The purpose of this study was to investigate the effects of extract mixture of C. wilfordii and P. umbrosa (IPLUS-CWPU) on bone growth in 4-week old young male SD rats. To confirm the effect of IPLUS-CWPU, we measured the length of bone growth plate, the ratio of proliferative zone to the length of growth plate and the expression level of insulin-like growth factor, IGF-1. The IPLUS-CWPU treatment shows a significant increase of tibial and femoral growth plate and the ratio of proliferative zone in growth plate. Especially, the length increased by 13.9% and 25.3% in the tibia and femur, respectively, in the high-dose group compared to the normal group. Moreover, the expression of IGF-1 gene in liver was upregulated in IPLUS-CWPU treated groups. These results indicated that IPLUS-CWPU administration could increase the proliferative zone of bone growth plate in early developmental stage by upregulation of IGF-1 gene.

Background : To develop a new cultivation technique which can save the labor and harvest cost in Cynanchum wilfordii. Stable production is possible for soil and container cultivation. Therefore, this test was conducted to evaluate the reuse effect that can reduce the cost of culture soil. Methods and Results : Container standards were used plume tube of diameter 30 ㎝ × height 50 ㎝. The blending ratios of reuse cultivation soil were 60% cocopeat, 10% peatmoss, 6% pearlite, 6% vermiculite, 5% zeolite, and 5% char. To this, 0, 10, 20, and 30% of composts containing 45% of fowl droppings and 5% of cattle dung were added to the culture soil and mixed. In each treatment C. wilfordii seedling were planted, and the overground growing and yield ability were examined. After harvesting, the functional components contents (cynanoneside B, wilfoside C1N, and p-hydroxyacetophenone) of C. wilfordii conventional cultivation and container cultivation (culture soil) were analyzed. As a result, the growth of overgrown crops showed better tendency as the compost content increased. However, the growth and yield of undergrowth increased by 182% (164.7 ㎏/10 a → 282 ㎏/10 a) compared to no addition at 20% mixing ratio. The content of functional ingredients according to cultivation method and skin was not different all in cynanoneside B, wilfoside C1N, and p-hydroxyacetophenone. Therefore, it was a larger tendency that the classification according to whether it was skined (existence and nonexistence) than the cultivation method. In the economic analysis, it was found that when the cultivation soil was reused by adding 20% compost, the profit of 680,000 won/10 a was increased. Conclusion : Soil and container cultivation was effective saving method for harvesting ease of C. wilfordi, harvest yield was increased by 66%. Also, container cultivation was effective the cost cutting of installation dismantlement and redres since no need to move the cultivation area. As the cultivation soil can be reused, it can be expected to maximize the utilization in container cultivation of deep rooting medicinal crops as well as Cynanchum wilfordii.

Cynanchum wilfordii Radix (CWR), Arctium lappa L (ALL), and Dioscorea opposite (DO) have been known to improve blood lipid profile, blood pressure, and inflammation. To find the optimal combination ratio of CWR, ALL, and DO in terms of vascular health improvement, we compared the effects of various combinations on gene expression of Vascular cell adhesion protein 1 (VCAM-1) in human aortic smooth muscle cells (HASMC). VCAM-1 mediates endothelial leukocyte adhesion and is upregulated in atherosclerosis. Cells was stimulated by TNF-α (10 ng/㎖, 2h) and treated with various combinations for 24 h. A combination (CADM5, CWR:ALL:DO = 2:1:1) showed the strongest suppression of VCAM-1 so that CADM5 was chosen for further experiments. We performed cell viability test with CADM5 (0, 3.125, 12.5, 25, 50, and 100 ㎍/㎖) and no cytotoxicity was found. We also investigated the effect of CADM5 on protein expression of VCAM-1, ICAM-1, Nrf-2, and HO-1 using western blotting. We found that CADM5 diminished the expression of VCAM-1 and increased the expression of Nrf-2 and HO-1. Therefore, we concluded that CADM5 (CWR:ALL:DO = 2:1:1) effectively improves vascular health by regulating the expression of VCAM-1.

Background : Cynanchum wilfordii and Cynanchum auriculatum belong to the Asclepiadaceae family and appear morphologically similar. In order to discriminate them, it is needed to find the presence of sap and the leaf shapes: C. auriculatum has a blade ovate leaf comparing to C. wilfordii. However, in the herbal medicine market, they have been handled as cut and dried roots. Due to their similar morphology, it is limited to distinguish the roots of C. wilfordii and C. auriculatum. Recently in Korea, it has been a critical issue to misuse these two roots in the herbal market and food industry. Thus, it is required to establish a robust tool for the discrimination and quality control of them. Methods and Results : To separation and characterization of flavor compounds, C. wilfordii and C. auriculatum samples were analyzed by head space solid phase micro extraction (HSS) fiber coupled with gas chromatography-mass spectrometry using Rtx-5MS (30 m x 0.25 mm x 0.25 μm) column. As a result, We have identified compounds of a few hundred in aliphatic aldehydes and aliphatic alcohols, alkenes and acids, aromatic compounds, aromatic compounds containing nitrogen & sulfur, etc,. In particular, The aliphatic and aromatic compounds had been clearly separated on the second dimensional direction by using two-dimensional GC. Conclusion : The volatile flavor compounds of C. wilfordii and C. auriculatum could easily analyzed without pre-treatment with improved resolution and sensitivity using HSS-GCxGC-TOFMS. We have identified compounds of a few hundred in C. wilfordii and C. auriculatum sample. And It was more accurately qualitative confirmed with separation of GCxGC and TSD. We have confirmed the PCA and PLS-DA Plot that was classified depending on C. wilfordii and C. auriculatum through multivariate statistical analysis of the identified flavor compounds.
