        본 연구에서는 쥐참외뿌리 추출물의 항산화 활성 및 미백, 항염증 활성에 대한 생리활성 효과와 기능성 소재로써의 활용 가능성을 확인해 보고자 하였다. 쥐참외뿌리 추출물의 DPPH 라디칼 소거 활성을 통한 항산화 활성, 멜라닌 세포인 B16F10 melanoma 세포에 대한 멜라닌 생성 억제능을 통한 미백 활성 효과, 대식 세포인 RAW 264.7 세포에 대한 NO 생성 억제능을 통한 항염증 활성 효과를 확인해 보고자 하였다. 연구결과, 쥐참외뿌리 추출물은 농도 의존적으로 DPPH 라디칼 소거활성이 확인되었으며, 양성 대 조군인 Ascorbic acid와 비슷한 DPPH 라디컬 소거활성이 확인되었다. 100 nM α-MSH로 유도된 멜라닌 생성 억제 활성과 LPS 1 ㎍/mL로 유도된 NO 생성 억제능을 유의하게 억제 하는 것을 확인되었다. 이에 따라 쥐참외뿌리 추출물은 항산화 및 미백, 항염증 효과를 가진 기능성 소재로써의 활용 가능성이 있음이 사료되어 진다.

        본 연구의 목적은 효소 가수분해 및 유산균 혼합발효기술을 이용하여 GABA 함량이 강화된 발효 다시마 추출물을 생산함에 있다. TLC 분석방법을 통해 GABA 생성능력이 우수한 균주 L. plantarum KCTC 21004, L. acidophilus KCTC 3164, L. sakei subsp. sakei KCTC 3598를 선정하였 다. 선정된 균주를 이용한 유산균 발효의 특성은 배양 시간이 증가 할수록 적정 산도는 증가하였고 pH 는 감소하는 경향을 나타내었다. GABA 생성능력이 우수한 3종의 균주 중 L. plantarum KCTC 21004 이 GABA 함량이 136.4 mg/100g으로 우수하였다. 이러한 연구 결과는 유산균을 이용한 GABA 생산과 정에 있어서 기초적인 자료로 제공될 것으로 판단된다.

        The purpose of this study was to confirm the possibility of using Korean purple yam (Dioscorea alata) as proximate composition, material, and antioxidant activity. In the proximate composition of the freeze-dried purple yam powder, the carbohydrate content was the highest at 86.67%, and in minerals, potassium showed the highest content at 1,765.69 mg/100 g. To study the antioxidant activity of purple yam, distilled water and 70% ethanol were used as extraction solvents. The total polyphenol, total flavonoid, and total anthocyanin contents were 1.3~1.6 times higher in 70% ethanol extract, than in the distilled water extract. As a result of ORAC, DPPH and ABTS radical scavenging activity, SOD activity, and reducing power, the 70% ethanol extract showed higher antioxidant activity than the distilled water extract in all results. As a result of freeze-drying purple yam and measuring antioxidant activity by extraction solvents, it is concluded that purple yam can contribute to the food industry as a natural antioxidant and health functional material.

        The purpose of this study was to evaluate the antioxidative activities of jeolpyun containing Cnidium officinale M extract (2%, 4%, 6%, 8%) by total polyphenol contents, electron donating ability on 2,2-diphenyl-1-picrylhydrazyl (DPPH), scavenging ability of superoxide anion radical and decomposing ability of hydrogen peroxide. In chromaticity analysis, the brightness significantly decreased with increasing Cnidium officinale M extract content. Jeolpyun containing 8% Cnidium officinale M extract revealing the highest value for the redness and the yellowness, 1.07, 12.70, respectively. The total polyphenol contents of jeolpyun containing 8% Cnidium officinale M extract were the highest content of 4,213 μg gallic acid equivalent (GAE)/mL. The total polyphenol contents revealed significant difference (p<0.05). Jeolpyun containing 8% Cnidium officinale M extract revealing the highest electron donating ability (83.55%). The electron donating abilities were significantly related at p<0.05. The scavenging abilities of superoxide anion radical for jeolpyun containing 4% Cnidium officinale M extract revealed the highest ability (0.01676). There was no significant difference. The hydrogen peroxide decomposing ability for jeolpyun containing 8% Cnidium officinale M extract revealed the most hydrogen peroxide decomposing ability (-0.193) and the hydrogen peroxide decomposing ability revealed a significant difference (p<0.05).

