Background : Platycodon grandiflorum has been used as famous medicinal vegetable and traditional medicine in Korea. Platycodon grandiflorum is used more for vegetable than it is used as a medicine. Proper storage conditons are required for distribution of Platycodon grandiflorum as vegetables. The purpose of this study was to find the appropriate storage temperature. Methods and Results : 2-years old Platycodon grandiflorum cultivated in Herbal Crop Research, RDA Eumseong was used. They were packed with plastic container box of 45 × 70 × 30 ㎝ and then stored at –2℃, 2℃, 5℃, 25℃, humidity 80 - 82%. During the storage, hardness, L a b values, weight loss, decay rate were measured every 30 days as quality indices. During the storage period, L value decreased but a and b values increased and same pattern was observed at all storage temperatures. Hardness decreased and same pattern was also observed at all storage temperature. Under constant humidity conditions, weight loss was larger at higher temperature. The higher the temperature, the larger the deviation at weight loss. There were little decay but 25℃ temperature stored showed that dacay rate was higher with time over. Conclusion : The favorable temperature for storage is –2℃ - 2℃ while maintaining constant humidity.

Background : Licorice (Glycyrrhiza uralensis F.) is one of the most widely used raw materials for food and pharmaceuticals, and is currently the No. 1 imported medicinal plant in Korea. Efforts to settle licorice cultivation in Korea have been carried out for hundreds of years but have not succeeded, and many people suspect that the reason is due to the inadequate cultivation environment. However, even until recently, most licorice has been procured by wild harvesting, so studies on licorice cultivation technique and cultivation environment have not been conducted much in the world. This experiment was conducted to investigate the germination characteristics of licorice by temperature and salt concentration in order to find the optimal cultivation environment conditions of licorice. Methods and Results : The germination rate of licorice seeds was high 77.5 - 90.5% at 10 - 3 6℃, but it started to drop from 66.5 - 68.0% at 38 - 40℃ and it decreased sharply from 9.0 - 15.0% at 42 - 46℃. No longer germinated at temperatures above 48℃. The average number of days of germination was 12.3 days at 10℃, 5.7 days at 20℃, 3.5 days at 30℃ and 4.1 days at 40℃. The germination start date was 2 to 4 days in the 18℃ - 46℃ range. The average germination rate was the highest at 32℃. The germination time of 40% was 2 - 4 days at 18 - 46℃. The germination of more than 80% took 3 - 5 days at 20 - 40℃, 6 - 8 days at 14 - 1 8℃ and 10 days at 10 - 12℃. Licorice was able to germinate even in the presence of salt. Seed germination rate was 78.5 - 85.5% at 0 - 1% salt concentration and 63% at 1.5% and 18% at 2.0%. In addition, 8.5% seeds germinated successfully even at 3.0% salt concentration. Conclusion : Licorice has been known as a low temperature crop, but germination was possible in a wide range of temperature and salt concentration. Especially, the germination characteristic was higher in the high temperature range of 30 - 36℃, which corresponds to the domestic summer temperature. Therefore, the results of this study are expected to be useful reference for exploring suitable cultivation area in Korea.

Background : Ginseng root rot is a devastating disease caused by the fungus, Ilyonectria mors-panacis that generally attacks younger roots (-2 years), leading to defects in root quality, ginsenoside accumulation and also life cycle of the plant. Hence, there is an indispensable need to develop strategies resulting in tolerance against ginseng root rot. The protective role of silicon during pathogen infestation is well documented in other plant systems and a previous study demonstrated that silica nanoparticles are absorbed and accumulated more than the bulk silica in maize. However, the role of silica in ginseng-root rot pathosystem is unknown. Methods and Results : In the present study, we evaluated the effect of silica nanoparticles (N-SiO2) in Panax ginseng during I. mors-panacis infection. Long term analysis (30 dpi) revealed a striking 50% reduction in disease severity index upon 1 mM and 2 mM treatment of N-SiO2. However, N-SiO2 did not have any direct antifungal activity against I. mors-panacis. Candidate genes and metabolites based approach revealed jasmonic acid (JA) mediated sterol accumulation and incresed ginsenside biosyntesis as the key transcriptional reprogramming events orchestrated by N-SiO2 during the fungal infection. Conclusion : In a nut shell, N-SiO2 administration induces transcriptional reprogramming in ginseng roots, leading to increased phytosterol and ginsenosides synthesis resulting in enhanced tolerance against I. mors-panacis.

