In this study, the effect of daily short-distance walking exercise on milk production and metabolic status of the lactating dairy cows was investigated. The experiment was conducted with 10 lactating dairy cows comprising 2 groups (5 dairy cows/group), which were walking activity and non-walking activity. The walking activity (WA) group lactating dairy cows walked outdoors for 1 km/d, and the non-walking activity (non-WA) lactating dairy cows were maintained within a cowshed from 18thApril to30th May. The activity volume of the neck was not significantly different between the groups, but the activity volume of the leg was higher in WA group compared with non-WA (p<0.05). The rectal temperature was higher in the WA group (38.6 ℃) compared with non-WA (38.2 ℃) (p<0.05). No significant differences in dietary NE intake, milk production and milk composition were found between the groups. The plasma cortisol concentration was higher in WA group (2.14 μg/dl) compared to non-WA (0.95 μg/dl) (p<0.05). However, plasma cortisol level of WA dairy cows was significantly decreased as compared with non-WA cows. On the contrary, the level of plasma melatonin was increased in WA cows than that of non-WA dairy cows. It may be interpreted as a change in physical fitness. From this study, we suggest that walking activity can be improved energy balance. Further investigation is needed to determine whether different combinations of distance, rate or doing in morning or afternoon will stimulate energy balance.
The main objective of this experiment was to investigate effects of two different feeding systems on body weight, milk yield, milk composition, and mineral and fatty acids content of Holstein dairy cows’ milk. Sixteen of 25 months-old Holstein dairy cows were assigned to two groups (n=8) to study effects of the feeding system for 150 days. Two feeding systems were compared for five months; Group 1 was housed indoors and mainly fed a concentrate diet, Group 2 was maintained outdoors for five-seven hours/day on various kinds grass in a pasture. The experiment was conducted June-October 2017. Results revealed the indoor-fed cows had higher body weight, that was significant compared with the outdoor-based feeding system of Holstein dairy cows (p<0.05). Indoor-raised milking cows had higher milk yield (32.45 kg) as compared with pasture-raised milk yield (26.44 kg). Cows fed indoors significantly increased milk yield, total protein content, lactose, citric acid level, and lowered level of total solid and free fatty acids relative to the pasture-fed milking cows (p>0.05). There were higher levels of mineral content and fatty acid content in the milk of indoor-fed dairy cows than the pasture-raised dairy cows (p>0.05). Our study results demonstrated the potential benefits of the indoor feeding system for increased body weight, milk yield, mineral and fatty acids content summer through autumn when low pasture growth rates and quality may otherwise limit production.
To solve the problem of raw milk surplus, the Korean government encouraged the farmstead milk processing industry. However, the hygiene of farmstead dairy products has not been evaluated. Therefore, the microbiological analysis of farmstead cheeses was performed in this study. Farmstead cheeses (Berg, Colby, Cottage, Gouda, Mozzarella, String, Tilsiter, and Quark) were purchased from 16 dairy farms. In qualitative analysis, the presence of foodborne pathogens (Bacillus cereus, Escherichia coli, Clostridium perfringens, Salmonella, Staphylococcus aureus, Listeria monocytogenes, and Campylobacter spp.) were examined. Also, total aerobic bacteria, Pseudomonas spp., coliform, and E. coli, and yeast/mold were enumerated. Seventeen samples (37.8%) of 45 farmstead cheeses were contaminated with B. cereus and the highest detection rate was observed in String cheese. Two samples (4.4%) were E. coli positive and seven samples (15.6%) were S. aureus positive. Four other foodborne pathogens were not detected in all farmstead cheeses. Also, the mean levels of total aerobic bacteria, coliform, E. coli, and yeast/mold were 4.3 Log CFU/g, 1.4 Log CFU/g, 1.1 Log CFU/g, and 3.8 Log CFU/g, respectively. These results indicate that the food safety of farmstead cheese is extremely poor and, thus, the hygiene management of farmstead cheese should be improved to provide safe farmstead cheese to consumers.
