        Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and α-melanocyte stimulating hormone(MSH) stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. This study the effects of Radix Trichosanthis on the basal melanogenic activities of B16/F10 mouse melanoma cells, and on the α-MSH induced melanogenesis. Radix Trichosanthis alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Radix Trichosanthis also suppressed the increase of α-MSH (10 nM) induced melanin content and tyrosinase activity. These results suggest that Radix Trichosanthis inhibits melanogenesis and abrogates α-MSH induced melanogenesis in B16 melanoma cells.

        Platelet activation is originated by the intracellular or/and extracellular Ca^(2+). Agonist-induced Ca^(2+) entry through a plasma-membrane pathway has been reported repeatedly, but the mechanisms has proven harder to elucidate. Recently, a number of natural products have been isolated from medicinal plants and marine organisms and have proved to be useful chemical tools for resolving the mechanism of cellular functions. In an attempt to understand the mechanism of platelet activation in Bupleuri Radix, we have studied some aspects of the isolation of active components and their dependence of external Ca^(2+) on platelet activation. Acetone extract of Bupleuri Radix has the most activity on platelet activation and it's active components were identified as saikosaponin a and d. Their optimal concentration was respectively 20 ug/ml and 5 ug/ml and their platelet activation was not dependent on external Ca^(2+), whereas optimal concentration of each agonist was arachidonic acid (10 uM), collagen (10 ug/ml), thrombin (0.1 unit/ml), PAF (5 uM), PMA (5 uM), ionophore A23187 (2 uM) and their dependence of external Ca^(2+) on platelet activation appeared to thrombin$gt;collagen≥PAF$gt;PMA$gt;arachdonic acid$gt;ionophore A23187. These results suggest that saikosaponin is different from each agonists in the dependence of external Ca^(2+) on platelet activation.

        더덕 물추출물이 면역세포에 미치는 영향에 대해 연구한 결과, 더덕 물추출물을 경구투여한 경우 흉선세포의 증식을 촉진하였으나. 흉선세포에 직접 처리한 경우는 그다지 영향을 주지 않았다. 경구투여시 T_H 세포를 활성화하였다. 더덕물추출물은 경구투여시 복강 매크로파지의 NO 생성을 억제하였고, 사람 PMN 세포의 phagocytosis를 증가시켰다. 이 결과는 더덕이 생체내에서 면역작용을 증강시킬 수 있는 것을 시사한다.

        Adriamycin의 독성 및 이의 경감을 위한 백렴의 항산화 활성을 검토한 결과 in vivo에서 adriamycin에 의한 독성발현의 1차적 반응은 지질 과산화에 의한 것이 아닌 다른 경로에 의한 것으로 생각된다. 또한 시료로 사용된 백렴의 체내 지질 과산화 억제에 대한 작용은 뚜렷하지 않았으며, in vivo에서의 thiol기 현저한 증가를 보이는 것으로 보아 시료의 항산화 활성은 체내 glutathione 및 이와 관련한 효소의 활성화의 관련이 있을 것으로 생각된다.

        Adenophorae Radix (AR) has been used as a traditional medicine for various diseases. However, the regulatory mechanisms of AR in allergic inflammation are not yet understood. The present study was conducted to investigate the effect and mechanisms of AR on the mast cell-mediated allergic response. To determine the pharmacological mechanisms of AR in allergic inflammation, we evaluated the effects of AR on the production of tumor necrosis factor (TNF)-α, interleukin (IL)-6, IL-1β and IL-8 as well as the activation of nuclear factor-κB (NF-κB) and caspase-1 in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). Our results demonstrated that AR effectively attenuated the PMACI-induced production of TNF-α, IL-6, IL-1β and IL-8 in stimulated HMC-1. Additionally, we showed that the inhibitory effect of AR on inflammatory cytokines in PMACI-stimulated HMC-1 cells involved the suppression of the activation NF-kB/caspase-1 in PMACI-stimulated HMC-1. Collectively, these findings provide experimental evidence that AR may be a useful candidate for the treatment of allergic inflammation.

