        The purpose of this study was to examine the quality characteristics of cookies made with Angelica gigas Nakai fermented by Lactobacillus paracasei. As the content of Angelica gigas Nakai fermented by L. paracasei (AFL) powder increased, the pH of the cookies decreased from 6.3 to 5.2. There was no significant difference in the moisture content depending on AFL powder content. The content of reducing sugar also increased with increasing AFL powder content. Regarding the surface color of the cookies, the L- and b-values decreased with increasing AFL powder content, whereas the a-value increased. As the AFL powder content increased, the total polyphenol content and ABTS and DPPH radical scavenging activities significantly increased. Cookies with 8% AFL powder (AC8) had the highest levels of 107.16 mg%, 38.69%, and 65.56%, respectively. The texture, adhesiveness, and cohesiveness of the cookies with various AFL powder levels were not significantly different, and hardness, springiness, gumminess, and chewiness showed no tendencies toward changes. Taken together, these results showed that when AFL powder was added to cookies, bioactivities such as antioxidant activity increased, making AFL powder a good material with high potential for use in commercially baked products.

        Antioxidant properties and antioxidant activities were analyzed for water extracts and 50% and 70% ethanol extracts of the leaf of Angelica gigas Nakai. The polyphenol and flavonoid contents in water, 50%, and 70% ethanol extract of the leaf of Angelica gigas Nakai, it was found that the polyphenol contents were 18.75 mg GAE/g, 28.95 mg GAE/g, and 34.73 mg GAE/g, respectively, and flavonoid contents were respectively. The DPPH IC50 scavenging activity was 45.84 mg/mL, 36.44 mg/mL, 19.11 mg/mL, respectively, and theABTS+radical scavenging ability (1 mg/mL) was 28.73%, 22.79%, and 12.70%, respectively. Tyrosinase inhibitory activity, 70% ethanol extract, 50% ethanol extract, and water extract 33.14%, and 4.53%, respectively. Nitrite scavenging activity, 70% ethanol extract, 50% ethanol extract, and water extract were in the order of 1 mg/mL scavenging activity, 36.43%, 34.80%, and 18.85%, respectively.

        Angelica gigas Nakai (A. gigas) easily changes its color during storage, and appropriate thermal treatment can improve storage stability through inactivation of enzymes such as polyphenol oxidase. Therefore, this study was performed to determine quality characteristics of dried A. gigas in response to high-temperature-short-time (HTST) treatment during storage. Dried A. gigas were treated at 120-180℃ for 10 min, the samples were stored at 4℃ and 50℃ for 10 weeks, and used for the analysis of qualities. Concerning the color values, the sample treated at 120℃ was similar to the control, and the color change was large when treated above 180℃. However, color difference (△E* ab) was lower in treated samples than in control. Browning index was similar for all the samples except for the sample treated at 180℃. Functional qualities (phenolics content, antioxidant activities, and level of major components) showed a slight difference according to storage periods in all samples without control, and nodakenin content was observed in control. The results of this study showed that HTST treatment improved storage stability such as stability of colors and browning index in dried A. gigas during storage, and the appropriate treatment temperature was 120℃ in terms of stability in color and browning index.

        Method development and validation of decursin for the standardization of Angelica gigas Nakai as a functional ingredient and health food were accomplished. The quantitative determination method of decursin as a marker compound of aerial parts of Angelica gigas Nakai extract (AAGE) was optimized by HPLC analysis using a C18 column (3×150 mm, 3 μm) with 0.1% TFA in water and acetonitrile as the mobile phase at a flow rate of 0.5 mL/ min and detection wavelength of 330 nm. The HPLC/PDA method was applied successfully to quantification of the marker compound in AAGE after validation of the method with linearity, accuracy, and precision. The method showed high linearity in the calibration curve at a coefficient of correlation (R2) of 0.9994 and the limit of detection and limit of quantitation were 0.011 μg/mL and 0.033 μg/mL, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 1.10% and 1.13%, respectively. Recovery of decursin at 0.5, 1, 5 and 10 μg/mL were 92.38 ~ 104.11%. These results suggest that the developed HPLC method is very useful for the determination of marker compound in AAGE to develop a health functional material.