Background : Sea buckthorn (Hippophae rhamnoides) is a multipurpose small tree with unique berries of high nutritional and pharmaceutical values has wide spread distribution from Eurasia to South east Asia. In recent times the medicinal benefits of vitamin tree are inclining, hence, efforts were taken to propagate them in vitro to exploit their medicinal property. Methods and Results : The tissue culture potential of them was investigated for the ability to induce shoot organogenesis in leaf explant, and induction of direct somatic embryogenesis from leaf tissue. Moreover, we also determined the effective induction medium for callus and somatic embryo production from H. rhamnoides. To induce the callus form leaf tissue, several phytohormone combinations such as α-naphthalene acetic acid (NAA) and 6-benzyladenine (BA), 2,4-Dichlorophenoxyacetic acid (2,4-D) and 6-benzyladenine (BA), and Kinetin (K) were tried with the Murashige and Skoog (MS) as well as woody plant medium (WPM). In MS basal medium, the combination of 2,4-D and K showed the best callus induction rate of 71%, whereas in WPM basal medium the combination of NAA and BA showed the best callus induction rate of 91%. The adventitious root induction form callus was also attempted by using MS and B5 medium with the phytohormone combinations of IBA 1 – 5 g/ℓ. In MS medium, root was induced only at 4 g/ℓ of IBA and 64%, 51% and 55% root induction results were obtained at 3 g/ℓ, 4 g/ℓ and 5 g/ℓ in B5 basal medium, respectively. The somatic embryos were induced only in half strength MS with the triple phytohormone ratio of 2:1:2 of NAA, BA, and K. Conclusion : The in vitro propagation of sea buckthorn was successfully employed by generating callus, adventitious roots as well as the induction of somatic embryos form the leaf tissues derived callus. Our results provided a valuable addition to the utilization of H. rhamnoides thus enabling their propagation.

Background : The most problematic disease in Boxthorn is Anthracnose in open field. New variety with Anthracnose-resistant is good for high yield and Safe fruit production in open field. Therefore it is necessary to develop a new variety with Anthracnose -resistant and high yield. Methods and Results : The new boxthorn line, CB10511-104 was selected from the cross between CB09480-18 and Chunghong (IT258916) to breed new variety with high yield, Anthracnose-resistant and good color in 2011. Its preliminary yield test was performed from 2013 to 2014 and the selected line was named Cheongyang 26. Its regional yield trials were carried out in Cheongyang, Yesan and Jindo for 3 years, from 2015 to 2017. Cheongyang 26 was registered as the new variety, Cheonggang, in 2017. The specific characteristics were summarized as follows. Tree shape is semi-open type and the leaf is lanceolate and green color. The flowers are normal size and purple. The fruit is normal size, round oval and redish. The flowering was June 17 with medium flowering. Number of sprout branch by pruning was generated less than the check variety, Cheongmeong. The infection rates on leaves to Eriophys macrodonis Keifer of Chunggang, 54.4 percent, was higher than the check variety, Cheongmeong. Anthracnose on fruits in open field was slightly strong compared to the check variety, but check variety was Anthracnose-resistant. The content of betaine and free sugar in dried-fruits were higher than that of the check variety. The dried-fruit yield was increased about 26 percent in open field compared Cheongmeong. Conclusion : This variety ‘Cheonggang’ was suitable for cultivation in open field and rain-shelter plastic greenhouse because of Anthracnose-resistant and good color. It is necessary the companion variety because of self-incompatible. It’s suitable ‘Cheonggeum’ in open field, ‘Cheongun’ in rain-shelter plastic greenhouse.

