        뿌리썩음증상은 병징에 따라 3가지로 대별되었다. 뿌리썩음증상은 Pythium aphanidermatum을 선접종 후 Fusarium solani를 후접종 하였을 때 가장 심하였다. 뿌리혹선충과 Phthium aphanidermatum과의 상호작용은 없었다. 근경 발아는 각 처리에서 현저히 증가하였고 Alliette 0.25% 처리에서 가장 높았다. 토양관주에 의한 치료 및 예방효과는 Alliette, Burgandy mixture, 다이센 M-45, 디포라탄이 우수하였으며, 수량도 증가하였다.

        강원도 고냉지 배추재배 지역에서 53개의 이병 뿌리와 22개 토양 표본을 채취하여 배추 생육 초기에 뿌리 썩음을 일으키는 병원균을 조사하였다. 이병된 뿌리에서 검출된 13개 곰팡이 중에서 Pythium ultimum, Pythium echinocarpum, Rhizoctonia solani의 3종이 자주 검출 되었으며 배추에 병원성이 있었다. 채취한 토양에 본엽 엽기의 배추를 이식하여 발명 유무를 온실에서 조사 하였을 때 토양 표본에 따라 5내지 100%의 식물이 이들 균에 의하여 감염 되었다. 토양내 이들 3종의 균의 밀도를 배추 식물의 감염 빈도에 의하여 조사 하였을 때 차항 2리 지역의 토양에서는 P. ultimum이 우점종 이었고 P. echinocarpum은 용산 2리, 횡계리, 문맥 지역에서 주로 밀도가 높았다. 매봉산 지역의 토양에서는 R. solani 균만이 단지 분포하고 있었으며 P. ultimum 과 P. echinocarpum이 동시 분포하는 지역에서는 대체로 P. echinocarpum 균의 밀도가 P. ultimum에 비하여 더 높았다. 주요 30품종의 유묘를 가지고 온실에서 품종 저항성을 조사한 결과 6개 품종이 P. ultimum 에 대하여 저항성이었고 3개 품종이 중도 저항성 이었다. P. echinocarpum에 대해서는 저항성 품종이 없고 2개 품종이 중도 저항성 이었으며 R. solani균에 대해서는 모두가 이병성 이었다.

        두 종류의 부리혹선충(M. incognita, M. hapla) 각각에 대한 우리나라 콩 장려품종들의 저항성을 검정한 결과 1. 인코그니타혹선충(M. incognita)에는 은대두, 백천, 동북대, 단엽콩이 저항성 이었으며 광두, 황금총, 부석은 중정도 저항성이었고 광교외 8품종은 감수성 이었다. 2. 당근혹선충(M. hapla)에는 장엽콩 한 품종만이 저항성이었으며 단엽콩, 힐, 동북태, 함안대립, 충북백은 중정도 저항성을 나타내었고 육우 3호 외 9품종은 감수성이었다. 3. 두 종류의 뿌리혹선충에 모두 저항성을 나타내는 품종은 없었으나 단엽콩과 둥북태는 인코니타혹선충(M. incognita)에는 저항성을 가지면서 당근혹선충(M. hapla)에는 중정도 저항성을 나타내는 좋은 품종이었다.

        The infection rate of soybean black root rot disease caused by Calonectria crotalariae was about . The isolated fungi from the infected soybean roots and stems were Calonectria crotalariae, Fusarium solani, F. roseum, Phomopsis sojae, Pythium aphanidermatum, Rhizoctonia solani and Macrophomina sp. Among them, C. crotalariae was the most virulent pathogen under the laboratory conditions. Mycelial growth and microsclerotial formation were good on PSA containing 1000cc of water, 100g of potato and 20g of sugar. Mycelial growth, sporulation and microsclerotial formation were good on sterilized root. Perithecial formation was better in the dark condition than in the light. Survival of macroconidia was not available between soil water content. Microsclerotia and mycelium in infected plant debris were survived for 4 months at to soil water content. The plant height, when inoculated with inoculum density, reached approximately half of uninoculated plants. Disease severity was much higher at nonsterilized soil than completely sterilized soil. It was determined that the host range of this pathogen includes soybean, peanut, green bean and red bean.