        Angelica gigas Nakai has been used for traditional Korean medicine which acts anti-inflammatories, protects the liver, and enhances the immune system. Subcritical water extraction (SWE) is an eco-friendly extraction method in which the pressure is applied between the boiling point and critical temperature of water that maintains the liquid state. The aim of study was to determine the optimal extraction conditions for subcritical water extraction of the decursin and nodakenin from Angelica gigas Nakai, comparing two scales, laboratory-scale (22 mL) and pilot-scale (8 L) devices to assess the feasibility of industrial applications. Extraction efficiencies were evaluated by measuring total polyphenol contents and DPPH radical scavenging activity with a spectrophotometer, extraction yields of decursin and nodakenin with a high-performance liquid chromatography. The subcritical water extraction conditions of pilot-scale were as follows: extraction temperature of 150, 170, 190 and 210°C; extraction times of 5, 10, 15, and 20 min. The highest yields of decursin and nodakenin were at 190°C/15 min (17.65 mg/g Angelica gigas) and 170°C/20 min (4.35 mg/g Angelica gigas), respectively. These were not significantly different from the maximum contents of decursin (13.49 mg/g Angelica gigas), and nodakenin (4.66 mg/g Angelica gigas) for lab-scale subcritical water extraction. The maximum values for total polyphenol content and DPPH scavenging activity of subcritical water extraction were 64.32 mg/g Angelica gigas and 74.4%, respectively. On the other hand, those of conventional extraction method using methanol (60°C/120 min) were obtained as 14.31 mg/g Angelica gigas and 41.9%. Therefore, the subcritical water extraction is a rapid and efficient method for extracting polyphenols and anti-oxidative compounds.

        We have hybridized Angelic gigas Nakai flower extract (AGNF) and two-dimensional layered double hydroxide (LDH) nanomaterials through reversible dehydration-hydration in order to obtain the nanopowder of natural extract. The Angelica gigas Nakai flower was treated with methanol to extract carbohydrate, polyphenol, and flavonoid components. LDH with an uniform size of 250 nm was prepared by hydrothermal method and calcined at 400ºC to obtain layered double oxide (LDO) precursor. For hybridization, AGNF in 40% methanol was reacted with LDO powder at various AGNF/LDO weight ratios: 0.15, 0.30, 0.85, and 1.70. The hybrids were obtained in fine powder which had enhanced hydrophilicity and water dispersity compared with dried AGNF. The X-ray diffraction and scanning electron microscopic results revealed that the house-of-cards structure of nanomaterials could encapsulate AGNF moiety inside their cavity. Quantitative analyses using UV-Vis spectra exhibited that the content of AGNF in hybrid increased upon AGNF/LDO ratio in reactant increased. According to 1,1-diphenyl-2-picrylhydrazyl radical scavenging assay, AGNF/LDO showed higher antioxidant activity compared with an equivalent amount of AGNF itself.

        본 연구는 생약재로 이용되는 참당귀의 다용도 기능성 소재 개발을 통한 이용가치를 높이기 위하여 효소처리를 활용한 기능성 다당체 분리방법에 대한 연구를 수행하였다. 참당귀의 세포벽 성분 분해를 위한 최적의 효소는 Viscozyme L로 선정되었다. 다당체 분리를 위한 효소처리는 단백분해효소(Alcalase 및 Flavorzyme)와 전분분해효소(Termamyl120L)를 Viscozyme L과 함께 복합적으로 처리한 VAFT처리구가 추출 수율과 총당 함량이 각각 12.20%, 76.80%로 다른 효소처리구(T, AFT 처리구)에 비하여 가장 높았으며, 제거된 전분 함량 역시 29.62%로 가장 높게 나타났다. 또한 비전분다당체 함량이 22.58%로 T 처리구의20.78%과 AFT 처리구의 21.98%에 비하여 유의적(p<0.001)으로 가장 높게 나타나 참당귀 다당체 분리를 위한 최적효소처리 조건으로 선정되었다. 비전분다당체의 주성분인arabinose, galactose의 함량은 대조구보다 T, AFT, VAFT의모든 효소처리구에서 유의적(p<0.001)으로 높게 나타났다.또한 참당귀 다당체의 분자량 분석을 통하여 대조구(491,000Da)에 비하여 VAFT 효소처리구가 13,000Da로가장 저분자화되는 것을 확인할 수 있었다. 따라서 VAFT효소처리에 의한 참당귀 기능성 다당류의 효율적인 분리방법은 참당귀를 활용한 새로운 산업적 기능소재 및 제품 개발에 활용 가능성이 높을 것으로 기대되었다.