        본 연구에서는 다양한 약리학적 활성을 가지는 것으로 알려진 감초 열수추출물(GRW)의 항암효능을 알아보기 위하여 인체 방광암 T24 세포에서 생존율 및 증식억제에 미치는 영향과 이와 연관된 apoptosis 유발 여부 및 관련 인자들의 발현 변화를 조사하였다. 먼저 GRW의 처리에 따른 증식억제 정도를 조사한 결과, GRW 처리 농도 의존적으로 생존율 및 증식억제 현상이 나타났으며, 핵의 형태 변화, DNA 단편화 및 apoptosis 유발에 관하여 조사한 결과 역시 GRW 처리 농도 의존적으로 증가됨을 확인할 수 있었다. 이는 GRW의 처리에 의한 암세포의 증식억제 및 형태적 변형이 암세포의 apoptosis 유발과 밀접한 관련이 있음을 시사하여 주는 것으로 사료된다. GRW 처리에 의한 apoptosis 유발에 관여하는 유전자의 탐색을 위하여 apoptosis와 연관성을 가지는 Bcl-2 family에 속하는 유전자의 발현을 조사한 결과 GRW 처리 농도 의존적으로 Bax 단백질의 발현증가와 더불어 Bcl-2 및 Bcl-xL 단백질의 발현감소가 관찰되었다(Fig. 3A). 이는 GRW에 의한 T24 세포의 apoptosis 유발에 Bcl-2 family에 속하는 유전자의 발현 조절이 중요한 역할을 하는 것으로 사료된다. 또한 GRW의 처리에 따른 MMP의 소실은 미트콘드리아 막의 교란이 유발되었음을 의미하는 것으로, 이러한 MMP 값의 변동은 Bcl-2 family 단백질의 발현 변화에 의한 것이라 추정된다. 한편 Apoptosis에 중요한 역할을 하는 것으로 알려진 caspase(-3/-8/–9)의 발현과 이들의 활성을 억제하는 IAP family (XIAP, cIAP-1, cIAP-2)의 발현에 GRW이 어떠한 영향을 미치는지를 조사한 결과, caspase-3, -8 및 -9의 활성형 단백질 발현 및 정량적 활성증가를 확인하였으며, IAP family 속한 3가지 단백질 모두 발현이 감소하는 것이 관찰되었다. 이상의 결과에서 GRW은 외인적 및 내인적 경로의 개시에 핵심적인 역할을 하는 caspase-8 및 -9의 활성을 모두 증가시켰으며, 이에 따른 caspase-3의 활성증가에 의하여 apoptosis가 유발되었음을 알 수 있었다. 이러한 두 경로의 동시 활성화에는 미트콘드리아의 기능 소실과 Bcl-2 및 IAP family의 발현 변화가 관여하고 있었으며, 특히 Bid의 발현 감소는 GRW에 의한 내인적 경로를 증폭시키는 효과로 작용했을 것이라 추정된다. 방광암의 치료에 보다 효과적인 생리활성을 갖는 물질을 발굴하고 그와 관련된 분자 및 세포수준에서의 기전을 밝히는 것이 중요하기에 본 연구의 결과는 향후 GRW로 수행될 추가 실험을 위한 기초자료로서 그 가치가 매우 높을 것으로 사료된다.

        Background: Taraxacum platycarpum has been used in traditional medicine in Korea to treat intoxication and edema and as a diuretic. According to previous reports, it has anti-cancer, anti-gastritis, and anti-inflammation effects. However, the improvement effect of T. platycarpum on rheumatoid arthritis has not been investigated. The anti-oxidative and anti-inflammation effects of the aerial parts of T. platycarpum are different from those of its subterranean parts. Thus, we evaluated the effect of the water extracts of Taraxaci radix (WTR) on type II collagen-induced rheumatoid arthritis (CIA) in animal models. Methods and Results: Rheumatoid arthritis was induced by type II collagen. WTR (100㎎/㎏ and 500㎎/㎏) was administered to the animal models. Methotrexate was used as the positive control. The levels of interleukin-6, TNF-alpha, and type II collagen IgG in the animals were measured by using enzyme-linked immunosorbent assay. Treatment with 500㎎/㎏ WTR decreased the serum levels of interleukin-6, TNF-alpha, and collagen IgG in the CIA models. Moreover, treatment with WTR diminished the arthritisinduced swelling of the hind legs and monocyte infiltration in the bloodvessels of the animal models. Conclusions: These results indicate that WTR has the potential to improve rheumatoid arthritis by reducing the levels of inflammatory cytokines such as interleukin-6 and TNF-alpha. However, further experiments are required to elucidate the influence of WTR on signal transduction in vitro and in vivo.

