In this study, we developed vinegar depending on the quantity consumed and type of peeled and unpeeled roots of Platycodon grandiflorum (PG) using Acetobacter pasteurianus A11-2, analyzed vinegar samples using colorimeter and HPLC for 15 days to assess the characteristics on quality, and evaluated their antioxidant activity using 1,1-diphenyl-2-picry1 hydrazy1 (DPPH) and 2.2’-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activities. The major result in PG vinegar was the high acidity of 6.39~6.74% and alcohol was totally converted on the 15th day of fermentation. When we fermented vinegar from peeled roots of 8% PG with a starter culture, we observed high contents of acetic acid, platycodin D, and total polyphenol and high antioxidant activity. Moreover, the vinegar fermented using 8% peeled roots of PG had the high intensity on umami and sour taste and low salty, bitter, and astringent tastes. Consequently, we could develop the PG vinegar with quality and functional characteristics from 8% peeled roots and A. pasteurianus A11-2.
This study investigated the quality characteristics and antioxidant activities of Makgeolli (a traditional Korean rice wine) made with Etteum bell flower root (0.5, 1, 1.5, and 2% with steamed rice) during fermentation. The pH values of Makgeolli with Etteum bell flower root powder decreased after 3 days of fermentation and then increased after 5 days of fermentation. Sugar content decreased after 3 days of fermentation. Color evaluation showed the L values of these drinks decreased during fermentation, whereas a and b values both increased. The alcohol content of Makgeolli increased after fermentation, reaching a maximum concentration of 7.90-8.07% by the end of fermentation. Total phenolic compound contents and DPPH radical scavenging activities increased as the ratio of Etteum bell flower root increased. Sensory scores of Makgeolli fermented with 1.5% Etteum bell flower root ware greater than those of Makgeolli prepared by other treatments. Therefore, Makgeolli added with 1.5% Etteum bell flower root added considered to be the most suitable for manufacturing.
Streptozotocin(STZ)을 45mg/kg.b.w의 용량으로 흰쥐의 미정맥에 투여 한 후 유발된 당뇨 흰쥐에게 1일 1회 7일간 1,000mg/kg의 용량으로 도라지 뿌리 에탄올 추출물을 투여 후 glucose 함량과 당대사에 관여하는 효소인 glucose-6-phosphatase(G-6-Pase) glucose-6-phosphate dehydrogenase (G-6-PDH), glucokinase(GK)활성과 glycogen 함량, triglyceride(TG), total cholesterol등의 지질대사에 관여하는 물질들을 측정하였다 그 결과 도라지 뿌리 에탄올 추출물 투여군이 glucose, TG, total cholesterol등의 함량과 G-6-Pase 활성의 유의적인 감소(p<0.05)를 나타내었으며 glycogen 함량과 HDL-cholesterol, G-6-PDH, GK의 활성이 유의적인 증가(p<0.05)를 나타내었다. 이와 같이 도라지 뿌 리 에탄올 추출물이 항당뇨 개선효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.
본 연구는 도라지의 임간재배 시 대나무용기를 이용하여 적절한 재식방법을 강구함으 로써, 뿌리 생장유도 및 규격화를 통한 품질 향상을 이끌어 내고자 수행되었다. 생존율 은 용기의 길이가 길수록 저조하였고 뿌리의 생중량 변화는 30 cm 용기의 산지황토처리 에서 가장 향상되었다. 뿌리의 길이생장은 70 cm 용기의 산지 부엽토와 황토, 그리고 50 cm 용기의 산지부엽토에서 우수하였으며, 부피생장은 30 cm 용기의 산지황토에서 뛰어났으나 정상적인 생장을 한 도라지의 뿌리길이가 30 cm 이상으로 나타나 적절한 대 나무 용기의 길이는 50 cm로 조사되었다. 뿌리형태의 발달은 50 cm 용기의 산지 부엽 토와 황토에서 우수하였다. 종합해 보면, 고품질 도라지의 규격화 생산을 위한 대나무용 기 길이는 50 cm가 적절하며 산지 부엽토 뿐만 아니라 황토에도 적용이 가능할 것으로 기대된다.
Background : When the Platycodon grandiflorum is applied before the rainy season, the increase of the incidence of the Platycodon grandiflorum root rot disease increases greatly. This experiment was carried out to reduce the incidence of rot root rot disease through the lodging protecting method.