        Currently, the storage temperature of the production factory of medicinal herbs (hGMP) is about 5~ 12℃. This temperature is possible to suppress pest but can not kill the pests. For this reason, we need to lower the temperature during lethal time because the pest has often occurred inside the packaging of medicinal herbs in the distribution process in summer. In order to solve this problem, we have investigated the lethal time of the storage insect(Tribolium castaneum Herbst) After we froze medicinal herbs(Cnidium officinale Makino and Angelica gigas N. ) at approx. –70℃ and –15℃ respectively. We then investigated the change of bioactive components and exterior characteristics of medicinal herbs in order to determine whether there is a change of quality. The results were as follows. The lethal time of Tribolium castaneum Herbst is about 2 minutes if processed at approx. –70℃, while the other time is about 16 minutes at approx. –15℃. We investigated the change of quality after the treatment of the two medicinal herbs in the similar way but could not confirm the difference of color and brightness and the bioactive components on statistics. Through this research, it has been verified that the process of quick freezing for pest control can not affect the main quality of the medicinal herbs. Therefore this technology can be introduced in the manufacturing process of medicinal herbs through additional research.

        Some biological activities such as an electron donating capacity, the contents of total polyphenol compounds and flavonoids, fibrinolytic activity and α-glucosidase inhibitory activity have been detected in hot water extracts of Ligularia fischeri and Angelica gigas Nakai. To increase the usefulness of the functional ingredients for prevention and improvement of some metabolic disorders, ethanol-treated hot water extracts of Angelica gigas Nakai were prepared. A hot water extract of Ligularia fischeri has 92% of electron donating capacity, 39.4 mg/g of total polyphenol compounds, 24.8 mg/g of flavonoids and 29.8% of α-glucosidase inhibitory activity, but no fibrinolytic activity. A hot water extract of Angelica gigas Nakai has 94.7% of electron donating capacity, 5.8 mg/g of total polyphenol compounds, 2.6 mg/g of flavonoids, 0.48 plasmin units of fibrinolytic activity and no α-glucosidase inhibitory activity. However, with partial purification using cold ethanol treatment, the α-glucosidase inhibitory activity of Angelica gigas Nakai was increased to 70.5%. Thus, we expected a more useful effect with the use of the addition of a cold ethanol-treated Angelica gigas Nakai extract. The L, b values of cold buckwheat noodles using a mixture of 0~3% of Ligularia fischeri powder and 0.5% of an ethanol-treated hot water extract of Angelica gigas Nakai were decreased with the addition of an increasing amount of Ligularia fischeri powder. Among the mechanical qualities, only adhesiveness was significantly higher in 3% Ligularia fischeri noodles. From sensory evaluation data, it was determined that these two functional ingredients did not ruin the color, texture, and overall acceptance of the cold buckwheat noodles. A higher amount of the extracts improved the quality of the product with little added cost.