        The study investigated the chemical components, antioxidant activities, and glucose-regulating enzyme activities of red chicory (Cichorium intybus L.). On a dry weight the contents of moisture, crude protein, crude fat, crude ash, crude fiber, and carbohydrate were 7.36, 28.0, 4.33, 19.63, 10.84 and 40.52%. The contents of mineral were Na (772.39 mg/100 g), K (5,686.03 mg/100 g), Mg (307.48 mg/100 g), Ca (804.94 mg/100 g), Fe (8.61 mg/100 g), and P (1,145.24 mg/100 g). Chlorophyll a, Chlorophyll b, total chlorophyll (793.80, 363.06, 1,156.86 mg/100 g,), total carotenoid (171.82 mg/100 g), and anthocyanin (79.49 mg/100 g) contents were measured. Antioxidant activities were analyzed for water and 70% ethanol extract. The polyphenol contents 23.67, 32.58 GAE mg/g, and flavonoid contents were 18.15, 28.76 RE mg/g, respectively. The DPPH scavenging activity IC50 was 694.48, 76.12, μg/mL, and the ABTS scavenging activity IC50 1,154.90, 566.00 μg/mL, respectively. In reducing power, the 70% ethanol extract showed a higher reducing power. In α-glucosidase and α-amylase inhibition activity IC50, the 70% ethanol extract (5.87, 17.91 mg/mL, respectively) showed a higher than water extract (8.87, 100 < mg/mL, respectively). This study is expected to provide basic data for developing functional food ingredients for red chicory.

        The leaves of Allium victorialis (AV) are known an edible perennial herb, which has been used in Korean traditional medicine. However, the beneficial pharmacological effects of AV extracts (AVE) on the antioxidant activity and atopic dermatitis (AD) have not been thoroughly examined. Therefore, the present study aims to investigate both antioxidant activity and anti-inflammatory effect of AVE on AD in vitro and in vivo. Antioxidant activity was evaluated by total polyphenol content and ferric reducing ability. AVE showed a level of polyphenol content and reducing power activity. The five-week-old BALB/c mice were used as an AD-like mouse model by treating them with 1-chloro-2, 4-dinitrobenzene (DNCB). Topical administration of AVE for 3 weeks to DNCB-treated mice significantly alleviated clinical skin lesion dermatitis severity and epidermal thickness. Histopathological analysis also demonstrated that AVE decreased eosinophil and mast cell infiltration into skin and ear tissue. These results suggest that topical application of AVE inhibits the development of AD-like skin lesion in mice by their antioxidant activity. Thus, AVE may be a potential therapeutic agent for AD.

        Aralia elata, Chaenomeles sinensis fruit, and Glycyrrhizae radix have been widely used as oriental medicinal plants in Korea, China and Japan and found to possess anti-oxidative and anti-inflammatory activities. The current study was conducted to investigate the neuroprotective effect of an ethanol extract of a mixture of A. elata, C. sinensis fruit, and Glycyrrhizae radix (ACG) against ischemia-induced brain injury in rats and excitotoxic and oxidative neuronal death in primarily cultured rat cortical neurons. Transient focal cerebral ischemia was induced by 2 h middle cerebral artery occlusion followed by 24 h reperfusion (MCAO/R) in rats. Oral administration of ACG (10, 25, and 50 mg/kg) 30 min before MCAO, after 1 h of MCAO, and after 1 h of reperfusion reduced MCAO/R-induced brain infarct and edema formation. ACG also inhibited development of behavioral disabilities in MCAO/R-treated rats. Exposure of cultured cortical neurons to 500 μM glutamate for 12 h resulted in neuronal cell death. ACG (1, 10, and 50 μg/mL) inhibited glutamate-induced neuronal death. Furthermore, ACG inhibited 100 μM hydrogen peroxide (H2O2)- and hypoxia-induced neuronal death. These results suggest that the neuroprotective effect of ACG against ischemia-induced brain damage might be associated with its anti-excitotoxic and anti-oxidative activity and that ACG may have a therapeutic role for prevention of neurodegeneration in stroke.

        쥐참외뿌리 추출물이 항산화 활성 및 피부 세포에서의 독성을 확인하여, 피부에 효과적으로 사 용할 수 있는 기능성 소재로써의 활용 가능성을 확인해 보고자 하였다. 쥐참외뿌리 추출물의 항산화 활성 의 지표가 되는 총 폴리페놀과 총 플라보노이드 함량을 확인하였고, 피부에서의 Neutral red assay를 이용 하여 세포 독성을 확인하였다. 연구 결과, 총 폴리페놀과 총 플라보노이드의 함량은 농도 의존적으로 증가 하였다. 섬유아 세포인 HDF cell에서의 높은 생존율이 확인되었으며, B16F10 melanoma cel와 RAW 264.7 cell에서는 5 μg/mL부터 세포 생존율이 유의하게 낮아지는 것이 확인되었다. 본 연구 결과는 쥐참 외뿌리 추출물의 항산화 활성 및 피부 세포에서의 기초적인 자료로 사용가능할 것으로 사료되어 진다.

