        Helicobacter pylori are known as a causative agent of gastritis, gastric duodenum and peptic ulcer, and gastric cancer, and multiple drug use is associated with various side effects in patients. The discovery of antibacterial substances against H. pylori from Korean resource plants is an important substitute for antibiotics. 52 species of Korean resource plants were collected and extracted with 50% ethanol, and antibacterial activity against H. pylori was measured using the disk diffusion method. The toxicity of plant extracts to human gastric adenocarcinoma(AGS) cells was measured by MTT assay, and the level of IL-8 secreted when gastric epithelial cells were inoculated with H. pylori was measured. As a result of measuring the antibacterial activity of H. pylori, antibacterial activity was confirmed in 38 plant extracts. The plant species with the strongest antibacterial activity were Chrysanthemum indicum, Rheum rhabarbarum, Patrinia scabiosaefolia and Petasites japonicus. C. indicum was not cytotoxic to H. pyroli-infected AGS cells and showed anti-inflammatory effects. This study's results can be used to develop healthy, functional foods and medical materials.

        헤스페리딘(Hesperidin, HD)은 다양한 식물체에 존재하는, 강한 항산화 기능을 가진 대표적인 flavonoid의 일종이다. 본 연구에서는 수용성 HD인 Hesperidin glucoside(HDG)가 가지는 세포손상 회복, 항염증 인자억제 및 melanin 생성억제 활성을 세포수준에서 비교하였다. HDG는 HD에 당전이 효소반응 으로 제조되었으며, HD에 비해 20,000배 이상 수용해도가 증가되었다. HaCaT 세포주에 대한 세포독성은 HDG가 HD에 비해 월등히 낮았다. HD와 HDG는 모두 자외선 조사된 HaCaT 세포에서 세포생존율 회 복효과를 나타내었다. 또한 HD와 HDG는 세포내 산화질소(NO), 종양괴사인자-α(TNF-α) 및 인터루킨 -6(IL-6)과 같은 염증 매개체 및 cytokine을 감소시켰으며, HD 보다는 HDG의 효과가 다소 우수하였다. Melanoma B16F10 세포주를 이용한 melanin 형성능과 tyrosinase 저해활성을 측정한 결과, HD와 HDG 모두 효과를 나타내었으며 HDG가 약간 우수한 결과를 보였다. 결론적으로, HD의 당전이체인 HDG는 HD에 비해 동등이상의 세포손상 회복, 염증성 매개체 및 cytokine 억제능과 melanin 형성억제능을 나타내 었으며, HDG의 높은 수용성과 낮은 세포독성 등의 특성은 다양한 분야에서의 용도를 확대시킬 수 있을 것으로 보인다.

        This study analyzed the chemical characteristics and physiological activity of five kinds of fresh vegetables produced in trees in early spring and tried to use them as basic data for wild vegetable producers and processed food manufacturers using wild vegetables. The crude protein, minerals, ascorbic acid, folate, total phenol, total flavonoid, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and ACE (angiotensin converting enzyme) inhibitory activity were determined. Five spring wild vegetables contain high protein and phosphorus, indicating that they are useful food ingredients as sources of protein and phosphorus. Vitamin C content was high in R. venicifera and C. sinensis shoots, and in particular, R. venicifera and A. cortex shoots have high folic acid (folate) contents of 1,903.91 ug% and 1,525.35 ug%, respectively, which is considered a good food for folic acid intake in spring. The total phenol content was between 0.52% and 1.27%, and it was the highest in C. sinensis of 1.27%, followed by the order of R. venicifera, A.cortex, K.pictus, and A. elata, which tended to be consistent with the total flavonoid content. As for DPPH radical scavenging ability, C. sinensis (55.93%) showed the highest activity, and ACE inhibitory activity showed the highest activity in A. cortex (88.04%).

        Steaming is a method that has traditionally been used for medicinal plant extraction. This study investigated nitrite oxide production, ferrous ion chelating activity, α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities of ethanol, acetone and hot-water extracts of Codonopsis lanceolata prepared by steaming seven times. MTT assay showed that each extract was non-toxic up to a concentration of 700 μg/mL confirming that there was no cytotoxicity in all extracts. The α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities exhibited by the hot-water extract obtained from steaming seven times were higher (83.1%) than the other extracts. Higher production of nitrite oxide and better ferrous chelating activity was recorded with hot-water extract compared to ethanol and acetone extracts. These results indicated that more steaming of Codonopsis lanceolata extracts would be required to validate the possibility of developing antioxidants. Also, further study is needed to determine if the components present in the tested extracts might be useful in the prevention of Alzheimer's disease. These results showed that hot-water extracts may be useful for their antioxidant and the production inhibitory activity of nitrite oxide. It will be helpful in the investigation of the constituent analysis of the steam-processed product of Codonopsis lanceolata.