Although milk, which is a representative western food, has a more than 100-year history in Korea, the preference is controversial. This study examined the milk history of Choseon in 1884~1938. This period was divided into 4 periods regarding the westerner’s records and advertisements. Westerners who visited Choseon in 1884~1895 (1st period) recorded the eating habits of Choseon,i.e., no milk consumed, even in insufficient food situation. Among the westerners, medical missionaries began to show or/and recommend condensed milk for sick children. In 1896~1909 (2nd period) newspapers, general shops in Hanseong for westerners showed advertisements of dairy products. In the 1900s, condensed and raw milk were advertised through newspapers. Domestic ranching systems to produce raw milk were established at that time mainly by Japanese. In the 1910s (3rd period), raw milk and condensed milk were advertised in newspapers. Since the mid-1920s (4th period), dairy products were bisected into condensed and powdered (dried) milk. Moreover, many Japanese manufacturers appeared in the advertisement in the 1920s. These results suggest that milk has been recognized as a symbol of an enlightenment food in Korea since the late 1900s, but the old negative wisdom, unfamiliar taste, and high price of milk at that time limited its appeal.
본 연구는 홀스타인 젖소 201,327두의 1산에서 5산까지의 검정기록 총 388,747개를 이용하여 유생산 형질에 대한 유전모수를 추정하고자 수행되었다. 1산차의 검정기록이 전체자료의 51.8%를 차지하였으며, 산차가 증가할수록 검정기록의 수는 현저히 감소하였다. 1산차의 유량, 유지방량과 유단백량에 대한 유전력은 각각 0.28, 0.26과 0.23으로 추정되었으며, 산차가 증가할수록 유전력은 일괄적으로 감소 하였다. 유량, 유지방량, 유단백량에서 산차간 유전상관계수의 범위는 0.83~1.00, 표현형상관계수의 범위는 0.21~0.51로 유전상관계수보다 항상 낮게 추정되었다. 연간 표현형 개량량은 유량, 유지방량과 유단백량에서 각각 121.50kg, 4.45kg과 4.65kg, 연간 유전적 개량량은 각각 13.70kg, 0.33kg과 0.36kg이었다.
This study attempted to establish an ultra performance liquid chromatography (UPLC) analysis method for standard determination of silymarin as a health functional food material in Silybum marianum extraxt (milk thistle). UPLC was performed on a Waters Acquity BEH C18 (50×2.1 mm, 1.7 μm) column using a gradient elution of distilled water and methanol at a flow rate of 0.21 mL/min and detection wavelength of 288 nm. The UPLC method showed high linearity in the calibration curve at a coefficient of determination (r2) of >0.9999, and limit of detection and quantitation for 6 flavonolignans were 0.0167-0.2469 and 0.1648-1.2931 μg/mL, respectively. The recovery of each flavnolignan was in the range of 99.96-100.81%, and the relative standard deviation for precision of each flavonolignan was less than 1.0%. The UPLC method established in this study was more specific for the quantitative determination of silymarin than the HPLC method. Also, since the UPLC method is shorter in the equipment operation time and smaller in the amount of used solvent than the HPLC method, UPLC is expected to have higher energy efficiency and lower environmental impact compared with HPLC.