Methods and Results : Three-years-old Platycodon grandiflorum was subjected to four treatments with no treatment, net installation, cutting stem, and removing bud. No treatment was used as a control, and as a further control, netting was used in the Platycodon grandiflorum to set up a treatment with almost no lodging. In the case of cutting stem, the stem was cut off in the middle of June, leaving more than 60 ㎝ before flowering. In the case of removing bud, blooming just before, the bud was removed. As a result, the coverage rate was the highest at 36.9% in the non - treatment area in the middle of July after the rainy season and 0.4% in the net installation. Compared with the case of cutting a lot of stems, 12.7% of the stem was covered with stones, whereas the stalk was 31.8%, which was close to the untreated stomach. As a result of the change of morbidity rate per treatment, it showed a morbidity rate of 49.7% in case of net installation, compared with 60% or more morbidity rate in case of untreated.
Conclusion : As a result, an anti-lodging technic has helped prevent the onset of root rot disease. Further research on how to prevent the lodging of Platycodon grandiflorum using cutting stem will be needed.
Background : Platycodon grandiflorum root (PGR) was one of the primary herbs used in a phlegm-relieving herb from the past. Platycoside compounds on PGR may exhibit neuroprotective, antimicrobial, anti-inflammatory, anti-cancer, anti-allergy, improved insulin resistance, and cholesterol-lowering properties. In order to developing a concentrate product that improved the functionality and preference of PGR, it was fermented using lactic acid bacteria (Lactobacillus plantarum N76-10 and 56-12). Methods and Results : The concentrate products were created by PGR-concentrate (PGRC, 60 ºBrix) mixed with fermented PGR-extract (FPGRE, 2 ºBrix) at the level of 0, 50, 100, 150 and 200%. Sweetness and preference were supplemented by other added materials including honey, oligosaccharide, concentrate of jujube (60 ºBrix) and pear (60 ºBrix), and cactus Chounnyouncho extract (2 ºBrix). The products were put into investigation for their preference of taste, antimicrobial activity in accordance with amount of FPGRE. When it comes to preference of taste, the most favor is adding 100% of FPGRE on PGRC. The product added 150% FPGRE exhibited a strong microbial anti-proliferation in all four kinds (Corynrbacterium diphtheriae, Klebsiella pnneumoniae, Staphylococcus aureus, and Streptococcus pyogenes) of bacteria inducing bronchus diseases and was higher antimicrobial activity than concentrate without FPGRE. In terms of the sensory evaluation (taste, texture and visco-elasticity), concentrate mixed with FPGRE (10), jujube concentrate (2), pear concentrate (10), cactus Chounnyouncho extract (10), oligosaccharide (2), honey (1) and xanthan gum (0.02) showed the highest scores. Conclusion : Thus, A PGR concentrate was made by adding FPGRE (100%) and it was increased organoleptic quality, antimicrobial activity. These studies may provide new product development for effective utilization on Platycodon grandiflorum root.
This study was conducted to elucidate the effects of mulching materials on root growth characteristics and weed occurrence of Platycodon grandiflorum transplants (3 years old) in organic farming. After transplanting, roots were mulched with one of three mulching materials (biodegradable film, sawdust, or rice husks) or a control (no mulching). The average root diameter of all mulching groups was 28.1 mm, which was 5.1 mm thicker than the no-mulching treatment. Roots with the biodegradable film treatment were the thickest at 30.9 mm. Root length was the shortest, 22.0 cm, with rice husk mulching. The number of rootlets was the highest with film mulching (36.0 rootlets/root) and the lowest with no mulching. For root fresh weight, which directly affects yield, the average of treated and untreated roots was 100 and 56 g/root, respectively. The highest fresh weight was in the biodegradable film plot (130 g/root). The mulching treatment groups showed a weed inhibition effect compared with the no-mulching treatment. The dominant weeds with the no-mulching treatment were Setaria viridis and Digitaria ciliaris, and that with the mulching treatment was Setaria viridis. Average annual weed occurrence across all treatments was 72%. Weed occurrence was the highest with the no-mulching treatment (125 weeds/m 2 ), and there were no weeds with the biodegradable film treatment.