Background : Several members of the genus Clausena have a great potential as a candidate for the identification of new drug lead molecules, but lack of their genomic information can be a hindrance for the verification of the genetic background for future use. To broaden and delve into the genomic features of this genus, Clausena excavata, an important medicinal plant in many Asian countries, was used for RNA-seq analysis. Methods and Results : A ten ㎍ of the total RNA was used for mRNA isolation using oligo-dT beads and random sheared mRNAs were used to prepare a cDNA library for a illumina hiseq 2500 analysis. In total, 17,580,456 trimmed clear reads from the illumina hiseq 2500 were used for de novo assembly using three assemblers, CLC genomics workbench, velvet-oases, and Trinity. A total of 16,638 non-redundant unigenes with an average length of 755 bp were generated by the assembly. The functional categorization of the identified unigenes by a gene ontology (GO) term resulted in 2,305 genes in the cellular component, 5,577 in the biological processes, and 8,056 in the molecular functions, respectively. The top sub-category in biological processes was the metabolic process with 4,374 genes. Among annotated genes, 3,006 were mapped to 123 metabolic pathways by KEGG metabolic pathway analysis tool. The search for simple sequence repeats (SSRs) resulted in 845 SSRs from 749 SSR-containing unigenes and the most abundant SSR motifs was AAG/CTT with 179 occurrences. Twelve SSR markers were tested for cross transferability among five Clausena species; eight of them exhibited polymorphism. Conclusion : In the present study, genomic resources of the genus Clausena were enriched through RNA-seq and SSR markers, which will serve as valuable resources for genomic/genetic studies of the genus Clausena and close relatives.

Background : For ginseng breed development, mostly the pure-line separation method that registers excellent varieties after performing the regional adaptation test with the varieties selected through the preliminary and main productivity tests on selected excellent individuals cultivated in farms or excellent lineages for growing from collected lineages is used. Recently, the quality of ginseng is degrading while the yield decreases by 15 - 20% due to the high-temperature injury, the physiological disorder and/or the diseases caused by the climate warming so that development of disaster tolerant varieties strong against high temperatures, salts and diseases are demanded. For such reason, three lineages showing excellent overground and underground growing were selected from the gene resources collected from 1997 to 2000. And the productivity test has been conducted with these lineages having Chunpoong and Yunpoong, the existing varieties as the reference varieties. For the lineages excellent in growing, the variety registration will be performed after the regional adaptation test. Methods and Results : The results of productivity tests on the 3 lineages (GS98-3-4, GS00-58 and GS97-69) selected for excellent growing while having Chunpoong and Yunpoong as the reference varieties show that on the 2-years-old dormant roots, all of the selected 3 lineages are better in growing compared to the reference varieties. Especially, GS98-3-4 shows excellent leaf lengths and stem lengths while GS97-69 shows excellent leaf widths. On the overground growing of 3-years-old ones, GS00-58 and GS97-69 lineages are exceedingly more excellent compared to the reference varieties while the selected 3 lineages showing superb underground growing. Especially, GS98-3-4 lineage shows great root weights and diameters. Conclusion : According to the productivity tests on the 3 lineages selected for excellent growing, the selected lineages are excellent in both overground and underground growing compared to the reference varieties of Chunpoong and Yunpoong. On the 2-years-old ginsengs, GS98-3-4 shows excellent overground growing while GS97-69 shows excellent underground growing. On the 3-years-old ginsengs, GS00-58 and GS97-69 show excellent overground growing while GS98-3-4 shows excellent underground growing.

Background : Rehmannia glutinose Liboschitz is a perennial herb belonging to the family of Scrophulariaceae. Its roots have been used as a traditional medicine. For cultivation of varieties of Rehmannia glutinose Liboschitz, the pure line selection method, which is to select the best line among those cultivated in farms for pedigree breeding, replicated yield test and regional adaptation test before registering as a new variety, is widely used. Until now, there are 8 registered varieties of Rehmannia glutinose Liboschitz in Korea. The needs for development of disaster-resistant varieties with better chances of surviving when high temperature, salts and disease are increasing. Therefore, this study focused on cultivation of disaster-resistant varieties among those selected for their disaster tolerance and salt tolerance through regional adaptation test. Methods and Results : As a result of examining the incidence of pest insects of the selected 1 - 2 years old varieties used in the study, among the 2-years-old crops, RGES 20 showed the lowest rate in ground parts. Among the 1-year-old crops, the lowest rate incidence of pest insects in ground parts was observed in RGES 23. In addition, RGES 20 and RGES 23 showed superior growth in under ground part with the highest dry root amount. Conclusion : Based on the above results, RGES 20 among the 2-years-old crops, RGES 23 among the 1-year-old crops showed superior growth in under ground part with the highest dry root amount.

