        이 연구는 복분자에서 분리된 Lactobacillus plantarum stain (GBL16 및 17)을 포함한 probiotic에 대해 수행되었으며, 장내 미생물 변화를 확인하였다. Balb/c 마우스모델에서 GBL 16, 17을 포함한 시제품인 PMC와 시판중인 유산균 과자 BFY를 8주간 경구 투여 하였다. PMC의 투여는 장내 미생물총의 균형을 향상시키고, 장내개선에 긍정적인 영향을 보였다. 맹장을 이용한 NGS 분석을 통해 확인한 결과, 스트레스 후 증가하는 양상을 보이는 균인 Lachnospiraceae균이 감소하는 경향을 보였다. Alistipes균의 증가는 염증성 장 질환에 효과가 있을 것으로 판단되나, 추후 더 많은 연구가 필요할 것으로 보인다.

        본 연구는 부패미생물의 종류와 차이를 알아보기 위해 유기농산물과 일반농산물(깻잎, 양배추, 상추)을 구입하여 4oC에 14일 저장한 후 일반목적배지를 사용하여 미생물을 분리 및 16S rRNA sequencing방법을 이용하여 동정하였다. 유기농산물 깻잎, 양배추, 상추의 총균수는 5-7 log10 CFU/g 수준이었고, 일반농산물의 총균수는 5-8 log10 CFU/ g 수준이었다. 유기농산물보다는 일반농산물이 총균수를 더 많이 내재하고 있으나 유의적인 차이는 나타나지 않았 다(p > 0.05). 유기농산물 깻잎에서 발견된 주요 균은 Microbacterium sp.이었으며, 일반농산물 깻잎에서 발견된 주요 균은 Stenotrophomonas maltophilia이었다. 또한, 유기농산물에서 부패를 일으킬 수 있는 균인 Klebsiella pneumonia 가 발견되었으며, 일반농산물보다 유기농산물에서 발견된 균의 종류가 더 많은 것을 확인할 수 있었다. 이에 본 연구 결과는 부패미생물에 대한 이해를 높이고 농산물의 신선도 및 유통기한을 늘리기 위한 새로운 살균소독제 및 부패 억제제를 개발하는데 좋은 기초자료로 사용될 수 있을 것으로 사료된다.

        To evaluate the indoor air quality of food manufacturing plants, the presence of viable bacteria and fungi was assessed in the indoor air of the facilities at which 9 food items were manufactured. Air samples were collected from the general zone, low clean zone and clean zone of each factory with an air sampler, in combination with plate counts agar using for bacteria, and dichloran-glycerol agar for fungi. The samples were incubated at 25℃for 4 to 7 days. After culture, the colony forming units (CFU) on each plate were counted and corrected with a positive hole conversion table. The average concentration of bacteria was 2.2 × 10³ CFU/㎥ in the general zone, 1.2 ×10³ CFU/㎥ in the low clean zone and 7.3 × 10² CFU/㎥ in the clean zone. The average concentration of fungal microbes was 2.5 × 10³ CFU/㎥ in the general zone, 2.6 × 10³ CFU/㎥ in the low clean zone, and 2.0 × 10² CFU/㎥in the clean zone. No meaningful differences were detected between the general zone and the low clean zone, but the clean zone had significantly lower concentrations than the other zones. Additionally, the identification of the fungi was performed according to morphological method using a giant culture and slide culture. The fungi were identified as belonging to 18 genera, and the genera Cladosporium(33%), Penicillium(29%) and Aspergillus(26%), predominated. Aspergillus isolates were identified to species level, and A. ochraceus, a mycotoxigenic species, was identified. As part of the effort to control the quality of the indoor air of food manufacturing plants, our results show that continued studies are clearly warranted.

