This study was conducted to determine the proximate compositions, nutritional components, and antioxidant effects of white and brown enoki mushrooms (Flammulina velutipes). The crude protein and carbohydrate contents were higher in the brown than white mushrooms, whereas the moisture, crude ash, crude lipid, and dietary fiber levels were lower. The mineral contents of the white mushroom was higher than levels obtained in the brown mushroom for the detected components (Ca, Cu, K, Mn, Na, and P). The amount of vitamin B3 in the brown mushroom was 1.51 mg/100 g, which was 4.5 times higher than that in the white mushroom. The major fatty acids detected were palmitic acid, linoleic acid, and α-linolenic acid. The total polyphenol and flavonoid contents were highest in 70% ethanol extracts of the white and brown mushrooms, respectively. For the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, a 70% methanol extract of the white enoki mushrooms showed an activity of 76.4% (p<0.05). For the ferric-reducing antioxidant power (FRAP) activity, a 70% methanol extract of the brown enoki mushrooms showed the highest value. Further, the total flavonoid contents were significantly correlated with the DPPH and FRAP activities.
This study was conducted to find out the optimal solvent extraction method [Distilled water (DW), 70% ethanol, 99% ethanol] of mushrooms, including Pleurotus ostreatus (Jacq.) Que, Pleurotus eryngii and Flammulina velutipes and improve their usability as natural antioxidants. To analyze antioxidant activities in each mushroom, total polyphenol, flavonoid contents, 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2?-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) cation radical (ABTS+) and fluorescence recovery after photobleaching (FRAP) were measured. All mushrooms showed the highest total polyphenol contents in DW mushroom extract (p<0.001). Total flavonoid contents were the highest in P. eryngii and F. velutipes DW and 70% ethanol mushroom extracts (p<0.05). All mushrooms showed the highest activities using DPPH and FRAP assays in the DW extraction method (p<0.001). P. ostreatus (Jacq.) Que and P. eryngii showed the highest ABTS+ radical scavenging activity in the DW extraction method, and F. velutipes showed the highest activity in the 70% ethanol extraction method (p<0.001). As a result of comparing IC50 values of DPPH and ABTS+ radicals and FRAP EC50 values, the DW P. ostreatus (Jacq.) Que extract showed high antioxidant activities (p<0.001). Pearson's correlation between total polyphenol contents and antioxidant activities showed a positive correlation in all mushrooms (p<0.01). Therefore, extraction of the mushrooms with DW can enhance the extraction of effective bioactive substances and antioxidant activity.
Centella asiatica (CA) is a vegetable that has been used for medicinal purposes for a long time, but it is not well known in Korea. In this study, the approximate analysis, physicochemical properties, free sugars, free amino acids and minerals of native and improved CA cultivated in Chungju area were measured. And the antioxidant contents and activities of CA solvent extracts were measured. There was no significant difference between native and improved species. As for the characteristics of CA, it was confirmed that the improved species had a large weight, size and a tough texture. Glucose was detected in native CA, fructose, glucose and sucrose were detected in improved species. A total of 15 free amino acids were detected and the content was slightly higher in improved species than in native species. The mineral content in CA was slightly higher in the improved species, and the detected minerals were Na, P, Ca, K, Mg and Fe. Polyphenols, ascorbic acid and flavonoids in each extraction solvent (water, 70% and 100% ethanol) were all higher in the improved species than in native species. And it was found that the antioxidant activities of ethanol extracts were higher than that of water.