A diverse of recommendation has been made for the structure and management of dairy cows, despite demanding research, the relationship between lactation number and various factors is yet to be established. The present study was aimed to investigate the covariance among lactation number, growth performance, calving interval, and milk production was considered to increase an efficiency of selection schemes and to manage more efficiently Holstein dairy cows that have been raised on small-scale family farms in Republic of Korea. For that purpose, the data were observed from 850 Holstein dairy cows, which a total of 3929 milking, since April 2016 - January 2017. We measured the body weight, height, age, calving interval, and milk production of the each dairy cow. Also, information about the date of lactation, calving interval, and milk production was recorded using an automatic milking system(AMS) with identification numbers. Milk production was calculated per udder quarter in the AMS. Our study results showed the increased average body weight(p>0.05) in 1, 2, 3, and 4th lactating dairy cows and afterwards, we noticed the tendency on the average body weight(p<0.05) per lactation progressed. There was no significant difference noticed on height measurement of dairy cows. From the processing data of 850 Holstein dairy cows, the lactation number 1 and 7 had a greater calving interval with significantly lowered milk production, and the lactation number 2, 3, 4, 5, and 6 had significantly lowered the calving interval(p<0.05) with a greater milk production. From our study results, we evidenced that there is a significant relationship between the lactation number, growth performance, calving interval, and milk yield, and the maximum production of milk occurring in the 3rd and 4th lactation dairy cows. The achieved results from this study can be used by the small-scale farmers to encourage the structure and management of growth performance, calving interval, and milk yield in Holstein dairy cows in Korea.
본 연구는 우유 중에서 덱사메타손을 효과적으로 정량 분석하기 위한 LC-MS/MS법을 확립하고 이를 검증하기 위해 수행되었다. 확립된 LC-MS/MS에 대해 특이성, 검출 한계, 정량한계, 정확도 및 정밀도에 대한 검증을 통하여 유효성을 확인하였다. 표준용액을 이용하여 검량성을 작성한 결과, r2 > 0.999 이상의 직선성을 확인하였고, 덱사 메타손에 대한 검출한계와 정량한계는 각각 0.15와 0.5 ng/ mL이었다. 또한, 회수율은 98.9-109.6%로 나타났으며, 상대표준편차는 1.7-4.4%로 나타나 정확성이 우수하였으며, 이는 식품의약품안전처의 잔류동물용의약품 분석법에서 제시한 기준에 모두 적합한 수준이었다. 따라서 본 연구를 통해 개발된 LC-MS/MS법은 향후 우유 중 덱사메타손을 분석하는데 효과적으로 활용될 수 있을 것으로 사료된다.
In order to verify the quality characteristics of soybean milk added chickpeas, the following characteristics were investigated: pH, solid contents, color, DPPH radical scavenging, as well as electric nose and sensory evaluation. Physicochemical and the sensory characteristics were analyzed based on the experimental data. The pH value was different in the control and the treatments (p<0.005). As the quantity of chickpea content increased, the solid content was augmented (p<0.0001). The L value was 56.86 in the control, and with the amount of chickpea addition increasing, the L value increased to 57.43 in 100% chickpea soybean milk (p<0.0001). The a value and b value also increased significantly (p<0.0001). However, the DPPH radical scavenging in the control was the lowest but the antioxidant activity of 100% chickpea milk was more than 2.5 higher than that of the control (p<0.0001). In the electric nose experiment, the flavor component of 20%, 30% and 100% chickpea treatment showed a significant difference compared to the control in the flavor components. In the sensory evaluation, for the score of flavor (p<0.001) and taste (p<0.0001), the score was higher in the treatments where 20% and 30% of chickpeas were added. In the sensory test of texture, there was no significant difference in the different experimental conditions except for the 100% chickpea addition treatment. In the overall acceptability test, the scores of 20% and 30% chickpea treatment were the highest results, compared to other treatments (p<0.0001). According to the correlation analysis, both antioxidant activity (0.797) and solid content (0.834) had shown high correlation to pH among the physiochemical characteristics (p<0.01). In the sensory evaluation, color, flavor, taste, texture and overall acceptability had shown a positive correlation to the amount of the soy bean milk added chickpea (p<0.01). In particular, the overall acceptability had shown the highest correlation to the taste (0.803), and it was the texture which resulted in the next highest correlation for overall acceptability (0.666).
