        To study the optimal cultivation for paddy soil, growth characteristics and ginsenoside content was investigated by both of poor drainage class (PDC) and imperfect drainage class (IDC) in three-year-old ginseng of varieties, Cheonpoong (CP), Yeonpoong (YP), Hwangsookjong (HS), and Jakyeongjong (JK). Root yield of IDC was higher than that of PDC by 3.6 times because stem length, leaf area, and chlorophyll contents were increased, while discolored leaf ratio was decreased. Root yield of HS in PDC was highest among four varieties because chlorophyll contents, leaf area, and survived plant ratio were relatively high. Root yield of CP in IDC was highest among four varieties because of high leaf area and survived plant ratio, and low discolored leaf ratio. Ratio of rusty-colored root showed significant difference by varieties, which was the highest in HS and the lowest in CP among four varieties irrespective of drainage classes. Total ginsenoside contents showed significant difference by drainage classes, which were high in IDC of good growth and low in PDC of poor growth. Total ginsenoside contents were high in JK and CP, while low in HS and YP both of drainage classes.

        We developed a surface washing system for ginseng. The washing system was developed using different treatments and conditions and characterized in terms of product hardness, weight loss, and change in temperature and color.         Cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.95), catalyzes the reduction of hydroxycinnamaldehydes to give hydroxycinnamyl alcohols, or "monolignols," the monomeric precursors of lignin. Lignins are important components of cell walls and lignified secondary cell walls play crucial roles in long distance transport of water and nutrients during plant growth and development and in plant defense against biotic and abiotic stresses. Here a cDNA clone containing a CAD gene, named as PgCAD, was isolated from a commercial medicinal plant Panax ginseng. PgCAD is predicted to encode a precursor protein of 177 amino acid residues, and its sequence shares high homology with a number of other plant CADS. The expression of PgCAD in adventitious roots and hairy roots of P. ginseng was analyzed using reverse transcriptase (RT)-PCR under various abiotic stresses such as salt, salicylic acid, wounding and chilling treatment that triggered a significant induction of PgCAD at different time points within 2-48 h post-treatment. This study revealed that PgCAD may help the plants to survive against various abiotic stresses.

        The parts of leaves, flowers and stems in ginseng were obtained for analyzing the component of saponin on 15th April, 25th April, 5th May, 25th May, which were considered as ginseng foliation stage. The total saponin content of the leaves were 97.29, 66.42, 67.61, 36.24 mg/g, respectively, in which the content of Re, Rb1 and Rd were more than 2/3 amount of total saponin. Especially, the saponin content of leaves decreased according to the sequential collection days, in which the similar results were observed from the flowers and stems of ginseng. The total saponin content of the flowers and stems were 141.09,143.84,139.25,133.47 and 13.32, 9.85, 8.00, 4.65 mg/g, respectively. Among them, the content of Re, Rd and Rb2 in flowers were more than 2/3 while the content of Re, Rg1 and Rd in stems showed more than 9/10 amount of total saponin. The total saponin content of individual leaf were 19.46, 28.56, 58.82 and 169.24 mg/plant, 2.53, 2.76, 5.20 and 12.32 mg/plant in stems, and 14.11, 30.21, 37.60 and 73.41 mg/plant in flowers. Therefore, the total saponin content of aboveground parts in ginseng were leaves 〉 flowers 〉 stems.

        Somatic embryos on growth regulator-free medium can be produced directly from cotyledon explants of Panax ginseng C. A. Meyer. When the embryo developmental stage was torpedo and cotyledon, the germination rate of embryos was quite high on MS medium supplemented with gibberellic acid (GA3). However, the percentage of plantlet formation at the cotyledon stage was higher than that at the torpedo stage. This result demonstrates that the embryo at the cotyledon stage was the most appropriate for increasing germination by GA3. Embryos cultured on medium including four levels of GA3 concentrations (3, 5, 10, or 20 mg/l) showed all quite high germination rates (87-91%). When the well-developed embryos were continuously cultured on media including high concentrations of GA3 from 10 to 20 mg/l, the percentage of formation of normal plantlets was lower than that seen under low concentrations from 3 to 5 mg/l. This treatment of high concentrations resulted in shoots with abnormal shape. The optimal GA3 treatment provides a basis for the efficient method obtaining healthy plantlets derived from ginseng somatic embryos.