        참당귀(Angelica gigas Nakai)는 미나리과(Apiaceae) 당귀 속(Angelica L.)에 속하는 다년생 식물로, 뿌리에 효능이 많아 약용 목적으로 재배된다. 본 연구에서는 주로 생산되는 참당귀 재배지의 특성에 대한 연구와 이에 따른 참당귀의 생장 및 지표 성분 함량과의 상관관계를 구명하여 참당귀의 안정적인 한약재 수급을 위한 생산량 및 유용성분 함량 증가에 적합한 재배 환경에 대한 정보를 제공하는데 있다. 참당귀의 근직경은 평창 두일리가, 지근 개수는 평창 탑동리가 가장 많았다. 생중량은 평창 탑동리가 가장 높았으며, 건중량도 이와 같은 경향을 보였다. 재배지의 토성은 사질식양토(Sandy Clay Loam)였으며, 유기물 함량은 탑동리에서 3.63%로 가장 높은 함량을 보였다. 전 질소 함량은 봉화 남회룡리에서 0.19%로 가장 높았으며, 유효인 산 함량은 고선리에서 높았다. 총 decursin 함량은 두일리에서 5.31%로 가장 높았다. 결과적으로 평창 두일리에서 참당귀의 생육이 가장 우수하였으며, decursin 함량 또한 높았다. 당귀의 수량이나 성분은 재배지역의 환경조건에 크게 영향을 받는데, 해발이 높은 지역에서 재배하는 것이 지표성분 함량과 생산량 증가에 유리하다고 사료되며, 환경조건과 생육 및 지표성분 함량과의 상관관계에 대한 기초자료가 될 것이라고 판단된다. 또한, 최적의 재배환경의 확립과 식물의 생리작용에 대한 연구의 기초가 될 수 있을 것이라고 판단된다.

        본 연구는 참당귀 재배 시에 발생하는 추대 참당귀를 부산물로 활용가능성이 있는지 평가하기 위해 수행하였다. 추대 참당귀의 지상부와 지하부의 총 무게는 수확기 비추대 참당귀의 지하부 무게 대비 약 98.5%로 높게 나타났다. 또한 추대 당귀의 decursin과 decursinol angelate의 총 함량은 주 생산품인 참당귀 신(身)의 지표성분 총 함량인 4.24%와 대비해 부위별로 0.64%~2.39%로 나타났으며, 이는 추출물로 이용 시 생산품 대비 15~56%에 해당하는 것으로 나타났다. 또한, 항상화 활성의 경우 추대 참당귀는 전반적으로 비추대 참당귀에 비해 우수한 것으로 나타났다. 추대 참당귀의 생산량, 지표성분 함량 그리고 항산화 활성 결과를 고려해 봤을 때, 추대 참당귀의 부산물은 사료 첨가제 및 화장품 용도의 추출물로 이용 가능성이 높은 것으로 생각된다.

        Angelica gigas Nakai (AGN) is a traditional medicinal herb especially in Korea. It contains pyranocoumarins, which are major active components including decursin (D) and decursinol angelate (DA). This study was carried out to determine the change in active component content and antioxidant activity depending on the root diameter of AGN. Several processing steps are involved to use AGN roots as medicine. The dried AGN roots are divided into body (B), thick root (TkR), medium root (MR) and thin root (TnR) according to their diameter before cutting into medicine. The recovery rates of each root parts per 100 ㎏ were measured as 32.3±2.5, 9.0±1.0, 39.3±2.1 and 15.0±1.0%, respectively and the mean diameters were measured as 51.95±4.55, 7.05±0.89, 2.88±0.49 and 1.57±0.32 ㎜. Two index components, D and DA, were analyzed. The change of both D and DA content showed a similar tendency. Both D and DA content were increased as the root diameter decreased (higher in TnR). In addition, antioxidant activity was higher in B and TnR, and lower in TkR and MR than control. This study showed that the thinner the root diameter, the higher the D and DA content in AGN roots and that TnR has excellent antioxidant activity compared to other root parts, suggesting that the thinner root part of AGN could be used as a potential material.