        In this study, baicalin, as a marker substance of Scutellariae Radix, was quantitatively analyzed by a high performance liquid chromatography-photodiode array detector (HPLC-DAD). We identified wogonoside, baicalein, and wogonin in the Scutellariae Radix by a high performance liquid chromatography-electrospray ionization-mass spectrometer (HPLC-ESI-MS). The baicalin was separated on a Xterra C18 column (5 ㎛, 4.6 x 250 ㎜) using mobile phase consisting of 38% acetonitrile in 0.68% phosphoric acid. The baicalin spectrum in the Scutellariae Radix extracts was coincided by comparing with UV-visible spectrum (200-550 ㎚) of baicalin standard in the library. The amount of baicalin in Scutellariae Radix was 10.46%, which is higher than KFDA’s guideline. The marker substances of Scutellariae Radix showed a strong base peak [M]+ in the positive detection mode following as; baicalin (m/z; 271 [MH+-sugar]+, 447 [M+H]+), wogonoside (m/z; 285 [MH+-sugar]+, 461 [M+H]+), baicalein (m/z; 271 [M+H]+), wogonin (m/z; 285 [M+H]+). These results are consistent with the fragment pattern and molecular weight of standard components from literature.

        본 연구에서는 하수오를 이용하여 50% 에탄올 추출물과 에틸아세테이트 분획물을 제조하고, 추출물 및 분획물의 항산화, 세포 보호 및 항균 효능을 평가하여 기능성 소재로서의 가능성을 확인하였다. 또한, 하수오에 주요하게 존재하는 성분의 활성도 검증하였다. HPLC-DAD, LC-EIS-MS를 통해 분석한 결과, 하수오의 주성분은 2, 3, 5, 4'-tetrahydroxystilbene 2-O-β-D-glucoside (THSG)이었다. 페놀류 및 THSG 함량은 에틸아세테이트 분획물이 에탄올 추출물 보다 각각 11.1 및 3.0배 높게 나타났다. DPPH 자유 라디칼 및 Fe3+-EDTA/H2O2 시스템에서 생성된 ROS에 대한 소거 활성 평가에서, 에틸 아세테이트 분획물은 에탄올 추출물 보다 뛰어난 소거 활성을 나타냈다. 특히 ROS 소거 활성 평가에서 에틸 아세테이트 분획물과 THSG은 L-ascorbic acid와 동등한 소거 활성을 나타냈다. 이러한 결과를 바탕으로 수행된 광증감 반응으로 유도된 적혈구의 산화적 손상에 대한 세포 보호 활성은 에틸아세테이트 분획물, 에탄올 추출물, THSG 순서로 나타났으며, 모든 실험군이 양성 대조군으로 사용한 (+)-α-tocopherol보다 우수한 활성을 나타냄을 확인하였다. 항균 활성 평가는 S. aureus, E. coli, P. aeruginosa, C. albicans 균주를 대상으로, disc diffusion assay와 broth microdilution assay를 이용하여 수행하였다. 그 결과 추출물, 분획물 및 THSG 모두 모든 균주에 대해 항균 활성을 나타냈으며, 특히 그람 양성균인 S. aureus에 대해 methyl paraben보다 우수한 항균력을 나타냄을 확인하였다. 본 연구의 결과는 하수오가 항산화, 세포 보호 및 항균력에 관한 천연 소재로의 활용될 수 있는 가능성을 시사한다.