        경제작물증산에 크게 영향을 미치는 뿌리혹선충에 간하여 1977년 경상남, 북도 경기도 제주도 등의 4개도 46개 지역에서 66개의 재료를 채집하여 조사한 결과 뿌리혹선충 종두는 Meloidogyne hapla. Meloidogyne arenaria, Meloidgyne incognita 그리고 Meloidogyne javanica등 4종이 발견되었으며 동정된 4종의 뿌리혹선충의 중요 형태적 특징을 기술했다. 이 중에 Meloidogyne javanica는 우리나라 미기록종으로 경기도 수원시 이목동 원예시험장의 감자, 경북 대구시 검사 동의 배추, 대구시 산격동의 오랑캐 꽃, 경남 남해군 초음리의 호박 등에서 발견되었다. 지역별 뿌리혹선충 종류별 분포상태는 제주도에서는 전부 Meloidogyne incognita만 발견되었고 다름 3종은 발견도지 않았다. 그러나 경기도에서는 Meloidogyne incognita는 전연발견되지 않았다. 경상북도와 경기도에서는 Meloidogyne hapla가 가장 많이 발견되었으며 경상남도에서는 4종 모두 발견되었으나 Meloidogyne hapla와 Meloidogyne incognita가 비슷하게 분포하고 있었다. 전반적으로 우리나라에서 가장 빈번히 광범하게 분포하고 있는 종은 Meloidogyne hapla로 전체 조사수의 를 차지하고 있으며 다음이 Meloidogyne incognita로 , Meloidogyne arearia , Meloidogyne javanica 의 순으로 나타났다. 지역별 피해상태는 제주도가 가장심하게 나타났고 다음이 경남, 경북, 경기 등의 순이었다. 뿌리혹선충의 피해를 받고 있는 직물은 24종이 발견되었으며, 그 중에 특히 중요한 농작물로는 콩, 땅콩, 감자, 토마토, 오이, 당근, 호박, 수박, 우엉, 고추, 가지, 배추, 상치, 담배, 인삼 등이었다. 전반적으로 뿌리혹선충방제를 위한 지도 계몽이 시급히 요구된다.

        벼뿌리선충의 벼뿌리갈변에 미치는 영향을 알아보기 위하여 벼가 자라는 배지조건을 달리하여 실내시험한 결과, 살균배지조건에서는 선충에 의하여 갈변도가 약간 높아졌으나 무살균조건에서는 선충의 존재여부에 관계없이 뿌리전체가 갈변되었는데 이것은 벼뿌리의갈변이 선충에 의한 다기보다 주로 그외의 토양미생물에 의하여 일어나는 것으로 보인다.

        인삼뿌리썩음병균, Cylindrocarpon destructans(Zins.) Scholten을 접종한 인삼절편으로 부터 얻은 섬유소분해효소 및 펙틴질분해광소의 역가는 그 농도, 시간에 비례하였다. Cellulase (Cx), endo-polygalacturonase(endo-PG), endo-poiymethylgalacturonase (endo-PMG), exo-polygalacturonase (exe-PG)와 exo-polymeth-ylgalacturonase(exo-PMG)의 역가는 접종후 4 일째 최대였으며 endo-PG와 endo-PMG는 1일 및 2일째는 전혀 검출되지 않았으나 exo-PG와 exo-PMG는 검출되었다. 접종후 6 일째에는 모든 펙틴질분해효소는 활성을 완전히 잃었으나 섬유소분해효소는 높은 역가을 유지하였다. Cx, endo-PG, endo-PMG, exo-PG 및 exo-PMG의 최적 pH는 각각 6.0, 5.5, 8.0, 7.0-7.5, 8.5이였다. Cx, endo-PG, endo-PMG, 및 exo-PG의 최적온도는 각각 exo-PMG 였다. 섬유소분해효소는 공시한 16개 이온중에서 0.05M 만이 억제하였다. 펙틴질분해효소는 이온에 따라 상이하였으나 과 이 가장 현저히 억제하였다. 공시한 8개 농약중에서 어느 것도 유효성분으로는 모든 효소작용을 억제하지 못했으며 exo-PG만은 모든 농약의 의하여 상당히 억제되었다. 그 중에서 Difolatan은 모든 펙틴질분해효소를 가장 잘 억제시켰다. 과 은 펙틴질분해효소를 거의 억제시켰으나 섬유소분해효소는 같은 농도에서 각각 에 이르렀다. Formalin은 exo-PG와 endo-PMG를 완전 억제시켰으나 다른 효소 특히 Cx는 그렇지 못했다. Difolatan은 모든 효소를 이상 억제하였으나 Cx는 그 이하였다. C. destructans가 분필하는 섬유소분해효소 및 펙틴질분해효소는 인삼뿌리썩음병과 밀접한 관계를 가지고 있으며 이 병을 효과적으로 방제하기 위하여 억제되어야 하겠다.