        점봉산 참당귀는 해발 750-965 m의 교목층은 들메나무, 고로쇠나무, 신갈나무가 분포하고, 아교목층에는 당단풍, 까치박달나무가 분포하는 계곡지로 유기물이 집적되기 쉬운 다습한 지형에 자생하였다. 참당귀 군락의 토양 분석 결과 토양pH는 4.9-6.0으로 우리나라 산림토양의 평균과 매우 유사한 값을 보였으며, 토양 양분의 지표인 유기물함량, 전질소 양이온치환용량, 치환성 Ca, K, Mg, Na의 함량은 산림토양의 평균보다 비교적 높은 값을 보였다. 생장 특성으로는 9월 말에는 뿌리 생장이 평균 47.9 g이나 10월 말에는 33.8 g으로 29% 감소하였으며, 유용 성분은 반대로 10월말에 decursin 3.8%, decursinol angelate 3.6%로 각각 26%, 67% 증가하였다. 참당귀 자생지 8개 집단의 생장 특성 및 유용성분 함량과 토양특성 간의 상관분석을 수행한 결과 뿌리 생장은 치환성 양이온인 K, Mg, Na과 높은 양의 상관관계를 보였고, 유용성분은 뿌리의 수분함량과 높은 음의 상관관계를 보였다.

        Background: The objective of this study was to make colloidal dispersions of the active compounds of radix of Angelica gigas Nakai that could be charaterized as nano-composites using hot melt extrusion (HME). Food grade hydrophilic polymer matrices were used to disperse these compound in aqueous media. Methods and Results: Extrudate solid formulations (ESFs) mediated by various HPMCs (hydroxypropyl methylcelluloses) and Na-Alg polymers made from ultrafine powder of the radix of Angelica gigas Nakai were developed through a physical crosslink method (HME) using an ionization agent (treatment with acetic acid) and different food grade polymers [HPMCs, such as HP55, CN40H, AN6 and sodium alignate (Na-Alg)]. X-ray powder diffraction (XRD) analysis confirmed the amorphization of crystal compounds in the HP55-mediated extrudate solid formulation (HP55-ESF). Differential scanning calorimetry (DSC) analysis indicated a lower enthalpy (ΔH = 10.62 J/g) of glass transition temperature (Tg) in the HP55-ESF than in the other formulations. Infrared fourier transform spectroscopy (FT-IR) revealed that new functional groups were produced in the HP55-ESF. The content of phenolic compounds, flavonoid (including decursin and decursinol angelate) content, and antioxidant activity increased by 5, 10, and 2 times in the HP55-ESF, respectively. The production of water soluble (61.5%) nano-sized (323 ㎚) particles was achieved in the HP55-ESF. Conclusions: Nano-composites were developed herein utilizing melt-extruded solid dispersion technology, including food grade polymer enhanced nano dispersion (< 500 ㎚) of active compounds from the radix of Angelica gigas Nakai with enhanced solubility and bioavailability. These nano-composites of the radix of Angelica gigas Nakai can be developed and marketed as products with high therapeutic performance.

        Background : The root of Angelica gigas Nakai is used as a traditional herbal medicine in Korea for the treatment of many diseases. Poor water solubility of the active compounds from A. gigas Nakai is a major obstacle to bioavailability. Methods and Results : This work aimed to enhance the solubility of active compounds by a chemical (viz. surfactant) and physical (HME, hot melt extrusion) crosslinking method (CPC) in A. gigas Nakai. Infrared fourier transform spectroscopy (FT-IR) revealed multiple peaks in extrudate solids representing new functional groups including carboxylic acid, alkynes and benzene derivatives. Differential scanning calorimetry (DSC) analysis of the extrudate showed lower glass transition temperature (Tg) and lower enthalpy (ΔH) [Tg: 43℃; ΔH: <6 (J/g)] compared to the non-extrudate (Tg 68.5℃; ΔH: 123.2) formulations. X-ray powder diffraction (XRD) analysis revealed amorphization of crystal materials in extrudate solid. In addition, enhanced solubility (53%), nanonization (403 ㎚), and higher amount of extracted phenolic compounds were achieved in the extrudate solid compared to non extrudate (36%, 1499 ㎚, respectively). Among the different extrudates, acetic acid and span 80 mediated formulations showed superior extractions efficiency. Conclusion : We conclude that the HME successfully enhanced the production of amorphous nano dispersions of phenolic compound including decursin from extrudate solid formulations. human dermal fibroblasts, which is, associated with the regulation of procollagen biosynthesis resulting from AMRP-induced TGF-β1 expression and the mitogenic activity in HDF cells, and therefore, is expected to reduce the age-dependent loss of extracellular matrix proteins.