Lignosus rhinocerotis, commonly known as Tiger Milk Mushroom, has been long extolled for its medicinal properties and used for treatment of asthma, cough, fever, cancer, liver-related illnesses, joint pains and as tonic. The history of usage for Tiger Milk Mushroom dates back to almost 400 years, ago but there were no records of scientific studies done due to inavailability of sufficient samples. Even when there were samples collected from the wild, the supply and quality was inconsistent. With the advent of cultivation success of one of the most utilized species of Tiger milk mushroom (L. rhinocerotis) in 2009 (known as TM02), scientific investigation was done to validate its traditional use and to investigate its safety for consumption, biochemical and biopharmacological properties. Preclinical toxicology evaluations showed that TM02 did not induce adverse effects on fertility. It also poses neither teratogenic effects nor genotoxicity. The no-observed-adverse-effect level (NOAEL) dose of the TM02 in 180-day chronic toxicity study is 1000 mg/kg which is equivalent to 162 mg/kg human dose, anticipated dose for a 60 kg adult is ~10g. The effective dosage of TM02 is as low as 0.5g per day for an adult. There is therefore, a huge margin for the safe consumption of TM02. TM02 is proven to be effective in supporting the lung and respiratory health, immune health, maintenance of joint health (caused by inflammation), improving vitality and stamina and improving the quality of life for cancer patients. Recent scientific findings have shown that TM02 contains various active components such as the polysaccharides-protein complexes, β-glucans, proteins, immuno-modulatory proteins all of which could play possible roles in rendering significant therapeutic properties such as anti-inflammatory, immuno-modulatory, anti-proliferative, anti-oxidative, neurite outgrowth stimulation etc. The revelation of the genome, transcriptome, proteome of L. rhinocerotis by MMRG has provided valuable insights into the biomolecule discovery and provided the foundation for future research and exploitation of L. rhinocerotis in pharmacological and functional food applications. These data forms a valuable foundation for future research in the exploitation of the L. rhinocerotis in pharmacological and industrial applications.
This study was conducted to examine the effect of corn (Zea mays L.) - soybean (Glycine max L.) silage prepared by intercropping method on the nutritive value of the silage, in vitro rumen fermentation characteristics, dry matter degradability, as well as milk yield and milk composition of dairy cows. In a couple of experiments intercropped corn-soybean silage (CSBS) was compared with corn silage (CS) and/or Italian ryegrass hay (IRG). Numerically, CSBS had higher crude protein, ether extract, and lactic acid contents compared to CS. In vitro rumen fermentation analysis demonstrated that up to a 24-h incubation period, both CS and CSBS showed higher total gas production, ammonia N concentration, and dry matter degradability compared to IRG (p<0.05). The investigation on animals was conducted in a commercial dairy farm located in Gyeongju, South Korea, employing 42 Holstein cows that were divided into 2 group treatments: CS and CSBS in a completely randomized design. Although no significant difference was observed in milk yield, animals fed on CSBS showed significantly higher milk protein (p<0.05) and milk fat content (p<0.01), compared to animals fed on CS. Taken together, our findings indicate that corn-soybean silage that is cultivated, harvested, and prepared through intercropping can improve the protein content of the silage, and can also enhance in vitro rumen fermentation, dry matter degradability, and performance of dairy cattle.