        In this study, polyphenol and flavonoid contents were measured, and DPPH, OH, H2O2 radical scavenging activity, and the α-amylase inhibitory activity were measured to study the antioxidant activity of 70% ethanol extract from Morinda citrifolia L. The polyphenol and flavonoid contents of noni 70% ethanol extract were 29.52 GAE/g and 12.48 CE/g, respectively. Also, the IC50 values of DPPH, hydroxyl radical, and hydrogen peroxide scavenging activity of 70% ethanol extract from noni were 18.70 mg/mL, 26.45 mg/mL, and 35.67 mg/mL, respectively. Measurement of the α-amylase inhibitory activity of 70% ethanol extract from noni showed 45% inhibitory activity at 10 mg/mL.

        The purpose of this study was to investigate the antioxidant activity and tyrosinase inhibitory activity of Codonopsis lanceolata 50% ethanol extract, and its solvent fractions (n-hexane, ethyl acetate (EA), n-butanol, water). The main components of the EA fraction were qualitatively analyzed using UPLC Q-ToF/MS. Additionally, a quantitative analysis was performed using UPLC. As a result, the total polyphenol content was 113.36 mg gallic acid/g in the EA fraction, which contained the largest amount of the C. lancolata solvent fractions. Also EA showed the highest antioxidant activity than other fractions. The IC50 of DPPH(2,2-diphenyl-1- picrylhydrazyl) radical scavenging activity was 0.03 mg/mL and the IC50 of ABTS [2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonate)] radical scavenging activity was 0.049 mg/mL. The EA fraction showed tyrosinase inhibitory activity than other fractions and especially inhibited monophenolase oxidase reaction higher than diphenolase oxidase reaction. The monophenolase oxidase inhibited 55% when the concentration of the EA fraction was 0.25 mg/mL. As a result of Q-ToF/MS analysis, it was confirmed that tangshenoside I and lobetyolin were the main components of EA fraction. Thus, these results suggest that C. lanceolata may be used as a potent source of cosmetic agents.

        The objective of this study was to evaluate the effect of fermented Kalopanax pictus (KP-F) on macrophage activation and its effect as a competitive inhibitor of LPS and inhibitory effect on endotoxemia. The results showed that KP-F could activate macrophage in a dose-dependent manner, and KP-F was confirmed to act as a ligand for TLR4. Also, it was found that KP-F did not exhibit the same biotoxicity as LPS in intraperitoneal injection, and that it could suppress the neutrophil migration induced by LPS administration. In normal mice, the body weight, tissue weight, and amount of nitrite and pro-inflammatory cytokines in serum showed no significant changes with KP-F diet for 2 weeks, confirming that administration of KP-F in normal mice did not lead to over activation of immune response and biotoxicity. In the mouse model of endotoxemia induced by LPS and D-galactosamine(D-GalN) in sub-lethal dose, the diet of KP-F effectively inhibited the amount of nitrite and cytokines in the blood, and thus was found to be able to relieve the hepatic and kidney injury. In addition, in the endotoxemia mouse model induced by LPS and D-GalN of lethal dose, the survival rate was increased by KP-F diet in a dose-dependent manner.