        초 록 1. 인삼묘포에 Captan, Difolatan, Zineb, Maneb와 PCNB를 일주일 간격으로 토양관주 하였다. 처리하기전과 2회 및 4회처리후에 각각 Rhizoctonia solani, Pythium debaryanum, Fusarium 그리고 Trichoderma 균총의 숫적 (수적)변화를 Boosalis의 특별잔사법에 의해서 조사하였다. 우리나라에서 인삼(Panax ginseng)의 모잘록병균으로서 Pythium debaryanum을 처음으로 기술하였다. 2. Rhizoctonia solani, Pythium debaryanum의 수는 토양관주에 관계없이 시일이 경과함에 따라서 점차로 감소하였고 Fusarium 및 Trichoderma는 오히려 증가하였다. Rhizoctonia solani의 수는 PCNB에 의해서 현저하게 감소하였고 다른 약제에 의해서도 대조구 보다 감소하였다. 3, Pythium debaryenum은 Zineb, Maneb, Captan, Difolatan의 순으로 고 수가 증가하는 반면 Fusarium은 일정한 경향을 볼 수 없었다. Tichroderma를 제외한 모든 균의 숫적 변화는 시일의 경과에 대하여 수준에서 유의성이 있었다. 4. 토양관주에 의한 약해는 Maneb, Zineb, Captan 구에서 나타났고 묘삼 뿌리의 생체 무게는 Difolatan, PCNB, Maneb구에서 대조구 보다 증가하였다.

        Ginseng (Pnanx ginseng C. A. Meyer) is famous worldwide, and is very important cash crop and medicinal herb in Korea. It takes four to five years to produce harvestable ginseng roots, and ginseng is attacked by several pathogens during cultivation. We investigated the disease rate caused by ginseng root rot from 6 years old ginseng cultivation fields (Chungnam; 9 fields, Chungbuk; 11 fields, Gangwon 5 fields). The highest disease severity was Dangjin D (2.9) and the lowest one was Gaesan C (0.6). Of the 625 isolations, 340 isolations were classified as Ilyonectria radicicola and Fusarium solani. Finally, genetic diversity of I. radicicola and F. solani was confirmed by sequence analysis. Among the I. radicicola group, I. mors-panacis, which is known as highly virulent pathogen, and I. liriodendri, I. robusta and I. cyclamicicola, which are weakly virulent pathogens, were identified. In the case of F. solani, it is divided into two groups, but it is necessary to conduct diversity research through genetic analysis and pathogenetic studies using various markers. Based on these results, it could be used as a basic data for control of ginseng root rot pathogens.