        Spinach (Spinacia oleracea L.), a green leafy vegetable, is well known as a functional food due to its biological activities. Vascular calcification is associated with several disease conditions including atherosclerosis, diabetes, and chronic kidney disease (CKD), and is known to raise the risk of cardiovascular diseases related morbidity and mortality. However, there are no previous studies that have investigated the effects of fermented spinach exract (FSE) against aortic and its underlying mechanisms. Therefore, this study investigated the effects and action of possible mechanisms of FSE on inorganic phosphate (PI)-induced vascular calcification in ex vivo mouse aortic rings. PI increased vascular calcification through calcium deposition in ex vivo aortic rings. FSE inhibited calcium accumulation and osteogenic key marker, runt-related transcription factor 2 (Runx2), and bone Morphogenetic Protein 2 (BMP-2) protein expression in ex vivo aortic rings. And, FSE inhibited PI-induced extracellular signal-regulated kinase (ERK) and p38 phosphorylation in ex vivo aortic rings. These results show that FSE can prevent vascular calcification which may be a crucial way for the prevention and treatment of vascular disease association with vascular calcification.

        This study was to investigate an analytical method for determining dieckol content in Ecklonia stolonifera extract. According to the guidelines of International Conference on Harmonization. Method validation was performed by measuring the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) of dieckol using high-performance liquid chromatography–photodiode array. The results showed that the correlation coefficient of calibration curve (R2) for dieckol was 0.9997. The LOD and LOQ for dieckol were 0.18 and 0.56 μg/mL, respectively. The intra- and inter-day precision values of dieckol were approximately 1.58-4.39% and 1.37-4.64%, respectively. Moreover, intra- and inter-day accuracies of dieckol were approximately 96.91- 102.33% and 98.41-105.71%, respectively. Thus, we successfully validated the analytical method for estimating dieckol content in E. stolonifera extract.

        The objectives of this study are to increase the contents of functional compounds and antioxidant activity obtained from ginger (Zingiber officinale Roscoe) extracts using fermentation and enzyme. The optimal conditions (1%, Aspergillus luchuensis) for extraction methods were determined through fermentation treatment of ginger (concentrations of 0.5 and 1%; Aspergillus oryzae and Aspergillus luchuensis). The quality characteristics of the ginger extract for each treatment method (HW, hot water treated; EZ, enzyme; AL, fermentation; ALEZ, fermentation and enzyme) were observed using the optimal conditions. The water-soluble index (WSI) and total sugar content of the ALEZ increased by 2.6 times and 6.5 times, respectively, compared with that of the HW. However, the sum of gingerol and shogaol contents of ALEZ were 73.9% in ratio compared with lower than that of the HW. Antioxidant contents were generally higher in ALEZ, similar to the WSI trend. Therefore, ALEZ was more effective in enhancing antioxidant activity than EZ and AL. The fermentation and enzymatic approach described in this study would be beneficial to food industries for developing ginger functional products and materials.

        This study investigated optimal extraction conditions for applying Capsicum annuum L. leaf as a functional food resource. Capsicum annuum L. leaf was extracted at different extraction solvents (water and 95% ethanol), extraction temperatures (80oC and 100oC), and e xtraction times ( 30, 60, and 9 0 min), a nd t he extracts were e valuated for extraction yield, luteolin content as a major flavonoid component, antioxidant activity, and α-glucosidase inhibitory activity. The extraction yield, luteolin content, DPPH and ABTS radical scavenging activities, and α-glucosidase inhibitory activity of the hot water extract were higher than those of the ethanol extract. In evaluating the extraction temperature of Capsicum annuum L. leaf, the antioxidant activities were similar, but the extraction yields, luteolin contents, and α-glucosidase inhibitory activities were higher at 100oC extraction. In evaluating the extraction time of Capsicum annuum L. leaf, extraction yield increased as the extraction time increased, but antioxidant activity and α-glucosidase inhibitory activity were the highest at 60 min of extraction. These results suggest that the optimum extraction conditions of Capsicum annuum L. leaf are hot water for 60 min at 100oC, and the extracts can be used as a functional food resource.