        Background: The objective of this study was to make colloidal dispersions of the active compounds of radix of Angelica gigas Nakai that could be charaterized as nano-composites using hot melt extrusion (HME). Food grade hydrophilic polymer matrices were used to disperse these compound in aqueous media. Methods and Results: Extrudate solid formulations (ESFs) mediated by various HPMCs (hydroxypropyl methylcelluloses) and Na-Alg polymers made from ultrafine powder of the radix of Angelica gigas Nakai were developed through a physical crosslink method (HME) using an ionization agent (treatment with acetic acid) and different food grade polymers [HPMCs, such as HP55, CN40H, AN6 and sodium alignate (Na-Alg)]. X-ray powder diffraction (XRD) analysis confirmed the amorphization of crystal compounds in the HP55-mediated extrudate solid formulation (HP55-ESF). Differential scanning calorimetry (DSC) analysis indicated a lower enthalpy (ΔH = 10.62 J/g) of glass transition temperature (Tg) in the HP55-ESF than in the other formulations. Infrared fourier transform spectroscopy (FT-IR) revealed that new functional groups were produced in the HP55-ESF. The content of phenolic compounds, flavonoid (including decursin and decursinol angelate) content, and antioxidant activity increased by 5, 10, and 2 times in the HP55-ESF, respectively. The production of water soluble (61.5%) nano-sized (323 ㎚) particles was achieved in the HP55-ESF. Conclusions: Nano-composites were developed herein utilizing melt-extruded solid dispersion technology, including food grade polymer enhanced nano dispersion (< 500 ㎚) of active compounds from the radix of Angelica gigas Nakai with enhanced solubility and bioavailability. These nano-composites of the radix of Angelica gigas Nakai can be developed and marketed as products with high therapeutic performance.

        Background : Phlomidis radix is known to be effective for pain relief and anti-inflammation. Phlomidis radix (Han-Sok-Dan) is likely to be confused because it is similar in name to Dipsaci Radix (Cheon-Sok-Dan), or is similar in morphology to other Phlomioides species. Therefore, we investigated the origins and morphological characteristics of the origin plant of Phlomidis radix in order to cultivate and consume the correct one. Methods and Results : We reviewed North and South Korean and Chinese Pharmacopoeia and the books on oriental herbal medicine and botanical illustrations. In South Korea, P. umbrosa was defined as the origin plant of Phlomidis radix, whereas in North Korea, P. maximowiczii was defined. In China, it was not used as an oriental medicine. In the past, P. umbrosa, which is native to the Korean peninsula, or P. maximowiczii, which is native to the northern part of the Korea was used instead of Dipsacus asperoides for Dipsaci Radix. However, after the publication of the Korean Pharmacopoeia, Phlomidis radix and Dipsaci Radix have been distinguished, and only P. umbrosa was defined as the origin plant of Phlomidis radix. Therefore, Phlomidis radix and Dipsaci Radix should be distinguished from each other, the origin plant of Phlomidis radix is considered to be P.umbrosa. Furthermore, we investigated the external morphological and growth characteristics of Phlomoides species in Korea. There are three species of Phlomoides in Korea, such as P. umbrosa, P. maximowiczii, P. koraeinsis. P. umbrosa has no basal leaves but only stem leaves, while P. maximowiczii and P. koraeinsis have basal leaves. Also, P. maximowiczii is distinguishable from seeds with hairs while P. koraeinsis from hairless seeds. Conclusion : We considered P. umbrosa as the origins of Phlomidis radix. As a result of investigation of morphological and growth characteristics, P. umbrosa, P. maximowiczii and P.koraeinsis could be distinguished by the presence or absence of basal leaves and the difference in whether the seeds have hair or not. These results could be used as a basis for standard varieties research and cultiviation of P. umbrosa.