Background : The several studies on the characteristics of Korean ginseng cultivars and breeding lines have already been carried out the level of molecular classification analysis in Korea. In spite of where Geumsan is a representative place of Korean ginseng, Geumsan native species (breeding lines) have not yet been carry out analysis of morphological, genetic characteristics and relationship. We have plan to carry out morphological, genetic characteristics and relationship for Geumsan native species, breeding lines. Furthermore, We could be used diverse genetic resources for Ginseng breeding. Methods and Results : In this study, a total of 71 breeding lines and variety (GS97-1 - Geumwon) consisting of native ginseng collections from Geumsan was analyzed to identify for Korean ginseng variety respectively, and clustered for the selection of Geumsan native ginseng in Korea using DNA markers. We collected 71 Ginseng breeding lines from Geumsan. Analyses of the genetic characteristics of the collection were conducted for extraction gDNA using sprout. We were measured DNA concentration using QIAxpert (QIAGEN). Each DNA sample was quantified at the final DNA concentration of 5 ng/㎖ using sterilized distilled water. Korean ginseng 14 variety and 57 Ginseng breeding lines from Geumsan could be identified polymorphism using the selected 6 primer (MFGp183, MFGp130, MFGp110_E, UFGp163, MFGp108 and UFGp156). Conclusion : These finding could be used for morphological and genetic characteristics for produced native ginseng in Geumsan area. Futhermore, we could be used diverse genetic resources for Ginseng breeding.

Background : Recently, there is a urgent demand for development of new varieties with enhanced resistance to various biotic/abiotic stresses. In order to develop ginseng varieties with such traits, systematic breeding program and comprehensive field studies are prerequisite. Methods and Results : ‘Jinwon' seeds were collected in 2003 from the farmer field of Pocheon. Physiological investigation and propagation were conducted from 2008 to 2010. It was given the name 'Eumseong No. 11 through the observed yield trial from 2011 to 2013 and local adaptability was carried out from 2014 to 2017. All phenotypes including agronomic characteristics, seed yield, and physiological response to biotic/abiotic stresses were investigated according to the ginseng GAP and UPOV guidelines. Red leaf and red berry at maturing stage were observed. The time of emergence, flowering and berry maturity of the ‘Jinwon’ were faster than those of ‘Chunpoong’. Stem length of ‘Jinwon’ was longer than that of ‘Chunpoong’, whereas stem diameter was thicker than that of ‘Chunpoong’. Main root length was similar but main root diameter is thicker than that of ‘Chunpoong’. The yield performance of this variety was 576 ㎏/10 a in local adaptability test for two years, which is 9% higher than that of ‘Chunpoong’. ‘Jinwon’ showed strong resistance at high temperatures. Conclusion : Our study demonstrated that ‘Jinwon’ is an ideal variety with heavier root weight and enhanced stress resistance and contribute will enhance biotic/abiotic stress resistance and increase the farmers' income.

Background : The Codonopsis genus belongs to the Campanulaceae, and it is recorded that there are four species of Codonopsis genus in Korea, such as Codonopsis lanceolata, Codonopsis pilosula, Codonopsis minima, and Codonopsis ussuriensis. C. lanceolata has been proved to be safety and efficacy, and has been widely used for medicinal and edible purposes for a long time in East Asian countries including Korea, China and Japan. However, little genetic research has been done. Methods and Results : Ten species of Codonopsis plants were collected and DNA was extracted using CTAB (cetyl trimethylammonium bromide) method. The extracted DNA was diluted to 5 ng/㎕ for the PCR (polymerase chain reaction) process. C. lanceolata genome was used to develop molecular markers by searching insertion and deletion regions (InDel) in the chloroplast sequence. The developed markers were applied to 4 individuals per Codonopsis species. PCR amplification was carried out using a denaturation at 94℃ for 30 sec, annealing at 58℃ for 30 sec and extension at 72℃ for 30 sec, repeated for 35 total cycles. The PCR products were separated in a 4% agarose gell at 100 V for 40 min. Conclusion : Using the molecular markers developed in this study, genetic diversity of Codonopsis genus was tested, and at the same time, a specific molecular marker was developed to differentiate C. lanceolata from the Codonopsis plants.