        Background : A major medicinal plant, Angelica gigas Nakai, have using root parts and mainly cultivated the cool temperature region of high alpine as wild type in the northeast of China, Japan and Korea. The roots of grow it’s rapidly form August, and harvested from October to November. This study performed that the growth characteristics on soil mulching vegetation cultivation field of 300 m altitude of under Larix leptolepis (S. et Z.) Gordon in Bonghwa-gun, Gyeongsangbuk-do. Methods and Results : After one-year seeding of the same size (7 - 8 ㎜) were transplanted in three types soil mulching of cultivation (plastic, straw and non-mulching) overground growth surveyed in three times (May to August), To confirm of soil condition compared the average. Soil condition of plot were observed to pH 5.56, OM 3.22%, T-N 017%, and growth increment were higher observed to that straw and non-mulching then plastic mulching of such as plant length 46.8 ㎝, stem diameter 5.9 ㎜, leaf length 24.4 ㎝ and width 28.9 ㎝, number of radical leaf 3.1. But from August, when the average temperature was high, the growth characteristics such as plant length, petiole Length, leaf length and width were reversed compared to those of plastic and non-mulching. Conclusion : In case of cultivation of A. gigas in the environmental conditions of mountainous area, considering the growth from August to October when the roots growth bigger, It suggested that strew mulching is better than plastic and non-mulching, would be done through further research on underground growth characteristics of A. gigas according to harvesting time confirmed the relationship to the growth characteristics of A. gigas according to environmental conditions and soil mulching mountainous plantations.

        Background: The root of Angelica gigas Nakai is used as a traditional herbal medicine in Korea for the treatment of many diseases. However, the poor water solubility of the active components in A. gigas Nakai is a major obstacle to its bioavailability. Methods and Results: This work aimed at enhancing the solubility of the active compounds of A. gigas Nakai by a chemical (using a surfactant) and physical (hot melt extrusion, HME) crosslinking method. Fourier transform infrared spectroscopy revealed multiple peaks in the case of the extrudate solids, attributable to new functional groups including carboxylic acid, alkynes, and benzene derivatives. Differential scanning calorimetry analysis showed that the extrudate soilid had a lower glass transition temperature (Tg) and enthalpy (ΔH) (Tg : 43℃, ΔH : < 6 J/g) as compared to the non-extrudate (Tg : 68.5℃, ΔH : 123.2) formulations. X-ray powder diffraction analysis revealed the amorphization of crystalline materials in the extrudate solid. In addition, enhanced solubility (53%), nanonization (403 ㎚), and a higher amount of extracted phenolic compounds were achieved in the extrudate solid than in the non-extrudate (solubility : 36%, nanonization : 1,499 ㎚) formulation. Among the different extrudates, acetic acid and span 80 mediated formulations showed superior extractions efficiency. Conclusions: HME successfully enhanced the production of amorphous nano dispersions of phenolic compound including decursin from extrudate solid formulations.

        Background: Angelica gigas Nakai is a perennial herb belonging to the family Umbelliferae. Its roots are utilized in traditional medicine. The aim of this study was to increase the yield of and the content of two indicator components (decursin and decursinol angelate) in A. gigas Nakai. Methods and Results: The roots of A. gigas Nakai were harvested 4-times from late August to late October in 2017. Two agents (trace element-TE, and plant hormone-HM) were applied 4 times at intervals of 2 weeks. The content of the two indicator components were analyzed by high performance liquid chromatography. The HM treatment showed the greatest increase in underground part yield and root diameter. The content of the two indicator components in the control (non-treatment) group was the highest in the underground part, but was higher in the aerial parts in the agent treatment group. After treatment with the agents, the content of the indicator components tended to decrease in the underground part. However, the total content of the indicator components in the two agent treatment groups exceeded the level of 6% set by the Korean Pharmacopoeia. Conclusions: The highest underground part yield was found in the HM treatment group, while the highest content of decursin and decursinol angelate were found in the control group. This study provides basic information for yield improvement in A. gigas.