Centella asiatica (CA) has been widely used as herbal plants. It is a valuable resources. The aim of the present study was to evaluate physiological activities of solvent extracts from CA cultivated in Chungju, Korea (Good tiger care). After preparing water (cold-water, CA-WE; hot-water, CA-HWE) and EtOH extracts (50% EtOH, CA-50E; 70% EtOH, CA-70E), their total polyphenol, flavonoid, and triterpenoid contents, and anti-oxidant activities, and anti-inflammatory activities were examined and compared. CA-70E showed the most potent anti-oxidant activity based on ABTS radical scavenging activity and reducing ability assays, while CA-50E and CA-70E showed the highest DPPH radical scavenging activity. Among major triterpenoid glycosides present in CA, madecassoside and asiaticoside contents were found to be the highest in CA-70E, and madecassic acid and asiatic acid were the highest in CA-50E. In LPS-stimulated RAW 264.7 cells, CA-70E showed the highest TNF-α inhibitory activity, although CA-50E and CA-70E similarly inhibited nitric oxide production. Ethanol extracts significantly inhibited IL-6 and IL-8 production more than water extracts using TNF-α/IFN-γ-stimulated HaCaT cells, indicating its better inhibitory against skin inflammation. Therefore, Chungju-CA EtOH extract, especially 70% EtOH extract, has high physiologically active ingredients and potent anti-oxidant and anti-inflammatory activities, suggesting its industrial application as a functional material.
본 연구는 열수와 에탄올 용매에 따른 복령 및 산수유 추출물의 농도별로 항산화 활성, 항염증 효과 및 신경세포 보호효과에 미치는 영향을 살펴보고자 진행하였다. 추출물들의 총 polyphenol 함량은 복령 열수 추출물에서 가장 높았고, 복령 에탄올 추출물, 산수유 열수 추출물, 산수유 에탄올 추출물 순으로 유의적으로 감소되는 경향을 나타냈다. DPPH 및 ABTS radical 소거 활성능은 추출물의 농도 의존적으로 증가하였고 산수유 열수 추출물은 모든 추출물에서 항산화 활성이 가장 높게 나타났으며, 복령 열수 추출물은 에탄올 추출물에 비해 높은 활성이 확인되었다. 복령과 산수유에 함유된 총 polyphenol과 항산화 성분을 추출하기 위해서는 에탄올 추출방법 보다는 열수 추출방법이 효과적인 방법이라고 사료된다. LPS로 염증이 유도된 RAW 264.7 세포에서 복령추출물이 산수유 추출물에 비해 NO 생성 억제 효과가 우수한 것으로 확인되었다. MPP+에 의해 유도된 SH-SY5Y 신경세포에 복령 열수 추출물은 산수유 열수 추출물에 비해 산화적 스트레스로부터 신경세포 보호효과가 우수하게 나타났다. 복령과 산수유 추출방법에 따른 항산화, 항염 및 산화적 스트레스로부터의 신경세포 보호효과를 갖는 기능성 소재로서 가능성이 있다고 사료된다.
In order to examine the functionality of broccoli sprout (Brassica oleracea, BS), solvent extracts were prepared and their anti-oxidative and immunomodulating activities were compared with those of broccoli (B). EtOH extracts (E) were potently higher than hot-water extracts (HW) in the antioxidant contents and radical scavenging activity. In particular, the total polyphenolic contents in addition to ABTS and DPPH radical scavenging activity were significantly higher in EtOH extract of broccoli sprout (BS-E; 9.15 mg GAE/g, 4.52 mg AEAC/g, and 1.14 mg AEAC/g) compared with that of broccoli (B-E; 7.83 mg GAE/g, 3.63 mg AEAC/g, and 0.97 mg/AEAC/g). Whereas, total flavonoid content was significantly higher in B-E (1.60 mg QE/g) than BS-E (1.43 mg QE/g). Anti-inflammatory activity was investigated using LPS-induced cell line model at a concentration of 10~100 μg/mL, in which all solvent extracts of both broccoli sprouts and broccoli were not toxic to RAW 264.7 cell lines. In anti-inflammatory activity of broccoli sprouts, EtOH extracts also showed significantly more potent activity than hot-water extracts in all sample concentrations tested. In addition, BS-E (100 μg/mL) inhibited nitric oxide (NO) and IL-6 production to 60.9% and 68.9% compared with the LPS inflammation group (without extracts), whereas B-E inhibited 49.6% and 54.9%. On the other hand, in immunostimulating activity by splenocytes and macrophages, hot-water extract showed significantly higher activity than EtOH extract. Especially, BS-HW stimulated the splenocyte proliferation (1.2-fold against saline group) and IFN-γ production (264.39 pg/mL) at 100 μg/mL, and the production of IL-6 (1.33-fold), IL-12 (1.09-fold) and TNF-α (1.49-fold) from macrophages was also significantly enhanced over broccoli. In conclusion, broccoli sprouts showed more potent anti-oxidative, anti-inflammatory and immunostimulating activity than broccoli, suggesting the possibility of using broccoli sprouts as functional food materials.