        This study was conducted to assess water movement in paddy-upland rotation soil scheduled for ginseng cultivation through the measurement of infiltration and permeability of soil water. Soil sample was divided with four soil layers. The first soil layer (to 30cm from top soil) was loamy sand, the second and the third soil layers (30~70 ㎝) were sand, and the fourth (〈 120 ㎝) was sandy loam. The soil below 130 ㎝ of fourth soil layer was submerged under water. The shear strength, which represents the resisting power of soil against external force, was 3.1 kPa in the first soil layer. This corresponded to 1/8 of those of another soil layer and this value could result in soil erosion by small amount of rainfall. The rates of infiltration and permeability depending on soil layers were 39.86 cm hr-1 in top soil, 2.34 cm hr-1 in 30~70 ㎝ soil layer, 5.23 cm hr-1 and 0.18 cm hr-1 in 70~120 ㎝ soil layer, with drain tile, and without drain tile, respectively. We consider that ground water pooled in paddy soil and artificial formation of soil layer could interrupt water canal within soil and affect negatively on water movement. Therefore, we suggest that to drain at 5 m intervals be preferable when it makes soil dressing or soil accumulation to cultivate ginseng in paddy-upland rotation soil to reduce failure risk of ginseng cultivation.

        This study was conducted to assess the response of LED (Light-emitting diode) irradiation on the growth characteristics and saponin contents of Panax ginseng C. A. Meyer. LED irradiation showed a positive effect for most of the parameters studied. The content of chlorophyll a in leaves was increased by 4.9~36.5%, under LED and fluorescent light conditions compared to the control. The content of chlorophyll b was also increased by 44.4~55.6% under blue and red LED compared to the control except under the red plus blue LED condition. The shoot and root weight were increased by 20~60% and 14.8~59.3%, respectively under LED and fluorescent light conditions compared to the control. The total saponin content was increased by 1.8% under blue LED compared to the control, while total saponin content was decreased by 8.8~11.5% under red LED, red plus blue LED and fluorescent light conditions.

        본 연구는 5 년근의 인삼에서 차광시설의 재배위치에 따른 사포닌의 함량 차이를 검정하였다. 재식 위치별 전행 (1~2행) 에서 연풍과 재래종 주근의 총 사포닌 단위함량은 각각 15.01 mg/g, 21.79 mg/g, 지근은 각각 35.93 mg/g, 43.32 mg/g, 세근은 각각 87.85 mg/g, 105.51 mg/g이었다. 중행 (3~5행) 에서 연풍과 재래종 주근의 총 사포닌 단위함량은 각각 18.73 mg/g, 23.19 mg/g, 지근은 각각 44.92 mg/g, 43.50 mg/g, 세근은 각각 92.97 mg/g, 110.70 mg/g이었다. 후행 (6~7행) 에서 연풍과 재래종 주근의 총 사포닌 단위함량은 각각 21.88 mg/g, 26.68 mg/g, 지근은 각각 38.41 mg/g, 44.89 mg/g, 세근은 각각 101.03 mg/g, 107.06 mg/g이었다. 재배위치에 따른 연풍과 재래종의 주근, 지근 및 세근의 총 사포닌과 주요 사포닌 함량의 차이는 연풍의 주근을 제외하고 모든 부위에서 크지 않았다. PD/PT 비율은 전행 인삼은 중행과 후행에 비하여 낮았으며 뿌리의 부위별로도 차이가 컸다.