        Background : Brain waves are generated by the neurons in the brain. Many scientists fixed the electrodes of the electroencephalogram (EEG) on the scalp to record the signal expression of continuous cerebral activity. EEG is widely used in clinical diagnosis. It can be used to screen epilepsy, depression, ementia, stroke, encephalitis, ADHD and other brain diseases. Also, many local and foreign studies on aromatherapy focused on the discussion of human skin absorbing essential oils. This study discusses the level of relaxation efficacy of inhaled essential oils via the human olfactory organs. Thus, this study intended to observe the effects of essential oil of Aucklandiae Radix and Cyperi Rhizoma on EEG activity. Methods and Results : We evaluated the effects of essential oil of Aucklandiae Radix and Cyperi Rhizoma, on human EEG activity. For this purpose, essential oil was extracted from Aucklandiae Radix and Cyperi Rhizoma by the supercritical extraction method. The effect of its inhalation on EEG activity was evaluated by measuring the EEG power spectrum in 20 healthy participants. EEGs signals were recorded using QEEG-8 system (LAXTHA Inc., Daejeon, Korea) from 8 grounding electrodes. The subjects were instructed to sit quietly, close their eyes and to breathe normally during the measurement. The fragrance stimulus was dipped in a paper (14 ㎝2) then placed about 3 cm in front of the subject's nose. EEG was recorded before and during the fragrance exposure for 30 seconds. The mean power values (μV) calculated for fast fourier transform (FFT) analysis indicators. The results were as essential oils of Aucklandiae Radix and Cyperi Rhizoma decreased the overall brain relative theta (θ waves), but increased relative beta (β waves), absolute gamma (γ waves), spectral edge frequency (SEF) and spectral edge frequency 95% (SEF95). Conclusion : The changes in the EEG activities following inhalation of Aucklandiae Radix and Cyperi Rhizoma suggest that the oil can improve psychological well-being by increasing attention and relaxation.

        Background : Polygonati rhizoma is used for medicine using rhizome of Polygonatum sibiricum Redoute belonging to Liliaceae. This study was carried out to find proper method to establish the guideline of GAP post-harvest management. Methods and Results : Six-years-old polygonati rhizoma harvested in march in Geumsan was used for this study. Steaming was treated for 1, 2, and 3 hours respectively and drying was treated at 45℃, 50℃, 55℃, and 60℃ using hot-air drying machine with checking moisture content at intervals of a day. Moisture contents of polygonati rhizoma according to drying temperature after steaming showed that drying was faster in higher drying temperature treatment after steaming. Moisture content was below 10% as 9.6% and 9.4% at 55℃ drying treatment after steaming for 1 hour and 2 hours respectively. Color value according to treatments was as follows, L-value was lower in longer steaming and higher drying temperature. a-Value was higher in steaming an higher drying temperature. Conclusion : In steaming and hot-air drying treatment to establish the guideline of GAP post-harvest management, moisture content was most proper as 9.4% below 10% in 2-hour steaming and 55℃ hot-air drying for 6 days. L-value was 27.49 and a-value and b-value were 4.38 and 9.73 in this treatment and dried rhizome looked glossy and transparent with higher quality. Proper drying method for polygonati rhizoma was considered as 2-hour steaming and 55℃ hot-air drying for 6 days.

        Paeoniae Radix has been used as a traditional medicine for various diseases including hepatic disease. However, the inhibitory effect of Paeoniae Radix on intestinal inflammation has not been fully understood yet. The aim of this study was to evaluate the effect of Paeoniae Radix on colitis induced by dextran sulfate sodium in mice. To investigate the protective effects of Paeoniae Radix, the colitis mice were induced by drinking water containing 5% dextran sulfate sodium for 7 days. Mice were randomized into groups receiving Paeoniae Radix (100 ㎎/㎏), sulfasalazine (150 ㎎/㎏) as a positive control, or water as a negative control. We evaluated the effects of Paeoniae Radix on clinical signs induced by dextran sulfate sodium, measuring weight loss, colon length, and disease activity index. Additionally, to find a possible explanation for the anti-inflammatory effects of Paeoniae Radix, we evaluated the effects of Paeoniae Radix on the interleukin-6 and cyclooxygenase-2 levels in colitis tissue. The results indicated that mice treated with dextran sulfate sodium showed measurable clinical signs, including weight loss and reduced colon length. However, Paeoniae Radix treatment significantly improved the weight loss and disease activity index as clinical symptoms. Moreover, Paeoniae Radix inhibited the interleukin-6 and cyclooxygenase-2 expression levels in colon tissues treated with dextran sulfate sodium. Taken together, the findings of this study suggest that Paeoniae Radix may be useful for treating intestinal inflammation, including ulcerative colitis.