Background : Angelica species are representative medicinal plants and it has been used in traditional medicinal methods, especially, in the traditional Asian medicine. The Angelica species used in conventional medicine varies by country according to specific regulations, i.e. A. gigas Nakai in Korea, A. sinensis Diels in China, and A. acutiloba Kitagawa in Japan. Because of the similarity between the names among Angelica, they can be confused in the market. Methods and Results : In this study, twenty-four chloroplast insertion or deletion (cpInDel) markers were developed from chloroplast DNA of A. gigas Nakai and tested for the classification of Angelica species. Primer sets were designed from flanking sequences of the discovered InDel loci from chloroplast DNA of A. gigas Nakai using CLC Main Workbench with the following parameters : primer length = 18 - 26 bp (Opt. 23 bp); GC% = 50 - 70% (Opt. 60%); Ta = 55 - 62℃ (Opt. 58℃); product size range = 120 - 300 bp. Polymorphism and genotype analysis of 13 Angelica species were performed using the developed cpInDel markers. Conclusion : The 24 cpInDel markers developed in this study could be used for genetic diversity analysis and classification of Angelica species.

Background : Jujube (Zizyphus jujuba. Mill) is a broad-leaved shrub belonging to the family Seagull. Its origin is India and its height is about 5 m. The flowers are gathered in two to three in May-June, with five petals and yellowish green. Leaves are alternate, egg-shaped or long egg-shaped, with clearly visible three veins. The fruit, called jujube, is an elliptical nucleus with the seed wrapped in a solid nucleus. It is 2.5 - 3.5 ㎝ in length, green at first, ripened in brown or reddish brown in September-October. Jujube uses the bud mutation to breed and spreads through grafting. Therefore, there is little difference in phenotype between cultivars. However, because of the lack of research on jujube molecular biology, there is no standard to distinguish the variety at the DNA level. In order to overcome such difficulties and to create a research foundation of jujube, we have developed molecular markers from jujube. Methods and Results : We collected 12 jujube varieties include Bogjo and extracted DNA using CTAB method. The DNA was diluted to 10 ng/㎕ and kept at -20℃. We designed the primer sets using CLC Main Workbench based on DNA InDel regions between the varieties. PCR and electrophoresis were performed to confirm the polymorphism. We designed 26 primer sets from 23 InDel regions. Eighteen of 26 primer sets amplified the amplicon from the primer screening. Eight primer sets were selected for polymorphism assays. All primer sets showed polymorphism. The domesticated cultivars were divided into two groups and the Japanese and Chinese varieties were separated. Conclusion : The InDel markers developed in this study could be good tools to differentiate the jujube cultivars cultivated in Korea.

Background : Solanum nigrum Lin. is an annual herbaceous plant which belongs to the family Solanaceae as known the important medicinal herb and health-benefiting plant. Phytochemical investigation of whole plant reported that contain total alkaloids, steroid alkaloids, steroidal saponins, flavonoids, tannins, glycosides, proteins, carbohydrates, coumarins and phytosterols, and glycoproteins, exhibiting antitumor activity. Methods and Results : The accumulation of anthocyanin known as synthesis by flavonoid biosynthesis pathway is increased by AtPAP1 transcriptional factor. To identify the effect of AtPAP1 in S. nigrum, we generate AtPAP1-overexpressed S. nigrum and investigate total anthocyanin in these transgenic lines. The transgenic lines resulted in increasing the total anthocyanin amount compared with control line. Thus, the transgenic lines appeared purple colored leave as well as light purple in flower petal and dark purple in flower anther. To identify that AtPAP1 affects the genes profiling of anthocyanin biosynthetic pathway in S. nigrum, we harvested the AtPAP1-overexpressed S. nigrum transgenic lines and proceeded to qRT-PCR analysis. The result indicated that AtPAP1 gene highly induces in transgenic lines and the most upstream pathway genes of flavonoid such as SnPAL, SnC4H, Sn4CL, SnCHI and SnHCT are induced by AtPAP1 overexpression, excepting UGT75C1. According to these results, those functional genes may be regulated by AtPAP1 transcription factor. Thus, the purple color appearing in AtPAP1-overexpressed S. nigrum plants may be regulated by those genes. Conclusion : When AtPAP1 is overexpressing, the most genes in flavonoid pathway upregulate and activate resulting in accumulation of anthocyanin compound in leave and flower organs in S. nigrum. Ultimately, our data will be helpful for further research in S. nigrum molecular and physiology and better strategies for exploiting its phytochemical properties.