        일반전두부와 비교하여 인삼분말을 첨가한 전두부는 일반성분, 색도, 수분함량에서의 유의적인 차이는 나타나지 않았다. 미생물의 경우 인삼분말 2%첨가 전두부는 저장 15일에서 만 일반세균 3.51 LogCFU/g이 나타났고, 일반전두부는 12일에 일반세균 4.48 LogCFU/g, 15일에 4.81 LogCFU/g로 각각 나타내었으며 대장균의 경우 일반전두부에서만 15일에 3.57 LogCFU/g로 나타나 인삼분말 2%첨가 전두부가 일반전두부와 비교

        논토양에서 적응하는 품종 선발과 재배기술을 개발하기 위해 배수등급이 다른 논토양(배수 불량지와 배수 약간불량지)에서 천풍, 연풍, 황숙종, 자경종 등 4 품종을 사용하여 2년생 인삼의 생육특성 및 수량성과 뿌리의 진세노사이드 함량을 조사한 결과는 다음과 같다. 1. 배수등급에 따라 지상부 생존율과 황증 발생율은 뚜렷한 차이를 보여 배수불량지에서는 지상부 생존율이 저하되고 황증 발생율이 증가하였는데, 배수불량지에서 황숙종은 지상부 생존율이 가장 높고 황증 발생율이 가장 낮았으며, 연풍은 지상부 생존율이 가장 낮고 황증 발생율이 가장 높았다. 2. 배수불량지에서는 배수약간불량지에 비해 지상부가 일찍 고사된 관계로 주당 근중이 작고 수량성도 뚜렷이 감소되었는데, 배수불량지에서의 수량성은 자경종 > 천풍 > 연풍 > 황숙종 순으로 자경종과 천풍의 수량성이 높았으며, 배수약간불량지에는 천풍 > 연풍 > 자경종 > 황숙종 순으로 천풍과 연풍의 수량성이 높았다. 3. 적변율은 배수등급보다는 품종 간에 더 큰 차이를 보였는데, 천풍과 자경종의 적변율은 비교적 작은 반면 연풍과 황숙종은 매우 큰 특징을 보였다. 4. 근 비대가 불량했던 배수불량지는 배수약간불량지보다 진세노사이드 함량이 증가되었는데, 배수불량지에서 품종별 총 진세노사이드 함량은 연풍 > 천풍 > 자경종 > 황숙종 순이었고 배수약간불량지에서는 연풍 > 자경종 > 천풍 > 황숙종 순으로 배수조건에 관계없이 연풍이 가장 높고 황숙종이 가장 낮았으며, 천풍과 자경종은 서로 비슷하였다.

        본 연구는 박피시간에 따른 백삼의 색택과 일반성분 및 사포닌 함량의 변화에 미치는 영향을 조사하였다. 백삼 표피의 색택은 박피시간이 길어질수록 양호하였다. 일반성분인 조지방, 조단백질, 지방산 및 아미노산은 박피에 따라 약간 증가하였으나 탄수화물 및 유리당은 감소하였다. 조사포닌과 ginsenosides 함량은 박피에 따라 크게 감소하였다. 박피 인삼의 ginsenosides는 무박피에 비하여 약 20-30% 감소 하였다. 이러한 결과는 백삼의 박피가 색택을 좋게 하지만 인삼의 주요 유효성분인 사포닌 함량의 현저한 감소를 초래하였다.

        인삼 4년생을 대상으로 해가림 색상을 달리하여 재배 시험한 결과, 대조구인 4중직 PE 차광망(청색 3 + 흑색 1중직)이나 청색 4중직 차광망에서 재배된 인삼의 뿌리 근중은 적색 4중직 차광망보다 높게 나타났으며, 대조구보다 청색과 적색에서 동체의 비율이 낮고 지근의 비율이 높아 투광량이 많아지면 동체의 생장이 감소하고 지근의 생장은 증가하였다. 뿌리에 함유된 진세노사이드 함량은 적색 해가림재배에서 가장 높게 나타나 투광량이 증가할수록 진세노사이드 함량이 증가되었으며, 특히 진세노사이드 Rg1의 함량이 유의적으로 더 높았다. 색상에 따른 진세노사이드 함량차이는 해가림 색상 차이에 의해 생합성 과정이 영향을 받은 것이 아닌 생육량의 차이에 따른 결과로 판단되었다.