        After infection by Gram negative bacteria, neonatal patients or patients in intensive care unit usually suffer from endotoxin shock, leading to death finally in spite of eradication of bacteria by potent antibiotics. The purpose of this study that is the pilot study to prevent endotoxin shock and nosocomial infection, is to offer basic data on species and concentrations of airborne microbes in hospitals. Therefore, this study collected and analyzed articles and documents on airborne microbes in hospitals in Korea that were published in relevant journals from 1980 to 2004. Studies on the concentration of airborne microbes in hospitals as a part of study on the nosocomial infection have been performed from the latter half of 1970s. Pooled average concentrations of total suspended bacteria in hospitals by RCS air sampler and Cascade air sampler were 194.85 CFU/㎥ and 367.72 CFU/㎥, respectively. They were lower than the guideline concentration(800 CFU/ ㎥) of the Korean Ministry of Environment. Pooled average concentrations of total suspended fungi in hospitals by RCS air sampler and Cascade air sampler were 20.66 CFU/㎥ and 193.34 CFU/㎥, respectively. They were lower than the guideline concentration(150 CFU/㎥) of the World Health Organization(WHO). Pooled average concentrations of total suspended bacteria in operating room and intensive care unit(ICU) were 324.75 CFU/㎥ and 182.43 CFU/㎥, respectively. They again were lower than the guideline concentration of the Korean Ministry of Environment. Staphylococcus spp., Micrococcus spp., and Lactobacillus spp. for Gram-positive bacteria, and Pseudomonas spp., Klebsiella spp., and Escherichia spp., for Gram-negative bacteria were shown in hospitals. Also, Aspergillus spp., Penicillium spp., Mucor spp., Epidemophyton spp., Trichophyton spp., and Cladosporium spp. for fungi were found in hospitals. Staphylococcus spp. was shown frequently in operating rooms, intensive care units and wards. Penicillium spp. and Aspergillus spp. were shown commonly in them.

        In order to screen the antagonistic bacteria which inhibit the growth of the apple pathogen, Penicillum expansum, we isolated an effective bacterial strain and investigated into the antifungal activity of the antagonist and it's identification. The eleven strains of bacteria which strongly inhibited P. expansum were isolated from the nature, and the best antagonistic bacterial strain designated as CH142, was selected. The antagonistic strain CH142 was identified to be the genus Bacillus subtilis based on morphological and biochemical characterization. The CH142 showed 55.9% of antifungal activity against the growth of P. expansum. By the treatment of the culture broth and the heat treated culture filtrate of it, the B. subtilis CH142 showed 90% and 15% of antifungal activity, respectively.

        백김치를 담근 후 15℃에서 60일간 밀폐상태로 발표시키면서 발효일수 경과에 따른 pH, 총산함량 및 미생물수의 변화를 조사하고, 주요 미생물군집을 분리·동정하였다. 담금 즉시의 김치는 pH 6.15, 총산 0.03%였고, Gram 음성 간균류가 최우점 미생물이었으며 이들의 주요 속 구성은 Pseudomonas 55%, Enterobacter 40%, Erwinia 5%였다. 젖산균은 발효 2일 이후부터 최우점 미생물이 되었으며, 발효 4일, 12일 및 50일에 젖산균의 1차, 2차 및 3차 정상기를 각각 나타내었다. 젖산균 2차 정상기의 김치는 pH 3.51, 총산 0.59%로 적숙상태에 해당되며, 이 때 주요 젖산균의 종 구성은 Lactobacillus bavaricus 55%, Leuconostoc mesenteroides subsp. mesenteroides 42.5%, Leuconostoc paramesenteroides 2.5%였다. 젖산균 3차 정상기의 김치는 pH 3.40, 총산 1.10%로 과숙상태에 해당되며, 이 때 주요 젖산균의 종 구성은 Lactobacillus plantarum 65%, Lactobacillus brevis 35%였다. 젖산균들을 동정할 때에 Leuconostoc mesenteroides subsp. mesenteroides, Leuconostoc paramesenteroides. Lactobacillus plantarum 및 Lactobacilus brevis로 동정된 균주들은 기존의 분류체제에 비교적 잘 일치하였으나, Lactobacillus bavaricus로 동정된 균주들은 일치하지 않는 점들을 많이 나타내었다. 젖산균의 수가 증가할 때에 효모의 수가 감소하였고, 젖산균의 수가 감소할 때에 효모의 수가 증가하였다. 효모의 정상기는 젖산균의 제2정상기와 제3정상기의 사이인 30일에 나타났으며, 이 때 주요 효모의 속은 모두 Saccharomyces였다.