        Angelica gigas Nakai (AGN) is a traditional medicinal herb especially in Korea. It contains pyranocoumarins, which are major active components including decursin (D) and decursinol angelate (DA). This study was carried out to determine the change in active component content and antioxidant activity depending on the root diameter of AGN. Several processing steps are involved to use AGN roots as medicine. The dried AGN roots are divided into body (B), thick root (TkR), medium root (MR) and thin root (TnR) according to their diameter before cutting into medicine. The recovery rates of each root parts per 100 ㎏ were measured as 32.3±2.5, 9.0±1.0, 39.3±2.1 and 15.0±1.0%, respectively and the mean diameters were measured as 51.95±4.55, 7.05±0.89, 2.88±0.49 and 1.57±0.32 ㎜. Two index components, D and DA, were analyzed. The change of both D and DA content showed a similar tendency. Both D and DA content were increased as the root diameter decreased (higher in TnR). In addition, antioxidant activity was higher in B and TnR, and lower in TkR and MR than control. This study showed that the thinner the root diameter, the higher the D and DA content in AGN roots and that TnR has excellent antioxidant activity compared to other root parts, suggesting that the thinner root part of AGN could be used as a potential material.

        Background: Taraxacum platycarpum has been used in traditional medicine in Korea to treat intoxication and edema and as a diuretic. According to previous reports, it has anti-cancer, anti-gastritis, and anti-inflammation effects. However, the improvement effect of T. platycarpum on rheumatoid arthritis has not been investigated. The anti-oxidative and anti-inflammation effects of the aerial parts of T. platycarpum are different from those of its subterranean parts. Thus, we evaluated the effect of the water extracts of Taraxaci radix (WTR) on type II collagen-induced rheumatoid arthritis (CIA) in animal models. Methods and Results: Rheumatoid arthritis was induced by type II collagen. WTR (100㎎/㎏ and 500㎎/㎏) was administered to the animal models. Methotrexate was used as the positive control. The levels of interleukin-6, TNF-alpha, and type II collagen IgG in the animals were measured by using enzyme-linked immunosorbent assay. Treatment with 500㎎/㎏ WTR decreased the serum levels of interleukin-6, TNF-alpha, and collagen IgG in the CIA models. Moreover, treatment with WTR diminished the arthritisinduced swelling of the hind legs and monocyte infiltration in the bloodvessels of the animal models. Conclusions: These results indicate that WTR has the potential to improve rheumatoid arthritis by reducing the levels of inflammatory cytokines such as interleukin-6 and TNF-alpha. However, further experiments are required to elucidate the influence of WTR on signal transduction in vitro and in vivo.

        This study was conducted to evaluate resistance of 10 different cowpea cultivars which were introduced from the United States against two major root-knot nematodes, Meloidogyne arenaria and M. incognita, that affect greenhouse crop cultivation in Korea. The results showed that the resistance of the tested cultivars to the root-knot nematodes varied with the cultivar. Texas Cream 40 showed moderate resistance to M. arenaria whereas the other cultivars were susceptible. Purple Hull Pinkeye and Texas Pinkeye Purple Hull were found to be resistant to M. incognita, whereas Mississippi Silver showed moderate resistance and the other cultivars were susceptible. As the cultivars exhibit resistance to M. arenaria and M. incognita, such cultivars resistant to root-knot nematodes should be considered during the cultivation of cowpea as a green manure crop.

        Background: Atractylodes radix is a well-known medicinal crop having many physiological effects. This study was conducted to select useful Atractylodes japonica × Atractylodes macrocephala (AJM) cultivars by comparing anti-oxidative and anti-inflammatory efficacies. Methods and Results: Seven extracts from AJM cultivars were used to treat lipopolysacchride (LPS)-treated BV2 cells, and the effects on cell viability and inhibition on reactive oxygen species (ROS) and nitric oxide (NO) production were analyzed. In vitro scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and peroxynitrite (NOO−) radicals were also investigated. Contents of total phenol, atractylenolide I, and atractylenolide III in the AJM extracts were measured using high performance liquid chromatography (HPLC) or spectrophotometry. The experiments show that none of the seven extracts was cytotoxic above 89.2% at 20 - 250㎍/㎖. Extracts of Gowon, Dawon, Sangchul, and Huchul inhibited ROS generation in a dose-dependent manner, and Sangchul extract showed the highest inhibition on ROS production. All the AJM extracts showed effective inhibitory activity after on NO release in the LPS-treated BV2 cells, and Sangchul extract showed the highest activity. Sangchul extract had the most potent scavenging activities for NOO− and had some DPPH radical scavenging effect. Sangchul extract also had the highest content at total phenol and atractylenolide I content. Atractylenolide III was not detected in the AJM extracts. Conclusions: The results suggested that Sangchul was the most useful anti-oxidative and anti-inflammatory resource among the AJM cultivars.

