        본 연구의 목적은 포졸란의 항생제 내성균과 장내 유해 미생물 억제에 관한 체외배양 항균 활성을 조사하는 것이었다. 대조군 (CON, 포졸란 무첨가 대조군)과 DP0.3 (증류수와 포졸란 분말 0.3%를 혼합하여 제조한 배지군), DP0.5 (증류수와 포졸란 분말 0.5%를 혼합하여 제조한 배지군), PE (포졸란 분말 추출물을 이용하여 제조한 배지군)으로 구분하였다. Lacctobacillus casei 균수는 CON과 비교할 때 DP0.3 처리군에서 유의하게 높았으나 (P<0.05) 기타 처리구 사이의 차이는 없었다. Clostridium butyricum, E. coli, Salmonella typhimurium 균수는 CON과 비교할 때 포졸란 처리구가 유의하게 낮았다 (P<0.05). Clostridium butyricum, Salmonella typhimurium 균수는 DP0.3, DP0.5와 PE 사이, E. coli 균수는 DP0.5와 PE 사이의 유의차가 있었다 (P<0.05). MRSA와 VRE의 균수는 CON과 비교할 때 포졸란 처리구가 유의하게 낮았다 (P<0.05). MRSA 균수는 DP0.5와 DP0.3, PE 사이의 차이가 있었고, VRE 균수는 PE> DP0.3> DP0.5> CON 처리구 순서로 유의하게 높았다 (P<0.05). 이와 같은 결과를 통해 포졸란의 항생제 내성균 및 장내 유해 미생물에 대한 항균활성이 확인됨에 따라 가축용 천연항균제, 보조사료 규산염제로써의 활용이 가능할 것으로 보인다.

        Indoor air contaminated with various pollutants commonly poses a risk to human health, and the need for installing air purifiers has been increasing. However, in commercial air purifiers pollutants-removal efficiency and durability are generally low. Since silver nano-composites are known to have catalytic oxidation and antibacterial capacities, it was anticipated to be applicable for indoor air purifiers. In this study, silver nano-composites were applied to granular activated carbon and scrubber solutions to treat a mixture of three air pollutants including toluene, formaldehyde, and bioaerosol. In the activated carbon deposited with silver nano-particles, the specific surface area decreased, resulting in a 10% loss of adsorption capacity for toluene. However, the removal efficacy of formaldehyde and bioaerosol increased by 10% due to the catalytic oxidation and antibacterial capacities. In the scrubber operation with silver nano-particles, the removal rates of formaldehyde and bioaerosol improved by 20%, while toluene removal was not observed. When the activated carbon column and the scrubber was connected in series, toluene was mainly removed by the activated carbon, and the removal rates of formaldehyde and bioaerosol increased in the presence of silver nano-particles. Consequently, for the improvement of indoor air quality, it is deemed appropriate to apply silver nano-material to indoor environments contaminated with pollutant mixtures.

        We prepare ZnO nanoparticles by environmentally friendly synthesis using Cyathea nilgiriensis leaf extract. Various phytochemical constituents are identified through the assessment of ethanolic extract of plant Cyathea nilgiriensis holttum by GC-MS analysis. The formation of ZnO nanoparticles is confirmed by FT-IR, XRD, SEM-EDX, TEM, SAED and PSA analysis. TEM observation reveals that the biosynthesized ZnO nanopowder has a hexagonal structure. The calculated average crystallite size from the high intense plane of (1 0 1) is 29.11 nm. The particle size, determined by TEM analysis, is in good agreement with that obtained by XRD analysis. We confirm the formation of biomolecules in plant extract by FT-IR analysis and propose a possible formation mechanism of ZnO nanoparticles. Disc diffusion method is used for the analyses of antimicrobial activity of ZnO nanoparticles. The synthesized ZnO nanoparticles exhibit antimicrobial effect in disc diffusion experiments. The biosynthesized ZnO nanoparticles display good antibacterial performance against B. subtilis (Gram-positive bacteria) and K. pneumonia (Gram-negative bacteria). Bio-synthesized nanoparticles using green method are found to possess good antimicrobial performance.