        Background : Ginsenosides, the main ingredient of ginseng roots can be confirmed various physiological activity such as anticancer, antioxidant, a natural ginsenosides is there a structure to be absorbed into the body does not work well absorbed through this process biologically active thus a high conversion ginsenosides. β-glucosidase enzyme is observed in several of the microorganism with an enzyme that serves to convert a ginsenoside prosper that is absorbed into the body. Methods and Results : To view a primary β-glucosidase activity, the bacteria were innoculated in esculin agar medium and the color change of the media were measured by the time and degree of changing color. In the other method, 5 mM of p-nitrophenyl-β-D-glucopyranoside (pNPG) containing 25 mM phosphate buffer solution (pH 7.0) was added to 50 ul enzyme solution. Then the solution was added to 50 ul reaction for 5 min at 30°C. The amount of p-nitrophenol liberated measured at 405 nm absorbance. The experimental results showed higher β-glucosidase activity in Pediococcus pentosaceus, Leuconostoc mesenteroide, Leuconostoc mesenteroides subsp. cremoris, and Paenibacillus polymyxa by using esculin agar medium method. Similarly in second method, β-glucosidase activity was higher in P. pentosaceus 402.32±11.43 unit/l, L. mesenteroide 353.73±14.64 unit/l, Lactobacillus sakei 198.4±15.47 unit/l Lactobacillus plantarum subsp. plantarum 164.1±8.12 unit/l. Conclusion : The result that the β-glucosidase activity was higher in P. pentosaceus, L. mesenteroide, and L. plantarum subsp. plantarum as compared to tested microbes. Therefore selected bacteria can be used in the industry of functioned foods and beverage to improve human healths.

        This study investigated the effect of electron beam (EB) treatment on the microbial reduction of dried laver (Porphyra tenera) and identified EB-resistant bacteria from the treated dried laver. After EB treatments of 4 kGy and 7 kGy, the numbers of total bacteria and EB-resistant bacteria were measured using tryptic soy agar and mannitol salt agar, respectively. The morphological and biochemical characteristics of each isolated EB-resistant bacteria were investigated and these bacteria were identified. Compared to the control (1.5±0.2)×106 CFU/g, the total bacterial number was significantly decreased to (5.4±0.5)×104 CFU/g and (1.1±0.6)×104 CFU/g after EB treatments of 4 kGy and 7 kGy, respectively. With a higher EB dosage, the number of red colonies was almost same, whereas the number of yellow colonies was significantly decreased to (3.3±1.2)×103 CFU/g and 0 CFU/g for 4 kGy and 7 kGy, respectively. All red and yellow colonies were gram-positive cocci, catalase-positive, and resistant to 3% and 5% NaCl media. From the 16S rDNA sequence analysis, yellow and red colonies were identified as either Micrococcus flavus or M. luteus, with 99% similarity for the yellow colonies, and Deinococcus proteolyticus and D. piscis, with 99% and 97% similarity for the red colonies, respectively.

        In this study, we isolated microorganisms from dried persimmon in Sangju and obtained 15 strains of microorganisms as the basic research to prevent the quality changes during drying and storage of dried persimmon. Contaminated microorganisms were separated using seven species of medium. Viable cell counts of dried persimmon from Sangju was 5.18×102∼1.68×107 CFU/g. Green mold K2-1 accounted for the highest percentage in the contaminated dried persimmons and identified as a major causative microorganism. Light violet and creamy yeasts were the second largest contaminated microorganisms. Green mold K2-1 strain was identified as Penicillium sp. and fungus K-1 and K-3 were identified as Caldosporium sp. and Aspergillus sp.

        Three biphenyl-degrading microorganisms were isolated from polluted soil samples in Sasang-gu, Busan. Among them, isolate DS-94 showing the strong degrading activity was selected. The morphological, physiological, and biochemical characteristics of DS-94 were investigated by API 20NE and other tests. This bacterium was identified as the genus Pseudomonas by 16S rDNA sequencing and designated as Pseudomonas sp. DS-94. The optimum temperature and pH for the growth of Pseudomonas sp. DS-94 were 25℃ and pH 7.0, respectively. This isolate could utilize biphenyl as sole source of carbon and energy. Biphenyl-degrading efficiency of this isolate was measured by HPLC analysis. As a result of biological biphenyl-degradation at high biphenyl concentration (500 mg/L), biphenyl-removal efficiency by this isolate was 73.5% for 7 days.

        The high surface area polymer, AquaMatsⓇ was used for the leachate purification process originated from the Samsan Dong Landfill in Ulsan Metropolitan City. And then, three species of dominant microbes were isolated and identified from AquaMatsⓇ. Gram staining revealed these microbes to be Gram-negative rod strains: They were identified as Agrobacterium radiobacter, Pseudomonas cepacia, Flavobacterium indologenes. All they showed no growth on media in which the leachate was added alone, but a rapid proliferation rate on media with yeast extract as nutrient requirements.