Background : The roots of Codonopsis pilosula (Franch.) Nannf are used as medicines such as expectorants, and they are edible. Codonopsis pilosula (Franch.) Nannf currently has a very small cultivation area, and there are no breeding cultivars, most of the native species are cultivated in the farm house. Therefore, it is important that identification of collecting species characteristic in order to breeding strategy. In this study, morphological features of the leaves and stems of the collecting species in the area were observed. Methods and Results : The morphological characteristics of mansam collected from three areas including Cheolwon (41 accession), Jeongseon (19 accession), and Pyeongchang (15 accession) were investigated at the flowering stage on leaf and stem. As a result of investigation of leaf characteristics, The leaf shape was the most frequent with 78.9% in the kidney type, 8.5% in the cardiac type, 7.0% in the wide-elliptical type, and 2.8% in the egg and egg-oval type. The leaf length was the most frequent with 67.1% in case of short, 28.6% in case of middle, and 4.3% in case of long. The leaf width was 67.1% in case of middle, 30.0% in case of broad, and 2.9% in case of narrow. Leaf hair was 47.1% in case of plenty, 27.1% in case of middle, and 25.7% in case of small number. The degree of greenness of the leaves was 60.0% in case of middle, 31.4% in case of darkness, and 8.6% in case of lightness. The degree of pigmentation of anthocyanin in petiole was 41.4% in case of lightness, 31.4% in case of darkness, and 27.1% in case of middle. As a result of investigating the characteristics of stem, The presence and absence of hair on the stem was 95.7% in case of little, 2.9% in the middle, and 1.4% in case of plenty. The degree of staining of anthocyanin in the stem was 42.9% in case of middle, 30.0% in case of darkness, and 27.1% in case of lightness. Conclusion : We intend to use this data as a basic data for cultivating varieties by supplementing investigation of traits such as expansion of collection area and investigation object and flower shape in the future.

Background : Atractylodes japonica koidz. (AJ) and Atractylodes macrocephala koidz. (AM) belong to Atractylodes genus (Asteraceae) and their rhizomes are used as traditional medicine ‘Baek-chul’. The ‘Baek-chul’ is considered to be effective for the treatment of stomach disorder, virus, diuresis, inflammation, arthritis and Atractylenolide I, Atractylenolide II and Atractylenolide III are the major active ingredients of these. Previously, we developed 8 hybrid cultivars with disease resistance, high yielding ability and high active ingredients by interspecific hybridization between AJ and AM. In this study, growth characteristics of 8 hybrid cultivars were investigated in RDA experimental field. Methods and Results : Growth characteristics of 8 hybrid cultivars were investigated in RDA experimental field. Among these cultivars, ‘Sanwon’ had the highest fresh weight of rhizome (149.5 g/plant), followed by ‘Dachul’ (145.8 g/plant). Most of hybrid cultivars showed higher fresh weight than and AM (108.7 g/plant). In addition, active ingredients (Atractylenolide I, Atractylenolide Ⅱ and Atractylenolide Ⅲ) were investigated using HPLC. As a result, The sum of active ingredients were the highest in ‘Dachul’ (0.40 ㎎/g) that was 2 times higher than AM (0.22 ㎎/g). Conclusion : In this study, hybrid cultivars showed higher agronomic performance than AM. Particularly, ‘Dachul’ could be the superior cultivar with high atractylenolides as well as high yield ability.

Background : Atractylodes japonica koidz (AJ) is a perennial herb that belongs to Atractylodes genus. The dried rhizome of AJ is known as ‘Baek-chul’. The ‘Baek-chul’ is used as important traditional medicine in north-east Asia. It is considered to be effective for the treatment of stomach disorder, virus, diuresis, inflammation, arthritis. AJ is heavily depend on import from china and only few studies have been carried out. In this study, we develop SSR marker to build a foundation of breeding, to analyze genetic diversity and to construct core collection. Methods and Results : AJ resources was collected from each different place. To find simple sequence repeat (SSR) marker, we sequenced genomic DNA of AJ resources using Illumina HiSeq 2000 System. As a result of next generation sequencing (NGS), we obtained putative SSR loci. From these SSR primers, 553 SSR primer sets were designed successfully and confirmed polymorphism by in silico analysis. Nucleotide motifs ranged from tri- to penta-. Among these, 48 primer were tested in 4 individuals by capillary electrophoresis. Finally, selected 28 SSR marker were showed clear band and polymorphism by Electrophoresis. Conclusion : In this study, we developed 28 polymorphic SSR marker using NGS, and it could be used for analyzing genetic diversity of A. japonica. These marker would be useful for breeding of new cultivar in the future.