        An advanced extraction method by ultrasonic extraction with applied solid phase extraction (SPE) has been developed for the determination of simultaneous eight major ginsenosides, namely ginsenosides Rg1, Re, Rf, Rb1, Rg2, Rc, Rb2, and Rd in the root of Panax ginseng. Four extraction methods including n-BuOH reflux extraction (Method A), 70% EtOH reflux extraction (Method B), 50% MeOH reflux extraction with SPE (Method C), and 50% MeOH ultrasonication with SPE clean-up process (Method D) were investigated for the determination of eight major ginsenosides. Total contents of ginsenosides were highest by extraction of Method C as 2.408±0.011%. However, Method D was evaluated as relatively simpler and more efficient method due to short extraction time, small solvent consumption and less expensive, compared to conservative reflux method. Ginsenosides were also satisfactorily separated with good resolution and the accuracy range was between 1.05 and 4.06% as relative standard deviation (RSD) by Method D. SPE condition and HPLC condition were further optimized for determination of eight major ginsenosides by the ultrasonic extraction method. Conclusively, ultrasonic extraction of 2 g sample of ginseng using ultrasonic bath and 1 loading for SPE was evaluated as proper condition for extraction of ginseng.

        To selection of optimal shading material, two-year-old ginseng (Panax ginseng C. A. Meyer) of new cultivar, 'Cheonpoong' (CP), and native species 'Hwangsookjong' (HS) were cultured under three kinds of shading materials such as three-layered blue and a one-layered black PE (polyethylene) net (TBSB), blue PE sheet (BS), and aluminium coated PE sheet (AS) in imperfectly drained paddy soil. Growth characteristics, yield and ginsenoside contents were investigated under three shading materials. Yield and ginsenoside contents of ginseng were distinctly affected by intensity and quality of sunlight penetrated through shading materials. Light transmission ratio, air and soil temperature according to shading materials were higher in order of BS, AS, and TBSB. However, ratio of aerial phase and porosity of the soil were higher in order of AS, BS, and TBSB, respectively. There was no significantly difference in the ratio of rusty colored root by shading materials. CP showed higher stem length, leaf area, and root weight than that of HS, while the former showed distinctly lower discolored leaf ratio than that the other. Eight kinds of ginsenosides content of CP were higher than that of HS in Rg1, Re, Rf, Rb1 and Rc except Rg2, Rb2, and Rb3. Total ginsenoside contents of CP was distinctly higher than that of HS. Total ginsenoside contents as affected by shading materials was higher in order of BS, TBSB, and AS in CP, while TBSB, BS, and AS in HS.

        This study was conducted to investigate the influence of shading material on the chlorophyll fluorescence, photosynthesis, transpiration, stomatal conductance and its any correlations in Panax ginseng C.A.Meyer. Fo was higher in polyethylene shade net than in silver-coated shading plate, but this treatment caused a lower Fm in comparison with silver-coated shading plate. Also, Fv/Fm and PhiPS2 showed higher in silver-coated shading plate than in polyethylene shade net. The relationship between net photosynthetic rate and transpiration, stomatal conductance were increased as the PAR (Photosynthetic active radiation) was increased and reached maximum at the 200-400 μmol/m2/s of PAR in all of leaves, and the higher in silver-coated shading plate than in polyethylene shade net. A linear equation was obtained between net photosynthetic rate and transpiration, net photosynthetic rate and stomatal conductance. SPAD was higher in silver-coated shading plate than in polyethylene shade net.