The quality and shelf life of sliced root of Platycodon grandiflorum (Doraji) treated by ozon-microbubble-heat shock (OMH) were investigated by combining modified-atmosphere packaging [MAP, (50% O2+15% CO2+35% N2)]. The study was based on microbiological (total viable bacteria, fungi, Enterobacteriaceae and coliforms numbers), physicochemical and sensory changes. OMH treatment was effective in reducing microbial populations of the sliced Doraji, especially Enterobacteriaceae and coliforms reduced by 2 log CFU/g. However OMH-MAP treatment remained the aerobe and fungi numbers. Regarding the color, OMH-MAP delayed the change of Hunter b* and the sliced Doraji by OMH-MAP treatment exhibited lower decrease of flavor and overall acceptability compared to those by polypropylene packaging after tap water treatment (Control). The OMH and 50%O2+15%CO2 MAP treatment gave better sensory quality and extended shelf-life for sliced Doraji (~3 days longer shelf-life than Control). Flavor was significantly related to overall acceptability at both Control and OMH-MAP, whereas total coliforms prevalence was associated with overall acceptability at only OMH-MAP. Therefore microbubble-heat shock treatment may improve microbial safety and sliced Doraji by OMH treatment can stored under 50%O2+15%CO2 treatment for up to 7 days. Thus, OMH and MAP treatment may be used in maintaining the storage quality and marketability of sliced Doraji.
This study was performed to compare the effect of Platycodi Radix (PG) and Black Platycodi Radix (BPG; developed from fresh Platycodon grandiflorum root by steaming nine times at 80°C for 36 hr and drying nine times at 30°C for 24 hr , at which point it becomes black in color.) on anti-oxidant effect, anti-bacterial properties and ovalbumin (OVA)-induced allergic asthma mouse model by calculating serum cytokines and IgE. To induce the allergic asthma, in the control group and the sample treated group, mice of each group were sensitized intraperitoneally with OVA solution at the 1st, the 7th and the 14th day. Sensitization was performed by aerosol allergen challenges with 1% OVA solution intratracheally at the 21th, the 23th, the 25th and the 27th day. At the 29th day, the mice were killed and the levels of interferon-γ, interleukin-4, 5 and 10, total IgE and OVA-specific IgE were measured. IFN-γ was not different among the PG, BPG, and control groups. IL-4, IL-5, IL-13, total IgE, and OVA-specific IgE were significantly in the BPG group compared with the control group. Considering the above experimental results, this study showed that nine times-repitative steaming process on the roots of Platycodon grandiflorum could enhance reduction of the allergic reaction by reducing level of OVA specific IgE and immune cell infiltration and restoring Th1/Th2 cytokine imbalance.

To evaluate preference and functionality of PGR extracts, PGR was fermented by several lactic acid bacteria. Lactic acid bacteria used were Leuc. mesenteroides N12-4 and N58-5, L. plantarum N76-10 and 56-12, L. brevis N70-9 and E3-8. Methods and Results : This study was performed in order to investigate the changes of platycosides, as well as the antimicrobial activities on bronchus diseases inducing bacteria(C. diphtheriae, K. pnneumoniae, S. aureus, S. pyogenes) of Platycodon grandiflorum root(PGR) fermented by using lactic acid bacteria(Leuc. mesenteroides N12-4, Leuc. mesenteroides N58-5, L. plantarum N76-10, L. plantarum N56-12, L. brevis N70-9, L. brevis E3-8). Growth of L. plantarum on PGR was the most active during lactic acid fermentation by some different strains. Total platycoside, platycoside E, platycodin A, polygalacin D2, polygalacin D and diapioplatycoside E contents of PGR fermented for 96 hours at 37℃ by Leuc. mesenteroides and L. plantarum were increased, while platycodin D and platycodin D3 were decreased. The antimicribial activity on PGR fermented by L. plantarum N56-12 exhibited a strong microbial proliferation in all four kinds of bronchus diseases inducing bacteria and was higher than non-fermented PGR extract. Conclusion : Thus, this results showed antimicrobial activities on bronchus diseases inducing bacteria and platycosides content of PGR by L. plantarum N56-12 were higher than non-fermented PGR extract.