        Background: Some phenolics detected in the soil may inhibit the seed germination and seedling growth of ginseng (Panax ginseng). This study investigated the effect of irrigation and ginseng root residue addition on the soil microbial community and root rot disease in 2-year-old ginseng. Methods and Results: Each 20 ℓ pot was filled with soil infected with ginseng root rot pathogens, and irrigated daily with 2 ℓ of water for one month. After the irrigation treatment, ginseng fine root powder was mixed with the irrigated soil at a rate of 20 g per pot. In descending order, NO3 −, electric conductivity (EC), exchangeable Na (Ex. Na) and K (Ex. K) decreased due to irrigation. In descending order, NO3 −, EC, Ex. K, and available P2O5 increased with the additon of ginseng powder to the soil. The abundance of Trichoderma crassum decreased with irrigation, but increased again with the incorporation of ginseng powder. The abundance of Haematonectria haematococca increased with irrigation, but decreased with the incorporation of ginseng powder. The abundance of Cylindrocarpon spp. and Fusarium spp., which cause ginseng root rot, increased with the incorporation of ginseng powder. The abundance of Arthrobacter oryzae and Streptomyces lavendulae increased with irrigation. The abundance of Streptomyces lavendulae decreased, and that of Arthrobacter spp. increased, with the incorporation of ginseng powder. Aerial growth of ginseng was promoted by irrigation, and ginseng root rot increased with the incorporation of ginseng powder. Conclusions: Ginseng root residues in the soil affected soil nutrients and microorganisms, and promoted ginseng root rot, but did not affect the aerial growth of ginseng.

        Background : The causative organism of Astragalus membranaceus (Hwanggi) root rot disease was known as Pythophthora drechsleri. In this study, a new species of root rot causative organism has been identified and reported. Methods and Results : Hwanggi, which has root rot disease, was collected in Jungsun-gun, Gangwon-do. The diseased root was cut into 1 ㎝ size and disinfected with 1% sodium hypochlorite solution for 1 minute and washed with sterile distilled water. The dried samples were dipped in the PDA medium and cultured in at 25℃ incubator. The 21 isolates were cultured under the same conditions and genomic DNA was extracted. The genomic DNA of isolated strains was extracted using a kit of Biofact. The isolated strains were identified using internal transcribed spacer (ITS) region sequencing. The ITS region was amplied using the primer pair ITS1 (5’-TCCGTAGGTGAACCTGCGG-3’) and ITS4 (5’-TCCTCCGCTTATTGATATGC-3’). The PCR was performed in a final volume of 50 ㎕ containing 10 mM Tris-HCl (pH 8.3), 50 mM KCl, 1.5 mM MgCl2, each dNTP at a concentration of 0.2 mM, 1.25 IU of Taq polymerase, each primer at a concentration of 10 pmol, and 2ul of the DNA template. Sequencing was performed by asking Macrogen. The analyzed sequences were compared using the Blast program provided by NCBI. As a result of identification of 21 strains, 13 Fusarium spp., 1 Peronospora sp. (causative organism of downy mildew), 3 Acremonium spp., 3 Uncultured fungus, 1 Phyrtophthora sp.. The main pathogen of wilt disease was known as Fusarium sp., a soil-borne pathogen. After analyzing the systematic taxonomic relationships, the systematic diagram was created using the neighbor-joining method. Conclusion : We have finally identified Phytophthora cryptogea as a causative organism of Hwanggi root rot disease and will check whether the disease has occurred on the inoculation through pot test.