This study was designed to investigate the carryover effects of high-forage to concentrate (F: C) diet in bred heifers on feed intake, feed efficiency (FE) and milk production of primiparous lactating Holstein cows. The experiment was conducted for 589 days (d) from onset of pregnancy through to the end of first lactation. Twenty-four bred heifers (Body weight: BW= 345.8 ± 45.4 kg and 15 ± 1.2 mon of age) randomly assigned to two groups of 3 pens containing 4 heifers each and fed high forage (HF) diet with F: C ratio of 91.7: 8.3% and low forage (LF) diet with F: C ratio of 77.8: 22.2% throughout the pregnancy period. After calving, lactating cows were fed total mixed ration (TMR) based diet. No differences (p > 0.05) were observed in dry matter intake (DMI) of bred heifers and primiparous lactating cows in both HF and LF groups. The FE of mid-to-late lactation period was higher (p< 0.05) in HF than LF group. However, the HF group showed higher (p < 0.05) milk yield, 4 % fat corrected milk (FCM) and energy corrected milk (ECM) than LF group during the 305 d lactation. The LF group showed higher (p < 0.05) milk fat, crude protein (CP), milk urea nitrogen (MUN), solid not fat (SNF) and somatic cell count (SCC) than HF group. It is concluded that restriction of F: C ratio to 91.7: 8.3% to bred heifers has the potential carryover effects to maintain higher milk yield and FE with no adverse effect on feed intake and milk composition of primiparous lactating Holstein cows.
This study was conducted to investigate effects of feeding fermented milk on growth, intestinal microorganisms and fecal noxious gas emission in suckling pigs. A total of a hundred birth piglets (Landrace×Yorkshire×Duroc) were randomly assigned into feeding group and control group during suckling period that ten pigs per sow. Fermented milk contained 3.0×108/g of Bacillus and 3.5×108/g of Lactobacillus, and was supplied by top dressing method. Fermented milk fed to the sulking pigs indicated tendency to increase weaning body weight (p=0.052) and average daily gain (p=0.094). Total microbial flora and Escherichia coli in the feces were lower (p<0.05) in the feeding group than the non-feeding group. Reversely, Lactobacillus was higher (p<0.01) in feces of the pigs fed fermented milk than the pigs of the control group. Hydrogen sulfide emitted in feces was decreased in feeding group compared with control group (p<0.05). Similarly, fecal total mercaptans was diminished in the feeding group than the control group (p<0.01). Therefore, the fermented milk fed to the sulking pigs may improved growth and can influence positively intestinal microorganisms and fecal noxious gas emission.