The objective of the present study was to determine the total phenol and flavonoid contents and to evaluate the antioxidant potential, of different solvent extracts (ethyl acetate, n-butanol, chloroform, and water) from Robus idaeus in various radical scavenging models (DPPH activity, superoxide dismutase (SOD) activity, reducing power, and nitrite scavenging activity), along with their antimicrobial potential. Measurement of total phenol and flavonoid content of the ethyl acetate extract of R. idaeus was found to be significantly higher than those of the other extracts. The ethyl acetate extract (at a concentration of 1,000 μg/mL) showed significantly higher reducing power and DPPH radical scavenging activity as compared to the other extracts. Results were dose-dependent. Moreover, the ethyl acetate extract of R. idaeus (1,000 μg/mL) showed potent antioxidant efficacy (85.5±1.18%) as evidenced by nitrite scavenging ability at pH 1.2. All solvent extracts of R. idaeus showed lower SOD-like activity (13.72~20.54%). In addition, the antimicrobial activity of all solvent extracts except water extract showed strong inhibition (inhibitory zones in mm) of Staphylococcus aureus (19.40±1.00 mm) and Bacillus cereus (20.50±0.21 mm) growth. In particular, ethyl acetate extracts (100 mg/mL) showed antimicrobial activity comparable to that of tetracycline (0.01 mg/mL), which was used as a positive control. The results of this study indicate that the ethyl acetate extract of R. idaeus is a natural antioxidant and antimicrobial, with enriched phenols and flavonoids concentration, that has potential in the development of health-enhancing food products.
본 연구에서는 국내에 유통되는 생약 원료에 대하여 유기 용매 추출 시에 돌연변이원성이 나타나는 지에 대한 조사를 실시하였다. 추출방법으로는 70% ethanol로 10가지 생약원료 를 모두 추출하여 돌연변이원성을 Ames test를 이용하여 측 정하였고, 또한 70% ethanol 추출물로부터 dichloromethane 및 ethyl acetate를 이용하여 각각 추출한 분획에 대해서도 조사 를 실시한 결과, 70% ethanol에서는 S9 mix가 첨가된 경우, 황기, 천궁 및 산약에서 돌연변이원성이 나타났고, 70% ethanol 추출물로부터 dichloromethane 및 ethayl acetate로 분획한 것 에서는 당귀, 황기, 천궁, 숙지황 및 산약에서 돌연변이원성 이 나타났다. 위의 모든 생약의 경우, S9 mix가 첨가되지 않 은 경우는 돌연변이원성이 나타나지 않았고, S9 mix가 첨가 한 경우에만 돌연변이원성이 나타나는 것으로 보아, 간접돌 연변이원이 본 시험에서 사용한 유기용매에서 용출되는 것 으로 사료된다.
The purpose of this study was performed to determine the whitening effect of organic solvent extracts from the centipede, Scolopendra subspinipes mutilans. We prepared different concentrations (50%, 70% and 100%) of ethanol, methanol, 100% ethyl acetate and water extracts. We tested melanin inhibitory effect and tyrosinase activity using B16/F10 melanoma cell. As a result, treatment of organic solvent extracts is decreased the biosynthesis of melanin and tyrosinase activity to 30~60%. Especially the 70% ethanol extracts was the most effective in B16/F10 melanoma cells. In the study on melanogenic protein expression, 70% ethanol extracts of Scolopendra subspinipes mutilans blocked glycosylation of tyrosinase. Therefore this result suggests that 70% ethanol extracts could be developed as a skin whitening agents.