        This study was investigated the synergistic antibacterial effects from the combination of sulfamethazine (SMZ) and tiamulin (SMT) against Salmonella Typhimurium (S. Typhimurium) and Actinobacillus pleuropneumoniae (A. pleuropneumoniae) isolated from pigs. Minimum inhibitory concentrations (MICs) and fractional inhibitory concentration (FIC) index were determined for 10 bacteria from 13 swine farms in Gyeongsangnam province by the National Committee for Clinical Laboratory Standards broth microdilution procedure. Antimicrobial agents tested included SMZ, tiamulin and SMT. Against S. Typhimurium and A. pleuropneumoniae, the MIC ranges of SMZ, tiamulin and SMT were 125-500, 15.625-62.5 and 15.625-62.5 μg/mL, respectively. In addition, the FIC index ranges of SMT against S. Typhimurium and A. pleuropneumoniae were 0.19-0.38 and 0.19-0.62, respectively. In conclusion, SMT has a high antibacterial activity against S. Typhimurium and A. pleuropneumoniae, then the combination may be applied for the treatment of swine diseases caused by S. Typhimurium and A. pleuropneumoniae.

        To investigate the effects of antioxidant activities and hair loss prevention of extracts from Platycodon grandiflorum, we’ve prepared chloroform (CF) and ethylacetate fractions (EA) extracted from P. grandiflorum. In the results of DPPH radical scavenging assay, the two fractions showed dose-dependent antioxidant activities. Furthermore, in the ABTS assay, the two fractions exhibited the inhibitory effect over 90% at 10, 50, 100, 200 mg/mL. To investigate the inflammation inhibitory effect, we used RAW264.7 cells, these extracts were inhibited inflammatory reaction by suppressing the production of nitric oxide (NO) and prostaglandin E2 (PGE2) in dose-dependent manner. In the assay of HaCaT cells’ proliferation, the 24 hr treatment of the extracts significantly accelerarted cell proliferation in the range of concentrations used. The two fractions inhibited the proliferation of Malssezia furfur, the bacteria induce the dandruff. Finally, the CF could significantly inhibit the DHT production at 1, 10, 50, 100 μg/mL, but EA showed the inhibitory effect at the concentration over 50 μg/mL. The overall results of this study suggest that the chloroform (CF) and ethylacetate fractions (EA) from P. grandiflorum could be a useful raw material for the hair loss prevention products.

        Lactic acid bacteria (LAB) were isolated and analysed its fermentation ability in triticale powder at different moisture levels. Furthermore, the antibacterial activity of fermented silage extract against cattle pathogens was also studied. The isolated strains were P. pentosaceus (TC48) and L. brevis (TC50) that confirmed based on biochemical and 16srRNA sequences methods. Extract from LAB fermented silage showed higher antibacterial activity (inhibition zone diameters: 18~24.2 mm) against E. coli P. aeruoginosa, S. aureus and E. Fecalis than the non-inoculated silage extract. TC48 and TC52 strains exhibited high tolerance to artificial gastric, duodenal and intestinal fluids. In summary, lactic acid bacteria mediated fermentation of triticale silage extract showed great antibacterial activity with significant probiotic characteristics might be an effective and safe way to provide new strategies for reducing the incidence of pathogenic bacteria associated diseases in animals.

        바이오존의 대기압 플라즈마를 사용한 쥐치포(Stephanolepis cirrhifer) 중 미생물안전관리 대상인 B. cereus F4810/72 와 S. aureus ATCC 6538의 저감화 정도 및 품질 특성을 관찰하였다. 대기중의 공기를 활용하여 플라즈마 발생을 유도하여 0 분, 1 분, 3 분, 5 분, 10 분 및 20 분 처리하였다. 이후 대상 미생물의 살균력 평가를 위해 표준평판 법을 사용하여 로그 감소값을 계산하고 Color difference meter를 사용하여 Hunter “L”(명도), ”a”(적색도) 및 “b”(황색도)의 차이를 분석했다. 대기압 플라즈마 3분 처리만으로도 이 두 세균을 1 로그 감소(=90% 감소) 시킬 수 있음이 확인되었다. 아울러 최대 20 분 처리시 쥐치포의 명도, 적색도 및 황색도의 차이는 관찰되지 않았다. 그러나, 본 연구에 사용된 대기압 플라즈마의 기타 수산 건어포와 일반 식품에 대한 산업적 적용을 위해서는 추가적인 목적 미생물의 살균력 연구와 각 품질의 특성을 고려한 품질평가 및 관능평가 등의 추가적인 연구의 확대가 필요하다고 사료된다.

        Perennial need for bactericides requires cost-effective nanomaterials with strong antibacterial activities even in the absence of external irradiation. Hence, in this work, we have synthesized f-CFT (fluorinated TiO2- doped, glycine-functionalized MWCNTs) and have studied its antibacterial activities. Both gram-negative and gram-positive bacteria were analyzed in the absence of photo-activation. Zone of inhibition of 15 mm for S. aureus, and 11 mm for P. aeruginosa was observed for f-CFT owing to the synergistic bactericidal properties of functionalized MWCNTs, fluorine and nano-TiO2. Anatase phase TiO2 with average crystallite size as 35 nm was observed in XRD. Scanning electron microscopic images showed uniform mixed cubic structures. Hydroxyl groups observed in FT-IR along with the glycine MWCNTs interface aid the inhibition of bacterial growth even in the absence of photo activation. A desired higher gram-positive bacterial inhibition opens new gates for better antibacterial agents.