본 조사연구는 충남 예산지역에 위치하고 있는 젖소 사 육농가 중 10개 전업 검정농가를 대상으로 2015년 1월부터 2015년 12월까지 농협중앙회 젖소개량사업소의 검정성적 을 통하여 유량 및 유조성분을 조사·분석하여 젖소농가의 경영지표 설정과 농가의 경영기술 개선을 위한 기초적 자 료를 얻고자 실시한 결과를 요약하면 다음과 같다. 평균 산유량은 33.62kg로 나타났다. 산차별로는 유의한 차이 (p<0.05)를 보였으며 1산차는 29.60kg로 가장 낮은 산유량 을 나타났으나, 2산차 이상부터 높은 산유량을 보였다. 산 유량별로도 유의한 차이를 보였으며(p<0.05), 계절별 평균 산유량은 봄에 34.51kg으로 다른 계절보다 높은 유량을 보 였다(p<0.05). 산차별의 유지율은 유의성 없이 1산에서 6산 차까지 비슷한 결과를 보였다. 산유량별 간에 유지율은 유 의한 차이(p<0.05)가 있었고, 계절별 간에는 유의한 차이 (p<0.05)를 보였는데, 겨울에 유지율 4.15%로 가장 높은 성 적을 보였으나, 여름에 3.61%로 가장 낮은 함량을 보였다. 산차별 간의 단백율은 유의한 차이 없이 평균치와 비슷한 수준을 보였다. 산유량별 간의 유단백율은 유의한 차이 (p<0.05)로 1일 30kg미만 군에서는 3.51% 높은 유단백율을 보였고, 40kg이상 군은 3.14%로 가장 낮은 함량으로 나타났다. 계절별 간의 유단백율은 가을과 겨울은 각각 3.40% 와 3.41%으로 높았으나, 봄과 여름은 각각 3.28%와 3.27% 로 낮은 수준를 보였다(p<0.05). 총고형분은 산차별 간에 1 산차가 8.92%로 가장 높은 함량를 보였으나(p<0.05), 산차 가 커질수록 감소현상을 보였다. 산유량별에 따른 총고형 분율은 1일 30 kg미만 군에서 8.92%로 높았으나, 40kg이상 군은 8.62%로 낮은 총고형분율을 나타났다(p<0.05). 계절 별 간의 총고형분율은 가을과 겨울에 각각 8.87%와 8.84% 으로 높은 함량이 었으나, 봄과 여름에는 각각 8.77%와 8.75%로 낮았다(p<0.05). 산차별간의 체세포수는 1산차가 100.75천개/ml로 가장 낮은 수를 나타냈으나, 산차가 커질 수록 체세포수는 증가현상을 보였다(p<0.05). 산유량별로 는 1일 30kg미만 군은 157.00천개/ml의 높은 체세포수로 유의한 차이를 보였으나(p<0.05), 40kg이상 군의 123.64천 개/ml로 낮은 수로 나타났다. 산유량은 유지율, 유단백율 및 총고형분율과 각각 -0.378, -0.534 및 –0.370으로 음(-)의 상관계수를 나타냈다. 유지율은 유단백율 및 총고형분율과 의 상관계수는 각각 0.600과 0.493으로 매우 높은 양(+)의 관계를 보였다. 유단백율과 총고형분율 간에는 0.833으로 양(+)의 매우 높은 상관관계를 보였다. 산유량과 유지율의 단순회귀모형은 y=-0.0297x+4.907(R2=0.1426), 유지율과 유 단백율은 y=0.2824x+2.2482(R2=0.3606), 유단백율과 총고형 분율은 y=0.8564x+5.9404(R2=0.6942)를 나타냈다. 따라서 젖소의 산유량과 유성분은 계절적 요인, 그리고 체세포수 는 산차와 산유능력에 영향을 받는 것으로 나타나 적절한 젖소의 산유량과 유질개선에 각별한 관리가 요구된다.
본 연구는 「축산물의 가공기준 및 성분규격」에 기준 규격은 설정되어 있지 않으나, 국제 기준과의 조화를 위해 규격 신설 검토 대상 성분인 비오틴의 분석법을 마련하고자 수행하였다. 조제유류에 미량으로 함유된 비오틴함량 분석을 위해 선택성과 정밀성이 뛰어난 LC-MS/MS를 이용한 분석법을 확립하고 시중에 유통중인 제품을 대상으로 적용성을 검토하였다. 비오틴 표준품을 이용하여 LC-MS/MS를 이용한 기기분석조건을 확립하고, 식품의 기준 및 규격 분석법을 참고로 0.01 M 인산이수소칼륨(pH 4.8) 용액을 이용하여 시료중의 비오틴을 추출하여 분석하였다. 분석법 검증은 특이성, 직선성, 검출한계 및 정량한계, 정확성, 정밀성에 대해 수행되었다. 5~60 ng/mL의 농도범위에서 R2 = 0.999 이상의 우수한 직선성을 확인할 수 있었으며, LOD와 LOQ는 각각 0.10, 0.31 ng/mL 이었다. CRM (NIST SRM 1849a) 및 표준물질 첨가법을 이용하여 정확성을 검토하였으며, CRM에서 103%, 조제분유에서 101~104%, 조제우유에서 99~101%의 회수율을 확인할 수 있었다. 정밀성을 검토한 결과 시료 채취량에 따른 상대 표준편차가 조제분유 0.4~0.9%, 조제우유1.4~1.6%로 확인하였으며, 실험일자에 따른 재현성은 조제분유에서 1.3%, 조제우유에서 1.2%로 확인하였다. 본 연구에서 확립된 분석법을 적용하여 조제분유 39건, 조제우유 3건, 성장기용조제분유 23건 등 국내 유통중인 조제유류 65건에 대해 적용성 검토를 실시한 결과 전체 시료에서 분석이 용이하였으며, 모두 표시기준에 적합함을 확인하였다.