The roots of Platycodon grandiflorum are massively used in traditional herbal medicine as a remedy for pulmonary disease and respiratory disorders. However, in spite of its potential medicinal significance, the molecular mechanism of its roots is still unknown. In the present study, high throughput proteome approach was conducted to profile proteins from 3, 4 and 5 months aged diploid and tetraploid roots of Platycodon grandiflorum. Two dimensional gels stained with CBB, a total of 68 differential expressed proteins were identified from diploid root out of 767 protein spots using image analysis by Progenesis SameSpot software. Out of total differential expressed spots, 29 differential expressed protein spots (≥ 2-fold) were analyzed using LTQ-FTICR MS whereas a total of 24 protein spots were up regulated and 5 protein spots were down-regulated. On the contrary, in the case of tetraploid root, a total of 86 differential expressed proteins were identified from tetraploid root out of 1033 protein spots of which a total of 39 differential expressed protein spots (≥ 2-fold) were analyzed using LTQ-FTICR MS whereas a total of 21 protein spots were up regulated and a total of 18 protein spots were down-regulated. It was revealed that the identified proteins from the explants were mainly associated with the nucleotide binding, oxidoreductase activity, transferase activity. In that way, the exclusive protein profile may provide insight clues for better understanding the characteristics of proteins and metabolic activity in various explants of the economically important medicinal plant Platycodon grandiflorum.
도라지를 수확 후 진공포장, 밀폐포장, 질소가스포장 후 저온과 상온에 100일 동안 저장하면서 저장 50일과 100일에 도라지 경도와 사포닌 함량을 조사한 결과를 요약하면 아래와 같다.1.저장온도에 따른 도라지의 경도는 저온저장이 상온저장보다 높게 나타났으며, 저장기간이 길어질수록 도라지의 경도는 낮아졌다. 2.저장방법에 따른 도라지의 경도는 저장 50일차 지퍼팩을 이용한 밀봉에서 1,765 g으로 가장 높게 나타났으며, 저장온도와 저장방법과의 상호작용에서는 고도의 유의성(P≤0.01)이 인정되었다.3.저장온도에 따른 platycodin D3, polygalacin D, 그리고 deapioplatycodin D의 함량조성이 다르게 나타났다.4. 저장기간에 따른 platycodin D3와 polygalacin D의 함량은 저장 50일 이후 감소하는 경향을 보였지만, deapio-platycodin D는 저장기간이 길어질수록 증가하는 경향을 보였다.
We developed an efficient protocol for transformation of balloon flower (Platycodon grandiflorum) root cultures by using cotyledon leaf explants that were infected by Agrobacterium rhizogenes. We found that four different strains of A. rhizogenes differed in their ability to transform P. grandiflorum hairy root cultures. We also found correct antibiotics concentration for selection after transformation by Agrobacterium. Our results demonstrate that use of suitable strain of A. rhizogenes and correct level of antibiotics for the hairy root culture, and may allow to study and apply of valuable metabolites like phenolic compounds from P. grandiflorum hairy root culture.
본(本) 시험(試驗)은 1997년(年)부터 1998년(年)까지 2년간(年間) 사각 pot를 이용하여 식양토, 미사질양토, 사양토 등 3토성(土性)을 처리하여 년수별 생육과 수량 및 품질에 미치는 영향을 알고자 시험하였던 바 얻어진 결과를 요약하면 다음과 같다. 토성처리간 년수별 길경(桔硬) 경장의 경시적(輕視的) 변화를 보면 1년생(年生)은 7월(月) 10일(日)에서 8월(月) 10일(日) 사이, 2년생(年生)은 5월(月) 20일(日)에서 6월(月) 20일(日) 사이에서 최고로 성장 하였고 엽수, 분얼수, 분지수는 식양토가 양호하였으며, 사양토, 미사질양토 순(順)으로 많았다. 1년생(年生) 길경(桔硬)의 출현기 는 토양처리간 차이가 없었고 개화기는 식양토가 가장 빠르고 엽수 및 분얼수도 많았으며 주근장, 지근수, 주근경도 식양토가 사양토, 미사질양토보다 크고, 2년생(年生)도 같은 경향을 보였다. 건근수량(乾根收量)은 식양토에서 1년생(年生)은 274kg/10a, 2년생(年生)은 934kg/10a로 가장 많았고, 다음이 사양토. 미사질양토 순(順)이었고, 주근(主根) 표피색(表皮色)은 식양토는 담갈색, 사양토, 미사질양토 갈색이었으며, 주근육색(主根肉色)은 식양토가 황백색이였다. 길경(桔硬) 1년색(年生), 2년생(年生)의 엑스 함량은 사양토가 1년생(年生) 47. 1%, 2년생(年生)은 36.3%로서 가장 높았고, 다음이 미사질양토, 식양토 순(順)이었으며, 반면에 조(粗)사포닌 함량은 식양토가 1년생(年生)은 3.0%, 2년생(年生)은 3.6%로서 많은 편이었으나 토양 처리간 유의성은 인정되지 않았다.
