룹 (EG)에서 각각 119.43mg/dL, 305.25mg/dL로 나타났고, 에탄올과 혼합 추출물을 동시에 투여한 그룹 (100, 200mg/kg body weight + 25% ethanol 5g/kg body weight, ME100, ME200)에서 각각 272.76mg/dL, 234.60mg/dL로 감소하였다. 혈중 에탄올 함량은 EG에서 4.08mg/dL를 나타내었고 ME100, 200에서 각각 3.85mg/dL, 3.08mg/dL로 감소하였으며, 혈중 아세트알데하이드 함량은 18.72mg/dL에서 각각 15.76mg/dL, 15.16mg/dL로 감소하였다. 또한 ME100, ME200은 혈청 내의 생 리학적 지표에서 간 독성 지표인 glutamine pyruvic transaminase(GPT), glutamic oxaloacetic transaminase(GOT)와 신장 독성 지표인 blood urea nitrogen(BUN), creatine(CRE), 혈중 total cholesterol(TCHO), triglyceride(TG)의 함량이 유의하게 감소하였다. 뇌 조직에서 에탄올에 의해 acetylcholinesterase(AChE)가 EG(116.10%)에서 NG(100.00%)와 비교하였을 때 증가된 활성을 나타냈 으나, ME에서 각각 109.00%와 108.47%로 유의적으로 감소하였다. ME에서 EG에 비해 간과 뇌 조직에 서 superoxide dismutase(SOD)의 함량이 증가하였고, oxidized glutathione(GSH)/total GSH 비율과 malondialdehyde(MDA)의 함량이 감소하였다. 이러한 결과들을 통해 지구자를 포함한 혼합 추출물은 간, 뇌 조직 및 혈액 등에서 만성 에탄올 섭취에 의해 유발된 산화적 스트레스를 효과적으로 보호할 수 있는 제품으로의 개발이 가능할 것으로 판단된다.

        Human tissue-type plasminogen activator (t-PA) is responsible for fibrin-specific plasminogen activation and plays a key role in fibrinolysis thereby aiding breakdown of blood clots in the vasculature. In the present study, in order to develop a system for production of recombinant st-PA and t- PAHis6 proteins in transgenic rice seeds, a DNA fragment encoding t-PA gene was selected and cloned to a plant binary vector (pMJ21) harboring a rice GluB1 promoter, an N-terminal signal peptide of the rice glutelin B1 protein and a Pin II terminator. The constructed plasmid was transformed into Agrobacterium tumefaciens LBA4404 (pSB1) to facilitate introduction into rice callus. The insertion of the st-PA and t-PAHis6 genes into the genome of transgenic rice seeds and their transcripts were confirmed using PCR, and Southern blot as well as RT-PCR, respectively. The highest level of recombinant st-PA expression as determined by enzyme-linked immunosorbent assay (ELISA) was found to be 2,916 ng/total soluble protein (mg) in transgenic rice seeds. The amount of recombinant proteins expressed in transgenic plants was estimated to range from 634 ~ 2,916 ng/TSP mg (st-PA) and 925 ~ 2,640 ng/TSP mg(t- PAHis6), respectively. Immuno-blot analysis of transgenic rice seeds revealed single bands of approximately 68-kDa representing recombinant st-PA and t-PAHis6 proteins. These results demonstrate the expression and in vivo activity of recombinant st-PA and t-PAHis6 in transgenic rice seeds. This study is a promising endeavor for production of recombinant pharmaceutical proteins using rice seed system.

        Effectiveness of transgene transfer into genome is crucially concerned in mass production of the bio-pharmaceuticals using genetically modified transgenic animals as a bioreactor. Recently, the mammary gland has been considered as a potential bioreactor for the mass production of the bio-pharmaceuticals, which appears to be capable of appropriate post-translational modifications of recombinant proteins. The mammary gland tissue specific vector system may be helpful in solving serious physiological disturbance problems which have been a major obstacle in successful production of transgenic animals. In this study, to minimize physiological disturbance caused by constitutive over-expression of the exogenous gene, we constructed new retrovirus vector system designed for mammary gland-specific expression of the hEPO gene. Using piggyBac vector system, we designed to express hEPO gene under the control of mammary gland tissue specific and lactogenic hormonal inducible goat β-casein or mouse Whey Acidic Protein (mWAP) promoter. Inducible expression of the hEPO gene was confirmed using RT-PCR and ELISA in the mouse mammary gland cells treated with lactogenic hormone. We expect the vector system may optimize production efficiency of transgenic animal and reduce the risk of global expression of transgene.

        폴리카프로락톤(PCL)에 NaCl을 혼합한 용액을 블레이드법에 의하여 막형태로 제조한 후 NaCl을 추출하는 염출 법을 이용하여 조직공학적으로 사용할 3차원 다공망을 갖는 멤브레인 형태의 지지체를 제조하였다. 본 연구에서는 성형된 멤 브레인의 건조조건과, NaCl 입자의 크기, NaCl의 혼합량을 각각 다르게 하여 제조하였다. 별도로 제작한 고분자용액 공급장 치를 이용하여 PCL/클로로포름(CHCl3) 용액에 NaCl 입자가 균일하게 혼합된 용액을 유리판에 분주하여 필름 어플리케이터 를 사용하여 블레이드법에 의한 멤브레인을 제조하였다. 멤브레인 지지체에는 NaCl 입자에 의한 거대기공과 거대기공을 이 루는 구조벽에서는 CHCl3의 증발에 의한 미세기공이 함께 복합적으로 상호 연결되어 형성되었다.

        Antibiotic Detection Kit (Combination I), a lateral flow immunoassay (LFIA) developed for detecting antibiotic residues in milk, was utilized for the analysis of antibiotic residues in the muscle tissue of olive flounder. After 5-h treatment of samples by placing them in water dosed with sulfadimethoxine (SDM; 200 g/ton water), the residue depletion of SDM was investigated in 25 cultured olive flounders (Paralichthys olivaceus). Muscles from fish were sampled before treatment and on the 1st, 2nd, 3rd, 4th and 5th days after treatment. The concentration of SDM in the muscle was then determined by LFIA. The absorbance ratio of the sample to the control blank (Bs/Bo) was employed as an index to determine the residue in olive flounder muscle. To investigate the recovery rate, standard solutions were added to muscle samples to obtain final concentrations of 25 and 50 ng/mL in the muscle. The recovery rates of all spiked samples were >96.6% of the spiked value. SDM was detectable in the muscle of fish treated with the drug until the 1st day of the withdrawal period. The present study shows that the LFIA can be easily adopted to detect SDM residues in the tissue of farmed fish.

        배추속은 전세계적으로 재배되는 중요한 경제작물을 포함하고 있으며, 종의 진화 및 배수성 연구를 위해 학문적으로 중요한 가치를 가지고 있다. 그러나 대부분의 작물이 교잡육종을 통해 종자를 생산하기 때문에 고정 계통을 육성하는 데 많은 시간과 노력이 필요하다. 이러한 문제점을 해결하기 위하여 인위적인 방법으로 반수체 식물체를 유도하여 형질이 고정된 계통을 단시간에 육성함으로써 연구 및 육종 소재로 활용할 수 있는 다양한 기내배양법이 시도되었다. 본 논문에서는지금까지 배추속 작물에서 시도된 기내배양법의 연구현황을살펴보고 반수체 생산에 영향을 미치는 요인들을 조사하여 효율적인 반수체 생산 기술에 대한 정보를 제공하고자 하였다.