        β-mannanase from Trichoderma reesei was purified by DEAE Sephadex A-50 ion exchange chromatography and Sephadex G-100 gel chromatography. The molecular weight was determined to be 54 kDa by tricine SDS-PAGE. Trigonella foenum-graecum galactomannan was hydrolyzed by the purified β-mannanase, and then the hydrolysates were separated by Bio-Gel P2 chromatography. The main hydrolysates were composed of D.P. (degree of polymerization) 2, 3, and 4, 6 galactomanno-oligosaccharides. To investigate the effects of Trigonella foenum-graecum galactomanno- oligosaccharides on in vitro growth of L. plantarum, were cultivated individually on a modified-MRS medium containing carbon sources such as low- and high-molecular-weight galactomanno-oligosaccharide. Lactobacillus plantarum grew 1.8-fold after treatment with high- and low-molecular-weight galactomanno-oligosaccharides, compared to 1.3-fold for those with standard MRS medium. Bacteriocin was purified by Sephadex G-100 gel chromatography and determined to be 122 kDa by tricine SDS-PAGE. The bacteriocin activated doubly more effectively after treatment with galactosmanno-oligosaccharides compared to those with standard MRS medium. Bacteriocin showed antimicrobial activity against Staphylococcus aureus. The inhibitory compound lost activity when heated to temperatures greater than 30oC and when inhibited to pH changes that lowered the pH below 4 or raised it above 5. Furthermore, its effects were inhibited by treatment with proteolytic enzymes.

        Insect peptides have been extensively studied due to beneficial effects in the treatment of infectious diseases. Melittin, a fundamental component of honeybee venom produced by European honeybee Apis mellifera, has applied to prevent various inflammatory disease and bacterial infections in human. However, the therapeutic application of melittin is limited due to its low stability, hemolytic activity and expensive manufacturing costs. In this study, we aimed to discovery unknown peptides from the Apis mellifera and evaluate its antibacterial activity against Escherichia coli KACC 10005. A total 15,853 peptide sequences were diciphered using Illumina HiSeq 2500 next-generation sequencing (NGS) platform and analyzed based on the Apis mellifera official Gene Set Version 3.2 (amel_OGSv3.2) and the Collection of Anti-Microbial Peptides (CAMPR3) database. All the peptide sequences and annotation data sets were combined and sorted by physicochemical features of antimicrobial peptides (AMPs), such as short peptide length <=50, positive charge, isoelectric point (8.0<=pl<=12), and aggregation propensity (in-vitro: <=500, in-vivo: –40<= Na4vSS <=60). Among the screened peptides, four unknown peptide candidates, named AMP1-4, were chemically synthesized and tested for antimicrobial activity in comparison with a reference peptide, melittin. Inhibition of bacterial growth was observed in the AMP4 treated group from 6 hours to 48 hours post-treatment against E. coli. These results suggest that honeybee-derived peptide sequences can be applied as natural resources to acquire novel AMPs and the peptide sequences derived parameters are enough to recognize antibacterial peptides. In addition, the selected novel peptide candidate, AMP4, has antibacterial activity.

        This study is Antimicrobial activity for bacteriocin produced by Lactobacillus acidophilus. Production of lactacin B was pH dependent, with maximum activity detected in broth cultures maintained pH 6. As a result of the antimicrobial activity against p. acnes in the pH culture, the clear zone was the largest at pH 6 and persisted for a long time.

        This research was conducted to elucidate the optimum conditions for the antibacterial activity of konjak jelly using the evolutionary operation-factorial design technique. In the first set of experiments, concentration of a coagulation agent, soaking liquid, and temperature of water were set to 0.4%, 0.6×10-2 N, and 65℃ as a central point, respectively. The highest antibacterial activity was acquired at E21, in which the number of bacteria was 1.25 log cfu/g. Because the code of changes in the main effect was (-), it could be decided that the central point of the first set was not the optimum point. Although antibacterial activity in the second set was improved, the values of the main effect were higher than that of changes in the mean effect. The central point of third set was concentration of coagulation agent 0.8%, concentration of soaking liquid 1.0×10-2 N, and temperature of water 65℃. It was found that the antibacterial activity of central point in the third set was highest among all the tested set. Further, all the necessary conditions were appropriate to reach the optimum condition. The antibacterial activity of the central point in third set was more than 1,000 times higher than that of E11, in first set.