        본 연구에서는 기능성 식의약품 및 화장품 소재로써 흰색 느티만가닥버섯(Hypsizygus marmoreus)의 이용 가능성을 조사하기 위해서 흰색 느티만가닥버섯 열수 추출물과 메탄올 추출물의 항산화 활성 및 tyrosinase 저해 효과를 비교하였다. 열수 추출물과 메탄올 추출물의 총 폴리페놀 함량은 각각 8.4±3.27 mg GAE/g과 7.3±2.85 mg GAE/g이었고 플라보노이드 함량은 각각 4.8±3.81 ug/mg과 2.5±1.95 ug/mg이었으며 총 폴리페놀과 플라보노이드 함량 모두 메탄올 추출물보다 열수 추출물에서 높게 나타났다. Tyrosinase 저해 활성은 추출물의 농도에 따라 증가하는 경향을 나타내었으나 양성 대조구로 사용한 2% 알부틴 (arbutin)비해 40 mg/ml의 높은 농도에서도 열수 추출물은 69.72%, 메탄올 추출물은 52.67%의 낮은 저해 활성을 나타내었다. 항산화 활성은 DPPH에 의한 라디칼소거 활성을 측정하여 확인하였으며 열수 추출물과 메탄올 추출물의 DPPH 라디칼 소거능은 40 mg/ml의 농도에서도 각 각 80%와 74%로 낮게 나타났다. 추출물의 세포독성은 WST-1 assay (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H- 5-tetrazolio]-1,3-benzene disulphonate)를 이용하여 추출물의 처리농도에 따른 B16BL6 melanoma cell의 세포생존율로 확인하였으며 열수 추출물과 메탄올 추출물을 각각 0-40 mg/ml의 농도로 처리하였을 때 B16BL6 melanoma cell은 90% 이상의 생존율을 나타내었으므로 열수 추출물과 메탄올 추출을 모두 B16BL6 melanoma cell에 독성을 나타내지 않는 것으로 판단된다.

        최근 소득이 증가하고 건강에 관심이 증가함에 따라 단순한 미용 화장품보다 여러 가지 기능성을 가진 화장품에 대한 관심이 높아지고 있다. 이에 따라 각종 천연물에 대하여 항산화, 항염 및 미백 등의 활성을 검증하는 다양한 연구가 진행되고 있으며, 본 연구에서도 천연물의 활성과 더불어 다양한 약리작용이 있는 영지버섯 균사체와의 발효에 의한 시너지 효과를 규명하고자 하였다. 인진쑥 영지균사발효 추출물의 DPPH radical 소거 활성은 영지버섯 균사추출물과 유사하게 농도 의존적으로 증가하는 것이 확인되었으나, 저농도의 인진쑥 추출물 보다는 다소 감소하는 것이 확인되었다. 인진쑥 영지균사발효 추출물의 RAW 264.7 세포의 NO 생성 억제활성을 통한 항염활성 평가에 앞서 세포 생존율을 평가한 결과, 흥미롭게도 인진쑥 추출물은 150 μg/mL의 농도부터 RAW 264.7 세포에 대한 생존율을 유의적으로 크게 감소시켰으나, 영지 균사 발효 시 세포 생존율이 크게 증가하는 것이 확인되었다. 영지 균사 발효 시 인진쑥의 독성이 크게 감소하는 것이 확인되었다. 또한 인진쑥 추출물 25 μg/mL의 처리는 동일 농도의 인진쑥 영지버섯 균사발효 추출보다 오히려 유의적인 수준에서 RAW 264.7 세포의 NO생성을 억제하는 것이 확인되었다. 그러나 인진쑥 영지버섯 균사발효는 인진쑥 추출물 자체의 RAW 264.7 세포에 대한 독성을 저감하는 효과가 있어 이로 인해 더욱 높은 농도의 영지버섯 균사발효 추출물 처리를 가능할 것으로 사료된다. 영지 균사 발효에 의한 미백 기능성 소재 발굴이라는 본 연구의 취지에 부합하는 결과가 인진쑥 영지균사발효 추출물 처리구에서 확인되었으며, 200 μg/mL 농도에서는 양성 대조군인 arbutin 보다 약 3.66배의 활성을 보였다. 이러한 결과는 향후 인진쑥 영지균사발효 추출물이 tyrosinase활성을 억제하는 기능성 소재로서의 활용가능성이 크다는 것을 의미하는 것이다.