        The main goal of this work was to investigate changes in microbiological quality and dominant bacterial communities present in minimally processed Platycodi radix (Doraji) under vacuum packaging during cold storage using matirix-assisted laser desorption/ionization time of flight (MALDI-TOF) mass spectrometry fingerprinting. Microbiological analysis of raw Platycodi radix throughout the manufacturing process highlighted the presence of high numbers of total viable cells both before and after storage. Further, the dominant bacterial communities changed during processing and storage, with greater richness detected in Enterobacteriaceae; specifically, Serratia fonticola and Rahnella aquatilis became abundant in Platycodi radix after processing. During storage at 5 and 15℃, the quality diminished and the relative abundance of Leuconostoc spp. increased; Leuconostoc pseudomesenteroides and Leuconostoc citreum were the main bacterial communities in decayed Platycodi radix at the end of storage. Microorganisms present in Platycodi radix products during processing and storage are potentially involved in deterioration. The identification of dominant bacteria can be helpful in the effective establishment of packaging and storage conditions to prolong the freshness of minimally processed Platycodi radix products.

        이 연구는 도라지의 유기재배를 위하여 3년근 이식 예정지에 풋거름 작물 재배에 따른 도라지 뿌리의 생육특성 및 사포닌 함량에 미치는 영향을 구명하고자 실시되었다. 생육특성에서는 기재배지에서의 뿌리의 직경 및 곁뿌리 수는 호밀 재배구에서 가장 굵거나 많았으며, 뿌리 생체중은 풋거름작물 재배구간 차이는 없었으나, 대조구에 비해 높게 나타났다. 도라지 뿌리의 무기성분 함량에서는 무기성분별 유의성이 있는 것으로 나타났으며 특히, 기재배지에서의 호밀 처리구에서 높은 함량이 나타났다. 사포닌 함량의 변화에서는 기재배지에서는 호밀 재배구에서 모든 사포닌 함량이 가장 높은 함량으로 나타났으며, 새재배지에서는 헤어리베치 재배구에서 높은 함량을 나타냈다. 대조구에 비해 풋거름작물 재배구의 기재배지와 새재배지 모든 재배구에서 사포닌의 함량이 높게 나타났다. 따라서 도라지 유기재배 시 풋거름작물에 따른 뿌리의 생육 특성 및 약리성의 변화 등의 유의적인 차이가 나타나는 것을 확인할 수 있었으며, 연차 간 재배 시 호밀 재배구에 생육 및 약리성이 우수한 것으로 나타났다.

        Background : To evaluate the quality of Glycyrrhizae radix (licorice), a popular herbal medicine in Korea, it is necessary to investigate the component profiles of the Glycyrrhiza species distributed in Korean market. Specifically, to control the quality of Korean Glycyrrhizae radix preparations including raw materials, glycyrrhizin (> 2.5%) and liquiritigenin (> 0.7%) has been used as a standard marker compound in Korea Food and Drug Administration (KFDA). To the best of our knowledge, there have been only a few studies regarding the fingerprinting techniques and metabolomics approach of the phytochemicals in Glycyrrhizae radix in Korean market. Therefore, it is necessary to develop a validated method for botanical raw material (BRM) standardization. Methods and Results : Twenty-one Glycyrrhizae radix (G. glabra, G. uralensis, and G. glabra × G. uralensis) were analyzed by UPLC-QTOF-MS and were estimated to statistical and multivariate analysis. Large scale multi-targeted metabolic profiling and fingerprinting techniques were utilized to access a broader insight into the chemical composition of Glycyrrhiza species. The UV/VIS spectra, and MS data (accurate mass and fragmentation pattern in negative mode) of the components in the BRM of G. glabra, G. uralensis, and G. glabra × G. uralensis (incorporation and hybridization) were analyzed with multivariate analyses. Interestingly, UPLC-QTof-MS derived PCA analysis and fingerprinting techniques was found to be effective techniques in the classification of species based on genetic and/or geographical origins. In addition, the revealed metabolite markers could be utilized as a guidance for the BRM standardization as well as a database for quality control of commercial products containing Glycyrrhizae radix. Conclusion : From the above results, the revealed metabolite markers could be utilized as a guidance for the BRM standardization as well as a database for quality control of commercial products containing Glycyrrhizae radix.