        The aim of this work is to assess the safety of fungicide tolclofos-methyl, difenoconazole and azoxystrobin in ginseng sprayed by safe use guideline. When tolclofos-methyl was sprayed on ginseng by safe use guideline, the residue amounts (MRL) of it in ginseng was 0.13 mg/kg which is below than 0.3 mg/kg, maximum residue limit established by Korea Food & Drug Administration (KFDA). The residue amounts of ginseng parts, head part was 0.37 mg/kg and main body part was 0.13 mg/kg. In case of difenoconazole, the residue amounts in ginseng was 0.81 mg/kg. which was exceed the MRL, 0.2 mg/kg. By the analyze results of ginseng part, the residues of head and main body part were 3.01 and 0.40 mg/kg, respectively. In experiment of vinyl mulching, the residue amount of difenoconazole in ginseng was 0.05 mg/kg. The residue amounts of azoxystrobin in ginseng sprayed by safe use guideline was 0.14 mg/kg. This residue was not exceed the MRL 0.5 mg/kg. The residue amounts by ginseng parts was 0.51 mg/kg for head part and 0.28 mg/kg for main body part. In case of vinyl mulching, the residue amount of azoxystrobin was 0.02 mg/kg.

        PPD ginsenosides in ginseng leaf were analyzed to determine effects of either FIR heat or steaming heat treatment. Among the PPD ginsenosides, Rb1, Rc and Rb3 forming four glycoside-attached aglycons were increased as FIR heat temperatures were increased from 60 to 120℃, while Rb3 was decreased. In addition, FIR heat treatment was effective to increase Rd forming a three glycoside-attached aglycon. Rg3 and Rh2 were not increased by the FIR heat treatment. In steaming heat treatment, Rb1 was significantly decreased, while Rb2 was increased. Rd was also increased by increased steaming temperature, yet its content was lower than in the FIR heat treatment. However, the steaming heat treatment increased yields of Rg3 and Rh2, which were not observed in the FIR heat treatment. Thus, FIR heat treatment was beneficial to efficient products of Rb1, Rc, Rb3 and Rd. Steaming heat treatment was effective to higher collection of Rb2, Rg3 and Rh2.

        최근 들어 발열량이 매우 적으며 식물체의 생장 및 형태 형성에 영향을 미치는 파장역만을 갖고 있는 발광다이오드 (Light- Emitting Diode, LED)를 이용한 광 조사 시스템에 대한 관심이 고조 되고 있다. 인삼은 반음지성 식물로 고온에서는 광합성량이 급속도로 감소하며, 온도가 낮은 이른 아침에 광합성 효율이 높은 것으로 알려졌다. 본 연구는 충남대학교 비닐하우스에 이식한 인삼포에 LED의 청색, 적색, 청색과 적색 혼합광, 백열등을 설치하여 인삼의 생육과 수량에 미치는 영향을 조사하기 위하여 실시하였다. LED 처리는 해뜨기 전 3시간 동안 매일 처리하였으며 인삼 특성은 9월10일에 조사하였다. 근장은 무처리구 20.7cm, 백열등 처리구 21.3cm, 청・적색 혼합광 처리구 20.2cm, 적색광 처리구 20.6cm, 청색광 처리구는 21.7cm로 청색광 처리구가 가장 높았으며, 근직경은 무처리구 6.5mm, 백열등 처리구 7.6mm, 청・적색 혼합광 처리구 7.2mm, 적색광 처리구 6.7mm, 청색광 처리구는 7.8mm로 청색광 처리구가 가장 높았다. 근중은 무처리구 2.7g, 백열등 처리구 4.0g, 청・적색 혼합광 처리구는 3.3g, 적색광 처리구는 3.1g, 청색광 처리구는 4.3g이며, 무처리구에 비해 백열등 처리구 48.1%, 청・적색 혼합광 처리구 22.2%, 적색광 처리구 14.8%, 청색광 처리구 59.3% 증가하여 청색광에서의 생육이 좋음을 알 수 있었다. chlorophyll a는 무처리구 4.1(mg/100g)에 비해 청색광 처리구가 5.6(mg/100g)으로 다른 처리구에 비해 높았으며, chlorophyll b는 무처리구 1.8(mg/100g)에 비해 청색광 처리구가 2.8(mg/100g)로 가장 높았다.