        Background : Development of new real-time PCR diagnosis method for simultaneous diagnosis of Cylindrocarpon destructans and Fusarium solani, causative fungi of ginseng root rot disease. C. destructans and F. solani are known to be the major pathogens of ginseng root rot disease. Root rot caused by these pathogens is a disease that is difficult to control because the disease progresses slowly and it is difficult to diagnose early and even when symptoms of plant seeding are present, the disease is already spread in the roots. Diagnostic methods to detect the presence or absence of ginseng roots rot fungi in soil before ginseng cultivation are currently being used as a method for controlling. However, commercialized soil extraction kits and PCR diagnostics have cost, diagnostic time, and single diagnostic problems, and need to develop new diagnostic methods. Methods and Results : Primers and probes in the beta-tubulin 2 gene were designed for species-specific detection. In silico analysis, the detection rate of C. destructans was 100% and the detection rate of F. solani was 95%. The multiplex real time PCR optimization conditions including the internal control were established. The analytical sensitivity using positive samples was 10 copies/㎕ for C. destructans and 10 copies/㎕ for F. solani. As a result of performance comparison test with conventional PCR diagnosis methods, it was confirmed that the developed multiplex real time PCR method has the same or better performance in terms of sensitivity. In the developed soil extraction kit, the extraction time was reduced and the extracted DNA quality was improved, compared to the used soil extraction kit. Conclusion : From the above results, we expect that the developed C. destructans / F. solani multiplex real time PCR diagnosis method and soil extraction kit will be useful for real-time monitoring of ginseng root rot pathogenic fungi in the soil of ginseng cultivation area and diagnosis of suitability of ginseng cultivation area.

        Background : Root rot is a major factors of replanting failure in ginseng cultivation. Some of the phenolics detected in the soil could inhibit the seed germination and seedling growth of ginseng. Methods and Results : Water of 2 ℓ was irrigated per pot (20 ℓ) into the soil infected with ginseng root rot pathogens for one month every day. After the irrigation treatment, the powder of ginseng fine root of 20 g per pot was mixed with the irrigated soil. NO3 -, electric conductivity (EC), exchangeable Na (Ex. Na) and K (Ex. K) were decreased in descending order by irrigation. NO3 -, EC, Ex. K, and available P2O5 were increased in descending order by incorporation of ginseng powder into soil. Aerial growth of ginseng was promoted by irrigation, and ginseng root rot was increased by incorporation of powder. Conclusion : The residues of ginseng root in the soil affected soil nutrients and microorganisms, and promoted ginseng root rot, but did not affect the aerial growth of ginseng.

        Background : Aralia continentalis Kitag. (syn. = A. cordata Thunb., Araliaceae) is a traditional medicinal herb spread widely in northeastern Asia, including Korea, China, and Japan. Many constituents from its root extracts, including diterpene and essential oils, have been isolated as active components for antioxidant, anti-inflammatory, analgesic, sedative, antifungal, anti-thrombotic, and growth inhibition. In this study, we describe the structural determination of the two new compounds. Materials and Results : The roots of A. continentalis were collected in Namwon-si, Jeollabuk-do, Korea, in March 2018 and identified by one of the authors (Dr. S. S. Hong). The roots of A. continentalis were extracted with 70% EtOH two times at room temperature. The concentrated residue was subsequently suspended in H2O and partitioned with n-hexane, CH2Cl2, EtOAc and n-BuOH. The n-hexane, CH2Cl2, and EtOAc layer were subjected to sequential column chromatography over silica-gel, RP-18, MPLC and preparative HPLC to isolated the compounds 1 - 30. Consequently, a new nor-ent-pimaran diterpenoid (11) and a new 8-O-4′ type neolignan (25) along with 28 known compounds that included diterpenoids, phenolic derivates, and polyacetylenes have been isolated from the ethanol extract of the roots of A. continentalis. The structures of the new compounds were established by spectroscopic data interpretation, particularly HRESIMS, 1D and 2D NMR data including HSQC and HMBC. Also, those of the known compounds were identified by spectral comparison with those of the reported values. Conclusion : The structures of two new compounds were determined as 18-nor-ent-pimara-9 (11),15-diene-4β-ol (11) and 8-O-4-dehydrocoumaroyl-ferulic acid (25). To our knowledge, compounds 10, 14, 15, 19, 20, 22 – 24, and 26 were isolated and identified from Aralia genus for the first time.