천연 살초성분 해발을 위한 기초적 연구로 발아 및 생장억제물질 동정을 위하여 몇 가지 유기용매에 따른 황련의 분획 추출물에 대한 다른 식물의 종자발아 및 유묘생장에 미치는 영향을 조사하였다. 추출물에 대한 검정식물의 발아율은 들깨, 배추, 피, 참깨 순으로 억제되었으며, 2,000ppm ethyl acetate추출물에서 가장 높은 발아억제를 보였다. 초장 및 뿌리생육은 참깨, 들깨, 배추, 피 순으로 억제되었으며, 특히 뿌리신장은 H2O, butyl alcohol, hexane, ethyl acetate 추출물 순으로 억제정도가 심하였다 추출물에 대한 발아 및 유묘생육의 억제정도는 참해 및 배추에서 종특이적으로 작용하였으며, 특히 물 추출물은 종자발아 및 뿌리 신장에 천연살초활성을 보였다. 이러한 결과들은 금후 천연제초제 개발을 위한 기초자료로 이용될 것으로 기대되었다.
천연첨가물검색의 일환으로 초피의 용매 추출물의 항산화성과 항균성을 실험하고, 소비자의 수용도를 검토하기 위하여 초피를 첨가한 육원전의 관능 검사를 실시한 결과는 다음과 같았다. 1. 초피 용매 추출물들의 옥수수유에 대한 항산화 효과는 가장 많이 사용되는 합성항산화 제인 BHT과 같은 농도에서 BUex는 월등히 높았으며, MEex, EAex는 유사한 경향을 나타냈다. 2. 초피의 용매 추출물의 중 CHex와 BUex가 Escheria coli, Salmonella typhimurium, Psudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus 및 Listeria monocytogenes 모두에 대하여 항균효과를 보였다. 3. 초피 가루를 식품에 첨가하여 제조한 육완자전에 대한 소비자의 기호도를 검사한 결과는 0.1%를 첨가한 경우는 초피를 첨가하지 않은 본래의 육완자전과 같은 수준의 기호도를 나타내므로써(P〈0.05) 초피 가루에 의한 이미(異味)가 없어 육와자전 제조에 초피가루를 사용 할 수 있을 것으로 사료된다.
The extraction yield of MeOH extract of green teas, oolong tea and black tea were 3 to 5, 4 to 5, and 5 to 7 fold higher than those of EtOH and EtAC extract, respectively. The amount of total catechins of EtAC extract of the black tea, and of the green teas and oolong tea were three- and two-fold higher than that of EtOH or MeOH extract of the corresponding teas, respectively. The antioxidative activities of EtOH, MeOH and EtAc extract were considerably higher than that of BHT and dl-α-tocopherol at 200 ppm level. The antioxidative activities of EtOH and MeOH extract at 200 and 500 ppm level, and of EtAc extract at 200 ppm level varied depending on the type of tea as follows : green tea I $gt; green tea II $gt; green tea III $gt; oolong tea $gt; black tea. The antioxidative activity increased as the content of EGC increased. But the antioxidative activity of MeOH extract at 1000 ppm level, and of EtAc extract at 500 and 1000 ppm level were not affected by the content of EGC and EGCG.
The extracts of Quercisemen(Quereus acutissima Carruthers seed) powder was obtained by the extraction with distilled water, water : acetonet(1:1, v/v), ethanol and ethyl ether, respectively. As a result this experiment, the antioxidative activity of each solvent extracts on linoleic acid were examined as follows: 1. Each fraction extracted by the acetone : water(1:1, v/v), water and ethanol respectively showed high antioxidative activity. 2. A fraction of the tannin extracted from the solvent, acetone, water(1:1, v/v) showed even more antioxidative activity than that of the α-tocopherol or BHT. 3. Acceleration of peroxide reaction by Fe++ and Cu++ on the linoleic acid was strongly inhibited by adding the tannin, 4. Organic acid, such as malic acid, citiric acid and tartaric acid with the tannin were showed the synergistic effect fo the antioxidation reaction.