        Tenebrio molitor beetles have been widely used in traditional and folklore medicines to treat various human diseases worldwide. Alzheimer’s disease (AD) is a neurodegenerative disorder and the most prevalent form of dementia in developed and developing countries. The human β-amyloid cleaving enzyme (BACE-1) is a target for AD treatments. This study was performed to determine BACE-1 inhibitory activity of 12 compounds that were extracted from T. molitor adults and their 65 related compounds using a fluorescence resonance energy transfer-based enzyme assay. The results were compared with those of two positive controls for BACE-1, the cell-permeable isophthalamide BACE-1 inhibitor IV and the natural BACE-1 inhibitor, epigallocatechin gallate (EGCG). Based on IC50 values, linoleic acid, ergosterol, palmitoleic acid, and tryptopol (28.41‒46.16 μM) were the most potent BACE-1 inhibitors and the anti-BACE-1 activity of these compounds was lower than either inhibitor IV (13.13 μM) or EGCG (1.12 μM). Multiple regression analysis of the anti-BACE-1 activities of compounds was examined using their IC50 values and the values of the physical parameters (molecular weight (MW), log P, and molecular refraction (MR)) for the 41 fatty acids (R2 = 0.674 (P = 0.044)) and 29 flavonoids (R2 = 0.587 (P = 0.063)). Correlation coefficient (r) analysis showed that MW, log P, and MR may be positively correlated with IC50 of the fatty acids (MW, r = 0.658; log P, r = 0.471; and MR, r = 0.713) and IC50 of the flavonoids (MW, r = 0.547; log P, r = 0.644; and MR, r = 0.591). Further studies will warrant possible applications of T. molitor adults as therapeutic BACE-1 blocker.

        본 연구는 적작약 꽃 추출물의 활성산소 억제와 항염증 및 MMP-1 발현 억제능 효과에 관해 알아보고 기능성 화장품 소재로써의 가능성을 확인하고자 하였다. 본 실험 방법으로는 세포 내 ROS 측정을 통한 활성산소 억제효과와 세포 독성 평가 및 항염증 측정, HDF 세포에서의 MMP-1의 발현 억제능 효과를 측정하고자 하였다. 실험 결과, RAW 264.7 세포와 HDF 세포 내에서 ROS로 억제효과를 확인하였고, 세포 독성평가는 적작약 꽃 추출물 5, 10 μg/mL 처리 농도에서 90% 이상의 세포 생존율, 그 외 처리 농도에서는 80% 이상의 세포 생존율을 확인하였다. 또한 RAW 264.7 세포에서의 NO 생성 억제와 HDF 세포에서 MMP-1의 발현 억제능이 유의하게 억제되는 것을 확인하였다. 이상의 결과를 종합하면, 적작약 꽃 추출물의 세포 내 ROS 생성억제와 NO 생성 억제로 항산화와 항염증 효과, 피부세포에 대한 낮은 독성, MMP-1의 발현 억제를 통한 노화 효과가 확인됨에 따라 기능성 화장품 소재로써의 활용 가능성을 확인할 수 있었다.

        Ramie (Boehmeria nivea M.) has been used for fiber materials in Korea traditionally, but in recent years, the concern with ramie leaves for the food industry such as tteok (a kind of Korean rice cake) industry has been increasing, so a study for eatable ramie is required for the expansion of ramie consumption. Moreover, the ramie varieties for the food industry are not established, so the natural species are cultivated in general; therefore, it is very important to select the ramie varieties for the food industry such as rice cakes, tea, beverage and so on. This study was undertaken to compare the physiochemical properties among 9 ramie lines selected in the Yeonggwang-gun Agricultural Technology Center to select the eatable ramie varieties for the food industry. The contents of the protein among 9 ramie lines was 6.21~7.56% and had the highest content in the YG55. The folic acid (folate) and vitamin C content had varying differences among the 9 lines; the content of folate showed 771.52~1,978.84 μg%, that of vitamin C showed 149.42~275.34 mg%. The ACE inhibitory activity appeared to be the highest in YG88 (21.5%) among the 9 ramie varieties tested.

        This study was performed to determine the optimal time of harvest for ramie leaves with the two varieties (Seocheon Seobang and Seoncheon Baekpi) by comparison of physiological activity and physicochemical characteristics. The crude protein, minerals, ascorbic acid, folate, chlorophyll, ACE inhibitory activity and AChE inhibitory activity were determined. The amount of crude protein in ramie leaf, which was collected in Seocheon-gun, Chungcheongnam-do, grew up steadily from early May to September. The content of calcium in was higher in Baekpi than in Seobang. Seobang displayed its highest value of 3,569.90 mg% in September, while Baekpi displayed its highest value of 3,163.84 mg% in October. Although, folate and vitamin C contents in the two varieties were slightly different, they were higher as the growth date grew in October. The highest value of chlorophyll content was observed in October, which was later in the vegetative state. ACE inhibitory activity and AChE inhibitory activity appeared to be higher in Baekpi than in Seobang. Between June and August, ACE inhibitory activity was highest in Baekpi variety.