Background : Schisandrae Fructus (Omija) has been widely used for medicinal herbs because it is known for its various medical functions and antimicrobial activities. This study was conducted to investigate the antimicrobial activities of extracts from Schisandrae Fructus against six foodborne pathogens (Bacillus cereus, E. coli, Listeria monocytogenes, Salmonella enteritidis, Staphylococcus aureus, Vibrio parahaemolyticus) and figured out the efficient extraction methods. Methods and Results : Traditionally, Schisandrae Fructus is soaked or boiled in water when people drink it like tea. In this study, we extracted Schisandrae Fructus by soaking in water and ethyl alcohol for 24 hours, ultrasonicating in methyl alcohol and water for 60 minutes, and boiling in water for 30, 60, 90, 120 minutes. Each extract used for paper disc test that estimates antimicrobial activities by measuring the inhibition zone. The blank discs that absorbed with extracts were placed on the agar plates that smeared with each foodborne pathogen, then incubated overnight at 37℃ incubator. Then, antibacterial extracts formed clear zones around the disc. In this study, Vibrio parahaemolyticus was the most sensitive to the all kinds of extracts and methyl alcohol extracts that were obtained by ultrasonicating showed the most strong antimicrobial activities against six foodborne pathogens (23, 12, 29, 18, 23, 28 ㎜). Hot water extracts and soaking extracts in water showed the similar results, and the extracts that was boiled for 60 minutes was the best results (13, 8, 21, 10, 13, 17 ㎜) against all the pathogens. Conclusion : The extracts from Schisandrae Fructus inhibited the growth of six foodborne pathogens. Soaking and boiling in water are good methods for drinking Schisandrae Fructus like tea because they showed antimicrobial activities in the paper disc test. According to this study, the best boiling time for effective antimicrobial activity is 60 minutes.

Background : Natural substance extracted from plant is going increased to apply for agricultural industry to improve crop product. Thus, hydrosols of lavender used to cosmetic materials was treated in plant to investigate growth effect. Methods and Results : The young plants grown for 7 days from seed were transferred to pot. They have foliage dressed with lavender hydrosols diluted to 500 × at intervals of 2 days. The results of N. benthamiana grown by foliage dressing for 2 weeks were 11.37 ± 0.31 ㎝ of leaves length, 11.07 ± 0.47 ㎝ of leaves width, 14.43 ± 0.90 ㎝ of plant length. These values were investigated to more increase than control. In the results measuring plant length at 4 weeks, there had no difference in two treatments. The activities of DPPH scavenging and SOD were checked to examine the antioxidant activity in two treatments. The DPPH scavenging activity was not shown a difference in two treatments. However, SOD activity displayed at high to level of 2 times in extracts from 100% ethanol of N. benthamiana treated with lavender hydrosols in compared to control. The RT-PCR preceeded to investigate the transcriptional changes of genes related to antioxidant activity using the respective primers for 5 genes such as NbNOA1, NbNR, NbPAL, NbAPX, NbSOD. These 5 genes were known to detect as a high level in N. benthamiana treated with lavender hydrosols in transcriptionl expression. Conclusion : The influence on plant growth through lavender hydrosol treatment was represented as a promoting effect in early growth stage.

Background : Obesity, a global health problem and a chronic diseases, is associated with increased risk of developing type 2 diabetes and coronary heart diseases. A wide variety of natural remedies have been explored for their obesity treatment potential. To elucidate the anti-obesity effect of ginsenoside Rg5 : Rk1 (Rg5 : Rk1), a mixture of protopanaxadiol type ginsenosides isolated from Panax ginseng Meyer in a 3T3-L1 adipocytes. Methods and Results : In order to determinate the anti-obesity effect of Rg5 : Rk1, Oil Red O staining and triglyceride (TG) content was assessed. Furthermore, to elucidate the possible mechanism whether Rg5:Rk1 affects lipid accumulation, mRNA and protein expression analyses of adipocyte markers such as STAT3, PPARγ, CBEPα and ap2 were carried out. Rg5:Rk1 treatment showed an inhibition of lipid droplet accumulation and decrease on TG content. In addition, expression of STAT3, PPARγ, CEBPα and ap2 were decreased in dose dependent manner. Similar to these results, Rg5:Rk1 treatment reduced PPARγ and CEBPα protein expression. Conclusion : Rg5 : Rk1 treatment exhibits anti-adipogenic activity by down-regulation of the STAT3PPARγ/CEBPα pathway in 3T3-L1 adipocyte cell line.
